海洋生境贝莱斯芽孢杆菌TCS001的鉴定及抑真菌活性   总被引:1,自引:1,他引:0  
为研究海洋生境芽孢杆菌TCS001的分类地位和抑菌活性,通过形态和生理生化特征观察,并结合gyrA序列同源性分析对菌株进行了鉴定;通过平板对峙培养法测定了菌株TCS001对多种植物病原真菌的抑菌谱;采用菌丝生长速率法和凹玻片法,测定了不同浓度TCS001菌株发酵滤液对靶标菌黄瓜灰霉病菌Botrytis cinerea菌丝生长和孢子萌发的影响。结果显示:该菌株为贝莱斯芽孢杆菌Bacillus velezensis,其对6种供试病原菌均有一定的抑制效果,其中对黄瓜灰霉病菌的抑制率最高,达87.66%。不同稀释倍数下,TCS001发酵滤液对黄瓜灰霉病菌均有一定的抑制作用,其中,稀释5倍时对菌丝生长和孢子萌发的抑制率最高,分别为96.24%和98.05%,稀释20倍时抑制率也均达90%以上。形态学观察发现,TCS001发酵滤液可导致黄瓜灰霉病菌孢子萌发芽管中间或顶端膨大畸形。研究表明,海洋生境贝莱斯芽孢杆菌TCS001极具开发为微生物农药的潜能。  相似文献

The effects ofSclerotinia sclerotiorum live and autoclaved sclerotia, and sclerotial exudates, and commercial oxalic acid were testedin vitro on sevenConiothyrium minitans strains differing in aggressiveness towardsS. sclerotiorum. Only sclerotial exudates and autoclaved sclerotia affected the mycelial growth rate of almost all the strains tested, whereas a change in theC. minitans mycelial growth pattern was observed in the presence of autoclaved sclerotia and live sclerotia germinating by the myceliogenic eruptive germination. In addition, sclerotial exudates had a stimulatory effect on spore germination.  明确黄脉爵床棒孢霉叶斑病病原菌及其生物学特性,为防控提供理论依据。通过病原菌分离、形态特征观察、致病性测定、rDNA-ITS序列分析、生物学特性及寄主范围测定等研究,证明该病病原菌为山扁豆生棒孢Corynespora cassiicola;菌丝生长及产孢适宜温度20℃~28℃,孢子萌发适宜温度24℃~32℃,菌丝致死温度49℃处理10 min;菌丝生长适宜pH 6~10,产孢适宜pH 4~8,孢子萌发最适pH 8;光暗交替适合菌丝生长与产孢,连续光照可抑制菌丝生长;菌丝生长量由少到多培养基顺序为CA、PCA、PDA、CMA、OA,而PDA上产孢最多;刺伤接种,病菌可侵染喜树(Camptotheca acuminata)等植物。黄脉爵床棒孢霉叶斑病病原为山扁豆生棒孢C. cassiicola,病菌易产孢,寄主广,潜育期短。该病菌侵染黄脉爵床为首次报道。  相似文献

为筛选出有效防治番茄叶霉病的药剂,采用生长速率法及平板涂布法测定6种三唑类杀菌剂对番茄叶霉病菌菌丝生长及分生孢子萌发和芽管伸长的毒力,评价其对番茄植株的安全性和对叶霉病的田间防效。结果表明,己唑醇、苯醚甲环唑、戊唑醇和氟硅唑对番茄叶霉病菌菌丝生长的毒力均较高,EC_(50)分别为0.50、0.55、0.80、2.42 mg/L,其次为腈菌唑和四氟醚唑,EC50分别为6.92、15.08 mg/L。6种杀菌剂抑制病菌分生孢子萌发及芽管伸长的作用均较弱,对芽管伸长的抑制活性高于对孢子萌发的抑制活性;戊唑醇和四氟醚唑抑制孢子萌发的作用相对较强,100 mg/L处理的抑制率为60%~70%,戊唑醇、四氟醚唑和己唑醇抑制芽管伸长的作用相对较强,100 mg/L处理的抑制率均在70%以上。己唑醇和戊唑醇200 mg/L处理番茄植株,显著抑制其株高,苯醚甲环唑和腈菌唑对其影响相对较小,这4种杀菌剂对番茄植株的叶色及形态均无明显影响;且这4种杀菌剂对番茄叶霉病的田间预防效果均高于治疗效果,其中150 mg/L己唑醇的预防效果和治疗效果均最高,分别为90.67%和85.58%;苯醚甲环唑的最低,300 mg/L时预防效果为80.16%,治疗效果为71.68%。  相似文献

为明确山东省辣椒炭疽病菌对吡唑醚菌酯的敏感性,延缓其抗性发展及降低防治成本,在该省主要辣椒产区采集并经单孢分离获得175株尖孢炭疽复合种,采用菌丝生长速率法和孢子萌发法测定了病原菌群体对吡唑醚菌酯的敏感性,同时测定了吡唑醚菌酯与甲基硫菌灵、多菌灵、代森锰锌、戊唑醇和咯菌腈5种杀菌剂分别混用对尖孢炭疽复合种的联合毒力。结果表明:吡唑醚菌酯对尖孢炭疽复合种菌丝生长和孢子萌发的EC50值范围分别为0.056~0.530和0.002~0.027 μg/mL,平均值为(0.273 ± 0.067)和(0.014 ± 0.019)μg/mL,其敏感性频率分布呈连续单峰曲线,符合正态分布,可作为田间抗性监测的敏感基线;不同采集地菌株对吡唑醚菌酯的敏感性存在一定差异,其中采自菏泽市的菌株敏感性最高,而采自潍坊市的菌株敏感性最低;吡唑醚菌酯与甲基硫菌灵按体积比1:3和3:1复配,对抑制尖孢炭疽复合种的菌丝生长和分生孢子萌发均表现为增效作用,其中按体积比1:3混用时增效作用最明显,增效系数分别是4.82和3.94。结果可为吡唑醚菌酯的合理使用提供理论依据。  相似文献

本文对辣椒褐斑病病原菌Cercospora capsici Heald et Wolf的生物学特性进行了研究,结果表明,不同的营养、温度、pH等条件对菌丝生长和孢子萌发有显著影响。病菌菌丝的生长以PDA培养基为最适;适宜温度为20~25℃,最适温度为25℃;最适pH为8.0~9.0,光照对菌丝生长没有明显的促进作用,菌丝致死温度及时间为55℃ 10 min。分生孢子萌发适宜碳源为1%的蔗糖溶液,适宜氮源为1%的甘氨酸溶液;孢子萌发适宜温度为20~30℃,最适温度25℃;最适pH为5~6,光照对孢子萌发没有明显的促进作用,分生孢子致死温度及时间为52℃10 min。  相似文献

为探讨硅酸钠在室内和田间对葡萄炭疽病的防治效果,以巨峰葡萄为材料,对葡萄炭疽菌孢子萌发、菌丝生长速率及室内和田间防治炭疽病效果等进行测定。结果表明:不同浓度硅酸钠对葡萄炭疽菌的孢子萌发和菌丝生长均有抑制作用,且随浓度升高抑制作用增强;室内接种炭疽病菌5 d后,15、20和25 mmol/L硅酸钠对葡萄炭疽病的防治效果分别为30.58%、44.37%和52.71%,且抑制效果差异极显著;田间最后1次喷药10 d后,15、20和25 mmol/L硅酸钠对葡萄炭疽病的防治效果分别为66.94%、67.16%和73.46%,喷施10%苯醚甲环唑6次的防治效果为76.84%,喷施3次的防治效果为64.29%;25 mmol/L硅酸钠与喷施6次苯醚甲环唑的防治效果差异不显著,但与喷施3次苯醚甲环唑的差异极显著。研究表明,在生长季可利用硅酸钠与常规药剂交替使用来防治葡萄炭疽病。  相似文献

The effects of temperature, relative humidity (RH), leaf wetness and leaf age on conidium germination were investigated for Spilocaea oleagina, the causal organism of olive leaf spot. Detached leaves of five ages (2, 4, 6, 8 and 10 weeks after emergence), six different temperatures (5, 10, 15, 20, 25 and 30°C), eight wetness periods (0, 6, 9, 12, 18, 24, 36 and 48 h), and three RH levels (60, 80 and 100%) were tested. Results showed that percentage germination decreased linearly in proportion to leaf age (P < 0.001), being 58% at 2 weeks and 35% at 10 weeks. A polynomial equation with linear term of leaf age was developed to describe the effect of leaf age on conidium germination. Temperature significantly (P < 0.001) affected frequencies of conidium germination on wet leaves held at 100% RH, with the effective range being 5 to 25°C. The percent germination was 16.1, 23.9, 38.8, 47.8 and 35.5% germination at 5, 10, 15, 20 and 25°C, respectively, after 24 h. Polynomial models adequately described the frequencies of conidium germination at these conditions over the wetness periods. The rate of germ tube elongation followed a similar trend, except that the optimum was 15°C, with final mean lengths of 175, 228, 248, 215 and 135 μm at 5, 10, 15, 20 and 25°C, respectively after 168 h. Polynomial models satisfactorily described the relationships between temperature and germ tube elongation. Formation of appressoria, when found, occurred 6 h after the first signs of germination. The percentage of germlings with appressoria increased with increasing temperature to a maximum of 43% at 15°C, with no appressoria formed at 25°C after 48 h of incubation. Increasing wetness duration caused increasing numbers of conidia to germinate at all temperatures tested (5–25°C). The minimum leaf wetness periods required for germination at 5, 10, 15, 20 and 25°C were 24, 12, 9, 9 and 12 h, respectively. At 20°C, a shorter wetness period (6 h) was sufficient if germinating conidia were then placed in 100% RH, but not at 80 or 60%. However, no conidia germinated without free water even after 48 h of incubation at 20°C and 100% RH. The models developed in this study should be validated under field conditions. They could be developed into a forecasting component of an integrated system for the control of olive leaf spot.  相似文献

不同杀菌剂对黄瓜靶斑病菌的毒力作用特性比较   总被引:2,自引:2,他引:2  
为筛选防治黄瓜靶斑病的有效药剂,采用菌丝生长速率法、孢子萌发法和芽管伸长法测定了19种杀菌剂对黄瓜靶斑病菌的毒力,探究了4种毒力较高杀菌剂对病原菌菌丝形态的影响。结果表明:氟啶胺、吡唑萘菌胺、咯菌腈和四霉素对黄瓜靶斑病菌菌丝生长、孢子萌发和芽管伸长各生长发育阶段均有较强的抑制活性,其中,4种药剂抑制菌丝生长的平均EC50值分别为0.516 3、1.538、1.605和0.648 2 mg/L,抑制孢子萌发的平均EC50值分别为0.164 4、1.359、1.127和0.002 5 mg/L,抑制芽管伸长的平均EC50值分别为0.189 6、0.144 7、0.060 4和0.001 5 mg/L;啶酰菌胺、百菌清和肟菌酯3种不同作用机制杀菌剂对黄瓜靶斑病菌孢子萌发和芽管伸长的抑制活性较强,3种药剂抑制孢子萌发的平均EC50值分别为1.564、0.373 0和0.021 3 mg/L,抑制芽管伸长的平均EC50值分别为0.629 5、0.233 4和0.405 0 mg/L,但对菌丝生长的抑制作用较弱,平均EC50大于16 mg/L;氟啶胺、吡唑萘菌胺、咯菌腈和四霉素可引起黄瓜靶斑病菌菌丝不同程度的异常生长,主要表现为菌丝隔膜间距变小、菌丝节间肿大、主菌丝上二次分枝发生频繁、菌丝变形黏连、菌丝新分枝处生长受抑制或分生孢子生长变形。研究表明,氟啶胺、吡唑萘菌胺、咯菌腈和四霉素对黄瓜靶斑病菌具有较高的毒力,在黄瓜靶斑病的田间防治中具有较好的开发应用潜力。  相似文献

采用大孔吸附树脂提取和硅胶柱层析分离技术,从黄瓜炭疽病菌Colletotrichum orbiculare代谢物中获得一个抗真菌化合物,采用高分辨质谱、核磁共振波谱和红外光谱等技术鉴定了其化学结构,并评价了其对番茄灰霉病菌、西瓜枯萎病菌、烟草赤星病菌、苹果炭疽病菌和玉米弯孢病菌的孢子萌发和菌丝生长的抑制作用。结果表明:该化合物为间二羟基苯甲酸大环内酯类抗生素——根赤壳菌素;其对所有供试真菌的孢子萌发均未表现出明显的抑制活性,但对番茄灰霉病菌Botrytis cinerea的菌丝生长表现出很强的抑制作用。  相似文献

An isolate of Trichoderma harzianum Rifai from an infected cacao pod produces and secretes nonanoic (pelargonic) acid into a liquid culture medium. Nonanoic acid (NA) was very inhibitory to spore germination and mycelial growth of two cacao pathogens, Crinipellis perniciosa Stahel and Moniliophthora roreri Cif. H.C. Evans. It was highly active causing 75% inhibition of spore germination in an in vitro assay at a rate as low as 0.09 μM for M. roreri and 0.92 μM for C. perniciosa. Mycelial growth was comparatively less sensitive to inhibition, but still there was a 75% reduction in growth with 0.62 μM in M. roreri and 151 μM NA in C. perniciosa. In contrast, NA did not affect Trichoderma mycelial growth or spore germination at concentrations that were inhibitory to the pathogens. 6-pentyl-α-pyrone was also produced and secreted into the medium by T. harzianum, however; it was not antagonistic to the cacao pathogens. Although a number of metabolites produced by Trichoderma spp. have been identified in the past, this is the first report of NA production and secretion by any Trichoderma. The results suggest that NA may play a role in the successful use of some Trichoderma spp. isolates in the biocontrol of fungal diseases of plants.  相似文献

为了研制高效、无毒的生物农药,采用自主分离的寡雄腐霉生防菌株Pythium oligandrum CQ2010制备发酵液,研究其对温室番茄生长及灰霉病的防治作用。结果表明,寡雄腐霉发酵液(Pythium oligandrum broth,POB)能显著提高番茄叶绿素含量,增强根系活力,增加氮、磷、钾吸收,促进植株生长,使生物量比对照提高65.14%;同时能激活与叶片抗病性相关的过氧化物酶和过氧化氢酶活性,诱导植株产生抗病性反应,提高抗病能力,其效果优于寡雄腐霉卵孢子制剂;而且POB能显著抑制离体灰葡萄孢菌的菌丝生长和孢子萌发;施用POB能使番茄灰霉病发病率下降22.28%~31.05%,病情指数从52.5(对照)下降至22.5~27.5,相对防治效果达47.62%~57.14%,叶片丙二醛含量显著降低,说明POB减轻了灰葡萄孢菌对细胞膜的伤害。表明POB兼具促进番茄生长及防治灰霉病的作用。  相似文献

报道了新型Qo I类杀菌剂苯噻菌酯(试验代号Y5247)的生物学特性。在含50μg/m L水杨肟酸(SHAM)旁路氧化专化性抑制剂的AEA培养基上,该杀菌剂抑制水稻纹枯病菌、稻瘟病菌、油菜菌核病菌及草莓灰霉病菌菌丝生长的有效中浓度(EC50)分别为0.004、0.009、0.016和0.023μg/m L;其抑制辣椒炭疽病菌和草莓灰霉病菌孢子萌发的EC50值分别为0.448和0.019μg/m L。苯噻菌酯对防治小麦白粉病具有保护和治疗作用,EC50值分别为0.991和1.823μg/m L。其在小麦叶片上内吸输导性差,但具有一定的渗透性、良好的粘着性、耐雨水冲刷和较长的持效期。用有效成分为25μg/m L的苯噻菌酯药液喷雾处理的麦苗,14 d后接种小麦白粉病菌,其防效仍达72.48%。  相似文献

红海榄赤斑病病原菌鉴定及其生物学特性研究   总被引:6,自引:0,他引:6  
 本文对来自红海榄(Rhizophora stylosa)赤斑病同一病斑上获得的2个菌株PⅠ-1和PⅡ-3进行了鉴定和生物学特性研究。结果表明,PⅡ-3菌株的致病力比PⅠ-1的稍强;2个菌株均具有一定的寄主专化性。经形态特征观察与rDNA-ITS序列分析,2个菌株均鉴定为异色拟盘多毛孢(Pestalotiopsis versicolor)。生物学特性测定结果表明,温度、pH值和光照对2个菌株菌丝生长及孢子萌发的影响差异不大,生长温度均为10~32℃(最适25℃),pH值为3.0~10.9(最适4.0);孢子萌发温度均为10~35℃(最适25~28℃),pH值为2.5~8.0(最适3.0);黑暗条件有利于2个菌株菌丝生长和孢子萌发。但2个菌株的产孢特性存在明显差异,其中PⅠ-1产孢的最适温度为28℃而PⅡ-3为20℃;PⅠ-1产孢的最适pH值为5.1而PⅡ-3为9.0;PⅠ-1产孢对光照不敏感,而PⅡ-3在黑暗条件下几乎不产孢。另外,在相似条件下,PⅠ-1产孢量比PⅡ-3大,而PⅡ-3菌丝生长速度比PⅠ-1快,孢子萌发率也比PⅠ-1高。由此可见,2个菌株PⅠ-1和PⅡ-3虽属于同一个种,但某些生物学特性存在一定的差异。  相似文献

The effect of environmental factors on the development of each stage ofPeronospora destructor (Berk.) Caspary on onions is reviewed. For sporulation to take place, a period of light must precede the period of darkness and high humidity in which spores are formed. Spores are discharged when the relative humidity (RH) is increasing or decreasing, and over a wide range of temperatures. Their discharge is triggered by exposure to red-infrared radiation and by vibration of the leaf. Dissemination of spores follows a daily periodic cycle and spores can be blown by wind over long distances. Duration of spore survival depends on temperature, RH and, especially, the absence of strong radiation. The rate of spore germination is highest at 10°C and declines with the rise in temperature. Germ tubes develop in liquid water, and a continuous period of wetness is required for infection to be completed. Systemic infection is common in cooler climates, where necks of onion bulbs are slow to dry. The principal sources of downy mildew infection by wind-borne spores are systemically infected propagation material, onion volunteer plants, and neighboring older crops.  相似文献

8种杀菌剂对核桃炭疽病病原菌胶孢炭疽菌的室内毒力   总被引:4,自引:1,他引:4  
由胶孢炭疽菌Colletotrichum gloeosporioides引起的核桃炭疽病是目前危害山东省核桃生产的主要病害。为筛选防治该病害的高效药剂,在确定了最适菌丝生长和分生孢子萌发温度的基础上,采用菌丝生长速率法和孢子萌发法,测定了8种常用杀菌剂对胶孢炭疽菌的毒力,评估了温度对咪鲜胺和戊唑醇毒力的影响,检测了17个胶孢炭疽菌菌株对咪鲜胺和戊唑醇的敏感性,同时评价了这2种杀菌剂的预防和治疗作用。结果显示:在胶孢炭疽菌菌丝生长和孢子萌发的最适温度28℃下,供试8种杀菌剂中,咪鲜胺、戊唑醇和三唑酮对供试菌株菌丝生长的抑制作用最强,平均EC50值分别为0.07、1.45和3.04 mg/L;异菌脲、代森锰锌和咪鲜胺对分生孢子萌发的抑制作用最强,平均EC50值分别为21.07、25.14和25.18 mg/L。咪鲜胺对菌丝生长的抑制作用受温度影响较小,而戊唑醇对菌丝生长的抑制作用随温度降低而增强;供试17个菌株对咪鲜胺和戊唑醇均有较高敏感性,平均EC50值分别为0.08 和 1.03 mg/L。建议在核桃炭疽病发病的不同时期,轮换使用咪鲜胺、戊唑醇、三唑酮、异菌脲和代森锰锌等杀菌剂,以有效控制该病害。  相似文献

This work presents an analysis of the relationship between components of partial disease resistance (PDR) detected using in vitro detached leaf and seed germination assays, inoculated with Microdochium majus, and Fusarium head blight (FHB) resistance to Fusarium graminearum assessed using point inoculation, termed Type II resistance. Relationships between in vitro-determined PDR components and FHB resistance using techniques which inoculate the wheat spike uniformly, termed Type I resistance (incidence and severity), have been reported previously. In this study shorter incubation periods, longer latent periods and shorter lesion lengths in the detached leaf assay and higher germination rates in the seed germination assay were related to greater FHB resistance measured by single point inoculation (Type II), collectively explaining 54% of the variation. Overall the relationships observed for Type II FHB resistance were similar to previous findings for Type I resistances. However, the relative magnitude of effects of the individual PDR components determined in vitro varied between FHB disease resistance parameters. Resistance in seed germination and latent period in the detached leaf assay were more strongly related to resistance assessed by point inoculation (Type II) and severity-Type I as opposed to incubation period which was most strongly related to disease incidence-Type I. The results provide evidence that individual components of partial disease resistance differentially affect aspects of FHB disease progression in the wheat spike. This work supports the view that the current model of types of resistance is an oversimplification of the interacting mechanisms underlying expression of FHB resistance.  相似文献

以蓝莓毛色二胞枝枯病菌假可可毛色二胞Lasiodiplodia pseudotheobromae为靶标菌,通过稀释平板法从山东青岛的蓝莓种植园根际土壤中分离出20株细菌,采用对峙平板法和菌丝生长速率法筛选得到一株对靶标菌具有明显抑制效果的拮抗菌株HMQAU140045,采用凹玻片法和菌丝生长速率法测定该菌株发酵滤液对靶标菌孢子萌发和菌丝生长的影响。结果表明,稀释50倍的发酵滤液的抑制率可达100%。离体枝条试验结果表明,发酵液不同组分对蓝莓毛色二胞枝枯病的防治效果均可达90%以上。菌丝生长速率法测定结果表明,该菌株抑真菌谱广,对包括4种蓝莓枝枯病菌在内的15种植物病原真菌具有良好的拮抗作用。通过形态观察、生理生化特性以及gyrB的序列分析,确定该菌株为解淀粉芽孢杆菌Bacillus amyloliquefaciens。试验结果表明,解淀粉芽孢杆菌菌株HMQAU140045具有较好的抑真菌活性。  相似文献

为探讨大蒜根系分泌物对烟草尖孢镰刀菌的抑菌活性,采用琼脂法培养大蒜,利用气相色谱—质谱技术(GC-MS)测定其主要成分,研究大蒜根系分泌物对烟草尖孢镰刀菌根腐病的影响。对菌丝生长速率和孢子萌发的测定表明,当大蒜根系分泌物粗提物浓度为1 g/mL时,其菌丝生长抑制率为30.56%,孢子萌发抑制率高达100.00%。大蒜根系分泌物主要成份2,6—二异丙基苯酚和邻苯二甲酸二丁酯分别对其菌丝生长和孢子萌发具有较强的抑菌作用。邻苯二甲酸二丁酯的防控效果较好,其浓度为5 mmol/L,接菌21 d后其病情指数为55.00,防控效果达32.38%。大蒜根系分泌物及主要成分对尖孢镰刀菌所引起的烟草根腐病有显著的抑制作用,这为研制含大蒜成分的植物保护剂和间作套种大蒜防控烟草尖孢镰刀菌根腐病提供科学参考。  相似文献

对山葵墨入病病株进行分离鉴定,明确其病原菌为山葵茎点霉菌(Phoma wasabiae Yokogi)。对该菌生物学特性研究结果表明,适宜该菌生长的培养基为PDA培养基,不同的温度、pH、光照等条件对山葵茎点霉菌的菌丝生长和产孢有显著的影响。该菌适宜的生长温度为19-25℃,最适温度为22℃;适宜生长的pH为6.0-9.0,最适pH7-8;该菌在黑暗条件下比在光照和光暗交替处理下生长快,且产孢的能力相对强;致死温度为55℃。10min。  相似文献
