In March 1991, large chlorotic patches appeared in an autumn-sown barley crop growing near Thessaloniki, Greece. Leaves had characteristic mosaic symptoms and immunosorbent electron microscopy and enzyme-linked immunosorbent assay confirmed the presence of both soil-borne mosaic viruses of barley, barley mild mosaic and barley yellow mosaic bymoviruses. In the following year, similar symptoms appeared in a crop at Souroti, 30 km east of Thessaloniki but the disease has not been found in other areas of Macedonia. This report is the first record of these viruses from Greece and is the most southerly European record.  相似文献

About 90 barley cultivars mostly of European or Japanese origin, were grown for 2–5 years at eight sites in China where barley yellow mosaic virus was known to occur. The sites were selected because they had previously been used to screen breeding lines and some differences between them in cultivar response had been suspected. ELISA tests showed that symptomless plants were not infected by the virus and the proportions of plants with symptoms were therefore recorded as a measure of susceptibility. European cultivars carrying the ym4 gene, which confers resistance to the common European strain, were usually resistant at two sites but susceptible at the others, but one (cv. Energy) was resistant at all sites. Eleven of the Japanese cultivars showed differential responses between sites but there was no correspondence with strains recognized in Japan. There are probably several distinct Chinese strains but further experiments would be needed to identify them. The Japanese cultivars Chosen, Hagane Mugi, Iwate Mensury 2 and Mokusekko 3 seem to be resistant to all known virus strains and are probably the most useful for plant breeders.  相似文献

Samenvatting Een aantal isolaten van het gerstegeelmozaïekvirus, afkomstig van verschillende plaatsen in Engeland, werden met elkaar vergeleken. Uit de resultaten, verkregen met ELISA, goudmerking en mechanische overdracht, kan worden geconcludeerd, dat tenminste twee stammen van het virus in Engeland voorkomen. The BaYMV resistance of German cultivars like Diana, Franka, Gloria or Sonate is due to one recessive gene (‘German gene’), located on barley chromosome 3. This gene and the gene Ym1 are most probably allelic (or tightly linked). Resistance of the American cv. Anson barley is inherited independently of the ‘German gene’ and Ym1. The haploidy technique is an efficient means for approaching major breeding goals: (1) to improve quality characteristics of cultivars carrying‘German resistance' (2) to adapt exotic germplasm carrying the gene Yml to European growing conditions; (3) to broaden the genetic base of BaYMV resistance by incorporating additional ‘new’ resistance genes.  相似文献

南芥菜花叶病毒是我国进境植物检疫性有害生物,为建立快速灵敏的检疫检测方法,本研究用生物素标记一条引物,用荧光素(或地高辛)标记另一条引物,经RT-PCR扩增产生双标记的扩增产物,建立了PCR扩增产物的胶体金层析检测和ELISA检测方法。胶体金层析检测方法在15min即可获得检测结果,ELISA方法需20h,后者检测的灵敏度可比前者分别提高100倍和5000倍。所建立的2种方法都免去了普通PCR检测的溴化乙锭染色和电泳的过程。  相似文献

Infection by barley yellow mosaic virus (BaYMV) was greater in a crop sown in October than in November and in cv. Maris Otter than in cv. Sonja. However, a greater proportion of plants of Sonja than of Maris Otter were infected, and more severely, with the soil-borne fungus vector of BaYMV. Polymyxagraminis. Indifferent years the yield of individual plants of Maris Otter was decreased by 15% and of Sonja by 40%. The spring barley cultivars Athos. Porthos. Georgie and Ark Royal were all infected with BaYMV when sown in autumn in infected soil. Infected winter barley cv. Igri lost up to 50% of yield when this was measured across a naturally infected patch of crop.  相似文献

利用双抗夹心酶联免疫吸附测定(DAS-ELISA)的方法对甘肃出入境南瓜、西葫芦种子及采自河西地区显症病株叶片进行检测,在种子及病叶组织中均检测到ZYMV病毒,其中南瓜种子带毒批次占12.5%,西葫芦种子带毒批次占11.8%。根据已报道的小西葫芦黄花叶病毒(Zucchini yellow mosaic virus)基因组核苷酸序列,设计引物扩增其外壳蛋白(CP)基因,以ELISA阳性种子或病叶组织总RNA为模板,进行RT-PCR扩增,对预期大小的扩增产物进行测序,结果表明扩增获得的核苷酸序列与世界各地的ZYMV分离物CP基因具有高度一致性,综合ELISA检测和RT-PCR的结果,确定南瓜、西葫芦种子可携带ZYMV,且ZYMV是侵染甘肃瓜类作物的重要病毒种类。  相似文献

Improved diagnosis of barley yellow mosaic (BaYMV) and barley mild mosaic (BaMMV) viruses was obtained by adjusting the buffers used in immunospecific electron microscopy (ISEM) to ensure a pH ≥ 7-0 and in ELISA by replacing ovalbumin with 10 g/l full cream milk powder.
Over 70% of samples of winter barley with symptoms of mosaic received from different sites in the UK during 1987-90 had BaYMV and 37% had BaMMV, with 11% containing both viruses. BaMMV was much more common on malting cultivars than on those grown for feed and this resulted in an easterly bias to the geographical distribution of the virus. Both viruses were, however, widely distributed in areas where winter barley is grown intensively. A small number of BaYMV records were from cultivars previously regarded as resistant and these are probably a distinct strain of the virus.  相似文献

Barley yellow mosaic virus was first reported in the United Kingdom in 1980. Since then cumulative records of reports of disease, confirmed at the laboratory of the Agricultural Development and Advisory Service of the Ministry of Agriculture, Fisheries and Food, show how infection has spread from the east of England where it first occurred in quantity, to the west. Areas of concentrated infection appear to relate to areas of intensive winter-barley cultivation. Records suggest that infection may have occurred as early as 1976. Patterns of local spread in fields as defined in aerial photographs reflect the means by which the viruliferous vector may be passively moved in the soil. Other patterns reflect previous cropping. The higher frequency of infection on the sites of long-removed hedgerows is not explained.  相似文献

小麦黄花叶病是由禾谷多黏菌传播的小麦黄花叶病毒引起的病害,近年在黄淮麦区呈蔓延加重趋势。为了给病害的防治和抗病育种工作提供依据,本研究利用分级评价方法对黄淮地区推广的小麦品种进行了田间抗病性鉴定。两年鉴定结果表明,在145个供试小麦品种中,‘濮优938’、‘新麦208’、‘豫麦416’、‘新原958’、‘豫麦70-36’、‘泛麦5号’等70个品种表现为免疫,占总数的48.28%;‘豫麦47’和‘邯6172’表现为抗病,占总数的1.38%;‘洪育2号’、‘花培2号’、‘偃展4110’、‘豫麦41’、‘郑麦9023’等48个品种表现为中抗,占总数的33.10%;‘兰天06129’、‘兰天0591’、‘徐麦9158’、‘徐麦0054’、‘徐麦1108’等25个品种表现为感病,占总数的17.24%。研究结果为指导小麦黄花叶病区合理选择小麦品种提供了科学依据。  相似文献

ABSTRACT Molecular markers linked to quantitative trait loci conditioning tolerance to barley yellow dwarf virus (BYDV) were identified in oat (Avena sativa) using amplified fragment length polymorphism (AFLP) analysis. Near-isogenic and recombinant inbred lines (NILs and RILs, respectively) derived from a cross of Clintland64 (BYDV-sensitive) and IL86-5698 (BYDV-tolerant) were evaluated for their responses to an Illinois isolate of the PAV strain of BYDV. Individual markers identified in the analysis of the NILs explained up to 35% of the variability seen in the tolerance response. Single-point analysis of the marker data from the RIL population identified 24 markers in three linkage groups that were associated with tolerance to BYDV infection at P /= 3.0. These loci explained about 50% total of the variation in BYDV tolerance in multimarker regression analysis in both years. The BYDV tolerance loci A, C, E, and R were mapped to hexaploid oat restriction fragment length polymorphism linkage groups 2, 8, 36, and 5, respectively, by analyzing the segregation of the AFLP markers in the Kanota x Ogle RIL population.  相似文献

In 1976 distinctive patterns were identified on aerial photographs of winter wheat crops in East Anglia. The patterns consisted of numerous dark foci which were found to be associated with sooty moulds (mostly Cladosporium spp.) in the crops. Both green and mature crops were photographed in 1977 and the patterns were matched with crop inspection data. These suggested that barley yellow dwarf virus (BYDV) was the cause of the patterns and that sooty moulds secondarily invaded the infected plants. A different type of focal pattern appeared to be associated with damage caused by aphid feeding. All crops showing the pattern associated with BYDV were sown before 13 October but there was no relationship with sowing date for the crops damaged directly by aphids. Yield losses within foci were estimated by sampling and the total crop areas affected were calculated using an image analysis computer: yield losses were estimated to range from 10% (BYDV) to 2% (aphid feeding). The epidemiology of BYDV is discussed and reasons are suggested for the development and appearance of the foci.  相似文献
