Disease incidence and severity was studied for winter wheat variety Bezostaya 1 and susceptible checks based on data from international nurseries from 1969 to 2010 and from 51 countries across major winter wheat production regions totalling 1,047 reports. The frequency of leaf rust and stripe rust occurrence was stable over time with marked increases in severity in 2001–2010 especially in Europe and Central and West Asia. Substantial global reductions in stem rust occurrence were recorded and attributed primarily to use of resistance genes although the recent emergence of race Ug99 makes wheat more vulnerable. The occurrence of powdery mildew remained globally stable over time. It was the most important foliar disease in Western and Southern Europe, where the frequency was very high for all time periods coupled with slight increases in severity during 2001–2010. The durable resistance of variety Bezostaya 1 to all four diseases was demonstrated in the study using comparisons of disease severities of Bezostaya 1 and the most susceptible entries. The Lr34/Yr18/Pm38 pleiotrophic set possessed by Bezostaya 1 is currently an important target for selection because it is now amenable to molecular selection. Increased use of genes like Lr34 combined with strategies to minimize cultivation of extremely susceptible varieties will contribute to long term maintenance of low and non-damaging disease levels. The durable disease resistance of Bezostaya 1, combined with its adaptability and good end-use quality, was a significant reason for its huge impact in agriculture over the last 50?years.  相似文献

Push–pull involves intercropping of cereals with Desmodium as a “push” crop and planting Napier grass/Brachiaria as the “pull” crop at the border. The technology has been reported to effectively control stemborers, striga weed, and fall armyworm (FAW), and to improve soil nutrition, resulting in increased grain yield. This study evaluated the impact of stemborer and FAW management using this technology on incidence of maize ear rots and preharvest contamination of grains with aflatoxin and fumonisin in western Kenya. The study was conducted during three cropping seasons on maize grown under the push–pull system and as a monocrop. Incidence of stemborer and FAW damage was determined as percentage of damaged plants, while incidence of ear rots was determined as percentage of ears with symptoms. At harvest, fungi were isolated from kernels and aflatoxin and fumonisin were quantified using enzyme-linked immunosorbent assay. Stemborer and FAW damage was significantly (p = .001) reduced by over 50% under the push–pull system. There was also a significant (p < .001) reduction in the incidence of Fusarium verticillioides (60%) and Aspergillus flavus (86%), which was reflected in a reduced incidence of ear rots (50%) with the push–pull system (p = .001). Fumonisin in maize from push-pull farms was significantly (p = .048) reduced (39%) but the technology had no significant (> .05) effect on aflatoxin. The study showed that push–pull is an effective strategy for managing maize ear rots and fumonisins, and therefore could play a role in improving food safety among smallholder maize farmers in the region.  相似文献

A study was undertaken to examine the main source of inoculum of Bipolaris sorokiniana responsible for its reappearance in rice–wheat cropping regions of eastern India. Soil samples were collected at monthly intervals during April–October in the years 2000 and 2001 from fields having rice–wheat cropping. Bipolaris sorokiniana conidia were isolated and their viability was found to decline sharply with the onset of flooding in the month of August. In contrast to 82% in April, viability was 4% and <1% in August and September, respectively. Viable conidia were multiplied in the laboratory and inoculated on to susceptible cv. Sonalika under controlled conditions for test of pathogenicity. Appearance of symptoms typical to spot blotch were recorded after 7 days. Twenty-two different species (weeds and grasses) normally found to be associated with rice–wheat fields were tested for the presence of B. sorokiniana to evaluate their possible role as alternative hosts. Only three species, i.e. Setaria glauca, Echinochloa colonum and Pennisetum typhoids, were found to naturally harbour B. sorokiniana. Isolates from these hosts were tested for pathogenicity and also for their possible spread to wheat. When reisolated from these hosts, the pathogen did not infect wheat. Seeds of 25 different wheat genotypes were tested for B. sorokiniana infection. All genotypes were infected and the incidence of infection varied from 26% to 86%. Five isolates of wheat and one isolate from each of the three species (S. glauca, E. colonum and P. typhoids) were subjected to RAPD analysis. Two broad clusters were formed, suggesting that the wheat isolates were different from the isolates originating from other hosts. The results indicate that seeds are the most important source of inoculum for the reappearance of spot blotch of wheat in rice-wheat cropping systems in eastern India.  相似文献

Samara is the reproductive organ (seed) for many tree species in arid land in northwestern China. It is ecologically important in population development due to its dispersal function. However, information on its photosynthesis and effect of environmental stresses on its photosynthesis is still very limited. In the present study, responses of photosystem II (PSII) activity in samara and leaf of Siberian maple to short-term chilling/freezing and subsequent recovery potential were comparatively investigated by using polyphasic fluorescence test. The samara had more efficient photosynthesis (Fv/Fm and PIABS) and more efficient electron transport (φEo) but lower energy dissipation (DIo/RC) than leaf. Generally, the PSII performance and the electron transport for both samara and leaf were inhibited under low temperature stress, accompanied by an increase of energy dissipation in PSII reaction centers (RCs). PSII of both samara and leaf was not markedly affected by chilling and could acclimate to chilling stress. Short-term freezing could completely inhibit PSII activity in both samara and leaf, indicated by the drop of values of Fv/Fm, PIABS, φEo to zero. PSII functional parameters of short-term dark frozen samara could be largely recovered whereas those of frozen leaf could not be recovered. The higher tolerance of samara to short-term low temperature stress than leaf is of great ecological significance for seed development, population establishment of Siberian maple.  相似文献

A mitogen-activated protein kinase (MAPK) pathway has been demonstrated as a key pathway in plant defense against pathogen attacks. With proteomics approaches, we specifically studied activation events downstream of a MAPK kinase, tMEK2, in tomato. Overexpression of a constitutively activated tomato MAPK kinase gene (tMEK2MUT) enhanced resistance of transgenic tomato lines to the virulent bacterial pathogen Pseudomonas syringae pv. tomato. Pathogenesis-related genes, PR1b1, β-1,3-glucanase, and endochitinase were up-regulated by tMEK2MUT. Two-dimensional electrophoresis and matrix-assisted laser desorption/ionisation-time-of-flight-mass spectrometry analysis of total soluble leaf proteins indicated that β-1,3-glucanase and endochitinase are among the up-regulated proteins in these transgenic plants. Co-expression studies using a transient gene expression system have indicated that β-1,3-glucanase and endochitinase genes up-regulated by tMEK2MUT were down-regulated by different specific phosphatases through dephosphorylation of certain downstream signaling molecules. Our observations indicate that increased products of β-1,3-glucanase and endochitinase genes downstream of tMEK2 may play an important role in achieving disease resistance.  相似文献

Proximal remote sensing is being widely studied as a noninvasive method to partially automate diagnostics of plants and insects. The hypothesis that proximal remote sensing can be used to differentiate specimens of adult beet leafhoppers (Circulifer tenellus) that were nonviruliferous or viruliferous for beet curly top virus (BCTV) was tested. A key aspect of applications of proximal remote sensing is the ‘robustness’ or repeatability of input reflectance data. Many factors may contribute to low input reflectance data robustness; these include: (i) issues related to the consistency of proximal remote sensing conditions (light intensity and spectral composition, ambient temperature), (ii) insect specimen preparation (projection angle, storage and handling), and (iii) insect specimen characteristics (age, growing conditions, variety/biotype, host plant). This study demonstrates that nonviruliferous and viruliferous specimens of adult beet leafhoppers possess unique body reflectance features and, therefore, can be differentiated. However, insect specimen preparation (removal of wings and placement) markedly affected the classification accuracy. Addition of experimental noise to input reflectance data was conducted to simulate varying degrees of input reflectance data robustness. The potential of developing reflectance-based diagnostic tools for detection of plant pathogenic viruses in insects is discussed, with an emphasis on input data robustness.  相似文献

A total of 80 single–lesion isolates of Phytophthora infestans were collected from tomatoes and potatoes in several locations in Chiang Mai and Tak provinces in 2000–2002. These isolates were analyzed for mating type, metalaxyl sensitivity, mitochondrial DNA (mtDNA) haplotype RFLP pattern as determined by probe RG57, and for microsatellite markers. All isolates were A1 mating type. Isolates from tomato were usually sensitive to metalaxyl, but isolates from potato were usually resistant to metalaxyl. With one exception, all tomato isolates were related to the US-1 clonal lineage. With two exceptions, all potato isolates were related to two European lineages. In these two provinces, the populations of P. infestans on tomatoes are clearly different from those on potatoes.  相似文献

Rhyzopertha dominica causes extensive damage to stored wheat grains. α-Amylase, the major insect digestive enzyme, can be an attractive candidate to control the insect damage by inhibiting the enzyme through α-amylase inhibitors. R. dominica α-amylase (RDA) was purified to homogeneity by differential ammonium sulphate fractionation, Sephadex G-25 and Sephadex G-100 column chromatography. The homogenous α-amylase has a molecular weight of 52 kDa. The pH optima was 7.0 and temperature optima was 40 °C. Activation energy of RDA was 3.9 Kcal mol−1. The enzyme showed high activity with starch, amylose and amylopectin whereas dextrins were the poor substrates. The purified enzyme was identified to be α-amylase on the basis of products formed from starch. The enzyme showed Km of 0.98 mg ml−1 for starch as a substrate. Citric acid, oxalic acid, salicylic acid, HgCl2, tannic acid and α-amylase inhibitors from wheat were inhibitors whereas; Ca2+ and Mg2+ were the activators of RDA. Ki values calculated from Dixon graphs with salicylic acid, citric acid, oxalic acid and wheat α-amylase inhibitors were 6.9, 2.6-8.2, 3.2 mM and 0.013-0.018 μM, respectively. The Lineweaver-Burk plots with different inhibitors showed mixed type inhibition. Wheat α-amylase inhibitor showed mainly competitive inhibition with some non-competitive behaviour and other inhibitors showed mainly non-competitive inhibition with some un-competitive behaviour. Feeding trials with salicylic acid, citric acid, oxalic acid and wheat α-amylase inhibitor showed significant effect of salicylic acid and oxalic acid along with wheat α-amylase inhibitor in controlling the multiplication of R. dominica.  相似文献

Recently, Elsinoë piri has become an important pathogen in unsprayed and organic apple orchards. The increasing demand for organic apples as well as the limited use of fungicides imposed by European legislation has turned this pathogen, which has been known for over a century in Europe, into a resurgent phytosanitary threat. It also represents a new trait to be taken into account in apple breeding programmes. As E. piri is extremely difficult to isolate and causes symptoms that can be confused with those caused by other pathogens, a quantitative PCR (qPCR) test has been developed based on the internal transcribed spacer (ITS) region of the rDNA gene. This test, which displays a low limit of detection, allows the early detection of the fungus in apple leaves. As a quantitative method, it is a promising tool for breeding purposes as it can be used on leaves to assess the level of quantitative resistance of apple cultivars to the disease. The molecular test is also useful to gain knowledge on the epidemiology of this re-emerging fungal disease. Spore trapping conducted in 2015, 2016 and 2020 using Rotorod devices and the qPCR test showed that airborne inoculum is released at the end of the growing season. Tests were also carried out to demonstrate that the pathogen was detected in buds during winter. A comparison of 18 E. piri isolates based on the sequencing of five genome regions highlighted a high genotypic diversity. All these isolates were detected with the qPCR test developed in this study.  相似文献

Induction of the microsomal oxidase system by dietary phenobarbital and β-naphthoflavone was examined in three blowflies, Phormia regina (Mg.), Lucilia illustris (Mg.), and Eucalliphora lilica (Walk.). Responses were similar in adults and larvae of all species. Phenobarbital increased cytochrome P-450 levels up to 9-fold and aldrin epoxidase up to 138-fold. Increases in cytochrome P-450 and aldrin epoxidase caused by β-naphthoflavone were minor relative to those produced by phenobarbital. In toxicity experiments with carbaryl and propoxur tolerance was associated with the amount of microsomal oxidase activity. Using piperonyl butoxide to synergize carbaryl and propoxur there was no clear indication for the use of either the synergist ratio or synergist difference as an indicator of microsomal oxidase activity.  相似文献

Semi-thin sectioning and transmission electron microscope techniques were employed to investigate the cuticle thickness, integument structure, and fat body of larvae from susceptible and resistant strains of Bactrocera dorsalis. The results showed that the cuticle of β-cypermethrin-resistant strains (25.96 ± 1.00 μm) was thicker than that of susceptible strains (19.36 ± 0.82 μm). The number of chitin layers in the endocuticle of β-cypermethrin-resistant strains (98.00 ± 3.61 layers) was more than that in susceptible strains (75.67 ± 2.40 layers). Compared with susceptible strains, the laminated structure of the chitin layers in the endocuticle of resistant strains revealed higher density and more distinctive structure, and the interspace of epidermal cells was thicker. Fat body in the resistant insects contained more fat granules than those in susceptible insects. Moreover, HPLC analysis showed that the cuticular penetration of β-cypermethrin into larvae of resistant strains was slower than that of susceptible strains. In addition, the metabolism of β-cypermethrin in resistant strains was faster than that in susceptible strains, indicating that the resistant strains could enhance detoxification metabolism. These results indicated that cuticle thickness, fat body, laminated structure of the chitin layers, and interspace of epidermal cells might be correlated with cuticular penetration between susceptible and resistant strains, suggesting that the resistant strains could decrease the rate of penetration of insecticide into the internal cavity.  相似文献

Abstract

In hybrid sorghum ‘CSH-1’ severe infestation of the earhead caterpillars Cydia sp., Ectomyelois sp., Eublemma sp. nr. gayneri Roths., Heliothis armigera Hub., and Euproctis limbata Wlk, was recorded at Jabalpur (Madhya Pradesh) during 1968. The first two were relatively more serious. The loss in grain yield was estimated to be 18.26 per cent, i.e. about 717 kg/ha.

Among different insecticides tested, carbaryl 0.2 per cent spray proved most effective, followed by endosulfan 0.05 per cent spray. Dichlorvos 0.05 per cent gave good initial kill, but showed little persistent effect.  相似文献

Sixty-four isolates of Bean yellow mosaic virus (BYMV) from cultivated and naturalized gladioli were divided into two pathogenic groups, necrotic spot (NS) and chlorotic spot (CS) groups on Chenopodium quinoa. NS-type isolates (S-22N and E-24N), CS-type isolates (S-22C and E-92C), and broad bean isolates (Sb-50C and Sb-12C) differed in their pathogenicity on Antirrhinum majus, Nicotiana benthamiana, Phaseolus vulgaris, Spinacia oleracea and Vigna unguiculata. The four gladiolus isolates were different from BYMV-B, -P, -O and C1YVV-N in their pathogenicity on these plants, while the two broad bean isolates were similar to BYMV-B, originally from broad bean. The nucleotide (nt) sequences of the 3′-terminal region of the BYMV RNA genome of the two NS-type isolates, the two CS-type isolates, the two broad bean isolates and BYMV-B, -P and -O were determined. In a phylogenetic tree based on the CP amino acid (aa) sequences, the two NS-type isolates clustered together (identity 98.4% and 98.2% at the nt and aa level, respectively). The two CS-type isolates clustered with BYMV-O (93.2 to 99.3% nt identity and 95.6 to 98.5% aa identity). The two broad bean isolates clustered with BYMV-B (99.0 to 99.5% nt identity and 98.9 to 99.6% aa identity). BYMV-P clustered with BYMV-CS (identity 97.7% and 99.3% at the nt and aa level, respectively). The obtained sequences were compared with those of the 3′-terminal regions of seven published BYMV isolates. Lolium isolate TP2 of Pyricularia oryzae, causal agent of gray leaf spot of perennial ryegrass (Lolium perenne), is virulent on perennial ryegrass, but avirulent on wheat cultivars. Genetic analysis of wheat F2 populations revealed that the resistance of wheat cultivars Chinese Spring, Shin-chunaga, and Norin 4 to TP2 was conditioned by two genes, R1 and R2. R1 was highly effective, while R2 was less effective. The strong resistance gene R1, designated Rmg6, was mapped on chromosome 1D using microsatellite markers. For revealing genetic mechanisms of avirulence, TP2 was crossed with Triticum isolate Br48. Segregation analysis of their F1 progenies revealed that the avirulence of TP2 on the three wheat cultivars was conditioned by two unlinked genes, one (A1) highly effective and the other (A2) less effective. These results suggest that the incompatibility between TP2 and the common wheat cultivars is conditioned by two gene pairs; the Rmg6A1 interaction results in strong resistance, and the R2A2 interaction results in moderate resistance.  相似文献

The northeastern coast of the Caspian Sea is one of the youngest territories for plant colonization in Kazakhstan,and the flora of the most recent portion of this coastal area,the New Caspian marine plain,was chosen to study phytogeographical characteristics of plant species,especially as related to alien origin and invasive potential.Because of the recent formation of this flora,I expected that a large proportion of the species may be alien and invasive.After compiling a comprehensive species list from previous research,I identified the taxonomic,life form,ecological,and geographical structures of the flora.The area belongs to the northern type of deserts with continental climate and low annual precipitation.Taxonomic and life form structures show that the flora is specific to Central Asian deserts.Ecological structure demonstrates a high percentage of halophytes,which resulted from their adaptation to soil salinity and shallow ground waters that occur throughout the study area.Geoelements of the flora indicate allochtonous properties of the flora(i.e.species derived from areas outside the study area) with a low proportion of native(Caspian) plants.Alien species are estimated to comprise 12%-19% of the flora.Although some species have invasive features,severe arid environmental conditions and high salinity of soils and ground waters decrease invasive capacity of exotic plants.However,a threat of expansion of potentially invasive plants exists in the New Caspian marine plain due to the fast growth of anthropogenic habitats associated with recent oil developments.  相似文献
