本研究针对大蒜提取物对印度明对虾(Fenneropenaeus indicus)黑腮病病原多重耐药哈维氏弧菌(Vibrio harveyi)体外和体内的抗菌活性进行了评价。先利用市售的几种抗生素记录致病性哈维氏弧菌的抗菌谱。然后,利用琼脂扩散法来研究大蒜对哈维氏弧菌的抗菌活性。将哈维氏弧菌接种在琼脂平板上,对每种大蒜提取物(鲜榨大蒜提取物—FSGE,冻干大蒜提取物—FDGE以及用甲醇提取的大蒜提取物—MGE)设计三种不同的浓度。结果表明,鲜榨大蒜提取物对哈维氏弧菌的抑菌活性明显优于冻干大蒜提取物和用甲醇提取的大蒜提取物。本研究分别在对虾基础饲料中添加0%(对照)、0.5%和1%浓度梯度的大蒜来评价大蒜治疗感染哈维氏弧菌的对虾的效果。结果表明,综合大蒜提取物的短期和长期防治(1%含量)的效果分析,对虾累积死亡率降低了75%。本研究探讨了鲜榨大蒜提取物替代抗生素在印度明对虾饲养防病上的优点。  相似文献

Herbals such as Cynodon dactylon, Piper longum, Phyllanthus niruri, Tridax procumbens, and Zingiber officinalis were extracted with acetone, benzene, butanol, and petroleum ether and screened against the pathogen Vibrio harveyi isolated from the infected Grouper Epinephalus tauvina. Among the different solvent extractions screening to V. harveyi, petroleum ether extracts were suppressed significantly (P < 0.05). Equal proportions of the all-plant extracts were mixed with the artificial feeds at concentrations of 100, 200, 400, and 800 mg kg−1 of diet and fed to grouper juveniles of 20 ± 2 g average weight for a period of 60 days. Every 20 days, fish juveniles were challenged with V. harveyi and the immune response was studied. The herbal diets significantly (P < 0.05) increased the survival, growth, and immune responses compared to the control group. The herbal diets were significantly improved (P < 0.01) in immune parameters such as phagocytic activity and albumin–globulin (A–G) ratio. Among the different concentrations of the herbals in the diet, the 400 mg kg−1 diet was the most effective in the experiment.  相似文献

The pathways by which pathogens invade Fenneropenaeus indicus, the potential colonization in various tissues and the disease transmission mechanisms are unclear. The aims of the present study were to visualize the colonization and pathogenesis of GFP‐tagged Vibrio parahaemolyticus, in various tissues of F. indicus to evaluate the pathogen interaction. Among the three strains isolated, a virulent strain VpDAHV2 was tagged with green fluorescent protein (GFP‐VpDAHV2) and validated for both its growth characteristics and its virulence as a genuine model for F. indicus infection. VpDAHV2 was positive for toxR and tlh genes and negative for tdh genes. CLSM images revealed that maximum colonization was observed in the haemolymph of the F. indicus challenged with GFP‐VpDAHV2. The haemolymph was the primary site for the colonization of GFP‐VpDAHV2 in F. indicus. The enteric localization occurred independently of the flagellum or motility of GFP‐VpDAHV2 through the intestinal route. The F. indicus infection model suggests that the haemolymph and the intestine represent the sites of infection by GFP‐VpDAHV2, and hence are the active sites of pathogen interactions. GFP tagging of V. parahaemolyticus is a new and systemic approach to determine the presence of bacteria in vivo for the confirmation of host pathogen interactions in aquaculture studies.  相似文献

Effect of ambient copper on the immune, antioxidant and lipid peroxidation parameters of Fenneropenaeus indicus and its susceptibility to white spot syndrome virus under heavy metal (Cu2+) exposure has been studied. Adult shrimps were acclimated to 25‰ salinity for a period of 7?days, and after 12?h of starvation, shrimps were dosed with 0.075, 0.150, 0.225 and 0.30?ppm Cu2+ by adding appropriate quantities of copper sulphate solution. After 14?days of metal exposure, the shrimps were challenged with white spot syndrome virus through oral administration and the immune and antioxidant parameters were analysed. Analysis of variance showed that there were significant differences (P?<?0.05) in the immune and antioxidant parameters and lipid peroxidation product in different treatment groups of F. indicus compared to untreated group. The immune parameters and antioxidant enzymes have been significantly lowered (P?<?0.05) in the haemolymph of shrimps dosed with higher concentrations of copper sulphate leading to lipid peroxidation and accumulation of malondialdehyde. However, there was less oxidative stress in shrimps exposed to 0.075?ppm Cu2+. The present study showed that high levels of Cu2+ enhanced the mortality of F. indicus with concomitant reduction in immune and antioxidant parameters. A concentration of 0.075?ppm Cu2+ in the rearing water was found to have beneficial effect in shrimps in terms of immunostimulation and higher survival against WSSV infection. This can be adopted as a pharmacological approach towards shrimp health management in culture systems.  相似文献

Bacterial subcellular components and probiotics were successful for the stimulation of immunity and the prevention of Vibrio harveyi infections in rainbow trout, Oncorhynchus mykiss (Walbaum). Rainbow trout were immunized with whole inactivated cells of V. harveyi to obtain polyclonal antibodies against specific antigens. Western blotting showed a unique reactive band (∼93 kDa) between serum and bacterial proteins from outer membrane proteins (OMP) and extracellular products (ECP). Probiotics were selected according to their capability to inhibit V. harveyi . Two of these bacteria, i.e. A3-47 and A3-51, showed cross-reactivity with V. harveyi antiserum. Their OMPs and ECPs were reactive with V. harveyi antiserum in bands of ∼93 kDa for A3-51 and higher for A3-47. In vivo tests determined that fish fed with A3-51 produced cross-reactive antibodies against V. harveyi and also, the survival of these fish infected with V. harveyi was high, being similar to the level achieved with vaccinated fish. Thus, the probiotics, when administered as live preparations, were capable of producing cross-reactive antibody against specific bacterial pathogens.  相似文献

A comparative study was undertaken to assess the effect of sous vide (SV) processing, vacuum, and ordinary air packaging on the quality and shelf life of condiment-incorporated Indian white shrimp (Fenneropenaeus indicus) during chilled storage. The pasteurization value (PV) for SV-processed shrimp was 4.02 min at 90ºC. The initial K value of shrimp (2.17%) increased to 5.26% with SV processing and further increased to a value of 46.9% on the day of sensory rejection, compared with 64.3% and 62.7% for vacuum and air-packed samples, respectively. A nearly 3 log reduction in total mesophilic count was observed for SV-processed shrimp. Formation of volatile bases, indole, and lipid oxidation was significantly (p < 0.05) inhibited in SV-processed shrimp compared to others. SV processing extended the shelf life of condiment-incorporated shrimp up to 28 days, compared to only 15 and 8 days for vacuum and air-packed samples, respectively.  相似文献

哈维氏弧菌(Vibrio harveyi)是导致养殖对虾暴发弧菌病的重要病原之一。从我国南方养殖凡纳滨对虾(Penaeus vannamei)分离、鉴定了10株哈维氏弧菌(Vh00947、Vh00949、Vh11011、Vh11014、Vh21217、Vh21218、Vh21220、Vh21229、Vh21231和Vh31487),分别肌注感染健康凡纳滨对虾后发现,菌株Vh21229致病性很弱,Vh00949其次,其它菌株毒力较强;对8种哈维氏弧菌常见毒力基因(Vh1、Vh2、Vh3、Vh4、tox S、hcp、zot和pap6)的检测显示,南方虾源哈维氏弧菌无论强弱毒株都很少携带zot、pap6、toxs和4种Vh基因(≤10%),表明这10个菌株的致病性与这7种常见毒力因子的相关度很低;hcp基因在所有菌株中均有检出,其中强毒株中检出率较高(50%),在较弱毒株(Vh00949)中也存在,表明hcp基因与这些菌株的致病性密切相关,但不是决定性因子。因此,这10株南方虾源哈维氏弧菌菌株的致病性差异应由这8种常见毒力因子以外的未知或未检测到的因子所决定。生化特征分析显示,所有菌株中只有弱毒株Vh21229不能利用D-甘露糖、蔗糖、D-海藻糖,而且只有其赖氨酸脱羧酶反应至阳性,而其它菌株均为阴性,推测导致哈维氏弧菌菌株生化特性改变的某种因子可能对菌株的致病性也产生了影响。药敏实验表明,10株哈维氏弧菌均对环丙沙星、氯霉素、恩诺沙星、美罗培南、头孢曲松、多西环素、头孢吡肟、诺氟沙星敏感,而对氨苄西林耐受性强,表明在虾类养殖过程中应当严格规范和控制抗菌药物尤其是青霉素类药物的使用。  相似文献

Vibriosis is a severe infection occurring in many commercially important marine fish species. In this study, vaccines containing Vibrio harveyi recombinant outer membrane protein K (rOmpK), outer membrane protein U (rOmpU) and rOmpK‐OmpU fusion protein in addition to the metabolizable MontanideTM ISA 763 A VG adjuvant were developed and evaluated in the orange‐spotted grouper. The results indicate that recombinant V. harveyi protein‐based vaccines resulted in a remarkably higher expression of IL‐1β and IL‐8 at 24 hr, and greater antibody production, as early as 2 weeks postimmunization. Notably, enhanced immune responses and significant protective efficacy against V. harveyi infections were observed in the fusion protein vaccine‐injected fishes with relative per cent survival value of 81.8%. Additionally, the rOmpK‐OmpU antisera presented a high bactericidal effect on not only V. harveyi, but also Vibrio parahaermolyticus and Vibrio alginolyticus. Our results demonstrated that the fusion protein rOmpK‐OmpU was an effective vaccine candidate that exhibited potentially great versatility for controlling vibrio infections.  相似文献

This experiment was carried out to investigate the effect of feeding low protein diets (~20BFd20.13; ~22BFd22.20; ~24BFd24.32, and ~26BFd26.44) in the presence of the biofloc on the growth performance and feed utilization of (Fenneropenaeus indicus) in comparison with commercial feed of 35% CP as control. The present study was based on application of genetic, nutritional and pathological tools. The biofloc was developed in the low protein diet using molasses as a carbon source. Fifty juveniles F. indicus with an average body weight of 0.52?±?0.03 g were stocked in 50 L plastic tanks. Low protein diets and the control were tested in three replicates over a 112 days feeding trial. There were significant differences in protein, amino acids profile, lipids and fatty acids profile between the control and low diets group in the presence of the biofloc. However, less variation was noticed in the whole body composition (protein, amino acid profile, lipids and fatty acid profiles) of the shrimp between the control and low diets groups in the presence of the biofloc. There were no significant differences in final weight between control and low diets groups. Same trend was noted in the SGR, which did not vary significantly between low protein diets and control. However, the utilization of the biofloc by shrimp as a feed source was evident by the significant differences (P<?0.05) in FCR between low protein diets and control. There was a significant difference in the shrimp survival (%) (P<?0.05) between the low protein diets and the control which ranged between 86.66 % and 66.66 %, respectively. The development of biofloc significantly reduced the TAN, nitrate NO3-N and nitrite NO2-N levels in the low protein diets tanks. Diagnostic of mortality cases revealed that biofloc can serve as a biosecurity system for shrimp farmimg. RAPD-PCR were used to study the epidemiology of Vibrio parahaemolyticus responsible for early mortality syndrome (EMS) isolated from the water of culture system. The OPC5 (GATGACCGCC) primer produced bands ranged from 1 to 8 with sizes from 0.2–5.0 kb.  相似文献

Feeding aquatic animals with bacterial encapsulated heat‐shock proteins (Hsps) is potentially a new method to combat vibriosis, an important disease affecting aquatic animals used in aquaculture. Food pellets comprised of shrimp and containing Escherichia coli overexpressing either DnaK‐DnaJ‐GrpE, the prokaryotic equivalents of Hsp70‐Hsp40‐Hsp20, or only DnaK were fed to juveniles of the white leg shrimp Penaeus vannamei, and protection against pathogenic Vibrio harveyi was determined. Maintaining pellets at different temperatures for varying lengths of time reduced the number of live adhering E. coli, as did contact with sea water, demonstrating that storage and immersion adversely affected bacterial survival and attachment to pellets. Feeding P. vannamei with E. coli did not compromise their survival, indicating that the bacteria were not pathogenic to shrimp. Feeding P. vannamei with pellets containing bacteria overproducing DnaK (approximately 60 cells g?1 pellets) boosted P. vannamei survival twofold against V. harveyi, suggesting that DnaK plays a role in Vibrio tolerance. Pellets containing DnaK were effective in providing protection to P. vannamei for up to 2 weeks before loss of viability and that DnaK encapsulated by these bacteria enhanced shrimp resistance against Vibrio infection.  相似文献

以初体重为(7.20±1.38)g的凡纳滨对虾(Litopenaeus vannamei)为研究对象,在室内养殖箱进行3周的养殖实验和2周的哈维氏弧菌(Vibrio harveyi)人工感染实验,其中对照组每日投喂普通商品饲料,实验组每日投喂在普通商品饲料中添加地衣芽孢杆菌(Bacillus licheniformis)1.0×10~8 CFU/m L配制成的实验饲料。目的是研究饲料中添加益生菌对凡纳滨对虾肠道菌群、Toll受体及溶菌酶基因表达量和抗哈维氏弧菌能力的影响。实验结果表明,在饲料中添加地衣芽孢杆菌可显著提高对虾抗哈维氏弧菌感染的能力,其相对免疫保护率为22.22%。与对照组相比,实验组可显著降低对虾肠道内弧菌数量(P0.05)。感染哈维氏弧菌后,实验组溶菌酶(lysozyme,LZM)m RNA的相对表达量在12 h、18 h、24 h、36 h、48 h、72 h均显著高于对照组(P0.05)。实验组感染哈维氏弧菌后在6 h、12 h、18 h、24 h、36 h、48 h、72 h、7 d的Toll受体(Toll receptor)m RNA的相对表达量均显著高于对照组(P0.05)。实验结果提示:饲料中添加地衣芽孢杆菌可有效提高对虾抗哈维氏弧菌感染的能力,这种能力的提高可能是通过降低对虾肠道内的弧菌量,并提高抗病相关基因的表达量实现的。  相似文献

刺激隐核虫是一种原生纤毛类寄生虫,可以感染几乎所有海水硬骨鱼类并导致死亡,给海水鱼类养殖业造成巨大经济损失。由于刺激隐核虫体表寄生的特性,使其成为研究鱼类黏膜免疫机制的良好病原模型,可为高效疫苗的研发提供理论依据。本文综述了鱼类抗刺激隐核虫感染的黏膜免疫研究进展,以期为开展海水鱼类抗刺激隐核虫感染的免疫防控措施研究提供理论支撑。已有研究表明,受刺激隐核虫感染后,斜带石斑鱼皮肤黏液或其培养上清液能使幼虫发生阻动,由皮肤中的抗体分泌细胞产生的特异性IgM抗体在抗寄生虫感染中发挥重要作用;同时在刺激隐核虫感染时,多种免疫细胞如NCC细胞、中性粒细胞等在寄生虫周围聚集,趋化因子以及趋化因子受体表达量在寄生虫感染部位上调,暗示其调节的免疫细胞也参与抗虫免疫;此外,研究发现黄斑蓝子鱼对刺激隐核虫具有天然抗性,其血清和皮肤黏液对刺激隐核虫幼虫和滋养体均具有杀灭作用,已从其血清中分离到一种天然抗虫蛋白—L-氨基酸氧化酶,为刺激隐核虫病的防控提供新的途径;在理论研究的基础上,通过免疫实验证实疫苗防控刺激隐核虫病是可行的。  相似文献

Immune cells were identified and their interaction towards Vibrio alginolyticus, V. parahaemolyticus and V. anguillarum was studied in vitro in the penaeid shrimp, Penaeus indicus. Haemocytes were divided into agranulocytes, semi-dense granulocytes and dense granulocytes according to their morphology. Agranulocytes (100%) and 0.3–0.7% of granulocytes were actively involved in coagulation. Granulocytes were involved in in vitro phagocytosis and encapsulation of foreign materials. Phagocytosis was enhanced by prior opsonization of bacteria with cell-free shrimp haemolymph. Semi-dense granulocytes were phagocytic towards V. alginolyticus with and without opsonization at the rate of 91.1% and 83.1%, respectively ( P < 0.05 ). Granulocyte death observed after 2 h with opsonized haemolymph was 26.1%. About 64.5% of dense granulocytes and 23.2% of semi-dense granulocytes were actively involved in encapsulation, forming capsules. A spectrophotometric nitroblue tetrazolium (NBT) reduction assay was used to demonstrate the production of superoxide anions (O2) by shrimp haemocytes. All the Vibrio spp. were able to induce superoxide anions (O2) during phagocytosis. Live Vibrio sp. induced O2 production in haemocytes in a dose-dependent manner. Significant activity was detected with a 40:1 bacteria to haemocyte ratio ( P < 0.05 ). NBT reduction assay for measuring the post-phagocytic killing mechanism in shrimp haemocytes might be a valuable tool for monitoring shrimp health and immunological studies.  相似文献
