1. The effects of spraying linseed oil on naturally clean eggs, subsequently stored for 24 d at 28 °C were studied. 2. Oiling slowed the decline in albumen height (Haugh units) to that which occurred in unoiled eggs stored at 12 °C. 3. Oiling slowed the increase in albumen pH equally at 28 and 12 °C. 4. Oiling markedly decreased the weight loss of eggs stored at either temperature. 5. 1. A study was undertaken into the storage of turkey semen in vitro for 24 h. 2. Turkey semen storage for 24 h at 10 °C was not noticeably improved by using a buffered diluent compared with an unbuffered (pH 7.3) diluent. This is different from the case of the fowl and probably reflects differences in the metabolism of spermatozoa between the species. 3. Adverse effects on the fertilising ability of turkey spermatozoa were observed with diluents buffered around pH 7.4 and 5.8 when compared with the use of diluent either buffered at pH 7.1 or unbuffered. 相似文献

1. The aim of this study was to investigate the stability of immunoglobulin-Y (IgY) in stored eggs from immunised hens. 2. Eggs from individual hens were randomised and stored for up to one month at room temperature, or for up to 6 months at +4°C. IgY was extracted from the egg yolks and the antibody activities were tested by ELISA. 3. There was no significant reduction in antibody titres with egg storage under these conditions. 4. Egg yolks of immunised chickens provide an inexpensive source of large amounts of polyclonal antibodies for use in immunotherapy and immunoassays. By collecting eggs from different immunised hens and pooling their yolks, it should be possible to reduce batch-to-batch variation. 相似文献

1. Fowl and turkey spermatozoa were found to utilise oxygen when stored in tubes at 5 and 10 °G respectively; the storage medium becoming rapidly anaerobic. 2. This aerobic metabolism was encouraged by continuous introduction of air into the semen during storage. 3. Fertility of turkey semen stored at 10 °C for 24 h in a diluent containing glucose was increased 24‐fold by simple aeration and reached 99% of that achieved with fresh semen. 4. Fertilities of greater than 90% were also achieved with fowl semen stored aerated for 48 h at 5 °C in a diluent with or without glucose. Background: Hypothermic preservation of boar semen is considered a potential method for omitting antibiotics from insemination doses, thereby contributing to the global antibiotic resistance defence strategy. The main challenges are chilling injury to spermatozoa and bacterial growth during semen storage leading to reduced fertility.Objectives: To examine chilling injury and the number and type of bacteria in boar semen stored at 5 ℃ in the absence of antibiotics, and to assess the applicability of hypothermic semen storage under field conditions.Material and methods: Boar ejaculates were extended with AndroStar~? Premium, stored at 17 ℃ with and at 5 ℃ without antibiotics and tested for functional sperm parameters by flow cytometry. Raw semen and extended samples were investigated bacteriologically. Fertility was evaluated after once-daily inseminations of 194 sows in a field study.Results: Lethal sperm damage assessed by motility and membrane integrity was low throughout storage in both experimental groups. Sublethal chilling effects based on the decrease of viable spermatozoa with low membrane fluidity were higher(P < 0.05) up until 72 h in sperm stored at 5 ℃ compared to 17 ℃ but did not differ after 144 h.After 72 h, incubation in capacitating medium for 60 min induced a similar decrease in viable sperm with high mitochondria membrane potential and low cytosolic calcium in both groups. In semen stored at 5 ℃, bacteria counts were below 103 CFU/mL and the bacteria spectrum was similar to that of raw semen. In 88% of 34 boars,cooled semen fulfilled the requirements for insemination. Fertility was high and did not differ(P > 0.05) between sow groups inseminated with semen stored antibiotic-free at 5 ℃ and semen stored at 17 ℃ with antibiotics.Conclusion: Despite subtle chilling effects and low bacterial numbers, antibiotic-free hypothermic storage of boar semen offers the possibility to reduce the use of antibiotics in pig insemination. However, strict sanitary guidelines must be maintained and further evidence of efficiency under field conditions is considered desirable. 相似文献

ObjectiveTo assess the cardiopulmonary effects caused by reverse Trendelenburg position (RTP) at 5° and 10° in sevoflurane-anesthetized yearling steers.Study designProspective, experimental study.AnimalsEight Holstein steers aged (mean ± standard deviation) 12 ± 2 months and weighing 145 ± 26 kg.MethodsIn the first phase of the study, the individual minimum alveolar concentration (MAC) of sevoflurane was determined using electrical stimulation. In the second phase, the effects of RTP were assessed. The animals were anesthetized on three separate events separated by ≥7 days in an incomplete crossover design: control treatment using a table without tilt (RTP0); treatment with the table at 5° RTP (RTP5) and table tilted 10° RTP (RTP10). Subjects were physically restrained in dorsal recumbency on the table, which was already tilted according to each treatment. Anesthesia was induced with sevoflurane at 8% in 5 L minute–1 oxygen via face mask followed by maintenance with sevoflurane at 1.3 MAC and spontaneous breathing. Cardiopulmonary variables were obtained immediately after instrumentation (T0) and then after 30, 60, 120 and 180 minutes (T30, T60, T120 and T180, respectively).ResultsThe mean sevoflurane MAC for the eight steers was 2.12 ± 0.31%. Cardiac output was lower at all time points and the systemic vascular resistance index was higher at T120 and T180 in RTP10 compared with RTP0. Oxygen consumption was lower at T0 and at T180 in RTP10 compared with RTP0 and at all time points except T30 compared with RTP5. Oxygen extraction was lower at T0 in RTP10 compared with RTP0 and RTP5, and at T60 and T180 compared with RTP5.Conclusions and clinical relevanceRTP 5° and 10° did not improve ventilatory and oxygenation variables in sevoflurane-anesthetized steers when compared with no tilt, however the cardiovascular variables were adversely affected in RTP10. 相似文献

1. A comparison was made of” oven ready “ duck carcasses stored at 2 or — 1 °C and wrapped in either a low‐density oxygen‐permeable polyethylene film or a heat‐shrunk oxygen‐impermeable film. 2. At spoilage the main organisms at 2 and — 1 °C on the carcasses wrapped in the oxygen‐permeable film were pseudomonads, producing unacceptable “off odours” when their numbers were >108/cm2. This occurred in about 10 d at 2 °C but in about 19 d at ‐ 1° C. 3. The effect of wrapping in the heat‐shrunk oxygen‐impermeable film was to delay or inhibit the growth of pseudomonads and thus extend the shelf‐life by more than 50% at either temperature. The predominant organisms isolated from the spoiling carcasses were atypical lactobacilli and enterobacteria. 4. Sensory assessment of the carcasses stored at — 1 °C by a trained panel indicated that, although less obvious “ off odours “ were produced by the micro‐organisms growing on the carcasses wrapped in the impermeable film, differences were detected at 33 d when the numbers of bacteria reached about 107/cm2 whilst at 41 d the meat was described as rancid. 相似文献

Cauda epididymal spermatozoa were obtained from testicles collected from abattoir(s). The pooled sperm samples were divided into four aliquots. Each aliquot was washed separately with the buffer of respective extender and finally extended with the four extenders viz. egg yolk–citrate (EYC), egg yolk–citrate–fructose (EYCF), Tris–citric acid–egg yolk–fructose (TCEYF) and egg yolk–Mcillvaine glucose (EYMG) and preserved at 4°C. The per cent sperm motility for EYC, EYCF, TCEYF and EYMG at 0 h was 50.83%, 56.67%, 75.00% and 31.67%, respectively, and at 72 h was 24.17% (EYC), 30.83% (EYCF), 51.67% (TCEYF) and 7.50% (EYMG). The corresponding figures for live sperm count at 0 h was 83.17%, 86.33%, 90.42% and 81.75% and at 72 h was 64.75%, 73.92%, 76.00% and 57.67%. The corresponding figures for mean per cent intact acrosome at 0 h was 95.33%, 95.50%, 90.92% and 97.25% and at 72 h was 86.17%, 83.92%, 77.58% and 86.33%. The sperm motility was significantly (p < 0.05) higher for TCEYF at different h of preservation from 0 h through 72 h. The sperm motility, live sperm count and per cent intact acrosome declined significantly (p < 0.05) with the advancement of storage time in all the four extenders. Our study concluded that TCEYF was best out of the extenders studied for preservation of cauda epididymal spermatozoa after double centrifugation and extension at 4°C up to 72 h of preservation. However, EYCF also has better potential for the preservation of cauda epididymal spermatozoa as viability was in close proximity and acrosomal integrity was higher compared with TCEYF extender. 相似文献

Background: The time from sampling to analysis can be delayed when blood samples are shipped to distant reference laboratories or when analysis cannot be readily performed. Objective: The objective of this study was to evaluate the stability of hematologic analytes in blood samples from monkeys, rabbits, rats, and mice when samples were stored for up to 72 hours at 4°C. Methods: Blood samples from 30 monkeys, 15 rabbits, 20 rats, and 30 mice were collected into EDTA‐containing tubes and were initially analyzed within 1 hour of collection using the ADVIA 120 analyzer. The samples were then stored at 4°C and reanalyzed at 24, 48, and 72 hours after collection. Results: Significant ( P<.0003) changes in hematologic analytes and calculations included increased HCT and MCV and decreased MCHC and cell hemoglobin concentration mean (CHCM) at 72 hours and increased MPV at 24 hours in monkeys; increased MCV at 72 hours and MPV at 48 hours and decreased monocyte count at 24 hours in rabbits; increased MCV and decreased MCHC, CHCM, and monocyte count at 24 hours in rats; increased MCV, red cell distribution width, and MPV and decreased MCHC, CHCM, and monocyte count at 24 hours in mice. Conclusions: Although most of the changes in the hematologic analytes in blood from monkeys, rabbits, rats, and mice when samples were stored at 4°C were analytically acceptable and clinically negligible, the best practice in measuring hematologic analytes in these animals is timely processing of blood samples, preferably within 1 hour after collection. 相似文献

Egg yolk-Tris is most commonly used semen extender; however, its use involves hygienic risk, interference with fertility and poor microscopic examination. Therefore, replacement of egg yolk with a plant-based component with protective effects on spermatozoa would be advantageous. In present study, we observed effect of soya milk-based extenders on dilution and liquid preservation of Murrah buffalo bull semen at 5°C up to 72 h in comparison with conventional egg yolk-Tris extender (Ext.1). In experiment one, a total of 32 buffalo semen ejaculates from four animals were extended and preserved at 5°C for 72 h in soya milk-based extender (Ext.2) with different percentages (10%, 15%, 20%, 25% and 30%) of soya milk for optimization of soya milk concentration. Semen quality was assessed for individual motility, viability, membrane integrity and acrosome integrity at 0, 24, 48 and 72 h of liquid preservation. The results of experiment one indicated that 25% soya milk is an optimum concentration for buffalo bull semen extender preparation. A modified method was used to prepare another soya milk-based extender (Ext.3). In the second experiment, two soya extenders (Ext.2 and 3) with optimized concentration (25%) of soya milk were comparatively assessed with egg yolk-Tris extender (Ext.1) for semen quality parameters at 0, 24, 48 and 72 h of liquid preservation. The individual sperm motility at 0 and 24 h following dilution were found non-significant among extenders. However, after 48 h of dilution, individual motility in Ext.3 was observed significantly (p < 0.05) higher than Ext.1. After 24, 48 and 72 h of dilution sperm membrane integrity in Ext.3 was found significantly (p < 0.05) higher than Ext.1. Overall, comparative evaluation of sperm parameters obtained revealed that Ext.3 containing 25% soya milk can be used as a substitute of egg yolk-based extender for buffalo semen liquid preservation. 相似文献

1. Studies were conducted to determine the effects of anticoagulants and storage (4°C) on the PCV of blood samples from Nigerian domestic fowl (DF) and the guinea fowl (GF). 2. Citrate significantly reduced pre‐storage PCV of the DF in comparison with the effect of ethylenediamine tetra‐acetic acid (EDTA). 3. It further decreased (P<0.05) the PCV of the blood of DF and GF over 3 d of storage; this was similar to the effect of EDTA on the PCV of the GF blood. 4. Citrate and oxalates induced haemolysis of blood of the DF and the GF in storage faster than EDTA, but overall the haemolysis was more pronounced from red cells of the GF than from those of the DF. 5. The mean fall in the PCV of the DF was significant at 3.0 mg EDTA/ml of blood in contrast to the fall in the PCV of the GF blood. 相似文献

To evaluate a novel feed additive, hops β-acids, a pen trial using 1,440 one-day-old broiler chickens, in a randomized design with 6 treatments and 6 replicates of 40 birds/pen, was conducted. The experimental treatments were a negative control (NC) basal diet, a positive control (PC) basal diet supplemented with zinc 30 mg/kg of bacitracin, and 4 additional treatments consisting of the basal diet supplemented with 30, 60, 120, and 240 mg/kg of hops β-acids in a microencapsulated form. All birds were vaccinated against coccidiosis by eye drops. Productive performance, jejunum morphology, and Clostridium spp. count in jejunum and cecum contents were evaluated. At 21 d of age, the birds fed the PC, 30, or 60 mg/kg of β-acids had the same weight gain of those fed the NC, whereas the highest level of β-acids decreased the gain. In addition, feed intake was decreased and FCR was improved in all treatments when compared with the NC. At 42 d, compared with the NC, the treatments containing 30 or 240 mg/kg of β-acids and the PC resulted in improved FCR. No differences were found for other performance variables. No difference was observed among the treatments on villus length, crypt depth, and villus-to-crypt ratio. Except for one sample, all others had negative results for Clostridium spp. based on the methodology of sulphite reduction. In this study, we demonstrated positive effects of supplementation of hops β-acids on productive performance of broilers, and the best results were obtained with addition of 30 mg/kg. 相似文献

The influence of phthalate esters di-2-ethylhexyl phthalate (DEHP) and mono-2-ethylhexyl phthalate (MEHP) on uterine prostaglandin (PGF2α and PGE2) and ovarian oxytocin secretion was investigated. Endometrial, granulosa, and luteal cells from cows on days 8–12 of the estrous cycle were treated with DEHP or MEHP (0.1, 1, or 10 ng/mL). We found that DEHP and MEHP stimulated ( P < 0.05) secretion of PGF2α and inhibited ( P < 0.001) secretion of PGE2 from endometrial cells. The ratio of PGF2α to PGE2 was markedly altered. The endocrine disrupting chemicals also enhanced secretion of oxytocin ( P < 0.05) from ovarian cells. Our results indicated that DEHP and its metabolite MEHP could affect the process of the estrous cycle by impairing secretion of prostaglandin from the uterus and oxytocin from the ovary. 相似文献

The aim of this study was to investigate the levels of acute-phase proteins and minerals as indicators for the reactivity in 1-year-old sheep. A total of 26 Chios breed sheep were fed colostrum-breast milk (control, n = 15)or were deprived afterseparation from their mother immediately after birth(experimental, n = 11). Mannheimia (Pasteurella) haemolytica serotype A1 was inoculated intratracheally and blood samples were taken in vacuumed sera on days 0, 1, 4, 7, 10, 13, 16, 19 and 22. Antibiotic treatment was initiated after blood sampling on day 22, and blood samples were taken on days 1, 4 and 7 after the treatment. The levels of C-reactive protein (CRP), haptoglobin, ceruloplasmin, fibrinogen, zinc, iron and calcium, which are the indicators of immune function and infectious diseases were analysed. No significant difference between the control and trial groups before and after the infection was determined. However, serum CRP, haptoglobin, ceruloplasmin and fibrinogen levels were increased in the course of the infection. These levels were restored to normal following treatment. 相似文献

Production of the Th1 cytokine interferon gamma (IFNγ) is associated with resistance to intracellular pathogens, including Rhodococcus equi. While neonatal foals are initially deficient in IFNγ production, expression of this cytokine increases throughout their first year of life. This is presumably the result of stimulation by environmental antigens including pathogen associated molecular patterns (PAMPS) signaling through toll-like receptors (TLR). This increased expression of IFNγ is likewise associated with an age-related resistance to R. equi infection. While immunostimulants containing PAMPS have been administered to adult horses in an attempt to modify their immune response, the effect of these materials on IFNγ expression in foals is unknown. Red seabream iridovirus (RSIV), a member within genus Megalocytivirus ( Iridoviridae), causes serious economic losses to marine fish aquaculture industry in East Asia. In this study, we established a Blue Striped Grunt Haemulon sciurus fin (grunt fin; GF) cell line persistently infected with RSIV (PI-GF RSIV) by subculturing GF cells that survived RSIV inoculation. PI-GF RSIV cells were morphologically indistinguishable from naive GF cells. They could stably produce RSIV at approximately 10 4.9 ± 0.5 genomes per microliter after 24 passages over 18 months. The optimum temperature to produce RSIV in PI-GF RSIV cells was 25°C. These cells also produced RSIV at 15, 20, and 30°C with multiple subcultures. The amount of RSIV yielded from PI-GF RSIV cells decreased gradually by multiple subculturing at 15°C or 30°C. Red seabream iridovirus was no longer detected from PI-GF RSIV cells after subcultures at these temperatures. These PI-GF RSIV cells freed from RSIV infection exhibited a level of RSIV productivity similar to those of naive GF cells after inoculation with RSIV. Therefore, we consider that these PI-GF RSIV cells were no longer infected with RSIV after multiple subculturing at 15°C or 30°C. This study was performed to investigate the effects of β-hydroxy-β-methylbutyrate (HMB) administration on skeletal system properties in turkeys. Thirty-two males were randomly divided into two groups at the age of 35 days of life. The first group included control turkeys (n = 16) treated with placebo, while the second group of birds (HMB group; n = 16) was administered orally with calcium salt of HMB during the last 15 weeks of life. The turkeys were sacrificed at the age of 20 weeks and tibia was isolated for analysis of bone geometrical parameters, volumetric bone mineral density (vBMD) and mechanical properties. Furthermore, assessment of free amino acid concentrations in plasma was performed. The results showed a 6.3% increase of vBMD of tibia in response to HMB treatment (p < 0.01). Cross-sectional area, second moment of inertia, maximum elastic strength and ultimate strength of tibia were significantly increased in HMB-treated turkeys by 21.3%, 49.0%, 27.2% and 28.3%, respectively (p ≤ 0.01). β-hydroxy-β-methylbutyrate administration increased plasma concentrations of proline,glutamate, leucine, isoleucine, valine, alanine, aspartate, phenylalanine and cysteic acid (p < 0.05). These results indicate that long-term administration of HMB improves vBMD, and geometrical and mechanical properties of skeletal system in turkeys, and that these effects are associated with improved plasma amino acid concentrations. 相似文献

BackgroundAccumulating evidence suggests that transfusion of packed red blood cells ( PRBCs) stored for >14 days is associated with increased rates of sepsis, multiple organ dysfunction, and mortality in human patients. ObjectiveTo determine if duration of PRBC storage has an effect on morbidity and mortality in dogs after transfusion. AnimalsDogs admitted to the Matthew J Ryan Veterinary Hospital of the University of Pennsylvania. MethodsA retrospective case review of dogs identified through blood bank logbooks that received PRBC transfusions (minimum, 5 mL/kg) between 2001 and 2010. Dogs were categorized according to major cause of anemia (eg, hemorrhage, hemolysis, ineffective erythropoiesis) for analysis. ResultsA total of 3,095 dogs received 5,412 PRBC units. Longer duration of PRBC storage was associated with development of new or progressive coagulation failure ( P = .001) and thromboembolic disease ( P = .005). There was no association between duration of PRBC storage and survival for all dogs overall. However, a logistic regression model indicated that for dogs with hemolysis, 90% of which had immune‐mediated hemolytic anemia, longer duration of PRBC storage was a negative risk factor for survival. For every 7 day increase in storage, there was a 0.79 lesser odds of 30 day survival (95% CI, 0.64–0.97; P = .024). Conclusions and Clinical ImportanceDuration of PRBC storage does not appear to be a major contributing factor to mortality in the overall canine population. However, longer duration of PRBC storage may negatively impact outcome in dogs with immune‐mediated hemolytic anemia, thus warranting further investigation with prospective studies. 相似文献
