At 35 days of age, chickens which as 1-day-old chicks were inoculated with the infectious bursal disease virus (IBDV) had significantly lower antibody titers against Mycoplasma synoviae, Newcastle disease virus, and infectious bronchitis virus than did those never inoculated with IBDV. The IBDV also had a marked effect on the development of air-sac lesions. Birds infected with IBDV that were later inoculated with M synoviae (day 14), Newcastle disease virus (days 14 and 28) experienced an increased incidence and greater seversity of airsacculitis than did chicks which were not exposed to IBDV.  相似文献

SUMMARY: Australian lentogenic Newcastle disease viruses were evaluated as uninactivated vaccines in Australian chickens, the response being evaluated by the production of haemagglutination-inhibition (HI) antibodies. Two viruses, V4 and PM9, induced high levels of antibody and were readily transmissible between chickens by contact exposure. Three other viruses were poorly immunogenic and poorly transmissible. Chickens vaccinated intramuscularly with the V4 strain produced higher HI antibody titres than chickens vaccinated by the orotracheal, intranasal and intraocular routes. HI antibody titres in chickens vaccinated with the V4 strain reached peak levels 3 to 5 weeks after vaccination and waned considerably during the next 2 to 4 weeks. However, low levels of HI antibody persisted for at least 36 weeks after vaccination. Intramuscular vaccination with the V4 strain of one-day-old chicks lacking maternal antibody to Newcastle disease virus resulted in 42–70% mortality and the survivors developed very high titres of HI antibody. Similar chickens inoculated orotracheally showed signs of depression and developed high titres of HI antibody, but there were no mortalities. Chickens 1-, 2-, 3- and 4-weeks-old and lacking maternally derived HI antibody to Newcastle disease virus suffered no adverse reaction to intramuscular or orotracheal vaccination. The antibody response of the 1-week-old chickens was considerably poorer than that of the older chickens. Following orotracheal vaccination with the V4 strain, chickens with low levels of maternally derived antibody responded with low levels of HI antibody. On the other hand, in the progeny of hens hyperimmunised with the V4 strain the production of active antibody following orotracheal vaccination was delayed until the level of passive antibody had declined considerably. There was no response to intramuscular vaccination in congenitally hyperimmune chickens. The minimum HI antibody inducing dose of V4 vaccine, when measured 3 weeks after vaccination of 6-weeks-old chickens, was 105.6 50% egg infectious doses.  相似文献

The safety and efficacy of the cell-associated (C-A) vaccine prepared by chicken embryo fibroblast (CEF) cells infected with the tissue-culture-modified strain of infectious laryngotracheitis (ILT) virus were studied in chickens. Over seventy percent of chickens inoculated with the C-A vaccine by the subcutaneous (S.C.) or intramuscular (I.M.) route at 1 day of age was protected against challenge with a virulent strain of ILT virus without any clinical signs. Chickens vaccinated with the C-A vaccine at 1 day of age acquired immunity within 6 days after vaccination, and the protection rate maintained more than 60% until 10 weeks post-vaccination. The C-A vaccine was invariably effective for chickens at various age. There was no evidence that the development of immunity was hindered by further vaccination with Newcastle disease and infectious bronchitis combined live vaccine. In addition, the C-A vaccine was safe when chickens were inoculated with 10 doses. In the field trials of the C-A vaccine, no adverse reaction was observed, and over 65% of vaccinated chickens was protected against the challenge of the virulent ILT virus at 8 weeks after vaccination.  相似文献

The efficacy of green-coloured (GC) I-2 Newcastle disease vaccine was determined in the present study. I-2 vaccine was mixed with a green coloured dye and stored at 4°C for 6 months while assayed for the virus infectivity at a monthly interval. Chickens were vaccinated with the GC vaccine by eye drop. Serum samples were collected from all birds before and after vaccination at weekly interval for 4 weeks and tested for haemagglutination-inhibition (HI) antibody against Newcastle disease virus (NDV). These chickens were challenged with NDV virulent strain four weeks after vaccination. The results showed that there was no difference between the infectivity titres of GC and uncoloured vaccines. However, chickens vaccinated with GC vaccine produced higher HI antibody titres than chickens vaccinated with uncoloured vaccine. Results from the challenge trial showed that all vaccinated chickens survived whereas all unvaccinated chickens died. The findings from this study have shown that the GC vaccine is safe and produced protective antibodies against NDV in vaccinated chickens. Wambura, P. N., 2008. Chickens vaccinated once or twice with inactivated oil-emulsion vaccine against Mycoplasma gallisepticum (MG) or left unvaccinated were challenged intratracheally with the R strain of MG. The population of MG organisms was determined by enumerating tracheal cultures periodically up to 28 weeks postchallenge (PC). The number of organisms in the respiratory tract increased rapidly after 4 days PC, and the number tended to decrease after 4 weeks PC. Tracheal populations of MG varied considerably among individual chickens. Bacterin-vaccinated chickens had numerically lower populations of MG in their tracheas up to 8 weeks PC, but the differences were small and considered of little practical significance.  相似文献

Eighty-three-week-old table egg layers with swollen sinuses were presented with a history of increased mortality. Serology revealed positive titers to Mycoplasma gallisepticum (MG). The birds were part of a flock in which some birds had been vaccinated with 6/85 live MG vaccine at 18 wk of age. Tracheal cultures were obtained from both vaccinated and unvaccinated birds within the flock. The cultures were indistinguishable from 6/85 vaccine by both random amplified polymorphic DNA analysis and DNA sequence analysis. Challenge studies were performed to compare the field isolates with 6/85 vaccine and the R strain of MG. The field isolates produced a greater antibody response by serum plate agglutination than did the 6/85 vaccine. The isolates effectively colonized the trachea without increasing the tracheal mucosal thickness; however, they did not extensively colonize the air sacs or cause airsacculitis in the experimental birds.  相似文献

SUMMARY An Australian strain of Newcastle disease virus, was evaluated for use as a vaccine following its administration by drinking water, aerosol and spray to chickens at 1 and 21 days of age. Haemagglutination inhbition antibody was produced and persisted for 11 weeks. Aerosol vaccination induced higher levels of haemagglutination inhibition antibody than the other methods of vaccination. No respiratory disease was observed following vaccination. Chickens vaccinated by aerosol and spray were fully protected when challenged at 5, 7 and 11 weeks of age with virulent Newcastle disease virus. Mortality of 10 to 30 per cent was observed in chickens vaccinated by drinking water and intranasally following challenge.  相似文献

Avridine, a lipoidal amine with interferon-inducing and adjuvant properties, was an effective adjuvant for Newcastle disease antigen (NDA) in chickens. Eleven vaccine lots were evaluated: 2 commercial water-in-oil vaccines, 4 experimental oil emulsion vaccines, 4 avridine-containing vaccines, and a control lot of nonadjuvanted antigen. Avridine significantly enhanced the immunologic responses of chickens against NDA. Chickens vaccinated with the avridine-containing vaccines had significantly higher antibody titers (hemagglutination inhibition) than did chickens vaccinated with the commercial vaccines. Experimental oil emulsion vaccines prepared from the same antigens as avridine-adjuvanted vaccines induced higher hemagglutination inhibition antibody titers after primary but not after booster vaccination. Use of avridine as an adjuvant for NDA in vaccines for chickens induced immunologic protection rates similar to those induced by oil emulsion vaccines, without causing the reactogenic and tissue residue problems associated with the use of oil vaccines in chickens.  相似文献

A temperature sensitive (ts) vaccine strain designated ts-11 was selected after exposure of a low passage culture of the immunogenic Australian field isolate (strain 80083) of Mycoplasma gallisepticum to 100 mg/ml of N-methyl-N-nitro-N-nitrosoguanidine. Viable counts (assayed as colour changing units (CCU)/25 microliters) of a thawed stock culture of ts-11 were typically log10 3 to log10 5 higher when incubated at 33 degrees C (the permissive temperature) than duplicate viable counts incubated at 39.5 degrees C (the restrictive temperature). Doses of approximately 2 x 10(7) CCU of ts-11 caused no gross lesions or loss of egg production when inoculated into the air sacs of susceptible chickens and no clinical or pathological signs of sinusitis when inoculated into the infraorbital sinuses of susceptible turkey poults, whereas the parent strain 80083 was demonstrably pathogenic. However, 1 of 10 poults inoculated intra-abdominally with approximately 2 x 10(7) CCU of ts-11 did show signs of mild airsacculitis. Eight-week-old pullets were vaccinated by eye drop with up to 1.4 x 10(7) CCU of ts-11 and simultaneously subjected to several stressful management practices, without apparent ill effects. Administration by coarse aerosol of 5 ml of ts-11 vaccine/25 day-old broilers, with or without 25 doses of infectious bronchitis virus vaccine caused no obvious signs of respiratory disease. The non virulent ts phenotype was maintained after 3 passages of strain ts-11 in chickens. A series of trials was conducted in which specific-pathogen-free (SPF) leghorn chicks were exposed to various isolates of Alcaligenes faecalis. Chicks were inoculated with A. faecalis alone or in combination with Newcastle disease/infectious bronchitis (Nc/Br) vaccine, laryngotracheitis vaccine, infectious bursal disease virus, or Mycoplasma gallisepticum. The response was evaluated by morbidity, mortality, airsacculitis, reisolation of A. faecalis, and histopathological lesions of tracheas. Although A. faecalis was recovered up to 42 days postinoculation in some cases, no clinical signs were directly attributed to simple A. faecalis infection. None of the other agents significantly increased the severity of A. faecalis signs or lesions, except that A. faecalis-infected chicks that were given Nc/Br vaccine had prolonged microscopic tracheal lesions. In another trial, the effects of A. faecalis in young SPF leghorns, non-SPF broilers, and turkeys were compared. Broiler-type chicks were more susceptible than leghorns and less susceptible than poults. Consequently, the use of leghorns as a model for studying this infection is questioned.  相似文献

Broiler chicks were administered vaccines against Newcastle disease and infectious bronchitis (both Arkansas and Massachusetts strains) at 2 weeks of age as either primary or secondary vaccinations. The vaccine was administered as a spray at 2 weeks of age to chicks that had received Newcastle disease vaccine alone, bronchitis vaccine alone, both vaccines in combination, or no vaccine at day 1 in the hatchery. The Newcastle disease hemagglutination-inhibition response was significantly lower in chicks receiving Newcastle disease vaccine as a secondary vaccine at 2 weeks than in those receiving the vaccine as a primary vaccination at that age. In contrast, the bronchitis hemagglutination-inhibition response was significantly higher in chicks receiving bronchitis vaccine as a secondary vaccination at 2 weeks than in those receiving the vaccine as a primary vaccination at that age.  相似文献

Infectious bronchitis virus CA99 serotype was isolated from several broiler flocks in Northern California. The virus caused late-onset respiratory disease and increased airsacculitis condemnation in affected flocks despite the use of an established infectious bronchitis virus vaccination program. An experimental study compared Holland/Arkansas and Massachusetts/Arkansas vaccination protocols to determine the efficacy of commercial infectious bronchitis virus vaccines in reducing respiratory disease and airsacculitis lesions found at processing that were associated with a CA99 field isolate. All vaccination groups were given Massachusetts/Connecticut strains of infectious bronchitis virus vaccines at age 1 day followed by vaccination with either Holland/ Arkansas or Massachusetts/Arkansas vaccine strains at 18 days of age. Birds were challenged at age 31 days with a CA99 field isolate. Gross pathology, histopathology, and virus isolation were evaluated. Chickens vaccinated with Holland/Arkansas had marginally better protection against CA99 challenge than chickens vaccinated with Massachusetts/Arkansas, although differences were not statistically significant.  相似文献

Newly hatched chickens were immunized with a temperature-sensitive (TS) Mycoplasma gallisepticum (MG) mutant (TS 100). Immunized chickens resisted challenge with the virulent S6 strain. The dose of TS MG needed for protection was less than 3.3 X 10(4) colony-forming units. After immunization with TS 100, chickens were subjected to a variety of virus infection and immunosuppressive treatments. Neonatal bursectomy or thymectomy, infectious bursal disease virus infection, and infectious bronchitis virus vaccination or a combination of infectious bronchitis virus and Newcastle disease virus vaccination did not contribute to the development of air-sac lesions in TS MG-immunized chickens. Infectious bronchitis virus vaccination enabled MG to migrate from the nasal cavity to the trachea but not to the air sacs. The TS MG vaccine appears to be a safe immunogen.  相似文献

An international effort (sponsored by the Australian Centre for International Agricultural Research) is being made to develop oral vaccines that will protect village chickens against Newcastle disease. The vaccines being used are derivatives of the avirulent Australian V4 strain that have been selected for enhanced heat resistance. The present study, undertaken in Sri Lanka, used local processed (parboiled) rice as a vehicle for the vaccine. Chickens receiving two doses of vaccine on cooked, parboiled rice were completely protected against contact challenge with the virulent SL 88/1 Sri Lankan strain of Newcastle disease virus Chickens kept in contact with these vaccinated chickens were similarly protected. Lower levels of protection were achieved with vaccine given on uncooked parboiled rice. V4 vaccine administered intranasally also gave complete protection. Serums from vaccinated chickens that survived challenge were tested for haemagglutination-inhibition antibodies, using both vaccine virus and challenge virus as antigens. Attenuated derivatives (delta cya delta crp mutants) of an O2 and an O78 avian septicemic Escherichia coli strain were used to immunize broiler chickens by spray to determine the safety, immunogenicity, and efficacy of the derivatives in single- and double-dose regimens. In the safety and immunogenicity studies, groups of 10 chickens were vaccinated by spray (droplet size approximately 20 microm) with the parent E. coli, the mutant organisms, or phosphate-buffered saline (PBS) at 14 days of age and euthanatised 21 days later. There was no deaths or gross pathologic finding in any of the chickens immunized with the vaccine strains. Compared with the levels in chickens exposed to PBS, there were significantly higher levels of immunoglobulin (Ig) G antibody in serum and air sac washings and of IgA antibody in air sac washings in response to the virulent parent strains than to the vaccine strains. In efficacy studies, chickens were immunized with the O2 or the O78 vaccine strain or PBS at day 14 and with the O2 vaccine strain or PBS at days 10 and 14 and challenged with the parent strain 10 days after the last vaccination. There was no significant difference in local IgA and IgG and serum IgG responses between vaccinated and control groups. Chickens vaccinated with the O2 strain, but not the O78 strain, had significantly lower air sac lesion scores compared with those of the unvaccinated groups in both single- and double-dose regimens. We conclude that the mutant O2 strain provided moderate protection against airsacculitis.  相似文献

Twelve-day-old chickens were vaccinated once with different Newcastle disease (ND) vaccines ( F, La Sota and Mukteswar) by two different routes (intraocular and drinking water). Chickens from a seventh group were uninoculated controls. At weekly intervals for 7 weeks after vaccination, 20 chickens from each vaccinated group and 20 chickens from the control group were examined for the production of haemagglutination-inhibition (HI) antibodies and for protection as assessed after challenge with velogenic, viscerotropic ND virus.

La Sota ND vaccine used intraocularly ranked the best and Mukteswar vaccine by the drinking water route the worst for their HI antibody titres prior to challenge. Differences between the treatments in protection were examined. For all three vaccines intraocular vaccine produced higher protection than drinking water vaccine. An inverse relationship between prechallenge and postchallenge HI titres was also recorded.  相似文献

Serial passage of two infectious bronchitis virus (IBV) vaccine strains in chickens enhanced their capacity to increase the incidence and severity of Mycoplasma synoviae (MS) airsacculitis. Included in this report were the mild Massachusetts-type Connaught strain and the Arkansas 99 vaccine strain of IBV. The Connaught strain and one of two Ark 99 vaccine strains passaged in chickens increased the incidence of airsacculitis markedly compared with nonpassaged virus. The other Ark 99 vaccine virus already exacerbated MS airsacculitis, before passage in chickens, and its influence did not increase on passage. All IBV strains studied to date have either possessed this trait or reacquired it on passage in the natural host.  相似文献

Two experiments were conducted to evaluate the virulence and vaccination efficacy of a Mycoplasma gallisepticum (MG) isolate designated MG Intervet 6/85. Virulence of the strain was determined by evaluation of airsacculitis scores following aerosol exposure to the isolate before and after 10 sequential passes in either commercial broiler chickens or commercial turkeys. Two-week-old specific-pathogen-free chickens were vaccinated by aerosol exposure. The birds were challenged with the R' strain of MG at either 4 or 8 weeks post-vaccination. Efficacy was evaluated by airsacculitis scores determined 21 days after challenge. Ten repetitive back-passes of the isolate in chickens and turkeys did not substantially increase the virulence. Virulence for both chickens and turkeys was minimal, while protection elicited by aerosol vaccination in young chickens against virulent R' strain was significant (P less than or equal to 0.05) compared with unvaccinated controls.  相似文献

本研究以新城疫病毒(NDV)V蛋白羧基端结构域(Vc)的重组蛋白为包被抗原,建立了用于检测NDV V蛋白抗体的间接ELISA方法,并采用该方法检测了鸡群免疫或接毒后血清中的V蛋白抗体水平。结果显示:两组不同NDV灭活疫苗组在免疫后的3周内检测结果均为阴性;两组灭活疫苗免疫3周后再人工感染NDV强毒的鸡群,攻毒后第7、14和21 d,NDV阳性率分别为60%、80%、70%和50%、80%、70%;两组不同的NDV弱毒疫苗免疫组鸡群,仅在免疫后第21 d阳性率分别为20%和10%。以上结果表明,NDV疫苗免疫组与强毒感染组的V蛋白抗体阳性率存在明显差异,本方法可在群体水平上区分新城疫疫苗免疫与强毒感染鸡群,为NDV血清学诊断和流行病学调查提供了一种新的检测手段。  相似文献
