Mutations in the human neurotrypsin gene are associated with autosomal recessive mental retardation. To further understand the pathophysiological consequences of the lack of this serine protease, we studied Tequila (Teq), the Drosophila neurotrypsin ortholog, using associative memory as a behavioral readout. We found that teq inactivation resulted in a long-term memory (LTM)-specific defect. After LTM conditioning of wild-type flies, teq expression transiently increased in the mushroom bodies. Moreover, specific inhibition of teq expression in adult mushroom bodies resulted in a reversible LTM defect. Hence, the Teq pathway is essential for information processing in Drosophila.  相似文献

Sleep is believed to play an important role in memory consolidation. We induced sleep on demand by expressing the temperature-gated nonspecific cation channel Transient receptor potential cation channel (UAS-TrpA1) in neurons, including those with projections to the dorsal fan-shaped body (FB). When the temperature was raised to 31°C, flies entered a quiescent state that meets the criteria for identifying sleep. When sleep was induced for 4 hours after a massed-training protocol for courtship conditioning that is not capable of inducing long-term memory (LTM) by itself, flies develop an LTM. Activating the dorsal FB in the absence of sleep did not result in the formation of LTM after massed training.  相似文献

Priming is a nonconscious form of human memory, which is concerned with perceptual identification of words and objects and which has only recently been recognized as separate from other forms of memory or memory systems. It is currently under intense experimental scrutiny. Evidence is converging for the proposition that priming is an expression of a perceptual representation system that operates at a pre-semantic level; it emerges early in development, and access to it lacks the kind of flexibility characteristic of other cognitive memory systems. Conceptual priming, however, seems to be based on the operations of semantic memory.  相似文献

Two distinct forms of consolidated associative memory are known in Drosophila: long-term memory and so-called anesthesia-resistant memory. Long-term memory is more stable, but unlike anesthesia-resistant memory, its formation requires protein synthesis. We show that flies induced to form long-term memory become more susceptible to extreme stress (such as desiccation). In contrast, induction of anesthesia-resistant memory had no detectable effect on desiccation resistance. This finding may help to explain why evolution has maintained anesthesia-resistant memory as another form of consolidated memory, distinct from long-term memory.  相似文献

The controlled motion of a series of domain walls along magnetic nanowires using spin-polarized current pulses is the essential ingredient of the proposed magnetic racetrack memory, a new class of potential non-volatile storage-class memories. Using permalloy nanowires, we achieved the successive creation, motion, and detection of domain walls by using sequences of properly timed, nanosecond-long, spin-polarized current pulses. The cycle time for the writing and shifting of the domain walls was a few tens of nanoseconds. Our results illustrate the basic concept of a magnetic shift register that relies on the phenomenon of spin-momentum transfer to move series of closely spaced domain walls.  相似文献

A model for sorted patterned ground shows that some types arise from density-driven Rayleigh free convection that occurs during thawing of water-saturated recurrently frozen soils. The regularly spaced convection cells result in an uneven melting of the underlying ice front. Frost action causes stones to be upthrusted and to form in a pattern on the ground surface that mirrors the corrugation in the underlying ice front. The implications of the water circulation direction in the cells on the sorting process are considered.  相似文献

Quartz grains that appear to have been shock-metamorphosed occur within three closely spaced shale beds from the uppermost Triassic ("Rhaetian") Calcare a Rhaetavicula in the Northern Apennines of Italy. The upper shale coincides with the abrupt termination of the distinctive, uppermost Triassic Rhaetavicula fauna and is overlain by the Hettangian (Lower Jurassic) Calcare Massiccio; no extinctions appear to be associated with the two lower layers, which occur 1.2 and 2.4 meters below the boundary shale. Approximately 5 to 10% of the quartz grains within these layers exhibit one or more sets of planar deformational features whose orientations cluster around the rational crystallographic planes (basal, omega, and pi) most commonly observed in shocked quartz. Textural and stratigraphic observations support an interpretation of at least three closely spaced impacts at the end of the Triassic.  相似文献

Associated locally with well-known tufa mounds and towers of Mono Lake, California, are subvertical, concretionary sand structures through which fresh calcium-containing artesian waters moved up to sites of calcium carbonate precipitation beneath and adjacent to the lake. The structures include closely spaced calcite-impregnated columns, tubes, and other configurations with subcylindrical to bizarre cross sections and predominantly vertical orientation in coarse, barely coherent pumice sands along the south shore of the lake. Many structures terminate upward in extensive calcareous layers of caliche and tufa. Locally they enter the bases of tufa mounds and towers. A common form superficially resembles root casts and animal burrows except that branching is mostly up instead of down. Similar defluidization structures in ancient sedimentary rocks have been mistakenly interpreted as fossil burrows.  相似文献

The series of precisely spaced, sharp spectral lines that form an optical frequency comb is enabling unprecedented measurement capabilities and new applications in a wide range of topics that include precision spectroscopy, atomic clocks, ultracold gases, and molecular fingerprinting. A new optical frequency comb generation principle has emerged that uses parametric frequency conversion in high resonance quality factor (Q) microresonators. This approach provides access to high repetition rates in the range of 10 to 1000 gigahertz through compact, chip-scale integration, permitting an increased number of comb applications, such as in astronomy, microwave photonics, or telecommunications. We review this emerging area and discuss opportunities that it presents for novel technologies as well as for fundamental science.  相似文献

Computer simulations of flow and sediment transport in the swash zone on a beach demonstrate that a model that couples local flow acceleration and alongshore surface gradient is sufficient to produce uniformly spaced beach cusps. The characteristics of the simulated cusps and the conditions under which they form are in reasonable agreement with observations of natural cusps. The self-organization mechanism in the model is incompatible with an accepted model in which standing alongshore waves drive the regular pattern of erosion and deposition that gives rise to beach cusps. Because the models make similar predictions, it is concluded that currently available observational data are insufficient for discrimination between them.  相似文献

Little is known about the neuronal mechanisms that subserve long-term memory persistence in the brain. The components of the remodeled synaptic machinery, and how they sustain the new synaptic or cellwide configuration over time, are yet to be elucidated. In the rat cortex, long-term associative memories vanished rapidly after local application of an inhibitor of the protein kinase C isoform, protein kinase M zeta (PKMzeta). The effect was observed for at least several weeks after encoding and may be irreversible. In the neocortex, which is assumed to be the repository of multiple types of long-term memory, persistence of memory is thus dependent on ongoing activity of a protein kinase long after that memory is considered to have consolidated into a long-term stable form.  相似文献

Until recently, it has been impossible to approach learning with the techniques of cell biology. During the past several years, elementary forms of learning have been analyzed in higher invertebrates. Their nervous systems allow the experimental study of behavioral, neurophysiological, morphological, biochemical, and genetic components of the functional (plastic) changes underlying learning. In this review, we focus primarily on short-term sensitization of the gill and siphon reflex in the marine mollusk, Aplysia californica. Analyses of this form of learning provide direct evidence that protein phosphorylation dependent on cyclic adenosine monophosphate can modulate synaptic action. These studies also suggest how the molecular mechanisms for this short-term form of synaptic plasticity can be extended to explain both long-term memory and classical conditioning.  相似文献

Poor memory after brain damage is usually considered to be a result of information being lost or rendered inaccessible. It is assumed that such memory impairment must be due to the incorrect interpretation of previously encountered information as being novel. In object recognition memory experiments with rats, we found that memory impairment can take the opposite form: a tendency to treat novel experiences as familiar. This impairment could be rescued with the use of a visual-restriction procedure that reduces interference. Such a pattern of data can be explained in terms of a recent representational-hierarchical view of cognition.  相似文献

Two-dimensional (2D) vibrational echo spectroscopy has previously been applied to structural determination of small peptides. Here we extend the technique to a more complex, biologically important system: the homodimeric transmembrane dimer from the α chain of the integrin α(IIb)β(3). We prepared micelle suspensions of the pair of 30-residue chains that span the membrane in the native structure, with varying levels of heavy ((13)C=(18)O) isotopes substituted in the backbone of the central 10th through 20th positions. The constraints derived from vibrational coupling of the precisely spaced heavy residues led to determination of an optimized structure from a range of model candidates: Glycine residues at the 12th, 15th, and 16th positions form a tertiary contact in parallel right-handed helix dimers with crossing angles of -58° ± 9° and interhelical distances of 7.7 ± 0.5 angstroms. The frequency correlation established the dynamical model used in the analysis, and it indicated the absence of mobile water associated with labeled residues. Delocalization of vibrational excitations between the helices was also quantitatively established.  相似文献
