Smyrnium olusatrum (Apiaceae), well known as wild celery, is a biennal celery-scented plant used for many centuries as a vegetable, then abandoned after the introduction of celery. In the present work, the essential oil obtained from inflorescences and the amounts of its main constituents isofuranodiene, curzerene and germacrone were analyzed by GC as well as by HPLC because of their degradation (Cope rearrangement) occurring at high temperatures. The oil and the main constituents were assayed for cytotoxic activity on the human colon cancer cell line (HCT116) by MTT assay. Flower oil and isofuranodiene showed noteworthy activity on tumor cells with IC50 of 10.71 and 15.06 μg/ml, respectively. Analysis of the cytotoxic activity showed that wild celery oil and isofuranodiene are able to induce apoptosis in colon cancer cells in a time and concentration-dependent manner suggesting a potential role as models for the development of chemopreventive agents.  相似文献

Five ophiobolane sesterterpenes, ophiobolins P–T, and three known compounds, 6-epi-21,21-O-dihydroophiobolin G, 6-epi-ophiobolin G and 6-epi-ophiobolin K, were isolated from the acetone extract of the endolichenic fungus Ulocladium sp. by using OSMAC method. Their structures were elucidated on the basis of spectroscopic analysis. The absolute configuration of the 18,19-diol moieties in ophiobolin Q was assigned using the Frelek's method. The cytotoxic effects on KB and HepG2 cell lines, antibacterial activity against Bacillus subtilis, methicillin-resistant Staphylococcus aureus, and Bacille Calmette–Guerin were evaluated for all isolated compounds. Ophiobolin T and 6-epi-ophiobolin G exhibited the most potent cytotoxic activity against HepG2 with IC50 of 0.24 and 0.37 μM, respectively. In antibacterial assay, ophiobolins P and T showed moderate antibacterial activity against B. subtilis and meticillin-resistant S. aureus. Ophiobolin T also displayed moderate antibacterial activity against the Bacille Calmette–Guerin strain.  相似文献

The methanol extracts of Astragalus plumosus var. krugianus Chamb. & Matthews afforded sixteen cycloartane glycosides among which krugianoside A, was never reported before. All compounds were evaluated for their cytotoxic activity in human skin fibroblast WS1 cells. For compounds exhibiting no significant effect on WS1 viability, the antioxidant potential was examined. Compounds 1 and 8 prevented elevation of ROS induced by t-BOOH, suggesting the potential activity of these compounds to protect fibroblasts from oxidative stress.  相似文献

A novel xanthene (1), blumeaxanthene II, was isolated from the aerial parts of Blumea riparia (Bl.) DC., a Chinese medicinal plant with hemostatic properties, and its structure was determined by extensive spectroscopic analyses. This compound is the first example of a halogenated xanthene from a natural plant, and a biosynthetic pathway was proposed. Blumeaxanthene II was also tested against Bel-7404 cells in vitro but was found to be only weakly cytotoxic.  相似文献

Where there is limited availability of conventional fertilizers, the use of organic materials is considered a viable alternative to increase the productive capacity of soils. Many potential plant residues remain underutilized due to limited research on their use as a nutrient source. In this study, the nitrogen supplying capabilities of ten rarely-used leaf biomass sources (Acacia auriculiformis, Baphia nitida, Albizia zygia, Azadirachta indica, Senna siamea, Senna spectabilis, Tithonia diversifolia, Gliricidia sepium, Leucaena leucocephala and Zea mays) were tested based on their nutrient content, N mineralization patterns and effect on maize yield (in comparison with inorganic fertilizer). N mineralization was studied in the laboratory using an incubation experiment. Field trials were also established using a randomized complete block design. Plant residues were applied at 5 t dry matter ha?1 a week before planting maize while fertilizer was split-applied at 90 kg N ha?1 on designated plots. From the results on plant residue chemistry, most of the plant residues recorded relatively high N concentration (≥24.9 g kg?1) and low C/N ratio (≤20.1) although neither N content nor C/N ratio significantly (p > 0.05) affected their N mineralization patterns. Leaf biomass application of B. nitida, A. auriculiformis, A. zygia and maize stover resulted in an initial net N immobilization that lasted for 14 days. Application of all plant materials significantly increased the biological yield and N uptake of maize with G. sepium and T. diversifolia producing the greatest impact especially in the major rainy season. Relative to the control, total grain yield after four cropping seasons was comparable between inorganic fertilizer (9.2 t ha?1), G. sepium (8.8 t ha?1) and T. diversifolia (9.4 t ha?1) treatments. The results on maize biological yield were significantly correlated with the effects of the treatments on N uptake. The findings suggest that in locations where inorganic fertilizers are limited, leaf biomass from G. sepium and T. diversifolia could offer the most suitable option in comparison with the other species used in this study.  相似文献

A phytochemical study of the ethanolic extract of Chrysosplenium carnosum Hook. f. et Thoms. led to the isolation of two new oleanane-type triterpenoids, 6β-hydroxy-3-oxoolean-12-en-27-oic acid (1) and 3β, 21α-dihydroxyolean-12-en-27-oic acid (2), along with fourteen known compounds (316), all of which were isolated from this plant for the first time. The structures of these compounds were established on the basis of spectroscopic methods. Compounds 14 were evaluated for their in vitro anti-tumor activities on B16F10, SP2/0 and Hep-G2 cells lines. Compounds 1, 3 and 4 exhibited strong inhibitory activity against B16F10 and SP2/0 cells' growth, compared with moderate cytotoxic activity against Hep-G2 cells. However, compound 2 showed to be inactive against these tumor cells.  相似文献

The aim of this study was to investigate the cytotoxic and anticancer activities of stylosin, a monoterpene extracted from an edible plant, Ferula ovina, on 5637 and HFF3 cells using MTT and comet assays and DAPI staining.To assess stylosin effects, cells were cultured in the presence of various concentrations of stylosin during three days; the IC50 of stylosin on cancerous 5637 cells was less than its value on HFF3 normal cells, indicating that it might have anticancer properties. Investigating the mechanism of stylosin action revealed that it quickly induced DNA lesions and increased the number of apoptotic cells.  相似文献

Four paclitaxel derivatives were afforded by preparative HPLC separation of two pairs of diastereoisomers, which were obtained by catalytic hydrogenation and epoxidation of the C-13 side-chain double bond of cephalomannine, a naturally occurring paclitaxel analog. The four paclitaxel derivatives were analyzed using NMR, CD spectroscopy, and side-chain hydrolysis in order to measure their optical rotations and GC characteristics. In this way, the stereoconfigurations of the products were determined. Evaluation of the compounds' activity indicated that they had differing cytotoxic activities: compound 5 had superior activity in BCG-823 tumor cells compared to paclitaxel, while compound 7 had superior activity in HCT-8 and A549 tumor cells compared to paclitaxel. These results indicate that the stereoconfiguration of the paclitaxel N-acyl side chain has a significant impact on its activity.  相似文献

Plant defensin is a small, cationic, cysteine-rich broad-spectrum antimicrobial peptide with four or five disulfide bridges and has been shown to be a component of the innate immunity system in plants. In the present study, the defensin gene (TvD1) from Tephrosia villosa was overexpressed in tobacco and characterized. Two high-expression (T1, T26) and one low-expression (T13) plant lines were selected through semi-quantitative RT-PCR and used for bioassays along with non-transgenic controls. The high-expression plant line exhibited strong in vivo anti-fungal and anti-feedant activity against the pathogen Rhizoctonia solani and the first and second instar larvae of the Spodoptera litura (F.), the tobacco cutworm, respectively. The low-expression plant line showed a moderate level of tolerance/resistance in the bioassays. The recombinant peptide (rTvD1) exhibited toxicity to tobacco pollen grains in the germination assay, but transgenic plants produced copious fertile pollen and set capsules with viable seeds. The results of this study demonstrate that the single gene (TvD1) effectively controls both fungal and insect pests and, hence, it can be used for crop transformation.  相似文献

Five new amino γ-butenolides, fritenolide A (1), B (2), C (3), D (4), and E (5), along with four known compounds, were isolated from the bulbs of Fritillaria unibracteata. Their structures were determined by spectroscopic analysis, including 1D NMR, 2D NMR, HRESIMS, HRESIMS/MS, IR, and CD techniques. All isolates were evaluated for the protective activity on injured hepatocytes and cytotoxic activity on human cancer cells in vitro. The effects of understory plant litter on dominant tree litter decomposition are not well documented especially in semi-arid forests. In this study, we used a microcosm experiment to examine the effects of two understory species (Artemisia scoparia and Setaria viridis) litter on the mass loss and N release of Mongolian pine (Pinus sylvestris var. mongolica) litter in Keerqin Sandy Lands, northeast China, and identified the influencing mechanism from the chemical quality of decomposing litter. Four litter combinations were set up: one monoculture of Mongolian pine and three mixtures of Mongolian pine and one or two understory species in equal mass proportions of each species. Total C, total N, lignin, cellulose and polyphenol concentrations, and mass loss of pine litter were analyzed at days 84 and 182 of incubation. The chemistry of pine litter not only changed with the stages of decomposition, but was also strongly influenced by the presence of understory species during decomposition. Both understory species promoted mass loss of pine litter at 84 days, while only the simultaneous presence of two understory species promoted mass loss of pine litter at 182 days. Mass loss of pine litter was negatively correlated with initial ratios of C/N, lignin/N and polyphenol/N of litter combinations during the entire incubation period; at 182 days it was negatively correlated with polyphenol concentration and ratios of C/N and polyphenol/N of litter combinations at 84 days of incubation. Nitrogen release of pine litter was promoted in the presence of understory species. Nitrogen release at 84 days was negatively correlated with initial N concentration; at 182 days it was negatively correlated with initial polyphenol concentration of litter combinations and positively correlated with lignin concentration of litter combinations at 84 days of incubation. Our results suggest that the presence of understory species causes substantial changes in chemical components of pine litter that can exert strong influences on subsequent decomposition of pine litter.  相似文献

We studied the effect of dense populations of red and fallow deer on a burned Mediterranean plant community in Portugal. We sampled a set of 12 open and 12 fenced plots 2, 3, 4, and 8 years after fire occurrence. Each plot was sub-sampled using a quadrat grid to estimate the relative plant frequency (proportion of quadrats with plants). The effect of deer on temporal trends of overall plant frequency and diversity, and on the abundance of the four most common woody species, was modeled using Generalized Linear Mixed Models. The effect of deer on plant assemblage composition was assessed by use of Redundancy Analysis. In open plots the relative plant frequency increased along the study period from 0.59 to 0.85, whereas in fenced plots there was a significantly faster recovery from 0.35 to 0.96. Cistus salvifolius and Rubus ulmifolius were significantly favored by fencing but a similar effect was not observed for Erica scoparia and Ulex jussiaei. The Redundancy Analysis showed a divergent floristic composition of the two sets of plots. The effect of deer was significant for the Evenness index but not for the Shannon and the Richness indices. Our results show that a high deer density may significantly delay the post-fire recovery of a Mediterranean community of woody plants and cause shifts in the direction of plant succession.  相似文献

The process of solid-state fermentation was used to produce a cocktail enzyme of Trichoderma reesei ATCC 66587 and Aspergillus tubingensis KRCF 700-33. Wheat bran, corncob, and sugi pulp were supplemented with ammonium sulfate as an enzyme-producing medium using T. reesei and A. tubingensis. The corncob blend ratio, duration of incubation, and ammonium sulfate concentration were optimized for enhancing cellulase production from T. reesei using a Box-Behnken design. Filter paper degrading activity more than tripled when T. reesei was grown in the optimized medium, as compared with the initial medium. The highest activity of 4.03 FPU/ml (about 29 FPU/g of dry material) was obtained with a cocktail enzyme having a 25 % content of A. tubingensis and 75 % of T. reesei. The sugi pulp was then fermented to ethanol with the cocktail enzyme and thermotolerant yeast (Saccharomyces cerevisiae BA-11) under simultaneous saccharification and fermentation (SSF) at 40 °C. An ethanol concentration of 4.48 % (w/v) was achieved using the cocktail enzyme (4 FPU/g-pulp) that was produced on-site with a substrate loading level of 12.5 wt %, which achieved an ethanol yield of 76 % after 72 h.  相似文献

Acne vulgaris is the most common skin disease in the world, and the number of antibiotics resistant to acne-inducing bacterial strains has been increasing in the past years. Natural substances from plants are promising candidates to treat this disease. The methanol and 50 % (v/v) ethanol extracts of 29 plant species traditionally used in Sudan for treatment of a variety of diseases were tested in vitro for their potential anti-acne activity. The activities of these extracts were determined using an antibacterial assay against Propionibacterium acnes, a lipase inhibitory assay, and l,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity assay. The results showed that methanol and 50 % ethanol extracts of Terminalia laxiflora Engl & Diels wood exhibited good antibacterial activity (minimum inhibitory concentration 0.13 mg/ml). The 50 % ethanol extracts of Abrus precatorius L. seed, T. laxiflora Engl & Diels and methanol extract of Acacia nilotica (L.) pods showed lipase inhibitory activity more than 70 % at 500 μg/ml. The methanol extracts of A. nilotica (L.) pods showed the best DPPH radical scavenging activity (IC50 1.32 μg/ml). Total phenolic, flavonoid and total tannin contents of selected plant extracts shown anti-acne activities were investigated. Almost all selected extracts contained phenolic compound. The highest level of flavonoids (38.87 μg/mg) was detected in T. brownii bark, whereas the highest amount of tannin was detected in A. nilotica (L.) bark (88.01 %).  相似文献

The aqueous extract of Tilia cordata flowers showed in vitro stimulatory effects on lymphocyte proliferation (maximum stimulatory concentration: 20 μg/ml). This effect was mimicked by Ro 5-4864, an agonist of the peripheral benzodiazepine receptor, and by Pk 11195, an agonist/antagonist of the same receptor. These results suggest that Tilia extract could exert its stimulatory effect acting as an agonist of the peripheral benzodiazepine receptor.  相似文献

Stachys alopecuros subsp. divulsa (Lamiaceae), a perennial herb endemic to central Italy growing on mountain pastures, was investigated for the first time for the content of secondary metabolites, for the micromorphology and histochemistry of glandular trichomes, and for the biological activity of the volatile oil, namely cytotoxic, antioxidant and antimicrobial. The plant showed the molecular pattern of iridoids, among which a new iridoid diglycoside (4′-O-β-d-galactopyranosyl-teuhircoside) was detected, together with a sterol glucoside and a phenylethanoid glycoside. The essential oil from the flowering aerial parts was characterized by a high proportion of sesquiterpene hydrocarbons (65.1%), with (E)-caryophyllene (33.2%) as the most abundant, while other main components were germacrene D (7.6%), α-humulene (6.4%) and the oxygenated cis-sesquisabinene hydrate (10.2%). Taken together, polar and apolar chemical profiles support the classification of the species within the section Betonica of the genus Stachys. Micromorphological study revealed three types of glandular hairs secreting different classes of compounds, with type A peltate hairs producing the bulk of the essential oil. MTT assay revealed the potential of the volatile oil in inhibiting A375, HCT116 and MDA-MB 231 tumor cells (IC50 values below 20 μg/ml).  相似文献

Various plants are well known for their insecticidal activity and their use was maintained for millennia throughout all the agricultural regions of the world. In a current context, the use of Botanical insecticides represents one of the best alternatives to chemicals for the development of environmental-friendly strategies for stored grain pest control. Datura alba Nees is a plant found extensively in the warmer regions of the world and it is used as a medicinal plant. This study aims to assess the contact toxicity and the trans-generational effect of D.?alba leaf extract (DLE) against two important insect pests on stored rice, Trogoderma granarium and Sitophilus oryzae, under laboratory conditions. Filter papers were soaked in three DLE concentration solutions and in two control treatments: water and acetone. The survivor specimens (F0) were transferred to a new untreated feeding substrate and the population build up of the two following generations (F1 and F2) were counted after 30 and 60?days, respectively. The highest DLE concentration (2.5?%) induced the significantly highest mortality with 33.5 and 45?% mortality in T.?granarium and S.?oryzae after 7?days of exposure, respectively. The DLE long-term effect toward both tested species was also proved by the high demographic decrease in the F2 generations, when compared to control groups. This study is the first step toward establishing a scientific basis for the effective application of D.?alba plant materials as biorational tools to control stored grain pests.  相似文献

Tribolium castaneum is a pest of stored foods that causes serious economic losses. Understanding insect nutritional requirements and the effects of different diets on insect development can provide tools for developing strategies to control insect and integrated pest management systems. In this work we studied the performance of the T. castaneum on different plant diets composed of Vigna unguiculata, Phaseolus vulgaris and wheat flours. The diets interfered differentially with development and insect survival. Larvae size was reduced about 60 % in larvae fed with V. unguiculata flour. A delay in pupae formation was also observed. The larval mortality of 100 % was observed in the P. vulgaris diet. Carbohydrate and triglyceride showed variations during development and with different diets. Larvae fed with wheat diets showed a high concentration of carbohydrates 21 days after hatching (DAH) and triglyceride at 15 and 21 DAH. Larvae fed with wheat diets showed α-amylase activity during development. At 15 DAH the activity of larvae fed with V. unguiculata increased about 50 %. Cysteine protease activity in larvae fed with wheat remained high throughout development. Serine protease activity also varied according to diets. These results show that the nutritional state of developing larvae varies greatly with respect to triglycerides, carbohydrate and digestive enzymes depending on the diet and larval stage. Altogether, these results show that changes in dietary nutrients can interfere with insect physiology. Therefore changes in diet composition that may deprive insects of essential nutrients or include toxic compounds might be a good control strategy for stored product pests.  相似文献

Two new abietane diterpenoids, ramentoxide (1) and ramentoxidone (2) and a new icetexane diterpenoid, amentonone (3) were isolated from the barks of Amentotaxus formosana. The structures of 1-3 were determined by spectroscopic methods. Known compounds brevitaxin (4), and (+)-ferruginol (5) and ent-kaur-16-en-15-one (6) isolated from this plant revealed potent cytotoxic activity against human breast adenocarcinoma cells, MCF-7 cells with an IC50 value of 0.08 ± 0.05 μg/mL, and significant anti-inflammatory activities, respectively.  相似文献
