Tea fruit peel is the main byproduct during manufacture of tea seed oil. The great increase in tea seed oil production in recent years brings the challenge of finding application for tea fruit peel. The aims of this study were to obtain tea fruit peel extracts enriched with bioactive compounds by several solvent extraction methods and to evaluate their antioxidant activity and inhibitory potential against α-glucosidase and α-amylase in vitro. Flavonoids and phenolics were accumulated in ethyl acetate, butanol, and chloroform fractions, and these fractions possessed much better antioxidant activities, including scavenging effect on 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH) and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) cation radicals (ABTS+), and reducing activity, compared with those of the 75% ethanol extract and water fraction. Moreover, the ethyl acetate, butanol, and chloroform fractions exhibited excellent inhibitory activity against α-glucosidase, much stronger than that of the positive control (acarbose). Wheat flour is generally supplemented with α-amylases to increase maltose levels in bread dough and increase loaf volume. While the preference of yeast for glucose and fructose over maltose as substrate for fermentation is well documented, the impact of maltose versus glucose producing enzymes on bread dough fermentation kinetics and bread sugar levels is ill documented. Hence the impact of α-amylase, α-glucosidase and glucoamylase action on both aspects was investigated. Glucoamylase and α-amylase increase the total fermentable sugar content of dough, while α-glucosidase only affects the glucose/maltose ratio. Due to their effect on total fermentable sugar levels, addition of α-amylase or glucoamylase prolongs the total productive fermentation time, while this is not the case for α-glucosidase. In contrast to α-amylase, both glucoamylase and α-glucosidase supplementation leads to higher CO2 production rates during the initial stages of fermentation. In the final bread product, different sugar levels are observed depending on the dosage and type of starch-degrading enzyme. The results of this study imply that long and short fermentation processes benefit from α-amylase and α-glucosidase addition, respectively, while glucoamylase supplementation is suitable for both long and short fermentation times. 相似文献

Bis (2,3-dibromo-4,5-dihydroxy-phenyl)-methane (BDDPM) is a natural bromophenol compound derived from marine algae. Previous reports have shown that BDDPM possesses antimicrobial activity. In the present study, we found that BDDPM has cytotoxic activity on a wide range of tumor cells, including BEL-7402 cells (IC50 = 8.7 μg/mL). Further studies have shown that prior to the onset of apoptosis, the BDDPM induces BEL-7402 cell detachment by decreasing the adherence of cells to the extracellular matrix (ECM). Detachment experiments have shown that the treatment of BEL-7402 cells with low concentrations of BDDPM (5.0 μg/mL) significantly inhibits cell adhesion to fibronectin and collagen IV as well as cell migration and invasion. High doses of BDDPM (10.0 μg/mL) completely inhibit the migration of BEL-7402 cells, and the expression level of MMPs (MMP-2 and MMP-9) is significantly decreased. Moreover, the expression of β1-integrin and focal adhesion kinase (FAK) is found to be down-regulated by BDDPM. This study suggests that BDDPM has a potential to be developed as a novel anticancer therapeutic agent due to its anti-metastatic activity and also indicates that BDDPM, which has a unique chemical structure, could serve as a lead compound for rational drug design and for future development of anticancer agents. 相似文献

In order to determine the absolute configuration of naturally occurring alloxanthin, a HPLC analytical method for three stereoisomers 1a–c was established by using a chiral column. Two authentic samples, (3S,3′S)- and meso-stereoisomers 1b and 1c, were chemically synthesized according to the method previously developed for (3R,3′R)-alloxanthin (1a). Application of this method to various alloxanthin specimens of aquatic animals demonstrated that those isolated from shellfishes, tunicates, and crucian carp are identical with (3R,3′R)-stereoisomer 1a, and unexpectedly those from lake shrimp, catfish, biwa goby, and biwa trout are mixtures of three stereoisomers of 1a–c. 相似文献

2,5-bis(3′-Indolyl)pyrroles, analogues of the marine alkaloid nortopsentin, were conveniently prepared through a three step procedure in good overall yields. Derivatives 1a and 1b exhibited concentration-dependent antitumor activity towards a panel of 42 human tumor cell lines with mean IC50 values of 1.54 μM and 0.67 μM, respectively. Investigating human tumor xenografts in an ex-vivo clonogenic assay revealed selective antitumor activity, whereas sensitive tumor models were scattered among various tumor histotypes. 相似文献

The study aimed to characterize and identify anti-diabetic and anti-hypertensive bioactive peptides generated upon enzymatic hydrolysis of quinoa protein isolates. Different quinoa protein hydrolysates (QPHs) were produced using food grade enzymes like Bromelain, chymotrypsin and Pronase E at a hydrolysis interval of 2 h up to 6 h. QPHs were characterized for their physicochemical properties using degree of hydrolysis, SDS-PAGE, and their anti-diabetic properties via inhibition of dipeptidyl peptidase-IV (DPP-IV) and α-glucosidase (AG), and anti-hypertensive property via inhibition of angiotensin converting enzyme (ACE) were explored. IC50 for DPP-IV, AG and ACE inhibitory activities of QPHs were in the range of 0.72–1.12, 1.00–1.86 and 0.18–0.31 mg/mL, respectively. The chymotrypsin derived 6 h hydrolysate (QC6) was sequenced for peptides identification and 136 peptides were identified among which 35 peptides were predicted as potential bio-active peptides (BAPs) based on their Peptide Ranker score. Results showed that identified peptides were predicted to possess high potential in inhibiting the DPP-IV, AG and ACE. In particular, QHPHGLGALCAAPPST was found to bind to the highest number of active hotspots of the target enzymes that are involved in their enzymatic activities. In conclusion, quinoa protein hydrolysates were identified as potential sources of BAPs with inhibitory properties towards key enzymes involved in the control of type 2 diabetes and hypertension. 相似文献

A rapid method for characterization and online detection of surfactin isomers was developed based on HPLC-MS(n) (n = 1, 2, 3) analyses, and many surfactin isomers were detected and characterized from the bioactive fraction of the mangrove bacterium Bacillus sp. Inhibitory activities of surfactin isomers on the overproduction of nitric oxide and the release of TNF-α and IL-6 in LPS-induced macrophages were systematically investigated. It was revealed that the surfactin isomers showed strong inhibitory properties on the overproduction of nitric oxide and the release of IL-6 on LPS-induced murine macrophage cell RAW264.7 with IC(50) values ranging from 1.0 to 7.0 μM. Structure-activity relationship (SAR) studies revealed that the existence of the free carboxyl group in the structure of surfactin isomers was crucial. These findings will be very helpful for the development of this novel kind of natural product as new anti-inflammatory agents. 相似文献

Pheromone-responsive neurons of insects not only require specific receptors but in addition several auxiliary components, including the “sensory neuron membrane protein,” SNMP. Accordingly, SNMP is considered as a marker for neurons responding to pheromones. For the desert locust Schistocerca gregaria, it is known that the behavior, including aggregation behavior and courtship inhibition, is largely controlled by pheromones. However, little is known about pheromones, their receptors, and the pheromone-responsive cells in locusts. In this study, we have identified two SNMP subtypes, SNMP1 and SNMP2, and compared their phylogenetic relationship and primary structure motifs with SNMPs from other species. Both SNMPs were found in chemosensory tissues, especially the antennae. Employing double in situ hybridization, we identified and localized the SNMP-expressing cells in the antennae. Cells expressing SNMP1 were localized to sensilla trichodea but also to sensilla basiconica, which in locust respond to pheromones. One or a few cells express SNMP1 within the multineuron clusters from sensilla basiconica, whereas the SNMP2 subtype was expressed in cells surrounding the neuron clusters, possibly supporting cells. Based on the finding that SNMP1 is expressed in distinct neurons under chemosensory sensilla, it is conceivable that these cells may represent pheromone-responsive neurons of the desert locust. 相似文献

Bioavailability trials of β-carotene from fresh and dehydrated drumstick leaves (Moringa oleifera) in a rat model 总被引:1,自引:0,他引:1
Male albino rats (Charles Foster, n = 40) were fed a synthetic diet deficient in vitamin A for 4 weeks. Six rats died during the depletion period. Of the 34 surviving, 5 rats were continued on the vitamin A deficient diet for 4 more weeks and 24 were repleted with vitamin A (4000 IU/kg diet) in the form of vitamin A acetate (group A, n = 8), fresh drumstick leaves (group B, n = 8) or dehydrated drumstick leaves (group C, n = 8) for 4 weeks. The remaining 10 rats were continued on the vitamin A adequate diet for 4 (n = 5) and 8 weeks, respectively (n = 5). A marked reduction in food intake, body weight, accompanied by clinical signs of vitamin A deficiency and a decline in serum vitamin A (29.2 to 19.1 g/dL) and liver vitamin A (3.7 to 2.0 g/dL) were seen at the end of 4 weeks of feeding a vitamin A deficient diet. On repletion significant improvements in clinical signs, food intake and body weights were noted in the three groups compared to the baseline (n = 5) and at the end of 4 weeks of depletion. The gain in body weight was highest for the group repleted with dehydrated drumstick leaves. Among the repleted groups, the serum vitamin A was highest for group A (34.7 g/dL) given synthetic vitamin A, compared to group B (25.8 g/dL) and group C (28.2 g/dL) given drumstick leaves. All these were significantly higher than the serum vitamin A values seen at the end of 4 weeks of depletion (19.1 g/dL). A significant improvement was also observed in the liver retinol levels on repletion for 4 weeks in the three groups, compared to the vitamin A depleted rats. These results imply that -carotene from drumstick leaves was effective in overcoming vitamin A deficiency although serum vitamin A levels remained somewhat lower compared to the group repleted with vitamin A acetate. In terms of growth parameters, the fresh and dehydrated drumstick leaves were better than the synthetic vitamin A. It is therefore concluded that in the developing countries like India, sources of vitamin A such as drumstick leaves are valuable in overcoming the problem of vitamin A deficiency. 相似文献

This study aimed at surveying the local calliphorid and sarcophagid species in
Maranhão State (Brazil) to determine their distribution and abundance, as well
as the distribution of exotic Chrysomya species. In total, 18,128
calliphorid specimens were collected, distributed in 7 genera and 14 species. The
species Hemilucilia semidiaphana (Rondani, 1850) and
Paralucilia paraensis (Mello, 1969) were new state records.
Chrysomya albiceps (Wiedemann, 1819) and Cochliomyia
macellaria (F., 1775) were the most abundant species, and the exotic
species of Chrysomya together contributed more than 50% of
total blow fly abundance. The abundance distribution of the calliphorid community
conformed to a log series model, characterized by a steep curve that reflects an
assemblage with a high degree of dominance. For the Sarcophagidae, a total of 14,810
specimens were collected and distributed in 15 genera, 11 subgenera, and 52 species.
Tricharaea (Sarcophagula)
occidua (F., 1794) and Peckia
(Sarcodexia) lambens (Wiedemann, 1830) were the
most abundant species. The abundance distribution of the species followed a log
normal model, with a gentler slope, consistent with a more uniform community. The
cumulative species curve for the sarcophagids did not reach the asymptote.
Forty-three sarcophagid species were new state records and 22 were new records for
the Brazilian northeast, which emphasizes the need for a continued survey in this

Background
Understanding the responses of rice to environmental stresses such as unscheduled submergence is of pressing important owing to increasing severity of weather thought to arise from global climate change. When rice is completely submerged, different types adopt either a quiescence survival strategy (i.e., minimal shoot elongation) or an escape strategy (i.e., enhanced shoot elongation). Each strategy can prolong survival depending on the circumstances. While submergence responses have been studied in rice typical of lowland and flood-prone areas, few studies have explored the physiological and molecular properties of upland rice under submergence. Here, we use seedlings of the upland rice ‘Tung Lu 3’ (‘TL3’) to analyze physiological and molecular responses to submergence. We compare them with those of ‘FR13A’, a lowland rice that tolerates submergence by adopting the quiescence strategy.Results
Plant height and distance between leaf sheaths, increased rapidly in ‘TL3’ under submergence. Although this indicated a strong escape strategy the seedlings remained totally underwater for the duration of the experiments. In contrast, ‘FR13A’ elongated much less. Consequently, after 4 days complete submergence followed by drainage, ‘TL3’ lodged much more severely than ‘FR13A’. After 10 d complete submergence, 55% of ‘TL3’ seedlings survived compared to 100% in ‘FR13A’. Chlorophyll a, b and total chlorophyll concentrations of the 2nd oldest leaves of ‘TL3’ were also significantly above those of ‘FR13A’ (but were lower than ‘FR13A’ in the 3rd oldest leaves) and less hydrogen peroxide accumulated in ‘TL3’. Peroxidase activity in submerged ‘TL3’ was also greater than in ‘FR13A’ 1 day after submergence. Quantitative RT–PCR showed increased expression of sucrose synthase 1 and alcohol dehydrogenases 1 after 2 days complete submergence with significantly higher levels in ‘TL3’ compared to ‘FR13A’. Expression was also higher in ‘TL3’ under non-submerged conditions.Conclusions
The upland rice line ‘TL3’ gave a stronger elongation response than ‘FR13A’ to complete submergence. This escape strategy is widely considered to prejudice survival when the plant remains totally submerged. However, contrary to expectations, ‘TL3’ survival rates were substantial although below those for ‘FR13A’ while physiological, biochemical and molecular parameters linked to adaptation differed in detail but appeared to be broadly comparable. These findings highlight that submergence tolerance is determine not only by the adoption of quiescence or escape strategies but maybe by metabolic and physiological properties unrelated to the underwater elongation rate.17.
Anthonomus signatus, the strawberry bud weevil, is active in early spring coinciding with the bloom period of host plants and when managed and unmanaged pollinators are active. Female A. signatus cause injury to the host plant during egg laying when they deposit a single egg inside an unopened flower bud and then girdle or “clip” the bud at the pedicel. Past efforts to control A. signatus populations have relied on the use of broad spectrum insecticides. Fields are either treated at the first signs of damage, or precautionary treatments may be applied prophylactically in fields with a history of damage. Because A. signatus damage occurs during bloom, there is concern about the potential harm to pollinators caused by these treatments. In order to identify materials more compatible for use during bloom, the efficacy of reduced risk pesticides against A. signatus was tested in semi-field bioassays over two years. Beauveria bassiana, cyantraniliprole, novaluron, and pyrethrins were assessed in the first year, and none caused significant A. signatus mortality. Acetamiprid, flonicamid, and spinosad were compared in the second year, acetamiprid and spinosad had higher A. signatus mortality than an untreated control, and exposure to spinosad resulted in similar mortality to bifenthrin, an industry standard material. 相似文献

Biodegradable block copolymers containing poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) and poly (ɛ-caprolactone) (PCL) units (PHCLs) with different contents of PCL block were synthesized successfully by using telechelic
hydroxylated PHBV (PHBV-diol) with low molecular weight as a macroinitiator to initiate ring-opening bulk polymerization of
ɛ-caprolactone (ɛ-CL). The chemical structure and molecular weight were characterized by 1H NMR, FTIR and GPC. The PHBV and PCL blocks in PHCLs
were miscible in amorphous state, and formed separate crystalline phases with lower crystallinity than corresponding homopolymers,
which was characterized by DSC and WAXD. The results of TGA showed that PHCLs underwent a two-step thermal degradation process.
The thermal degradation process of PCL blocks was significantly different from PCL homopolymers. The activation energies of
thermal degradation of PCL blocks calculated by Horowitz and Metzger method were much higher than that of each step of thermal
