Ageratum conyzoides L. is an annual herb in the tropics and subtropics whose extracts are known to possess pharmacological and biocidal activity. We report on the bioactivity of a secondary metabolite (a chromene) isolated from the shoots ofA. conyzoides against some plant pathogenic fungi. Organic solvent extracts from the shoots were tested for antifungal activity against the plant pathogenic fungiRhizoctonia solani, Sclerotium rolfsii, Botryodiplodia theobromae, Phomopsis theae andFusarium species growingin vitro on potato dextrose agar medium. The cruden-hexane extract completely inhibited the growth ofR. solani andS. rolfsii. Then-hexane extract was chromatographed over a column of silica gel followed by activity-guided fractionation to give an antifungal principle. Structure elucidation by detailed analysis of1H,13C NMR and mass spectroscopy identified the active compound as precocene II. The growth ofR. solani andS. rolfsii was completely inhibited by precocene II at a concentration of 80–100 ppm. The sclerotia ofR. solani andS. rolfsii were also completely suppressed by 150 ppm of precocene II. Sub-culture of these inhibited fungi onto precocene II-free medium restored growth of the fungus, indicating that precocene II is fungistatic. A collection ofTrichoderma isolates, with different biocontrol capabilities, were identified by molecular methods. Arbitrarily-primed PCR (ap-PCR) using repeat motif primers was performed on DNA from aTrichoderma spp. collection of 76 isolates, and representative isolates were further characterized into three main clades by internal transcribed spacer (ITS) sequence analysis. Abstrast  Three-hundred-twenty-five isolates ofRhizoctonia (215R. solani and 110 binucleateRhizoctonia) were obtained from roots and crowns of alfalfa, sainfoin and common vetch grown in Erzurum, Turkey. The isolates were assigned to five anastomosis groups (AG) ofR. solani (AG-2-1, AG-3, AG-4, AG-5, and AG-10) and two anastomosis groups of binucleateRhizoctonia (AG-I and AG-K). In pathogenicity tests on alfalfa, sainfoin and common vetch, the highest disease severities were caused by isolates of AG-4 and AG-5. Isolates of AG-10 and AG-I were not pathogenic on the three tested forage legumes, whereas isolates of AG-K on alfalfa and sainfoin, and of AG-2-1 on sainfoin, were moderately virulent. Alfalfa isolate AG-3 was moderately virulent on sainfoin. This is the first report ofR. solani AG-3, AG-5, AG-10 and binucleateRhizoctonia AG-I on alfalfa. Sixteen isolates belonging to 11 species of Trichoderma (T. asperellum, T. ceramicum, T. andinensis, T. orientalis, T. atroviride, T. viridescens, T. brevicompactum, T. harzianum, T. virens, T. koningii and T. koningiopsis) were evaluated for biological control of potato (Solanum tuberosum) stem rot caused by Sclerotinia sclerotiorum. In dual culture tests, all antagonists significantly reduced sclerotia formation, and were able to inhibit radial growth of the pathogen. Growth inhibition by production of volatile and non-volatile inhibitors was also measured in in vitro tests. In screening the most efficient species of Trichoderma, establishment of mycelium on sclerotia and sclerotia lysis were also considered as important biocontrol qualities. Excluding T. asperellum, T. brevicompactum, T. andinensis and T. harzianum, all tested Trichoderma species were able to lyse sclerotia. The sclerotia-destroying species of Trichoderma and one isolate of Talaromyces flavus were tested in greenhouse tests and during 2 years of field experimentation during the 2007 and 2008 cropping seasons. After one aerial application of spore suspension in greenhouse trials, T. koningii, T. virens, T. ceramicum and T. viridescens were the most effective bio-agents and reduced significantly disease severity, and the least biocontrol efficacy was observed in T. flavus. Under field conditions and after five soil and foliar applications of spore suspension, all tested antagonists reduced significantly disease incidence. T. viridescens followed by T. ceramicum showed the best results. T. flavus and T. orientalis were less effective than other tested antagonists in both field trials.  相似文献

Four-hundred-sixty-eightFusarium andFusarium-like isolates were obtained from crowns and subcrown internodes of winter wheat grown in Erzurum, Turkey. Of these isolates, 34.8% wereFusarium acuminatum, 32.3% wereF. equiseti, 16.9% wereF. oxysporum, 15.0% wereMicrodochium nivale (formerlyFusarium nivale), 0.6% wereF. tabacinum and 0.4% wereF. solani. In pathogenicity tests on wheat, the highest disease severity was caused by isolates ofM. nivale, whereas isolates ofF. acuminatum, F. equiseti, F. oxysporum andF. solani were slightly virulent; isolates ofF. tabacinum were nonpathogenic. Crude ethanolic seed extracts ofAnnona muricata, A. squamosa (Annonaceae),Lansium domesticum andSandoricum koetjape (Meliaceae) collected from different locations and years in Maluku, Indonesia, were screened for inhibition of larval growth against the polyphagous lepidopteranSpodoptera litura (Noctuidae). Extracts ofA. squamosa were significantly more active (20-fold) than those ofA. muricata. A. squamosa collected from Namlea yielded the extracts with the greatest inhibitory activity. There were significant differences among locations for bothA. squamosa andA. muricata but not forL. domesticum andS. koetjape. Extracts ofA. squamosa, collected from Namlea, inhibited larval growth in a dose-dependent manner, with a dietary EC50 (effective concentration to inhibit growth by 50% relative to controls) of 191.7 ppm fresh weight. Extracts ofA. squamosa collected from individual trees in Namlea also varied in growth inhibitory effect againstS. litura andTrichoplusia ni larvae. The study deals with the tachinid parasitoids ofAncyrosoma leucogrammes (Gmelin) (Heteroptera: Pentatomidae):Clytiomya dupuisi Kugler,Clytiomya sola (Rondani) andGymnosoma clavatum (Rohdendorf) (Diptera: Tachinidae). All species are recorded for the first time as parasitoids ofA. leucogrammes, andC. dupuisi was reared for the first time from a host. Fusarium head blight of wheat, often associated with mycotoxin contamination of food and feed is caused by various Fusarium species. The efficacy of fungicide sprays for the control of the disease and mycotoxins varies from being highly effective to even increasing mycotoxin levels. The potential role of antagonistic fungi in this variability was investigated assessing sensitivity of Fusarium species and saprophytic fungi colonizing wheat kernels to fungicides. Saprophytes were tested for their antagonistic activity to the prevalent Fusarium species Fusarium avenaceum, Fusarium culmorum, Fusarium graminearum, and Fusarium poae. Fungal isolates from mature winter wheat kernels were Alternaria alternata, Arthrinium sp., Aspergillus niger, Epicoccum sp., Microdochium spp., Rhizopus oryzae and Trichoderma sp. In dual culture A. niger, R. oryzae and Trichoderma hamatum were more effective in reducing mycelial growth of Fusarium species than Microdochium majus; A. alternata and Epicoccum sp. were ineffective because of slow growth rates. Saprophytic fungi were sensitive to triazoles; however, prothioconazole and tebuconazole had stronger effects on mycelial growth of Fusarium species. ED50 values also indicated significant differences in the sensitivity of Fusarium species to triazoles (range 0.1–1.7 mg l−1). Azoxystrobin and fluoxastrobin were largely ineffective in inhibiting in vitro growth of Fusarium spp.; sensitivity of the other fungi was generally lower, except for M. majus which was highly sensitive. Due to differences in fungicide sensitivity among Fusarium spp. and ear-colonizing fungi antagonistic to Fusarium spp. fungicides are likely to modify the balance within the mycoflora of wheat ears which may also affect the mycotoxin contamination of grain.  相似文献

Biological control of Rhizoctonia solani with Trichoderma harzianum has been demonstrated in several studies. However, none have reported the dynamics of expression of defence response genes. Here we investigated the expression of these genes in potato roots challenged by R. solani in the presence/absence of T. harzianum Rifai MUCL 29707. Analysis of gene expression revealed an induction of PR1 at 168 h post-inoculation (hpi) and PAL at 96 hpi in the plants inoculated with T. harzianum Rifai MUCL 29707, an induction of PR1, PR2 and PAL at 48 hpi in the plants inoculated with R. solani and an induction of Lox at 24 hpi and PR1, PR2, PAL and GST1 at 72 hpi in the plants inoculated with both organisms. These results suggest that in the presence of T. harzianum Rifai MUCL 29707, the expression of Lox and GST1 genes are primed in potato plantlets infected with R. solani at an early stage of infection. Adult Mediterranean fruit flies (Ceratitis capitata), collected in the field, were infected with entomophthoralean fungi. The fungi sporulated poorly on the cadavers, and resting spores, rhizoids and cystidia were not observed. Measurements of conidia and nuclei and counts of nuclei per conidium from different specimens suggest that the causative agents wereEntomophthora muscae sensu stricto andEntomophthora schizophorae, species recently separated from theEntomophthora muscae species complex. Fourteen species in two families of the order Hymenoptera: Ichneumonidae (1 sp.) and Eulophidae (13 spp.), were recorded during the study ofCameraria ohridella parasitoids at eight localities in Serbia. Among the observed parasitoids, the most significant species wereMinotetrastichus frontalis, Pediobius saulius, Pnigalio agraules andClosterocerus trifasciatus. In addition, at some localities the speciesMinotetrastichus platanellus andCirrospilus talitzkii were also significant. Trichoderma harzianum is an effective biocontrol agent against the devastating plant pathogen Rhizoctonia solani. Despite its wide application in agriculture, the mechanisms of biocontrol are not yet fully understood. Mycoparasitism and antibiosis are suggested, but may not be sole cause of disease reduction. In the present study, we investigated the role of oxidant-antioxidant metabolites in the root apoplast of sunflower challenged by R. solani in the presence/absence of T. harzianum NBRI-1055. Analysis of oxidative stress response revealed a reduction in hydroxyl radical concentration (OH; 3.6 times) at 9 days after pathogen inoculation (dapi), superoxide anion radical concentration (O2•−; 4.1 times) at 8 dapi and hydrogen peroxide concentration (H2O2; 2.7 times) levels at 7 dapi in plants treated with spent maize-cob formulation of T. harzianum NBRI-1055 (MCFT), as compared to pathogen-inoculated plants. The protection afforded by the biocontrol agent was associated with the accumulation of the ROS gene network: the catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx) and ascorbate peroxidase (APx), maximum activity of CAT (11.0 times) was observed at 8 dapi, SOD (7.0 times) at 7 dapi, GPx (5.4 times) and APx (8.1 times) at 7 dapi in MCFT-treated plants challenged with the pathogen. This was further supported by the inhibition of lipid and protein oxidation in Trichoderma-inoculated plants. MCFT stimulated the accumulation of secondary metabolites of phenolic nature that increased up to five-fold and also exhibited strong antioxidant activity at 8 dapi, eventually leading to the systemic accumulation of phytoalexins. These results suggest that T. harzianum–mediated biocontrol may be related to alleviating R. solani-induced oxidative stress.  相似文献

The relationships between biotic changes and local decrease in soil conduciveness in disease patches towards the disease incited by Rhizoctonia solani AG 2-2 in a sugar beet field in France were investigated. Soil samples from healthy and diseased areas were analysed for bacterial and fungal densities, molecular and physiological microbial community structures, and antagonistic abilities of Trichoderma isolates collected from diseased and healthy areas. Although the inoculum density was higher inside the disease patches, the respective soil was less conducive towards disease incited by R. solani AG 2-2. It was concluded that the pathogen was present in healthy areas but did not incite disease in field conditions. Conversely, the response of the microflora to previous development of R. solani in diseased areas prevented further pathogenic activity. Indeed, genetic and physiological structures of the fungal communities and physiological structures of the bacterial communities were modified in disease patches compared to healthy areas. The terminal restriction fragment length polymorphism (T-RFLP) analysis revealed that the peaks corresponding to R. solani and Trichoderma spp. were higher inside the patches than in the healthy areas. Trichoderma isolates from the disease patches were more antagonistic than those from the healthy areas. These results suggest that disease caused by R. solani AG 2-2 induced changes in genetic and physiological structure of microbial populations and development of antagonists. The decreased conduciveness inside the patches may help explain patch mobility in the following season.  相似文献

Leaves of tomato and barley were inoculated with conidia of Blumeria graminis f. sp. hordei race 1 (R1) or Oidium neolycopersici (KTP-01) to observe cytological responses in search of resistance to powdery mildew. Both conidia formed appressoria at similar rates on tomato or barley leaves, indicating that no resistance was expressed during the prepenetration stage of these fungi. On R1-inoculated tomato leaves, appressoria penetrated the papillae, but subsequent haustorium formation was inhibited by hypersensitive necrosis in the invaded epidermal cells. On the other hand, KTP-01 (pathogenic to tomato leaves) successfully developed functional haustoria in epidermal cells to elongate secondary hyphae, although the hyphal elongation from some conidia was later suppressed by delayed hypersensitive necrosis in some haustorium-harboring epidermal cells. Thus, the present study indicated that the resistance of tomato to powdery mildew fungi was associated with a hypersensitive response in invaded epidermal cells but not the prevention of fungal penetration through host papilla.  相似文献

Tachinid parasitoids ofTraumatocampa ispartaensis Doğanlar & Avcı (Lepidoptera, Thaumetopoeidae), which was found to be a new species, were collected from the cedar forests around Isparta-Kapıdağ. The species found wereBlondelia nigripes (Fall.),Compsilura concinnata (Meig.),Pales processioneae (Ratz.),Phryxe caudata (Rond.),Exorista segregata (Rond.) andCarcelia iliaca (Ratz.). Populations of pear psylla,Cacopsylla pyri (L.) (Rhynchota: Psyllidae), and its natural enemies were sampled on pears in Antalya province during the years 2000–2002. Thirty-two species of predators and three species of parasitoids were detected to be associated withC. pyri in Antalya province. The heteropterans (especially anthocorid species) were the most abundant predator group. Among the anthocorids,Anthocoris nemoralis (Fabricius) (Heteroptera: Anthocoridae), whose population was closely related to the dynamics of the pear psylla population, was generally the principal antagonist ofC. pyri. Other species recorded during the study were the miridDeraeocoris spp., the chrysopidChrysoperla carnea L., and many coccinellid species. Although these predators are polyphagous, as are theOrius species, their populations were related to the psylla fluctuations. Among the parasitoid complex belonging to the order Hymenoptera, the encyrtidTrechnites psyllae (Ruschka) was the only primary parasitoid. Two hyperparasites,viz., Syrphophagus mamitus (Walker) andPachyneuron aphidis (Bouché) (Encyrtidae and Pteromalidae, respectively), were detected in the surveys. Also, this study revealed new information concerning the pear psylla parasitoid complex in Turkey. WhileT. psyllae andS. mamitus are new records to Turkish parasitoid fauna,C. pyri is a new host record forP. aphidis. Significant antibacterial activity was observed in the essential oil (E.O.) ofRosa damascena Mill. and the Minimum Bactericidal Concentration (MBC) of the E.O. was determined as 1386.5 μg ml−1 forErwinia amylovora, the causal agent of fire blight disease. Ooze formation on immature pears and lesion formation in artificially inoculated shoots were completely (100%) prevented by the essential oil ofR. damascena (1500 μg ml−1), essential oil ofThymbra spicata var.spicata (500 μg ml−1) and streptomycin (100 μg ml−1). Copper oxychloride plus maneb significantly reduced ooze formation and lesion formation, but the control was less than that obtained with the essential oils or streptomycin. Studies were carried out on the egg material ofTraumatocampa ispartaensis Doganlar & Avcı (Lep.: Notodontidae) collected onCedrus libani A. Rich. A total of 95 egg-batches were sampled over two annual generations ofTr. ispartaensis. The number of eggs in each egg-batch varied between 39 and 245 and the length of the batches varied between 7 and 36 mm. The mean number of eggs per batch was found to be 119 and 122 in 1999 and 2000, respectively. Based on field data the oviposition period continued from mid August to mid September. The mean hatching rate of the host was found to be 85.8% and 88.9%, whereas the impact of egg parasitoids accounted for 11.3% and 7.4%, respectively in the 2 years.Ooencyrtus pityocampae (Mercet) was observed as the most abundant egg parasitoid, followed byOoencyrtus sp. nearmasii (Mercet) andTrichogramma brassicae Bezdenko. Botryodiplodia theobromae, Colletotrichum gloeosporioides andGliocephalotrichum microchlamydosporum are the causal fungi of three rambutan postharvest diseases, stemend rot, anthracnose and brown spot, respectively. Two different treatments of rambutan fruits to control the diseases were investigated: application of potassium metabisulphite (250 ppm) or culture filtrate ofTrichoderma harzianum (TrH 40) followed by controlled atmosphere storage (CA) at 13.5°C and 95% r.h. Potassium metabisulphite at 250 ppm under CA effectively controlled the incidence and severity of the three postharvest diseases and maintained the eating quality and color of the fruit for 21 days. The greatest effect of this treatment was on brown spot disease, caused byG. microchlamydosporum. Artificial suppression of radish damping-off disease was induced by repeated soil inoculations with Rhizoctonia solani, binucleate Rhizoctonia (BNR) and Sclerotium rolfsii in pot systems. Soils repeatedly inoculated with R. solani and BNR showed suppressive to disease caused by R. solani and S. rolfsii, while soils repeatedly inoculated with S. rolfsii were suppressive to disease caused by S. rolfsii but not by R. solani. Species of Trichoderma were consistently isolated from soils repeatedly inoculated with R. solani, BNR and S. rolfsii. These Trichoderma spp. accumulated selectively in relation to the fungal species that was repeatedly added to the soils. The ratios of the frequencies of T. viride, T. harzianum and T. hamatum were 5:2:2 and 8:5:2 in soils repeatedly inoculated with R. solani and BNR, respectively. In S. rolfsii- inoculated soils, T. koningii was predominantly isolated. T. viride, T. harzianum and T. hamatum isolates obtained from either R. solani or BNR after repeated additions to the soils suppressed radish damping-off disease caused after challenge inoculations with R. solani or S. rolfsii. Among the Trichoderma species, T. viride consistently yielded high levels of suppression. However, isolates of T. koningii obtained from S. rolfsii-infested soils suppressed disease caused by S. rolfsii but failed to suppress disease caused by R. solani. Generally, the species of Trichoderma accumulated in a selective pattern that was closely related to the species of fungal pathogen used to induce the suppressive soil.  相似文献
