The follicle stimulating hormone beta-subunit (FSHβ) gene plays an important role in piglets. Marker-assisted selection (MAS) in conjunction with traditional selection methods is most effective for improving the piglets breeding traits. To find the new SNPs, the polymorphism of it in six pig breeds (Min, Landrace, Yorkshine, Duroc, wild boar, and wild boar × Landrace) was analyzed by using PCR-SSCP and was further compared with each other. Polymorphism was found and the sequencing results showed that there was one silent mutation on exon2 (C48T) and two mutations on exon3 (T422C and A514G). Genotype distribution of FSHβ in Min and wild boar on exon3 was in accordance with Hardy-Weinberg Law.  相似文献

为寻找与贵州小香羊繁殖力相关的分子标记提供基础数据,采用单链构象多态(SSCP)方法对在生殖中起重要作用的FSHβ基因外显子2的序列进行多态性检测,并对具有SSCP多态性的DNA片段进行测序分析。结果表明,在该特异性扩增产物中存在基因多态性,在94bp处检测到1个多态位点,该位点发生了G→A突变,出现3种基因型(AA、AB和BB)并导致编码的氨基酸由丙氨酸变为苏氨酸,A等位基因频率为62.3%,B等位基因频率为37.7%。  相似文献

The method of PCR-RFLP was used to analyze the effects of ESR and FSH β genotypes on the litter size, especially the corresponding changes with the difference of genotype distributions in the Min, Sanjiang and Junmu I pigs. The results show that the ESR and FSH β genes are the major genes of litter size ; the heterozygous genotype for ESR as well as for FSH β locus has a more litter size than others. However, the combined genotype ABDD means a more litter size than other genotypes,the frequencies of the heterozygous genotypes in Sanjlang and Junmu I are lower than in Min which maybe one of the most important reasons for the little litter sizes of those two breeds.  相似文献

[目的]探讨PPARγ-2基因多态性与广西巴马小型猪2型糖尿病易感性的关系.[方法]利用高糖高脂日粮饲喂24头广西巴马小型猪,利用PCR方法扩增PPARγ-2基因外显子2部分序列,测定其空腹血糖和胰岛素(INS)含量并进行糖耐量试验.[结果]克隆的PPARγ-2基因外显子2部分序列存在1处SNP位点(19813A/G),有AG(7头)和AA(17头)2种基因型.AG型广西巴马小型猪的空腹胰岛素含量和糖耐量试验中60 min血糖值显著高于AA型(P<0.05),而AG型广西巴马小型猪2型糖尿病发生率(71.4%)明显高于AA型(5.9%).[结论]广西巴马小型猪PPARγ-2基因外显子2的19813A/G多态性与2型糖尿病易感性有关.  相似文献

Grain texture in barley (Hordeum vulgare L.) is an important quality character. The single nucleotide polymorphisms (SNPs) in Hordoindoline a (hina) gene and its relationship with hardness index among North American harley cultivars were investigated. The hina genes in 36 cultivars were sequenced for the SNP analyses, and 17 sets of SNP primers were designed to detect the SNP variations of hina locus in 92 North American barley cultivars. SNP detection indicated that there were four haplotypes in the hina genes of 92 barley cultivars, and haplotype 01 and 02 were shared by 68 and 14 cultivars, respectively, suggesting that there was a very limited diversity in hina genes among North American barley cultivars. Despite the wide range in hardness exists in 92 barley cultivars, however, unlike wheat, where a clear relationship has been demonstrated between a number of SNPs in the wheat hardness genes and quality (soft or hard wheat), there was no such relationship for barley. The genotypes used in this study demonstrated that there was a low level of polymorphism in hina gene in North American barley cultivars and these polymorphisms had no impact on grain hardness.  相似文献
