Comparative virulence of blue-stain fungi isolated from Japanese red pine   总被引:2,自引:0,他引:2  
 We made inoculations to determine the comparative virulence of blue-stain fungi isolated from Japanese red pine (Pinus densiflora Sieb. & Zucc.). Based on the symptoms caused by each fungus, the most virulent fungus was Leptographium wingfieldii. Also, Ophiostoma minus was relatively virulent while lesions induced by some species did not differ from those in the control treatment. Based on the results, we concluded that the pathogenicity of the blue-stain fungi varied widely, and thus their impact on the host most likely varies too. Black stain root disease (BSRD) caused by Leptographium wageneri var. ponderosum (Verticicladiella wageneri) is a destructive disease in many Jeffrey pine (Pinus jeffreyi) and ponderosa pine (Pponderosa) forests of the western United States. Overcrowding is a pervasive problem in these forests, unless alleviated by thinning. In an overcrowded BSRD‐infested second‐growth Jeffrey/ponderosa pine forest in northern California, a large study (16.2 hr; 40 acres) was designed to determine the impact of thinning to four levels—6.9 m2/ha (30 ft2/acre), 13.8 m2/ha (60 ft2/acre), and 20.7 m2/ha (90 ft2/acre) of basal area and a control with no thinning—on the incidence of BSRD‐caused mortality. Ten years after this experiment was initiated, results indicate that thinning to any of these levels lowered the incidence of BSRD‐caused mortality as compared with BSRD‐caused mortality in unthinned control plots.  相似文献

The pinewood nematode (PWN), Bursaphelenchus xylophilus, is the causal agent of pine wilt disease (PWD), which is a major problem in East Asia and West Europe. Quick identification of PWN is needed to prevent the dispersal of PWD to healthy forests. Various detection methods of PWN have been developed using anatomical characters and molecular markers. These methods are not suitable for rapid diagnosis because it is difficult to distinguish B. xylophilus from the non‐pathogenic species Bursaphelenchus mucronatus based on morphological characters without expertise in nematode taxonomy and most PCR or isothermal amplification detection methods require time‐consuming processes. In this study, we developed an on‐site PWN detection method using a recombinase polymerase amplification (RPA) assay with a novel extraction buffer (DAP buffer). This new PWN detection method is able to extract genomic DNA from PWN in pinewood by simple buffer consisting of sodium hydrate, polyethylene glycol 200 and dimethyl sulfoxide in 10 min without using the experimental devices and able to distinguish between B. xylophilus and other Bursaphelenchus spp. by amplifying the species‐specific 5S rDNA fragment of B. xylophilus in 10 min. Taken together, our protocol can obtain the result for the detection of PWN in pine tree samples within 30 min. This result suggests that RPA/DAP assay is much faster, easier and cheaper than the conventional methods for detecting PWN.  相似文献

In Europe, fungal pathogens have reduced the overall productivity of sweet chestnut (Castanea sativa) stands and continue to threaten the economic viability of forestry operations. Chestnut Red Stain (CRS) in north‐eastern Spain, locally referred to as Roig, is capable of decreasing the market value of chestnut timber to the point of rendering chestnut coppices uneconomical. Despite its economic importance, the specific cause of this red discolouration is unknown. With the objective of verifying the presence of fungi within the symptomatic wood, and identifying the fungus or suite of fungi associated with the red stain, wood samples were collected and cultured from 37 stumps found in eight recently harvested stands in the Montseny and Montnegre‐Corredor Natural Parks. To separate the fungi associated with CRS from other species inhabiting the chestnut wood, the origin of each fungal culture was mapped in every stump. The fungi were isolated from cultures and identified by sequencing the ITS region. The results provide insight into the fungal community inhabiting chestnut wood and the potential cause of CRS; nine species were identified including two species known to cause decay in chestnut. One of them, Fistulina hepatica, appears to be a likely candidate for the causal agent of CRS. This is the first study reporting the fungi associated with CRS and opens the door to new epidemiological studies focused on F. hepatica.  相似文献

Bark beetle‐vectored ophiostomatoid fungi, Leptographium terebrantis, is inoculated on the roots and lower stems of stressed Pinus species during the feeding activity of bark beetle. To determine the exact host response following inoculation, it is critical to challenge the host with a realistic amount of fungal inoculum. Thus, we designed a series of stepwise experiments using L. terebrantis colonized toothpicks which focused on the inoculum transfer from the toothpicks to excised Pinus taeda stem segments and living saplings, respectively, at different inoculum densities. The toothpicks served as a substrate for fungal growth and sporulation and the inoculation showed their utility in eliciting host's response to the pathogen. The inoculated fungus caused blue‐stain and sapwood occlusions in P. taeda stems and saplings, respectively. The volume of occluded, visually damaged sapwood increased by 1.96 cm3 per radial inoculation point on average. Fungal colonized toothpicks can be used as a suitable alternative to agar discs for studying bark beetles vectored fungi and their host interactions.  相似文献

从5种不同松树上分离到拟松材线虫24个虫株,并在高倍显微镜下对其雌成虫的12个形态指标进行了测计,并以这些形态指标值作为形态学变异研究的依据。结果表明,拟松材线虫雌成虫的平均体长约为954.41μm,最长的能达1 312.50μm,最短为527.80μm;24个不同虫株的拟松材线虫雌成虫同一形态指标的变异系数在不同的虫株间各不相同,这反映了拟松材线虫形态多样性是很丰富的。但雌成虫的生殖系统指标(V’、V)和消化系统口针长度变异度较小,可以作为拟松材线虫的重要鉴定特征。不同寄主树种上的拟松材线虫雌成虫体长的均值大小依次为黑松、华山松、湿地松、马尾松、火炬松。借助于SPSS 13.0方差分析表明,不同寄主树种上的拟松材线虫雌成虫绝大部分的形态指标在0.01水平上差异显著,也就说寄主树种因素对拟松材线虫形态的分化有显著性影响。  相似文献

Lawson cypress (Chamaecyparis lawsoniana), an important landscape tree, is widely planted in gardens and parks throughout Iran. Crown gall disease on Lawson cypress trees was observed in Sari and Juybar Counties, Mazandaran province, northern Iran, in 2017. Isolation from galls on potato dextrose agar (PDA) containing CaCO3 yielded bacterial colonies, the predominant types of which were purified and selected for characterization. The isolates were Gram‐negative, oxidase positive, able to grow in 2% NaCl and produced 3‐ketolactose. They hydrolysed esculin, casein and arbutin but not starch, gelatin or Tween 80. Two representative isolates were selected for PCR amplification and sequencing of DNA gyrase subunit B (gyrB) gene. In the phylogenetic tree based on the partial sequence of the gyrB gene, isolates KH1 and KH2 clustered with Agrobacterium pusense. The pathogenicity of all isolates was confirmed by inoculation on Jimsonweed (Datura stramonium) and carrot discs (Daucus carota). Confirmation of the presence of genes involved in pathogenicity was made by performing PCR with the virD2A/virD2C and VCF/VCR primer pairs which resulted in amplification of the expected 224 and 730 bp fragments in all studied isolates, respectively. A. pusense was therefore identified as the causal agent of crown and stem gall of Lawson cypress. This appears to be the first report on the natural occurrence of crown gall disease on Lawson cypress and the first record of a plant disease caused by A. pusense.  相似文献

The presence of quarantine pathogen Lecanosticta acicola, the causal agent of brown spot needle blight, was confirmed using molecular methods for the first time in northern part of Poland on the coastal area adjacent to the Baltic Sea. This area includes sandy beaches, where Pinus mugo and P. sylvestris were planted. Symptomatic needles were collected in 2017 from 20 P. mugo trees growing in one stand in Ustka. Typical symptoms of brown spot needle blight infection, including dead needle tips and central zones with yellow or reddish brown, circular spots in green tissue, were observed on all samples. Only, the asexual stage of L. acicola was obtained during this work. The pathogen species identity was confirmed using classical morphological methods (microscopic examination of the infected needles), real‐time and species‐specific priming (SSPP) PCR, and ITS sequencing. Analysis of mating‐type (MAT) genes showed the presence of both mating types in northern Poland.  相似文献

Dwarf mistletoes (Arceuthobium spp.) are a group of obligate, hemiparasitic plants that infect numerous species in the Pinaceae in North America. Wildland fire is considered to be the primary natural agent influencing the population and distribution of dwarf mistletoes across landscapes. Based on this understanding, prescribed fire has been suggested as a potential method for dwarf mistletoe sanitation and control; however, experimental work has primarily focused on prescribed surface fire. In this study, we report long‐term impacts of three experimental crown fires on dwarf mistletoe severity in infested lodgepole pine stands in Colorado 33 years post‐fire. The three fires achieved tree mortality rates ranging from 20% to 100%. Our results suggested a significant negative relationship between the amount of fire‐caused tree mortality and future dwarf mistletoe severity. These findings supported the presumed natural role of fire in altering dwarf mistletoe populations, which perhaps exhibits a linear relationship between fire‐caused host tree mortality and future dwarf mistletoe severity.  相似文献

Diplodia sapinea and Diplodia scrobiculata are opportunistic pathogens of Pinus species. Several studies about taxonomy, impact and epidemiology of these fungi have been conducted in previous years, which have provided useful information and have raised new issues. These diseases produce a considerable impact on plantations resulting in significant economic losses. The main aims of this study are to increase the knowledge of the potential of genetic exchange and the relative aggressiveness of these organisms that can persist in healthy tissues of asymptomatic trees. A collection of 250 isolates among which are 149 strains collected from Pinus radiata plantations in Basque Country (Spain) and 101 strains from different countries was included in this work. Mating type ratios were analysed and compared using the structure of the MAT locus (MAT1‐1‐1 and MAT1‐2‐1). Inoculations of Pinus radiata seedlings were performed in a biosafety greenhouse (P2) to confirm pathogenicity of isolates and compare their aggressiveness. The frequency of occurrence of both idiomorphs of D. sapinea in Basque Country isolates was close to 1:1, however, for collection of isolates of this fungus from around the world, the ratio was 1:2. Furthermore, the spatial distribution of the two mating types in the Basque Country was random. Despite no detection of a sexual state, these results could suggest sexual reproduction behaviour. The pathogenicity of all strains in the collection was confirmed. Although aggressiveness (in terms of lesion lengths resulting from inoculation) varied greatly, no statistically significant effects of MAT type or pathogen species were detected.  相似文献

In western Ukraine, forest decline and dieback of several broadleaved tree species have become increasingly evident during recent years, and surveys in some areas have shown symptoms indicative of Phytophthora infections. In this study, we aimed to determine the occurrence and diversity of Phytophthora species associated with several broadleaved tree species (Alnus glutinosa, Betula pendula, Castanea sativa, Fagus sylvatica and Quercus robur) from forest stands where dieback has been observed. Rhizosphere soil samples were collected from 14 forest stands during 2017 and 2018 and tested for the presence of Phytophthora species using morphological and molecular methods. Seven Phytophthora species (P. bilorbang, P. cactorum, P. gallica, P. gonapodyides, P. lacustris, P. plurivora and P. polonica), and two other clade six taxa were detected from the various forest types, several of which are probable agents responsible for decline. Four of the Phytophthora species (P. bilorbang, P. gallica, P. plurivora and P. polonica) have previously never been reported from broadleaf forests in Ukraine.  相似文献

As a result of the detection of the pinewood nematode Bursaphelenchus xylophilus in Portugal, and its subsequent spread to Spain, intense surveys were conducted to screen for the presence of Bursaphelenchus species in Romania. Herein, we report recent surveys of insects potentially vectoring Bursaphelenchus species collected using trap trees or pheromone‐baited traps placed in the forest. Trap felled spruce trees (Picea abies) and pheromone‐baited traps were installed in six different counties in Romania (Bra?ov, Sibiu, Suceava, Hunedoara, Timi? and Dâmbovi?a). Ten different species of insects distributed among Curculionidae and Cerambycidae were obtained. Nematodes were extracted from insects and observed to validate the presence of Bursaphelenchus specimens. One female identified as Monochamus sutor was the only specimen carrying nematodes in the genus Bursaphelenchus. Nematodes were identified as B. mucronatus based on morphological and molecular features. This is the first detection and report of natural spread of B. mucronatus in Romania. The absence of B. xylophilus was confirmed in the areas of Romania surveyed in this work.  相似文献

In this study, a total of 68 endophytic bacteria were isolated from different tissues of walnut trees. About 55% and 22% of the strains had the ability to produce indole acetic acid and gibberellic acid, respectively. The capability of isolates to solubilize phosphate, growth on N‐free medium, siderophore, protease and lipase production was varied. Based on phenotypic grouping and plant growth promotion properties, twelve isolates were selected and 16S rRNA gene‐based phylogenetic analysis revealed that strains showed 99%–100% similarity to Pseudomonas, Bacillus, Arthrobacter, Roseomonas and Streptomyces genera. Amongst the selected strains, PS12, KS54, JS66 and KS74 showed root and shoot growth enhancement of poplar cutting. NS70, KS54 and PL36 strains showed antagonistic activity against Pseudomonas syringae pv. syringae;RR47, KS74 and NR69 strains had inhibition effects against Brenneria nigrifluens; and JS66 and RR26 strains had antagonistic activity against both phytopathogens under in vitro conditions. This is the first reported study to elucidate the endophytic bacterial diversity associated with walnut trees with beneficial attributes.  相似文献

In addition to Hymenoscyphus fraxineus, two fungi identified as Diaporthe eres aff. and Fusarium sambucinum aff. were also isolated from necrotic bark lesions on declining one‐year‐old Fraxinus excelsior in a forest stand in Montenegro. To examine their involvement in ash decline, a pathogenicity test was performed using under bark inoculations on one‐year‐old Fraxinus excelsior. Hymenoscyphus fraxineus was included as comparison. All three fungal species proved highly pathogenic towards one‐year‐old seedlings although lesion sizes differed significantly between the different species. Hymenoscyphus fraxineus was most aggressive, followed by F. sambucinum aff., while D. eres aff. caused the smallest lesions. This study demonstrates for the first time the ability of isolates in the D. eres and F. sambucinum species complexes to cause decline on one‐year‐old common ash seedlings.  相似文献

In February 2015, an unexpected windstorm downed five hectares of a European black pine Pinus nigra subsp. laricio forest formation located close to Vallombrosa, Florence (Central Italy). In the following spring, an extensive survey was conducted in the area. Felled trees, stumps and all the suitable plant material were screened for the presence of the pinewood nematode (PWN), Bursaphelenchus xylophilus, by sampling wood and bark. Bark beetles were then collected from the gallery systems on the inner side of bark samples and observed in the laboratory. The following bark beetles were morphologically identified: Ips sexdentatus, Orthotomicus erosus, O. laricis and Pityogenes bidentatusa. The dissection of Ips sexdentatus allowed the extraction of numerous nematodes that were morphologically and molecularly identified as Ektaphelenchoides pini. Conversely, only few nematode specimens were isolated from either pine bark or wood. These individuals could be only molecularly identified and belonged to an undescribed nematode taxon. Even though no PWN was recorded in the investigated sites, our survey allowed the detection of a new association between E. pini and I. sexdentatus on P. nigra.  相似文献

Fungal abundance and diversity were assessed in two differently managed forest soils. In soil 1, chipped woody debris had been spread on the top soil and mixed with the soil by standard deep ploughing to 50 cm. In soil 2, woody debris was removed and there was no ground preparation. Ten years later, the different methods of management resulted in unexpectedly distinctive habitat heterogeneity with a high variation in structure of the mycobiota communities. Incorporation of woody debris into soil resulted in lower abundance of fungi but greater fungal diversity (which may enhance ecosystem stability) and was beneficial for the growth of Scots pines. Removal of post‐harvest woody debris from the surface of the clear‐cut site with no subsequent ground preparation, compared with incorporating chipped debris and deep ploughing, resulted in the soil of the subsequent 10‐year‐old Scots pine plantation having: (a) 159‐fold greater abundance of fungi, (b) lower fungal diversity, (c) different fungal community composition, (d) higher proportion of mycorrhizal fungi, (e) no root rot pathogens, (f) less tree biomass. The study showed that removal of post‐harvest woody debris and no ground preparation is potentially beneficial for the health of young coniferous trees.  相似文献

Basal stem rot disease caused by the white‐rot fungus Ganoderma boninense is a major threat to the oil palm industry, and hence, the ability to detect infections at an early stage of development is desired. In this study, a headspace solid‐phase microextraction (HS‐SPME) method coupled with gas chromatography–mass spectrometry (GC‐MS) was employed to analyse the volatile organic compounds (VOCs) released from G. boninense cultures and infected oil palm wood. We examined VOCs released from three types of samples: G. boninense mycelium, oil palm wood and oil palm wood colonized by G. boninense. This preliminary study led to the tentative identification of 57 VOCs, including alcohols, alkanes, volatile acids, ketones, aldehydes, esters, sesquiterpenes and polycyclic aromatic hydrocarbon groups. Aliphatic compounds with eight‐carbon atoms, such as 1‐octen‐3‐ol, 3‐octanone, 1‐octanol and (E)‐2‐octenal, were the most abundant constituents of the Ganoderma samples, whereas furfural and hexanal were the major compounds detected in the oil palm wood samples. Chemometric analyses using cluster heat maps and principal component analyses were used to discriminate between the VOC profiles. The results indicated that the novel method described here could be used to detect Ganoderma disease and, more generally, for chemoecological studies of plant–pathogen interactions.  相似文献

Pine wilt disease is of major concern as it has destroyed pine forests in East Asia and Europe. Several studies have suggested that invasion by the pinewood nematode (PWN) Bursaphelenchus xylophilus, which causes this disease, evokes an excessive defence response in pine trees, resulting in tree death. However, few studies have quantitatively evaluated the correlation between PWN distribution and tree defence responses. Therefore, the present study aimed to quantify the number of PWNs and expression levels of putative pathogenesis‐related (PR) genes in different positions of Japanese black pine (Pinus thunbergii) seedlings over time. To quantify the number of PWNs in the seedlings, we used TaqMan quantitative real‐time PCR (qPCR) assay. During the early phase of infection, most PWNs were distributed around the inoculated sites, with only a small number being detected at distant sites, but the expression levels of PR genes were highly upregulated throughout the seedlings. Both the number of PWNs and expression levels of PR genes then increased drastically throughout the seedlings, all of which exhibited external symptoms. Thus, it appears that the rapid migration of PWNs induces a defence response throughout the seedling; however, this may not be effective in controlling these parasites, thereby ultimately leading to plant death.  相似文献

Bud rot disease or “Pudricion del cogollo” (PC) of oil palm is a major constraint on production in Colombia and neighbouring countries such as Brazil, Costa Rica, Ecuador, Nicaragua, Panama, Peru and Surinam. To date, there are no documented reports of Phytophthora disease of oil palm in South‐East Asia. This research, therefore, was conducted to determine the pathogenic potential of Phytophthora palmivora and Phytophthora nicotianae on oil palm using both in vitro and nursery inoculation experiments. In vitro inoculation of both Ppalmivora and P. nicotianae on immature oil palm leaflets caused discoloration within 2 days of inoculation and incubation at 25 ± 1.5°C, 100% RH. Similarly, in nursery trials, lesions formed on the buds (unopened leaflets) 3 days after inoculation with Ppalmivora or P. nicotianae zoospore suspensions. No lesions developed on untreated leaflets in either in vitro or nursery inoculation experiments. Phytophthora spp. were re‐isolated from leaflet lesions and confirmed as the inoculated pathogens.  相似文献
