采用RACE技术克隆获得中国明对虾(Fenneropenaeus chinensis)谷氨酸脱氢酶GDH基因(Fc GDH)。Fc GDH基因全长1779 bp,包括1个1659 bp的开放阅读框(ORF),编码552个氨基酸,预测分子量大小为61.3 k Da,理论等电点为6.54。同源性分析显示,Fc GDH氨基酸序列与其他动物高度保守,其中,与凡纳滨对虾最为相似,高达98%,其次为中华绒螯蟹,为89%。系统进化树分析显示,Fc GDH氨基酸序列与凡纳滨对虾GDH聚为一支,之后依次为:中华绒螯蟹、黑腹果蝇、埃及按蚊。组织表达分析发现,Fc GDH基因在肌肉、鳃、肝胰腺、胃、肠、淋巴和血淋巴中均有表达,其中,肌肉中表达量最高。氨氮胁迫后,Fc GDH基因在肌肉和肝胰腺组织中变化显著,在胁迫后期,Fc GDH基因表达量均上调,且与对照组相比差异显著(P0.05),说明Fc GDH基因在氨氮解毒代谢过程中发挥了重要作用。  相似文献

为发掘达氏鳇的功能基因,采用新一代高通量测序技术对达氏鳇肌肉组织转录组进行测序。结果获得原始数据14 447 211 200 bp,拼接获得了55 531条单基因序列(unigene),长度范围300~32 613 bp,平均长度941 bp。利用生物信息学方法对unigene进行了非冗余蛋白质数据库(Nr)相似搜索,此外还进行了GO的功能注释和KEGG代谢通路分析。结果一共有20 735条unigene(37.34%)与Nr数据库中的己知基因同源;根据GO功能可分为生物过程、细胞组分和分子功能3大类56分支;依据KEGG代谢通路分析可以分成290类。  相似文献

草鱼样品体重(1543±213)g。采用离体消化培养和茚三酮比色方法,以草鱼前肠、中肠、后肠和肝胰脏的粗酶液作为酶源,对鱼粉、豆粕、菜粕和棉粕进行酶解,并测定酶解14h内,豆粕酶解液中氨基酸生成量随时间的变化,以及0至4h,内4种饲料蛋白质酶解液中氨基酸的生成量。结果表明,(1)在0至4h内,4种饲料酶解液OD570随时间变化的线性关系较好。(2)对于同种蛋白质饲料原料,以中肠组织粗酶液的酶解速度最大,其余依次为前肠、后肠及肝胰脏粗酶液。表明草鱼中肠对饲料蛋白质的酶解能力强于前肠和后肠,同时,肠道酶液的酶解速度均大于肝胰脏。(3)对于4种不同的蛋白质饲料原料,以鱼粉酶解氨基酸生成速度最大,其他依次为豆粕、菜粕、棉粕。(4)草鱼肝胰脏粗酶液对豆粕的酶解氨基酸生成速度最大,表明草鱼对豆粕进行酶解消化的能力较强。实验证实,饲料原料种类的差异和饲料蛋白质组成与性质的差异使其酶解氨基酸生成量和生成速度有差异。  相似文献

采用垂直板聚丙稀胺凝胶电泳方法 ,对鲤鱼 (LyprinusCarpio)的肠道组织、肠内微生物谷氨酸脱氢酶 (GDH)同工酶进行分析 ,并对GDH活性进行测定 ,结果显示 ,摄食添加尿素饲料的鲤鱼肠组织及肠道微生物GDH同工酶谱与对照组有明显不同。试验组多一条酶谱且酶带染色深 ,表明鲤鱼在一定程度上能利用尿素合成 -谷氨酸 ,其合成可以通过肠组织和肠内微生物两条途径进行。而对GDH酶活性测定表明 ,试验组和对照组的肠组织和肠内微生物之间的酶活性均无显著性差异  相似文献

达氏鳇(Huso dauricus)是黑龙江流域土著鲟,近几十年来野生资源急剧下降,被确定为濒危物种之一。本研究采用线粒体DNA的Cyt b基因和D-loop区域的多态性信息评估了黑龙江抚远段野生群体、北京房山国家级鲟鱼原种场的保种群体及浙江衢州国家级鲟鱼良种场的繁殖群体等3个达氏鳇群体的遗传多样性水平。所有测试个体在Cyt b基因位点的核苷酸水平上没有检测到多样性,均具有相同的Cyt b单倍型,而在D-loop区域中发现了9种单倍型,对于D-loop区,单倍型多样性(H_d)达到0.593,但核苷酸多样性(π)仅为0.00213。在8尾野生个体中检测到6种D-loop单倍型,2个养殖群体共计58尾个体共计检出5种D-loop单倍型个体。分析结果显示:野生达氏鳇群体遗传多样性极低,历史上可能经历过严重的遗传瓶颈,同时达氏鳇人工繁殖过程中每批次可能只有极少个体参与了繁殖。此外,基于Cyt b基因部分序列的分析结果提示,达氏鳇与其他太平洋鲟类的亲缘关系较近,而与欧鳇(Huso huso)关系较远,传统上鳇属(Huso)的分类地位得不到有效的分子生物学数据支持。  相似文献

为了确定达氏鳇(Huso dauricus)幼鱼适宜的投饵率,选取同一种饲料,设置的5个投饵率水平(分别为体重的0.5%、1.0%、1.5%、2.0%、2.5%),在水温17~20℃、溶氧6.2mg/L以上的环境下,对达氏鳇幼鱼生长进行为期30d的试验。应用SPSS软件对各实验组相应的数据进行分析。结果表明,5个投饵率水平的达氏鳇幼鱼相对增重率和特定生长率先升高、后下降、再升高、再下降,而饲料转化率则逐渐下降;达氏鳇幼鱼在该条件下的适宜投饵率为1.0%。  相似文献

实验克隆了中华鲟(Acipenser sinensis)热休克蛋白hsp30基因cDNA的全长、分析了其分子结构与特征,并研究了其在高温胁迫下的表达水平。结果显示,中华鲟hsp30基因cDNA序列全长为1 037 bp,其中开放阅读框(ORF)636 bp,5′端非编码区(5′UTR)38 bp,3′端非编码区(3′UTR)363 bp,共编码211个氨基酸。氨基酸多序列比对发现含有一个保守的α晶状体结构;系统进化分析显示,中华鲟HSP30与鱼类HSP30聚为一支,与小体鲟HSP30氨基酸序列相似性最高,为79%。荧光定量PCR结果表明,中华鲟hsp30基因在皮肤中的表达量最高,肝脏次之,在肠中的表达量最低。高温胁迫后,心脏、脾脏、肾脏和皮肤中hsp30基因表达量均显著增加,表明这些器官在中华鲟应对高温胁迫中可能起着重要作用。  相似文献

用离体消化方法研究了凡纳滨对虾胃、肝胰脏和肠道的粗酶液对鱼粉、豆粕、菜粕和花生粕酶解8 h的体外消化率及0~4 h 4种饲料蛋白质酶解液中氨基酸的生成量.试验结果表明,凡纳滨对虾胃、肝胰脏和肠道对鱼粉、豆粕、菜粕、花生粕的干物质和粗蛋白质总消化率分别为36.50%、50.78%、42.02%、39%和46.28%、56.45%、43.28%、60.40%;0~4 h,4种饲料酶解液的氨基酸生成量随时间变化的线性关系较好.对于同一原料,肝胰脏粗酶液的酶解速度最大,其他依次为肠道、胃粗酶液,表明肝胰脏对饲料蛋白质的酶解能力强于胃和肠;对于不同原料,以花生粕酶解氨基酸生成速度最大,其他依次为鱼粉、豆粕、菜粕,表明凡纳滨对虾对花生粕酶解消化的能力较强.饲料原料种类的差异和饲料蛋白质组成与性质的差异使其酶解氨基酸生成量和生成速度有差异.  相似文献

Apparent digestibility coefficients (ADCs) of dry matter, crude protein, lipid, energy, phosphorus and amino acids in white fish meal, soybean meal, rapeseed meal and peanut meal were determined for Synechogobius hasta (28.65 ± 0.43 g, mean ± SD), using a reference diet with chromic oxide indicator and test diets that contained 70% reference diet, by weight, and 30% of feed ingredients. The juvenile S. hasta were held in 300 L tanks at a density of 30 fish per tank respectively. Faeces were collected from three replicate groups of fish by siphoning. The ADCs of dry matter and energy were the highest in white fish meal and the lowest in rapeseed meal ( P <0.05). Crude protein ADC was the lowest in rapeseed meal ( P <0.05) and showed no significant differences among other treatments ( P >0.05). The highest phosphorus ADC was observed in white fish meal and differences were not marked for other treatments ( P >0.05). Lipid ADC were above 90% and showed no significant differences among the treatments ( P >0.05). Amino acid availability values for the test ingredients followed similar trend to values of protein digestibility. All amino acids were more available from fish meal than from plant protein ingredients. Among three plant meals, the availability of amino acids was higher in peanut meal and lower in rapeseed meal.  相似文献

摘要:通过8周网箱实验评价了利用豆粕、菜粕和棉粕作为苏氏圆腹鱼芒 (Pangasius sutchi)饲料中鱼粉替代蛋白源的潜力。配制了7种等氮、等能饲料,其中基础饲料含45%鱼粉,按等量蛋白替代的原则,在其余6种饲料中分别添加31%和46%豆粕替代基础饲料中鱼粉的50%和75%,或添加20%和40%菜粕替代基础饲料中鱼粉的25%和50%,或添加19%和39%棉粕替代基础饲料中鱼粉的25%和50%。实验中所用的苏氏圆腹鱼芒初始体重为11.3 g。实验结果表明:添加豆粕将饲料中鱼粉含量从45%降低到23%,添加菜粕或棉粕将鱼粉含量降低到34%,对鱼成活率、摄食、鱼体增重、特定生长率(SGR)、饲料系数(FCR)、饲料蛋白储积率、脏体指数和红血细胞比积(Hct)未产生显著不良影响。添加豆粕将鱼粉含量降低到11%导致鱼摄食、鱼体增重和SGR下降,添加菜粕将鱼粉含量降低到23%导致FCR和鱼体能量储积率下降,添加棉粕将鱼粉含量降低到23%导致鱼体增重、SGR和Hct明显下降。上述结果显示,可通过添加豆粕(31%)将苏氏圆腹鱼芒 鱼种饲料中鱼粉含量降低到23%,或通过添加菜粕(20%)和棉粕(19%)将饲料中鱼粉含量降低到34%。  相似文献

以2%棉粕酶解蛋白肽(以下简称蛋白肽)分别等质量替代基础日粮中2%鱼粉和2%植物蛋白原料(1.0%豆粕+0.5%菜粕+0.5%棉粕),考察蛋白肽对(48.56±1.39)g建鲤(Cyprinus carpio var.Jian)生长性能、血清及肠道相关酶活的影响。结果表明:蛋白肽替代鱼粉或植物蛋白后试验组增重率和特定生长率均显著性高于对照组(P<0.05),同时饵料系数显著降低(P<0.05);蛋白肽可降低脏体比和肝体比,并显著性提高鱼体粗蛋白含量(P<0.05);同时可提高试验组建鲤血清溶菌酶、碱性磷酸酶和肠道中蛋白酶的活性。  相似文献

Apparent digestibility coefficients of dry matter, crude protein, lipid and energy, and amino acids availability in white fish meal, brown fish meal, meat meal, fermented soybean meal, soybean meal and rapeseed meal were determined for loach (Misgurnus anguillicaudatus) (12.05 ± 0.21 g), using a reference diet with 5 g kg?1 chromic oxide and test diets that contained 700 g kg?1 reference diet, by weight, and 300 g kg?1 of the test feed ingredients. The juvenile loach was held in 300‐l tanks at a density of 30 fish per tank. White fish meal, brown fish meal, meat meal and fermented soybean meal had highest apparent digestibility coefficients (ADCs) of dry matter, crude protein and gross energy among ingredients tested, ranged from 50.4% to 60.9% for dry matter, from 64.6% to 88.4% for crude protein and from 57.9% to 79.0% for gross energy. The apparent digestibility coefficients of dry matter ranged from 61.0% to 66.9% for animal products and 50.4% to 60.7% for plant products. For crude protein, apparent digestibility coefficients of protein exceeding 80% were observed for white fish meal, brown fish meal, meat meal and fermented soybean meal, and the apparent digestibility coefficients of protein in rapeseed meal were the lowest among all the treatments. Lipids from both animal and plant feedstuffs were poorly digested by loach, ranging from 64.0% to 77.6%. The apparent digestibility coefficients of energy were similar to those of dry matter and protein, and the highest and lowest ADCs of energy were found in WFM and RM, respectively. The loach used dietary phosphorus from the animal feedstuffs more efficiently than from plant feedstuffs (soybean meal and rapeseed meal), with ADC‐values ranging from 42.3% to 53.1% and from 25.1% to 32.7%, respectively. For the animal products, the availabilities of amino acids in white fish meal and brown fish meal were higher than that in meat meal, expect for Met, Asp, Pro, Gly, and Cys. Among all the plant products, the availabilities of amino acids in fermented soybean meal were higher than in soybean meal and rapeseed meal, and thus had a greater potential to be used as a dietary replacement of fish meal in loach diets.  相似文献

The purpose of the experiment was to evaluate effects of partial replacement of fish meal with soybean meal on digestibility in Atlantic cod and effects of biotechnological processing of soybean meal. Other objectives were to find out if apparent digestibility in cod was affected by age, temperature and/or faecal sampling methods, and to map digestion and absorption patterns along the intestinal tract for different age classes and diets. A control diet with fish meal as the main protein source (FM), a diet with 24% of crude protein from conventional extracted soybean meal (SBM), and a diet with 24% of crude protein from a bioprocessed soybean meal (BP-SBM) were used. Yttrium oxide was added as an inert marker for digestibility determination. The diets were fed to duplicate groups of 534 g (1-year-old) and 1750 g (2-year-old) cod in seawater. Faeces were stripped from the distal-most section (distal chamber) of the intestine after 48 and 84 days of feeding. In addition, intestinal contents were removed by dissection after 84 days. The dissected intestine was divided into 5 sections and contents were analysed separately. Dietary soy did not alter the viscosity of the digesta. The replacement of fish meal protein with soybean protein reduced the apparent digestibility of crude protein, fat, energy and almost all amino acids, independent of type of soybean meal processing. Larger fish had higher digestibility of starch, ash and several amino acids than smaller fish. A temperature decrease from 9.1 °C at feeding day 48 to 6.8 °C at day 84 resulted in lower digestibility of starch, ash and several amino acids. Absorption of amino acids was quantitatively terminated in the distal part of the mid intestine when feeding the soy-containing diets, but continued into the distal chamber when feeding the FM diet. Absorption of fat was completed in the proximal part of the mid intestine when feeding the soy-containing diets, but continued into the middle part of the mid intestine when feeding the FM diet. Soy did not affect absorption pattern of starch along the intestinal tract. Faecal sampling by dissection resulted in higher apparent digestibility of macronutrients than stripping in the 2-year-old cod, and higher apparent digestibility of protein in the 1-year olds.  相似文献

An 8‐week feeding trial was conducted to investigate the effects of dietary rapeseed meal (RM) and glutathione supplementation (400 mg kg?1) on growth and intestinal microbiota of grass carp (Ctenopharyngodon idellus). Dietary 450 g kg?1 RM decreased growth and feed intake, while increased the activities of serum alkaline phosphatase (ALP), aspartate aminotransferase (AST) and alanine transaminase (ALT). The hepatic damages were also observed the significant lower condition factor (CF), hepatosomatic index (HSI) and the symbol of parenchymal damage. Glutathione supplementation increased weight gain and feed intake while significantly decreased the three serum biochemistry parameters compared with the RM group. In addition, HSI, CF and pathology reports revealed that glutathione supplementation alleviated the damage caused by RM on the fish liver. In addition, glutathione had a positive effect on the intestinal digestive capacity. PCR‐denaturing gradient gel electrophoresis (DGGE) analysis indicated a higher similarity between dietary glutathione supplementation and control group. In conclusion, we demonstrated dietary 450 g kg?1 rapeseed meal lead to negative health outcomes, while glutathione supplementation neutralize the adverse effect of rapeseed meal presented in aqua feed through alleviating the damage factors on the fish liver.  相似文献

分析商品南极磷虾粉、自制酶解磷虾粉的主要营养成分(氨基酸和脂肪酸)组成,同时采用生化方法比较了5种鲽形目鱼肉蛋白的氨基酸组成。以5种鲽形目鱼肉蛋白为参比蛋白,利用必需氨基酸指数、氨基酸比值系数分和关联度分析法评价南极磷虾粉作为鲽形目鱼饲料蛋白源的营养价值,同时对2种磷虾粉的氟含量进行了分析。结果显示,鲽形目鱼肉(干样)中含有16种常见氨基酸,其中,7种为人体必需氨基酸,4种为呈味氨基酸(总量分别为28.52%–38.03%、25.26%–33.56%);5种鲽形目鱼氨基酸组成均符合 FAO/WHO 的理想模式。南极磷虾粉和酶解磷虾粉的粗蛋白分别为60.84%和68.60%,粗脂肪分别为12.08%和10.79%,达到了规定鱼粉的一级品甚至特级品的指标。酶解处理后蛋白含量显著升高(P<0.05),脂肪含量显著下降(P<0.05),灰分含量无差异(P>0.05)。从必需氨基酸指数(EAAI)来看,EAAI 均大于0.95,磷虾粉的必需氨基酸与鲽形目鱼的必需氨基酸拟合度较高。从氨基酸比值系数分(SRC)和关联度分析法来看,2种磷虾粉对于鲽形目鱼的必需氨基酸平衡性较好,相关系数与鱼粉相差不大。氟含量方面,酶解后磷虾粉的氟含量[(331.21±6.70) mg/kg]显著降低(P<0.05),低于欧盟标准(350 mg/kg)。综合来看,2种磷虾粉氨基酸平衡性较好、营养价值较高,是优质蛋白源。  相似文献

为研究复合蛋白源替代鱼粉对杂交鳢生长、体组成、抗氧化与生化指标的影响,在基础饲料中用复合蛋白源(由棉粕+菜粕+蚕蛹+桑叶发酵而成)分别替代饲料中0%、12%、24%、36%和48%的鱼粉,配制成5种等氮等脂(含粗蛋白CP 42%,粗脂肪EE 8.5%)的实验饲料,在室内循环水族箱内饲喂杂交鳢(初始体质量22 g)8周。结果显示,当复合蛋白源替代鱼粉水平大于12%时,杂交鳢的终末体质量(FBW)和特定生长率(SGR)显著降低,在48%替代水平达到最低。36%和48%替代水平蛋白质效率(PER)显著降低,而饲料系数(FCR)显著升高。各实验组摄食率(FR)无显著差异。替代水平组全鱼粗脂肪含量以及脏体比(VSI)和肠脂比(IPF)显著降低,而水分含量显著增加。随替代水平的增加,胰蛋白酶活性显著降低,而肠脂肪酶活性显著升高。替代水平24%以上肝脏谷丙转氨酶(GPT)、过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性显著升高。替代水平12%以上血清中总胆固醇(CHO)含量显著降低,而替代水平36%以上血糖(GLU)含量会显著降低。各实验组血清甘油三酯(TC)、总蛋白(TP)和白蛋白(ALB)含量无显著变化。研究表明,复合蛋白源替代12%鱼粉不会影响杂交鳢的生长,而高水平替代会抑制杂交鳢的生长,但可以改善杂交鳢机体糖脂代谢,提高机体的抗氧化能力。  相似文献

The effect of dietary protein level on the apparent digestibility coefficient (ADC) of meat and bone meal and rapeseed meal for Nile tilapia Oreochromis niloticus L. was examined. Three reference diets were formulated to contain 240 (RD24), 350 (RD35) and 450 (RD45) g kg?1 crude protein, and chromic oxide was added at 10 g kg?1 in the reference diets as an inert indicator. Six test diets were formulated by mixing one of the reference diets with each test ingredient at a ratio of 70:30. Fish (initial body weight 101.0 ± 0.6 g) were fed reference diets for 3 days and then fed test diets in the following 4 days cyclically for 4 weeks. Faeces of fish fed each reference diet or test diet was collected from the third day of each diet conversion. Dietary protein level significantly affected the ADC of protein and energy of reference diets, test diets and test ingredients. The ADC of protein and energy of meat and bone meal and rapeseed meal was the highest when these ingredients were mixed with diet RD35. The scope of variation in the ADC of protein and energy of meat and bone meal was 8% (from 69.9% to 78.3%) and 8% (from 70.2% to 78.0%), and the scope of variation in the ADC of protein and energy of rapeseed meal was 4% (from 79.8% to 83.3%) and 5% (from 56.0% to 60.7%) when the dietary protein level was increased from 300 to 480 g kg?1. This indicates that the ADC of protein and energy of meat and bone meal and rapeseed meal for Nile tilapia varied considerably at different dietary protein levels.  相似文献
