Summary Pasteurella haemolytica causes two distinct disease syndromes in sheep. P. haemolytica biotype A causes septicaemia in young lambs and pneumonia in all ages of sheep. Biotype T produces an acute systemic disease affecting principally the upper alimentary tract and lungs in young adult sheep. The antibody response to various combined polyvalent Pasteurella haemolytica vaccines was studied in sheep and cattle. In sheep, certain oil adjuvant vaccines gave rise to a better antibody response to P. haemolytica than an A1(OH)3-adsorbed vaccine. This finding, however, was not consistent for all serotypes, and with respect to P. multocida, oil adjuvants had no advantage. Furthermore, it was found that the removal of all the culture supernatant fluid during the production process had no deleterious effect on the antigenicity of the product. In cattle, good responses were obtained with both alum-precipitated and A1(OH)3-adsorbed vaccine where all culture supernatant fluid was not removed during the production process. No advantage was gained with oil emulsion vaccines. The degree of immunity afforded to mice and the antibody response to different serotypes of P. haemolytica varied considerably. Further detailed studies with respect to specific serotypes of P. haemolytica are therefore required.  相似文献

A streptomycin-dependent, live Pasteurella haemolytica vaccine was given in 1 or 2 doses to 2 groups of weaned calves; 2 other groups of calves were not vaccinated. All calves in the vaccinated groups and calves in 1 of the nonvaccinated groups were stressed by transport, intratracheally inoculated with bovine herpesvirus type-1 (Cooper strain), and then intratracheally inoculated with P haemolytica type A1. The 4th group of calves (nonvaccinated controls) was not stressed and were not intratracheally inoculated with virus or bacteria. Mean daily weight gains, total clinical sign scores, lung lesion scores, plasma fibrinogen concentrations, and antibody titers against P haemolytica were determined at various intervals. Calves that had been vaccinated twice had greater mean daily weight gains and lower total clinical sign scores and lung lesion scores than did nonvaccinated, challenge-exposed calves, but the difference was not significant (P greater than 0.05). Calves vaccinated once had the greatest mean daily weight gains, the lowest total clinical sign scores, and the lowest lung lesion scores when compared with the other 2 challenge-exposed groups of calves. Mean daily weight gains and total clinical sign scores of calves vaccinated once were significantly different (P less than 0.05) than those of calves vaccinated twice. Nonvaccinated, nonchallenge-exposed control calves did not develop clinical signs of disease, did not develop lung lesions, and had consistently positive daily weight gains, and had scores in these areas that were significantly different (P less than 0.05) from those of all challenge-exposed groups of calves. Lipopolysaccharides (LPS) were extracted from a serotype of each of 2 species of Pasteurella isolated from sheep with respiratory tract infections. Lipopolysaccharides from P haemolytica 82-25 (serotype 1A) or P multocida P-1573 (serotype 12) were mixed with sheep lung surfactant and were incubated for 6 hours at 37 C. After incubation, LPS-surfactant mixtures were centrifuged overnight in sucrose density gradients, and fractions were analyzed. Binding occurred between LPS and surfactant vesicles resulting in a stable complex with densities greater than those with the surfactant alone. The surfactant alone had a density of 1.052 to 1.060 g/ml. Diffuse bands of surfactant had a density of 1.075 to 1.092 when incubated with P haemolytica LPS and a density of 1.069 to 1.105 when incubated with P multocida LPS.  相似文献

A live Pasteurella haemolytica vaccine efficacy trial   总被引:3,自引:0,他引:3  
A live Pasteurella haemolytica serotype 1 vaccine was used in an efficacy trial conducted on 100 lightweight feeder calves purchased from a Florida ranch. Forty-one calves were inoculated with the vaccine intradermally in the neck. Fifty-nine calves served as nonvaccinated controls. Fourteen days later, the calves were shipped to an order buyer in eastern Tennessee, where the calves were mixed with 60 local calves in a community sale barn for 72 hours. After 3 additional days, the calves were shipped to a research feedlot in Bushland, Tex. They remained in the feedlot for 56 days, and the test was concluded 76 days after vaccination. The P haemolytica vaccine had no significant effect on performance, morbidity, or mortality. There was no significant difference between the vaccinated and nonvaccinated calves in the number of times Pasteurella was isolated. The calves became seropositive to bovine viral diarrhea virus, respiratory syncytial virus, and infectious bovine rhinotracheitis (IBR) virus during the 76-day experiment. All calves initially were seropositive to parainfluenza-3 virus. A virulent outbreak of IBR occurred 30 days after the calves arrived at the feedlot. Before the onset of IBR, the isolation of P haemolytica serotype 1 from nasal turbinates was rare (2 of 500 nasal swabs). After the IBR outbreak, P haemolytica serotype 1 was isolated from 40 of 92 calves.  相似文献

Three vaccines containing Pasteurella haemolytica serotype A2 antigens were tested for their ability to protect sheep against a homologous challenge. A crude cytotoxin preparation in combination with a sodium salicylate extract (SSE) or crude cytotoxin alone were highly protective (98 and 86%, respectively), whereas SSE alone was poorly (47%) protective. These findings indicated that the crude cytotoxin was an essential component of a protective vaccine. Protection correlated with serum cytotoxin-neutralising (CN) titres and bactericidal activity, which were stimulated by antigens in the crude cytotoxin preparation.  相似文献

A group of Caesarian-derived, colostrum-deprived lambs was inoculated intranasally and intratracheally with a virulent Mycoplasma ovipneumoniae isolate selected from ovine mammary studies and propagated in an ovine mammary gland. Other groups of lambs were inoculated with M. ovipneumoniae in combination with Pasteurella haemolytica type Al or P. haemolytica alone. The M. ovipneumoniae isolate alone did not induce any specific pneumonic lesions in the lambs and when combined with P. haemolytica type Al did not increase the severity of the P. haemolytica-type lesions. Fifty percent of lambs inoculated with P. haemolytica developed a purulent and exudative bronchopneumonia with pleurisy and high titres of P. haemolytica were recovered from these lesions.  相似文献

Twelve Pasteurella-free Holstein-Friesian calves were used in a study to test the efficacy of a live streptomycin-dependent Pasteurella multocida A:3 and streptomycin-dependent Pasteurella haemolytica A1 vaccine. The calves were inoculated intramuscularly twice at 14-day intervals with either the streptomycin-dependent vaccine, containing 1 X 10(6) colony forming units/mL P. multocida and 4 X 10(8) colony forming units/mL P. haemolytica, commercial bacterin, or phosphate buffered saline. Two weeks following the second vaccination, all calves were challenged by intranasal inoculation of 10(8) TCID50/4.0 mL infectious bovine rhinotracheitis virus followed three days later by intratracheal injection with 2.3 X 10(7) colony forming units/mL of a 16 hour culture of P. multocida A:3 and 2.6 X 10(8) colony forming units/mL of an 8 hour culture of P. haemolytica A1. Seven days after challenge with Pasteurella, calves were killed for collection of tissues at necropsy. Each calf was given a score based on macroscopic and microscopic lesions. The scores for the calves receiving live vaccines were significantly lower (p less than 0.025) than those for the controls. Also, the calves receiving live vaccines had a significant (p less than 0.05) increase in the level of serum antibody to P. haemolytica. The results of this preliminary study showed that the streptomycin-dependent vaccine offered better protection than the commercial bacterin against a virulent homologous challenge.  相似文献

A vaccine against pneumonic pasteurellosis was evaluated for efficacy at two dilutions in lambs transported by sea from New Zealand to Saudi Arabia. The experimental vaccine was a killed Pasteurella haemolytica serotype A1 and A2 preparation. There was no evidence of either dilution of the vaccine leading to a lower pneumonia death or lesion rate than for the control group. However, bacteriological examinations to establish the causality of the pneumonia cases showed Pasteurella multocida to be the dominant organism, while P. haemolytica types A1 and/or A2 occurred at only a very low incidence.  相似文献

Ovine isolates of the 15 known serotypes found within the A and T biotypes of Pasteurella haemolytica were cytotoxic for sheep bronchoalveolar macrophages (BAM). Weaker toxicity for the same target cells was also expressed by non-serotypable ovine isolates of P. haemolytica. Six Rocky Mountain bighorn sheep were raised in captivity from birth (n = 5) or taken from the wild as a lamb (n = 1). After the bighorn sheep were in captivity for over a year, 6 clinically normal domestic sheep were placed on the 2 ha of pasture on which the bighorn sheep were kept. Nasal swab specimens were obtained from all sheep at the time the domestic sheep were introduced. Pasteurella haemolytica was isolated from swab specimens obtained from 4 of 6 domestic sheep, but not from specimens obtained from the bighorn sheep. All 6 bighorn sheep died of acute hemorrhagic pneumonia after exposure to domestic sheep. Death in the bighorn sheep occurred on days 4, 27, 27, 29, 36, or 71 after initial exposure to domestic sheep. Pasteurella haemolytica was isolated from respiratory tract tissue specimens of all bighorn sheep at the time of death. None of the domestic sheep were clinically ill during the study. At the end of the study, 3 of 6 domestic sheep were euthanatized, and at necropsy, P haemolytica was isolated from 2 of them. The most common serotypes in bighorn and domestic sheep were P haemolytica T-3 and A-2. Other serotypes isolated included P haemolytica A-1, A-9, and A-11 in bighorn sheep and A-1 in domestic sheep. On the basis of results of this study and of other reports, domestic sheep and bighorn sheep should not be managed in proximity to each other because of the potential fatal consequences in bighorn sheep.  相似文献

A field trial compared a modified Pasteurella haemolytica biotype A serotype 1 leukotoxin vaccine to a commercial vaccine during March-July 1995 in a Natal Midlands, South African, feedlot. Weaners/long weaners purchased by the feedlot were allocated systematically into test vaccine and control vaccine groups of 1241 and 1240 head, respectively, and fed in groups of approximately 200 head. Morbidity and mortality were monitored until the animals were marketed. Details of pleuritis and pneumonia at veterinary meat inspection were recorded for 409 test-vaccinated and 424 control-vaccinated cattle. An increase in morbidity but not mortality risk of respiratory disease was shown between test (13.8% morbidity) and control (11.4% morbidity) groups. Cattle with a processing weight <245 kg were 1.4 times more likely to develop respiratory diseases than cattle with a processing weight > or =245 kg. Cattle bought on auction were 1.6 times more likely to develop respiratory disease than cattle bought at private sales. A partial farm budget incorporating Latin Hypercube sampling of uncertain variables was done to obtain the distribution of possible financial outcomes if the test vaccine were used. Impact (sensitivity) analyses indicated that median weight of carcass cut away had the greatest impact on the profit margin. The partial farm budget highlighted the importance of reducing sub-clinical lesions in a feedlot.  相似文献

An enzyme-linked immunosorbent assay utilising direct binding of capsular polysaccharide antigens to polystyrene immunoassay plates was used to measure sheep antibodies to Pasteurella haemolytica A1, A2 and A6 serotypes. Low level cross reactivity occurred between A2 antigen and heterologous antisera. A strong unilateral cross reaction between A1 antigen and anti-A6 serum was abolished by absorption. These reactions suggest shared capsular antigens between serotypes.  相似文献
