Salmonella enteritidis phagetype 8 was isolated from ill humans, milk-line filters, milk from a bulk tank, and milk from the right hind quarter of a five-year-old Holstein cow on a dairy farm in southern Alberta. The affected animal was removed from the herd and continued to shed S. enteritidis from this quarter during a seven-month interval in an isolation facility. Milk from the affected quarter was visually normal, and no other pathogen was isolated from the udder during the investigation. After removal of the infected cow from the herd, milk from the bulk tank was culturally negative for Salmonella sp. during the succeeding 15 months.  相似文献

A three-year study of Salmonella dublin infection in a closed dairy herd   总被引:3,自引:0,他引:3  
Over a period of three years, Salmonella dublin was isolated occasionally from the faeces of nine adult cattle in a closed dairy herd. The organism was also isolated from 12 of the samples collected after parturition; isolations were made from newborn calves on 11 occasions, from a vaginal swab once and from a milk sample once. Nine of the isolations from the calves were made from swabs obtained within 24 hours of birth. Throughout the investigation isolations were made from heifers, steers and older calves and 11 infected animals were detected. S. dublin was widespread in the farm environment and it was concluded that environmental contamination was an important source of infection for animals of all ages, some of which may have become latent carriers. The family history of one cow, seven of whose offspring were infected with S dublin, suggested the possibility of vertical transmission. Without reliable tests to detect latent carriers, it is suggested that control of this infection must be based on improved hygiene and the use of vaccination to improve the immunity of the herd.  相似文献

The purpose of this study was to assess changes in dairy herd milk production and milk composition associated with changes in Salmonella contamination of bulk milk on dairy farms in southwestern Ontario. Twenty-three dairy farms that had submitted milk filters for culture from which Salmonella were isolated (cases) and 23 farms that submitted Salmonella-negative milk filters (controls) were included in the study. The rolling herd averages for milk and fat of case and control farms for the months of December 1985, December 1986 and April 1987 were compared and no significant differences were detected. Case and control farms were divided into three groups (A,B,C) on the basis of Salmonella culture results of milk filters submitted at various time periods throughout the study. Daily and monthly changes in milk production and composition parameters that reflected the time periods of milk filter culture were compared. The following unconditional associations between a changing Salmonella infection status on dairy farms and changes in milk production or composition variables were significant (p less than or equal to 0.05): group A: case farms had higher plate loop counts than control farms; group B: case farms had younger cows than control farms; group C: case farms had cows with longer average days in lactation than control farms. After analytical control of confounding variables, the disappearance of Salmonella from bulk milk supplies of dairy farms was associated with a decrease in percent fat and in somatic cell count.  相似文献

Clostridium difficile was investigated as a possible cause of enteritis in calves. The organism and its toxins (TcdA and TcdB), respectively, were found in 25.3% and 22.9% of stool samples from diarrheic calves. Culture positive samples were more likely than culture negative samples to be toxin positive. However, toxin positive stools were more common among nondiarrheic calves, but diarrheic calves were nearly twice as likely to be culture positive. Ribotype 078 was dominant among isolates. Salmonella sp. was isolated from both diarrheic and nondiarrheic calves, but large numbers of E. coli were found more commonly in diarrheic calves than in nondiarrheic animals. Prevalence rates for coronavirus and Cryptosporidium sp. were substantially higher in nondiarrheic calves than in diarrheic, but rates of detection of rotavirus and Giardia sp. were more nearly equal between groups. Lesions in naturally infected calves included superficial mucosal erosion with associated fibrinous exudates. Neutrophils and eosinophils infiltrated lamina propria. Large Gram-positive rods morphologically compatible with C. difficile were abundant in the colonic lumen and the organism was isolated by bacteriologic culture. Toxins were found throughout the colon. Purified toxins A and B (individually and conjointly) caused comparable lesions, as well as fluid accumulation, in ligated intestinal loops. Our findings are in substantial agreement with those of others [Rodriguez-Palacios, A., Stampfli, H.R., Duffield, T., Peregrine, A.S., Trotz-Williams, L.A., Arroyo, L.G., Brazier, J.S., Weese, J.S., 2006. Two beef cow herds were investigated to determine the prevalence of subclinical mastitis and to determine whether it contributed to decreased weaning weight of calves nursed by affected cows. Quarter samples obtained about 30 days after calving were evaluated by the California mastitis test and by direct microscopic cell count. In 1 herd, a 92-cow confinement operation, 17 quarters of 12 cows were infected with Staphylococcus aureus, Streptococcus sp other than agalactiae, or Klebsiella sp. Mean 205-day adjusted weights were 203.9 kg and 218.2 kg for calves nursed by infected vs noninfected cows. In the other herd, a 75-cow range-pasture operation, 8 quarters of 8 cows were infected, all with S aureus. Mean 205-day adjusted weights were 162.5 kg and 174.6 kg for calves nursed by infected vs noninfected cows.  相似文献

Acute epidemic salmonellosis was investigated in the winter of 1978 in 4 dairy herds in the Waikato district. The outbreaks involved pregnant or recently calved cows. Calves were affected on only one farm. Salmonella bovismorbificans was isolated from the faecal, uterine and environmental samples from the herd most severely affected. In this herd 20, (20%) of the cows died; 10 calves were either born dead or died soon after birth; and 10 cows were culled as unlikely to return to production. S. typhimurium was involved in the outbreaks in the other 3 herds and from one of these farms salmonellae were isolated from the faeces of the 2 farm dogs and from a 12-month-old infant with dysentery. Salmonellae were recovered from all of the clinically ill cows and from 36% of the asymptomatic animals. Salmonellae were widespread on the properties and were isolated from 81% of soil samples; 36% of drain swabs; and from all the pairs of rubber boots worn while collecting samples.  相似文献

OBJECTIVE: To evaluate therapeutic efficacy of a high extralabel dose of ceftiofur for treatment of experimental salmonellosis in neonatal calves. ANIMALS: Forty-two 1- to 4-day-old Holstein bull calves. PROCEDURE: 36 calves were orally challenged with Salmonella enteritica serovar Typhimurium (6.5 x 10(8) colony-forming units). Six additional calves were retained as nonmedicated nonchallenged control calves. Four days following Salmonella challenge, surviving calves were randomly allocated to ceftiofur-treated (5 mg/kg, IM, q 24 h) or nonmedicated control groups. Calves assigned to the treated group were medicated daily for 5 days starting on day 4 after challenge. Calves were monitored for 18 days following Salmonella challenge. Outcome assessments included clinical parameters (attitude, appetite, fecal characteristics, and rectal temperature), mortality rate, and quantitative Salmonella culture of fecal samples, mesenteric lymph nodes, and cecal contents. RESULTS: Ceftiofur treatment was associated with a significant decrease in rectal temperature and diarrhea. Three of 15 medicated calves and 4 of 14 non-medicated calves died or were euthanatized between days 4 and 18. A significant decrease in fecal shedding of Salmonella organisms was observed in treated calves, compared with nonmedicated calves. Salmonella organisms were isolated from all 10 non-medicated calves at necropsy, whereas no Salmonella organisms were isolated from 5 of 12 medicated calves. CONCLUSIONS AND CLINICAL RELEVANCE: Treatment of salmonellosis in neonatal calves with a high extralabel dose of ceftiofur (5 mg/kg, IM, q 24 h) promotes animal welfare, reduces fecal shedding of Salmonella organisms, and may promote clearance of Salmonella infections when plasma ceftiofur concentrations are maintained above minimal inhibitory concentrations.  相似文献

The overall purpose of this study was to provide information on animal and occupational health associated with the infection of a diary herd with Salmonella Muenster that would be useful in the management of dairy herds so infected. This retrospective, longitudinal report records a 2-year infection of a 140-cow dairy herd with S. Muenster, which was likely introduced by additions to the herd. Six cows aborted or had diarrhea due to salmonellosis in the last trimester of pregnancy. Additions to the herd and the presence of animals that had not received an Escherichia coli bacterin-toxoid were risk factors for salmonellosis. One neonate died, and 24 of 36 calves born between November 1998 and May 1999 had diarrhea by 1 mo of age. Initially, over 60% of the cows were fecal positive; within 6 months, all cows but I had become infected. The intermittent shedding of the organism and the eventual zero prevalence highlight the inappropriateness of extensive culling as an eradication strategy. Cultures of the bulk-tank milk filters were more sensitive than cultures of the bulk-tank milk samples at detecting S. Muenster. Two months after the index case, S. Muenster was cultured from the milk of 7.8% of the cows. Positive fecal or milk cultures were not associated with impaired health or production. The herd's milk was a zoonotic risk, but contact with infected animals was not. The organism spread easily between operations, likely via manure-contaminated clothing and footwear.  相似文献

OBJECTIVE To identify the common serotypes and antimicrobial resistance patterns of Salmonella spp. associated with diarrhoea in Australian dairy calves under the age of 6 weeks. DESIGN Cross-sectional study. METHODS Faecal samples were collected from outbreaks of diarrhoea in dairy and dairy beef calves less than 6 weeks old. Samples were screened for Salmonella using standard enrichment culture techniques. The antimicrobial susceptibility to 12 commonly used veterinary and human antimicrobials was assessed using the Kirby-Bauer disk diffusion method and the susceptibility profiles of dairy and dairy beef properties were compared using Fisher's exact test. RESULTS Salmonella ser. Dublin, S. ser. Typhimurium and S. ser. Bovismorbificans were the three most common salmonella serotypes isolated. The majority of properties had one serotype. Most of the Salmonella isolates were not resistant to any of the antimicrobials tested. No resistance was seen to amikacin and nalidixic acid, and only one isolate was resistant to ceftiofur or amoxicillin-clavulanic acid. The most common antimicrobial resistance was to streptomycin, ampicillin or combination sulfonamides. Multi-drug resistance was detected in S. ser. Anatum, S. ser. Bovismorbificans, S. ser. Muenster, S. ser. Newport and S. ser. Typhimurium. Isolates from dairy beef properties were more likely to be resistant to ampicillin, kanamycin, neomycin, sulfamethoxazole/trimethoprim and tetracycline (P < 0.05) and were more likely to exhibit multi-drug resistance. CONCLUSION The prevalence of antimicrobial resistance in Salmonella isolates from dairy calves in Australia is low compared with that reported overseas. From a human health perspective, resistance to antimicrobials used in the treatment of human salmonellosis was infrequent.  相似文献

An epidemiological investigation of a calf rearing premises and a closely associated dairy herd was carried out after the isolation of Salmonella enterica serovar Paratyphi B variant Java phage type 3b variant 2 from clinically diseased calves on the premises. The isolate was resistant to ampicillin, chloramphenicol, streptomycin, sulphonamides, tetracyclines, trimethoprim and cefoperazone. The organism was widespread on the calf unit and was also recovered from the dairy premises, mainly from groups of weaned calves. The investigation was extended to 10 epidemiologically linked farms but no S Java was isolated from any of the 40 to 60 samples collected from each premises. Molecular studies showed that the S Java isolates were genetically most similar to isolates from cases of human disease associated with ornamental fish tanks or feed. Long PCR and resistance gene profiling identified a resistance island which was indistinguishable from the human 'fish tank' strain of S Java and animal and human epidemic strains of S Typhimurium DT104. The isolates were clearly distinguished from multi-resistant S Java strains commonly associated with continental poultry. This is the first report of S Java with this resistance pattern in Great Britain.  相似文献

Three red deer calves (Cervus elaphus) died with a haemolytic disease associated with infection by Leptospira interrogans serovar pomona. Infection within the herd was more prevalent than disease. Sera from 16 herd mates were tested by the microscopic agglutination test (MAT) and 12 had leptospiral titres, the majority to serovar pomona. A few calves had titres to balcunica and hardjo. Urine was obtained for culture from six of these calves and serovar pomona was isolated from five with titres to pomona, and hardjo from one with a titre to hardjo but not pomona. A fourth calf died with severe nephritis but a diagnosis of leptospirosis was not confirmed in this case.  相似文献

Aromatic-dependent Salmonella typhimurium as modified live vaccines for calves   总被引:29,自引:0,他引:29  
Strains of Salmonella sp with complete nonreverting aromatic biosynthesis (aro) defects are expected to be nonvirulent, in respect to invasive infection, because they need the aromatic metabolites paraaminobenzoate (for making folate) and dihydroxybenzoate (for making enterochelin) which are not available in host tissues. Derivatives with transposon-generated complete nonreverting aro-defects were prepared from 3 mouse-virulent strains of S typhimurium, namely, FIRN, WRAY, and UCD. The latter 2 parent strains originally were isolated from calves and are known to be calf-virulent. The resultant aromatic-dependent (aro-) strains were used to vaccinate 27 calves (2 to 3 weeks old), usually giving 2 doses by the IM route (10(9) bacteria) or orally (1.5 X 10(11)). Vaccination did not cause severe ill effects in any calf. Thus aro- defects cause loss of virulence for calves, as previously shown for mice. Vaccinated and control calves were challenge exposed, usually at 5 weeks of age, by feeding 1.5 X 10(11) cells of 1 of 2 calf-virulent S typhimurium strains, either UCD 108-11 or SL1323. Of the 16 challenge-exposed control calves, all became anorectic and depressed (CNS), and 15 had diarrhea. Fourteen of the 16 died; all tested tissues were bacteriologically culture-positive for Salmonella at necropsy. A modified, double-antibody, enzyme-linked immunosorbent assay (ELISA) was developed to detect the K99 pilus antigen of enterotoxic Escherichia coli (ETEC) in feces of calves. Extremely high positive to negative ratios (greater than 200) were obtained by using monoclonal antisera as the primary antibody. Strong positive reactions were obtained with strains of E coli known to produce the K99 antigen; however, non-enteropathogenic E coli (strains not producing the K99 antigen), Salmonella, Proteus, Klebsiella, Pseudomonas, Staphylococcus, Streptococcus, and rotavirus produced negative results. Seventy-five fecal samples, 8 from healthy calves and 67 from calves with neonatal calf diarrhea were examined with the K99 ELISA for the presence of ETEC. Rotavirus test and fecal culture results were available on feces from calves with diarrhea and were used with the K99 ELISA results to determine the specific cause of the disease. Enterotoxic E coli was the predominant agent detected in the feces of 29 diarrheal calves less than 5 days of age. Mixed infections of rotavirus and ETEC were also common in these calves, but rotavirus infections alone were not detected. In 38 calves greater than or equal to 5 days, rotavirus was detected without ETEC. Of these calves, only 2 produced positive tests with the K99 ELISA. Salmonella sp and Proteus sp were detected from 5 of 67 calves with diarrhea.  相似文献

Epidemiologic investigations of Salmonella infections in dairy cattle often rely on testing fecal samples from individual animals or samples from other farm sources to determine herd infection status. The objectives of this project were to evaluate the effect of sampling frequency on Salmonella isolation and to compare Salmonella isolation and serogroup classification among sample sources on 12 US dairy farms sampled weekly for 7-8 weeks. Three herds per state were enrolled from Michigan, Minnesota, New York and Wisconsin based upon predefined herd-size criteria. Weekly samples were obtained from cattle, bulk tank milk, milk filters, water and feed sources and environmental sites. Samples were submitted to a central laboratory for isolation of Salmonella using standard laboratory procedures. The herd average number of cattle fecal samples collected ranged from 26 to 58 per week. Salmonella was isolated from 9.3% of 4049 fecal samples collected from cattle and 12.9% of 811 samples from other sources. Serogroup C1 was found in more than half of the samples and multiple serogroups were identified among isolates from the same samples and farms. The percentage of herd visits with at least one Salmonella isolate from cattle fecal samples increased with overall herd prevalence of fecal shedding. Only the three herds with an average fecal shedding prevalence of more than 15% had over 85% of weekly visits with at least one positive fecal sample. The prevalence of fecal shedding from different groups of cattle varied widely among herds showing that herds with infected cattle may be classified incorrectly if only one age group is tested. Testing environmental sample sources was more efficient for identifying infected premises than using individual cattle fecal samples.  相似文献

Faeces samples from calves with diarrhoea in 45 outbreaks were examined for six enteropathogens. Rotavirus and coronavirus were detected by ELISA in 208 (42 per cent) and 69 (14 per cent) of 490 calves respectively; calici-like viruses were detected by electron microscopy in 14 of 132 calves (11 per cent). Cryptosporidium were detected in 106 of 465 (23 per cent), Salmonella species in 58 of 490 (12 per cent) and enterotoxigenic Escherichia coli bearing the K99 adhesin (K99+ E coli) in nine of 310 calves (3 per cent). In the faeces of 20 per cent of calves with diarrhoea more than one enteropathogen was detected; in 31 per cent no enteropathogen was found. Faces samples from 385 healthy calves in the same outbreaks were also examined. There was a significant statistical association of disease with the presence of rotavirus, coronavirus, Cryptosporidium and Salmonella species (P less than 0.001). Healthy calves were not examined for calici-like viruses and the association of K99+ E coli with disease was not analysed because there were too few positive samples. Rotavirus infections were more common in dairy herds and single suckler beef herds whereas Salmonella infections were more often found in calf rearing units. Cryptosporidium were more common in single and multiple suckler beef herds. K99+ E coli were found in one dairy herd and one multiple suckler beef herd both with unhygienic calving accommodation. Variations in coronavirus detection among different farm types were not statistically significant. In this survey rotavirus was the most commonly detected agent in calf diarrhoea and Cryptosporidium were found in approximately one quarter of affected calves. Infection with Salmonella species was widespread, but K99+ E coli infections were less common in the United Kingdom than in other countries.  相似文献

Prevalence of enteric pathogens in the feces of healthy beef calves   总被引:5,自引:0,他引:5  
Fecal specimens from 136 healthy beef calves (1 day to 12 weeks of age) were examined for the presence of infectious agents known to cause enteric disease in calves. The calves were selected from 22 herds in which all calves were free of clinically apparent enteric disease. Salmonella sp, enterotoxigenic Escherichia coli, Cryptosporidium, and coronavirus were not detected in any of the calves. Three calves were infected with rotavirus and 1 calf was infected with Yersinia enterocolitica. Campylobacter-like bacteria were isolated from 50 of 130 calves, with 36 of the calves positive for C jejuni. Seemingly, clinically normal calves may be infected more often with enterotoxigenic E coli, Cryptosporidium, coronavirus, or rotavirus in herds in which some calves have enteric disease than in herds free of major enteric disease. Campylobacter jejuni was well adapted to the bovine host and was of similar prevalence in diarrheal and nondiarrheal calves. The K99 positive, nonenterotoxigenic E coli was isolated from the feces of 16 healthy calves. Information in addition to the presence of K99 antigen is useful when diagnosing enterotoxic colibacillosis in calves.  相似文献

Between October 1999 and February 2001 the salmonella status of 449 dairy farms in England and Wales was determined by environmental sampling on up to four occasions. Data were collected through interview-based questionnaires, and multivariable analyses were used to identify risk factors associated with the farms either being Salmonella positive (prevalence data) or becoming Salmonella positive (incidence data). Region, herd size, month of visit and the lack of a clean visitor parking area were significantly associated with the prevalence of Salmonella species, and there was a significant trend towards an increased risk in late summer and autumn. The introduction of six- to 24-month-old cattle into a herd was associated with a reduced prevalence, but the introduction of adult cattle only, or calves with other cattle, was associated with an increased (but not significant) risk of farms being Salmonella positive. Month of visit, the lack of a clean visitor parking area, the use of part-time workers and not feeding calves whole milk, but not region or herd size, were associated with an increased incidence of salmonella.  相似文献

Fecal samples were collected from 200 feeder-calves on farms in Tennessee, after assembly at a Tennessee auction market, and after transport to a Texas feedyard. A final fecal sample was collected from each calf after 30 days of feedyard confinement. The fecal samples were cultured for the presence of Salmonella spp. Salmonella isolates were serotyped and antimicrobial drug-resistance patterns determined. The number of calves fecal culture-positive for Salmonella spp increased from 0 on the Tennessee farms and auction market to 3/200 (1.5%) at entry into the Texas feedyard, and 16/200 (8%) after 30 days of feedyard confinement. Salmonella serotypes isolated and the number of isolates of each serotype were S reading (8), S cerro (4), S newbrunswick (3), S anatum (2), and S typhimurium (copenhagen; 2). All Salmonella isolates were resistant to 5 or more of 13 antimicrobial drugs tested. Salmonella reading isolates were resistant to 10 or 11 of 13 antimicrobial drugs. The results indicated that the calves could have been infected with Salmonella spp prior to or during the course of the study, and that marketing stress as they moved from farm through feedyard may have induced fecal excretion of salmonellae. In addition, the pattern of antimicrobial drug resistance in the Salmonella isolates was broad.  相似文献

The in vitro susceptibility of Escherichia coli, Salmonella sp, Pasteurella multocida, and Pseudomonas aeruginosa strains isolated from young calves and their environment to kanamycin and its semisynthetic derivative--amikacin, was compared by the tube dilution method. The minimal inhibitory concentration of amikacin was two- to fourfold lower than the minimal inhibitory concentration of kanamycin for the Salmonella group B, Ps aeruginosa and P multocida strains examined, whereas the E coli and Salmonella group D strains were eqaully susceptible to the 2 antibiotics. The concentrations of amikacin and kanamycin in the serum of calves after single intramuscular injections at 10, 25, and 50 mg/kg were determined, and differences between the 2 antibiotics were not observed. Intramuscular dosage schedules were calculated, based on the sensitivity of the different bacterial strains and the duration of effective serum drug concentrations.  相似文献
