The humoral immune response and immunity conferred in chicks were compared following separate and combined oral vaccination with F strain of Newcastle disease virus (NDV) and HP1 strain of fowl pox virus. The haemagglutination inhibition (HI) antibody titre against NDV and passive haemagglutination (PHA) antibody titre against fowl pox virus were comparable in two respective groups. The serum IgG concentration increased significantly after the second vaccination in all the groups. The NDV vaccine induced significantly higher IgG production as compared to fowl pox virus vaccine. There was no significant difference in serum IgG concentration produced by combined vaccine and separate F strain vaccine. The protection afforded by combined and separate vaccinations did not vary significantly against challenge with virulent strains of NDV and fowl pox virus at different stages.  相似文献

The intra- and extracellular virus of three strains of fowl pox virus, when precipitated in succession with different saturation of ammonium sulphate revealed three antigens in gel diffusion test in the precipitates obtained at 25%, 50% and 75% of saturation. Further analysis of each positive antigen by dot ELISA revealed that the extracellular virus of FS-8 and HP 1 strains possessed excess antigenic protein at 50% saturation compared to their intracellular viruses. While no difference between extra- and intracellular viruses of FS-4 strain was observed.  相似文献

Smallpox vaccination and bioterrorism with pox viruses   总被引:1,自引:0,他引:1  
Bioterrorist attacks occupy a special place amongst the innumerable potential types of terrorist attack, with the intentional release of pox viruses being especially feared in this connection. Apart from the variola virus, the agent responsible for smallpox in humans, the monkeypox virus and numerous other animal pox viruses pose potential risks for humans and animals. This risk scenario also includes recombinations between the various pox viruses, changes in hosts and genetically engineered manipulations of pox viruses.

For over 200 years, the method of choice for combatting smallpox was via vaccination with a reproductive, original vaccinia virus. Worldwide eradication of smallpox at the end of the 1970s and the discontinuation of routine smallpox vaccination in 1980 can be credited to such vaccination. Unfortunately, these vaccinations were associated with a large number of postvaccinal impairments, sometimes resulting in death (e.g. postvaccinal encephalitis). The only way to restrict such postvaccinal complications was to carry out initial vaccination within the first 2 postnatal years. Initial vaccination at a later age led to such a sharp increase in the number of vaccines with complications that vaccination had to be discouraged.

The dilemma of the smallpox vaccine stocks stems from the fact that a large portion of these stocks are produced with the same vaccinia strains as before. This is irresponsible, especially as the percentage of immune-suppressed persons in the population, for whom vaccination-related complications pose an especial threat, is increasing.

One solution to the dilemma of the smallpox vaccine stocks is the MVA strain. It is harmless, protects humans and animals equally well against smallpox and can be applied parenterally.  相似文献

Conventional Newcastle disease vaccines are not suitable for application to village chickens in tropical countries of Asia. Trials with food-based vaccines are being initiated and the following experiments were performed to evaluate oral vaccination with Newcastle disease virus. Experimental chickens were vaccinated orally with the avirulent V4 strain of Newcastle disease virus and haemagglutination-inhibition antibody responses were measured. V4 virus was introduced into the crop by tube and total faecal output was collected daily and assayed for Newcastle disease virus. Virus was recovered on Days 5 and 6 after vaccination from most chickens that had received 10(7.4) and 10(6.4) 50% egg-infectious doses (EID50) of virus. There was no recovery of virus from birds receiving a lower dose of vaccine. Groups of chickens kept in cages with wire floors were given various doses of vaccine into the crop. Higher antibody titres were achieved with higher doses of virus. This dose responsiveness was not observed when various doses of vaccine were presented on food pellets and the groups of chickens were kept on concrete floors. Similar antibody responses were then seen with nominal doses of 10(5.2) and 10(8.2) EID50 per bird, possibly as a result of excretion and re-ingestion of the vaccine virus. Spread of the vaccine virus was demonstrated when control chickens and chickens receiving 10(7.7) EID50 of V4 virus on food pellets were housed together on a concrete floor. Similar antibody titres were achieved in both vaccinated and in-contact chickens.  相似文献

Cellular immune response of 6-week-old chickens infected with fowl adenovirus type-1 was evaluated by both in vivo and in vitro assay. In vitro assay, enumerating peripheral T lymphocyte by alphanaphthyl acetate esterase staining, revealed that cellular immunity appeared as early as 1 week post-infection and was maintained up to 5 weeks post-infection. In vivo assay by phytohaemagglutin skin test showed cellular immunity until 2 weeks post-infection. Thereafter no immunity was observed by this assay.  相似文献

Cellular and humoral immune responses to parenteral vaccination with an aromatic-defined (aroA) Salmonella typhimurium and to oral challenge with the S. typhimurium parent strain were examined in pigs. The effectiveness of aroA S. typhimurium vaccination for prevention of clinical disease following challenge was also evaluated. A split litter model was utilized and analysis of variance was by least squares. The statistical model accounted for the effects of vaccination and litter. Parenteral vaccination of pigs with the aroA mutant induced a significant O-polysaccharide (O-ps) specific lymphocyte blastogenic response as well as a significant antibody response to O-ps, lipopolysaccharide and killed bacteria. The aroA strain was avirulent in pigs, was not shed in the feces and significantly reduced the severity of diarrhea following oral challenge.  相似文献

Five-week-old chickens were inoculated with fowl pox (FP) virus and killed on various days through day 30 postinoculation (PI). The trachea was examined with a scanning electron microscope (SEM), a transmission electron microscope (TEM), and a light microscope (LM). From day 3 PI, small focal lesions of the mucosa were detected. On day 7 PI, upon formation of cytoplasmic inclusion bodies, epithelial cells proliferated profusely, enlarged, and formed clusters like papillomata. The disease proceeded to the gradual disruption of the lesions owing to the collapse of individual degenerating epithelial cells. Total desquamation of the lesions was observed. Ultrastructural examination revealed that the surface degenerating epithelial cells of the lesions ruptured and had virus particles inside. These changes were accompanied by severe inflammatory reaction. Thereafter, epithelial cells regenerated actively and the mucosa recovered by day 27 PI.  相似文献

In this study, the immune responses to and protective efficacy of a live attenuated Salmonella-delivered vaccine candidate secreting the papA, papG, iutA, and clpG antigens of Escherichia coli were evaluated against infection with avian pathogenic E. coli (APEC) in layer chickens. Primary vaccination was done at age 7 d and booster vaccination at age 5 wk. The levels of intestinal secretory immunoglobulin A specific to the 4 antigens were significantly higher in the vaccinated group than in the control group. A potent lymphocyte-proliferation response and increased levels of interferon-γ, interleukin-2, and interleukin-6 in the plasma and in culture supernatants of antigen-stimulated lymphocytes from the vaccinated group suggested significant induction of the cell-mediated immune response in this group compared with the control group. Upon challenge with a virulent APEC strain at 8 wk of age, the vaccinated group had no deaths, whereas the control group had a 15% mortality rate. In addition, the morbidity rate was significantly higher in the control group (55%) than in the vaccinated group (15%). Thus, giving primary and booster vaccination with the Salmonella-delivered APEC vaccine candidate significantly elevated both mucosal and cellular immune responses, which protected the chickens against colibacillosis.  相似文献
