The objective of this study was to investigate serum lipids, metabolic parameters and activity of the anti-oxidative enzyme paraoxonase-1 (PON1). The study was conducted on non-pregnant heifers with optimal health status and on healthy dairy cows in the period of intensive lactation, assuming that the energy and metabolic demands during lactation reduce anti-oxidative protection. Total cholesterol and HDL-cholesterol concentrations were significantly higher (P < 0.05) in lactating cows than in heifers. Bilirubin concentration and γ-glutamyltransferase (GGT) activity were also significantly higher in lactating cows (P < 0.05), indicating increased hepatic efforts of cows to meet energy requirements for lactation. Significantly lower PON1 activity and PON1/HDL ratio in lactating cows compared to heifers (P < 0.05) showed that metabolic efforts during pregnancy, parturition and lactation influence PON1 activity due to oxidative stress. Concurrent increase in total and HDL-cholesterol during lactation indicated that the HDL particle is a major carrier of cholesterol in cows.  相似文献

用低维生素E和添加不饱和脂肪酸的日粮饲喂雏鸡,建立VE缺乏实验动物模型,采用荧光偏振法检测红细胞膜流动性,高效薄层层析法检测红细胞膜组分。结果表明,VE缺乏组雏鸡红细胞膜磷脂酰胆碱(PC)、磷脂酰乙醇胺(PE)含量降低,神经鞘磷脂(SM)、磷脂酰丝氨酸(PS)含量升高,膜流动性下降,组间差异显著或极显著(P<0.05,P<0.01)。提示VE对维持雏鸡红细胞膜结构和功能起重要作用。  相似文献

Recent work has suggested that the recommended intakes of vitamin E for dairy cattle need to be increased, particularly in dry cows. However, these suggestions are based on data from cattle in the USA, which may have significantly different oxidative stresses than European cattle. This study, which involved 594 cattle on three dairy farms, was designed to determine the effect of increased vitamin E supplementation on the health and fertility of UK dairy cows. Cattle were randomly allocated to receive either two intramuscular injections of 2100 mg of vitamin E (and 7 g of sodium selenite) 2 weeks before calving and on the day of calving, or no additional vitamin E supplementation. Although supplementation had no effect on milk yield, reproductive efficiency, or incidence of uterine infections, supplemented cattle had a lower risk of culling and a lower rate of mastitis. These figures were economically significant but not statistically significant at the 10% level. Supplementation reduced the incidence of retained fetal membranes from 6.5% to 3%, an effect which was almost significant at the 5% level. If these data are representative they suggest that vitamin E recommendations for UK cattle should be reassessed.  相似文献

In addition to its basic role in the metabolism of purine nucleotides, xanthine oxidoreductase (XOR) is involved in the generation of oxygen-derived free radicals and production and metabolic fate of nitric oxide (NO). Growth hormone (GH) and Vitamin E (E) have been shown previously to modify immune response to infection. Our objective was to determine in heifers the effect of endotoxin challenge (LPS; 3.0 μg/kg BW, i.v. bolus, Escherichia coli 055:B5) on xanthine oxidase (XO) activity in plasma and liver and the modification of this response by daily treatment with recombinant GH (0.1 mg/kg BW, i.m., for 12 days) or GH+E (E: mixed tocopherol, 1000 IU/heifer, i.m., for 5 days). In experiment 1, 16 heifers (348.7±6.1 kg) were assigned to control (C, daily placebo injections), GH, or GH+E treatments and were challenged with two consecutive LPS injections (LPS1 and LPS2, 48 h apart). After LPS1, plasma XO activity increased 290% (P<0.001) at 3 h, reached peak (430%) at 24 h and returned to basal level by 48 h after LPS2. XO responses (area under the time×activity curve, AUC) were greater after LPS1 than LPS2 (P<0.001). Total plasma XO responses to LPS (AUC, LPS1+LPS2) were augmented 55% (P<0.05) over C with GH treatment but diminished to C responses in GH+E. There was a linear relationship (r2=0.605, P<0.001) between total response in plasma XO activity and plasma nitrate+nitrite concentration. In experiment 2, 24 heifers (346±6 kg) were assigned to C or GH treatments and liver biopsy samples were obtained at 0, 3, 6, and 24 h after a single LPS challenge. Hepatic XO activities increased 63.3% (P<0.05) 6 h after single LPS challenge and remained elevated at 24 h (100.1%, P<0.01) but were not affected by GH treatment. Results indicate that LPS-induced increases in plasma XO activity could be amplified by previous GH treatment but attenuated by E administration. The data also suggest that E may be effective in controlling some mediators of immune response associated with increased production of NO via the effect on XO activity and its production of superoxide anion as well as uric acid.  相似文献

Background

Increasing the polyunsaturated fatty acid (PUFA) content and decreasing the saturated fatty acid (SFA) content of mutton can help to improve its nutritional value for consumers. Several laboratories have evaluated the effects of vitamin E on the fatty acid (FA) composition of muscle in sheep. However, little information is available on wool sheep, even though wool sheep breeds are an important source of mutton, especially in northern China where sheep are extensively farmed. The present study was designed to address the effects of vitamin E on muscle FA composition in male Aohan fine-wool sheep.

Methods

Forty-two male Aohan fine-wool lambs (5 mo old) with similar initial body weight were randomly divided into seven groups and fed diets supplemented with 0 (control group), 20, 100, 200, 1,000, 2,000, or 2,400 IU/sheep/d vitamin E for 12 mo. Three lambs from each group were slaughtered to measure vitamin E and FA content in the longissimus lumborum (LL) and gluteus medius (GM) muscles.

Results

Vitamin E concentrations in the LL and GM increased significantly after 12 mo of vitamin E supplementation (P < 0.05). However, this increase did not occur in a dose-dependent manner because the muscle vitamin E concentration was highest in the 200 IU/sheep/d group. Dietary vitamin E supplementation also caused a significant reduction in SFA content and an increase in monounsaturated FA (MUFA) content in the LL and GM (P < 0.05). All six doses of vitamin E significantly increased cis9 trans11-conjugated linoleic acid (c9t11-CLA) content in the LL compared with the control group (P < 0.05).

Conclusions

Dietary supplementation with vitamin E increased muscle vitamin E content and improved the nutritional value of mutton by decreasing SFA content and increasing MUFA and c9t11-CLA contents in Aohan fine-wool sheep. These effects were greatest in sheep fed a diet containing 200 IU/sheep/d vitamin E.  相似文献

目的:研究裙带菜膳食纤维对大鼠红细胞膜及血小板细胞膜流动性,为裙带菜膳食纤维利用提供实验依据。方法:将60只正常大鼠分为正常对照组,水不溶性裙带菜膳食纤维低剂量组(5%)、高剂量组(10%),连续30d颈总动脉插管取抗凝血,测定其荧光偏振度ρ、求得微粘度η、细胞膜流动LFU。结果:裙带菜膳食纤维组的血小板细胞膜的流动性明显高于对照组(P〈0.05),而荧光偏振度及血小板细胞的微粘度明显低于对照组(P〈0.05,P〈0.01)。结论:裙带菜膳食纤维可增强大鼠红细胞膜及血小板细胞膜的流动性。  相似文献

Four dogs with poor semen quality, low seminal plasma superoxide dismutase (SOD) activity and low blood plasma testosterone (T) levels were orally administered one vitamin E tablet containing 50 mg α-tocopheryl acetate per dog daily for 4 weeks. The mean values of semen quality were temporarily improved after the start of vitamin E treatment and the values of 4, and 5 weeks after that were significantly different from those before the treatment (P<0.05–0.001). The mean blood plasma T and seminal plasma SOD activity values slightly increased in the 4 dogs after the treatment. The results of the present study indicate that poor semen quality in dogs with low seminal plasma SOD can be improved by vitamin E treatment.  相似文献

We assessed the effect of an anionic salt and supplementary administration of vitamin E and Se throughout the dry period, on the risk of retained fetal membranes (RFM), milk yield and reproductive performance of dairy cows. Data were collected from 456 dairy cows in three commercial farms. Each animal entering the dry period was assigned to one of two groups (treated and control group). All animals were then fed the same ration, which included 80 IU/kg vitamin E acetate and 0.2 ppm Se, but animals of the treated group also received an additional blend containing ammonium chloride (60 g), vitamin E [1000 IU (dl-α-tocopheryl acetate)] and Se (0.05 ppm). Calving ease was evaluated and no manual removal of placenta was attempted. Cows that retained their fetal membranes for more than 12 h after calving were considered to suffer from RFM. All animals experienced a 50-day voluntary waiting period before the first artificial insemination (AI). Treatment resulted in a decrease in the estimated average risk of RFM (10.6% vs 17.8%). Stratifying on farm the Mantel–Haenszel Relative Risk (MH-RR) estimate of RFM in treated animals relative to controls was 1.68 (95% confidence interval, CI, 1.05 to 2.68) and the Mantel–Haenszel test P-value was 0.028. Among cows that had not required assistance during delivery the average risk of RFM was greater for those without treatment (risk estimate: 10/208 or 9.7%) as compared to those with treatment (risk estimate: 20/207 or 4.8%). The MH-RR estimate of RFM in treated vs. control animals was 2.95 the respective 95% CI: (0.96 to 4.17) and the MH test of association P-value was 0.058). Conversely, among cows that did require assistance during calving, the average risk of RFM estimates were 77.8% (14/18) and 91.3% (21/23) in treated and non-treated animals, respectively (MH-relative risk estimate of RFM = 1.21, 95%CI: 0.91 to 1.60). Using analysis of variance with farm as a random effect, treatment did not appear to have an effect on milk production of cows without RFM either at 30 or 60 days postpartum (P > 0.10). Time intervals (in days) between parturition and first oestrus expression (POI), parturition and first AI (PFAI) and parturition and conception (PCI) were recorded for each animal. For each time-to-event variable, survivor functions were estimated for treated and control groups using the Kaplan–Meier methodology and survival curves of treated and control groups were compared using the log-rank test, separately for the animals that did or did not experience RFM in each farm. There were significant differences in the time-to-event survival curves in only one of the three farms. Median time between parturition and first oestrus expression among animals without RFM was 67 days in treated animals and 75 in control animals, and the survival curves were statistically significantly different (P = 0.021). Similarly, in the same farm, among animals that did not experience RFM the median time between parturition and conception was 114 in treated animals and 145 in controls, and the survival curves were statistically significantly different (P = 0.002). In conclusion, daily administration of a blend containing ammonium chloride, vitamin E and Se throughout the dry period seems to be safe and resulted in a decrease of RFM occurrence, without any effect on milk yield.  相似文献

As a result of research conducted in the US, recommendations for dry cow vitamin E intakes have increased seven fold there, however there has been no change to recommendations in the UK. As part of a larger study comparing the impact of existing UK and new US recommended vitamin E intakes on the health and fertility of commercial dairy cows in the UK, a study was set up to investigate the effect of route of supplementation and stage of lactation, over a 21 day period, on the response to mega-supplementation of cattle receiving supposedly adequate vitamin E. The study assessed the response of dry, peak lactation and mid lactation cows to in-feed or parenteral vitamin E supplementation (7 animals per treatment/lactation stage group) by measuring plasma and milk vitamin E concentrations, blood glutathione peroxidase (GSH-Px) activity and milk yields over a 21 day period. Plasma vitamin E concentrations were significantly influenced by a time, stage and treatment interaction (P = 0.046). Both dry and lactating animals had significantly higher plasma vitamin E concentrations at some time points in the parenteral supplemented cows compared to the in-feed supplementated animals (P ≤ 0.011 and P < 0.01, respectively). Milk vitamin E concentrations did not significantly differ between lactation stages but treatment had a significant effect on concentrations (P < 0.008) when lactation stage was removed from the model. There was no significant difference in milk yield between treatment groups. A significant relationship between plasma and milk vitamin E concentrations was only found in the parenterally supplemented cows (r = 0.435, P < 0.001). In cattle with intakes greater than the ARC recommendations, measurement of plasma vitamin E concentration may be of limited value in determining whether there has been a response to supplementation. The relationship between plasma and milk vitamin E concentrations is too poor for milk vitamin E concentrations to be used as a proxy for plasma vitamin E.  相似文献

Fifteen healthy dogs received a basal diet supplemented with either 12.4 g of sunflower oil, 0.6 g of sunflower oil and 7 g of menhaden fish oil, or 0.6 g of sunflower oil and 7 g of menhaden fish oil plus 0.18 g of alpha-tocopherol acetate for twelve weeks. There was no significant diet effect on platelet aggregation, lipid peroxidation, or standard hematologic and biochemical parameters, with the exception of decreased triglycerides in dogs supplemented with fish oil. These data demonstrate that this level of fish oil supplementation in dogs does not require vitamin E supplementation above recommended dosage and may prove beneficial in the treatment of hyperlipidemia in the dog.  相似文献

This study was conducted to exploit ameliorative effect of additional vitamin E and/or zinc supplementation on immune response of peripartum Sahiwal cows. Thirty‐two pregnant dry Sahiwal cows were blocked into four treatment groups (n = 8), namely control, zinc (Zn), vitamin E (Vit E) and zinc + vitamin E (Zn + Vit E). Feeding regimen was same in all the groups except that the Sahiwal cows in the zinc‐, vitamin E‐ and zinc + vitamin E‐fed groups were additionally supplemented with 60 mg Zn/kg DM, 1000 IU vitamin E and 60 mg/kg + 1000 IU Zn + vitamin E, respectively, from day 60 pre‐partum to day 90 post‐partum. Blood samples were collected on days ?60, ?45, ?30, ?15, ?7, ?3, 0, 3, 7, 15, 30, 45, 60, 90 and 120 with respect to day of parturition and analysed for total immunoglobulin (TIG), immunoglobulin G (IgG), interleukin‐2 (IL‐2), vitamin E (Vit E) and zinc (Zn) status. Before calving, cows showed a decrease in plasma TIG, IgG, IL‐2, Vit E and Zn levels. However, increased levels of plasma TIG, IgG, IL‐2, Vit E and Zn were observed after calving. After calving, Sahiwal cows supplemented with Zn + Vit E had higher plasma TIG, IgG and IL‐2 in comparison with cows of control and Zn + Vit E‐fed groups. In the present study, plasma vitamin E level was higher in Vit E‐fed and Zn + Vit E‐fed cows; however, zinc level was higher in Zn‐ and Zn + Vit E‐supplemented cows. In conclusion, a reduced immune response during peripartum period in Sahiwal cows was ameliorated by dietary vitamin E and zinc supplementation.  相似文献

The study aimed to investigate the effects of vitamin E injection for the prevention of transport stress on ewes. Kivircik ewes (2–3 years old, = 24) were randomly separated into three groups; G1 (Control) and G2 treated with 14 ml. saline as the placebo, G3 treated with 2100 IU/ind. DL-alpha-tocopherol acetate prior to transport. G2 and G3 were transported at 80 km/h for 4 h on a truck. Serum samples were obtained before (T0) and after (T1) transport. Serum cortisol, catalase, IgG, ceruloplasmin, C-reactive protein, complement component 4, interleukin-1 beta, tumour necrosis factor-alpha, glutathione peroxidase (GPx), superoxide dismutase, malondialdehyde analyses performed by ELISA, and serum alpha-tocopherol concentrations were evaluated by HPLC-UV. Wilcoxon and Kruskal–Wallis tests were used for statistical assessments (p < 0.05). Alpha-tocopherol concentrations were founded 1.22 ± 0.82, 0.27 ± 0.14 and 0.14 ± 0.07 µmol/L, respectively, in G1, G2 and G3 at T1. Alpha-tocopherol concentration decreased significantly in G2 between T0 and T1. GPx concentrations were increased twofold in G2 and G3 between T0 and T1 (p < 0.01). As a result, G2 alpha-tocopherol concentrations decreased but, the stress and oxidative parameters tested in this study were not affected by treating 2100 IU/ind. The aim of the study was to investigate the ability of vitamin C to increase the antioxidative and immunomodulating potential in healthy dogs. Fifteen dogs were tested for the effects of orally administered vitamin E (60 mg dl‐α tocopheryl acetate) in combination with vitamin C (0, 30 or 60 mg ascorbic acid crystalline). Three treatments (0, 30, 60 mg vitamin C) were tested in a 3 × 3 cross‐over study in three periods of 36 days. Pre‐prandial blood samples were taken for analysis of vitamins C, E, A, retinyl palmitate and stearate, antioxidant status [Thiobarbituric acid reactive substances (TBARS) and uric acid], biochemical and haematological analysis. Subpopulations of lymphocytes, mitogen‐induced peripheral blood mononuclear cell proliferation (PBMC) and serum IgA and IgG concentrations were determined. There was a trend (p = 0.056) for an increased plasma vitamin C concentration by vitamin C supplementation. There was no evidence that dietary treatment altered neither the other plasma vitamin concentrations nor TBARS and uric acid concentrations nor the subpopulations of the lymphocytes except for the number of CD4+ lymphocytes that increased with vitamin C supplementation. There was no effect of vitamin C on serum IgA and IgG concentration. A significant time × treatment interaction was demonstrated on PBMC’s to pokeweed, with an increase observed by 30 mg vitamin C supplementation but a decrease by 60 mg vitamin C supplementation. There was no clear evidence for an effect of dietary vitamin C on antioxidative capacity in healthy dogs fed a diet with vitamin E concentrations well above the recommendations. Yet, a limited number of immunological parameters were slightly affected.  相似文献

The aim of this study was to evaluate the stress response to long-term transportation and to analyze the effects of vitamin A + D + E and vitamin E + selenium administrations on oxidative stress induced by long-term transportation in Holstein dairy cows. Control group (n = 10) received 0.9% saline (placebo), Group I (n = 10) received vitamin A + D + E, and Group II (n = 10) received vitamin E + selenium. After the transport, adrenaline levels were high in all groups (P < 0.05). Noradrenaline level was low in Group I (P < 0.05) and tended to be low in Group II. Mean levels of serum cortisol decreased significantly in all groups (P < 0.05) when comparing pre-transport data with post-transport data. Glucose levels increased in Control group (P < 0.05) and in Groups I and II (P < 0.01). The malondialdehyde level significantly increased in the Control group (P < 0.05) after the transport and the levels were lower in Group I (P = 0.076) and Group II (P < 0.05) than in the Control group. As a result, vitamin E + selenium and vitamin A + D + E have been determined to prevent lipid peroxidation and oxidative stress, associated with long-term transportation stress in cattle.  相似文献

Pregnancy is a physiology state accompanied by high energy and oxygen demand that may lead to increased level of oxidative stress and development of metabolic and reproductive disorders in pregnant water buffaloes. In the present study, the alterations in serum cortisol and erythrocyte lipid peroxides and superoxide dismutase activities were examined in 28 pregnant water buffaloes supplemented with antioxidant nutrients, Vitamin E and selenium. Another 14 buffaloes did not receive any treatment during pregnancy to serve as negative control. Results of the present study suggested that pregnancy is associated with oxidative stress and supplementation of vitamin E and selenium may be beneficial by alleviating oxidative stress in water buffaloes.  相似文献

Twenty dairy cows with left abomasal displacement were used to investigate the effects of vitamin E and selenium treatment on thiobarbituric acid reactive substances (TBARS) and blood cortisol in dairy cows stressed by omentopexy. The cows were randomly divided into two groups. Ten hours before surgery 6 g of DL‐α‐tocopheryl acetate (6 mg/kg) and 67 mg of natrium selenite (0.1 mg/kg) in volume of 40 ml (Vitaselen®) were administered subcutaneously to 10 cows; the control animals (n = 10) received an equivalent volume of injectable water (40 ml). The injection of vitamin E and selenium produced a rapid rise (p < .05) in blood α‐tocopherol and selenium concentrations. The serum vitamin E increased several times 10 hr after vitamin E and Se injection and raised continuously to the highest average concentration 21.6 mg/L at hr 24 after the surgery. The highest selenium concentration was seen 10 hr after selenium administration with holding the increased concentrations in comparison with initial ones during the whole study. Two‐way ANOVA did not show significant treatment effect on plasma concentrations TBARS in the study. The plasma concentrations of thiobarbituric acid reactive substances reached the maximum value of 0.18 μmol/L in the control group 5 hr after the surgery. Twenty‐four hours after the surgery, the TBARS values returned to the initial ones. Serum cortisol increased in both groups after surgery. The highest cortisol concentrations were reached at 1 hr after surgery in the experimental and control group (56.7 ± 28.8 and 65.3 ± 26.1 μg/L respectively). A return to the levels similar to the initial ones was recognized 24 hr after the surgery. The ANOVA revealed a significant effect of vitamin E and selenium injection on plasma cortisol (p < .05). In conclusion, we have demonstrated that abdominal surgery resulted in typical stress changes with no significant effects of a single vitamin E/Se injection on blood lipid peroxidation. In addition, a weaker cortisol response to the abdominal surgery was recognized in animals treated with vitamin E and selenium.  相似文献

The effects of vitamin E and vitamin E-selenium combination on seminal plasma arginase activity and nitric oxide level and some spermatological properties in rams were investigated in this study. For control group, animals were injected intramuscularly with physiological saline. For vitamin E group, rams were injected intramuscularly with 300 mg/ram vitamin E. For vitamin E + selenium group, animals were injected intramuscularly with 5 ml/ram vitamin E + selenium. The semen was collected by artificial vagina at 1st, 4th, 24th, 48th and 72nd hr after administration in each group. Significant decreases in seminal plasma arginase activity (at 1st, 24th and 48th hr), nitric oxide level (at 72nd hr) and abnormal sperm rate (at 1st, 24th and 72nd hr), and significant increases in semen volume (at 24th hr), semen mass activity (at 24th and 48th hr), sperm motility (at 24th, 48th and 72nd hr) and concentration (at 1st hr) were observed in vitamin E group compared with control group. Similarly, significant increase in semen volume (at 1st, 24th and 48th hr), mass activity, (at 48th hr), motility (at 48th and 72nd hr) and concentration (at 4th, 24th and 48th hr), and significant decrements in abnormal sperm rate (at 1st, 24th, 48th and 72nd hr), seminal plasma nitric oxide level (at 1st, 4th, 24th and 48th hr) and semen pH (at 24th and 48th hr) were detected in vitamin E + selenium group in comparison to the control group. As a result, it is suggested that vitamin E and/or vitamin E + selenium applications may improve reproductive performance.  相似文献
