AIM:To observed the effect of quercetin on NLRP3 inflammasome activation in the rats with diabetic cardiomyopathy (DCM) and its protective effect on the myocardium. METHODS:Male SD rats (n=40) were randomly divided into normal control group (n=10) and model group (n=30). The rats in model group were intraperitoneally injected with streptozotocin at 60 mg/kg to establish the model of diabetes mellitus (DM). Blood glucose was measured weekly. After 4 weeks, the rats with random blood glucose ≥ 16.6 mmol/L were selected as DM animals. The rats with DM were randomly divided into 3 groups:DM group, DM+vehicle group and DM+quercetin group. The rats in DM+quercetin group were intragastric infusion with quercetin at 100 mg/kg per day. The cardiac function was measured at the end of the 16th week. The methods of Masson staining and HE staining were used to observe the morphological changes of the myocardial tissues. Western blot, ELISA and immunohistochemistry were used to observe the changes of NLRP3, ASC, caspase-1, interleukin (IL)-1β and IL-18. TUNEL staining was used to observe myocardial apoptosis. RESULTS:Quercetin significantly inhibited the activation of NLRP3 inflammasome in the myocardium of the DM rats (P<0.05). The levels of IL-1β and IL-18 in DM+quercetin group were significantly decreased, quercetin reduced cardiac tissue apoptosis, and the cardiac function in DM+quercetin group was significantly improved (P<0.05) compared with DM group and DM+vehicle grpup. CONCLUSION:Quercetin significantly inhibits the activation of NLRP3 inflammasome, and reduces the levels of inflammation and myocardial apoptosis, thus protecting the heart function of DCM rats.  相似文献

AIM: To investigate the serum levels of inflammatory factors and adiponectin in type 2 diabetic retinopathy (DR) patients.METHODS: One hundred and ten cases of type 2 diabetic patients were divided into 3 groups: no diabetic retinopathy group (DM, n=35), non-proliferative diabetic retinopathy group (NPDR, n=45), and proliferative diabetic retinopathy group (PDR, n=30). Other 40 normal persons served as controls (NC group). The physical examinated was performed for each patient. Serum levels of fasting plasma glucose (FPG), glycated hemoglobin A1c (HbA1c), 2 h postprandial plasma glucose (2hPG), fasting insulin (FINS), total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), adiponectin, intercellular adhesion molecule-1(ICAM-1), tumor necrosis factor-α (TNF-α) and high-sensitivity C-reactive protein (hs-CRP) were measured. Insulin resistance index (HOMA-IR) was also calculated.RESULTS: The systolic blood pressure, body-mass index, waist-hip ratio, serum levels of TG, LDL-C, FPG, 2hPG, HbA1c, ICAM-1, TNF-α, hs-CRP and HOMA-IR were significantly higher in DM group, NPDR group and PDR group than those in NC group (P<0.05). The systolic blood pressure, serum levels of ICAM-1, TNF-α, hs-CRP and HOMA-IR were higher in NPDR group and PDR group than those in DM group (P<0.05). The serum concentration of adiponectin was lower in DM group, NPDR group and PDR group than that in NC group (P<0.05), and that was also lower in NPDR group and PDR group than that in DM group (P<0.05). The negative correlations between adiponectin and ICAM-1 (r=-0.735,P<0.01), TNF-α (r=-0.781,P<0.01), hs-CRP (r=-0.768, P<0.01) or HOMA-IR (r=-0.752, P<0.01) were observed. The relationships between HOMA-IR and ICAM-1 (r=0.857,P<0.01), TNF-α (r=-0.906, P<0.01) or hs-CRP (r=-0.888,P<0.01) were positive.CONCLUSION: The results suggest that inflammatory refactors and adiponectin play important roles in the pathophysiology and progression of DR. The protective effects of adiponectin on DR may be related with its anti-inflammatory reactions to improve insulin resistant.  相似文献

AIM: To explore the relationship between 3-nitrotyrosine (3-NT) level in hearts or blood and myocardial cell apoptosis in rat diabetic cardiomyopathy (DCM). METHODS: Sixty Sprague-Dawley (SD) rats (male, 8-week-old) were randomly divided into 4 groups: normal group, diabetic cardiomyopathy group (DCM group), diabetic rats treated with valsartan (40 mg·kg-1·d-1, D+V group) and DCM rats treated with valsartan (40 mg·kg-1·d-1, DCM+V group). Apoptotic index (AI) of rat cardiac myocytes was examined by TUNEL. The expression index (EI) of 3-NT in rat cardiac myocytes was examined by immunohistochemistry. The 3-NT concentration in rat serum was examined by ELISA. RESULTS: (1) Significant differences of the heart weight indexes among the 4 groups were observed (P<0.01). The heart weight indexes in DCM group and DCM+V group were higher than those in normal group and D+V group (P<0.01). (2) The EI of 3-NT in the cardiac myocytes was positively correlated with the AI of the cardiac myocytes in the same group (P<0.01), but the concentration of 3-NT in blood had no correlation with the AI of cardiac myocytes (P>0.05). (3) The difference of AI of cardiac myocytes among the 4 groups had statistical significance (P<0.01). The arrangement from high to low of AI was DCM group > D+V group and DCM+V group > N group (P<0.05). (4) The EI of 3-NT in DCM group was the highest as compared to other groups (P<0.05). (5) No statistical difference of 3-NT concentration in blood among the 4 groups was observed (P>0.05). CONCLUSION: (1) The expression of 3-NT in DCM myocardial tissues in SD rats is significantly increased and closely correlated with the apoptosis in myocardial cells. Valsartan inhibits 3-NT expression in DCM myocardial cells, thus inhibits the DCM myocardium apoptosis. (2) The 3-NT level in blood can not be true for reflection of 3-NT expression in DCM myocardial tissues and its effect on myocardial cell apoptosis.  相似文献

AIM: To observe the changes of heart function and the expression of serum cardiac troponin I(cTnI) in early type 2 diabetic rats, and to explore the role of cTnI in the development of type 2 diabetes and early diabetic cardiomyopathy.METHODS: The type 2 diabetes rat model was established by an injection of streptozotocin after high fat diet(5 weeks). The rats were randomly divided into control group, model group of 2 weeks, and model group of 4 weeks. M-mode echocardiography was performed for echocardiographic measurements. Fasting blood glucose(FBG), total cholesterol(TC), triglyceride(TG), high density lipoprotein-cholesterol(HDL-C), low density lipoprotein- cholesterol(LDL-C), fasting insulin(FINS) and cTnI levels were tested. HE staining was used to observe the pathological changes of myocardial structures. The alteration of cTnI in myocardium was determined by Western blot.RESULTS: Compared with normal group, the levels of TC, TG and LDL-C in type 2 diabetic rats were significantly increased, HDL-C levels were significantly reduced. Cardiac histological analysis revealed that type 2 diabetes induced cardiomyocytes degeneration and necrosis. The expression of cTnI increased significantly in diabetic groups compared to control group, and that in model group of 4 weeks increased far more than that in model group of 2 weeks(P<0.05).CONCLUSION: The increased level of cTnI and the change of the heart function may be associated with the development diabetic cardiomyopathy. These changes are valuable for the early clinical diagnosis of myocardial injury in diabetic cardiomyopathy.  相似文献

AIM: To explore the effect of Yiqi-Yangyin recipe on myocardial ischemia-reperfusion injury (MIRI) in rats with diabetes mellitus (DM) and the possible mechanism. METHODS: The rats were divided into normal group (control group), DM sham operation (DM-S) group, DM+MIRI group, low-, medium-and high-dose Yiqi-Yang-yin recipe (TL, TM and TH) groups (7.5, 15 and 30 g/kg decoction of Yiqi-Yangyin recipe by gavage), and Nrf2 inhibitor (bardoxolone methyl) group (30 mg/kg bardoxolone methyl by intragastric administration). The gavage volume was 1 mL/kg. There were 15 rats in each group, and they were administered continuously for 7 d. The tail vein blood was collec-ted after the last administration to detect the blood sugar and lipid levels in the rats. The serum levels of cardiac troponin I (cTnI), tumor necrosis factor-α (TNF-α), interleukin (IL)-1β and IL-10 were measured by ELISA. Echocardiography was used to detect the changes of cardiac function in the rats after blood collection. After cardiac function test, the rats were sacrificed to obtain cardiac tissues, and the volume changes of myocardial infarction were assessed by triphenylte-trazole chloride staining. The histopathological changes of myocardium was observed by HE staining. The cardiomyocyte apoptosis was determined by TUNEL assay. The protein levels of phosphorylated extracellular signal-regulated kinase (p-ERK), nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) in the myocardium were determined by Western blot. The myocardial activity of superoxide dismutase (SOD) was measured by nitro blue tetrazolium method, the content of malondialdehyde (MDA) was tested by thiobarbituric acid method, and the production of reactive oxygen species (ROS) was analyzed by iron ion reduction method. RESULTS: Compared with control group, the levels of fasting blood glucose (FBG), total cholesterol (TC) and triglyceride (TG) in DM-S group and DM+MIRI group were significantly elevated, while the level of high-density lipoprotein cholesterol (HDL-C) was significantly lowered (P<0.05). Compared with DM-S group and DM+MIRI group, the levels of FBG, TC, TG in TL, TM, TH and bardoxolone methyl groups were significantly decreased, while HDL-C level was significantly increased (P<0.05). Compared with control group and DM-S group, heart rate (HR) and left ventricular end-diastolic pressure (LVEDP) were increased in DM+MIRI group, mean arterial pressure (MAP), left ventricular systolic pressure (LVSP) and left ventricular ejection fraction (LVEF) were decreased, serum levels of cTnI, TNF-α, IL-1β and IL-10 were increased, the myocardial infarction volume percentage was increased, the myocardial cell breakage and necrosis were increased, the myocardial cell apoptotic rate was increased, the protein levels of p-ERK1/2, Nrf2 and HO-1 were decreased, MDA and ROS levels were increased, and the activity of SOD was decreased (P<0.05). Compared with DM+MIRI group, HR and LVEDP were decreased in TL, TM, TH and bardoxolone methyl groups, MAP, LVSP and LVEF were increased, the serum levels of cTnI, TNF-α, IL-1β and IL-10 were decreased, the myocardial infarction volume percentage was decreased, myocardial cell breakage and necrosis were decreased, myocardial cell apoptotic rate was decreased, the protein levels of p-ERK1/2, Nrf2 and HO-1 were increased, the MDA and ROS levels were decreased, and the activity of SOD was increased (P<0.05). CONCLUSION: Yiqi-Yangyin recipe protects the myocardial tissue of DM+MIRI rats from injury and reduces the oxidative stress level, which may be achieved by activating ERK/Nrf2/HO-1 pathway.  相似文献

AIM: To observed the protective effect of diminazene aceturate (DIZE), an angiotensin-converting enzyme 2 (ACE2) activator, on rats with diabetic cardiomyopathy (DCM). METHODS: Male Wistar rats (n=30) were randomly divided into normal control group, DCM group and DIZE treatment group (DIZE group). The rats in DCM group and DIZE group were intraperitoneally injected with streptozotocin (65 mg/kg) to establish diabetic model. After 12 weeks, the diabetic rats were infused with DIZE at 15 mg·kg-1·d-1 or the same volume of saline for 4 weeks using osmotic minipump. The cardiac function was measured at the end of the 16th week. The methods of Mason staining and HE staining were used to observe the morphological changes of the myocardial tissue. Western blot, ELISA and immunohistochemistry were used to observe the changes of ACE2, angiotensin (Ang)Ⅱ, Ang-(1-7), interleukin (IL)-1, IL-6 and connective tissue growth factor (CTGF). RESULTS: DIZE significantly improved the expression of ACE2 in diabetic rats (P<0.05). Compared with DCM group, the levels of IL-1 and IL-6 in DIZE group were significantly decreased, and the cardiac function in DIZE group was significantly improved (P<0.05). CONCLUSION: ACE2 endogenous agonist DIZE significantly increases the ACE2 level and reduces the level of inflammation, thus protecting the heart function of DCM rats.  相似文献

ATM: To explore the influence of Tangshen formula (TS) on endothelial function and blood rheology in diabetic nephropathy (DN) rats. METHODS: The DN rat model was established by intravenous injection of low-dose (30 mg/kg) streptozotocin (STZ) after having the high-fat/high-glucose diets for one month. The animals were divi-ded into DN model group, TS group and valsartan group. Fasting blood glucose (FBG), serum total cholesterol (TC), serum triglyceride (TG), renal cortex blood flow and hemorheology were monitored. The content of von Willebrand's factor (vWF) and plasminogen activator inhibitor-1 (PAI-1) in the serum was determined by ELISA. RESULTS: Compared with normal group, FBG,TC,TG, vWF and PAI-1 were increased in DN model group (P<0.05), and no significant difference of FBG was observed. Compared with normal group, plasma viscosity, Casson viscosity, whole blood high/medium/low-shear viscosity, erythrocyte aggregation index, erythrocyte rigidity index and erythrocyte electrophoresis time were increased, and erythrocyte deformation index was decreased in DN model group (P<0.01). Compared with DN model group, plasma viscosity, Casson viscosity, whole blood high/medium/low-shear viscosity, erythrocyte aggregation index, erythrocyte rigidity index and erythrocyte electrophoresis time were decreased (P<0.05), but there was no significant difference for erythrocyte deformation index in TS group. Compared with normal group, the renal cortex microcirculation blood flow in DN model group was significantly decreased. Compared with DN model group, the renal cortex microcirculation blood flow was significantly increased in TS group (P<0.05), and no significant change in valsartan group was found.CONCLUSION: Tangshen formula plays a protective role in the kidney of diabetic rats by improving the blood rheology and endothelial function, thus ameliorating the renal cortex microcirculation blood flow in experimental diabetic rats.  相似文献

AIM: To observe the effects of liraglutide on the level of microRNA-33 (miR-33) and the expression of AMP-activated protein kinase (AMPK) and apoptosis-related proteins in mice with type 2 diabetes mellitus (T2DM), and to explore its possible mechanism. METHODS: High-fat diet and intraperitoneal injection of streptozocin were used to establish the type 2 diabetic model in C57BL/6 mice. The mice were randomly divided into 4 groups (n=15):in control group, the normal mice were subcutaneously injected with equivalent volume of saline; in model group, the T2DM mice were subcutaneously injected with equivalent volume of saline; in low-and high-dose liraglutide treatment groups, the T2DM mice were subcutaneously injected with 100 and 200 μg·kg-1·d-1, respectively. After 4 weeks of administration, the levels of FBG, TG, TC, HDL-C, LDL-C, ALT and AST were determined. HE staining was used to observe the pathological changes of the liver tissues. The protein level of cleaved caspase-3 in the liver tissue was detected by the technique of immunofluorescence. The protein levels of p-AMPK/AMPK and apoptosis-related proteins were detected by Western blot. The expression of miR-33 in the liver tissues was detected by real-time PCR. RESULTS: Compared with model group, the contents of FBG, TG, TC, LDL-C, ALT and AST were decreased significantly, while the content of HDL-C was increased significantly in low-dose liraglutide group and high-dose liraglutide group (P<0.05). The protein levels of phosphorylated AMPK and Bcl-2 were up-regulated significantly, and the expression of cleaved caspase-3 was down-regulated significantly (P<0.05). The level of miR-33 was decreased significantly (P<0.01). CONCLUSION: Liraglutide alleviates liver injury in type 2 diabetic mice, and the mechanism may be associated with reducing the level of miR-33 and increasing the phosphorylation of AMPK in the liver tissues, thereby inhibiting hepatocyte apoptosis.  相似文献

AIM:To explore the effects of Shenmai injection on myocardial fibrosis in a rat model of diabetic cardiomyopathy (DCM). METHODS:Wistar rats (n=30) were randomly divided into control group, diabetes group and treatment group. Single intraperitoneal injection of streptozotocin was utilized to establish a rat model of DCM. The rats with DCM in treatment group were intraperitoneally injected with Shenmai injection. Ventricular cannulation was applied to assess the cardiac functions. The formation of collagen in the cardiac tissues was assessed by Masson staining. The generation of reactive oxygen species (ROS) in the cardiac tissues was detected by dihydroethidium staining. The expression levels of matrix metalloproteinase 2 (MMP-2), tissue inhibitor of metalloproteinase 2 (TIMP-2) and collagen I in the cardiac tissues were determined by Western blotting. RESULTS:Compared with control group, the cardiac functions were deteriorated in diabetes group (P<0.05), which was improved in treatment group as compared with diabetes group (P<0.05). Compared with control group, the formation of collagen and ROS increased significantly in diabetes group (P<0.05), which was decreased in treatment group as compared with diabetes group (P<0.05). Compared with control group, the expression level of MMP-2 in the cardiac tissues was deceased and TIMP-2 was increased significantly in diabetes group (P<0.05), but reversed significantly in treatment group (P<0.05).CONCLUSION:Shenmai injection attenuates cardiac fibrosis in the rats with DCM by inhibiting the generation of ROS.  相似文献

AIM: To observe the inhibitory effect of recombinant human endostatin (rhES) on plaque angiogenesis, and to explore the regulatory mechanism of Dll4/Notch pathway in the anti-angiogenic effect of rhES. METHODS: Male Wistar rats were randomized into 3 groups:normal control group (N group), atherosclerotic model group (AS group), and rhES treated group (AS+rhES group). The rats in N group were fed a normal diet, while the remaining 2 groups were established to atherosclerotic rat model via high-cholesterol diet, intraperitoneal injection of vitamin D3 and aortic balloon injury. The rats in AS+rhES group received intraperitoneal injection of rhES. The blood total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), C-reactive protein (CRP), interleukin-1 (IL-1) and troponin I (TnI) were measured. The atherosclerotic abdominal aortas were taken for pathological observation. Immunohistochemical staining was used to measure the density of neovessels in the plaques, which were marked by CD31. The protein levels of Dll4 and Notch1 in the aortas were analyzed by Western blot. RESULTS: The levels of blood TC, TG, LDL-C, CRP and IL-1 in AS group and AS+rhES group were much higher than those in N group (P<0.05), and no statistical difference between AS group and AS+rhES group was observed. The expression of CD31 in AS group was the highest among all groups. Compared with AS group, the density of neovessels in the plaques of AS+rhES group decreased significantly (P<0.05). The protein expression of Dll4 and Notch1 in AS group was lower than that in N group (P<0.05). Compared with AS group, the protein expression of Dll4 and Notch1 increased significantly (P<0.05). CONCLUSION: rhES has the ability to inhibit plaque angiogenesis in rats. The activation of Dll4/Notch pathway may be the mechanism of rhES in inhibiting plaque angiogenesis.  相似文献

AIM: To study the distribution of 936C/T polymorphism in 3’- untranslated region of vascular endothelial growth factor (VEGF) gene in Chinese Han population and to analyze the relationship between the polymorphism and diabetic retinopathy (DR). METHODS: Two hundred and fifty-four patients with type 2 diabetes mellitus recruited in this study were divided into NPDR group (non-proliferative diabetic retinopathy), PDR group (proliferative diabetic retinopathy) and DM (diabetes without retinopathy) group. The normal control group consisted of 120 subjects. Genotypes were identified by PCR-RFLP among all the subjects, while other clinical parameters were measured. Serum levels of VEGF were tested by the method of ELISA. RESULTS: The frequencies of both genotype CC and allele C were significantly higher in NPDR group and PDR group than those in NC group (2=9.934, 2=4.899, P<0.05 and 2=10.895, 2=5.714, P<0.05) and DM group (2=7.490, 2=4.448, P<0.05 and 2=8.333, 2=5.227, P<0.05). However, the frequencies of genotype (TT+CT) and allele T were significantly lower in NPDR group and PDR group than those in NC group (2=9.934, 2=10.895, P<0.01 and 2=4.899, 2=5.714, P<0.05) and DM group (2=7.490, 2=8.333, P<0.01 and 2=4.448, 2=5.227, P<0.05). Multivariate logistic regression analysis showed that the levels of glycohemoglobin(HbA1c), total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C) and plasma VEGF presented a positive correlation with DR, respectively, and the 936C/T mutation of VEGF exhibited a negative correlation with DR (β=-1.027, OR=0.343, P<0.01, CI: 0.157-0.723). CONCLUSION: Allele 936C of VEGF may be a genetic marker susceptible to DR, while allele 936T may be a protective genetic marker of DR. The 936C/T mutation of VEGF may be a protective factor against DR.  相似文献

AIM:To investigate the effects of resveratrol (RSV) on cardiac dysfunction and acid sphingomyelinase (ASMase)-ceramide pathway in diabetic rats. METHODS:Type 2 diabetes mellitus (T2DM) model was established by a high-fat diet combined with STZ intraperitoneac injection (30 mg/kg). SD rats (n=20) were randomly divided into control group, T2DM group; T2DM+RSV group (diabetic rats were given resveratrol at 100 mg·kg·d-1 by intragastric administration for the treatment) and RSV group (some of control rats were selected to give the same dose of RSV for drug control group). The M-mode Doppler ultrasonography was performed to observe the changes of cardiac function and structure in the rats. The levels of serum glucose, lipid and superoxide dismutase (SOD) activity, malondialdehyde (MDA) content in heart tissues were measured. Oil red O staining and Sirius red staining were performed to observe lipid accumulation and cardiac fibrosis in heart tissues. The cardiac ceramide concentration in diabetic rats was analyzed by high-performance liquid chromatography. The protein expression of ASMase and peroxisome proliferator-activated receptor-γ co-activator 1α (PGC-1α) in the hearts was determined by Western blot. RESULTS:Compared with the control group, the levels of fasting blood glucose, total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) were significantly elevated in T2DM group. The values of ±dp/dtmax, fractional shortening and ejection fraction were declined, and the left ventricle internal dimension at end-systole (LVIDs) and left ventricle internal at end-diastole (LVIDd) were increased. Furthermore, increased MDA content and more lipid accumulation were also observed in diabetic hearts, while the SOD activity, ATP content and PGC-1α expression were reduced in diabetic hearts. However, all these parameters were reversed by addition of RSV, concomitant with decreased ASMase expression and ceramide content. CONCLUSION:RSV dramatically alleviates diabetes-induced cardiac dysfunction and cardiac fibrosis, which may attribute to inhibition of ASMase-ceramide activation.  相似文献

AIM:To observe the protective effect of delivery of acidic fibroblast growth factor (aFGF) to myocardium by ultrasound-targeted microbubble destruction (UTMD) on left ventricular function in diabetic cardiomyopathy (DCM) rats and to investigate the possible mechanisms. METHODS:Twenty-four rats were intraperitoneally injected with streptozocin to induce DCM and were randomly divided into DCM group and aFGF treatment group. Twelve healthy rats served as normal controls. The rats in aFGF treatment group were infused with SonoVue-aFGF mixed fluid through tail vein and UTMD was simultaneously performed. Four weeks after intervention, all rats underwent cardiac catheterization to mea-sure left ventricular end-systolic pressure (LVESP), left ventricular end-diastolic pressure (LVEDP) and the maximal increase/decrease rate of left ventricular pressure (LV±dp/dtmax). The microvessel density (MVD) of rat myocardial tissues was measured by immunohistochemical staining for CD31. The myocardial collagen volume fraction (CVF) was determined by improved Masson staining. The apoptotic index (AI) was detected by TUNEL method. RESULTS:Four weeks after intervention, the LVESP and LV±dp/dtmax in aFGF treatment group were significantly increased compared with DCM group (P<0.01), while the LVEDP in aFGF treatment group was significantly lower than that in DCM group (P<0.01). The MVD in aFGF treatment group was significantly increased compared with DCM group (P<0.01), but the CVF and AI in aFGF treatment group were significantly lower than those in DCM group (P<0.01). CONCLUSION: Delivery of aFGF to diabetic myocardium by UTMD could improve the left ventricular function of DCM rats and may be a new feasible therapeutic method for DCM.  相似文献

AIM: To investigate the effects of total triterpenoids from Psidium guajava leaves (TTPGL) on blood glucose and lipids in type 2 diabetic rats. METHODS: The diabetic rats were induced by intraperitoneal injection of streptozotocin at dose of 35 mg/kg and feeding with high-fat diet. The animals were divided into 5 groups: diabetic model control group (model), TTPGL treatment groups (with the doses of 60, 120 and 240 mg/kg, respectively) and rosiglitazone treatment group (3 mg/kg). Another 12 normal SD rats were used as the normal controls. The rats received daily treatment for 6 weeks, and then the levels of fasting blood glucose (FBG), fasting insulin (FINS), triglyceride (TG), total cholesterol (TCH), free fatty acid (FFA), glycosylated hemoglobin (GHb) and glycosylated serum proteins (GSP) were measured. The protein expression of peroxisome proliferator-activated receptor γ (PPARγ) in adipose tissues was detected by Western blotting. RESULTS: Compared with normal control group, the levels of FBG, GHb and blood lipids were increased in type 2 diabetic rats. The FINS, insulin sensitivity index, and the protein expression of PPARγ in adipose tissues were decreased. Compared with model group, the levels of FBG and GSP were decreased,and the FINS, insulin sensitivity index, and the protein expression of PPARγ in adipose tissues significantly increased in TTPGL treatment groups (with the doses of 120 and 240 mg/kg). The levels of serum TG,TCH and FFA were significantly lower in TTPGL treatment groups (P<0.01 or P<0.05) as compared with the model controls. CONCLUSION: TTPGL decreases the levels of blood glucose and lipids in diabetic rats. TTPGL also increases serum insulin level and improves insulin sensitivity. The action mechanism of TTPGL may be related to the increase in the protein expression of PPARγ.  相似文献

AIM: To explore the changes of plasma levels of soluble vascular endothelial growth factor receptor 2 (sVEGFR2) and superoxide dismutase (SOD) in hypertensive patients and hypertensive diabetic patients. METHODS: In this cross-sectional study, 88 cases were enrolled, which were divided into hypertensive group (n=31), hypertensive diabetic group (n=31) and control group (n=26). Blood pressure was obtained from each participant with mercury sphygmomanometer. The levels of sVEGFR2 and SOD were measured by ELISA. Meanwhile, the levels of plasma glucose, glycosylated hemoglobin A1c (GHbA1c) and lipid profile were detected. RESULTS: The levels of total cholesterol (TC) and body mass index (BMI) were significantly higher in hypertensive group than those in control group (P<0.05). The levels of TC, low-density lipoprotein cholesterol (LDL-C), triglyceride (TG), BMI, waist circumference were significantly higher in hypertensive diabetic group than those in control group (P<0.05). The plasma levels of sVEGFR2 and SOD in both hypertensive diabetic group and hypertensive group were significantly decreased compared with control group (P<0.05), while the mean plasma levels of sVEGFR2 and SOD in hypertensive diabetic group were significantly decreased compared to the hypertensive group (P<0.05). A significantly positive correlation between sVEGFR2 and SOD in the whole study population (P<0.05) was observed. CONCLUSION: The plasma level of sVEGFR2 is decreased in both hypertensive and hypertensive diabetic patients, and more significantly decreased in hypertensive diabetic patients. Decreased SOD level may be associated with to the reduction of sVEGFR2.  相似文献

AIM:To investigate the effect of JNK/MCP-1 signaling pathway on anti-diabetic neuropathic pain by curcumin in type 2 diabetic rats. METHODS:The male Sprague-Dawley rats were induced as the model of the type 2 diabetic neuropathic pain rats, they were randomly divided into 6 groups (n=27): type 2 diabetic neuropathic pain (DNP) group, type 2 diabetic neuropathic pain and intraperitoneal injection of curcumin (Cur) group, type 2 diabetic neuropathic pain and solvent control (DSC) group, type 2 diabetic neuropathic pain and JNK inhibitor (DJ) group, type 2 diabetic neuropathic pain and JNK inhibitor solvent control (DJS) group, type 2 diabetic neuropathic pain and monocyte chemoattractant protein 1 (MCP-1) agonist (DM) group. Another 27 normal SD rats were selected as control group. Mechanical withdrawal threshod and thermal withdrawal latency were measured at 3rd d, 7th d and 14th d after dosing, then the lumbar segment 4~6 of the spinal cord and L4~6 DRG were removed at the same time. ELISA was used to measure MCP-1 level. The expression of p-JNK was determined by Western blotting. RESULTS:Compared with DNP group, p-JNK was significantly decreased at 7th d and 14th d in Cur group, DJ group and DM group after treatment (P<0.05). Compared with C group, the MCP-1 was significantly declined in other 6 group after streptozotocin injection (P<0.05). Compared with DNP group, MCP-1 were significantly increased at 7th d and 14th d in Cur group and DJ group after treatment (P<0.05), and that in DM group was greatly decreased (P<0.05). CONCLUSION: The expression of p-JNK and MCP-1 was increased in DNP rats with spinal cord and dorsal root ganglion. The mechanism of curcumin reducing the neuropathic pain in type 2 diabetic rats might be through regulating the JNK/MCP-1 pathway.  相似文献

AIM: To study whether the pulmonary infection of Escherichia coli (E. coli) interferes the glucolipid metabolism in high-fat diet-induced obese mice. METHODS: High-fat diet-induced obese mice (n=48) and normal chow-fed control mice (n=48) were intranasally infused with 40 μL fluid containing 4×109 CFUs E. coli. The serum, periepididymal adipose tissue and liver were obtained at 0 d, 1 d, 2 d, 3 d and 4 d after infection. The body mass, periepididymal adipose tissue and liver were weighed, and the levels of fasting blood glucose (FBG), fasting blood insulin (FINS), free fatty acid (FFA) and very-low-density lipoprotein (VLDL) were measured by ELISA. The serum total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C), and hepatic TG contents were detected, and the hepatic steatosis was observed under microscope with oil red O staining. RESULTS: Compared with day 0, the body mass, fat mass and fat index were decreased significantly from day 1 to day 4 after infection (P<0.05). The levels of FBG, FINS and HOMA-IR were apparently raised from day 2 to day 4 after infection (P<0.05). The contents of serum FFA, TG and VLDL were increased markedly from day 1 to day 4 after infection (P<0.05). However, the concentrations of serum TC, LDL-C and HDL-C were decreased obviously from day 1 to day 3 (P<0.05). The liver mass, liver index and TG content were significantly increased from day 1 to day 4 (P<0.05). Consistently, the lipid droplet accumulation in the liver cells was increased obviously at day 2 and day 4 after infection. Compared with control group, except the levels of serum TC, LDL-C and HDL-C in obese group substantially decreased, the other indexes were increased by different degrees during the whole experiment period (P<0.05). CONCLUSION: Pulmonary infection of Escherichia coli exacerbates the disorder of glucose and lipid metabolism in high-fat diet-induced obese mice, which contributes the development of insulin resistance and hepatic steatosis.  相似文献

AIM:To explore the effects of carnosine (CAR) on cardiac dysfunction in type 1 diabetic mellitus rats and the underlying mechanism. METHODS:The SD rats were randomly divided into 4 groups:control (C) group, control+carnosine (C+CAR) group, diabetes mellitus (DM) group and diabetes mellitus+carnosine (DM+CAR) group (n=10). The rats were sacrificed after 12 weeks. The cardiac function was assessed by ventricular cannulation. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) were assessed by ELISA. The mRNA levels of tumor necrosis factor α(TNF-α), interleukin-1β(IL-1β) and IL-6 were measured by real-time PCR. The distribution of connexin 43 (Cx43) was examined by immunofluorescence. The protein levels of Cx43 and protein kinase C (PKC) were determined by Western blot. RESULTS:Compared with the C group, the left ventricular end diastolic pressure (LVEDP) was increased whereas the left ventricular pressure maximum rise/fall velocity (±dp/dtmax) was decreased in the DM group (P<0.01). The activity of SOD decreased while the MDA increased in the left ventricular tissues (P<0.01). The mRNA levels of TNF-α, IL-1β and IL-6 were increased (P<0.01). The Cx43 distribution was irregular. The protein levels of phosphorylated Cx43 and PKCε were elevated (P<0.01). Compared with the DM group, the cardiac function of LVEDP and ±dp/dtmax in DM+CAR group was ameliorated (P<0.01), with increased SOD activity and decreased MDA content (P<0.05). The mRNA levels of TNF-α, IL-1β and IL-6 were reduced (P<0.01). The Cx43 distribution was improved and the protein levels of phosphorylated Cx43 and PKCε were decreased (P<0.01). CONCLUSION:CAR treatment can improve the cardiac function by its anti-oxidative and anti-inflammation effects and suppression of Cx43 abnormalities through PKCε in DM rats.  相似文献

AIM:To study the effects of betaine on glial fibrillary acidic protein (GFAP), glycine (Gly) and glycine receptor (GlyR) expression in the hippocampus of rats with epilepsy induced by pentylenetetrazole (PTZ). METHODS:Forty-eight healthy male Wistar rats were randomly divided into control group, PTZ (35 mg·kg-1·d-1, intraperitoneal injection) group, PTZ+betaine (450 mg·kg-1·d-1, intragastric administration) group, PTZ+betaine (225 mg·kg-1·d-1, intragastric administration) group, PTZ+betaine (112.5 mg·kg-1·d-1, intragastric administration) group and PTZ+sodium valproate (200 mg·kg-1·d-1, intragastric administration) group. The rats in control group were intraperitoneally injected with saline at the same volume as PTZ injection, and those in control group and PTZ group received intragastric administration of saline at 1.0 mL·d-1. Rat behavior was recorded. Serum homocysteine (Hcy) level was measured. The expression of GFAP in the hippocampus was measured by immunofluorescence. Hippocampal Gly content was measured by an amino acid analysis system. The expression of GlyR was detected by immunofluorescence and Western blotting. RESULTS:There was no difference in the latency of grand mal seizures among groups (P>0.05). However, betaine treatment significantly decreased the duration of the first grand mal seizure compared with PTZ group (P<0.01). Serum Hcy level in PTZ group was significantly lowered compared with control group (P<0.01), and further decreased after betaine treatment (P<0.05). GFAP in PTZ group was significantly higher than that in control group (P<0.01), and decreased after betaine treatment (P<0.05). Gly in PTZ group was significantly lowered compared with control group (P<0.01), and increased after betaine treatment (P<0.05). The content of GlyR among groups showed the same trend as Gly. CONCLUSION:Betaine treatment shows antiepileptic effect, which may be related to its effects on the metabolites of Hcy and Gly.  相似文献

AIM: To observe the effect of Tangshenfang (TS) on the liver protection and the levels of silent information regulator 1 (SIRT1) and peroxisom proliferator-activated receptor γ coactivator-1α (PGC-1α) in the liver tissue. METHODS: The rat model of diabetes mellitus (DM) was established by intravenous injection of streptozotocin (STZ;30 mg/kg) after having the high fat/high glucose diets for 1 month. The diabetic rats were randomly divided into DM group, DM with high-dose TS (TSHi) group, medium-dose TS (TSMed) group and low-dose TS (TSLow)group. The normal rats were served as control group. There were 8 rats in each group. After treatment with TS for 12 weeks, the serum biochemical indices including fasting blood glucose (FBG), triglyceride (TG), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were tested. Fasting insulin (FINS) was also detected by radioimmunoassay, and homeostatic model assessment for insulin resistance (HOMA-IR) was calculated. The serum levels of tumor necrosis factor-α (TNF-α) and interleukin-1 (IL-1) were measured by ELISA. The activity of SOD and content of MDA in the liver tissues were measured by the methods of hydroxylamine and thiobarbituric acid. The liver pathological changes were observed under light microscope with HE and Masson staining. The protein expression of SIRT1and PGC-1α in the liver tissues was determined by Western blot. RESULTS: In DM group, serum FBG, TG, ALT, AST, FINS, HOMA-IR, TNF-α and IL-1 were obviously increased compared with the control group (P<0.01). The fatty changes, local necrosis, inflammation and fibrosis in the liver tissues were observed. The content of MDA in liver increased, while the activity of SOD decreased markedly. The protein expression of SIRT1 and PGC-1α was decreased (P<0.05). In TS treatment groups, all these changes in DM rats were markedly reversed by TS, and the protein expression of SIRT1 and PGC-1α in the liver tissues was markedly increased. CONCLUSION: TS may protect the rats from diabetic liver injury by increasing the expression of SIRT1 and PGC-1α, and thereby improving insulin resistance and oxidative stress.  相似文献
