The effect of inoculation with Frankia, a N-fixing actinomycete, on the growth of Alnus sieboldiana seedlings was studied on unsterilized soil from a nursery and an alder stand (forest of Alnus firma). The seedlings of A. sieboldiana were inoculated with Frankia before or after a 2-month culture on sterilized vermiculite, during which they nodulated, and transplanted to unsterilized soil from the nursery and the alder stand. The control seedlings were also cultured on sterilized vermiculite for about 2 months and transplanted to unsterilized soil without Frankia inoculation. The seedling growth, nodulation and N-fixing activity were measured 3, 10 and 16 weeks after the transplantation. Growth and nodule biomass of the seedlings inoculated with Frankia and those grown on the alder soil were better than those without inoculation with Frankia and grown on the nursery soil, respectively. The seedlings inoculated before spontaneous nodulation grew better than those inoculated at the transplantation. Nitrogen-fixing activity measured by acetylene reduction assay at 16 weeks after the transplantation was higher in the seedlings grown on the soil from the nursery than on the soil from the alder stand.  相似文献

Red pine and jack pine seedlings growing in styroblocks were inoculated 8 wk after sowing with mycelium/agar slurries of 3 mycorrhizal fungi (Laccaria bicolor, Scleroderma citrinum, and an unidentified basidiomycete), and one suspected mycorrhizal fungus (Cantharellula umbonata). Seedlings inoculated with L. bicolor developed mycorrhizae earlier and in greater numbers than the other inoculation treatments, with red pine out-performing jack pine in both respects. At 34 wk following sowing, seedlings were outplanted on a cleared xeric site in Baraga Co., in Michigan's Upper Peninsula. Seedlings inoculated with C. umbonata failed to form mycorrhizae and were not outplanted. Inoculation treatments did not affect shoot or root weight at outplanting. Red pine inoculated with L. bicolor averaged 21% and 19% greater survival compared with control seedlings after one and two years in the field, respectively. Other inoculation treatments failed to increase seedling survival for either tree species. Jack pine demonstrated higher overall survival than did red pine for both years in all corresponding inoculation treatments.  相似文献

We investigated the role of tetrapartite associations between an arbuscular mycorrhizal (AM) fungus (Glomus geosporum), phosphate solubilizing bacteria (Paenibacillus polymyxa), Frankia and Casuarina equisetifolia on growth, nutrient acquisition, nutrient utilization and seedling quality of C. equisetifolia. Seedlings of C. equisetifolia were grown in an Alfisol soil and inoculated with G. geosporum, P. polymyxa and Frankia either individually or in combinations. Inoculation of bioinoculants stimulated seedling growth, the efficiency of nutrient uptake and improved seedling quality. However, microbial inoculation generally reduced the efficiency of nutrient utilization in dry matter production (nutrient use efficiency). Inoculation of P. polymyxa or Frankia increased the extent of AM colonization, which resulted in the accumulation of the nutrients. Seedlings inoculated with Frankia and G. geosporum had more, and heavier nodules compared to seedlings inoculated with Frankia alone. Dual inoculation of microbes was more effective than individual inoculations. The growth response of seedlings to inoculation involving all the microbes was greater than the response to either individual or dual inoculations. The results of this study showed that the tetrapartite association could improve the growth, nutrient acquisition and seedling quality of C. equisetifolia under tropical nursery conditions.  相似文献

Experiments were made to determine the effect of inoculation withRhizobium, the ectomycorrhizal fungus Hebelomamesophasem and the arbuscular mycorrhizal fungus Glomuscaledonium on the growth and nitrogen fixation of black locust(Robinia pseudoacacia Linn.) seedlings grown invitro and in pot culture. The results showed that both mycorrhizalfungi and Rhizobium stimulated the growth and nitrogenfixing ability of inoculated seedlings. Inoculation with all three microbestogether produced the most beneficial effects on nitrogen fixation, mycorrhizaldevelopment and seedling growth.  相似文献

An investigation was carried out to screen and select efficient vesicular arbuscular mycorrhizal (VAM) fungi for inoculating the forest tree species, Casuarina equisetifolia. The seedlings were inoculated with 10 different VAM fungi, obtained from various sources. Inoculated seedlings generally had greater plant height, stem girth, biomass and P content than uninoculated plants. They also had more mycorrhizal root colonization and spore numbers in root zone soil. C. equisetifolia seedlings responded best (in biomass) to inoculation with Glomus mosseae (Nicolson and Gerdemann) Gerdemann and Trappe, closely followed by Acaulospora laevis Gerdemann and Trappe and G. fasciculatum (Thaxter Sensu Gerdemann) Gerdemann and Trappe; all the three being statistically on par with each other.  相似文献

Miyake-Jima, a round island of about 8 km in diameter, is located about 200 km south of Tokyo, Japan (34°08′ N, 139°53′ E). In July 2000, the central cone of this island collapsed and a new eruption started with the falling of volcanic ash and SO2 emission. In October 2001, we measured the distribution of bacteria, and Frankia, a symbiotic nitrogen-fixing actinomycete that forms root nodules, and ectomycorrhizal fungi associated with alder in newly deposited ash and its underlying soil by cultivation of Alnus sieboldiana seedlings in these soils. There were fewer bacteria growing on the nutrient broth agar and fewer bacteria growing on 100-fold diluted nutrient broth agar in newly deposited volcanic ash than there were growing in the buried old soil. In four out of five sites, little or no nodulation was observed in newly deposited ash; abundant root nodules were formed in the underlying soil. Ectomycorrhizae formed in the seedlings cultivated in the underlying soil. In May 2003, the distribution of Frankia and ectomycorrhizal fungi in soil at different depths (up to 160 cm) was also investigated. Frankia and ectomycorrhizal fungi were relatively abundant in surface soil layers.  相似文献

Commercially produced vegetative inocula of Laccaria laccata and Hebeloma crustuliniforme successfully formed ectomycorrhizae with Douglas-fir transplanted container (plug+1) seedlings. After 4.5 months in containers, 83% and 90%, respectively, of short roots were mycorrhizal. L. laccata- or H. crustuliniforme-inoculated seedlings had significantly more mycorrhizal and total short roots than Pisolithus tinctorius-inoculated (4% mycorrhizal root tips) or uninoculated control seedlings. No significant differences were detected in seedling growth at the end of the container phase.After transplantation and growth in nursery beds for 17 months, mean new short root colonization of all seedlings was 80%. H. crustuliniforme persisted as a dominant mycorrhizal fungus on seedlings initially inoculated with this fungus. L. laccata-inoculated seedlings had 40% of their short roots colonized by L. laccata and another 40% by native fungi Rhizopogon and Thelephora spp. All mycorrhizae of control seedlings and those inoculated with P. tinctorius were formed by fungi native to the nusery beds. A significant fungal treatment effect was detected for shoot height only. Three VA-mycorrhizal fungi; Glomus occultum, Glomus aggregatum (local isolates) and G. mosseae (strain from Bangalore, India) were inoculated to assess their effect on growth of Acacia mangium in lateritic soil. All inoculations enhanced growth with respect to shoot height, root diameter, leaf area, chlorophyll content and biomass of A. mangium significantly compared to uninoculated control seedlings. G. occultum proved most efficient among the three. The mycorrhizal dependency factor indicated that the growth of A. mangium was 57% dependent on G. occultum, 47% on G. mosseae and 46% on Glomus aggregatum.  相似文献

Our study aimed to assess the effect of dual inoculation with the selected Rhizobium strain KWN35 and the arbuscular mycorrhizal isolate BEG 176 (Glomus etunicatum) on the growth of Calliandra calothyrsus cultivated under irrigation in the field in Senegal (Dakar) over a period of 24 months. Although plants inoculated with both microsymbionts grew better (height and root collar diameter) than plants from three other inoculation treatments (control, single inoculation with KWN35 or BEG 176), these results were not statistically different except at 1 and 5 months after field transplantation. KWN35 was present in a relatively high percentage of nodules harvested from plants inoculated with this rhizobial strain either alone or with mycorrhiza (approximately 60% and 40% 1 and 2 years after field transplantation respectively). The percentage of mycorrhizal root infections was around 60% in the inoculated plants after 12 months with significantly higher N, P and K foliar contents of trees compared with the non-inoculated controls. However, as for nodulation, by 24 months after transplantation, there were no significant differences between treatments. We conclude that field inoculation of C. calothyrsus with Rhizobium strain KWN35 and arbuscular mycorrhizal isolate BEG 176 did not have a long-lasting effect on the growth of trees, even when a majority of nodules were occupied by the inoculated rhizobia and the roots infected by the mycorrhiza. Several reasons can be postulated for these results, such as a possible effect of soil fertility on the efficiency of the nodules.  相似文献

Nitrogen fixing and non-N2 fixing legumes such as Gliricidia speium and Senna siamea have been used in alley cropping systems for soil improvement and source of N for food crops. However their establishments could be limited by P and moisture deficiencies in degraded soils. Vesicular-arbuscular mycorrhizal fungi can help to overcome these deficiencies. We examined the effects of a vesicular-arbuscular mycorrhizal (VAM) fungus, Glomus deserticola, on the performance of sole hedgerow trees of Gliricidia sepium and Senna siamea and their mixtures (interplanted) in a fallowed alley cropping experiment on a degraded Alfisol in southwestern Nigeria. Percentage root infection by VAM fungi was higher in inoculated plants than in uninoculated ones irrespective of whether they were interplanted or non-interplanted. Inoculation with G. deserticola increased dry matter accumulation and nutrient uptake (N. P, Mg and K) but there was no significant interaction between mycorrhizal inoculation and interplanting for growth and nutrient uptake except for the uptake of P, Mg and K in G. sepium. Inoculation with G. deserticola reduced leaf shedding of G. sepium by 50% but did not have the same effect for S. siamea. For both tree species inoculated plants extracted more water from 0–30 cm depth than the uninoculated ones.  相似文献

To clarify the differences in susceptibility of six species in the Fagaceae (Quercus crispula, Quercus serrata, Quercus acutissima, Quercus phillyraeoides, Quercus glauca, and Castanopsis cuspidata var. sieboldii) to Raffaelea quercivora, we inoculated this fungus on seven potted 5-year-old seedlings of each species, observed symptom development, and measured xylem pressure potential (XPP) after inoculation. The first death was observed on the 11th day in Q. crispula and on the 56th day in Q. serrata. The number of dead seedlings of Q. crispula and Q. serrata were five and one, respectively, whereas no mortality was observed in the other four species. The XPP of inoculated seedlings in both Q. crispula and Q. serrata decreased after inoculation. In contrast, the XPP of inoculated seedlings of the other four species remained almost the same as in the control seedlings. These results indicate that R. quercivora is pathogenic to Q. crispula and Q. serrata and that the susceptibility of the six Fagaceae species in our study differed among species.  相似文献

Seedlings of Juglans nigra from three seed sources were grown in fumigated soil without vesicular-arbuscular mycorrhizal (VAM) fungi or inoculated with Gigaspora margarita, Glomus deserticola or Glomus etunicatum. Vesicular-arbuscular mycorrhizal development varied significantly between fungal symbionts within a black walnut source. Glomus deserticola and G. etunicatum produced the highest levels of root colonization in all sources. Significant differences in seedling shoot and root growth were attributed to root colonization by specific VAM fungi in each black walnut source. Glomus deserticola stimulated seedling leaf area and root weight 26 and 52%, respectively, in one seed source. Seedling leaf N, P and K concentrations were significantly improved by VAM in two seed sources. Juglans nigra seedlings respond favorably to VAM colonization. However, differences between seed sources suggest a strong host-symbiont interaction.  相似文献

To evaluate the effect of adjacent tree species on the susceptibility of Japanese black pine (Pinus thunbergii) to pine wilt disease, an inoculation experiment was conducted usingP. thunbergii seedlings potted with seedlings of six tree species,i. e. Alnus sieboldiana, Eurya japonica, Lespedeza bicolor formacutifolia, Pinus thumbergii, Robinia pseudo-acacia andSarothamus scoparius. About ten months after planting, they were inoculated with the pinewood nematode (Bursaphelenchus xylophilus) in early July 1992. After that, the proportion of pine seedlings with completely discolored foliage increased more quickly when the seedlings were potted withR. pseudo-acacia, S. scoparius orA. sieboldiana than when potted withP. thunbergii, L. bicolor orE. japonica. At the end of the study period, 17 weeks after inoculation, it reached 90.6%, 90.0%, 87.5%, 72.7%, 63.3%, and 59.4% when the pine seedlings were potted withR. pseudo-acacia, S. scoparius, A. sieboldiana, P. thunbergii, L. bicolor andE. japonica, respectively. This indicated that the susceptibility ofP. thunbergii seedlings to pine wilt disease was influence by the species of adjacent trees.  相似文献

[目的]通过板栗幼苗人工接种食用菌根菌固体菌剂,分析其对板栗幼苗生长指标和根系菌根化的效果,以验证大红菇(菌株编号2014-10)、红绒盖牛肝菌(菌株编号2014-14)和美味牛肝菌(菌株编号50559)这3种食用菌根菌固体菌剂作为板栗接种体的有效性及其促生效应,为进一步探索"板栗食用菌根菌新型经济林栽培模式"提供基础。[方法]在板栗播种时分别接种上述3种食用菌根菌固体菌剂,并设空白作对照。对当年生幼苗苗高、地径、根系干质量、侧根数、侧根总长、幼苗侵染率、吸收根侵染率等指标进行调查和分析,并制作菌根石蜡切片进行微观结构观察。[结果]表明:(1)上述3种食用菌根菌在人工接种下均能侵染板栗幼苗根系,平均幼苗菌根化率分别达95.8%、87.0%和96.7%,平均吸收根菌根化率分别达57%、53%和50%,达较高的侵染强度;经微观观察,板栗幼苗根系均形成典型的菌根结构——菌丝套和哈蒂氏网。(2)与对照幼苗相比,3个菌种处理幼苗的平均苗高分别提高32.5%、24.0%和22.7%,平均地径分别提高19.4%、14.1%和5.0%,差异显著(P0.05);平均根系干质量分别提高18.9%、25.3%和14.1%,平均侧根数分别提高16.0%、14.1%和12.1%,平均侧根总长分别提高26.8%、28.9%和44.8%,差异极显著(P0.01)。[结论](1)上述供试食用菌根菌均与板栗根系建立了共生关系,幼苗菌根化效果良好,吸收根菌根化强度高,形成了菌根的典型结构,即菌丝套和哈蒂氏网,验证了上述菌种固体菌剂的有效性,认为其可以作为板栗食用菌根菌栽培的接种体而用于生产实践,为探索"板栗食用菌根菌新型经济林栽培模式"提供了基础。(2)上述供试菌种接种后,对板栗幼苗各项主要生长指标均有不同程度的促进作用,说明上述食用菌根菌对板栗幼苗具有显著的促生效应。  相似文献

Some Acacia mangium Willd. plantations in Asia grow poorly due to low soil fertility and the absence of compatible mycorrhizal fungi. This legume tree can be colonized by arbuscular mycorrhizal (AM) and ectomycorrhizal (ECM) fungi but inoculation is not routinely practiced. This study aimed to compare the effectiveness of AM fungi and four isolates of the ECM fungus Pisolithus tinctorius (Pers.) Coker and Couch (Pt) in promoting growth of A. mangium seedlings under glasshouse conditions. AM inoculants were: Glomus etunicatum, G. fasciculatum, G. macrocarpum and Gigaspora margarita and mixed species extracted from rhizosphere soil of a Populus stand in Suwon (AMKFRI), and Carex (AMM6) and Populus (AMM7) growing in mine tailings in Korea. Pisolithus isolates were from Philippines (PtPhil) and Korea (PtKFRI, PtMKACC, PtKACC). Generally, ECM fungi promoted height and diameter growth of A. mangium more than the AM inoculants. The Korean Pisolithus increased plant dry weight by 122–145%, mixed AM inoculants by 61–97%, and Glomus and Gigaspora by 45–72% over the control. PtKACC gave the highest root colonization and promoted the highest growth and concentration of most nutrients. Mycorrhizal root colonization was positively correlated with plant dry weight, Na, Fe and Cu concentrations and N, P, K, Ca, Na, Fe and Cu contents. In conclusion, the results provide strong evidence for benefits of mycorrhizal inoculation on A. mangium seedlings under glasshouse conditions. The Korean Pisolithus isolates (particularly PtKACC), and two AM fungi (AMKFRI and AMM6) are potential mycorrhizal inoculants but their effectiveness and persistence should be determined on degraded lands in tropical countries where A. mangium is being planted for rehabilitation.  相似文献

The benefits of inoculation with six arbuscular mycorrhizal fungi (AMF) isolates (Glomus aggregatum, G. fasciculatum, G. intraradices, G. manihotis, G. mosseae, and G. verriculosum) were investigated on seedlings of Acacia senegal (L.) Willd., a multipurpose tree legume highly valued for arabic gum production. Mycorrhizal root colonization, plant growth and relative mycorrhizal dependency (RMD) were measured in A. senegal seedlings growing in soils from three geographical sites in Senegal (Dahra, Bambey and Goudiry) and two soil conditions (sterilized vs unsterilized) in the glasshouse. The impact of inoculation on mycorrhizal root colonization and plant growth depended on AMF isolates, soil origins and soil conditions. Mycorrhizal root colonization and plant growth were increased in sterilized soils regardless of soil origin and AMF isolates. The degree of RMD of A. senegal seedlings varied with soil origin, soil condition and AMF isolates. A. senegal showed the highest RMD values, reaching a maximum of 45 %, when inoculated with G. manihotis. However, in unsterilized soils, no significant effect of AMF inoculation on plant growth was observed despite significant root colonization with certain AMF isolates in Dahra and Goudiry soils. This indicates that the most infective AMF isolates were not the most effective and unsterilized soils may contain effective mycorrhizal propagules. In conclusion, it is important to consider the native mycorrhizal component of the soils before harnessing mycorrhizal inoculation programs for sustainable agroforestry systems.  相似文献

Inoculation of nursery beds with vegetative mycelia of P. tinctorius (Pt) significantly increased the number of salable 1–0 red oak seedlings produced in two of three years and the average stem height and diameter each year. The percentage of Pt mycorrhizal primary lateral roots on seedlings in the inoculated plots was significantly correlated with stem diameter and weight of lateral roots. The high rates of colonization of short roots by Pt in inoculated plots suggested the test fungus could successfully compete with indigenous fungi in forming mycorrhizae.  相似文献

The effect of two arbuscular mycorrhizal (AM) fungi, Glomus fasciculatum and G. macrocarpum on shoot and root dry weights and nutrient content of Cassia siamea in a semi-arid wasteland soil was evaluated. Under nursery conditions, mycorrhizal inoculation improved growth of seedlings. Root and shoot dry weights were higher in mycorrhizal than non-mycorrhizal plants. The concentration of P, K, Cu, Zn and Na was significantly higher in AM inoculated seedlings than in non-inoculated seedlings. Mycorrhization led to decrease in alkalinity of the rhizosphere soil from pH 8.5 to 7.4. Under nursery conditions, the degree of mycorrhizal dependency increased with age of C. siamea seedling. On transplantation to the field, the survival rate of mycorrhizal seedlings (75%–90%) was higher than that of non-mycorrhizal seedlings (40%). Besides better survival rate, AM inoculation improved the growth performance of seedlings in terms of height and stem diameter. Among the two AM fungi used, the efficiency of Glomus macrocarpum was higher than that of G. fasciculatum under both nursery and field conditions.This revised version was published online in May 2005 with corrected page numbers.  相似文献

The effect of 3 species of vesicular-arbuscular mycorrhizal (VAM) fungi on the growth of Leucaena leucocephala (Lam.) De Wit. in a phosphorus-deficient and aluminumsulfate (AIS)-treated medium was investigated in a screenhouse experiment. Plant height, root length, nodulation, phosphorus uptake and nitrogen fixation were used as indices of plant performance.While there were significant differences among mycorrhizal plants with respect to these indices, they outperformed their non-mycorrhizal counterparts in all respects except in nitrogen content. Of the 3 mycorrhizal species studied, Glomus etunicatum (Becker and Gerd) was the most efficient, followed by Glomus fasciculatum (Thaxter) Gerd and Trappe, and finally, Gigaspora margarita (Becker and Hall). Both aluminumsulfate and mycorrhizal treatments increased shoot dry weight.  相似文献
