Olive tree (Olea europaea L.) is an economically important woody fruit crop widely distributed in the Mediterranean regions. In this work the genome size of six Portuguese cultivars of olive (O. europaea ssp. europaea var. europaea) and wild olive (O. europaea spp. europaea var. sylvestris) was estimated for the first time. The nuclear DNA content of O. europaea cultivars ranged between 2.90 ± 0.020 pg/2C and 3.07 ± 0.018 pg/2C and the genome size of wild olive was estimated as 3.19 ± 0.047 pg/2C DNA. These results suggest a low intraspecific variation at least among the studied cultivars and between them and wild olive. This is not in accordance with previous results in some Italian cultivars where high genome size heterogeneity was found. The methodology presented here seems appropriate for genome size estimations within this genus and opens good perspectives for a large screening of estimation of nuclear DNA content among O. europaea cultivars and Olea species that could clarify this issue. 相似文献

The olive belongs to Olea, a complex genus and to a species with six subspecies. Subsp. europaea includes both the cultivated olive and the oleaster, the wild ancestor of the olive. Little is known on the phenotype of
fruits from subsp. cuspidata. We aimed to compare europaea and cuspidata trees for oil content and composition and to verify whether natural hybrids may exist between the two subspecies. Specimens
were from Kenya and putative natural hybrids between cuspidata and europaea were from Stellenbosch (South Africa). Cultivar and oleaster trees were sampled in France (continental and Corsica), Italy
(Continental and Sardinia) and Tunisia. We have examined the fruit (drupe) structure between subsp. cuspidata and europaea and extracted oil from the pulp, seed and total drupe. Comparison for oil content was made between the cultivated olive and
the oleaster and some cuspidata trees from Kenya. A few of cuspidata individuals from Botanical gardens do not enable phenotyping for oil content and composition. Oil composition analyses were
performed for the main fatty acids to compare the three taxa. We used microsatellite markers at 11 loci to compute genetic
distances between cuspidata, oleaster and cultivar trees and to reveal eventual hybrids. The SSR polymorphisms were huge between the two subspecies and
they revealed that putative hybrids were true hybrids sampled around olive orchards. The whole comparison of oil content shows
that olive cultivars display higher oil content than the subsp. cuspidata and that the oleaster trees are intermediate, whereas for oil composition of the drupe, cuspidata from Kenya shows less oleic acid than europaea. However, the cuspidata trees show seed oil composition similarities with the other two taxa. The discussion deals with possible reasons to explain
the differences and of the possible uses of these taxa for breeding both the olive and brown olive.

Common bean (Phaseolus vulgaris) is native to the Americas, and Rhizobium etli is the dominant microsymbiont in both the Mesoamerican and the Andean centers of genetic diversification. Wild common beans are not found in Brazil, although the legume has been cropped in the country throughout time and all but one of the rhizobial species that nodulate it (Rhizobium gallicum) have been broadly detected in Brazilian soils. However, the majority of the effective rhizobial strains isolated so far from field-grown plants belong to R. tropici. This study describes the analysis of symbiotic and non-symbiotic genes of 15 effective R. tropici strains, isolated from four geographically distant regions in Brazil. With RFLP-PCR of the 16S and 23S rRNA genes and sequence analysis of 16S rRNA, two clusters were observed, one related to R. tropici type A and another to type B strains. Diversity in ribosomal genes was high, indicating that type A strains might represent a new species. High intraspecies diversity was also observed in the rep-PCR analysis with BOX, ERIC and REP primers. However, in the RFLP-PCR analysis of nifH and nodC genes, all R. tropici showed unique combinations of profiles, which might reflect an evolutionary strategy to maximize N2 fixation. 相似文献

为探讨紫苏的耐镉生理机制, 通过水培试验, 研究了镉离子浓度为0 mg·L-1(对照)、2.0 mg·L-1、5.0 mg·L-1、10.0 mg·L-1 条件下紫苏的生长、镉富集作用及叶片的保护酶活性。结果表明, 2.0 mg·L-1、5.0 mg·L-1、10.0 mg·L-1 镉胁迫下, 紫苏单株生物量比对照分别减少28.4%、42.4%、66.0%, 不同处理间均存在极显著差异;3 个镉离浓度下紫苏的镉富集量依次为462.4 μg·株-1、705.0 μg·株-1、609.7 μg·株-1, 富集系数依次为91.9、70.0、51.1, 具备镉超积累能力。此外, SOD、POD、CAT 酶活测定结果显示, 随镉离子浓度升高, 紫苏叶片的SOD 活性和MDA 含量显著升高, POD 和CAT 活性显著下降, 破坏了体内保护酶系统的平衡, 抑制紫苏生长。 相似文献

The enormous losses suffered by the European elms during recent Dutch elm disease outbreaks led to concern over the conservation of elm genetic resources, and the subsequent establishment of a series of ex situ collections. However, as ex situ collections are inevitably finite in size, some consideration needs to be given to selecting which samples to include in them. To contribute towards this process for European ex situ elm collections we have undertaken genetic studies on a Europe-wide sample of 535 individuals. A major aim has been to use genetic markers to clarify the identification of samples to ensure that the ex situ collections contain a representative spread of taxonomic diversity. This is important given the paucity of mature elms in the landscape due to Dutch elm disease. The lack of mature material (critical for identification) compounds identification problems in what was already a taxonomically difficult group. Our data (derived from random amplified polymorphic DNA and inter-simple sequence repeats) have provided a useful supplement to morphology in undertaking such sample identifications. The molecular data served to highlight mis-identified samples and led to extensive revisions of sample identities within individual countries. Our results were less useful in detecting regional intra-specific genetic structure, and do not provide sufficient information for prioritising within-species sample selections. 相似文献

Eleven cadmium-tolerant bacterial strains were isolated from the root zone of Indian mustard (Brassica juncea L. Czern.) seedlings grown in Cd-supplemented soils as well as sewage sludge and mining waste highly contaminated with Cd. The bacteria also showed increased tolerance to other metals including Zn, Cu, Ni and Co. The isolated strains included Variovorax paradoxus, Rhodococcus sp. and Flavobacterium sp., and were capable of stimulating root elongation of B. juncea seedlings either in the presence or absence of toxic Cd concentrations. Some of the strains produced indoles or siderophores, but none possessed C2H2-reduction activity. All the strains, except Flavobacterium sp. strain 5P-3, contained the enzyme 1-aminocyclopropane-1-carboxylate (ACC) deaminase, which hydrolyses ACC (the immediate precursor of plant hormone ethylene) to NH3 and α-ketobutyrate. V. paradoxus utilized ACC as a sole source of N or energy. A positive correlation between the in vitro ACC deaminase activity of the bacteria and their stimulating effect on root elongation suggested that utilization of ACC is an important bacterial trait determining root growth promotion. The isolated bacteria offer promise as inoculants to improve growth of the metal accumulating plant B. juncea in the presence of toxic Cd concentrations and for the development of plant-inoculant systems useful for phytoremediation of polluted soils. 相似文献

Twenty-eight Rhizobium strains were isolated from the root nodules of faba bean (Vicia faba L.) collected from 11 governorates in Egypt. A majority of these strains (57%) were identified as Rhizobium leguminosarum bv. viciae (Rlv) based on analysis of a nodC gene fragment amplified using specific primers for these faba bean symbionts. The strains were characterized using a polyphasic approach, including nodulation pattern, tolerance to environmental stresses, and genetic diversity based on amplified ribosomal DNA-restriction analysis (ARDRA) of both 16S and 23S rDNA. Analysis of tolerance to environmental stresses revealed that some of these strains can survive in the presence of 1% NaCl and a majority of them survived well at 37 °C. ARDRA indicated that the strains could be divided into six 16S rDNA genotypes and five 23S rDNA genotypes. Sequence analysis of 16S rDNA indicated that 57% were Rlv, two strains were Rhizobium etli, one strain was taxonomically related to Rhizobium rubi, and a group of strains were most closely related to Sinorhizobium meliloti. Results of these studies indicate that genetically diverse rhizobial strains are capable of forming N2-fixing symbiotic associations with faba bean and PCR done using nodC primers allows for the rapid identification of V. faba symbionts. 相似文献

Burr medic (Medicago polymorpha L.) is an annual pasture legume that is widely distributed in southern Australian farming systems. Burr medic is nodulated by rhizobia (Sinorhizobium meliloti and Sinorhizobium medicae) that reside in many Australian soils, but the symbioses that develop are often sub-optimal in their rate of N2 fixation. We attempted to identify burr medic lines, which are able to form effective symbioses with the naturalised Sinorhizobium in Australian field soils, as potential parents for a breeding program. There were three glasshouse experiments. Initially, 222 lines (including the M. polymorpha cvv. Santiago, Serena and Circle Valley) were inoculated with extracts of two soils that had been collected near Waikerie (soil S109) and Lochiel (soil S142) in South Australia. These soils were used because they contained numerically large communities of naturalised Sinorhizobium spp. that produced sub-optimal rates of N2 fixation with cv. Santiago. None of the 222 lines of burr medic were able to form an effective symbiosis with the rhizobia from soil S109. However, when nodulated by the rhizobia from soil S142, some lines (e.g. SA8194) formed a very effective symbiosis, producing up to double the shoot dry matter (DM) of Santiago and eight times the DM of uninoculated plants. Seven promising lines were selected for further testing (with extracts of nine soils). Subsequently, two lines (SA20056 and SA8194) were selected and their symbiotic performance compared with that of Santiago, using extracts from 28 soils. While soil treatment had a major effect on mean shoot DM (soil N103=120 mg, soil N105=17 mg), the three medic lines performed similarly. Santiago, SA20056 and SA8914 all formed ineffective symbioses with the rhizobia in at least half of the 28 soils, even though >95% of the plants were nodulated. These experiments confirm that ineffective symbioses are common between burr medics and the rhizobia that have become naturalised in many Australian soils. Although some lines of burr medic were identified that were able to form more effective symbioses with the rhizobia in individual soils, none were able to form effective symbioses with a wide range of soil rhizobia. If a plant breeding approach is to be used to improve symbiotic performance of burr medic we propose that its hybridisation with other medic species, that have less specific rhizobial needs, will be required. 相似文献

The Emilia region (Northern Italy) is characterised by the occurrence of microclimates that permit olive growing. The presence
of the species, albeit sporadic, in these territories for several centuries as a fruit crop is well documented, by both archaeological
and written testimony, and by a large number of plants well over a century old, located in particular sites, favourable for
growth and development of the tree.
Olive genetic diversity was studied using RAPD and SSR techniques, on plants growing in the Emilia territory (Reggio Emilia
and Parma provinces). For genotype identification comparisons were made with 8 cultivars, some of which from Central Italy.
Screening was obtained analysing patterns produced by 20 RAPD primers and 3 SSR primers, developed by other authors; the primers
and we were able to discriminate olive cultivars with a sufficient degree of reliability. The dendrograms obtained from the
analysis show the genetic relationship among accessions present in the Parma-Reggio Emilia district. Our results demonstrated
the reliability of RAPDs and SSRs to identify all studied olive cultivars and to reveal the degree of their relatedness to
each other. The analysis also reveals the presence of an interesting amount of genetic diversity among the studied individuals. 相似文献

为明确西藏芥菜型油菜的性状演化与分支分类问题, 本文以67 份西藏芥菜型油菜为试验对象, 52 个形态学性状为依据, 白花菜科植物醉蝶花为外类群, 应用最大同步法, 对其进行了研究。结果表明: 芥菜型油菜的基生叶、伸长茎叶、薹茎叶和花器的性状, 均既有同期演化的, 也有非同期演化的, 有各自独特的演化规则; 西藏芥菜型油菜明显分为野生种和栽培种2 大类群, 无论是野生类群还是栽培类群中, 往往是来自于同一地区或气候相似区的材料聚在一起。表明西藏芥菜型油菜种质资源在形态演化上的相似性与其原产地的地理、气候背景密切相关。 相似文献

Soil compaction is of great importance in agriculture, because its high levels may adversely affect plant growth and the environment. Since mechanical methods are not very efficient and economical, using biological methods to alleviate the stress of soil compaction on plant growth may be beneficial. The objectives of this study were to: (1) evaluate the effects of soil compaction on corn (Zea mays L.) growth, and (2) test the hypothesis that applying arbuscular mycorrhiza (AM) with different origins can partially or completely overcome the stressful effects of soil compaction on corn growth under unsterilized and sterilized conditions. Corn was planted in unsterilized and sterilized compacted soils, while treated with three species of AM including, Iranian Glomus mosseae, Iranian Glomus etunicatum, and Canadian Glomus mosseae, received from GINCO (Glomales in vitro Collection), Canada. Plant growth variables and soil resistance parameters were determined. AM significantly increased root fresh (maximum of 94% increase) and dry (maximum of 100% increase) weights in the compacted soil. AM with different origins may improve corn growth in compacted soils, though its effectiveness is related to the level of compaction and also to the interaction with other soil microorganisms. 相似文献

This study provides evidence that egg-parasitic nematophagous fungi, Pochonia chlamydosporia, Pochonia rubescens and Lecanicillium lecanii, can also reduce root colonisation and root damage by a fungal pathogen. Interactions of nematophagous fungi with the take-all fungus, Gaeumannomyces graminis var. tritici (Ggt), and their influence on severity of the root disease it causes were studied in laboratory and pot experiments. In Petri dish experiments the three nematophagous fungi reduced colonisation of barley roots by Ggt and also reduced necrotic symptoms. On the contrary, root colonisation by nematophagous fungi was unaffected by Ggt. In growth tube experiments, the three nematophagous fungi again reduced Ggt root colonisation and increased effective root length of barley seedlings. This was true for both simultaneous and sequential inoculation of nematophagous fungi versus Ggt. In the pot experiments the inoculum of the tested fungi in soil was applied in the same pot, as a mixture or in layers, or in coupled pots used for wheat grown with a split-root system. The nematophagous fungi P. chlamydosporia (isolate 4624) and L. lecanii (isolate 4629), mixed with Ggt or in split root systems with the pathogen, promoted growth of wheat (i.e. increased shoot weight), although no disease reduction was found. In split root systems, lower levels of peroxidase activity were found in seedlings inoculated with Ggt in combination with the nematophagous isolates 4624 and 4629 than when the take-all fungus was applied alone.Our results show that nematophagous fungi reduce root colonisation by Ggt, root damage and stress induced senescence in Ggt-inoculated plants. 相似文献

Slow-release fertilizers are gaining acceptance to increase fertilizer use efficiency and reduce environmental impact. The release of nitrogen from methylene urea, a common slow release N fertilizer, is controlled by microbial decomposition. An enzyme hydrolyzing slow-release nitrogen fertilizer, methylene urea, was purified from Rhizobium radiobacter (Agrobacterium tumefaciens) to homogeneity using a four-step purification procedure with an overall yield of 3%. The active enzyme has a molecular mass of approximately 180 kDa determined by size exclusion chromatography, and the SDS page of the purified protein indicated three subunits of different sizes (62, 34 and 32 kDa). The N-terminal amino acid sequence of the 62 kDa fragment indicates identity with urease subunits from Mycobacterium tuberculosis (73%) and Helicobacter pylori (71%). However, for the internal amino acid sequences of the 62 kDa fragment no matches with known proteins were found. Some internal peptides in the smaller subunits (32 and 34 kDa) are homologous to urease subunits and unknown proteins in Agrobacterium tumefaciens. Based on the kinetic properties, substrate selectivity, and inhibition characteristics, the novel enzyme (MUase) is an intracellular enzyme complex with urease activity. The enzymatic mechanism of methylene urea breakdown was studied using a novel LC-MS method for MU analysis, which indicates that all cold-water soluble nitrogen forms of methylene urea are subjected to hydrolysis, and the hydrolysis proceeds via methylurea, urea and other yet unidentified hydrolysis-products, suggesting that the isolated enzyme complex performs a multistep hydrolysis. The microbiological and molecular data is useful in determining the soil factors affecting the efficacy of methylene urea as a slow release fertilizer in agricultural production systems. 相似文献

Growth promoting effects of corn (Zea mays) bacterial isolates under greenhouse and field conditions
Fertilizer costs are a major component of corn production. The use of biofertilizers may be one way of reducing production costs. In this study we present isolation and identification of three plant growth promoting bacteria that were identified as Enterobacter cloacae (CR1), Pseudomonas putida (CR7) and Stenotrophomonas maltophilia (CR3). All bacterial strains produced IAA in the presence of 100 mg l−1 of tryptophan and antifungal metabolites to several soilborne pathogens. S. maltophilia and E. cloacae had broad spectrum activity against most Fusarium species. The only strain that was positive for nitrogen fixation was E. cloacae and it, and P. putida, were also positive for phosphate solubilization. These bacteria and the corn isolate Sphingobacterium canadense CR11, and known plant growth promoting bacterium Burkholderia phytofirmans E24 were used to inoculate corn seed to examine growth promotion of two lines of corn, varieties 39D82 and 39M27 under greenhouse conditions. When grown in sterilized sand varieties 39M27 and 39D82 showed significant increases in total dry weights of root and shoot of 10-20% and 13-28% and 17-32% and 21-31% respectively. Plants of the two varieties grown in soil collected from a corn field had respective increases in dry weights of root and shoot of 10-30% and 12-35% and 11-19% and 10-18%. In sand, a bacterial mixture was highly effective whereas in soil individual bacteria namely P. putida CR7 and E. cloacae CR1 gave the best results with 39M27 and 39D82 respectively. These isolates and another corn isolate, Azospirillum zeae N7, were tested in a sandy soil with a 55 and 110 kg ha−1 of nitrogen fertility at the Delhi research Station of Agriculture and Agri-Food Canada over two years. Although out of seven bacterial treatments, no treatment provided a statistically significant yield increase over control plots but S. canadense CR11 and A. zeae N7 provided statistically significant yield increase as compared to other bacteria. The 110 kg rate of nitrogen provided significant yield increase compared to the 55 kg rate in both years. 相似文献

Birch leaves and horse manure were used to determine the effects of food quality on growth and reproduction of laboratory-reared Lumbricus terrestris. Animals grew to maturity within 6 months but attained a significantly (p < 0.001) larger adult size with manure (6.17 g) versus leaves (4.20 g). Cocoon production by recently-mated adults maintained in isolation, fed with birch leaves or horse manure, resulted in 4.53 and 3.84 cocoons ind.−1 month−1 respectively, with an initial hatchability of 86%, falling to zero after 18 months. Re-mating of these known individuals permitted long term monitoring of reproductive output (to 30 months). For the whole experimental period, overall hatchability of the 2010 cocoons produced was 44.4%. Median incubation time of those cocoons that hatched within accepted norms (less than 5 months at 15 °C) was 103 days and was not influenced by adult food type. A proportion (35.5%) of cocoons took in excess of 12 months to hatch. Adult mortality was minimal (25%) during the long term experiment but abnormal cocoon production was recorded after 2 years. Overall results demonstrate that food quality can have a significant influence on somatic and reproductive production of L. terrestris and these data may aid construction of production models for this earthworm in ecosystems with contrasting food quality. 相似文献

Historical data on the distribution and status of the purple gallinule (Porphyrio porphyrio) in Portugal were collected by bibliographic research, from museum catalogues and from questionnaires to wetland managers, ornithologists and birdwatchers. The population suffered severe decline and range contraction at the end of the 19th century and most of the 20th century, due mainly to habitat loss and degradation and excessive hunting. However, the population has begun to recover in the last decade mainly through direct protection of the species and of the remaining habitat, together with creation of artificial wetlands and a reintroduction programme, and probably also due to immigration from Spain. The population size has increased about 10-fold and the range has expanded to most of its former area. The estimated Portuguese breeding population in 2002 was 49-67 pairs. Densities varied between 0.05 and 7 individuals/ha in recently colonized areas and stable (saturated) areas, respectively. Although the population is recovering, its small size, high fragmentation and several natural and anthropogenic threats lead us to consider it still a species of major concern in Portugal. 相似文献

In a declining oak forest, a thinning treatment was performed in August 2004, targeting trees belonging to two decline classes. The whole ectomycorrhizal (EM) community was dominated by the fungal symbionts Clavulina cinerea, Tomentella sublilacina and Russula spp. The potential activities of eight secreted enzymes, involved in mobilizing nutrients (N, P) from soil organic matter, were measured on these three EM types in winter and spring 2006 using multiwell microplate photometric and fluorogenic methods. The enzymatic activities recorded in winter were generally significantly higher than in spring. Most of the enzyme activities studied, and particularly phosphatase and β-glucosidase, changed according to both decline class and sylvicultural treatment. In spring, each anatomotype displayed different enzymatic profile according to the decline class. These results suggest that the potential enzymatic activity of ectomycorrhizae adapts to the changes resulting from the sylvicultural treatment and reacts to the anthropic disturbance by adjusting to the new resource structure. 相似文献

Mount Meiron Nature Reserve is the largest Mediterranean reserve in Israel. Since its establishment, goat grazing and woodcutting have been prohibited and dense oak maquis has developed by succession. The succession of vegetation poses the risk of losing species. The peony (Paeonia masculla) is a locally endangered species in Israel, growing only in a small population in the Mount Meiron Nature Reserve with only 5% of plants flowering. We sought a management practice that would increase the flowering percentage of the peonies and secure the future of its population. We found that flowering peonies grew mainly in locations with about 47% direct sun radiation, while most plants grew under heavy shade with only 27% direct radiation. Creation of small gaps increased the flowering to 15-20%. To ensure the future of the peonies in the Mount Meiron Nature Reserve, small gaps must be created—even if this conflicts with broader current management policies. 相似文献

为探讨减少盐碱地棉花铃脱落的途径, 田间条件下研究了土壤供应低氮(N 226.5 kg·hm-2)和高氮(N 346.5 kg·hm-2)条件下, 不打顶、打顶和打顶后涂抹生长素对棉花铃脱落和产量构成因素的影响。结果表明, 低氮和高氮水平下, 打顶、打顶后涂抹NAA 处理果枝落铃率都低于不打顶处理。打顶后涂抹浓度为0.3 mmol·L-1 的NAA 处理在低氮水平下的成铃率比传统打顶处理高1.3 个百分点, 而铃脱落率比传统打顶处理低0.8 个百分点, 皮棉产量比传统打顶处理高23.7%。高氮水平下, 与传统打顶处理相比, 打顶后涂抹浓度为3 mmol·L-1 的NAA 单株成铃率、铃脱落率、皮棉产量之间差异不显著。在同一处理中, 高氮水平的单株成铃数、单铃重和吐絮数均低于低氮水平, 而衣分无明显差异, 从而导致皮棉产量下降。上述结果说明, 打顶后涂抹一定浓度的NAA 可增加棉花单株现铃、成铃、吐絮数, 增加单铃重, 降低落铃率, 从而提高产量, 而过量施用氮肥会导致产量下降。 相似文献

室内采用正交试验设计进行4 因素多水平试验, 研究了温度、城市污泥和麦麸配比、饲养密度、幼虫体重对黄粉虫幼虫生长速度、死亡率以及幼虫重金属含量变化的影响, 探讨黄粉虫幼虫在城市污泥中的生长状况和对城市污泥重金属的积累作用。结果表明: 随着温度的升高, 幼虫生长率和死亡率都呈上升趋势, 饲养密度、污泥和麦麸配比对黄粉虫幼虫生长率、死亡率无显著影响; 15 ℃、密度为0.2 g·cm-2、污泥/麸皮为1∶2、幼虫体重为85.4±5.9 mg 时, 幼虫对Cu、Se、Hg 的积累作用较显著; 15 ℃、密度为0.4 g·cm-2、污泥/麸皮为1∶0、幼虫体重为85.4±5.9 mg 时, 黄粉虫幼虫对Cd 的积累能力显著; 20 ℃、密度为0.4 g·cm-2、污泥/麸皮为1∶0、幼虫体重为34.9±4.9 mg 时, Zn 的积累系数较大; 黄粉虫幼虫对Cu、Cd、Se、Hg 和Zn 等重金属元素有很强的积累作用。 相似文献
