Excess consumption of selenium (Se) accumulator plants can result in selenium intoxication. The objective of the study reported here was to compare the acute toxicosis caused by organic selenium (selenomethionine) found in plants with that caused by the supplemental, inorganic form of selenium (sodium selenite). Lambs were orally administered a single dose of selenium as either sodium selenite or selenomethionine and were monitored for 7 days, after which they were euthanized and necropsied. Twelve randomly assigned treatment groups consisted of animals given 0, 1, 2, 3, or 4 mg of Se/kg of body weight as sodium selenite, or 0, 1, 2, 3, 4, 6, or 8 mg of Se/kg as selenomethionine. Sodium selenite at dosages of 2, 3, and 4 mg/kg, as well as selenomethionine at dosages of 4, 6, and 8 mg/kg resulted in tachypnea and/or respiratory distress following minimal exercise. Severity and time to recovery varied, and were dose dependent. Major histopathologic findings in animals of the high-dose groups included multifocal myocardial necrosis and pulmonary alveolar vasculitis with pulmonary edema and hemorrhage. Analysis of liver, kidney cortex, heart, blood, and serum revealed linear, dose-dependent increases in selenium concentration. However, tissue selenium concentration in selenomethionine-treated lambs were significantly greater than that in lambs treated with equivalent doses of sodium selenite. To estimate the oxidative effects of these selenium compounds in vivo, liver vitamin E concentration also was measured. Sodium selenite, but not selenomethionine administration resulted in decreased liver vitamin E concentration. Results of this study indicate that the chemical form of the ingested Se must be known to adequately interpret tissue, blood, and serum Se concentrations.  相似文献

Twenty-one 6 months old female lambs were divided into 7 groups and fed a basal diet containing 0.13 mg Se/kg. The basal diet was further supplemented with 0, 0.1, 0.5 or 1.0 mg Se/kg either as sodium selenite or as selenomethionine, and was fed for 10 weeks. Both feed additives produced an increase in the selenium concentration in the tissues analysed. Significant correlations were found between the concentrations of selenomethionine or sodium selenite added to the feed and the subsequent tissue levels. However, the selenium levels seemed to plateau at approximately 0.5 mg Se/kg of supplemented sodium selenite. The total glutathione peroxidase (GSH-Px) activity of the tissues increased when the selenium supplementation increased from 0 to 0.1 mg/kg for both selenium compounds. With further increase in selenium supplementation the GSH-Px activity in the tissues plateaued except in the blood where the activity continued to rise with increasing selenomethionine supplementation. The selenium dependent GSH-Px activity in the liver rose with increasing selenomethionine supplementation, but approached a plateau when 0.1 mg Se/kg as sodium selenite was added to the feed. The selenium concentration in whole blood responded more rapidly to the selenium supplementation than did GSH-Px activity. The experiment indicates that the optimal selenium concentration in the feed is considerably higher than 0.1 mg Se/kg, and that selenium levels of 1.0 mg/kg in the feed do not result in any risk for the animals or the consumers of the products.Key words: dietary selenium, lambs, selenium concentrations, glutathione peroxidase activities, tissues  相似文献

Forty-eight Norwegian bred White Leghorn chickens were divided into 6 groups and fed a basal diet containing 0.30 mig Se/kg supplemented with 0, 0.1, 0.5, 1.0, 3.0 or 6.0 mg Se/ kg in the form of selenomethionine for 18 weeks. A supplement of only 0.1 mg Se/kg induced significantly higher selenium concentrations in breast muscle and eggs, particularly in the egg white. The increase of selenium in the tissue and egg was proportional to the amounts of selenomethionine added to the feed. In the group given 6.0 mg Se/kg, the selenium concentrations in all tissues and eggs analysed ranged from 4.8 to 7.3 μg Se/g. No signs of toxic effects were observed even at the highest intake of selenium. Excess supply of selenium as selenomethionine to chickens was shown to be more potent than sodium selenite in raising the selenium concentration in tissues and eggs. A supplementation up to 10 times the requirement did not increase the levels of selenium in poultry products to such a degree that they could be considered as a potential risk for human consumption.Key words: dietary selenium, laying hens, selenium concentrations, tissues, eggs  相似文献

1. Diets containing supplements of selenium (10 mg/kg as sodium selenite), copper (500 mg/kg as copper (II) sulphate) and sodium nitroprusside (SNP 300 mg/kg) as a source of cyanide, were fed singly and in all possible combinations to chicks from 14 to 35 d of age.

2. Both copper and SNP individually alleviated the growth depression caused by excess selenium but interacted adversely with one another. The effect of SNP was to decrease liver selenium but copper increased it. Despite this contrast it is suggested that both achieve their beneficial effects through influences on the fraction of liver selenium that can be reduced to volatile forms by hydrochloric acid and zinc.

3. In a second experiment the effects of diets containing additional selenium (2 mg/kg as sodium selenite or selenomethionine) with or without additional copper (100 mg/kg) or SNP (100 mg/kg) on selenium incorporation into eggs were compared.

4. SNP reduced incorporation from selenite whereas copper had no effect. However, copper reduced the incorporation from selenomethionine into the protein fractions of egg white as much as SNP.  相似文献

The effect of selenium (Se) on sheep lymphocyte response to mitogens was studied. In an indoor experiment lambs were fed a basal diet containing 0.13 mg Se kg-1, and supplemented with, respectively, 0.1 or 0.5 mg Se kg-1, either as sodium selenite or as selenomethionine. Enhancement of the proliferative response of lymphocytes to phytohaemagglutinin (PHA), pokeweed mitogen (PWM) and concanavalin A was found in lambs following selenium supplementation at the lower levels. The highest dietary selenium content, however, induced decreased mitogen response. Transient increases in lymphocyte response to PHA and PWM by ewes supplemented with selenium was demonstrated in one field study and a combined effect of selenium and vitamin E was seen in another. There was no stimulatory effect on the mitogen response of lymphocytes from sheep supplemented with dietary vitamin E alone.  相似文献

The effect of dietary riboflavin (B2) supplementation and selenium (Se) source on the performance and Se metabolism of weanling pigs was studied. Pigs fed a B2-supplemented (10 mg/kg) casein-glucose diet for 18 d gained faster than pigs fed the B2-unsupplemented diet. Percentage active erythrocyte glutathione reductase (GR) declined rapidly when pigs were placed on the B2-unsupplemented diet and was lower (P less than .01) than that of B2-supplemented pigs after 12 d on test. Percentage active erythrocyte GR values fell below 50% before other B2 deficiency signs became evident. Supplementation of diets with 10 mg B2/kg resulted in increased kidney and muscle glutathione peroxidase (GSH-Px) activity. The Se concentration of liver and heart increased and plasma Se levels decreased with dietary B2 supplementation. Riboflavin supplementation and Se source did not alter apparent Se absorption, but B2 supplementation decreased urinary Se and thus increased Se retention. Also, there was less urinary Se excretion when selenomethionine was the dietary Se source and consequently more Se was retained than when sodium selenite was the dietary Se source. In a final trial, B2 supplementation increased kidney, muscle, heart and brain GSH-Px activity when sodium selenite was the dietary Se source, but not when selenomethionine was the dietary Se source.  相似文献

Young chicks exhibited a 61% reduction in weight gain when a corn-soybean meal diet was supplemented with 15 mg/kg Se provided as Na selenite. The same level of Se provided as selenomethionine depressed weight gain by 32%. Supplementing the high selenite diet with isoarsenous (14 mg/kg As) additions of As2O5, As2O3, phenylarsonic acid, phenylarsine oxide and roxarsone ameliorated the Se-induced growth depression: As2O5 almost totally restored growth rate; As2O3, phenylarsonic acid and phenylarsine oxide gave intermediate responses; and roxarsone gave only a small ameliorative growth response. Arsanilic acid was without effect in stimulating growth rate of selenite-intoxicated chicks. Dietary addition of .4% L-cysteine produced a growth response in selenite intoxicated chicks that was somewhat greater than that obtained with roxarsone; administering both roxarsone and cysteine corrected growth better than either compound given singly. Both roxarsone and As2O5 also effectively ameliorated the Se-toxicity growth depression caused by selenomethionine (15 mg Se/kg) supplementation, but cysteine showed no efficacy against morbidity caused by this form of Se. Liver Se concentration was elevated 10-fold by selenite and 25-fold by selenomethionine supplementation. The arsenic compounds had varying effects on liver Se, whereas cysteine tended to increase Se concentration. These findings suggest that both inorganic and organic arsenicals as well as cysteine ameliorate selenium toxicity by different mechanisms.  相似文献

本试验旨在研究酵母硒和亚硒酸钠作为硒源对产蛋高峰期蛋鸡生产性能、蛋硒含量、蛋品质以及储存期内蛋品质的影响。选用27周龄600只产蛋高峰期海兰褐蛋鸡,随机分为2个处理,每个处理3个重复,每个重复100只鸡。在日粮中分别添加0.2 mg/kg的亚硒酸钠和酵母硒(试验日粮实测硒含量分别为0.227、0.233 mg/kg),预试期1周,正试期8周,对鸡蛋28 d内的鸡蛋蛋品质进行跟踪检测。结果表明:酵母硒和亚硒酸钠对海兰褐蛋鸡生产性能的影响不显著;与亚硒酸钠组相比,酵母硒组对28 d鸡蛋蛋品质无显著影响,但可显著提高56d鸡蛋的蛋白高度和哈氏单位(P<0.05);与添加亚硒酸钠相比,添加酵母硒减缓了储存期鸡蛋哈氏单位的降低,有利于延长鸡蛋货架期;酵母硒组蛋黄、蛋清中硒含量显著高于亚硒酸钠组(P<0.05)。由此可见,相较于亚硒酸钠,饲喂含酵母硒的日粮在延长鸡蛋货架期、提高蛋黄和蛋清中的硒沉积方面效果更好。  相似文献

试验旨在研究添加不同水平的亚硒酸钠和酵母硒对产蛋鸡生产性能和蛋品质的影响。试验选用23周龄北京油鸡和罗曼粉杂交一代产蛋母鸡648只,随机分为12组,每组6个重复,每个重复9只鸡,进行为期8周的饲养试验。试验1~6组在基础日粮中分别添加含硒0.3、1.0、2.0、3.0、4.0、5.0 mg/kg的亚硒酸钠,试验7~12组在基础日粮中分别添加含硒0.3、1.0、2.0、3.0、4.0、5.0 mg/kg的酵母硒。结果表明,在产蛋鸡的饲料中添加5.0 mg/kg以内的亚硒酸钠或酵母硒都有提高产蛋率的趋势(P>0.05),且对蛋品质无明显不良影响。若以提高产蛋率为目的,亚硒酸钠的添加水平以2.0 mg/kg为宜,酵母硒的添加水平以3.0 mg/kg为宜。酵母硒的使用效果优于亚硒酸钠。酵母硒组采食量和蛋壳强度显著高于亚硒酸钠组(P<0.05),亚硒酸钠组蛋白高度和哈氏单位显著高于酵母硒组(P<0.05)。  相似文献

This paper describes 3 experiments comparing the effect of 10, 25 and 40 mg Se/kg, as sodium selenite, in mineral mixtures and salt licks fed to sheep. The supplement was given during the indoor season from October to May to 7 different flocks, each consisting of 50 to 100 sheep, in areas with selenium deficiency problems. The average selenium level in the basic diets did not exceed 0.05 mg/kg. Selenium status was monitored in the blood of ewes and lambs, and in milk. Blood selenium in lambs correlated well with blood selenium in their dams (r = 0.85). Selenium levels in milk on day 1 (colostrum) correlated well with selenium levels in dams (r = 0.92) and in offspring (r = 0.87). Statistically significant differences were found between the different flocks. In areas with extreme selenium deficiency, 10 mg Se/kg in mineral mixtures and salt licks proved insufficient. A content of 25 mg Se/kg, providing a daily intake of about 0.4 mg selenium, resulted in selenium levels in ewes’ blood, ewes’ milk and in the offspring that should prevent selenium deficiency disease without causing any toxic effects.  相似文献

The objective of this study was to evaluate neutrophil oxidative metabolism and haemogram in sheep experimentally infected with Haemonchus contortus and supplemented with selenium and vitamin E. Twenty male Corriedale sheep were utilised and distributed into four experimental groups each with five animals infected with larvae: G1‐ supplemented with sodium selenite, 0.2 mg/kg body weight (bw) given intramuscularly (IM); G2‐ supplemented with sodium selenite and vitamin E, 0.2 mg/kg bw and 2000 IU per animal, respectively, both IM; G3‐ supplemented with vitamin E, 2000 IU per animal IM; G4‐ not supplemented. A haemogram and the number of parasite eggs were determined in samples of blood and faeces, respectively, on days zero (T0), 20 (T1), 40 (T2) and 60 (T3) and nitroblue tetrazolium (NBT) assays were performed on heparinized blood samples taken on days zero, 30 and 60. A lower total leucocyte count was detected in G1 in relation to G4 at T4. Lymphocytes were reduced in G1 in relation to G3 and G4 at T3. In both non‐stimulated (NBT‐NS) and stimulated (NBT‐S) dye reduction assays, there was reduced activity at 60 days, in relation to other times in the groups treated with selenium (G1 and G2). On the basis of results obtained, we conclude that supplementation with selenium provides better antioxidant protection to neutrophils.  相似文献

A bioavailability study was conducted in lambs following intravenous and oral administration of sodium selenite (0.4 mg selenium/kg body weight) with and without concurrent oral monensin. Two- or three-compartment open models with first-order absorption after oral administration adequately described plasma selenium disposition irrespective of whether monensin was being administered. No significant differences were observed between groups of lambs receiving intravenous selenium with or without monensin with respect to distribution or elimination half-lives of selenium, areas under the concentration-time curve (AUC), volumes of distribution (Vd(ss)), or clearances (Cl). In lambs given selenium per os, no significant differences were observed with animals receiving monensin as well with respect to absorption and elimination half-lives, Vd(ss), or the time at which peak selenium concentrations occurred (tmax). However, peak selenium levels (Cmax) and AUC values were significantly higher in the group given monensin. The bioavailability of selenium with and without monensin was estimated to be 60% and 43%, respectively.  相似文献

1. The deposition of selenium in egg components has been investigated in two experiments in which sodium selenite was added to a conventional cereal‐based layer diet.

2. Addition of graded amounts of selenite up to 4 mg Se/kg resulted in linear increases in the selenium content of egg white and yolk, and in protein fractions derived from them. The presence of selenium in yolk phosvitin indicates that deposition is not dependent upon the presence of cysteine.

3. Addition of sodium nitroprusside at 0–15 and 0–3 g/kg to diets having an addition of selenite at the highest concentration, 4 mg Se/kg, resulted in substantial reductions in the selenium concentration in egg components.

4. Samples from eggs laid by hens receiving a diet containing an additional 8 mg selenite Se/kg were subjected to dialysis against sodium hydroxide or cysteine, or subjected to reduction with hydrochloric acid and zinc under anaerobic conditions. Comparisons were made with similar samples prepared from eggs laid by hens on the control diet.

5. Both sodium hydroxide and cysteine were more effective at extracting additional diet‐derived selenium from whole white than from whole yolk. The proportion of selenium that could be extracted from the water‐soluble or the high density fractions of yolk by either reagent was similar for both control and high selenium samples. However, neither reagent was effective at removing selenium from the ovalbumin or globin fractions of white from control eggs but substantial amounts were extracted from high selenium samples.

6. Most of the selenium was present in non‐reducible forms in all samples. There was significantly more reducible selenium in ovalbumin from control eggs than from all other samples but even so non‐reducible selenium accounted for two thirds of the selenium present.

7. The differential responses to chemical treatment suggest that selenium can be deposited in eggs in an unspecified number of different forms. These have still to be characterised but site of formation of egg proteins, liver or oviduct, has a bearing on the forms of selenium deposited.  相似文献

The LD50-values of sodium selenite after i/m injection to rabbits was determined by means of probit-regression straight line. Simultaneously a combination of vitamin E and sodium selenite was tested for toxicity in order to examine the effect of vitamin E on the selenium toxicity. The LD50/24h was 2.53 mg sodium selenite/kg body mass and in combination with vitamin E (30 IU vitamin E/5 mg NaSeO3) 2.73 mg/kg. Furthermore the selenium concentration was determined in blood, liver, kidney and muscles of animals which died or were killed after 24 h respectively. Not only chemical analysis but also relations of time-dose-efficiency and significantly increased LD50-values show a certain protective effect of vitamin E on selenium intoxication.  相似文献

Three experiments were carried out, using sheep fed a marginally low selenium diet, to study the effect of selenium supplementation on the antibody response to tetanus toxoid and on the serum IgG concentration. Six groups of three six-month-old lambs were fed a basal diet containing 0.13 mg Se kg-1 supplemented with either 0.1, 0.5 or 1.0 mg Se kg-1, as sodium selenite or as selenomethionine. These animals generally showed enhanced antibody response to tetanus toxoid, parainfluenza-3 virus and Corynebacterium pseudotuberculosis, and their total serum IgG concentrations were higher than in unsupplemented control animals although few responses were statistically significant. In two field studies significantly higher titres to tetanus toxoid were detected in ewes injected with 100 mg selenium as barium selenate, although no influence on serum IgG concentrations was detected. Lambs from selenium supplemented ewes had significantly higher titres to tetanus toxoid than lambs from ewes in the control group. Dietary vitamin E supplementation had a similar effect on the antibody response to tetanus toxoid in ewes, though no additive effect was seen when vitamin E was given together with selenium.  相似文献

The effects of three dietary selenium (Se) levels (0.15, 0.35 and 0.5 mg/kg dry matter (dm) and of two Se-compounds (sodium selenite and Se-yeast) on the Se-status, liver function and claw health were studied using 36 fattening bulls in a two-factorial feeding trial that lasted 16 weeks. The claw health was assessed macroscopically and microscopically. Compared to the two control diets containing 0.15 mg Se/kg dm, the intake of the diets containing 0.35 and 0.50 mg Se/kg dm significantly (P < 0.05) increased the Se-concentration in serum, hair, liver and skeletal muscle. Compared to sodium selenite the intake of Se-yeast resulted in significantly (P < 0.05) higher Se-concentration in serum, liver and hair. Concerning the claw horn quality, there was no significant difference between the different groups; the animals receiving organic Se tended to have a better histological score (P = 0.06) at the coronary band than the groups fed with sodium selenite. The serum vitamin E level decreased significantly (P < 0.05) with increasing Se-intake, which had no influence (P > 0.1) on growth and liver function parameters. With the exception of the decrease of the serum vitamin E level indicating an oxidative stress caused by a high Se-intake, no negative effects of dietary selenium exceeding recommended levels for 4 months were observed.  相似文献

通过在公鸡日粮中添加不同水平亚硒酸钠,研究硒对公鸡睾丸组织结构及精原干细胞增殖的影响,以期为禽类繁殖性能的研究提供实验依据。选取80只体重接近、健康无疾病的海兰白成年公鸡,随机分成4组,每组20只,分别在日粮中添加0,0.5,1和2mg/kg的亚硒酸钠。通过免疫荧光观察精原干细胞数量。结果显示:日粮中添加适量的硒对睾丸的组织结构及精原干细胞的增殖有显著影响(P〈0.05),其中0.5mg/kg组显著高于对照组、1mg/kg组和2mg/kg组。结论:适量的硒可以促进鸡睾丸的发育及精原干细胞的增殖,过量的硒对精原干细胞的增殖有一定的抑制作用。  相似文献

选择高氟区生长的高氟黄牛,通过扫描电镜观察了红细胞损伤的超微结构。结果表明:长期处在高氟环境中的黄牛,红细胞变形,出现数目不等的棘球状、多角形或星茫状突起,发生了棘红细胞胞增多症(acan-thocytosis)。通过饲料单独添加硒(亚硒酸钠)0.25mg/kg、铜(硫酸铜)15mg/kg和联合添加硒0.25mg/kg+铜15mg/kg+镁(硫酸镁)1mg/kg,连续饲喂83d,结果加硒组高氟黄牛的变形红细胞不同程度地恢复原形,加硒铜镁  相似文献
