2008年我国部分麦区小麦白粉病菌群体对温度的敏感性   总被引:1,自引:1,他引:0  
 The sensitivity of 113 isolates of wheat powdery mildew (Blumeria graminis f. sp. tritici) sampled from 6 provinces or cities in 2008 to temperature was tested by detached leaf segment method with setting up 5 different temperatures indoor. The results showed the mean ET50 (which represents the temperature that is required to obtain 50% of the maximum effect) of all isolates tested was 23.02℃. ET50 values of 17.70% isolates were more than 24℃. The highest and the lowest ET50 of isolates were 25.22℃ and 19.42℃, respectively. There were a certain differences for isolates sensitivity to temperature among different provinces or cites. It was also found that when temperature increased during 22-26℃, the latent period of isolates prolonged, and the latent period of different isolates was different at the same temperature, too. These results will provide a reference for the oversummering division of wheat powdery mildew, as well as the effect of climate to the disease.  相似文献

茄科蔬菜立枯丝核菌的融合群鉴定   总被引:3,自引:0,他引:3  
 Sixty-five samples were collected from rhizosphere soil, hot pepper and tomato plants showing damping-off, root rot and stem rot in Taian, Shouguang of Shandong province and Zhouzhi, Taibai of Shaanxi province. Thirty-nine Rhizoctonia solani isolates were obtained from these samples. The results of anastomosis group (AG) identification and sequence analysis of 5.8S rDNA-ITS of the isolates showed that thirty-six isolates (92.3%) belonged to AG-4, while only three (7.7%) belonged to AG-5. The isolates of AG4 could further be divided into two subgroups of AG4-HG-Ⅰ and AG4-HG-Ⅲ. The 5.8S rDNA-ITS sequences of the selected isolates of the two subgroups had the 99%-100% identity with standard isolates of AG4-HG-Ⅰ and AG4-HG-Ⅲ (from GenBank). Among the analyzed isolates, AG4-HG-Ⅰ subgroup was the dominant with the frequency of 79.5%. Subgroup AG-4-HG-Ⅲ with the frequency of 12.8% was the second. A species-specific PCR assay was established for rapid and accurate detection of the oomycete pathogen Phytophthora tentaculata in diseased plant tissues and infected soil.A pair of species-specific primers Pt1/Pt2 were designed on the basis of Ras-related protein(Ypt1) gene sequences of the Phytophthora species.PCR amplification with the Pt primers resulted in a 386 bp product only from isolates of P.tentaculata.The detection threshold with Pt primers was 100 pg of genomic DNA.A nested PCR procedure was developed using Ypt1F/Ypt1R as the first-round amplification primers and Pt1/Pt2 as the second-round primers,which increased the detection sensitivity 100-fold to 1 pg.PCR using these Pt primers can also be used to detect P.tentaculata in naturally infected plant tissues and soil.The PCR-based method developed in this study provides a rapid and sensitive tool for detection of P.tentaculata.  相似文献

 FM4-64,a kind of membrane-selective fluorescent dye,was utilized to stain the membrane structure of Botrytis cinerea and Phytophthora capsici.The effect of FM4-64 on hypha was observed under confocal microscope.The results indicated that the plasma membrane,speta and vesicles of the hyphae could be stained with FM4-64 at the concentrations of 1 to 20 μmol/L.The optimal dye concentration was 6 μmol/L for the both pathogens.At the same concentration of FM4-64,the staining of organelles was in a time-dependent manner and the different organelles were stained in diferrent times.The staining of plasma membrane and vesicles should be completed in 5 minutes,as other organelles can be stained after 10 minutes.  相似文献

壳寡糖诱导烟草防御酶系活性变化及PR-1a基因表达研究   总被引:3,自引:0,他引:3  
 The activity change of defensive enzymes and PR-1a gene expression of tobacco (Nicotiana tabacum) seedling induced by chito-oligosaccharides were studied. The results showed that high level systemic acquired resistance (SAR) was expressed in tobacco plants treated with chito-oligosaccharides solution at the concentration of 50 μg/mL. PAL activity increased greatly with 2 peaks, the activity of SOD decreased initially followed by an increase with higher increment, and the activity of POD peaked early followed by a gentle fall in chito-oligosaccharide treated plants. The PR-1a gene was strongly expressed in tobacco due to systemic acquired resistance induced by chito-oligosaccharides. At 168 h after inoculation the expression quantity (co-pies/2 μL) of PR-1a gene was increased to 2 469.6 in treated tobacco leaf, reached 392.6% than that at 0 h after inoculation, it was increased 3.05 times of that in untreated control.  相似文献

 Factors of influence on conidium germination and appressorium formation of Setosphaeria turcica,such as light condition,conidial concentration,nutrient resources and pH value of conidial suspension were studied.There was no significant difference among light treatments.The optimal pH for conidium germination and appressorium formation was 5.0 to 7.0.The exogenous nutrient sources were not the indispensable factors for conidium germination,but 5% sugar solution were more favorable for appressorium formation than the others.Low conidial concentration in suspension (conidia ≤ 104/mL) was propitious to conidium germination and appressorium formation,which were inhibited significantly in higher concentration.It was suggested that the phenomenon was due to the self-inhibitor,a kind of lipophilic substance,existing in the site of conidium germination.  相似文献

广西黑皮冬瓜疫病的病原菌鉴定及其生物学特性   总被引:9,自引:0,他引:9  
 Isolate PD1 of Phytophthora was isolated from diseased black pericarp wax gourd in Guangxi in 2005. The morphological and biological characters of the isolate were examined, and its sequence of ribosomal DNA-ITS was analyzed. The range of growth temperature of the isolate was 10-37℃ and the optimal one was 31℃. The pathogen was sensitive to 10 mg/kg of malachite green, and its ability for starch utilization was weak. A wide host range of the pathogen was confirmed by artificial inoculation. The sizes of sporangia were (37.5-71.7) μm×(20.8-47.5) μm (av. 55.0 μm×30.4 μm). Microcycle conidiation of the pathogen was observed. The pathogen was identified as Phytophthora drechsleri Tucker based on its morphological and biological characters as well as ribosomal DNA-ITS sequence.  相似文献

 Four isolates of Rice black-streaked dwarf virus (RBSDV) were collected from the maize plants showing rough dwarf symptom in Linyi and Tai'an,Shandong province.The S10 genomic sequences of these isolates were determined and compared with those of 14 other RBSDV isolates.All of the four sequences were 1 801 base pairs (bp) long including the 5'-UTR of 21 bp and the 3'-UTR of 103 bp.They all contained an open reading frame of 1 677 bp (22-1698),encoding the coat protein (CP) of 558 amino acids.The sequences of these four RBSDV isolates and those of the major cp gene of 14 other isolates available in the GenBank were divided into two groups in the phylogenetic tree.Recombination analysis indicated that the isolate Lym2 was likely a recombinant of isolates Lym1 and Zhjs.  相似文献

 The 930 bp segment in 3' terminal region of Strawberry mild yellow edge virus(SMYEV) genome was amplified by 3' rapid amplification of cDNA ends(RACE).Ten Chinese isolates were sequenced,and 7 of them were the same.Nucleotide and amino acid identities and phylogenesis were analyzed between Chinese isolates and 24 isolates from other regions of the world.Sequence analysis of the 878 nt stretch within 3' terminal region of SMYEV genome showed that nucleotide acid identities ranged from 79.5% to 100%,deduced amino acid sequences of coat protein gene identity were 86.4% to 100%.Phylogenetic analysis showed that all isolates of SMYEV fell into four clades.To a certain extent,the clades were related with the geological distribution of SMYEV.Chinese isolates SY01 and SY04 lay in the same clade with European and American isolates,but formed a small separate branch.Isolates SY03 and SY02,derived from Fragaria×ananassa cv.Changhong-2 and F.pentaphylla respectively,had a far relationship with other isolates and fell into one clade.They were likely to be the special isolates that existed only in China.  相似文献

 The technique of TaqMan MGB real-time fluorescent PCR was established to detect Diaporthe phaseolorum var.caulivora (DPC) and D. phaseolorum var.meridionalis (DPM). The primers and TaqMan MGB probes were designed based on the ITS of DPC, DPM, D. phaseolorum var. sojae and Phomopsis longicolla. A series of genomic DNA dilution were used to detect sensitivity of the technique, the results showed that the limits of detection for DPM and DPC were 7 fg/μL and 6 fg/μL DNA respectively.  相似文献

油菜菌核病菌对多菌灵和乙霉威的抗药性机理   总被引:1,自引:0,他引:1  
生物测定结果表明,油菜菌核病菌田间菌株对多菌灵(MBC)敏感性表型呈多样性,即存在MBCS、MBCLRLR、MBCHRHR和MBCVHR表型.而对乙霉威(DIE)则只检测到DIES和DIEHR表型.MBCS、MBCLR和MBCHR菌株中除JD2-3菌株为DIES外,其余菌株均为DIEHR,MBCVHR菌株对DIE表现为DIES.MBC和DIE之间存在典型的负相关交互抗性.序列分析结果表明,表型为MBCVHRDIES菌株的β-微管蛋白基因,第198位氨基酸由Glu(GAG)突变为Ala(GCG);表型为MBCHRDIEHR菌株的β-微管蛋白基因的突变位点在第200位,由Phe(TTC)突变为Tyr(TAC);而表型为MBCSDIES和MBCLRDIEHR的菌株在所扩增的β-微管蛋白基因片段中未发生突变.初步表明,β-微管蛋白基因198和200位氨基酸的突变是引起油菜菌核病菌对多菌灵抗药性呈多样性的分子机理.  相似文献

从中国山西、内蒙古、山东、海南、江苏、福建和陕西省 (自治区) 不同市、县从未使用过啶菌噁唑的露地或保护地采集并经单孢分离获得 166 株番茄早疫病菌,采用菌丝生长速率法测定了病原菌群体对啶菌噁唑的敏感性。结果表明:啶菌噁唑对番茄早疫病菌的 EC50 值范围在 0.11~2.29 mg/L 之间,敏感性频率呈正态分布,其均值 (0.56 ± 0.39) mg/L 可作为番茄早疫病菌对啶菌噁唑田间抗性监测的敏感基线;不同采集地菌株对啶菌噁唑的敏感性存在差异,其中采自福建省的菌株敏感性最高,而山西省大同、晋中和长治市的菌株敏感性最低。200 mg/L啶菌噁唑对番茄早疫病防治具有优异的保护和治疗作用,且接菌前 4 d 内施药和接菌后 1~2 d内施药防效最佳。研究表明,啶菌噁唑对番茄早疫病防治具有良好的实际应用前景。  相似文献

系统比较了从云南省主要葡萄产区采集的57株葡萄胶胞炭疽菌Colletotrichum gloeosporioides对三唑类脱甲基抑制剂烯唑醇、腈菌唑及咪唑类脱甲基抑制剂咪鲜胺的敏感性、交互敏感性及敏感性与生存适合度的相关性等指标。结果表明:供试菌株对咪鲜胺的EC50值在0.01~1.58 mg/L之间,高于其对烯唑醇(EC50值为0.05~25.45 mg/L)和腈菌唑(EC50值为0.49~192.93 mg/L)的敏感性;部分菌株对烯唑醇和腈菌唑的敏感性显著降低,而对咪鲜胺仍保持较高的敏感性;供试菌株对烯唑醇和腈菌唑的敏感性具有显著的相关性,而对咪鲜胺的敏感性与对烯唑醇和腈菌唑的敏感性之间则无显著相关性。低敏感性菌株的致病力和菌落生长速率与敏感菌株无显著差异,具有较高的生存适合度。  相似文献

BACKGROUND: Carbendazim (MBC) has failed to control wheat scab, caused by Fusarium graminearum Schwabe, on the eastern coast of China in recent years after about 30 years of application. RESULTS: MBC resistance was found to be common in pathogen populations on the eastern coast and along areas of the Yangtze River. EC(50) and minimum inhibitive concentration (MIC) values of MBC inhibiting mycelium growth of wild-type isolates were less than 0.9 and 1.4 microg mL(-1) respectively, while EC(50) values of resistant collections averaged 7.02 +/- 11.86 microg mL(-1). The slope of the MBC dosage-response curve (DRC) for resistant isolates of F. graminearum was flat: 1 < b < 2.8 for resistant isolates and 3.5 < b < 11 for sensitive isolates). Both field resistant and sensitive MBC strains shared similar temperature sensitivity, fitness and virulence on ears. Field resistant strains and UV-induced mutants showed positive cross-resistance to other benzimidazole derivatives and were mainly at intermediate MBC resistance level. Highly resistant field MBC strains rarely appeared, but only some of the highly resistant MBC UV mutants were insensitive to N-phenylaminecarbamates. No mutation in beta-tubulin was found in F. graminearum, in contrast to mutation in this tubulin which has led to MBC resistance in other plant pathogens. CONCLUSION: MBC(R) isolates have high fitness and competition in field, conferred by a novel molecular mechanism.  相似文献

 引起小麦赤霉病的禾谷镰孢菌(Fusarium graminearum)在华东地区对多菌灵已出现了高水平抗药性。本文报道了F.graminearum对多菌灵的敏感性基线及其抗药性菌株生物学特性。离体条件下多菌灵对100个野生敏感型菌株的平均EC50和MIC值分别为0.5748±0.0133 μg/ml和 < 1.4μg/ml。而对50个抗药性菌株的平均EC50和MIC值则分别为9.2375μg/ml和 > 100 μg/ml。在麦穗上多菌灵对50个敏感菌株和抗药性菌株防效的平均EC50分别为282.6 μg/ml和 > 2 000μg/ml。从田间获得的抗多菌灵菌株对苯菌灵、噻菌灵、甲基托布津表现交互抗药性,但不同于室内突变菌株,对乙霉威不表现负交互抗药性。抗药性菌株的无性和有性繁殖后代以及在无药培养基上菌丝体转代培养后,抗药水平保持不变。抗药性菌株的菌丝生长速率、产孢能力及致病力等与敏感菌株相比没有差异。  相似文献

Sensitivity to tricyclazole of 129 single-conidial isolates of rice blast fungus, Magnaporthe grisea, was determined. EC50 values ranged from 0.06 to 1.12 mg/L with an average value of 0.46 ± 0.09 mg/L according to the detached leaf segment tests. No significant difference of sensitivity was observed between isolates from Guangdong and Jiangsu where decreased efficacy was reported, and from two other provinces where tricyclazole provided excellent disease control. In seedling tests, tricyclazole could control the most tolerant isolate GY-6 successfully with a efficacy of 81.5% at the concentration of 40 mg/L. Sensitivities of GY-6 and DY-2, the most sensitive isolate, to tricyclazole were both unstable in sub-cultured single-conidial offspring isolates, with respective mean EC50 values of 5.40 ± 0.97 and 4.50 ± 0.88 mg/L calculated from seedling tests. There was no amino acid difference between them in the coding sequences of 1,3,6,8-tetrahydroxynaphthalene reductase and 1,3,8-trihydroxynaphthalene reductase. These results suggested that the decreased control reported in Guangdong and Jiangsu could not be attributed to the occurrence of resistance. When continuously “inoculated-reisolated-reinoculated” under the selection of tricyclazole in vivo, sensitivity of DY-2 decreased 10-fold after 20 generations, although the sensitivity of GY-6 did not shift significantly.  相似文献

为明确江苏省葡萄炭疽病菌Colletotrichum gloeosporioides群体对苯醚甲环唑、吡唑醚菌酯、咯菌腈和戊唑醇4种杀菌剂的敏感性和抗性水平,本研究采用菌丝生长速率法测定了来自江苏省7个地区的68株葡萄炭疽病菌对上述4种药剂的敏感性,建立敏感基线,并分析菌株抗性水平和抗性频率。结果表明:苯醚甲环唑、吡唑醚菌酯、咯菌腈和戊唑醇对供试68株葡萄炭疽病菌的EC50范围分别为0.03~1.20、0.05~3.48、0.14~1.20 mg/L和0.50~12.30 mg/L;对苯醚甲环唑、吡唑醚菌酯和咯菌腈的敏感基线分别为(0.53±0.032)(1.00±0.15) mg/L和(0.38±0.024) mg/L。68株菌株对戊唑醇的EC50平均值为3.61 mg/L,因敏感性频率分布不符合建立敏感性基线要求,故未建立敏感性基线。供试菌株对苯醚甲环唑均表现为敏感,对吡唑醚菌酯和咯菌腈呈现一定的低抗水平,抗性频率分别为4.41%和3.03%;对戊唑醇呈低抗和中抗水平,抗性频率分别为16.18%和5.88%。该研究为防控江苏省葡萄炭疽病提...  相似文献

Fungicide sprays on soybean in Brazil have contributed to the selection of less sensitive isolates of Corynespora cassiicola. We collected 59 isolates of Ccassiicola from three Brazilian states and two isolates from Paraguay. We investigated their EC50 to quinone outside inhibitors (QoI) and methyl benzimidazole carbamate (MBC), any cross-resistance to compounds within QoI and MBC groups, and characterized the polymorphisms in their cytb and β-tubulin genes. Local associations of polymorphisms identified in each gene were statistically correlated with assays results. In total, 79% and 74% of the isolates were classified as resistant to QoI and MBC fungicides, respectively. There was positive cross-resistance to active ingredients within QoI and MBC groups. For QoI, all isolates presented heteroplasmy in G143A of cytb gene; the mutations F129L and G137R were not found. For MBC, 63% of isolates possessed E198A and 21% possessed F200Y mutations, associated with reduced control by MBC fungicides. Heteroplasmy was identified in two and one isolates from Brazil with E198A and F200Y mutations, respectively. The resistance factor for isolates with E198A (10.9) was statistically similar to the isolate with F200Y (8.8) mutation. Genic association analysis of the in vitro assays using discriminatory doses proved them to be accurate. Reduced sensitivity of Ccassiicola to QoI and MBC was also identified in isolates from Paraguay and resistance to QoI and MBC was widely present in Ccassiicola isolates from the main soybean-producing states in Brazil. Thus, integrated management measures should be adopted to manage soybean target spot in these countries.  相似文献

黑斑病是影响白术种植的主要病害之一,2015—2017年从浙江省磐安、天台、新昌和嵊州4地采集、分离得到137株白术黑斑病菌Alternaria alternata,采用菌丝生长速率法检测其对甲基硫菌灵和腐霉利的抗性。结果表明:供试的黑斑病菌群体 (n = 137) 对甲基硫菌灵的高水平抗性频率为77.4%,中等水平抗性频率为22.6%,未检测到敏感菌株;对腐霉利的抗性频率为20.4%,含21株低水平抗性菌株和7株中等水平抗性菌株。7种杀菌剂的室内毒力测定结果表明:咪鲜胺对白术黑斑病菌的活性最高,EC50值为0.15 mg/L。供试的137株白术黑斑病菌对咪鲜胺的EC50值分布在0.05~0.87 mg/L之间,平均EC50值为 (0.29 ± 0.11) mg/L,可作为敏感性基线。  相似文献
