The objective of this study was to evaluate the effect of reduced-calorie avocado paste on lipid serum profile, insulin sensitivity, and hepatic steatosis in rats fed a hypercholesterolemic-high fructose diet. Thirty five male Wistar rats were randomly separated in five groups: Control group (ground commercial diet); hypercholesterolemic diet plus 60 % fructose solution (HHF group); hypercholesterolemic diet plus 60 % fructose solution supplemented with avocado pulp (HHF+A group); hypercholesterolemic diet plus 60 % fructose solution supplemented with reduced-calorie avocado paste (HHF+P group); and hypercholesterolemic diet plus 60 % fructose solution supplemented with a reduced-calorie avocado paste plus fiber (HHF+FP group). The A, P, and FP were supplemented at 2 g/kg/d. The study was carried out for seven weeks. Rats belonging to the HHF group exhibited significantly (P?≤?0.05) higher total cholesterol, triglycerides, and insulin levels in serum as well as lower insulin sensitivity than the control group. Supplementation with reduced-calorie avocado paste showed a significant (P?≤?0.05) decrease in total cholesterol (43.1 %), low-density lipoprotein (45.4 %), and triglycerides (32.8 %) in plasma as well as elevated insulin sensitivity compared to the HHF group. Additionally, the liver enzymes alanine aminotransferase and aspartate aminotransferase decreased significantly in the HHF-P group (39.8 and 35.1 %, respectively). These results are likely due to biocompounds present in the reduced-calorie avocado paste, such as polyphenols, carotenoids, chlorophylls, and dietary fibre, which are capable of reducing oxidative stress. Therefore, reduced-calorie avocado paste attenuates the effects of a hypercholesterolemic-high fructose diet in rats.  相似文献

The methanolic extract of Cassia hirsuta L. seed materials, an underutilized food legume collected from India, was analyzed for antioxidant activity and health relevant functionality. The methanolic extract of raw seeds contained a total free phenolic content of 15.82?±?1.69 g catechin equivalent/100 g extract DM. Encouraging levels of ferric reducing/antioxidant power (FRAP, 1,446 mmol Fe[II]/mg extract), inhibition of ß-carotene degradation (48.81%) and scavenging activity against DPPH (64.40%) and superoxide (43.78%) radicals were exhibited by the raw samples. Further, 83.11% of α-amylase and 62.79% of α-glucosidase enzyme inhibition characteristics under in vitro starch digestion bioassay were also recorded. Sprouting + oil-frying caused an apparent increase on the total free phenolic content and a significant improvement in the antioxidant and free radical scavenging capacity of methanolic extract of C. hirsuta seeds, while soaking + cooking as well as open-pan roasting treatments showed diminishing effects. The analysis of the phenolic profile revealed the presence of gallic acid, p-coumaric acid and (+)-catechin in the methanolic extract of these seeds.  相似文献

Avocado seed contains elevated levels of phenolic compounds and exhibits antioxidant properties. We investigated the effect of Avocado Seed Flour (ASF) on the lipid levels in mice on a hyperlipidemic diet. The concentration of phenols was determined by high-performance liquid chromatography, antioxidant activity was evaluated using the Trolox equivalent antioxidant capacity method, and dietary fiber was measured using the Association of Official Analytical Chemists (AOAC) method. The LD50 of ASF was determined using Lorke’s method and hypolipidemic activity was evaluated in a hypercholesterolemic model in mice. Protocatechuic acid was the main phenolic compound found in ASF, followed by kaempferide and vanillic acid. The total phenolic content in the methanolic extract of ASF was 292.00 ± 9.81 mg gallic acid equivalents/g seed dry weight and the antioxidant activity resulted in 173.3 μmol Trolox equivalents/g DW. In addition, a high content of dietary fiber was found (34.8%). The oral LD50 for ASF was 1767 mg/kg body weight, and treatment with ASF significantly reduced the levels of total cholesterol, LDL-C, and prediction of the atherogenic index. Therefore, the antioxidant activity of phenolic compounds and dietary fiber in ASF may be responsible for the hypocholesterolemic activity of ASF in a hyperlipidemic model of mice.  相似文献

Cardiovascular diseases, especially atherosclerosis, have found to be the dreadful diseases worldwide and therapeutic interventions using plant sources have wide therapeutic value. Vigna unguiculata (VU) leaves have been used as food and therapeutics. Hence, our study was designed to evaluate the hypolipidemic as well as anti-atherogenic potential of VU leaves in normalizing atherogenic gene expression, cholesterol profile, generation of reactive oxygen species (ROS) and antioxidant enzyme system on cholesterol fed rabbit model. For the study New Zealand white rabbits were randomly divided into four groups of six animals each and experimental period was three months; group -i - ND [normal diet (40 g feed)], group-ii- ND (normal diet) +EAVU [ethyl acetate fraction of Vigna unguiculata (150 mg/kg body weight)], group -iii- ND [normal diet ]+ CFD [cholesterol fed diet (cholesterol 1 % of 40 g feed and cholic acid 0.5 % of 40 g feed)] and group-iv - ND [normal diet] +CFD [cholesterol fed diet ]+EAVU [ethyl acetate fraction of Vigna unguiculata (150 mg/kg body weight)]. Atherosclerosis was induced by feeding the rabbit with cholesterol (1 % of 40 g feed) and cholic acid (0.5 % of 40 g feed). Supplementation of EAVU normalized cholesterol profile, generation of reactive oxygen species (ROS), lipid peroxidation products like thiobarbituric acid reactive substance (TBARS), antioxidant system and important genes of cardiovascular diseases like interleukin-10 (IL 10), paraoxanase-1 (PON I), interleukin-6 (IL 6), and cyclooxygenase-2 (Cox 2) to near normal level as compared with normal diet. The result obtained showed the antioxidant as well as anti-atherogenic potential of Vigna unguiculata leaves in ameliorating cholesterol induced atherosclerosis, and thus it is good task to include VU leaves in daily diet for the prevention of cardiovascular diseases especially atherosclerosis.  相似文献

Camellia euphlebia (family, Theaceae) has been used for the prevention and treatment of cardiovascular diseases in Southern China. However, there has been no report on the hypolipidemic activity of Camellia euphlebia flower. This study evaluated the hypolipidemic activity of different preparation of Camellia euphlebia flower extracts using in vivo models. Mice intragastrically administered aqueous extract at 400 mg/kg dose or ethanol extract at 100 and 400 mg/kg doses of Camellia euphlebia flower for 28 days exhibited significant decreases in the levels of total cholesterol, triglycerides and low-density lipoprotein cholesterol, while displaying increased level of high-density lipoprotein cholesterol in the serum. The Camellia euphlebia flower extracts also improved the antioxidant ability of hyperlipidemic mice as well as protecting the animals against liver damage by lowering the level of glutamic-pyruvic transaminase activity. Furthermore, 400 mg/kg ethanol extract effectively down-regulated the mRNA levels of fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and glycerol-3-phosphate acyl transferase, suggesting that Camellia euphlebia flower extract may potentially inhibit lipid accumulation in the liver by regulating the expression of fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and glycerol-3-phosphate acyl transferase. These results provided support for the potential hypolipidemic activity of Camellia euphlebia flower and could partly explain the basis of using Camellia euphlebia for the treatment of hyperlipidemia.  相似文献

Hyperlipidemia and hepatic steatosis are frequent alterations due to alcohol abuse. Amaranth is a pseudocereal with hypolipidemic potential among other nutraceutical actions. Here we study the effect of Amaranthus hypochondriacus (Ah) seeds on serum and liver lipids, and the expression of genes associated to lipid metabolism and liver histology in male Wistar rats intoxicated with ethanol. The animals were divided into four groups; two groups were fed the American Institute of Nutrition 1993 for maintenance diet (AIN-93M), and the other two with AIN-93M containing Ah as protein source. One of each protein group received 20 % ethanol in the drinking water, thus obtaining: CC (control casein), EC (ethanol casein), CAh (control Ah) and EAh (ethanol Ah). When comparing EAh vs. EC, we found a positive effect of Ah on lipids, preventing the increment of serum cholesterol (p?<?0.001), through the higher expression of the LDL receptor (p?<?0.001); and it also decreased free (p?<?0.05) and esterified cholesterol (p?<?0.01) in liver, probably via the reduction of the 3-hydroxy-3-methylglutaryl coenzyme A reductase expression (p?<?0.001). We also observed that amaranth contributed to the decrease of fat deposits in liver, probably through the decrease in acetyl-CoA carboxylase alpha (p?<?0.01), glycerol-3-phosphate acyltransferase 1 (p?<?0.01) and diacylglycerol O-acyltransferase 2 (p?<?0.05) expression. The histological study showed a decrease in the fat deposits in the amaranth group when compared to casein; this is consistent with the biochemical and molecular parameters studied in this work. In conclusion, amaranth could be recommended to avoid the alterations in the lipid metabolism induced by alcohol and other harmful agents.  相似文献

The low incidence of cardiovascular disease in Mediterranean countries leads to an increased interest of the scientific community for the Mediterranean diet. Our aim was to evaluate total phenol and flavonoid contents, antioxidant capacity, free radical scavenging activity and potential antihypertensive effect of aqueous extract obtained from Thymus serpyllum L. (wild thyme, TE), an aromatic herb from the Lamiaceae family (highly present in Mediterranean diet), in spontaneously hypertensive rats (SHR) and in normotensive Wistar rats. Total phenol content of TE was 2008.33?±?10.6 mg/L GAE, and rosmarinic and caffeic acids were predominant phenolic compounds. The ferric reducing/antioxidant power and antioxidant capacity analysis revealed strong antioxidative properties of TE. In vitro nitric oxide-scavenging activity of 1 mg/l TE was 63.43 % with the IC50 value of 122.36 μg/ml. Bolus injection of TE (100 mg/kg body weight i.v.) induced significant decrease of systolic and diastolic blood pressure and total peripheral resistance in SHR, without effects on these parameters in normotensive Wistar rats. Cardiac index remained unchanged after TE treatment in all experimental rats. Given dose of TE did not show significant nitric oxide-scavenging activity in vivo. Our results indicate that TE may protect against hypertension in experimental model of essential hypertension.  相似文献

One of the most important strategy in the treatment of obesity includes the development of nutrient digestion and absorption inhibitors. Inhibition of digestive enzymes is one of the most widely studied mechanisms used to determine the potential efficacy of natural products as hypolipidemic and hypoglycaemic agents. In vitro studies here reported were performed to evaluate the inhibitory activity of five species (as hydroalcoholic extracts) of edible plants from Calabria region (Italy) on amylase and lipase by monitoring the hydrolysis of p-NPC and the hydrolysis of glycoside bonds in digestible carbohydrate foods. The formulation obtained from Clematis vitalba L. exhibited the strongest inhibitory effect on pancreatic lipase (IC50?=?0.99 mg/ml) and on α-amylase (IC50?=?31.52 μg/ml). In order to explore metabolome production HPTLC analysis of the extracts was performed, revealing the predominance of (±)-catechin, caffeic acid and chlorogenic acid in C. vitalba formulation at concentration of 23.18?±?3.14, 13.63?±?0.65 and 18.88?±?0.76 mg/g, respectively. The present study investigated the effect of dietary tempe, a fermented soy product, on the colonic environment of rats fed high-fat (HF, 30 % fat; experiment 1) or low-fat (LF, 6 % fat; experiment 2) diets. Growing male rats were fed the experimental diets with or without 25 % tempe for 21 days. Tempe consumption slightly but significantly increased the growth of rats fed both the HF and LF diets (P?<?0.05). With both the HF and LF diets, dietary tempe markedly reduced a harmful fecal secondary bile acid, lithocholic acid (a risk factor of colon cancer) (P?<?0.05), and markedly elevated fecal mucins (indices of intestinal barrier function) and immunoglobulin A (IgA, an index of intestinal immune function) (P?<?0.05). With the HF diet, dietary tempe increased cecal acetate, butyrate, propionate, and succinate concentrations (P?<?0.05). Analysis of the profile of cecal microflora revealed lower Bacteroides and higher Clostridium cluster XIVa levels in the tempe group of rats fed the HF diet (P?<?0.05). Compared with the control group, the fecal activity of β-glucosidase was markedly higher in the tempe group (P?<?0.05), while that of urease was lower (P?<?0.05) with both the HF and LF diets. A feeding trial was conducted in order to evaluate the potential effect on the lipid profile in a experimentally induced situation of hypercholesterolemia of a previously uninvestigated legume (Cicer aretinum L.) widely included in Mediterranean and Latinamerican human diets. Rats fed on a hypercholesterolemic diet containing saturated fat, cholesterol and cholic acid (H) had 123 percent higher serum cholesterol and 62 percent greater triacylglycerols levels than the animals receiving casein (C) protein. The LDL and VLDL cholesterol levels were 1330 percent and 35 percent higher, respectively, and HDL cholesterol 34 percent lower in the group of animals given the H diet as compared to controls. Further feeding of the hypercholesterolemic rats with animal protein (HC) resulted in a significant decrease of triacylglycerols (–70 percent), which reflected the decrease in the VLDL fraction. These effects on the lipid metabolism were more marked when the legumeCicer aretinum L. was present in the diet (HL). Significantly decreased concentrations of total cholesterol (–54 percent) and triacylglycerols (–70 percent) as well as the levels of LDL (–54 percent) and VLDL (–70 percent) were seen in rats fed chickpeas. In conclusion, a differential hypocholesterolemic effect between dietary casein and chickpea intake in a model of hypercholesterolemia induced by the diet was found, with beneficial effects on the lipid metabolism when legume was included in the diet as compared to casein. Arbutus unedo, Prunus spinosa, Rosa micrantha and Rosa canina are good sources of phenolic compounds, including anthocyanins. These compounds have potent antioxidant properties, which have been related to anticancer activity. Herein, the in vitro antioxidant and antitumor properties of enriched phenolic extracts (non-anthocyanin phenolic compounds enriched extract- PE and anthocyanins enriched extract- AE) of the mentioned wild fruits were evaluated and compared. PE gave higher bioactive properties than the corresponding AE. It was observed a high capacity of A. unedo phenolic extract to inhibit lipid peroxidation in animal brain homogenates (EC50?=?7.21 μg/mL), as also a high antitumor potential against NCI-H460 human cell line (non-small lung cancer; GI50?=?37.68 μg/mL), which could be related to the presence of galloyl derivatives (exclusively found in this species). The bioactivity of the studied wild fruits proved to be more related to the phenolic compounds profile than to the amounts present in each extract, and could be considered in the design of new formulations of dietary supplements or functional foods.  相似文献

To study the radiation utilization efficiency, latent heat flux, and simulate growth of rice during post-flood period in eastern coast of India, on-farm trial was conducted with three water regimes in main plots (W 1 = continuous flooding of 5 cm, W 2 = irrigation after 2 days of water disappearance, and W 3 = irrigation after 5 days of water disappearance) and five nitrogen levels in subplots (N 1 = 0 kg N ha?1, N 2 = 60 kg N ha?1, N 3 = 90 kg N ha?1, N 4 = 120 kg N ha?1, and N 5 = 150 kg N ha?1) on a rice cultivar, ‘Lalat’. Average maximum radiation utilization efficiency (RUE) in terms of above ground dry biomass of 2.09 (±0.05), 2.10 (±0.02), and 1.9 (±0.08) g MJ?1 were computed under W 1, W 2, and W 3, respectively. Nitrogen increased the RUE significantly, mean RUE values were computed as 1.60 (±0.07), 1.78 (±0.02), 2.060 (±0.08), 2.30 (±0.07), and 2.34 (±0.08) g MJ?1 when the crop was grown with 0, 60, 90, 120, and 150 kg ha?1 nitrogen, respectively. Midday average latent heat flux (on clear days) varied from 7.4 to 14.9 and 8 to 13.6 MJ m?2 day?1 under W 2 and W 3 treatments, respectively, at different growth stages of the crop in different seasons. The DSSAT 4.5 model was used to simulate phenology, growth, and yield which predicted fairly well under higher dose of nitrogen (90 kg and above), but the model performance was found to be poor under low-nitrogen dose.  相似文献

In addition to their favourable nutritional profile, legumes also contain a range of bioactive compounds such as phenolic compounds and phytosterols which may protect against chronic diseases including cancer and cardiovascular disease. Germination of some legume seeds has been previously reported to increase the concentration of the bioactive compounds. In this study, the effect of germination of Australian Sweet Lupin (ASL) seeds for 9 days on the concentration of some bioactive compounds and the radical scavenging activity in the resulting flour was determined. The concentration of total phenolic compounds in methanolic extracts of germinated ASL flour was determined using Folin Ciocalteu reagent and phytosterols in oil extracts were analyzed by gas–liquid chromatography. The methanolic and oil extracts were also used to determine radical scavenging activity toward 2,2-diphenyl-1-picrylhydrazyl. In the methanolic extracts of germinated ASL flour, phenolic contents and the antioxidant activity were significantly increased following germination (700 and 1400 %, respectively). Analysis of the oil extracts of germinated ASL flour revealed that the concentration of phytosterols and the antioxidant activity were also increased significantly compared to ungerminated ASL flour (300 and 800 %, respectively). The relative proportion of phytosterols in germinated ASL flour was: β-sitosterol (60 %), stigmasterol (30 %) and campesterol (10 %). Germination increases the concentration of bioactive compounds and the radical scavenging activity in the germinated ASL flour.  相似文献

Antioxidant properties of amaranth extracts isolated sequentially by acetone and methanol/water from defatted plant leaves, flowers, stems and seeds were assessed by ABTS+?, DPPH?, ORAC and total phenols content (TPC) assays. In addition, antioxidant properties of solid plant material were evaluated by the direct QUENCHER method using the same assays. Leaves and flowers of amaranth as well as their extracts possessed the highest antioxidant activities. Radical scavenging capacity in ABTS+? assay for leaves, flowers, stems and seeds evaluated by QUENCHER method were 144.24?±?2.41, 112.33?±?7.45, 19.05?±?1.13 and 21.82?±?1.06 μmol trolox equivalents in 1 g of dry weight, respectively. On-line HPLC-DPPH? assay was used to determine the activity of separated compounds and it was observed that rutin was the main radical scavenger in amaranth extracts. Preliminary screening of extract composition was performed by UPLC/ESI-QTOF-MS and rutin, nicotiflorin, isoquercitrin, 4-hydroxybenzoic and p-coumaric acids were identified by measuring their accurate mass and retention time.  相似文献

Mature cactus pears from Opuntia stricta have a dark purple color due to high betacyanin concentration, whose biosynthesis is initiated with the amino acid L-tyrosine as a primary precursor. This study followed the maturation and ripening processes of Opuntia stricta fruits to harvest them at high betacyanin and other antioxidant concentrations. Fruits lasted 9 months for final ripening. Physical and compositional changes at different maturation and ripening stages have been determined. Thus, ripe fruits were around 4.72?±?0.10 cm length, 2.94?±?0.05 cm diameter and 22.71?±?0.20 g weight; moisture and pH were maintained at 87.05?±?0.19 % and 3.37?±?0.12, respectively. Purple pigment production started in the ovary of immature fruits four months after anthesis (MAA). Concentration of all analyzed metabolites increased from immature (4 MAA) until ripe (9 MAA) stage. In ripe fruits, reducing sugars were 4.72?±?0.54 g/100 g ff and total phenols 135.17?±?0.68 mg gallic acid/100 g ff. Metabolites identified by HPLC were the betacyanins: betanin (60.17?±?1.08 mg/100 g ff), isobetanin (7.58?±?0.94 mg/100 g ff) and betanidin (13.48?±?0.87 mg/100 g ff). Also, L-ascorbic acid (35.03?±?1.06 mg/100 g ff) and L-tyrosine (4.43?±?0.73 mg/100 g ff) were determined. Furthermore, the addition of L-tyrosine or L-dopa to fruit pulp of moderately ripe fruits, increased betacyanin concentrations 17 (103.3?±?3.8 mg/100 g) and 32 % (114.3?±?4.1 mg/100 g), respectively.  相似文献

Phlomis lychnitis L. (Lamiaceae) is consumed as a traditional herbal tea in Spain. The antioxidant-protective effects as well as its phytoconstituents have never been established. The ability of the methanolic extract to protect cells from oxidative stress was evaluated in rat pheochromocytoma cells (PC12) using hydrogen peroxide as toxic agent. The viability of PC12 cells pre-treated with the methanolic extract of Phlomis lychnitis, determined by the MTT and LDH assays, was significantly improved at the highest dose (p?<?0.01). The antioxidant activity was confirmed evaluating the capacity of the plant to scavenge ABTS, DPPH, O2 · ? radicals and to inhibit XO. Bioassay guided fractionation led to antioxidant compounds. Qualitative HPLC/DAD/ESI/MS analysis reported phenylpropanoids, verbascoside being the major antioxidant constituent.  相似文献

In vitro effect of betulin-containing extract from Betula pendula Roth. bark on alpha-amylase activity was studied, the kinetic mechanism of interaction was proposed and in vivo effect of betulin-containing extract on weight gain and meat quality of broiler chickens was evaluated. The highest level of inhibitory activity (20 %) was detected in extract concentration of 1,000 mg/L. Increased extract concentration did not lead to increased enzyme inhibition. Using Dixon and Cornish-Bowden coordinates, the competitive mechanism of inhibition was demonstrated. Calculated kinetic parameters were: Km equal to 0.6 mg/mL, Vmax equal to 2.6 and 2.1 mM/min from Lineweaver-Burk and Dixon coordinates, respectively and Ki equal to 3,670?±?230 mg/mL. The partial inhibition of enzyme indicates the existence of low concentration of active inhibitory form, which reaches saturation level with increased extract concentration in applied suspension. Therefore, Ki has an apparent constant character. This partial inhibition of amylase activity observed in in vitro assay did not affect weight gain and meat quality of broiler chickens during in vivo assay. Rather, the tendency to increase the weight of edible parts and muscles compared to diet without additive suggests that the extract may be a potential food additive in poultry farming. Additionally, it could be a source for further pharmaceutical and pharmacological research.  相似文献

Sixty-five Solanum tuberosum group Andigena, Phureja and Stenotomum genotypes from an initial population of 1,500 were analyzed for phenylpropanoids, carotenoids, and antioxidant capacity. Total phenolic content ranged from 3 to 49 mg g?1 DW, total carotenoids from 4.1 to 154 μg/g DW, anthocyanins from 0.27 to 34 mg g?1 DW and antioxidant capacity from 60 to 1,767 μmol TE/g DW. HPLC analysis of phenolic extracts revealed that 5-O-chlorogenic acid (5CGA) was the most abundant polyphenol in all genotypes. Ten genotypes were independently grown out for more in-depth phytonutrient analysis. The Phureja genotypes RN 27.01 had the highest polyphenol, anthocyanin and antioxidant content, while RN 39.05 had the highest carotenoid content. The tuber percentage dry matter varied markedly among the ten genotypes, influencing the phytonutrient values when expressed on a dry weight basis. Chlorogenic acid concentrations ranged from 1.7 to 29.4 mg g?1 DW and kaempferol-3-rutinose was present up to 3 mg g?1 DW. Petunidin-3-O-coum-rutinoside-5-O-glu or pelargonidin-3-O-coum-rutinoside-5-O-glu were the most abundant anthocyanins. The principal carotenoids were lutein, zeaxanthin, violaxanthin, and antheraxanthin, but no one carotenoid was predominant in all genotypes. These findings further support utilization of Phureja group germplasm for phytonutrient enhancement efforts.  相似文献

The effect of supplementation of three fruits, Indian gall nut, bedda nut and gooseberry, on serum lipid levels and excretion of bile acids was investigated. Rats made hypercholesterolemic by feeding hypercholesterolemia inducing diet (HID) for a period of 30 days were used as the test model. Feeding of a dried powder of these fruits along with the HID resulted in significant (p<0.01) reduction in total cholesterol, LDL cholesterol and triglycerides. HDL cholesterol remained unchanged in groups fed gall nut and bedda nut. The effect of the administration of coriander seeds (Coriandrum sativum) on the metabolism of lipids was studied in rats fed a high fat diet with added cholesterol. The spice had a significant hypolipidemic action. The levels of total cholesterol and triglycerides decreased significantly in the tissues of the animals of the experimental group which received coriander seeds. Significant increases in -hydroxy, -methyl glutaryl CoA reductase and plasma lecithin cholesterol acyl transferase activity were noted in the experimental group. The level of LDL + VLDL cholesterol decreased while that of HDL cholesterol increased in the experimental group compared to the control group. The increased activity of plasma LCAT, enhanced hepatic bile acid synthesis and the increased degradation of cholesterol to fecal bile acids and neutral sterols appeared to account for its hypocholesterolemic effect.  相似文献
