The possible interaction of Cryptosporidium molnari and bacteria in gilthead sea bream (Sparus aurata) was studied. Epidemiological data from a pathological survey under farm conditions were analyzed. In addition, parasite and bacteria burdens were studied in experimental models in which naturally and experimentally parasitized fish were challenged with a particular strain of Vibrio harveyi (H57). All the bacteria species present were studied. Under farm conditions, the parasite was more prevalent when mortality or morbidity cases (study C) occurred than in randomly sampled fish (study B). In study C, parasite abundance was significantly higher in bacteria-negative fish, and total bacteria abundance was significantly higher within non-parasitized fish. V. harveyi and V. splendidus were the most prevalent among bacteria carriers in studies B and C, respectively. In study C, among bacteria carriers, most isolates were slightly more prevalent in parasitized than in non-parasitized fish. Two groups (G1, G2) of naturally parasitized fish were inoculated with H57 by intracoelomic injection (ICI) and by oral intubation (OI). H57 was recovered only in G1 inoculated fish, which had a significantly higher basal abundance of total bacteria, and where the only ones with mortalities. In G1, the mortality rate and the prevalence of other V. harveyi strains different from the H57 molecular type were higher in ICI than in OI fish, and the total bacteria abundance was also significantly higher in ICI fish. C. molnari abundance was significantly higher in G1 than in G2, and also in OI than in ICI fish within G1. When H57 was IC inoculated to fish (G3, from the same farm as G2) experimentally infected with C. molnari, H57 was not recovered from any fish. A low mortality was recorded, and only in those fish inoculated with both pathogens. Also in these fish, the prevalence of infection of C. molnari was higher and histopathological damage to the stomach was greater than in fish inoculated only with the parasite. Therefore, the impact of the parasite would be reduced notably when the bacterial burden or the intensity of parasite infection are low (G2, G3).  相似文献

The recent identification of multiple genotypes of the salmonid parasite Ceratonova shasta with different virulence levels in Chinook Salmon Oncorhynchus tshawytscha suggests that it is possible to immunize fish against subsequent infection and disease. We hypothesized that exposure of Chinook Salmon to the less-virulent parasite genotype (II) prior to the more virulent parasite genotype (I) would decrease disease and/or result in fewer mature parasites compared with fish only infected with the more virulent genotype. To test this hypothesis, fish were challenged in a combination of field and laboratory exposures, and we measured infection prevalence, percent morbidity, and mature parasite production. Neither mortality nor mature parasite production were reduced when fish were exposed to genotype II prior to genotype I compared with fish exposed only to genotype I, suggesting that protection against C. shasta using a less-virulent genotype of the parasite does not occur.

Although rainbow trout fry mortality syndrome caused by the bacterium Flavobacterium psychrophilum is widespread in fish farms it is difficult to reproduce infection of rainbow trout in the laboratory using immersion exposure with bacterial suspensions. It has therefore been speculated that ectoparasites could act as enhancers of bacterial infections under natural conditions. In the present study rainbow trout fry were exposed to infections with F. psychrophilum (immersion for 30 min or 10 h) alone, exposed to the ectoparasitic monogenean Gyrodactylus derjavini alone or exposed to both pathogens in combination. Infection levels and host mortality were subsequently monitored to elucidate if the ectoparasitic monogeneans could enhance infection of fish with the bacterium. Immersion of fish in bacterial suspensions alone did not result in infection. Only one fish became infected with the bacterium and this fish belonged to the combination exposure group. The parasite populations increased differently in the various groups and it was found that host mortality was correlated to gyrodactylid infection levels (r=0.94) but not to bacterial exposure. The results emphasise the pathogenicity of the parasite G. derjavini, the relative resistance of intact fish to direct exposure to F. psychrophilum but provide only a weak indication of a possible enhancement of bacterial invasion due to ectoparasitic infections. It cannot be excluded that higher parasite burdens and/or prolonged immersion (more than 10 h) in bacterial suspensions may result in bacterial invasion.  相似文献

Lamproglena clariae attaches to gill filaments of Clarias gariepinus, penetrates the gill tissue and consumes blood. Hence, the purpose of this study was to investigate the mechanical damage caused by this parasite on gill filaments of the host. Infestation intensity and haematocrit values of fish blood were determined and correlated. Fish were killed, gills dissected out and observed with a dissection microscope. Gill filaments with adult females in situ were fixed in a solution of alcohol, formaldehyde and acetic acid and preserved in 70% ethanol. Fixed specimens were studied by histological examination following staining with Heidenhein’s azan solution. Gross morphology revealed varying degrees of swelling of gill filaments near the attached specimens of L. clariae. Epithelial hyperplasia, resulting in fusion of gill lamellae and necrosis of the host tissue in the vicinity of the parasite were revealed through histological examination. No correlation was evident between the infestation intensity and fish haematocrit values. Although results showed that L. clariae causes localized infestation, high intensity of infestation may be detrimental to the fish. Hyperplasia of epithelial tissue resulting in fusion of lamellae would result in restriction of air passages and thus hinder the process of respiration of the host.  相似文献

Dietary fish oil has a beneficial effect on heart and some bacterial diseases and apart from other effects, some studies have revealed their ability to modulate the course of inflammatory and autoimmune diseases. The study here reported was designed to evaluate the possible influence of a fish oil supplement on the course of a Trichinella infection. Nutritional, parasitological and immunological parameters were analyzed. Two groups of 20 Wistar rats, one fed a standard diet and the other one a standard diet supplemented with fish oil, were infected with 1000 L1 larvae. Other two uninfected groups served as control. Results were as follows: fish oil diet intake and infection have, respectively, a positive and a negative effect on growth and food utilization. The negative effect is detected later in animals fed the fish oil diet. A reduction of 30.9 and 36.6% in the number of adult worms and L1 larvae, respectively, was observed in the fish oil group as compared to the standard diet group. Production of IFNgamma (Th1 response) and IL4 (Th2) response was measured in stimulated splenic cells. The fish oil diet increased both IFNgamma and IL4 levels. At 6 days after infection both IFNgamma and IL4 responses were detected, but at 36 days after infection only IL4 was detected in the standard group. The level of somatic and cuticular antibodies was not affected by the diet.  相似文献

An immuno-blot assay was used to investigate the serum antibody response in flounder injected with formalin-killed flagellates (immunized) and those injected with saline (control) and challenged with live T. bullocki after 21 days. Fish were held at 20 degrees C and at ambient temperature from October through June. At 20 degrees C immunized fish had significantly higher antibody titers than control fish, but immunized fish were not protected from infection with T. bullocki. At ambient temperature, after initial flagellate growth phase, antibody titer varied directly with temperature (2-25 degrees C) and T. bullocki intensity varied inversely with titer. Flagellates were eliminated from the peripheral circulation in both immunized and control fish when antibody titer peaked in May. Recovered fish were immune to homologous challenge for at least one year.  相似文献

Brown-marbled grouper Epinephelus fuscoguttatus which had been fed diets containing sodium alginate and kappa (kappa)-carrageenan at 5, 10 and 20gkg(-1), respectively after 0, 2, 4, 6, 8 and 14 weeks were examined for survival, growth, innate cellular and humoral responses as compared to the fish that fed control non-supplemented diet. Survival was 100% for the fish that fed all diets after 14 weeks and no significant difference in growth was observed among seven diets. The fish that fed a diet containing sodium alginate at 10gkg(-1), and fed a diet containing kappa-carrageenan at 5gkg(-1) over 2-8 weeks showed significantly increased leucocyte count, respiratory burst, phagocytic activity and phagocytic index, The fish that fed a diet containing sodium alginate at 5gkg(-1), and fed a diet containing kappa-carrageenan at 5gkg(-1) over 2-8 weeks showed significantly increased alternative complement activity (ACH50) and lysozyme activity. In another experiment, E. fuscoguttatus, which had been fed control diet, and all diets containing sodium alginate, and kappa-carrageenan after 14 weeks were challenged with Vibrio alginolyticus at 5.0x10(9) colony-forming units (cfu) fish(-1) and then placed in seawater of 34 per thousand. The survival of fish that fed a diet containing sodium alginate at 10gkg(-1) and kappa-carrageenan at 5gkg(-1) was significantly higher than that of fish which fed the control diet over 96-168h. It was concluded that E. fuscoguttatus which fed a diet containing sodium alginate at 10gkg(-1) or less, or fed a diet containing kappa-carrageenan at 5gkg(-1) enhance the innate immunity, and increase the resistance from V. alginolyticus infection.  相似文献

Olive flounder, Paralichythys olivaceus fed with pomegranate enriched diet and challenged with or without Philasterides dicentrarchi had a significantly higher white blood cell (WBC) count on weeks 2 and 4 than the infected group fed with non enriched diet (standard diet). Similarly the red blood cell (RBC) counts did not significantly change in control and treated fish on weeks 1 and 2. It was significantly increased in treated fish on week 2 when compared to the control. In both the groups the hemoglobin (Hb) and hematocrit (Ht) levels significantly increased on weeks 2 and 4. The mean corpuscular volume (MCV) did not significantly change at any time in both groups whereas mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) increased significantly on week 4 in the treated group. The leukocytes such as lymphocytes (Lym), monocytes (Mon), neutrophils (Neu), and biochemical parameters such as total protein (TP), glucose (GLU), and calcium (CAL) levels significantly increased in treated groups on week 2 or 4 as compared to the control. The scuticocidal activity and respiratory burst activity were significantly enhanced in treated groups with or without parasite on weeks 2-4. However, the serum lysozyme activity was significantly enhanced from weeks 1 to 4. The protective response in terms of cumulative mortality was low in groups fed with enriched diet against parasite when compared to control. Therefore, we suggest that pomegranate enriched diet following challenge with P. dicentrarchi restores the altered hematological and biochemical parameters, and improves the innate immune system in olive flounder against P. dicentrarchi.  相似文献

In serum most of the iron molecules are bound to transferrin (Tf), which is a highly polymorphic protein in fish. Tf is an essential growth factor for mammalian trypanosomes. We performed a series of experiments with Trypanoplasma borreli to detect putative correlations between different Tf genotypes of common carp (Cyprinus carpio L.) and susceptibility to this blood parasite. Five genetically different, commercially exploited carp lines (Israelian 'D', Polish 'R2' and 'K', Ukrainian 'Ur', Hungarian 'R0') and a reference laboratory cross ('R3xR8') were challenged with T. borreli and parasitaemia measured to determine susceptibility to the parasite. Among the commercial carp lines, Israelian 'D' carp were identified as most and Polish 'R2' carp as least susceptible, and used to produce a next generation and reciprocal crosses. These progenies were challenged with T. borreli and parasitaemia measured. We demonstrated significant effects of genetic background of the carp lines on susceptibility to T. borreli. This genetic effect was preserved in a next generation. We also observed a significant male effect on susceptibility to T. borreli in the reciprocal crosses. Serum samples from a representative number of fish from two infection experiments were used for Tf genotyping by polyacrylamide gel electrophoresis (PAGE), identifying DD, DG and DF as most frequent Tf genotypes. We could detect a significant association of the homozygous DD genotype with low parasitaemia in the least susceptible 'R2' (and 'K') carp lines and the lack of a such an association in the most susceptible 'D' carp line. Upon examination of parasite growth in vitro in culture media supplemented with 3% serum taken from fish with different Tf genotypes, we could show a faster decrease in number of parasites in culture media with serum from DD-typed animals.  相似文献

Discovery of fish exhibiting clinical signs of ceratomyxosis in Washington State prompted concern over the potential impact of the myxozoan parasite Ceratomyxa shasta on native stocks of steelhead Oncorhynchus mykiss (anadromous rainbow trout). To investigate these concerns, a survey of 16 freshwater systems within the Puget Sound watershed, including Lake Washington, was conducted by sentinel exposure of susceptible fish (cutthroat trout O. clarkii and rainbow trout). Fish were exposed for 7 d during September 2003 and May 2004 and then were returned to a holding facility for monitoring of disease signs. Mortality caused by the parasite occurred only in the exposure group held at the University of Washington Hatchery, which receives its water from Portage Bay of Lake Washington. Fish from all other sites were negative for C. shasta, both visually and by polymerase chain reaction (PCR) assay, except for a single fish held at the Tumwater Falls Hatchery in September 2003. A single deformed spore was detected in that fish, but infection could not be confirmed by PCR and the parasite was not detected from any other fish held at that site during either the September or the May exposure. From these results, we conclude that C. shasta is not likely to have contributed significantly to the decline of steelhead populations throughout Puget Sound.  相似文献

The protistan parasite Ichthyophonus sp. occurs in coastal populations of Pacific Herring Clupea pallasii throughout the northeast Pacific region, but the route(s) by which these planktivorous fish become infected is unknown. Several methods for establishing Ichthyophonus infections in laboratory challenges were examined. Infections were most effectively established after intraperitoneal (IP) injections with suspended parasite isolates from culture or after repeated feedings with infected fish tissues. Among groups that were offered the infected tissues, infection prevalence was greater after multiple feedings (65%) than after a single feeding (5%). Additionally, among groups that were exposed to parasite suspensions prepared from culture isolates, infection prevalence was greater after exposure by IP injection (74%) than after exposure via gastric intubation (12%); the flushing of parasite suspensions over the gills did not lead to infections in any of the experimental fish. Although the consumption of infected fish tissues is unlikely to be the primary route of Ichthyophonus sp. transmission in wild populations of Pacific Herring, this route may contribute to abnormally high infection prevalence in areas where juveniles have access to infected offal.

Abstract

Experimental feeding trials were performed to evaluate the efficacy of amoxicillin (AMX) in controlling Streptococcus iniae infection in blue tilapia Oreochromis aureus. The doses of AMX active ingredient tested were 0, 5, 10, 30, and 80 mg/kg of fish body weight (BW) per day. Administration of medicated feed started within 22–24 h postchallenge by waterborne exposure to S. iniae (after skin scraping) and continued for 12 consecutive days, followed by a 17-d posttreatment observation. Oral administration of AMX-medicated feed for 12 d at a daily rate of 10, 30, and 80 mg/kg BW significantly increased the survival of S. iniae?infected tilapia from 3.8% in the challenged, nonmedicated positive control to 45, 75, and 93.8%, respectively. The survival rate was significantly higher in the 80-mg treatment (93.75%) than the 10-mg treatment (45%) but did not differ significantly between the 10-mg (45%) and 30-mg (75%) treatments. At the conclusion of the experiment, no carriers were detected in any challenged group receiving AMX-medicated diet, whereas the bacterium was recovered from the nonmedicated, challenged survivors of the infection.  相似文献

Abstract

Evaluation of the susceptibility of bull trout Salvelinus confluentus fry to Myxobolus cerebralis infection using two different laboratory challenge models demonstrated that the method of administering the exposure dose affected infection prevalence. Administration of a low parasite dose (500 per fish) in a single exposure did not establish infection, but when the same cumulative dose was administered over 21 d the prevalence of infection was 45%. The results of challenges at a high exposure dose (5,000 per fish) were similar, infections being detected in 24% of the fish receiving a single dose and 40% of those administered the same dose in multiple exposures. Clinical disease was not detected in fish exposed via either challenge method. The susceptibility of yearling bull trout was tested by means of a single high dose of 10,000 per fish, and infection was detected in only 5% of those fish.  相似文献

Thelohanellus nikolskii infection of the common carp (Cyprinus carpio L.) has been a common parasitosis in the Central European fish farms since the first detection of the parasite about 20 years ago. This parasite, introduced from the Far East, causes intensive infection on the fins of fingerlings of the carp subspecies cultured in Europe (European carp, Cyprinus carpio carpio). This infection of the common carp occurs in the Hungarian fish farms every year. Until the present study, this parasite had not been recorded from the fins of koi or coloured carp (Cyprinus carpio haematopterus), a carp of Far Eastern origin, which is cultured in Hungary as an ornamental fish. A recent survey conducted in common carp, koi and goldfish stocks demonstrated that T. nikolskii infection of low prevalence and intensity occurs also in koi populations, but its prevalence and intensity are markedly lower than in common carp kept in the same ponds. It is suggested that the observed differences are due to disparities in the susceptibility of the two carp subspecies to T. nikolskii, and that the koi is less susceptible to this infection. Other signs of susceptibility can also be observed in the European subspecies, since in 15% of the fish plasmodium development was arrested at an early stage. Thelohanellus nikolskii infection could not be demonstrated on goldfish (Carassius auratus).  相似文献

In order to improve feeding and growth of juvenile great sturgeon Huso huso, the effects of various lipid sources in feed were examined. Three hundred juveniles (initial body weight 215 ± 10 g) were distributed into 15 flow-through 1100 l tanks (20 fish per tank). Five experimental diets containing various lipid levels were formulated: diet C (control; without added oil), SO(L) and SO(H) diets containing soybean oil concentrations respectively, (53.5 vs. 174 g/kg), FO(L) and FO(H) diets containing fish oil with low and high concentrations respectively (53.5 vs. 174 g/kg). Fish were fed three times daily based on their appetite for a period of 6 weeks. At the termination of the experiment, growth rate was improved with increasing oil content in the diets. There were significant differences in final weights among fish fed the different experimental diets. Lipid content of muscle and liver significantly increased with increasing dietary lipid levels for both soybean and fish oil. The lowest cholesterol level and the highest haematocrit value were measured in fish fed the FO(H) diet. No significant difference was found among dietary treatments in terms of triacylglycerol concentration, number of red blood cell, mean corpuscular volume, mean corpuscular haemoglobin (MCH) and MCH concentration (MCHC). According to the most growth indices, results of this study suggest that the best results were observed in FO(H) which can be attributed to the greater palatability of diet containing high fish oil, supply of fatty acid requirement and enhanced physiological performance.  相似文献

In this study, effect of ellagic acid on some haematological, immunological and antioxidant parameters in the blood and various tissues of rainbow trout (Oncorhynchus mykiss) were examined. Four groups of rainbow trout were fed experimental diets containing either no ellagic acid (control) or supplemented with ellagic acid at 50 mg/kg diet (EA‐50), 100 mg/kg diet (EA‐100) or 150 mg/kg diet (EA‐150) for 21 days. Samples of the blood and tissue (liver, kidney and spleen) were collected at the end of the experiment and analysed for their haematological profile (the red blood cell count, the haemoglobin concentration and the haematocrit level), immune response (the white blood cell count, the oxidative radical production (NBT activity), the total plasma protein and total immunoglobulin level) and oxidant/antioxidant status (the malondialdehyde level, the superoxide dismutase, catalase and glutathione peroxidase activity as well as the reduced glutathione concentration). The findings of this study demonstrated that ellagic acid had a positive effect on the haematological parameters, the immune response and the antioxidant enzyme activities of the fish.  相似文献

1. This study was conducted to determine the utility of a volatile oil (VO) mixture in quail diets as a natural growth promoter. Different levels of VO mixtures, derived from rosemary and oregano, were added to a basal diet to determine the effects of the mixture on live weight (LW), live weight gain (LWG), feed intake, feed conversion ratio (FCR), carcase yield (CY), lipid oxidation level in thigh meat samples, and blood constituents.

2. A total of 880 one-day-old Pharaoh (Coturnix coturnix Pharaoh) quails, including both males and females, were divided into 4 groups containing 220 quails and treated as follows: (1) a control treatment with 0?mg VO/kg of diet; (2) 100?mg/kg rosemary VO plus 100?mg/kg oregano VO (50:50%); (3) 140?mg/kg oregano VO plus 60?mg/kg rosemary VO (70:30%); and (4) 60?mg/kg oregano VO plus 140?mg/kg rosemary VO (30:70%). The diets were prepared fresh for each treatment. The experiment was carried out for 42?d.

3. The results of the study showed that the highest concentration of rosemary oil (140?mg/kg) caused a significant increase in live weight, live weight gain and carcase yields during the growing and finishing periods. However, feed intake and FCR were not significantly influenced by treatments.

4. The quails fed with rations containing the VO mixture derived from rosemary and oregano had reduced thiobarbituric acid levels (TBA) in raw breast meat samples at different storage times. There is possibly a synergistic effect between oregano and rosemary volatile oils in preventing lipid oxidation in stored meat.

5. In this study, the haemoglobin (PCV) and haematocrit values and the heterophile/lympohocyte (H/L) ratio increased in the blood samples taken from Treatment 2.

6. In conclusion, a volatile oil containing a mixture of rosemary and oregano oils could be a potential natural growth promoter for quails, depending on the plants from which the VOs were extracted, the dosage and the synergetic effects of the mixture.  相似文献

Infections by the digenetic trematode, Ribeiroia ondatrae, cause severe limb malformations in many North American amphibians. Ribeiroia ondatrae also infects fishes as second intermediate hosts, but less is known about the pathology and immune responses initiated in infected fish, even though reports of infected fish date back to early 1900s. To this end, we experimentally exposed juvenile Bluegills Lepomis macrochirus to three doses of R. ondatrae cercariae and monitored the pathology, parasite infection success, and humoral responses over 648 h. All exposed fish became infected with metacercariae, and the average infection load increased with exposure dose. Histologically, infection was associated with acute hemorrhages in the lateral line and local dermis at 36 h, followed by progressive granulomatous inflammation that led to the destruction of encysted metacercariae. Correspondingly, over the course of 648 h we observed an 85% decline in average infection load among hosts, reflecting the host's clearance of the parasite. Infection was not associated with changes in fish growth or survival, but did correlate with leukocytosis and neutrophilia in circulating host blood. Understanding the physiological responses of R. ondatrae in Bluegill will help to clarify the ecological effects of this parasite and provide a foundation for subsequent comparisons into its effects on behavior, individual health, and population dynamics of Bluegill.

Clarias gariepinus were collected from Lake Chivero, Zimbabwe, and examined for nematode parasites from November 2000 to May 2002. Of the 202 specimens collected, 42.6% were infected with third-stage larvae of Contracaecum sp. in the body cavity. The intensity of the infection was 1-7 worms per fish (mean intensity = 2.2). Seasonal variation in the prevalence of the parasite was not obvious and there was no significant difference in the prevalence of infection between males and females (chi 2 = 2.228; P > 0.05). No significant relationship between host size and prevalence was established. There was also no significant relationship between intensity and the body condition factor (r = 0.11; P > 0.05). The low parasite prevalence may have been caused by the disruption of the infection cycle since piscivorous birds, which are the final hosts of the parasite, do not feed on C. garieplnus in Lake Chivero.  相似文献

Gyrodactylids (Monogenea) are ectoparasites of fish, some of which negatively affect commercially valuable fishes. Temperature strongly regulates population dynamics of these viviparous flatworms in farmed and wild fish populations, with most gyrodactylid species showing positive temperature-abundance associations. In agreement with epidemiological theory, numerous laboratory studies demonstrate that these parasites cannot persist in confined fish populations without periodic introduction of susceptible hosts. Extinction of gyrodactylid populations is due to host immunity, which develops in several fish species. In this one-year study, we followed populations of the recognized pathogen Gyrodactylus cichlidarum infecting four genetic groups of confined tilapia (wild type Nile tilapia Oreochromis niloticus niloticus, red O. n. niloticus, Mozambique tilapia O. mossambicus and a red synthetic population called Pargo-UNAM) kept under farming conditions and subject to natural environmental fluctuations. Based on the antecedents given, we postulated the following three hypotheses: (1) parasite abundance will be regulated by water temperature; (2) parasites will induce host mortality, particularly during periods of rapid infrapopulation growth; and (3) gyrodactylid populations will eventually become extinct on confined fish hosts. We disproved the three hypotheses: (1) parasite numbers fluctuated independently of temperature but were associated to changes in microhabitat use; (2) although gyrodactylid populations exhibited considerable growth, no evidence was found of negative effects on the hosts; and (3) infections persisted for one year on confined fish. Microhabitat use changed over time, with most worms apparently migrating anteriorly from the caudal fin and ending on the pectoral fins. Gyrodactylid populations followed similar trajectories in all fish, aggregating and dispersing repeatedly. Several instances were found where increased parasite dispersion coincided with increased intensity of infection; as well as the opposite, where increased aggregation coincided with parasite population declines. Three alternative explanations could account for these observations: that parasites (1) experience differential mortality on different anatomical regions of the fish; (2) migrate to avoid intraspecific competition; and (3) migrate to escape localized immune responses induced by infection. Our data do not allow us to demonstrate which of these alternatives is correct, so we discuss the merits of each. We provide circumstantial evidence in support of the third explanation, because as shown in other fish host-gyrodactylid interactions where immune responses have been characterized, in this study worms progressively moved away from fins with high mucus cell density to those with low density - what would be anticipated if immune defenses occur and reach the fish surface through mucus.  相似文献
