During the occasional testing of Escherichiacoli from faecal samples of young calves we observed multi‐resistant isolates. Because of the significance of E. coli as an indicator bacterium for resistance trends we tested E. coli populations of young calves over a longer period. Here we present the results of a retrospective study comparing isolates from 1998 to 2000. Moreover, we compared, in a clinical study, the resistance rates of E. coli populations from 67 hospitalized calves both before and after hospitalization (with or without anti‐microbial therapy), and with their anamnestic data of antibiotic usage. The highest resistance rates were found to be more than 80% for tetracyclines, ampicillin, sulfonamide/trimethoprim combinations, and chloramphenicol. A significant increase or decrease over the years was not observed. In analysing the data of hospitalized calves, an increase of resistance to some anti‐microbials had to be registered that seemed to be connected with the selective pressure due to agents used in the clinic. In comparing anamnestic data and resistance rates it became obvious that reliable data are not easily available and that a number of potential anti‐microbial influence factors have to be taken into account.  相似文献

A comparative study of investigations from 1970 (before the prohibition against therapeutically used antibiotics as food additives for growth promotion) and from 1978 respectively proves that the prevalence of young pigs and calves with resistant E. coli has increased significantly from 61% to 92% and from 69% to 86% (tables I and II). Figures 1 and 2 show the prevalence of young pigs and calves with antibiotic resistant E. coli with resistance determinants towards sulfonamides, tetracycline, streptomycin, ampicillin, chloramphenicol, neomycin and furazolidone. This shows that the prevalence of young pigs with resistance determinants towards sulfonamides, tetracycline, streptomycin, ampicillin, and chloramphenicol has increased significantly from 1970 to 1978. The same facts apply to the calves. However, the increase in tetracycline resistance is not significant. Neomycin resistant E. coli for both young pigs and calves were only found in 1978. Figure 3 shows the distribution of the number of resistance determinants per young pig and calf with resistant E. coli. The figure shows the significant tendency that the pigs and calves investigated contain resistant E. coli with more resistance determinants in 1978 than in 1970. The distribution of R-factors among resistant E. coli seems to be reduced at first sight but a closer examination establishes that this only goes for R-factor mediated sulfonamide resistance (the experimental conditions may account for this). R-factor mediated streptomycin resistance has increased while R-factor mediated tetracycline resistance was at the same level. On the basis of literature the causes and risks of an increased distribution of antibiotic resistant E. coli are discussed.  相似文献

Escherichia coli isolates from calves were investigated by multiplex PCR assays for the presence of genes encoding K99, F41, F17-related fimbriae, heat-stabile enterotoxin a (STa), intimin (eae) and Shiga toxins (stx1 and stx2). A total of 120 E. coli isolates, 75 isolated from diarrhoeic or septicemic calves and 45 from clinically healthy calves aged between 1 day and 2 months were tested. Each isolate was obtained from different calves in different herds. Among the isolates from diseased animals, 12 (16%) isolates from 1- to 7-day-old diarrhoeic calves were detected as enterotoxigenic E. coli which possessed K99, F41 and STa in combination; F17-related fimbriae genes were detected in 33 (44%) isolates and they were found in combination with K99 + F41 + STa in two isolates. Of 120 isolates, 16 carried eae, eight stx1 and five stx2 genes alone or in combination. None of the eae- or stx-positive strains was identified as O157:H7. However, results indicate that calves may be carrier of Shiga toxin-producing E. coli which have potential as a human pathogen. Antimicrobial susceptibility of 75 isolates from diseased calves was determined by agar disk diffusion method for 14 antimicrobial agents. In 77.3% of the isolates, multiresistance was detected. Higher resistance rates were detected for cephalothin (72%), tetracycline (69.3%), kanamycin (69.3%), ampicillin (65.3%), nalidixic acid (53.3%), trimethoprim-sulphamethoxazole (52%) and enrofloxacin (41.3%), respectively. No resistance was found for ceftiofur and cefoxitin.  相似文献

In 2009, 1462 Escherichia coli isolates were collected in a systematic resistance monitoring approach from primary production, slaughterhouses and at retail and evaluated on the basis of epidemiological cut-off values. Besides resistance to antimicrobial classes that have been extensively used for a long time (e.g. sulphonamides and tetracyclines), resistance to (fluoro)quinolones and third-generation cephalosporins was observed. While in the poultry production chain the majority (60%) of isolates from laying hens was susceptible to all antimicrobials tested, most isolates from broilers, chicken meat and turkey meat showed resistance to at least one (85-93%) but frequently even to several antimicrobial classes (73-84%). In the cattle and pig production chain, the share of isolates showing resistance to at least one antimicrobial was lowest (16%) in dairy cows, whereas resistance to at least one antimicrobial ranged between 43% and 73% in veal calves, veal and pork. Resistance rates to ciprofloxacin and nalidixic acid in isolates from broilers were 41.1% and 43.1%, respectively. Likewise, high resistance rates to (fluoro)quinolones were observed in isolates from chicken meat and turkey meat. In contrast, ciprofloxacin resistance was less frequent in E.?coli isolates from the cattle and pig production chain with highest rate in veal calves (13.3%). Highest resistance rates to cephalosporins were observed in broilers and chicken meat, with 5.9% and 6.2% of the isolates showing resistance. In dairy cattle and veal, no isolates with cephalosporin resistance were detected, whereas 3.3% of the isolates from veal calves showed resistance to ceftazidime. Resistance to (fluoro)quinolones and cephalosporins in E.?coli isolates is of special concern because they are critically important antimicrobials in human antimicrobial therapy. The emergence of this resistance warrants increased monitoring. Together with continuous monitoring of antimicrobial usage, management strategies should be regularly assessed and adapted.  相似文献

OBJECTIVE: To compare antimicrobial susceptibility patterns of Escherichia coli isolates cultured from fecal samples from cows and calves on dairy farms that used organic (ie, no or severely limited antimicrobial use) versus conventional production methods. DESIGN: Cross-sectional study. SAMPLE POPULATION: Fecal samples from 10 cows and 10 calves on each of 30 organic dairy farms and 30 neighboring conventional dairy farms in Wisconsin. PROCEDURE: E. coli isolates obtained from the fecal samples were tested for susceptibility to 17 antimicrobials by means of a microbroth dilution test. Prevalence of antimicrobial resistance was compared between organic and conventional dairy farms. RESULTS: E. coli was isolated from 1,121 (94%) fecal samples. Farm type (organic vs conventional) and animal age (cow vs calf) were significantly associated with odds that E. coli isolates would be resistant to various antimicrobials. After controlling for age, logistic regression analyses indicated that isolates from conventional dairy farms had significantly higher rates of resistance to ampicillin, streptomycin, kanamycin, gentamicin, chloramphenicol, tetracycline, and sulfamethoxazole than did isolates from organic dairy farms. However, no significant differences were detected for the 10 other antimicrobials that were tested. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that compared with isolates from conventional dairy farms, E. coli isolates from organic dairy herds have significantly lower prevalences of resistance to 7 antimicrobials; however, prevalence of resistance was not significantly different for 10 other antimicrobials. Resistance was more common for isolates from calves than for isolates from adult dairy cows.  相似文献

河北省犊牛腹泻大肠杆菌致病性及耐药性分析   总被引:1,自引:0,他引:1  
为确定河北省犊牛腹泻大肠杆菌的致病性与耐药性,本研究对2018年1月至2019年6月期间从河北省石家庄、保定、承德、唐山、廊坊的部分肉牛场腹泻犊牛样品中分离到的50株大肠杆菌进行了致病性试验、药物敏感性试验、毒力基因和耐药基因检测。结果显示:50株分离菌均对小鼠具有致病性,致病菌占比100%(50/50)。毒力基因fyuA、irp2、eaeA、ler检出率分别为68.0%、66.0%、34.0%、34.0%。50株分离菌均对15种抗生素中的2种及以上表现为耐药,对10种及以上抗生素耐药的菌株占比达34%(17/50);分离菌对土霉素、磺胺对甲氧嘧啶、磺胺间甲氧嘧啶、替米考星4种药物表现为高度耐药,耐药率分别为90%、90%、94%、100%。除四环素类tetD基因检出率为0外,其它耐药基因均有检出,其中四环素类tetC、氨基糖苷类aadA1、喹诺酮类gyrA、gyrB基因检测率高达100%。试验的50株大肠杆菌均具有较强的毒力和多重耐药性。本研究为河北省大肠杆菌所致犊牛腹泻病的防治提供了实验依据。  相似文献

This study uses cluster analysis techniques to describe the antibiotic susceptibility patterns seen in calf fecal Escherichia coli (E. coli). Cohorts of 30 dairy calves at six farms were sampled at 2-week intervals during the pre-weaning period. At each sampling occasion five fecal E. coli isolates per calf were analyzed for antibiotic susceptibility to 12 antibiotics using the disk diffusion method. All isolates had a profile consisting of the aggregate measured inhibition zone size for each of the evaluated antibiotics. Several cluster analytic algorithms were assessed to partition the E. coli isolates. For our data, Ward's minimum variance method met the objectives of the study. Relative to the number of possible combinations of resistance clusters, a parsimonious set of 14 patterns was developed. This set of E. coli isolates exhibited a limited set of resistance patterns to the different antibiotics indicating that certain resistance genes may be linked.  相似文献

It is believed that the intensive use of antibiotics in the management of disease in pre-weaned calves contributes to high levels of antibiotic resistance in commensal and pathogenic bacteria. We described the temporal dynamics of antibiotic-susceptibility patterns seen in bovine enteric Escherichia coli in pre-weaned calves on dairy farms and dedicated calf-rearing facilities. Cohorts of 30 calves at each of six farms were sampled at 2-week intervals during the pre-weaning period. Faecal E. coli isolates were analyzed for antibiotic susceptibility to 12 antibiotics with the disk-diffusion method and grouped using cluster analysis of inhibition-zone patterns. The influences of calf age, farm-type, and individual-calf antibiotic therapy on the clusterings were assessed using stratified analyses and cumulative multinomial logistic regression using generalized estimating equation with antibiotic-resistance cluster as an ordinal-dependent variable. The model controlled for farm and cohort by a nested design and included a repeated measure on calf at each sampling occasion. E. coli from calves 2 weeks of age were more likely to be increasingly multiply resistant than E. coli from day-old calves (OR = 53.6), as were 4- and 6-week-old calves (OR = 29.8 and 16.4, respectively). E. coli from calves on dedicated calf-rearing facilities were more likely to be increasingly multiply resistant than E. coli from dairy-reared calves (OR = 2.4). E. coli from calves treated with antibiotics within 5 days prior to sampling were also more likely to be increasingly multiply resistant than E. coli from calves not exposed to individual antibiotic therapy (OR = 2.0).  相似文献

Rectal smears of calves, cows and young bulls, as well as cloacal smears of house sparrows (Passer domesticus), from farms at the villages of Sumice and Troskotovice, Czech Republic, were examined for E. coli resistant to 12 antimicrobials. The resistant isolates were tested for antimicrobial-resistance genes and integrons. Totals of 40% (n=183), 3% (n=95), 0% (n=33), and 9% (n=54) of Escherichia coli isolates from calves, cows, young bulls and house sparrows, respectively, were antimicrobial resistant. The following genes were identified in cattle E. coli isolates: tetA, tetB (isolates resistant to tetracycline), bla(TEM) (beta-lactams), strA, aadA (streptomycin), sul1, sul2 (sulphonamides), and cat, floR (chloramphenicol). Seven of 16 antimicrobial-resistant calf isolates from the Sumice farm possessed class 1 integrons with the aadA1 gene cassette integrated, 1 kb in size. On the Troskotovice farm, eight of 57 antimicrobial-resistant calf isolates possessed class 1 integrons. Integrons of 1.5kb with the dhfr1- aadA1 gene cassette were found in four isolates, followed by a 1kb integron with the aadA1 gene found in three isolates, and a 1.7kb integron with the dhfr17-aadA5 gene cassette and the phenotype ASSuTSxtNaCipCCfG. The prevalence of resistant E. coli in calves compared to adult cattle was much higher and probably was influenced by oral antimicrobial usage in calves, feeding with milk and colostrum from treated cows, as well as mechanisms unrelated to antimicrobial drug selection. Although house sparrows lived together with the cattle and came into contact with cattle waste on the farm, they were not infected by resistant E. coli isolates with the same characteristics as those found in cattle.  相似文献

A randomized, controlled, blinded clinical trial was performed at a research feedlot in western Canada. Auction-market-derived steers (n = 288) were randomly assigned to 1 of 3 treatments: 1) no antimicrobials on arrival; 2) oxytetracycline in the starter ration for 14 d; and 3) long-acting oxytetracycline subcutaneously on day 0. Minimal inhibitory concentrations of 7 antimicrobials were determined for 3 generic fecal E. coli isolates per animal on arrival and throughout the feeding period. There was a low prevalence of antimicrobial resistance in generic E. coli isolates from calves on arrival. There were increased proportions of cattle with resistant E. coli isolates early in the feeding period among calves in groups 2 and 3. Individual animal treatments were not associated with increased proportions of cattle with resistant E. coli isolates preslaughter. There was no difference in the proportion of animals with E. coli isolates resistant to tetracycline between the treatment groups preslaughter. However, there were significantly more animals with tetracycline resistant isolates of E. coli preslaughter than at arrival.  相似文献

Diagnostic laboratory data on antimicrobial susceptibility of Escherichia coli isolated from feces of spring calves were evaluated retrospectively for the 5-year period from 1999 to 2003. The antimicrobials to which resistance was most prevalent were tetracycline, ampicillin, and trimethoprim/sulphamethoxazole. Resistance to 3 or more antimicrobials was found in 52.5% [95% confidence interval (CI): 47.9 to 56.6] of the E. coli isolates. Incomplete records reduced the usefulness of the diagnostic laboratory data for surveillance. Standardized patient information submitted by veterinary clinics would increase the value of this data for surveillance.  相似文献

为了解畜禽源大肠杆菌的耐药状况,2011—2012年从患病和健康食品动物中分离鉴定大肠杆菌935株,包括健康猪源606株、患病猪源114株、患病鸡源51株、患病水禽源(鸭和鹅)164株。试验采用琼脂稀释法测定了不同动物来源的大肠杆菌对15种抗菌药物的敏感性。结果发现,大肠杆菌耐药严重,对大部分药物的耐药率超过70.0%,其中氨苄西林、复方新诺明和四环素的耐药率均达到了80.0%以上;仅对头孢西丁、黏菌素和阿米卡星较敏感,但患病水禽源大肠杆菌对阿米卡星的耐药率达到了52.4%,明显高于其他动物。健康畜禽源大肠杆菌对多数抗菌药的耐药率低于患病畜禽源大肠杆菌,表明抗菌药物的使用增加了病原菌对第3代头孢类药物和氟喹诺酮类药物的耐药率,势必增加临床治疗难度。  相似文献

The resistance to anti-microbial agents of bacteria isolated from pathological conditions of birds in Victoria, 1978 to 1983, was determined for isolates of Escherichia coli, Salmonella species, Staphylococcus aureus, Pasteurella multocida, P. anatipestifer, Yersinia pseudotuberculosis and Haemophilus paragallinarum. The isolates of E. coli had a high prevalence of resistance to tetracycline and sulphonamides, and a lower prevalence of resistance to furazolidone and sulphamethoxazole-trimethoprim. The isolates of Salmonella spp commonly had resistance to tetracycline, sulphonamides, furazolidone and sulphamethoxazole-trimethoprim. Almost half the isolates of S. aureus showed resistance to lincomysin and many showed resistance to penicillin. Resistance to tetracycline was found in isolates of P. multocida, P. anatepestifer and Y. pseudotuberculosis. Some isolates of H. paragallinarum showed resistance to sulphonamides, streptomycin and sulphamethoxazole-trimethoprim.  相似文献

Verotoxin-producing Escherichia coli O111:NM were isolated from two five week old Holstein calves with dysentery. On necropsy both calves had pseudomembranous ileitis, mucohemorrhagic colitis and proctitis. Large numbers of E. coli O111:NM were isolated from the colon and lesions typical of attaching-and-effacing E. coli were evident. The isolates from both calves had identical biochemical reactions and antimicrobial resistance patterns. Oral inoculation of a four day old colostrum deprived calf with 1 x 10(10) organisms of E. coli O111:NM produced a mild, focal colitis with typical attachment and effacement lesions. We conclude that the strain of E. coli O111:NM isolated from the clinical cases has the ability to produce colitis characterized by attachment and effacement of the colonic mucosa.  相似文献

The use of antimicrobial drugs in livestock is suspected to contribute to bacterial antimicrobial resistance (AR) development. Dairy farms experiencing recent outbreaks of salmonellosis involving multi-resistant (MR) Salmonella strains were compared to control farms with respect to AR among bovine commensal E. coli isolates. For most antimicrobials tested, the percentage of AR E. coli isolated from salmonellosis-affected farms was significantly higher than that from control farms. Calf E. coli from both case and control farms had greater levels of AR than cow isolates. Commensal E. coli isolates from case farms and calves tended to more frequently be MR. These data are consistent with the existence of higher antimicrobial selection pressure on farms with recent salmonellosis outbreaks, however, the directionality of the relationship remains to be elucidated. An improved understanding of the epidemiology of AR bacteria in livestock production, both at the herd and molecular level, is essential to mitigate risk to public health and food safety.  相似文献

OBJECTIVE: To determine whether duration of hospitalization in the intensive care unit (ICU) of a veterinary teaching hospital was associated with prevalence of antimicrobial resistance among rectal Escherichia coli isolates from dogs, whether antimicrobial treatment was associated with prevalence of antimicrobial resistance, and whether there were associations among antimicrobial drugs to which isolates were resistant. DESIGN: Prospective observational study. ANIMALS: 116 dogs hospitalized in an ICU for >or= 3 days. PROCEDURES: Rectal swab specimens were obtained every 3 days and submitted for bacterial culture for E coli. Isolates were tested for susceptibility to 12 antimicrobial agents by means of disk diffusion. RESULTS: For each additional day that a dog was hospitalized in the ICU, the odds of being colonized with an E coli isolate resistant to 1 or more of the 12 antimicrobials tested increased by a factor of 1.5, independent of antimicrobial treatment. Dogs that were treated with enrofloxacin were 25.6 times as likely to be colonized by a quinolone-resistant E coli strain as were dogs that did not receive any antimicrobials. Significant correlations were found for resistance to agents in the extended-spectrum cephalosporin group and the quinolone group. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that the proportion of rectal E coli isolates obtained from dogs housed for >or= 3 days in a veterinary teaching hospital ICU that were resistant to antimicrobial agents increased as the duration of hospitalization in the ICU increased. Thus, ICU hospitalization time should be as short as possible to prevent development of antimicrobial resistance among rectal E coli isolates.  相似文献

Within the set of 200 strains of E. coli isolated from healthy calves and 60 strains of E. coli isolated from calves suffering from diarrhoea we compared the incidence and transfer of determinants of antibioticoresistance, colicinogenesis and hemolytical activity. A significant difference in the incidence and independent mobility of the agents under examination in favour of E. coli from calves suffering from diarrhoea was determined in the case of resistance to chloramphenicol. The increased incidence and independent mobility of the chloramphenicol element in the antibioticoresistent strains of E. coli isolated from calves suffering from diarrhoea can be explained by the three to five-day therapy with a chloramphenicol product.  相似文献

Quinolone resistance is studied and reported increasingly in isolates from humans, food-producing animals and companion animals. Resistance can be caused by chromosomal mutations in topoisomerase genes, plasmid-mediated resistance genes, and active transport through efflux pumps. Cross sectional data on quinolone resistance mechanisms in non-pathogenic bacteria from healthy veal calves is limited. The purpose of this study was to determine the prevalence and characteristics of quinolone resistance mechanisms in Escherichia coli isolates from veal calves, after more than 20 years of quinolone usage in veal calves. MIC values were determined for all isolates collected as part of a national surveillance program on antimicrobial resistance in commensal bacteria in food-producing animals in The Netherlands. From the strains collected from veal calves in 2007 (n=175) all isolates with ciprofloxacin MIC ≥ 0.125 mg/L (n=25) were selected for this study, and screened for the presence of known quinolone resistance determinants. In this selection only chromosomal mutations in the topoisomerase type II and IV genes were detected. The number of mutations found per isolate correlated with an increasing ciprofloxacin MIC. No plasmid-mediated quinolone resistance genes were found. The contribution of efflux pumps varied from no contribution to a 16-fold increase in susceptibility. No correlation was found with the presence of resistance genes of other antimicrobial classes, even though all quinolone non-wild type isolates were resistant to 3 or more classes of antibiotics other than quinolones. Over twenty years of quinolone usage in veal calves in The Netherlands did not result in a widespread occurrence of plasmid-mediated quinolone resistance, limiting the transmission of quinolone resistance to clonal distribution.  相似文献

CS31A is a K88-related non-fimbrial adhesin first described on Escherichia coli strains isolated from diarrhoeic and septicaemic calves. In this report, CS31A antigen was screened by immunological methods and confirmed by PCR among bovine E. coli isolates. In addition, CS31A-producing strains were characterized with respect to different fimbrial antigens, O-serogroup and other properties related to virulence. Faecal or tissue specimens of 100 diarrhoeic or septicaemic calves and 27 older cattle with different pathologies from 71 outbreaks or individual cases that occurred in Buenos Aires province, Argentina, were examined. CS31A + E. coli strains were isolated from 21 (21.0%) calves from 16 outbreaks or individual cases. No CS31A + E. coli was detected in samples from cattle more than 1 year old. Fimbriae F5, F41, F17a and F17b were not detected among the CS31A-producing strains. Three (14.3%) of the CS31A+ E. coli strains expressed the F17c fimbria. All of the 21 isolates exhibited at least one property of septicaemic strains (resistance to serum, production of aerobactin or colicins) but none of them demonstrated heat-stable enterotoxigenic activity. CS31A + E. coli isolates belonged to 10 serogroups, more commonly O8, O7, O17 and O21. The results obtained here confirm the worldwide distribution of CS31A antigen in bovine E. coli strains. However, CS31A + or CS31A + /F17c + E. coli were less frequently isolated than they were in North hemisphere countries.  相似文献

In order to determine the drug resistance phenotype and prevalence of drug resistance genes in Escherichia coli (E.coli) isolated from sheep in Inner Mongolia, the minimum inhibitory concentrations (MICs) of the isolates to antibiotics commonly used in veterinary clinical were detected by micro-dilution method in vitro. The results showed that the highest resistance rates of the isolates to amoxicillin,cephalothin,sulfamethoxazole and polymyxin were up to 100.0%,respectively.To amoxicillin-clavulanic, tetracycline and ciprofloxacin were 50% to 80%. These isolates were sensitive to cefotaxime, meropenem trihydrate, neomycin, and their resistance rates were all less than 10%. Among the 108 strains of E.coli from sheep, 94.4% of them were resistant to more than 7 antimicrobial agents,15.6% of them were resistant to 13 antimicrobial agents, only one strain was sensitive to all antimicrobial agents. Six kinds of resistance genes among the 108 E.coli isolates were detected by PCR method.The results showed that detection rates of 4 kinds of drug resistance genes including blaTEM, proP-2, sul-Ⅰ and ampG were all over 50%,the detection rate of resistance gene aph (3')-Ⅰ was up to 40%, only resistance gene aac(3)-Ⅱ detection rate was 5.5%. Thus, the sheep E.coli isolates in Inner Mongolia produced various degrees of resistance to 13 kinds of antibiotics, and their multi-drug resistances were very serious. They carried aph(3')-Ⅰ, sul-Ⅰ, ampG, blaTEM, proP-2 and aac(3)-Ⅱ resistance genes.  相似文献
