Campylobacter is considered to be the leading bacterial etiologic agent of acute gastroenteritis in humans. Evidence implicates poultry as a major source of the organism for human illness; however, the pathways involved in Campylobacter contamination of poultry flocks, horizontal transmission and/or vertical transmission, remain unclear. Recent evidence implicates breeders as a potential source for Campylobacter contamination of the subsequent broiler offspring. In this investigation, Campylobacter isolated from feces, cloacal swabs, ceca, semen, and vas deferens of 12 breeder broiler roosters were genotyped by both flagellin A short variable region (flaA SVR) DNA sequence analysis and repetitive element (rep)-polymerase chain reaction (PCR). In 9 of 12 roosters, Campylobacter was isolated from multiple sites sampled. Comparison of multiple isolates obtained from individual roosters revealed variable results. In five of the nine roosters, all Campylobacter isolated demonstrated closely related flaA SVR DNA sequences as well as rep-PCR patterns; isolates from these roosters were collected from both the gastrointestinal and the reproductive tracts or from the gastrointestinal tract alone. The remaining four roosters had Campylobacter that were distinct by both typing methods. Isolates from two of these four roosters originated from both the gastrointestinal and the reproductive tracts. Isolates from the remaining two roosters originated from only the reproductive tract. Comparisons of all Campylobacter isolates recovered from a distinct sample type within either the reproductive tract or the gastrointestinal tract (feces, semen, cloaca, vas deferens, or ceca) were quite diverse. No relationship between the genotypes and the sample type could be ascertained. Further investigation is needed to determine the route of contamination and if the presence of Campylobacter within the rooster leads to contamination of the broiler offspring via the fertilized egg.  相似文献

Campylobacter are known to cause acute bacterial gastroenteritis in humans. Poultry products have been implicated as a significant source of these infections. Six experiments were performed to determine whether Campylobacter could be isolated naturally from the primary and secondary lymphoid organs, liver/gallbladder, and ceca of commercial broiler breeder hens. Broiler breeder hens were acquired from different commercial sources during the early, middle, and late lay cycles. The birds were euthanatized, defeathered, and aseptically opened. To reduce the possibility of cross-contamination between samples, the thymus, spleen, and liver/gallbladder were aseptically removed prior to removal of the ceca. Individual samples were placed in sterile bags, packed on ice, and transported to the laboratory for evaluation. In this study Campylobacter were found in 11 of 43 thymii, eight of 43 spleens, four of 43 liver/gallbladders, and 30 of 43 ceca. Overall, 28 of 53 isolates from the above samples were Campylobacter coli and 25 of 53 isolates were found to be Campylobacter jejuni.  相似文献

Campylobacter jejuni, a foodborne pathogen closely associated with market poultry, is considered to be the most frequent agent of human gastroenteritis in the United States. The pathways involved in the contamination of poultry flocks, vertical transmission and/or horizontal transmission, are unclear. In this study, Campylobacter isolates from two independent commercial broiler breeder flocks, as well as from their respective progeny, were characterized and compared by PstI ribotype analysis and by DNA sequence analysis of the short variable region (SVR) of the flaA gene (flaA SVR). Campylobacter isolates originating from one set of breeder hens and the feces from their respective progeny demonstrated identical ribotype patterns as well as identical flaA SVR DNA sequences, thereby suggesting that these isolates were clonal in origin. Ribotype analysis of Campylobacter isolates from the second set of breeder hens and processed carcasses from their offspring resulted in two patterns. Sequence analysis placed these isolates into two closely related groups and one distant group, similar to the ribotype analysis. These results demonstrate that Campylobacter isolates from commercial broiler breeder flocks and from the respective broiler progeny may be of clonal origin and that breeder hens can serve as a source for Campylobacter contamination in poultry flocks.  相似文献

1. The objective of this study was to determine a metabolisable energy (ME) requirement model for broiler breeder hens. The influence of temperature on ME requirements for maintenance was determined in experiments conducted in three environmental rooms with temperatures kept constant at 13, 21 and 30 degrees C using a comparative slaughter technique. The energy requirements for weight gain were determined based upon body energy content and efficiency of energy utilisation for weight gain. The energy requirements for egg production were determined on the basis of egg energy content and efficiency of energy deposition in the eggs. 2. The following model was developed using these results: ME = kgW0.75(806.53-26.45T + 0.50T2) + 31.90G + 10.04EM, where kgW0.75 is body weight (kg) raised to the power 0.75, T is temperature ( degrees C), G is weight gain (g) and EM is egg mass (g). 3. A feeding trial was conducted using 400 Hubbard Hi-Yield broiler breeder hens and 40 Peterson males from 31 to 46 weeks of age in order to compare use of the model with a recommended feeding programme for this strain of bird. The application of the model in breeder hens provided good productive and reproductive performance and better results in feed and energy conversion than in hens fed according to strain recommendation. In conclusion, the model evaluated predicted an ME intake which matched breeder hens' requirements.  相似文献

Photostimulation of retinal photoreceptors, which are sensitive to green light, appears to inhibit reproductive activity in birds, whereas photostimulation of extra-retinal photoreceptors, which are sensitive to red light, accelerates it. The objective of this study was to determine the effect of either retinal or extra-retinal photostimulation on reproductive activities of broiler breeder hens. At 23 wk of age, Cobb hens (N = 135) were divided into 9 rooms with individual cages (n = 15). At 24 wk of age, 3 rooms were photostimulated (14L:10D) with white light (Control, n = 45). Six rooms had 2 parallel lighting systems, red (660 nm) and green (560 nm), which were both on during 6 out of 14 h of the light period. Then, in 3 of these rooms, the green light was turned off and hens were exposed to a total of 14 h of red light (Red, n = 45), and in the other 3, the red light was turned off and green lighting continued for a total of 14 h (Green, n = 45). The Green group had reduced egg production; reduced plasma concentrations of ovarian steroids; reduced luteinizing hormone (LH)-β, vasoactive intestinal peptide (VIP), and prolactin mRNA expression; and greater retinal green opsin mRNA expression (P ≤ 0.05). The Red group had greater egg production; greater gonadotropin-releasing hormone-I (GnRH-I) and red opsin gene expression in the hypothalamus; and lesser green opsin gene expression in the retina (P ≤ 0.05). We suggest that selective photostimulation of extra-retinal photostimulation as opposed to retinal photostimulation is a key factor in the determination of successful reproduction of broiler breeder hens.  相似文献

Lameness and mortality of 0.55% to 1.00% per week occurred in 32-to-33-wk-old broiler breeder hens in a complex involving eight houses. Males were not affected. Laboratory evaluation of 19 birds, over 2 wk, revealed complete rupture or partial tear, primarily of the gastrocnemius tendon; the fibularis longus, fibularis brevis, and flexor muscles and their tendons were injured in a few birds. Rupture of the gastrocnemius tendon has been observed in broiler breeder chickens older than 12 wk of age and it is characterized by lameness secondary to unilateral or bilateral rupture of the tendon. The cause of this condition has been primarily attributed to reovirus infection but rupture by other causes can also occur. In this case, there was no evidence of synovitis or reovirus infection in the birds. The cause of the ruptures was attributed to aggressive males when the ratio of males to females was increased from seven or eight males per 100 females at 22-23 wk to nine or 10 per 100 at 31 wk of age.  相似文献

1. Broiler parent stock were fed daily allowances of 1.88, 1.73 or 1.52 MJ apparent metabolisable energy (AME) per bird at two different daily protein intakes (27, 21.3 g crude protein (CP) per bird) or daily protein intakes of 24.6 and 19.4 g CP per bird at a daily energy intake of 1.88 MJ AME per bird from 21 to 64 weeks of age.

2. Body‐weight gain and carcass fat and water content increased and fertility decreased with increasing energy allowance. Maximum egg production occurred at an energy intake of 1.73 MJ AME/bird d.

3. Differences in egg weight and hatchability were related to differences in both energy and protein intake. The highest egg weight occurred at the highest allowance of energy and protein. Hatchability was depressed where the daily allowances of protein and energy were in a ratio of more than 15 g protein: 1 MJ AME.

4. Apart from egg size no significant effects on reproductive performance were observed when dietary protein intake was varied from 27 to 19.5 g/bird d.

5. Requirements of broiler breeder hens for protein during lay may be lower than previously thought. For the strain used a protein intake of 19.5 g/bird d appeared adequate provided essential amino acid concentrations were maintained.

6. The close relationships between body weight and energy allowance and the latter and egg production make body‐weight gain a useful guide to management. A body‐weight gain of about 1.1 kg from 21 to 36 weeks of age was associated with optimum performance in this study.  相似文献

One of the greatest challenges for nutritionists is ensuring that breeder hens consume the quantity of nutrients necessary to ensure proper development while also ensuring the production of good quality chicks. However, new studies must investigate how nutrition can positively affect the performance of these poultry. The present study aimed to evaluate hydrolyzed yeast supplementation in the diet of Cobb 500 broiler breeder hens, starting from 25 wk of age, and to evaluate the performance of the hens and their progeny. Broiler breeder hens fed hydrolyzed yeast supplement exhibited better performance compared with the control group. The results of the progeny demonstrated improved performance in chicks from the breeder hens fed the hydrolyzed yeast supplement. However, no improvement was observed in breast or leg yields; only carcass yield improved in these broilers. The results demonstrate that supplementation with hydrolyzed yeast in the diet of broiler breeder hens improves the performance of hens and their progeny.  相似文献

Campylobacter and Salmonella are known to cause acute bacterial gastroenteritis in humans. Raw poultry products have been implicated as a significant source of these infections. Five trials were conducted to determine whether Campylobacter and Salmonella spp. exist naturally in the mature and immature ovarian follicles of late-life broiler breeder hens. Broiler breeder hens ranging from 60 to 66 wk of age were obtained from four different commercial breeder operations. For each trial, the hens were removed from the commercial operation and held overnight at the University of Georgia processing facility. The hens were euthanized, defeathered, and aseptically opened. To reduce the possibility of cross-contamination between samples, first the mature and immature ovarian follicles, then the ceca, were aseptically removed. Individual samples were placed in sterile bags, packed on ice, and transported to the laboratory for evaluation. Overall, Campylobacter was found in 7 of 55 immature follicles, 12 of 47 mature follicles, and 41 of 55 ceca. Campylobacter was found in at least one of each sample of mature follicles and in ceca in each of the five trials. Salmonella was found in 0 of 55 immature follicles, 1 of 47 mature follicles, and 8 of 55 ceca. In this study, the recovery rate of Salmonella from late-life broiler breeder hen ovarian follicles was relatively low. However, the recovery rate of Campylobacter from the hen ovarian follicles was reasonably high, suggesting that these breeder hens could be infecting fertile hatching eggs. Determining how Campylobacter contaminated these ovarian follicles and how many chicks could be colonized from this source are the next steps in helping to elucidate a better understanding of this ecology and the control of Campylobacter in poultry production.  相似文献

Myopathy of the pectoralis profundus and ambiens muscles was found in chickens from 5 flocks of broiler breeder hens. Histological lesions in the muscles varied from steatosis to loss of striations, fragmentation of fibers, and mineral deposition. A clinical response followed oral medication with vitamin E and selenium although analysis showed that selenium and vitamin E levels in the feed were normal. Tetramethylthiuramdisulfide (TMTD), the active ingredient in Arasan, fed to broiler breeder hens at 10, 20, or 40 ppm depressed egg production, reduced hatchability, and resulted in soft-shelled eggs. The progeny of these hens had anomalies manifested by bilaternal medial deviation of the toes, exophthalmia, and shortened femurs. Broiler breeder hens that consumed a ration containing 80 ppm of TMTD produced only soft-shelled eggs.  相似文献

Pooled semen samples from 12 groups of mature commercial broiler breeder roosters were analyzed for the presence of Campylobacter. Each of the 12 groups was comprised of eight individuals and was sampled weekly for five consecutive weeks. Once a day, roosters were allowed to have a restricted amount of feed after the semen samples were collected by abdominal massage. This feeding schedule reduced the amount of fecal contamination in and around the vent as well as in the semen sample. For replications 1, 2, and 3, the numbers of Campylobacter-positive groups were 8, 5, and 5, respectively, out of 12. For replications 4 and 5, 6 of 8 and 6 of 11 groups were positive, respectively. Only two groups were positive for Campylobacter at all sampling times, two groups were negative each time, and eight groups produced variable results. Also, fecal droppings, external swabs of the genitalia, and semen samples were taken from individual roosters between 49 to 65 wk of age. Of the total 275 semen samples collected, 9.47% contained naturally occurring Campylobacter, whereas 9.6% of 114 fecal droppings and 7.9% of the 114 genital swabs were positive. Levels of the organism present in the fecal samples ranged from 1.0 to 4.2 log colony-forming units (CFU)/g with an average of 2.9 log CFU/g feces. For semen, the levels ranged from as low as enrichment recovery only to as high as 3.1 log CFU/ml of semen with an average of 1.2 log CFU/ml. For swabs of genitalia, the levels of Campylobacter were so low that recovery was achieved only through enrichment. These data suggest that rooster semen may serve as a vehicle for transmission of Campylobacter to the reproductive tract of the hen and subsequently to the fertile egg.  相似文献

We previously reported the recovery of Campylobacter (naturally colonized) from the ductus deferens of 5 of 101 broiler breeder roosters, and four of those five positive roosters had previously produced Campylobacter-positive semen samples. Those results prompted further evaluation to determine if inoculation route influenced the prevalence or level of Campylobacter contamination of semen, the digestive tract, or reproductive organs. Individually caged roosters, confirmed to be feces and semen negative for Campylobacter, were challenged with a marker strain of Campylobacter jejuni either orally using 1.0 ml of a diluted cell suspension (log(10)4.3 to 6.0 cells), by dropping 0.1 ml of suspension (log(10)5.3 to 7.0 cells) on the everted phallus immediately after semen collection or by dip coating an ultrasound probe in the diluted cell suspension (log(10)4.3 to 6.0 cells) and then inserting the probe through the vent into the colon. Six days postinoculation, individual feces and semen samples were again collected and cultured for Campylobacter. Seven days postinoculation, roosters were killed, the abdomen aseptically opened to expose the viscera, and one cecum, one testis, and both ductus deferens were collected. The samples were then suspended 1:3 (weight/volume) in Bolton enrichment broth for the culture of Campylobacter. Samples were also directly plated onto Cefex agar to enumerate Campylobacter. Campylobacter was recovered 6 days after challenge from feces in 82% of samples (log(10)4.1 colony-forming units [CFU]/g sample), 85% of semen samples (log(10)2.9 CFU/ml), and on the seventh day postchallenge from 88% of cecal samples (log(10)5.8 CFU/g sample). Campylobacter was not directly isolated from any testis sample but was detected following enrichment from 9% (3/33) of ductus deferens samples. Roosters challenged with Campylobacter orally, on the phallus, or by insertion of a Campylobacter dip-coated ultrasound probe were all readily colonized in the ceca and produced Campylobacter-positive semen and feces on day 6 after challenge. The low prevalence of recovery of Campylobacter from the ductus deferens samples and failure to recover from any testis sample suggests that semen may become Campylobacter positive while traversing the cloaca upon the everted phallus. The production of Campylobacter-positive semen could provide a route in addition to fecal-oral for the horizontal transmission of Campylobacter from the rooster to the reproductive tract of the hen.  相似文献

Individually-caged broiler breeder hens were fed allowances of 1.88, 1.61, 1.32 or 1.13 MJ apparent metabolisable energy (AME) per bird d at four different protein intakes (27, 23, 19.5 or 16.5 g crude protein per bird d) from 21 to 60 weeks of age. A system of foot scoring was used to assess the extent and severity of foot damage. Damage increased as energy intake increased but protein intake had little effect on foot condition. Cushion floors or perches introduced into the cages at 27 weeks of age reduced the deterioration in foot condition observed on all dietary treatments during lay. Egg output was not affected by foot condition although severe foot damage was observed in several cases.  相似文献

1. Broiler breeder hens were used in an experiment lasting 10 weeks (29 to 38 weeks of age) to measure the responses to dietary lysine and methionine, the main objective being to determine whether the coefficients of response to these amino acids were the same for broiler breeders and for laying pullets. 2. The hens were offered 150 g/d of one of 20 dietary treatments, 10 being lysine-limiting and 10 being methionine-limiting. The diets were mixed by diluting one of two concentrate (summit) mixes with a protein-free dilution mixture. The lysine-limiting summit diet was designed to supply approximately 1300 mg lysine/bird d, while the other supplied 520 mg methionine/bird d, when fed at 150 g/bird d. 3. Birds on the 5 lowest concentrations of both lysine and methionine did not consume the allotted amount of food, the amount decreasing, in a curvilinear fashion, to approximately 105 g/bird d. 4. The minimum egg weight recorded was 0.8 of the maximum, whereas the rate of lay of birds fed on the diets with the lowest amino acid concentrations was 0.2 of the maximum. 5. Using the Reading Model, the coefficients of response were calculated to be (for lysine) 16.88 E and 11.2 W, and for methionine, 7.03 E and 1.52 W, where E = egg output, g/bird d, and W = body mass, kg/bird. An average, individual, broiler breeder of 3 kg, producing 45 g of egg output per day, would need 793 mg of lysine and 321 mg of methionine daily. This intake of methionine is similar to that estimated by means of coefficients used for laying pullets, but the lysine requirement would be underestimated by 0.18 if the coefficients for laying pullets were used. 6. The coefficients for maintenance for both lysine and methionine, determined in this experiment, are considerably lower than values published previously, whilst the coefficients for egg output are, in both cases, higher. The resultant flock response curves therefore differed significantly from those in which the coefficients of response for for laying pullets were used.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献
