Between August 1995 and June 1997 a survey to determine the distribution of tsetse-transmitted trypanosomosis was conducted in the Eastern Caprivi (Caprivi District, Namibia). A total of 1,481 adult cattle was examined at 33 sampling sites. Direct parasitological diagnostic tests were used and eluted blood spots were screened for the presence of anti-trypanosomal antibodies. Tsetse-transmitted trypanosomal infections were detected in 66 animals (4.5%) from 14 different locations. The parasitological and serological prevalence of trypanosomosis was highest in the Mamili area. Trypanosomosis was virtually absent in the Linyanti/Chobe area and the target barrier along the Kwando River had significantly reduced the prevalence of trypanosomosis in cattle grazing to the east of it. This suggests that anti-trypanosomal antibody prevalence data can be used to evaluate and monitor the effectiveness of tsetse control measures. Survey results suggest that in the Katima Mulilo area, trypanosomal infections were being acquired when cattle grazed along the Zambezi River. Moreover, survey results indicate that tsetse have not been able to establish themselves in the Katima Mulilo area. The parasitological prevalence in a herd and the respective prevalence of anti-trypanosomal antibodies was significantly correlated to the percentage of anaemic animals in that herd. Furthermore, the parasitological prevalence in a herd was positively correlated with the prevalence of anti-trypanosomal antibodies of that herd. It is concluded that the prevalence of anti-trypanosomal antibodies in a herd can be used as an additional indicator of the extent of infection in that particular herd. 相似文献

A study to assess the influence of re-invasion of Glossina pallidipes on the epidemiology of bovine trypanosomosis was conducted in Southeast Uganda. A total of 1,992 cattle were screened in villages, with (949) and without G. pallidipes (1043) for trypanosomosis using a combination of the BCT and HCT methods. The prevalence of trypanosomosis (15.5%), Trypanosoma brucei infection (1.4%), T. congolense infection (7.2%), T. vivax infection (5.3%) and mixed infection (1.6%) in cattle in villages with was significantly higher than in those without G. pallidipes: trypanosomosis (7.1%), T. brucei infection (0.6%), T. congolense infection (2.0%), T. vivax infection (3.3%) and mixed infection (1.2%) (overall trypanosome infection, chi2=35.5, d.f.=1, P<0.05; T. brucei infection, chi2=8.06, d.f.=1, P<0.05; T. congolense infection, chi2=22.8, d.f.=1, P<0.05 and T. vivax infection, chi2=6.4, d.f.=1, P<0.05). Infections of Trypanosoma congolense were predominant in cattle in villages with G. pallidipes, while T. vivax infections were predominant in cattle in villages without. In all villages, T. brucei infections were fewer than either T. congolense or T. vivax infections. The risk of transmission of T. brucei, T. congolense and T. vivax infections was 3, 2.7 and 1.6 times, respectively, higher in villages with G. pallidipes than in those without, despite the presence of G. f. fuscipes in either set of villages. The mean PCV (28.27+/-0.41, 95% CI) and mean herd size (3+/-0.46) of cattle in villages with G. pallidipes were significantly (P<0.05) lower than in those in villages without (mean PCV, 29.48+/-0.34; mean herd size, 4+/-0.72). It is evident that presence of G. pallidipes brings about an increase in the prevalence of T. congolense, which causes a more severe disease in cattle than other species of trypanosomes. This is a rare case of a re-invasion of a tsetse species whose disease transmission capability calls for refocusing of the traditional national tsetse and trypanosomosis control strategies to contain it. 相似文献

Study on comparative sensitivity of parasitological, serological, and molecular tests on 237 horses originating from two dourine-suspected
districts of Arsi-Bale highlands of Ethiopia was conducted to determine the prevalence of the disease and degree of agreement
of the diagnostic tests. Accordingly, the prevalence of the disease was found to be 4.6%, 36.7%, and 47.6% by parasitological
Woo test, RoTat 1.2 and 18S PCR tests, respectively. The seroprevalence of the disease was 27.6% in CATT/Trypanosoma evansi test. In Ethiopia, it was for the first time that trypanosomes from dourine suspected horses were demonstrated in 4.6% of
the animals using Woo test. The findings of the present study disclosed that dourine is highly prevalent and one of the major
diseases of horses in the area. There was no statistically significant difference (P > 0.05) in prevalence of the disease between districts, sexes, and age groups of the animals. However, there was a statistically
significant difference (P < 0.05) in the prevalence of the disease between emaciated and animals with good body condition. Assessment of the degree

Aiming to improve the diagnosis of canine leishmaniasis (CanL) in an endemic area of the Northwest region of S?o Paulo State, Brazil, the efficacy of parasitological, immunological and molecular diagnostic methods were studied. Dogs with and without clinical signs of the disease and positive for Leishmania, by direct parasite identification on lymph node smears and/or specific antibody detection by ELISA, were selected for the study. According to the clinical signs, 89 dogs attending the Veterinary Hospital of UNESP in Ara?atuba (SP, Brazil) were divided into three groups: symptomatic (36%), oligosymptomatic (22%) and asymptomatic (22%). Twenty-six dogs from an area non-endemic for CanL were used as negative controls (20%). Fine-needle aspiration biopsies (FNA) of popliteal lymph nodes were collected and Diff-Quick-stained for optical microscopy. Direct immunofluorescence, immunocytochemistry and parasite DNA amplification by PCR were also performed. After euthanasia, fragments of popliteal lymph nodes, spleen, bone marrow and liver were collected and processed for HE and immunohistochemistry. Parasite detection by both HE and immunohistochemistry was specifically more effective in lymph nodes, when compared with the other organs. Immunolabeling provided higher sensitivity for parasite detection in the tissues. In the symptomatic group, assay sensitivity was 75.61% for direct parasite search on Diff-Quick-stained FNAs, 92.68% for direct immunofluorescence, 92.68% for immunocytochemistry and 100% for PCR; the corresponding values in the other clinical groups were: 32, 60, 76 and 96% (oligosymptomatic), and 39.13, 73.91, 100 and 95.65% (asymptomatic). Results of the control animals from the CanL non-endemic area were all negative, indicating that the methods used were 100% specific. 相似文献

Baseline surveys were conducted in the Upper West Region of Ghana to assess the distribution and densities of tsetse species, as well as the prevalence of bovine trypanosomosis. The entomological survey was designed to cover the suitable tsetse habitats along the three main rivers in the study area (i.e. Black Volta, Kulpawn and Sissili). Results indicated the presence of Glossina tachinoides in all three river basins, whilst Glossina palpalis gambiensis was only found close to the southern limit of the study area. A random sampling of 1800 cattle of the West African Short Horn, Sanga and Zebu breeds from 36 randomly selected grid cells covering the study area showed substantial differences between parasitological and serological prevalences. The average parasitological prevalence was estimated at 2.5% (95% CI: 1.06-5.77) with the majority of the infections due to Trypanosoma vivax. Most of the infected cattle were found close to the major river systems. The serological prevalence, measured using Enzyme Linked Immunosorbent Assay (ELISA), test was 19% (95% CI: 14.03-25.35). A representative number of 217 camels (Camelus dromedarius) from different areas of western Rajasthan State, India, were examined from July 2002 to May 2003 for Trypanosoma evansi infection. The tests used were parasitological (wet blood film, WBF; stained thin blood smear, TBS), immunodiagnostic (double antibody sandwich enzyme linked immunosorbent assay for antigen detection, Ag-ELISA), and DNA amplification by polymerase chain reaction (PCR). These techniques were compared and the best efficiency was found for the last named (PCR). A prevalence of T. evansi infection was detected in 17.05, 9.67, 4.60 and 4.14% by PCR, Ag-ELISA, TBS and WBF with a sensitivity of 100, 56.75, 27.02 and 24.32%, respectively. PCR revealed a specific 227bp band in positive samples. The intensity of PCR bands was variable in different test samples depending upon the level of infection in the test samples. The history of intermittent fever, emaciation, oedema, poor body condition significantly correlated with positive serological status in ELISA as well as trypanosome DNA detection by PCR. 相似文献

A survey to update the distribution and clarify the epidemiology of bovine trypanosomosis in Malawi was conducted between 1995-97. Use was made of parasitological and serological (anti-trypanosomal antibody-detection Enzyme-Linked Immunosorbent Assay (ELISA) diagnostic methods. Trypanosomal infections were detected in cattle sampled adjacent to known tsetse foci. The distribution of cattle with anti-trypanosomal antibodies indicated that the distribution of bovine trypanosomosis was more widespread than the parasitological prevalence data suggested. This is attributed to the seasonal movement of tsetse (mainly Glossina morsitans morsitans and G. pallidipes) from its prime habitat and the presence of localized foci of G. brevipalpis. The odour-baited, insecticide-treated, target barriers along the edge of Kasungu National Park and the Nkhotakota Game Reserve appeared to be effective in preventing tsetse from moving outside the game areas and contacting cattle. The anti-trypanosomal antibody-detection ELISA proved to be a useful tool in establishing the distribution of bovine trypanosomosis. Moreover, the distribution and prevalence of cattle with anti-trypanosomal antibodies was instrumental in clarifying the epidemiology of bovine trypanosomosis in Malawi. The anti-trypanosomal antibody-detection ELISA had high sensitivity in detecting Trypanosoma congolense infections. In parasitologically negative animals, the average packed cell volume was higher in those that had no anti-trypanosomal antibodies. In this study five parasitological methods and a polymerase chain reaction (PCR) were compared for the diagnostic sensitivity for Trypanosoma evansi in experimentally infected water buffaloes over a period of 15 weeks. The combined estimates of sensitivity (CE(se)) of the PCR proved to be highest at 78.2%, closely followed by the mouse inoculation (MI), the micro-haematocrite centrifugation technique (MHCT) and the mini-anion-exchange centrifugation technique (MAECT) with CE(se) of, respectively, 74.0, 69.6 and 62.4%. The CE(se) of the buffy-coat technique (BCT) at 38.6% and the sodium dodecyl sulfate (SDS) clarification technique at 25.1% were considerably lower. PCR detected consistently all buffaloes infected from week 3 post-infection (PI) onwards. For MI this occurred after 5 weeks PI while for MHCT and MAECT these sustainable high levels were reached in the 7th week PI. BCT and SDS never detected all buffaloes infected. The influence of time and temperature on the viability of T. evansi in heparinized blood from water buffalo was also studied. In general we observed that the survival time tends to be longer when blood is kept at 4 degrees C. In samples kept in direct sunlight parasites became undetectable with the MHCT after 30min. After treatment of the water buffaloes with diminazene aceturate, the PCR signal disappeared within 24h. 相似文献

An epidemiological investigation was conducted on farms in Tororo and Soroti districts of Uganda from January to February 2000 to determine the cause of reported persistent mortality of cattle. Blood and faecal material of 98 cattle comprising of 33 Friesians, 58 Zebu and 7 Sahiwal were examined. Results revealed that seven (7.1 %) cattle had trypanosome infection, mainly due to Trypanosoma vivax and T. brucei, 17 (17.3%) Fasciola infection, 28 (28.6%) gastrointestinal nematode infection, 33 (33.7%) Theileria sp. infection and 13 (13.3%) Anaplasma marginale infection. Mixed infections were detected in 30%, 20.6% and 43 % of the Friesian, Zebu and Sahiwal cattle respectively. Anaemia (PCV < 25) was detected in 24%, 19% and 14% of the Friesian, Zebu and Sahiwal cattle respectively. Persistent mortality of cattle on these farms could have been due to either single or mixed parasitic infections probably exacerbated by malnutrition. 相似文献

This study assessed the prevalence of trypanosomes in cattle at the Kachia Grazing Reserve (KGR) in March and June 2004 and in February 2005. A total of 1293 cattle blood samples were collected at random. The samples were analysed using the buffy coat technique and Giemsa thin blood films for parasite detection and identification. The effects of herd pen location to watering and grazing point's distances (using the global positioning system (GPS)) were determined and mean Packed cell volume (PCV) assessed. Overall, the detected prevalence of trypanosomosis was 8.4%, much higher than the previous prevalence of 5.3% before the present study was conducted. The prevalences in the months of March, June (2004) and February (2005) were 2.3%, 11.6% and 15.4%, respectively. Increased prevalence was associated with proximity of herd pens to watering point's distances (χ2 for linear trend = 4.447, P < 0.05), but no association of herd pens to grazing point distances (χ2 = 2.186, P > 0.05); suggesting that hydrological network played an important part in trypanosomosis transmission. The mean PCV of parasitaemic and apparasitaemic cattle were respectively 25.99 ± 1.82% and 29.31 ± 1.70%. The drop in mean PCV was most in 0–1-year age group, 23.47 ± 3.10% and was statistically significant (P < 0.05), suggesting that anaemia was most pronounced in this age group. Factors that may have contributed to the increased prevalence obtained were collapse of control measures and breed susceptibility. Since, Zebu cattle were the predominant breeds in the reserve, the study advocates effective use of insecticide impregnated screens (traps and targets) with community participation in mind for sustainability. If government intervenes through PATTEC ground spraying of insecticides in the reserve is recommended. In addition, chemotherapeutic and chemoprophylaxis should be systematically used to fight the problem of trypanosomosis in the KGR towards improved livestock production. 相似文献

Livestock farmers' perception and epidemiology of bovine trypanosomosis in Kwale District, Kenya 总被引:1,自引:0,他引:1
We did cross-sectional surveys in Kwale District, Kenya to determine the epidemiology of bovine trypanosomosis and livestock owners' perceptions of the disease. The surveys involved relative importance of trypanosomosis, examination of the current disease constraints, current control practices and drug-use patterns. Informal meetings were held with farmers and cattle census undertaken. Tsetse-fly densities and trypanosomosis prevalences in cattle were determined. A total of 132 farmers were interviewed. Trypanosomosis, anaplasmosis, East Coast fever, foot-and-mouth diseases were reported to be the major constraints to livestock production. Trypanosomosis was the most important compared to other diseases. Chemotherapy was the most widely used method of controlling the disease. Farmer-based tsetse-control technologies were poorly adopted. Respondents were quite knowledgeable on the symptoms, causes and treatment of trypanosomosis. Glossina austeni, G. brevipalpis and G. pallidipes were found in the area; the latter was the most common (0.2-738 flies/trap). Trypanosoma congolense and T. vivax were found in cattle with the former more prevalent. Infection prevalences in cattle varied between 0 and 25% (median: 22%). 相似文献

A longitudinal study was conducted over a 1-year period in six selected villages in Petauke and Katete districts in the Eastern Province of Zambia. Starting in November 1997, 50 animals were sampled at random at each village every 2 months. The parasitological prevalence of trypanosomosis was determined by the haematocrit centrifugation buffy-coat technique, supplemented with thick and thin Giemsa-stained blood films. Serum samples also were collected for anti-trypanosomal antibody determination by indirect enzyme-linked immunosorbent assay.
Parasitological prevalence was highly variable between villages and between visits (range: 0–28.6%; median: 3.1%). Seroprevalence was also variable between villages (range: 0–80.8%; median: 50%), but was less variable between visits. Average annual parasitological prevalence and average annual seroprevalence for each village were highly correlated [R2 (adjusted for d.f.)=0.89, p<0.01]. Seroprevalence measured on any single visit to a study village was better than parasitological prevalence as a predictor of average annual parasitological prevalence. 相似文献

A pilot survey using a structured questionnaire was conducted in Tororo and Busia districts of Uganda on the knowledge and attitudes of cattle owners regarding tsetse fly and trypanosomosis control, in order to understand factors that hindered their full participation. A total of 81 cattle owners was randomly selected and interviewed, of which 92.5% were aware of tsetse flies and trypanosomosis and 87.6% recognised animal trypanosomosis as a problem in the area. Most cattle owners were aware of tsetse fly trapping (76.5%), isometamidium chloride use (55.5%), diminazene aceturate use (48%) and pour-on applications (18.5%). However, knowledge did not coincide with the application of control measures. Despite the widespread awareness, tsetse fly trapping and pour-on applications were used by only a small percentage of cattle owners (7.5% applied tsetse fly trapping while 76.5% were aware of it; 1.2% applied pour-on insecticides while 18.5% were aware of them). Differences between awareness and application were highly significant for tsetse fly trapping (chi2 = 67.8, d.f. = 1, P < 0.001) and pour-on applications (chi2 = 10.8, d.f. = 1, P < 0.05), but not for isometamidium chloride use (chi2 = 0.08, d.f. = 1, P = 0.77) and diminazene aceturate use (chi2 = 0.00, d.f. = 1, P = 1.00). Most cattle owners (97.5%) were willing to participate in future control programmes, but preferred participating on a group basis (85.2%) rather than individually (14.8%). The 4 most favoured control options in order of importance were: fly traps supplied by the government and maintained by cattle owners; contribution of labour by cattle owners for trap deployment; self-financing of trypanocidal drugs and self-financing of pour-on insecticide. The control options that should be selected in order to elicit full participation by cattle owners are discussed. 相似文献

We report hematological changes observed in natural cases of bovine trypanosomosis due to Trypanosoma vivax in beef and dairy cattle from Bolivian wetlands and Pantanal, Brazil. The main hematologic changes produced by T. vivax infections were anemia and severe leucopenia. The cattle presented macrocytic hypochromic anemia. The leukocyte changes were characterized by relative lymphocytosis and monocytosis and decrease in the neutrophil counts. The clinical signs were lachrymation, progressive weakness, marked weight loss, inappetence, diarrhea and abortions during the third trimester of pregnancy. 相似文献

The study was carried out to assess the economic burden of trypanosomosis in three villages of the Metekel zone in 2009. The
disease was found to cause substantial economic losses through cattle mortality, drug purchase, and draft power loss of infected
oxen. The farmers in the area were spending a significantly (p < 0.05) higher amount of money for the treatment of trypanosomosis than all other diseases combined. The overall mortality
rate of cattle due to trypanosomosis was 4.4%. The mortality was significantly higher (p < 0.05) in an area where trypanosomosis prevalence was also higher. Many of the farmers prioritized losses of draft power
as the most important impact of the disease. The overall prevalence of the disease was 12.1%. The disease burden was significantly
(p < 0.05) higher in the rainy season than at other times of the year. In general, farmers had good knowledge on the signs and
seasonality of trypanosomosis. Thus, tsetse suppression activities that involve the local community can be an important tool

A cross sectional survey to determine the distribution and prevalence of trypanosomosis was conducted in Kindo Koisha district, in the Wollaita zone in southern Ethiopia. A total of 1 008 adult cattle was examined at eight different localities. Dark field examination of the buffy coat, as well as stained thin blood film examination and packed cell volume (PCV) evaluation were the diagnostic techniques used. The overall prevalence of bovine trypanosomosis was 15 %. Among the positive animals, 108 (71.1%), 43 (28.4%) and 1 (0.6%) were due to Trypanosoma vivax, Trypanosoma congolense and mixed infection (T. vivax and T. congolense), respectively. The infection rate of T. vivax and T. congolense varied significantly (P < 0.01). The mean PCV of the positive and negative animals ranged between 18.3-32.1% and 26.8-33.4%, respectively. The mean PCV of negative animals (28 %) was significantly higher than the mean PCV of positive animals (22.3%) (P < 0.001). There was an inverse association of PCV with the prevalence of trypanosomosis (P > 0.05). The herd average PCV values of each site decreased with increasing proportion of the positive herds of that particular site. Of the diagnostic tests employed, the microhaematocrit buffy coat technique is relatively sensitive and it has an added advantage of indicating the general condition of the animal by haematocrit measurement. In view of the risk of trypanosomosis, a control intervention through the strategic application of appropriate trypanocidal drugs is recommended. A tsetse fly control scheme to reduce host-tsetse fly contact is equally as important as chemotherapy and chemoprophylaxis against trypanosomosis. 相似文献

Bacterial isolates (including 17 Haemophilus somnus isolates and an H. somnus-like isolate) from asymptomatic or diseased cattle and sheep, were evaluated for markers associated with virulence and host predilection. The isolates were separated into 6 distinct biovariants, 3 for sheep and 3 for cattle, based on reactions in a battery of 21 test media. Three bovine isolates associated with disease caused hemolysis of bovine blood. The rest of the isolates did not hemolyze either bovine or ovine erythrocytes. Protein profiles of all H. somnus isolates were similar with the exception of the major outer membrane proteins (MOMPs). The MOMPs of isolates associated with disease in cattle had a relative molecular weight of approximately 41 kDa compared with 33 kDa for the MOMPs of isolates from asymptomatic cattle. The MOMPs from sheep isolates were either slightly higher or lower than the 41 kDa MOMPs of bovine isolates. Major antigens detected by Western blotting were similar in all isolates except the H. somnus-like isolate. An immunodominant 40 kDa antigen was conserved in all H. somnus isolates. Antibodies to this antigen have previously been found to be protective in cattle and may also be protective for sheep. Marked differences between cattle and sheep isolates were revealed by use of restriction enzyme analysis, which separated the isolates into 12 ribotypes and 15 unique DNA profiles. Thus, cattle and sheep isolates in this collection had distinctive differences in biochemical reactions, MOMP profiles, and DNA analyses. Such differences have potential value for epidemiological studies and may also be used to evaluate host specificity of H. somnus isolates. 相似文献
