The pest control potential demonstrated by various extracts and compounds isolated from the kernels and leaves of the neem plant (Azadirachta indica) A. Juss. (Meliaceae) seem to be of tremendous importance for agriculture in developing countries. Laboratory and field trial data have revealed that neem extracts are toxic to over 400 species of insect pests some of which have developed resistance to conventional pesticides, e.g. sweet potato whitefly (Bemisia tabaci Genn. Diptera: Aleyrodidae), the diamond back moth (Plutella xylostella L. Lepidoptera: Plutellidae) and cattle ticks (Amblyomma cajennense F. Acarina: Ixodidae andBoophilus microplus Canestrini. Acarina: Ixodidae). The compounds isolated from the neem plant manifest their effects on the test organisms in many ways, e.g. as antifeedants, growth regulators, repellents, toxicants and chemosterilants. This review strives to assess critically the pest control potential of neem extracts and compounds for their use in the tropics. This assessment is based on the information available on the wide range of pests against which neem extracts and compounds have proven to be toxic, toxicity to non-target organisms, e.g. parasitoids, pollinators, mammals and fish, formulations, stability and phytotoxicity. 相似文献

为明确感染南方水稻黑条矮缩病毒(Southern rice black-streaked dwarf virus,SRBSDV)水稻对非介体褐飞虱取食行为的影响,在人工气候室通过刺探电位监测技术对褐飞虱在感染SRBSDV水稻植株和健康水稻植株上的取食行为进行持续监测和比较分析。结果表明,与对照相比,在感染SRBSDV水稻植株上取食的褐飞虱其取食路径波Nc(N2+N3)(口针在维管束组织移动)的总持续时间显著延长了116.69%,Nc(N2+N3)波形持续时间占总监测时间的百分数也显著提高了100.42%;而口针刺探次数和刺探时间、在韧皮部及木质部取食次数和持续时间等均无显著差异。表明水稻感染SRBSDV后对褐飞虱的取食行为影响不大,仅显著延长了其到达取食位点的时间。 相似文献

为筛选对立枯丝核菌Rhizoctonia solani具有拮抗作用的生防细菌,利用稀释涂布平板法,从吉林省水稻纹枯病样中分离筛选出对立枯丝核菌AG-1A具有高拮抗活性的生防菌株,通过gyrB基因序列分析鉴定其分类地位,并采用离体侵染试验和温室防治试验测定筛选生防菌株对水稻纹枯病的防效,在显微镜下观察生防菌株预处理后接种病原菌的水稻叶片表面的菌丝生长情况,分析其生防机理。结果显示,从水稻纹枯病样中共分离获得35株菌株,其中菌株ND11对立枯丝核菌AG-1A的抑制率和抑菌圈直径最大,分别为74.12%和31.50 mm。基于gyrB基因序列分析最终将菌株ND11鉴定为短小芽胞杆菌Bacillus pumilus;该菌株能够减缓立枯丝核菌AG-1A的侵染速度,抑制其侵染垫的形成,对水稻纹枯病的温室防效达70.69%以上。表明短小芽胞杆菌菌株ND11能够通过抑制立枯丝核菌侵染垫的形成来防治水稻纹枯病,且防效较好,具有开发为生防菌剂的潜力。 相似文献

The antimicrobial activity of leaf and callus extracts of Melia azedarach was tested on in vitro shoot cultures of the peach rootstoch ‘MRS 2/5’ (Prunus cerasifera × Prunus spinosa) that were heavily contaminated with Sphingomonas paucimobilis (Sp) and Bacillus circulans (Bc). The extracts were filter-sterilised and added at 0%, 1%, 5%, 10% and 20% to a modified Murashige and Skoog proliferation
medium previously autoclave-sterilised. Up to about 17% shoots died with 10–20% extract, except for Sp-contaminated shoots,
whose survival was reduced to 50% after treatment with 20% extract. No shoots died with 1% to 5% supplement. The undiluted
leaf extract showed bactericidal activity on plated Sp and Bc isolates. The homogenates of shoots randomly collected from
treated cultures were processed for bacterial colony counting. Thus the 10% supplement was the best treatment for ridding
Bc-contaminated cultures of bacteria (although 5% had a similar bactericidal effect), and allowing shoot growth and proliferation
comparable to controls at the fifth subculture on a standard medium, while 20% extract was needed to eliminate Sp, and could
induce higher growth and proliferation rates in surviving shoots than in untreated cultures. Callus extract was ineffective.
The bactericidal activity of the leaf extract seemed attributable to a synergistic effect of azadirachtin with other unidentified

为建立一种检测苹果褪绿叶斑病毒(Apple chlorotic leaf spot virus,ACLSV)的Taq Man探针实时荧光定量RT-PCR方法,根据ACLSV外壳蛋白基因(coat protein,cp)保守序列设计了特异性引物和Taq Man探针,以构建的ACLSV-cp重组质粒为阳性标准品绘制标准曲线,并对该方法的特异性、灵敏性、重复性进行检验。结果显示,以ACLSV-cp重组质粒为标准品建立的标准曲线相关系数达0.999,扩增效率为103.7%;建立的Taq Man探针实时荧光定量RT-PCR方法特异性好,与苹果茎沟病毒(Apple stem grooving virus,ASGV)、苹果茎痘病毒(Apple stem pitting virus,ASPV)、苹果锈果类病毒(Apple scar skin viroid,ASSVd)均无交叉反应;灵敏度为100拷贝/μL,比常规RT-PCR高100倍;批内和批间变异系数均小于0.84%。表明Taq Man探针实时荧光定量RT-PCR方法具有特异性强、灵敏性高、重复性好的优点,适用于实际样品中ACLSV的快速准确检测。 相似文献

利用昆虫细胞-杆状病毒表达系统(Bac-to-Bac baculovirus expression system)表达获得小菜蛾 Plutella xylostella(L.)中肠氨肽酶N2(aminopeptidase N,APN2)蛋白,成功建立了该蛋白在昆虫细胞中的表达体系。将对Cry1Ac毒素敏感和已产生抗性的小菜蛾中肠APN2基因插入表达载体pFAST Bac HTB中,并在草地夜蛾 Spodoptera frugiperda 细胞系(Sf9)中进行了重组表达。聚丙烯酰胺凝胶电泳(SDS-PAGE)及蛋白免疫印迹技术(Western-blotting)结果显示,在107 kDa处有1条特异性蛋白条带。研究表明,小菜蛾中肠APN2基因可在Sf9细胞中成功表达,这为继续研究其功能及抗性的关系奠定了基础。 相似文献

Neem oil (NO) and pungam oil (PO) based emulsifiable concentrate (EC) formulations,viz., neem oil 60 EC (acetic acid) [NO 60 EC(A)], neem oil 60 EC (citric acid) [NO 60 EC(C] and neem oil + pungam oil 60 EC (citric
acid) [NO+PO 60 EC(C)], which had been developed at Tamil Nadu Agricultural University, were evaluated for their efficacy
against sheath rot of rice. All three formulations effectively inhibited the mycelial growth of the pathogen,Sarocladium oryzae, underin vitro conditions. There was no significant difference between efficacy of the freshly prepared and stored formulations in arresting
the growth ofS. oryzae; efficacy was maintained even after 9 months of storage. These formulations effectively controlled rice sheath rot and led
to increased yield in five field trials. Among the various treatments, the formulation NO 60 EC(A) achieved the highest grain
yield in four out of five field trials, with a pooled mean grain yield of 4684 kg/havs 3882 kg/ha in the control. NO 60 EC(A) achieved the maximum cost-benefit ratio of 1:4.8, followed by NO+PO 60 EC(C), with

Greenhouse trials were carried out in order to test the efficacy of different seed treatments as alternatives to chemicals
against Colletotrichum lindemuthianum cause of anthracnose on bean and Ascochyta spp. cause of Ascochyta blights on pea, respectively. Resistance inducers, commercially formulated microorganisms, non-formulated
selected strains of different microorganisms (fungi, bacteria and yeasts) and plant extracts were applied as dry or liquid
seed treatments on naturally infested seeds. Seedling emergence and disease incidence and/or severity were recorded. Almost
all seed treatments turned out to be ineffective in controlling the Ascochyta infections, which is in line with the literature
stating that these pathogens are difficult to control. The only alternative treatments that gave some control of Ascochyta spp. were thyme oil and a strain of Clonostachys rosea. The resistance inducers tested successfully controlled infections of bean by C. lindemuthianum. Among the formulated microorganisms, Bacillus subtilis-based formulations provided the best protection from anthracnose. Some strains of Pseudomonas putida, a disease-suppressive, saprophytic strain of Fusarium oxysporum and the mustard powder-based product Tillecur also proved to be effective against bean anthracnose. However, among the resistance
inducers as well as among the other groups, certain agents caused a significant reduction of plant emergence. Different alternative
seed treatments can therefore be used for the control of C. lindemuthianum on bean, while on pea only thyme oil and a strain of Clonostachys rosea showed some effectiveness against Ascochyta spp. 相似文献

A virus was isolated fromStreptocarpus plants that showed colour breaking of the flowers. Initial diagnostic tests indicated that this virus was a member of the Tobamovirus genus. The virus could be transmitted mechanically to several test plants. Its stability in plant sap was in line with that of other tobamoviruses, i.e. infectivity was lost after 10 min incubation at 90 °C and after dilution to 10–8. In addition, the morphology of the virus was typical for tobamoviruses. The particles had a length of about 304 nm. On test plants, the virus fromStreptocarpus could be distinguished from 7 well-defined tobamoviruses.Nicotiana glutinosa showed the most characteristic symptoms. In agar double-diffusion tests and/or double antibody sandwich enzyme-linked immunosorbent assays, no cross reactivity was observed in heterologous combinations with these 7 and 3 other tobamoviruses. Mechanical inoculation of the virus to virus-freeStreptocarpus plants resulted in the appearance of flower breaking in about 50% of the plants. On the basis of these findings, it is concluded that the virus that causes flower breaking inStreptocarpus is a distinct member of the Tobamovirus genus, and the nameStreptocarpus flower-break virus is proposed. 相似文献

为开发研制新型植物源柑橘保鲜剂,以抑菌圈直径为指标,利用牛津杯法筛选出对柑橘采后意大利青霉有良好抑制作用的21种中草药的醇提物,并对白薇醇提物防治脐橙青霉病的效果进行了评价。结果表明:白薇、零陵香、鸦胆子、藏青果、白药子5种中草药乙醇提取物对意大利青霉均有较好的抑制效果,抑制圈直径达26 mm以上,其中白薇醇提物抑菌活性最佳,抑制圈直径可达38.39 mm,最小抑菌浓度和最小杀菌浓度分别为3.13 mg/m L和6.25 mg/m L,且抑菌效果与对照药剂1 000倍咪鲜胺无显著差异。25 mg/m L白薇醇提物处理脐橙7 d后,对脐橙青霉病的防治效果达35.93%,与1 000倍咪鲜胺的防治效果相当(36.37%);白薇醇提物处理后可抑制意大利青霉菌菌丝生长,导致菌丝肿胀、膨大、畸形、细胞膜透性增加、细胞内含物外渗;还可诱导脐橙体内过氧化物酶活性下降,而苯丙氨酸解氨酶和几丁质酶活性上升。表明白薇醇提物对脐橙采后青霉病具有较好的防治效果,可进一步开发为植物源柑橘保鲜剂。 相似文献

A phytotoxic protein that evokes the typical symptoms of Verticillium wilt disease in seedlings of Gossypium hirsutum L. (Upland cotton) was isolated from culture filtrates of Verticillium dahliae. The protein was purified by ammonium sulfate precipitation, Sephadex-G100 fractionation, and native PAGE. The 18.5 kDa protein, designated VD18.5, appears to be a single subunit protein with an isoelectric point between 3 and 5. VD18.5 induces symptoms of leaf dehydration, chlorosis, necrosis and stem discoloration in seedlings of the disease susceptible cotton cultivar Siokra 1–4. The LD50 of VD18.5 on protoplasts of Siokra 1–4 was 18 μg mL−1. VD18.5 had no noticeable effect on Pima S-7, which is a disease resistant cultivar. Phytotoxic activity was partially destroyed at high temperature and was abolished by digestion with proteinase K. Mass spectrometry fingerprinting and protein sequence data from VD18.5 yielded no significant matches when submitted to the Mascot search engine and NCBI non-redundant protein databases, respectively. These results suggest that VD18.5 is a novel protein that may be involved in the development of some of the symptoms associated with Verticillium wilt disease in the cotton plant. 相似文献

The potential of biocontrol products and plant extracts for control of late blight on potato plants, caused by Phytophthora infestans was evaluated in detached leaf assays and on potted plants. Based on an initial screening of 22 preparations and plant extracts, the 10 most active treatments were selected for further investigation. In the detached leaf assays the commercial preparations Elot-Vis, Serenade and Trichodex, and plant extracts of Rheum rhabarbarum and Solidago canadensis showed a significant effect on the level of infestation by P. infestans. However, none of the treatments was as effective as copper. In the case of Serenade, the metabolites produced by its active micro-organism, Bacillus subtilis, were demonstrated to be the effective component of the formulation, and not the micro-organism itself. In order to take curative and protective modes of action into account, the test substances were applied 24 h before, or 90 min after inoculation with P. infestans. Generally, better effects were obtained when the applications were made 24 h before inoculation. For defining the optimum time of application, potted plants were treated 72 or 24 h before, and 1 and 24 h after inoculation with P. infestans. In these tests, Trichodex showed no activity, while Elot-Vis gave best results when applied 1 day before inoculation. Serenade and the extracts of R. rhabarbarum and S. canadensis (all at 5% concentration) however, were effective when applied up to 3 days before and just after inoculation with P. infestans. The results of the experiments on potted plants indicated direct effects on the pathogen for all agents except the extract of S. canadensis, but other mode of actions, e.g. induced resistance, could not be ruled out. The distribution of theFrankliniella speciesF. occidentalis (Pergande) andF. intonsa (Trybom) (Thysanoptera: Thripidae), and of the predatory bugOrius niger (Wolff) (Hemiptera: Anthocoridae), in various organs of the cotton plant, as well as prey — predator interactions between
thrips andO. niger, were investigated over 6 years in cotton fields in the eastern Mediterranean region of Turkey. The highest number of larvae
ofFrankliniella spp. were found inhabiting bolls, whereas the adults colonized mainly flowers. The majority of predatory bug nymphs were
present on leaves, followed by bolls, whereasO. niger adults visited mostly flowers. The thrips larvae were most likely preyed upon on flowers and squares, and bolls were safe
plant parts for thrips, with a low predation rate. An intermediate but relatively high predation rate occurred on cotton leaves.
In further field experiments, the effects of insecticide treatment on the relationships betweenO. niger andFrankliniella spp. were investigated. These trials revealed that a higher correlation existed between the numbers of adultO. niger in flowers andFrankliniella spp. in non-treated cotton fields than in insecticide-treated fields. The proportions of prey/predator in flowers ranged
from 1.53 to 19.28 and were below four thrips per predator at most sampling dates in some of the non-treated cotton fields.
It is concluded thatO. niger is an effective predator that can play an important role in suppressing population increase ofFrankliniella spp. in cotton.

为获得对水稻纹枯病有生防潜力的菌株,从湖北省恩施土家族苗族自治州植物根际土壤中分离获得菌株,筛选对水稻纹枯病菌有拮抗作用的菌株,并测定其发酵液的抑菌活性、抑菌谱及生物学特性;基于形态学特征、生理生化特征及分子生物学特征对其进行分类鉴定,并通过离体叶片、室内盆栽和田间试验进一步评价其生防潜力。结果显示,从根际土壤中共分离获得162株菌株,其中生防菌株E12对水稻纹枯病菌的抑制率最高,达94.93%,且该菌株对常见的12种植物病原菌均有抑制作用,抑制率在59.84%~93.14%之间;生防菌株E12能分泌蛋白酶、嗜铁素和淀粉酶,具有固氮及形成生物被膜的能力;结合形态学特征、生理生化特征及分子生物学特征,将菌株E12鉴定为越南伯克霍尔德氏菌Burkholderia vietnamiensis;生防菌株E12处理离体水稻叶片后,其相对病斑长度较对照显著减小,对水稻纹枯病的室内盆栽和田间防治效果分别为62.51%和67.78%。表明越南伯克霍尔德氏菌E12在防控水稻纹枯病方面有较好的潜力。 相似文献

A spray inoculation of the first leaf of 2-leaf stage cucumber plants with a non-pathogenic isolate of Alternaria cucumarina or Cladosporium fulvum before a challenge inoculation with the pathogen Sphaerotheca fuliginea induced systemic resistance to powdery mildew on leaves 2–5. Systemic resistance was expressed by a significant (p < 0.05) reduction in the number of powdery mildew colonies produced on each leaf of the induced plants, as compared with water-sprayed plants. Systemic resistance was evident when a prior inoculation with each of the inducing fungi was administered 1, 3 or 6 days before the challenge inoculation with S. fuliginea. Increasing the inoculum concentration of A. cucumarina or C. fulvum enhanced the systemic protection and provided up to 71.6% or 80.0% reduction, respectively, in the number of colonies produced on upper leaves, relative to controls. Increasing the inoculum concentration of S. fuliginea used for challenge inoculation, increased the number of powdery mildew colonies produced on both induced and non-induced plants. Pre-treated plants, however, were still better protected than controls, indicating that the level of systemic protection was related to the S. fuliginea inoculum concentration. The induction of systemic resistance against powdery mildew by biotic agents, facilitates the development of a wide range of disease management tools. 相似文献

Plant products are receiving greater attention as prophylactics against several species of plant-parasitic nematodes. Numerous experiments have shown the potential nematicidal value of plant parts and their by-products when incorporated into soil or when the plants themselves are interplanted as seedlings among crop plants. Various products (oils, cakes, extracts, etc.) prepared from the leaves and seeds of the neem plant (Azadirachta indica A. Juss) (Family Meliaceae) have been reported as effective protectants against nematode pests when used as root-dips and seed treatments. Nemato-toxic compounds of the neem plant, especially the azadirachtins, are released through volatilization, exudation, leaching and decomposition. The modes of action of these compounds are complex, and a number of mechanisms in relation to nematode management are yet to be fully explored. 为明确水稻锯齿叶矮缩病毒(rice ragged stunt virus,RRSV)对褐飞虱Nilaparvata lugens生长发育和生殖的影响,分别比较带毒褐飞虱与未带毒褐飞虱的若虫发育历期、成虫寿命、性比、翅型及产卵量,并利用实时荧光定量PCR(real-time quantitative PCR,RT-qPCR)技术分析RRSV侵染后褐飞虱体内卵黄原蛋白(vitellogenin,Vg)与其受体(vitellogenin receptor,VgR)以及海藻糖代谢途径中相关基因表达量的情况。结果显示,带毒褐飞虱的若虫历期显著高于未带毒褐飞虱的若虫历期,分别为17.56 d和15.90 d;带毒褐飞虱的成虫寿命、雌虫比例和短翅比例均高于未带毒褐飞虱的,但两者间均无显著差异;带毒褐飞虱较未带毒褐飞虱单雌日产卵量显著提升,分别为11.26粒和6.45粒;带毒褐飞虱的Vg、VgR和海藻糖转运蛋白基因(trehalose transporter,TRET)表达量均显著高于未带毒褐飞虱的。表明RRSV侵染会显著延长褐飞虱若虫发育历期,同时可能通过上调Vg、VgR以及TRET的表达量以促进褐飞虱生殖。 相似文献
