One hundred and eighty-five Holstein-Friesian dairy cows received either sterile water or 250 micrograms of gonadotrophin releasing hormone intramuscularly on the fourth day after the first service postpartum. Heparinized blood samples were taken immediately prior to treatment (day 4) and on day 8 postinsemination for analysis of plasma progesterone concentration. Pregnancy diagnosis was carried out by rectal palpation at 42 days postinsemination and reconfirmed after 60 days postbreeding. The pregnancy rates after first, second or third service were not significantly different between gonadotrophin releasing hormone-treated and control cows. Plasma progesterone concentrations on day 4 and day 8 postinsemination, as well as the change in plasma progesterone concentration from day 4 to day 8, were similar for gonadotrophin releasing hormone-treated and control cows. The plasma progesterone concentrations on day 8 postbreeding were significantly higher (p less than 0.005) and the change in progesterone concentrations between days 4 and 8 were significantly greater (p less than 0.002) in pregnant cows compared to nonpregnant cows.  相似文献

Milk progesterone determination in buffaloes post-insemination   总被引:1,自引:0,他引:1
Milk progesterone levels were studied in 17 buffaloes between post-insemination and 30 days thereafter. Six animals were confirmed pregnant. Seven of 10 animals took greater than 24 days to return to oestrus and the pattern of change of progesterone levels indicated a steady increase from 1 ng/ml on day 2 to 20 ng/ml on day 14 post-insemination, falling subsequently to about 5 ng/ml, 2 days prior to oestrus. In pregnant buffaloes, the milk progesterone levels showed a continuous increase up to 20 days post-insemination but did not decline thereafter. In individual buffaloes returning to oestrus, a cut-off milk progesterone level of greater than 10 ng/ml was considered for pregnancy diagnosis, 20, 22 and 24 days post-insemination. The test was 60, 75 and 75% accurate for detection of pregnancy on days 20, 22 and 24 respectively and 100% for non-pregnancy diagnosis on all three days. Individual animals showed a highly variable oestrous cycle length, which has been suggested as a contributory factor for false pregnancy diagnosis by milk progesterone test in this species. Milk progesterone assay may also identify silent heats, and clinical disorders.  相似文献

Female guinea pigs were tested to determine whether they could serve as a model of zearalenone (ZEN) toxicosis during early pregnancy, as observed in domestic swine. Only 1 of 4 female guinea pigs that were given 21 mg of ZEN/kg of body weight orally during the first 8 days after mating was pregnant when examined 22 days after mating. Guinea pigs that were given 7 or 14 mg of ZEN/kg had normal fetal development. Serum concentrations of progesterone were less than 12 ng/ml in all guinea pigs 8 and 15 days after mating. Serum concentrations of progesterone were greater than 100 ng/ml in pregnant guinea pigs on day 22, but remained less than 12 ng/ml in nonpregnant guinea pigs. Three of 5 guinea pigs treated with 20 mg of ZEN/kg and only 1 of 4 guinea pigs given 30 mg of ZEN/kg on days 1 to 3 after mating were pregnant 22 days after mating. Female guinea pigs treated with 20 or 30 mg of ZEN/kg on days 4 to 5 or 6 to 8 after mating and female guinea pigs treated with 60 or 90 mg of ZEN/kg on days 4 and 5 after mating had normal pregnancies. Serum concentrations of progesterone were less than 10 ng/ml in all guinea pigs on day 15 and remained low on day 22 only in nonpregnant guinea pigs.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A study aimed at validating a human progesterone enzyme immunoassay kit was carried out on cattle at Bambui, Cameroon. Progesterone ELISA Kits (EH-511) were obtained from Clinpro International. Forty-one cows were selected, of which 19 were pregnant and 22 within 14 days post partum. Blood samples were analysed and progesterone levels were deduced from a curve obtained from standard absorbance values (A 450). Results show that 95.5% of postpartum cows had progesterone levels below 1 ng/ml, with the highest level being 0.75 ng/ml. The mean level was 0.5 ± 0.26 ng/ml. The cows in the ‘pregnant group’ had progesterone levels ranging from 3.5 to 17.5 ng/ml. This kit can be used for measuring progesterone levels in cattle. Levels of 1 ng/ml for two consecutive samples or one sample at or above 3 ng/ml are an indication of the presence of corpus luteum, while cows below 1 ng/ml will be in anoestrus.  相似文献

A field study was conducted to determine the usefulness of milk progesterone determination at the time of breeding to confirm estrus and at 21 days postbreeding to detect open cows. Twenty-seven dairy farmers collaborated in this study by providing milk samples on the day of breeding and 21 days later, together with pregnancy diagnosis data and information on herd reproductive management. Herd size ranged from 15 to 175 cows, the average being 65 milking cows. Six hundred and sixty-seven breeding-day samples and 472, 21-day samples were provided by the farmers. Analysis of milk samples for progesterone by a solid phase radioimmunoassay showed that only 32 (4.8%) of the services were performed when the cow was not in estrus (progesterone > 1 ng/mL). Of the 472, 21-day samples, 337 (71%) showed progesterone levels of > 1 ng/mL, while 135 (29%) showed progesterone levels of < 1 ng/mL. Subsequently, 243 (72%) of the cows with progesterone > 1 ng/mL and eight (6%) of the cows with progesterone < 1 ng/mL were diagnosed pregnant by transrectal palpation, giving a pregnancy rate of 53%. Progesterone concentration on the day of breeding was not associated with season or herd size. However, progesterone concentration at 21 days and pregnancy rate were associated with herd size. These results indicate that fertillzation failure and/or early embryonic mortality, rather than inaccurate detection of estrus, are the major reproductive problems encountered by the dairy farmers in British Columbia. Furthermore, progesterone values at 21 days were closely related to reproductive status and indicate the usefulness of milk progesterone assay for the early detection of open cows.  相似文献

Two hundred and fifty-one cows fresh at least 37 days with normal reproductive tracts and palpable ovarian structures which clinicians believed to be corpora lutea and which were presumed to be producing progesterone were treated with 500 micrograms of cloprostenol. The cows were stratified into three groups based on milk progesterone concentrations in whole milk samples taken at the time of examination. The low group had milk progesterone concentrations less than or equal to 1 ng/ml, the intermediate group had milk progesterone concentrations between 1 and 3 ng/ml, and the high group had milk progesterone concentrations greater than or equal to 3 ng/ml. The proportion of cows in each group inseminated within 5 days of treatment and the fertility at that breeding were compared. There were no significant differences among the groups with respect to the proportion of cows that came into estrus within 5 days. However, the conception rate of the high group (54%) was significantly greater than that of the intermediate group (26%). To study the relationship between practitioner experience and diagnostic accuracy the proportion of cows with low, intermediate and high concentrations of progesterone selected by clinicians with more than 3 years experience was compared to that for clinicians with 3 years of experience or less. Less experienced clinicians were significantly more likely to make a false positive diagnosis of functional luteal tissue in cows with palpable ovarian structures than were more experienced clinicians.  相似文献

In 160 cows with ovarian cysts as determined by rectal palpation, differentiation was made of follicular cyst, luteal cyst, and cystic corpus luteum on the basis of milk progesterone concentrations estimated by an enzyme immunoassay before and at 10 days after cows were treated with gonadotropin-releasing hormone. Cows having a progesterone concentration in skim milk less than 1.0 ng/ml were considered to have follicular cysts and those with concentrations of 1.0 ng/ml or higher were regarded as the cases of luteal cyst or cystic corpus luteum. Luteal cyst was characterized by progesterone values remaining high in the cows for 10 days after treatment, and cystic corpus luteum was characterized by a decrease in progesterone concentration after cows were treated. By the rectal palpation procedure it was impossible to differentiate luteal cyst and cystic corpus luteum from follicular cyst. The frequencies of follicular cyst, luteal cyst, and cystic corpus luteum were 65%, 19%, and 16%, respectively. Of 104 cows with follicular cysts as defined by milk progesterone assay result, 73 (70%) responded to the treatment with gonadotropin-releasing hormone, the milk progesterone concentration increasing from 0.7 +/- 0.2 ng/ml (mean +/- SD) to 1.8 +/- 1.1 ng/ml. The accuracy of rectal palpation 10 days after treatment for judgment of luteinization of follicular cyst confirmed by milk progesterone analysis was only 30% (48 cows of 160).  相似文献

The skim milk progesterone profile was assessed by radioimmunoassay, without extraction, from the day of insemination (day 0) until the cows were dried off on day 225 of gestation. A total of 418 samples were collected from 154 pregnant Holstein cows. The daily variation in skim milk progesterone was recorded from day 1 until day 45 of pregnancy to detect the commencement of progesterone secretion from the corpus luteum after insemination. Subsequent determinations were made every 2 weeks from day 46 until lactation ceased. On the day of artificial insemination and for the first 2 days after insemination, all the cows had a basal progesterone concentration <0.1 ng/ml. A rise in progesterone (0.2±0.1 ng/ml) was first detected on the third day after insemination. The progesterone values then increased significantly (p<0.001) until day 15.The values then remained nearly constant (2.5–3.5 ng/ml) until day 106 of pregnancy, when they began to decline. Between days 120 and 180 of gestation, progesterone was significantly decreased (2.2–2.9 ng/ml) before it rose again to the previous plateau (3.5–3.9 ng/ml) around day 180. The progesterone concentration then remained at the higher level until the animals were dried off.Abbreviations AI artificial insemination - RIA radioimmunoassay  相似文献

The present study investigated the changes of serum progesterone (P4) and its faecal metabolites in pregnant and non-pregnant cows (Expt 1) and the feasibilty of using faecal P4 metabolites for early screening of open cows post-insemination (Expt 2). In Expt 1, seven crossbred Holstein-Friesian (HF) cows were studied. Serum and faecal samples were collected once daily from the day of artificial insemination (AI) until 25 days after AI. In Expt 2, 27 crossbred HF inseminated cows were employed. Serum and faecal samples were obtained on the day of AI (day 0) and on days 19-22 post-insemination. Enzyme immunoassay measurements of serum P4 and faecal P4 metabolites were established. The low detection limit of the assay was 0.01 ng/ml and the amount of P4, resulting in a 50% reduction in the initial binding value, was 1.07 ng/ml. The intra- and inter-assay coefficients of variation were <8% and <14%, respectively. A positive correlation between the levels of serum P4 and faecal P4 metabolites was found in every single cow (r = 0.73-0.88, p < 0.001) and pooled data (r = 0.78, p < 0.001). The estimated value of faecal P4 metabolites at 100 ng/g of faeces was equal to the serum P4 levels of 1 ng/ml. The accuracies of pregnancy and non-pregnancy diagnosis based on the analyses of faecal P4 metabolites between day 0 and days 19-22 post-insemination, were 67% and 100%, respectively. In conclusion, the measurement of faecal P4 metabolites can be a potentially alternative method for early screening of open cows post-insemination with the same accuracy and precision, as measured by serum P4 assay.  相似文献

OBJECTIVE: To identify potential risk factors for embryonic loss before 35 to 42 days of gestation in dairy cattle. DESIGN: Prospective observational study. ANIMALS: 381 cows. PROCEDURE: Body condition score was determined at the time of artificial insemination (AI; day 0) and on days 20, 23, and 27 and between days 35 and 41; serum progesterone concentration was measured on days 0; 20 or 21; and 23, 24, or 25. Cows were excluded from analyses if day 0 serum progesterone concentration was > or = 1.0 ng/mL and classified as pregnant on day 23 if serum progesterone was > 1.5 ng/mL on day 20 or 21 and day 23, 24, or 25. Cows were examined via transrectal ultrasonography on day 27 or 28 and rectally palpated for pregnancy on days 35 to 41. RESULTS: 39% of cows that were pregnant on day 23 lost their embryo by day 27, and 18% of cows that were pregnant on day 27 or 28 were not pregnant on days 35 to 41. Breeding a pregnant cow posed the greatest risk for embryonic loss at both time periods. Mean serum progesterone concentrations on day 21 or 22 and day 23, 24, or 25 were lowest for cows that lost an embryo between days 24 and 28. Cows with a linear somatic cell count score > 4.5 before AI were twice as likely to lose the embryo by 35 to 41 days, compared with cows with a score < 4.5. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that embryonic loss could be reduced by more accurate estrus detection, reducing mastitis, and strategies to improve progesterone concentration after breeding.  相似文献

The effects of the rainy (RS) and the dry season (DS) on fertility, corpus luteum activity, and adrenal cortex response relationships were evaluated after first service (49 ± 6 d postpartum) in Carora cows, a dairy cattle of Venezuela raised in tropical conditions. Cows (n = 84 in RS and n = 98 in DS) were kept semistabled, had two or three calvings, body condition score 3.5 on a 5-point scale, and similar milk yield (2450 ± 560 kg of milk during the previous lactation). Cows were grouped retrospectively according to pregnancy status. A split-plot model with repeated measures over Days 5, 7, 10, 14, and 15 after insemination was used to analyze the effects of season, pregnancy status, and their interaction involving the day on: 1) serum concentrations of progesterone in four treatments: RS pregnant (n = 26), RS nonpregnant (n = 24), DS pregnant (n = 24), and DS nonpregnant (n = 20) cows; 2) serum concentrations of cortisol at Days 0, 10, 14, 15, and 16 postservice in the previous treatments (n = 9, 7, 6, and 8, respectively); and 3) concentrations of cortisol after 0.1 mg of adrenocorticotropin in these last four groups of cows at Day 14 postin-semination. Breeding during the DS decreased (P < 0.05) conception rate to first service and increased (P < 0.01) days in service. In addition, the DS decreased (P < 0.05) the percentages of cows with normal interestrous interval (20–22 d), expression of estrus, and (P < 0.01) luteal phase progesterone; but DS increased (P < 0.05) percentages of short and long estrous cycles, anovulatory estrus, and repeat breeding rate. Mean serum concentration of progesterone was lower (P < 0.05) at Days 10, 14, and 15 in DS nonpregnant than in DS pregnant cows, and lower during luteal phase (P < 0.05) in DS nonpregnant than RS nonpregnant cows. Serum cortisol concentration was greater (P < 0.05) at Days 10, 14, and 16 in DS nonpregnant than DS pregnant cows. A significant (P < 0.05) negative correlation (r = −0.78) between serum concentrations of progesterone and cortisol was found within DS nonpregnant cows. Concentrations of cortisol after adrenocorticotropin were greater (P < 0.05) in DS nonpregnant cows than in other groups. These results indicate that elevated concentrations of cortisol associated with the DS may decrease progesterone secreted by the corpus luteum and therefore mediate the negative effect of the DS on fertility.  相似文献

Blood from 171 pregnant and 65 nonpregnant cows was taken at slaughter for culture and examined during the culturing period for Trypanosoma theileri once a week for 5 weeks. Of the 171 fetuses from the pregnant cows, 119 (69.6%) were greater than or equal to 4 months gestation; blood samples from these fetuses were also taken for culture. Of 236 cows (81 of 171 [47.4%] pregnant and 20 of 65 [30.8%] nonpregnant cows), 101 (42.8%) were culture positive. More of the pregnant than nonpregnant cows were culture positive (P less than 0.05). More beef cows (48.0%) than dairy cows (34.1%) were culture positive (P less than 0.025). Two of the 119 (1.7%) fetus samples were found culture positive. The percentages of positive cultures from Brucella-reactor cows, 18 of 40 (45.0%), and from non-Brucella-reactor cows, 83 of 196 (42.3%), were similar. However, both of the culture-positive fetuses were from Brucella-reactor cows.  相似文献

Objectives were to determine associations between percentage pregnancy loss (PPL) in dairy cattle and: (i) pregnancy diagnosis by ultrasonography; (ii) pregnancy diagnosis by serum pregnancy‐specific protein B (PSPB) concentrations, with or without serum progesterone concentrations; and (iii) production and environmental factors. This study included 149 822 pregnancy diagnoses conducted over 13 years in Holstein‐Friesian cows in Hungarian dairy herds. The following were determined: PPL in cows diagnosed pregnant by transrectal ultrasonography 29–42 days after artificial insemination (AI; n = 11 457); PPL in cows diagnosed pregnant by serum PSPB 29–35 days after AI (n = 138 365); and PPL and its association with serum progesterone concentrations, PSPB and production/environmental variables. The definition of PPL was percentage of cows initially diagnosed pregnant based on ultrasonography or PSPB, but not pregnant when examined by transrectal palpation 60 –70 days after AI. The PPL was lower (p < 0.001) in cows following ultrasonographic vs PSPB diagnosis of pregnancy at 29–35 days (8.1 vs 19.3%, respectively), but was higher in cows following ultrasonographic pregnancy diagnosis on 29–35 vs 36–42 days (8.1 vs 7.1%, respectively, P < 0.05). Furthermore, 72.9% of pregnancies with ultrasound‐detected morphological abnormalities resulted in pregnancy loss. As a subset of PSPB data, a fully quantitative PSPB assay was used for 20 430 samples; PPL in cows with a high PSPB concentration (>1.1 ng/ml) was lowest (15.0%), whereas cows with low concentrations of both PSPB and progesterone (0.6–1.1 and <2 ng/ml, respectively) had the highest PPL (76.3%; p < 0.0001). Furthermore, PPL was higher in cows with advanced parity and with high milk production, when ambient temperatures were high, although body condition score (BCS) had no effect on PPL. Finally, there were no significant associations between serum PSPB and environmental temperatures or number of post‐partum uterine treatments.  相似文献

We gave gonadotropin-releasing hormone (GnRH) or an analog of GnRH to 264 lactating cows with cystic ovaries. The effects of milk progesterone concentration (MPC) and days after parturition (DAP) at diagnosis on clinical response 30 days after treatment and on subsequent fertility were examined. Palpation per rectum revealed ovarian cysts in 264 cows; 118 had true follicular cysts (MPC less than 1 ng/ml). Clinicians with more than 3 years of experience reported significantly more true cysts than did less experienced clinicians. Clinical response 30 days after treatment did not differ in 3 groups of cows (divided on the basis of MPC at treatment), but fewer cows with MPC less than 1 ng/ml were bred or conceived than were cows with MPC greater than or equal to 33 ng/ml. Days after parturition at diagnosis did not affect 30-day clinical response rate, but cows treated less than 35 DAP had significantly more days to first estrus and to conception than did cows treated greater than 90 DAP. Significantly more cows treated less than 35 DAP had MPC less than 1 ng/ml at treatment.

Behandlung i. m., 2. Contents Experiences with differing dosage forms of a novel progestin (Sa 45.249) for the induction of estrus in dairy cows with silent heat and simultaneous observations with the milk progesterone test. Oral administration of a novel progestin, Sa 45.249, 10 mg/day for 10 days, was effective in inducing heat and fertility in 20 dairy cows with silent heat. Within 9 days after the last medication 19 of 20 animals were in heat (10 with significant, 9 with weak or questionable symptoms) and 11 animals conceived from a. i. at the first heat. A total of 16 out of 20 animals became pregnant after an average of 17,4 days and an a. i. index of 1,3. A treatment of only two drug applications, one i. m. injection of 40 mg and an oral administration 72 hrs later of another 40 mg of Sa 45.249 was less effective. Within 9 days, only 17 of 25 treated animals showed heat (6 with significant, 11 with weak symptoms), and only 5 of 25 animals bred conceived at the first heat after treatment. 21 of all animals treated became eventually pregnant, after an average of 47,5 days, and an a. i. index of 2,2. The results from rectal ovarian palpation and milk progesterone values did not concur in 13,3 %. In 4 animals with no signs of functional activity at both ovaries, 3 showed progesterone values in milk indicative of luteal activities, while in 3 animals with corpora lutea progesterone values were at the baseline, below any level of c. 1. activity. In one animal progesterone values in milk after conception rose very slowly and reached only 2,6 ng/ml and 10,7 ng/ml at days 6 and 8 of gestation, respectively.  相似文献

Three hundred and sixty inseminated Holstein cows were monitored in a controlled trial for low milk progesterone concentration 21-23 days post-breeding. Information about low progesterone cows was given to herdsmen for the experimental group only. Of the cows in the experimental group with milk progesterone of less than or equal to 4.5 ng/ml, 56.0% were inseminated within 2 days of monitoring compared to 28.8% of the control animals. Of the cows reaching the 42-day rectal pregnancy diagnosis, 13.7% and 43.6% were negative for the experimental and the control groups respectively. Inter-service intervals were 34.5 +/- 14.5 days and 44.3 +/- 17.7 days for the experimental and control groups respectively. Pregnancy rates were the same for 264 cows served on routine progesterone monitoring and for 318 cows bred on behavioural oestrus when matched to farm and day of breeding. A gain of US $32.5 (an equivalent of 85 l milk) per lactation for a cow in the monitored herd was established using the data in a model of a 250-cow herd with a total pregnancy goal of 82.4%.  相似文献

The role of decreased luteal activity in embryonic loss after induced endotoxemia was studied in mares 21 to 35 days pregnant. Fourteen pregnant mares were treated daily with 44 mg of altrenogest to compensate for the loss of endogenous progesterone secretion caused by prostaglandin F2 alpha (PGF2 alpha) synthesis and release following intravenous administration of Salmonella typhimurium endotoxin. Altrenogest was administered daily from the day of endotoxin injection until day 40 of gestation (group 1; n = 7), until day 70 (group 2; n = 5), or until day 50 (group 3; n = 2). In all mares, secretion of PGF2 alpha, as determined by the plasma 15-keto-13,14-dihydro-PGF2 alpha concentrations, followed a biphasic pattern, with an initial peak at 30 minutes followed by a second, larger peak at 105 minutes after endotoxin injection. Plasma progesterone concentrations decreased in all mares to values less than 1 ng/ml within 24 hours after endotoxin injection. In group 1, progesterone concentrations for all mares were less than 1 ng/ml until the final day of altrenogest treatment. In 6 of 7 mares in group 1, the fetuses died within 4 days after the end of treatment, with progesterone concentrations less than 1 ng/ml at that time. In the mare that remained pregnant after the end of treatment, plasma progesterone concentration was 1.6 ng/ml on day 41 and increased to 4.4 ng/ml on day 44. In group 2, all mares remained pregnant, even though plasma progesterone concentrations were less than 1 ng/ml in 4 of 5 mares from the day after endotoxin injection until after the end of altrenogest treatment.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Two experiments were conducted to examine the temporal aspects of luteal resistance to the luteolytic effect of prostaglandin (PG) F2 alpha during early pregnancy. In Exp. 1, 14 pregnant and 12 nonpregnant ewes were treated with PGF2 alpha either on d 10 or 13 post-estrus. Jugular venous blood samples were collected at -30 min, 0, 6, 12, 18, 24, 30 and 36 h post-injection for quantification of progesterone. The difference (delta P) between pre-treatment and post-treatment concentrations of progesterone was calculated for each ewe. There was a significant interaction between pregnancy status and day of treatment on delta P (P less than .05). Pregnant and nonpregnant ewes treated on d 10 showed a large delta P. A large delta P also was observed in nonpregnant ewes treated on d 13 post-estrus. However, delta P in pregnant ewes treated on d 13 was smaller than in the other three groups (P less than .05). The temporal patterns of concentrations of progesterone in serum were different among treatment groups (P less than .05). A suppression in the concentration of progesterone was observed by 24 h post-injection in all four treatment groups. Progesterone returned to pre-treatment levels only in pregnant ewes treated on d 13. In Exp. 2, 47 pregnant ewes were treated with PGF2 alpha on d 10, 13, 16, 19, 22, 26 or 30 postestrus. Blood samples were collected and data were analyzed as described for Exp. 1.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
