Experiments consisted of three phases: spore production, spore storage and spore germination. Throughout the experiments temperature was near-optimal. In each phase various light and humidity treatments were given. The time at which a spore germinated was considered to be a stochastic variable. 3,000 lux inhibits germ tube elongation measurably; 7,600 lux inhibits germination measurably. Spores produced in darkness are more sensitive to light than spores produced in light, independent of the age of the pustules. Hydration of spores during storage increases light-sensitivity during germination. Samples of spores formed in darkness at low humidity are considered to consist of three sub-populations: a light-insensitive one presumably having profited from the foregoing light period, a light-sensitive sub-population and a group of spores which is inert or dead. The light-inhibited spores germinate rapidly after being placed in darkness with a germination rate little affected by the duration of light exposure. After four hours of light-exposure a gradually increasing proportion of the light-inhibited spores is killed, which means that the reversibility of the light-inhibition is limited. 相似文献

The accumulation of H2O2 (oxidative burst) and the progress of pathogen development were studied in compatible and incompatible wheat‐brown rust interactions. The accumulation of H2O2 was detected in 98·7% of guard cells with appressoria 8 h post inoculation (hpi). The reaction in both susceptible and resistant plants declined 2–3 days post inoculation (dpi). The second phase of the oxidative burst was observed in the mesophyll and/or epidermis. In susceptible plants it began 4–5 dpi and was detected only in the epidermis. In resistant plants the response was observed in the mesophyll. In moderately resistant plants it was induced 1–3 dpi, and the percentage of infection units reached 80–90% 8 dpi. This corresponded with severe necrotic symptoms. In highly resistant plants, the oxidative burst was short and transient. The percentage of infection units with H2O2 accumulation reached its highest level (60–70%) 2 dpi, and decreased thereafter. Four days later, the low percentage and weak DAB staining indicated very low H2O2 accumulation. The localization and the time‐course changes of the oxidative burst correlated with the profiles of the micronecrotic response, haustorium mother cell formation and pathogen development termination. An early and localized induction of oxidative burst followed by its rapid quenching correlated with high resistance and almost no disease symptoms. The possible correlation of the oxidative burst and pathogen development patterns with the level and durability of resistance conferred by Lr genes are discussed. 相似文献

A model simulating the progress of Puccinia recondita severity, expressed as a percentage of rusted leaf area (both as average and its 95% confidence interval) on individual wheat leaves over the course of a growing season, with a time step of one day, was elaborated using laboratory and field data from literature. Data on the stages of each infection cycle (uredospore germination, penetration, latency, uredium eruption and infectiousness) were transformed into model parameters by curve fitting, Montecarlo stochastic procedures, corrections and empirical assumptions. Data on host growth, like the timing of all phenological stages, the dynamic of the green area of each leaf from appearance to complete senescence, and tillering were obtained from a specific sub-model. Model validation was performed on actual data not used in model building and representing a wide range of conditions (several winter wheat cultivars grown at eight locations in northern Italy between 1990 and 1994) by using subjective, non-parametric and parametric tests: it revealed a satisfactory agreement between the data simulated by the model and actual data. 相似文献

The goal of this work was to establish which enzymes – peroxidases or NADPH oxidases – play the most important role in the resistance‐related oxidative burst response of wheat to infection by brown rust (Puccinia triticina). The expression of four peroxidases and two NADPH oxidases was analysed in the susceptible wheat cv. Thatcher and isogenic lines with different Lr resistance genes after pathogen inoculation. Of the peroxidases, TaPrx118 and TaPrx112 were induced several times more strongly than TaPrx103 and TaPrx107. The induction of peroxidases was more pronounced than that of NADPH oxidases. The patterns of peroxidase expression clearly differentiated moderately resistant from highly resistant lines and corresponded to oxidative response profiles. The possible involvement of peroxidases or NADPH oxidases was verified with enzyme‐specific inhibitors. The oxidative burst in the susceptible cv. Thatcher and in the lines TcLr24, TcLr25, TcLr9 was peroxidase‐dependent, while the response in line TcLr26 was NADPH‐oxidase‐dependent. It is postulated that class III peroxidases play a leading role in the formation of reactive oxygen species molecules during the response of wheat to pathogen infection. The results suggest a high level of redundancy of some peroxidase genes induced in biotic stress. The role of both enzyme systems in wheat response/resistance to brown rust is discussed in relation to the oxidative response, the efficiency of resistance, and the presence and origin of particular Lr resistance genes. 相似文献

由条形柄锈菌小麦专化型Puccinia striiformis f. sp.tritici(Pst)引起的条锈病,长期严重威胁小麦安全生产。Pst通过毒性变异产生新毒性小种或致病类型,能克服生产上已应用的抗病基因,导致抗病品种感病。因此,挖掘抗条锈病基因资源对小麦抗病遗传改良和加快抗病新品种培育具有重要意义。蛋白激酶在应答病原菌侵染,传递植物免疫信号中发挥重要作用。本研究通过小麦转录组数据分析,筛选到参与应答Pst、小麦白粉菌Blumeria graminis f. sp.tritici和赤霉病菌Fusarium graminearum侵染的蛋白激酶基因TaPiPK1。该蛋白激酶基因编码551个氨基酸,C末端含有一个STK激酶结构域。利用RT-qPCR检测发现,TaPiPK1在小麦与条锈菌非亲和互作的前期上调表达;TaPiPK1主要定位于细胞质与细胞膜。利用大麦条纹花叶病毒介导的基因沉默技术(BSMV-VIGS)瞬时沉默TaPiPK1,显著降低小麦‘水源11’对条锈菌非亲和小种CYR23的抗性,表明TaPiPK1参与小麦抗条锈病反应并发挥重要作用。因此,TaPiPK1有可能作为潜在的基... 相似文献

Members of a new class of fungicide containing benzylaminopyrimidine as a core structure were synthesized and their fungicidal potencies against wheat brown rust, Puccinia recondita, and barley powdery mildew, Erysiphe graminis, were assessed. Among these fungicides, N-(fluoroalkoxy or fluorophenoxybenzyl)-4-pyrimidinamines showed notable preventive activities. The potency of the new pyrimidines was increased when a difluoromethoxy or tetrafluorophenoxy group was introduced at the 4- or 3-position of the phenyl moiety and a methyl or ethyl group was introduced at the benzyl position. Promising wheat lines from FAO's International Program for Horizontal Resistance (IPHR) in Brazil and Zambia were tested in the Netherlands against brown rust (Puccinia recondita. f.sp.tritici). Race nursery tests were performed on isolated field plots using five monopustular isolates from the Netherlands representing at least four races. Mature plants of several lines showed high incomplete resistance. For most lines, the reactions to the different races showed a high uniformity. Apparently, lines selected according to the IPHR principles can perform well when exposed to brown rust isolates to which they have not been exposed during selection.Samenvatting Veelbelovende tarwelijnen uit het Internationale Programma voor Horizontale Resistentie (IPHR) van de FAO in Brazilië en Zambia werden in Nederland getoetst tegen bruine roest (Puccinia recondita f.sp.tritici). Hierbij werd gebruik gemaakt van de fysiovelden-techniek. Geïsoleerde fysiovelden werden geïnoculeerd met vijf Nederlandse isolaten, ieder afkomstig van een sporenhoopje. Deze vijf isolaten vertegenwoordigden ten minste vier fysio's. Bij verscheidene lijnen vertoonden de volwassen planten een hoge mate van onvolledige resistentie. De meeste lijnen vertoonden in hun reactie op de verschillende fysio's een hoge mate van uniformiteit. Kennelijk kunnen lijnen, geselecteerd volgens de beginselen van het IPHR goede resultaten geven wanneer zij getoetst worden met roest-isolaten, waaraan zij tijdens het selectieproces nooit zijn blootgesteld. 相似文献

ABSTRACT Stripe rust is one of the most important diseases of wheat and barley worldwide. On wheat it is caused by Puccinia striiformis f. sp. tritici and on barley by P. striiformis f. sp. hordei Most wheat genotypes are resistant to P. striiformis f. sp. hordei and most barley genotypes are resistant to P. striiformis f. sp. tritici. To determine the genetics of resistance in wheat to P. striiformis f. sp. hordei, crosses were made between wheat genotypes Lemhi (resistant to P. striiformis f. sp. hordei) and PI 478214 (susceptible to P. striiformis f. sp. hordei). The greenhouse seedling test of 150 F(2) progeny from the Lemhi x PI 478214 cross, inoculated with race PSH-14 of P. striiformis f. sp. hordei, indicated that Lemhi has a dominant resistance gene. The single dominant gene was confirmed by testing seedlings of the F(1), BC(1) to the two parents, and 150 F(3) lines from the F(2) plants with the same race. The tests of the F(1), BC(1), and F(3) progeny with race PSH-48 of P. striiformis f. sp. hordei and PST-21 of P. striiformis f. sp. tritici also showed a dominant gene for resistance to these races. Cosegregation analyses of the F(3) data from the tests with the two races of P. striiformis f. sp. hordei and one race of P. striiformis f. sp. tritici suggested that the same gene conferred the resistance to both races of P. striiformis f. sp. hordei, and this gene was different but closely linked to Yr21, a previously reported gene in Lemhi conferring resistance to race PST-21 of P. striiformis f. sp. tritici. A linkage group consisting of 11 resistance gene analog polymorphism (RGAP) markers was established for the genes. The gene was confirmed to be on chromosome 1B by amplification of a set of nullitetrasomic Chinese Spring lines with an RGAP marker linked in repulsion with the resistance allele. The genetic information obtained from this study is useful in understanding interactions between inappropriate hosts and pathogens. The gene identified in Lemhi for resistance to P. striiformis f. sp. hordei should provide resistance to barley stripe rust when introgressed into barley cultivars. 相似文献

Aegilops umbellulata, a non‐progenitor diploid species, is an excellent source of resistance to various wheat diseases. Leaf rust and stripe rust resistance genes from A. umbellulata were transferred to the susceptible wheat cultivar WL711 through induced homoeologous pairing. A doubly resistant introgression line IL 393‐4 was crossed with wheat cultivar PBW343 to develop a mapping population. Tests on BC2F7 RILs indicated monogenic inheritance of seedling leaf rust and stripe rust resistance in IL 393‐4 and the respective co‐segregating genes were tentatively named LrUmb and YrUmb. Bulked segregant analysis placed LrUmb and YrUmb in chromosome 5DS, 7.6 cM distal to gwm190. Aegilops geniculata‐derived and completely linked leaf rust and stripe rust resistance genes Lr57 and Yr40 were previously located in chromosome 5DS. STS marker Lr57/Yr40MAS‐CAPS16 (Lr57/Yr40‐CAPS16), linked with Lr57/Yr40 (T756) also co‐segregated with LrUmb/YrUmb. Seedling infection types differentiated LrUmb from Lr57. Absence of leaf rust‐susceptible segregants among F3 families of the intercross (IL 393‐4/T756) indicated repulsion linkage between LrUmb and Lr57. YrUmb expressed a consistently low seedling response under greenhouse conditions, whereas Yr40 expressed a higher seedling response. Based on the origin of LrUmb/YrUmb from the U genome and Lr57/Yr40 from the M genome, as well as phenotypic differences, LrUmb and YrUmb were formally named Lr76 and Yr70, respectively. These genes have been transferred to Indian wheat cultivars PBW343 and PBW550, and advanced breeding lines are being tested in state and national trials. 相似文献

The dose-response curve was quantified for the avian repellency of cinnamamide, a derivative of the plant secondary compound cinnamic acid, using a three-day, ‘short-term no-choice’ test. This test was designed to resemble more closely the availability of alternative food in the birds' natural environment than in previous no-choice studies. Cinnamamide reduced food consumption by feral pigeons (Columba livia Gmelin) at concentrations as low as 6.17 μmole g?1 food (0.09% w/w) and this reduction increased with concentration. An R50 index, the concentration at which food consumption was reduced by 50% was interpolated from the dose-response curve as 18.4 (±0.38) μmole g?1 food 10.26% w/w). This index can now be matched with those of related compounds in future structure-activity studies. The birds' response to cinnamamide changed over the three-day trial with respect to concentration. Repellency declined at concentrations < R50 and increased at concentrations > R50. This suggests that pigeons become habituated to cinnamamide at low concentrations but at high concentrations they may develop a conditioned aversion. This study provides quantitative confirmation of previous laboratory and field demonstrations of cinnamamide's effectiveness as a non-lethal chemical repellent suitable for reducing avian pest damage. 相似文献

Aegilops sharonensis (Sharon goatgrass) is a wild relative of wheat and a rich source of genetic diversity for disease resistance. The objectives of this study were to determine the genetic basis of leaf rust, stem rust, and powdery mildew resistance in A. sharonensis and also the allelic relationships between genes controlling resistance to each disease. Progeny from crosses between resistant and susceptible accessions were evaluated for their disease reaction at the seedling and/or adult plant stage to determine the number and action of genes conferring resistance. Two different genes conferring resistance to leaf rust races THBJ and BBBB were identified in accessions 1644 and 603. For stem rust, the same single gene was found to confer resistance to race TTTT in accessions 1644 and 2229. Resistance to stem rust race TPMK was conferred by two genes in accessions 1644 and 603. A contingency test revealed no association between genes conferring resistance to leaf rust race THBJ and stem rust race TTTT or between genes conferring resistance to stem rust race TTTT and powdery mildew isolate UM06-01, indicating that the respective resistance genes are not linked. Three accessions (1644, 2229, and 1193) were found to carry a single gene for resistance to powdery mildew. Allelism tests revealed that the resistance gene in accession 1644 is different from the respective single genes present in either 2229 or 1193. The simple inheritance of leaf rust, stem rust, and powdery mildew resistance in A. sharonensis should simplify the transfer of resistance to wheat in wide crosses. 相似文献

小麦条锈病是发生于甘肃陇南麦区小麦生产上的最主要病害,利用和种植抗病品种(系)是防治该病害最经济有效且有利于保护环境的措施。2011-2013年度连续两年在甘肃省农业科学院植物保护研究所甘谷试验站田间对86份来自贵州大学的贵协系小麦种质资源材料进行了成株期抗条锈病鉴定,结果发现仅有‘贵协1’、‘贵协2’、‘贵协3’、‘贵协7’、‘贵协011-2-6’、‘贵协011-3-16’、‘贵协011-3-29’、‘贵协011-3-31’、‘贵协011-3-34’等9份材料表现抗病,且抗病性相对稳定,可作为一线抗源材料在陇南抗病育种中充分利用。 相似文献

A lesion-mimic mutant was obtained from a mutagenic treatment performed with ethyl methanesulfonate on the Argentine bread wheat ( Triticum aestivum ) cultivar Sinvalocho M.A. The HLP (hypersensitive-like phenotype) mutant exhibited tiny, discrete, white lesions in the absence of any pathogen, resembling the typical hypersensitive response (HR). The lesions only became evident once the fifth or sixth leaf emerged, and spread at random along the leaf blades and leaf sheaths of the developing plant, including tissues of the spike. Because the lesion-mimic mutant showed no lesions at the seedling stage, the phenotypes of both the mutant and its mother line were identical at this point. Histochemical studies showed that spontaneous hypersensitive-like lesions in the HLP mutant corresponded to cell death. In leaf-rust ( Puccinia triticina ) infection experiments performed at seedling and adult-plant stages, adult HLP plants showed enhanced resistance to leaf-rust attack compared with plants of Sinvalocho M.A. of comparable developmental stage, suggesting that the HLP mutation may confer increased resistance to the fungus. Because enhanced resistance coincided with the presence of spontaneous HR lesions, activation of HLP plant defence responses appeared to be tightly linked to this phenomenon. Final plant height and yield components in the lesion-mimic mutant did not differ from those of the mother line, indicating that the HLP mutation caused no detrimental pleiotropic effects that significantly affected agronomic performance. These data support the direct use of mutations in disease-resistance breeding. 相似文献

为明确陕西省小麦条锈菌的群体结构、变异动态和新育成小麦品种(系)的抗病性,为病害流行预测、防治以及抗病育种提供依据,本研究于2002—2014年从陕西省8个市(区)的28个县(区)和毗邻的甘肃省、四川省和湖北省部分地区共采集鉴定小麦条锈菌标样2 779份,监测到条锈菌生理小种和致病类型45个,其中,CYR33和CYR32为目前陕西省小麦条锈菌主要流行小种,新致病类型G22-9和G22-14虽然目前出现频率不高,但对贵农系列、92R系列以及Moro均有毒性,且出现频率呈上升趋势。目前小麦条锈菌群体中Hybrid46致病类群和水源11致病类群占绝对优势,这与我国小麦品种抗病基因单一化有较大关系,应加强开发和利用新的、多元化的抗源材料。对2 952份陕西省新育成小麦品种(系)抗病性测试结果表明,其整体抗性水平呈上升趋势。综合条锈菌生理小种监测和抗病性分析结果,目前小麦抗条锈病育种应以抗CYR33和CYR32为主,同时注意对G22-9和G22-14的抗病性研究。 相似文献

在水源11、洛夫林10等7个品种上分别选择小麦条锈菌CY17、CY28等9个生理小种中非亲和小种进行诱发接种,亲和小种为挑战小种。观察发病后的病情指数并与单独接种诱发小种和单独接种挑战小种处理的病情指数进行比较。证明诱发抗病现象是比较普遍的,但表现程度因小麦品种、诱发小种和挑战小种而不同。诱发抗病性的表达时间可持续8 d,以诱发接种后1~2 d表达最强。诱发接种量与诱发抗病性表达呈指数函数关系,也间接证明这种诱发抗性是局部的。作者认为:诱发抗病性在品种、诱发和挑战小种不同组合之间的差异在小种动态中会起一定的作用。 相似文献
