The incidence and geographic range of Dothistroma septosporum, a serious foliar pathogen of pine, has increased dramatically in central Europe in the last three decades. In Poland, it was first detected in 1990, and by the mid-2010s became established throughout the country, meaning the population of D. septosporum in Poland is relatively young. In this study we analysed site populations of the pathogen in Poland using highly variable microsatellite markers in order to identify the genetic signatures of a recent migration and to track the mode of its introduction. As a result, we found that in Poland the population of D. septosporum is highly structured, the strongest division occurring between a portion of isolates from south-central Poland, including the original outbreak site, and the isolates from the rest of the country. Further genetic structure allowed identification of population clusters specific to particular regions of Poland. Random mating tests show that some population groups have a much lower tendency to reproduce sexually than others, a factor that helps maintain the strong population structure observed. Such a pattern of diversity indicates multiple independent introduction events/migration waves: a single introduction, probably human-mediated, at the original outbreak site, and at least two waves representing a general shift in the distribution of D. septosporum as a consequence of natural spread, resulting in most of the local populations having similar diversity parameters.  相似文献

In this study, evolution of the genetic structure of the oldest known population of Dothistroma septosporum in Poland was analysed. Dothistroma needle blight was first recorded in Poland in 1990 in a 5-year-old plantation, meaning the population is relatively young in terms of evolution and genetic development. The preservation of DNA extracts from other studies allowed examination of the genetic variation of the pathogen over the span of a decade with the use of highly sensitive microsatellite markers. A number of evolutionary factors were identified that have shaped the genetic structure of this population over time. First, indications of the founder effect and the impact of selection were observed. Secondly, structure analysis provided evidence of the introduction of a new genetic component to the population structure after the pathogen had established. Finally, the impact of the reproductive mode on various components of the Domiarki population was examined, revealing the presence of genetic groups and structure clusters with varying levels of sexual recombination. These results demonstrate how well established and genetically robust a new population of a pathogen may become in just a few decades, given favourable conditions, and how new introductions may facilitate this process.  相似文献

Since 1911, dothistroma needle blight, caused by Dothistroma septosporum, has been recorded in most European countries. In the Czech Republic, the fungus has become an important disease of pines since 2000, especially Austrian pines in plantations of Christmas and ornamental trees. The aim of this study was to analyse the population structure, gene flow and mode of reproduction of this pathogen. Microsatellite and mating‐type markers were analysed in a Dothistroma population in the southeastern part of the country using reference isolates from other European countries. The haplotypic diversity was high, with 87 unique and 13 shared haplotypes (probable clones) identified in 121 samples. Based on structure analysis, the isolates were divided into two populations, with an uneven distribution over the sampling sites. The grouping of the sites to the populations did not follow a geographical pattern because certain isolates that were sympatrically co‐occurring at the same site were placed in different populations. Tests for random mating (the index of association and a parsimony tree‐length permutation test) showed a significant clonal mode of reproduction in most cases, but the intrapopulation haplotypic diversity is unexpectedly high. Although a teleomorphic stage of D. septosporum has not been previously observed in the Czech Republic, the high intrapopulation haplotypic diversity can be explained by infrequent sexual reproduction consistent with the occurrence of both mating types.  相似文献

Grosmannia alacris is a fungus commonly associated with root‐infesting bark beetles occurring on Pinus spp. The fungus has been recorded in South Africa, the USA, France, Portugal and Spain and importantly, has been associated with pine root diseases in South Africa and the USA. Nothing is known regarding the population genetics or origin of G. alacris, although its association with root‐infesting beetles native to Europe suggests that it is an invasive alien in South Africa. In this study, microsatellite markers together with newly developed mating type markers were used to characterize a total of 170 isolates of G. alacris from South Africa and the USA. The results showed that the genotypic diversity of the South African population of G. alacris was very high when compared to the USA populations. Two mating types were also present in South African isolates and the MAT1‐1/MAT1‐2 ratio did not differ from 1:1 (χ2 = 1·39, = 0·24). This suggests that sexual reproduction most probably occurs in the fungus in South Africa, although a sexual state has never been seen in nature. In contrast, the large collection of USA isolates harboured only a single mating type. The results suggest that multiple introductions, followed by random mating, have influenced the population structure in South Africa. In contrast, limited introductions of probably a single mating type (MAT1‐2) may best explain the clonality of USA populations.  相似文献

Dothistroma pini is one of two pathogens causing Dothistroma needle blight (DNB), a foliar disease of pines. The species was redefined in 2004 and subsequently recorded in several European countries. In Slovakia, the first report of the pathogen was in 2013. In this study, the population structure, genetic diversity, and reproductive mode of 105 isolates collected from 10 localities and seven hosts were determined in Slovakia. Species-specific mating type markers, ITS haplotype determination, and 16 microsatellite markers were used to characterize and genotype the isolates. Overall, 15 unique multilocus haplotypes (MLHs) based on microsatellite markers and three ITS haplotypes were identified. Three independent methods (DAPC, STRUCTURE, EDENetwork) separated the isolates into two distinct population clusters corresponding with ITS haplotypes. A high level of clonality was recorded suggesting that conidia are the primary source of pathogen dispersal. The low genetic diversity, predominantly asexual reproductive mode of the pathogen, and the fact that most isolates were collected from introduced tree species and native species in artificially planted urban greenery, supports the hypothesis that D. pini has been recently introduced into Slovakia.  相似文献

Lasiodiplodia theobromae is one of the most frequent fungal pathogens associated with dieback, gummosis, leaf spot, stem-end rot and fruit rot symptoms in cashew, mango, papaya and grapevine. In this study, the variation in the genetic diversity of 117 L. theobromae isolates from northeastern Brazil (= 100) and Mexico (= 17), which were collected from these four crops, was analysed using microsatellite markers. The results revealed low genetic diversity among L. theobromae populations and the existence of two genetic groups. All Mexican isolates were grouped with Brazilian isolates, suggesting a low level of differentiation between these populations. Furthermore, no evident host or climate-based population differentiation was observed for L. theobromae in Brazil. The populations studied were mostly clonal, but additional studies are needed to better understand the mode of reproduction of the pathogen. The low genetic diversity of L. theobromae populations in northeastern Brazil suggests that resistant cultivars could be used as a durable management strategy to reduce the impact of the diseases caused by this pathogen.  相似文献

Pitch canker on plantation-grown Pinus species, caused by the fungal pathogen Fusarium circinatum, first appeared in the western and southern Cape regions of South Africa. However, outbreaks have subsequently been reported from the major plantation growing regions of KwaZulu-Natal and Limpopo in the eastern, summer rainfall regions of the country. It is more than 10 years since the last detailed population genetics studies on F. circinatum in the region were conducted. To shed light on the population biology of F. circinatum in this region of South Africa, we used microsatellite markers and mating-type assays to study a collection of 296 isolates from different nurseries and plantation sites. Our results showed that populations in the region are highly diverse, but strongly interconnected, with various genotypes shared across nursery and plantation collection sites. In contrast to nursery populations, those associated with pitch canker outbreaks were characterized by the presence of relatively small numbers of dominant genotypes that were generally widespread across the region. Opposite mating-type individuals occurred in most of the isolate collections, but multilocus linkage disequilibrium analyses pointed towards clonality being the main reproductive mode of F. circinatum in the region. Most of the pathogen's genetic variation could probably have resulted from multiple different introductions into the country and more specifically, into the summer rainfall region. Because the spread and establishment of invasive pathogens are typically driven by aggressive clones, the results of this study provide important considerations for current and future Pinus disease management strategies.  相似文献

Brown spot needle blight (BSNB), a disease of pine trees caused by the fungus Lecanosticta acicola, has been known in Slovenia since 2008 and in Croatia since 1975. Recent outbreaks in Slovenia prompted this study to compare L. acicola populations in these two neighbouring European countries. Sixty-nine isolates collected from three pine species (Pinus mugo, P. halepensis and P. nigra) were used to determine the phylogenetic relationships, genetic structure, and reproductive strategy of the pathogen. EF1-α sequences showed that Slovenian and Croatian isolates share a common ancestry with individuals from central and northern Europe. Population structure analysis revealed four distinct population clusters of L. acicola in these two countries, generally corresponding to their respective geographic location and host. An unequal ratio of mating types and a low overall genetic diversity in the population indicated a strong influence of asexual reproduction. Although some of the oldest recorded European occurrences of BSNB are from Croatia, this study provided no evidence that the population studied in Croatia was the source of the sampled outbreaks in Slovenia. Recent outbreaks of L. acicola in Slovenia are most likely due to introductions from other, yet to be identified, sources.  相似文献

Many riparian(Tugai) forests growing along rivers in arid and hyper-arid regions of Central Asia are dominated by the Euphrates poplar(Populus euphratica). Besides generative reproduction, which is only possible upon flooding events and at a distance to the groundwater of less than 2 m, this phreatophytic tree species also reproduces vegetatively by forming clones that can cover land surface areas of several hectares. Along a gradient of groundwater distances, we investigated whether the fraction of clones in P. euphratica stands(1) increases with increasing distance to the water table;(2) is higher if supplied with water via river cut-offs; and(3) approaches 100% at a short distance to the groundwater, but at high salt concentrations in the upper soil layers, which would prevent germination and establishment of seedlings. AFLP(Amplified Fragment Length Polymorphism) analyses were conducted on leaf samples taken from mature P. euphratica trees growing at the fringes of the Taklimakan Desert in stands with different distances(2–12 m) to the groundwater at two plots at the middle and the lower reaches of the Tarim River and in a stand close to Ebinur Lake, Xinjiang, China. Genetic diversity was large among plots, but considerably smaller within plots. We found the highest genetic diversity(caused by regeneration from seeds) at plots that have a short distance to the groundwater or are supplied with additional water. There was no significant relationship between groundwater distance and clonal fraction. All investigated trees at the saline Ebinur Lake site belonged to one single clone. Our results demonstrate that the genetic pattern of this widespread species is not easily predictable even over small distances as it is a result of a complex interplay of stand history and dispersal of propagules(pollen, seeds, and vegetative diaspores) by wind and water. In conservation and restoration schemes, P. euphratica stands with a high genetic diversity and stands that grow at short distances to the water table and are regularly subjected to flooding(which favors generative over clonal reproduction) should be prioritized.  相似文献

In a survey of Scottish potato late blight ( Phytophthora infestans ) populations from 1995 to 1997, nearly 500 isolates were collected from over 80 disease outbreaks in commercial potato crops and gardens/allotments. The isolates were characterized by mating type, resistance to the fungicide metalaxyl and almost 300 were examined by DNA-based AFLP fingerprinting. These data were examined alongside cropping details to determine the population structure in the context of existing disease management strategies. A1 and A2 mating type isolates were present in both commercial potato crops and gardens or allotments although they coexisted more frequently in the latter sites. One-fifth of the isolates collected were of the A2 mating type and the frequency was similar over the 3 years and amongst sites. In 1995 the proportions of isolates that were sensitive and resistant to metalaxyl were equal (∼40%) but, over the following 2 years, the frequency of resistant isolates decreased and that of intermediate isolates increased. The mating type response to metalaxyl differed markedly, with 52% of A1 and only 5% of A2 isolates being resistant. Considerable molecular diversity was observed, with over half of the isolates having unique AFLP patterns. Analysis of the molecular and phenotypic data revealed a broad clustering of the population into three groups. Many factors point to an A2 population restricted by its sensitivity to phenylamides. The majority of the A2 isolates were found in a single AFLP group, but the presence of mixed mating type samples, an increasing frequency of isolates of intermediate metalaxyl resistance and the extent of the AFLP diversity suggest occasional sexual recombination, and thus gene flow, between groups.  相似文献

Phytophthora pluvialis is the causal agent of red needle cast on Pinus radiata in New Zealand. It was first isolated in 2008 but had previously been recovered from tanoak (Notholithocarpus densiflorus) and Douglas‐fir (Pseudotsuga menziesii) trees in Oregon, USA in 2002. Phytophthora pluvialis was subsequently described as a new species in 2013 and classified as a clade III Phytophthora species. The aim of this study was to gain a better understanding of the genetic diversity, population structure and origin of this pathogen. A total of 360 P. pluvialis isolates were collected from the USA and New Zealand. The genome sequences of two P. pluvialis isolates were used to identify 27 single nucleotide polymorphism (SNP) markers that were then used to genotype the two populations. The genotypic data showed that the USA population of P. pluvialis had twice the genetic diversity and a greater number of multilocus genotypes (MLGs) compared to the New Zealand population, with 126 and 24 MLGs, respectively. The majority of the subpopulations within the USA and New Zealand showed linkage disequilibrium. All subpopulations had a negative fixation index, indicating that clonal reproduction is prevalent in both countries. A minimum spanning network (MSN) showed two unique clusters of isolates in the New Zealand population, suggesting two potential introductions of P. pluvialis into New Zealand from the USA. There was no significant structure within the New Zealand or USA populations. This study provides novel insight into the genetic structure of P. pluvialis in New Zealand and the USA.  相似文献

The aggressive Eucalyptus leaf pathogen, Teratosphaeria destructans, causes widespread damage in tropical and subtropical Eucalyptus-growing regions of Indonesia, China, Thailand, East Timor, Vietnam, Lao, and South Africa. Little is known regarding the origin, pathways of dispersal, or reproductive biology of this pathogen. The aim of this study was to investigate the genetic structure of a global collection of T. destructans isolates. This was achieved by developing and using polymorphic microsatellite markers. Low genotypic diversity and a limited number of private alleles were found in all investigated populations, with the highest maximum diversity of 10.7% in isolates from South Sumatra. This supports the hypothesis that T. destructans was introduced to these regions. High levels of clonality were common in all populations, especially in isolates sampled from the recent disease outbreak in South Africa, which were all identical. The global collection of isolates grouped into three distinct clusters, corresponding largely to their sampled regions. Low levels of genotypic diversity, high levels of clonality, and strong geographic structure suggest independent introductions into all the sampled areas from an unknown source. The results imply that strict biosecurity measures are needed to avoid introductions of additional genotypes in these areas.  相似文献

Dothistromin is a broad‐spectrum mycotoxin produced by the Dothideomycete pine needle pathogen Dothistroma septosporum. It accumulates in lesions, causing characteristic red bands on needles infected with this fungus. Dothistromin is similar in structure to the aflatoxin precursor versicolorin B and the biosynthetic pathways of these toxins share many common gene products. Although dothistromin is not essential for pathogenicity in dothistroma needle blight, its presence in infected needles suggests it might have a role in the disease process. The hypothesis that dothistromin is a virulence factor was tested by studying Pinus radiata infected with dothistromin‐deficient mutants of D. septosporum. The mutants were able to infect pine needles and complete their life cycle as previously shown, and were unaffected in spore germination, epiphytic growth or needle penetration. However, colonization of the mesophyll by the mutants was restricted compared to the wild type. Correspondingly, lesions produced by the mutants were smaller and produced significantly fewer spores than lesions produced by wildtype strains. Interestingly, ‘green islands’, in which chlorophyll was maintained at a higher level than in adjacent chlorotic and necrotic regions, surrounded early‐appearing lesions caused by both wildtype and mutant strains. At a later stage of disease development green islands were still present in the mutant but appeared to be masked by the extended dothistromin‐containing lesions in the wild type. Overall the results support a role for dothistromin in virulence in dothistroma needle blight.  相似文献

Sheath blight, caused by Rhizoctonia solani AG1‐IA, is one of the most serious diseases of rice. In this study, a total of 175 isolates of R. solani AG1‐IA were collected from five rice‐growing regions in China. Pathogenicity tests revealed that all isolates were virulent to five cultivars with different levels of resistance at the rice seedling stage in the greenhouse. There was considerable variation in aggressiveness, and the isolates were classified into three pathotypes based on disease severity, with moderately virulent isolates prevalent in the population. Forty‐three haplotypes were identified based on ITS sequencing, and 39 haplotypes were distinct among isolates. There were high levels of haplotype diversity and nucleotide diversity within the populations of Rsolani AG1‐IA. High gene flow (Nm = 1·63–5·22) was detected, consistent with relatively low differentiation between pairs of populations. Five populations were divided into two distinct clusters by the unweighted pair group method with arithmetic mean (UPGMA), and no spatial population differentiation was discernible. The majority (97·8%) of genetic diversity was distributed among isolates within populations, with only 2·2% of the genetic diversity attributed to differences among populations. The star‐like shape of the haplotype network provided evidence of signatures of population expansion in recent history. No significant relationships were found between the genetic diversity and aggressiveness or geographic origin among populations of R. solani AG1‐IA. These results highlight that the population characteristics of R. solani AG1‐IA should be taken into account in evaluating the germplasm resistance of rice cultivars to sheath blight.  相似文献

A genomic library was used to develop seven SSR markers for studying the population genetics of Alternaria solani, a pathogenic fungus causing early blight disease of potato and tomato worldwide. Population genetic analysis of 268 isolates of A. solani sampled from four locations, each representing one of four potato production systems in China, indicates that these SSR markers are moderately diverse, selectively neutral and possibly unlinked. Population genetic analysis also indicated that genetic variation of A. solani in China is high. About 2/3 of 123 genotypes were detected only once and genotype diversity measured by the standardized Shannon index ranged between 0·82 and 0·92 in the populations. Although clones were detected in multiple populations separated by thousands of kilometres, random association among SSR loci was found in half of the populations assayed. On average, nearly six copies of genetic material were exchanged among these populations each generation and no isolation by distance was detected. It is hypothesized that the joint effects of cryptic sexual reproduction and human‐mediated gene flow may account for the observed population genetic structure of A. solani in China.  相似文献

Phytophthora infestans populations can differ in composition as a result of host specialization on tomato and potato hosts. In Great Britain many amateur gardeners grow outdoor tomatoes but there is little or no commercial tomato production outdoors. This study analysed isolates of P. infestans from British gardens with 12 multiplexed simple sequence repeat markers that are used to monitor the disease on commercial potato crops. Samples of P. infestans from tomato hosts were collected in 3 years and from potato in 1 year from across Great Britain. Seven previously unreported clonal lineages were detected in garden populations and higher frequencies of unique clonal lineages (28–40%) were present compared with populations from British commercial potato crops reported elsewhere. Garden populations had a lower proportion (11–48% less) of the most common lineages (13_A2 and 6_A1) that together made up at least 86% of the commercial potato populations during the sampling period. Host species accounted for only 2·0% of molecular variance detected between garden potato‐ and tomato‐hosted samples. No significant difference in clonal lineage composition was found between host species in Great Britain and this could be due to the whole P. infestans population overwintering on potato. British garden populations on both hosts were much more diverse than those on commercial potato crops; this finding may be influenced by less frequent fungicide use by gardeners and a higher diversity of unsprayed susceptible potato cultivars, enabling metalaxyl‐sensitive and less aggressive genotypes to survive in gardens.  相似文献
