This study analyses anomalous cases of gestation ending in pregnancy loss during the early foetal period and their effect on progesterone and plasma pregnancy-associated glycoprotein-1 (PAG-1) concentrations. Data derived from a large-scale ultrasound pregnancy diagnosis programme in high producing dairy cows. Over a 3-year period (2004–2007), a very low incidence (0.5%: 15 of 3094) of anomalous pregnancies was recorded. The results revealed that the following anomalies were detected on days 35–41 of gestation in cows carrying singletons with one single corpus luteum: embryo death in eight cows (0.3%); and embryo in the uterine horn contralateral to the corpus luteum in seven cows (0.2%). All these animals suffered pregnancy loss during the early foetal period. In cows carrying dead embryos, no signs of conceptus degeneration were observed on pregnancy diagnosis. Amnion size (approximately 25 mm diameter) and uterine horn fluid contents were estimated to be similar to those of the normal pregnant cows in this period. In the contralateral gestations, live embryos were observed in all ultrasound checks before pregnancy loss. Uterine fluid contents increased in the two cows in which gestation continued for more than a week. In the cases of embryo death but not in those of contralateral gestation, a drop in PAG-1 levels was noted prior to pregnancy loss. Two cows carrying dead embryos increased with time allantoic fluid contents. The PAG-1 values increased with time in one cow bearing a dead embryo (from 2.31 to 6.79 ng/ml) and in two of the contralateral gestations (from 1.66 to 2.33 ng/ml and from 0.39 to 6.79 ng/ml, respectively). Results of this study indicate that the foetal membranes continue to undergo some activity following embryo death, and that contralateral pregnancy may determine failure of the gestation process.  相似文献

OBJECTIVE: A suboptimal uterine environment contributes to the bovine repeat breeder syndrome. Subclinical endometritis is a component, so the mechanism by which inflammation affects embryo survival was investigated by assessing the effect of non-cellular products of an inflamed endometrium on embryo development to blastocyst. DESIGN: Endometrial fluid from a lactating dairy cow was collected by lavage using embryo culture medium. Aseptic inflammation was then induced by infusion of glycogen and lavage was repeated 6 h later. The recovered fluid was used to culture Day 5 in-vitro-derived embryos for 2 days. Embryo development and quality were compared for two treatment groups (culture media conditioned by inflamed or non-inflamed endometrium) and two controls (control or control + serum). RESULTS: Development to blastocyst was higher for conditioned media or media + serum (inflamed 42.2%; non-inflamed 49.3%; control + serum 50.9%; control 16.9%; P < 0.001). Blastocyst cell numbers were lower for the conditioned-inflamed group (inflamed, 83.1; non-inflamed, 99.8; control + serum, 100.6; control, 110.1; P < 0.001). Trophectoderm cell number, but not inner cell mass number, was reduced in the conditioned-inflamed group and the inner cell mass:trophectoderm ratio was increased (P < 0.001). CONCLUSION: Altering the embryo culture environment with the products of endometrial inflammation had a subtle effect on embryo quality. An increased inner cell mass:trophoblast ratio is likely to negatively affect embryonic survival. Altered embryo quality is a mechanism for early embryonic failure in cows with subclinical endometritis. Culture of embryos with normal endometrial fluid may improve in vitro embryo production.  相似文献

Two Holstein heifers and a cow were diagnosed with White Heifer Disease by ultrasonography. Case 1 was a 14 month-old heifer with aplasia of both sides of the uterine horn. In case 2, a primiparous cow and case 3, an 18 month-old heifer, both showed aplasia of the right uterine horn. Case 2 became pregnant by artificial insemination at ipsilateral ovulatory follicle and corpus luteum in the left ovary, while case 3 became pregnant by embryo transfer at 7 days after oestrus with contralateral corpus luteum in the right ovary.  相似文献

CASE HISTORY: A routine ultrasonographic examination for pregnancy diagnosis in a 3-year-old ewe revealed a normal embryo located in the uterine cavity and an abnormal mass located in the abdomen. At the time of examination, the ewe was 4.5?months pregnant, but 10?months previously she had undergone a caesarean section due to dystocia.

CLINICAL FINDINGS: The abnormal mass (12?cm × 8?cm) was located outside the uterine cavity; it was anechoic at the periphery, and in the centre there was an echogenic embryo-like structure. Exploratory laparotomy was performed, which revealed an ectopic pregnancy through the caesarean scar.

TREATMENT: The ectopic foetus was excised and the caesarean scar was repaired. The ewe made a complete recovery and the intrauterine pregnancy was completed to full-term, resulting in the birth of a healthy female lamb 35?days after the removal of the ectopic foetus.

CLINICAL RELEVANCE: Ectopic pregnancies have previously been described in ewes. As far as the authors are aware, this is the first report of an ectopic pregnancy through a caesarean scar in sheep or any other farm animal species.  相似文献

Topical application of prostaglandin E2 (PGE2) gel to the surface of the uterine tubes via a laparoscopic procedure improved embryo recovery rates or pregnancy rates in 28 subfertile mares suspected of uterine tubal pathology. Gelatinous masses may occlude the lumen of the uterine tube and prevent sperm from reaching the site of fertilization or prevent embryos from reaching the uterus. PGE2 is secreted by the early equine embryo, promoting passage of the embryo into the uterus; topical administration of PGE2 onto the surface of the uterine tube has been shown to stimulate early transport of the embryo into the uterus. Embryos were produced or a pregnancy was obtained from 24 of the 28 barren mares treated with direct laparoscopic application of 0.2 mg of PGE2 to their uterine tubes. Mares had been barren for an average duration of 1.9 ± 0.6 years and an average of 6.9 ± 3.8 estrous cycles prior to treatment, without donating an embryo or becoming pregnant. Seventeen of 20 mares bred as embryo donors produced one or more embryos with an average of 2.1 ± 1.9 embryos collected per mare (0.45 embryos per cycle) after PGE2 treatment. Seven of 8 mares bred to carry their own pregnancy became pregnant within the first two cycles following PGE2 treatment. These 8 mares were bred an average of 5.6 ± 1.8 cycles without a pregnancy prior to treatment. The laparoscopic PGE2 procedure was performed during various stages of the estrous cycle; the stage varied among treated mares.  相似文献

Cattle are fed moderate amounts of long chain fatty acids (FA) with the objective to enhance lactation and growth; however, recent interest on lipid feeding to cows has focused on reproduction, immunity and health. Increasing the caloric density of the ration by fat feeding has generally improved measures of cow reproduction, but when milk yield and body weight losses were increased by fat supplementation, positive effects on reproduction were not always observed. Feeding fat has influenced reproduction by altering the size of the dominant follicle, hastening the interval to first postpartum ovulation in beef cows, increasing progesterone concentrations during the luteal phase of the oestrous cycle, modulating uterine prostaglandin (PG) synthesis, and improving oocyte and embryo quality and developmental competence. Some of these effects were altered by the type of FA fed. The polyunsaturated FA of the n-6 and n-3 families seem to have the most remarkable effects on reproductive responses of cattle, but it is not completely clear whether these effects are mediated only by them or by other potential intermediates produced during rumen biohydrogenation. Generally, feeding fat sources rich in n-6 FA during late gestation and early lactation enhanced follicle growth, uterine PG secretion, embryo quality and pregnancy in cows. Similarly, feeding n-3 FA during lactation suppressed uterine PG release, and improved embryo quality and maintenance of pregnancy. Future research ought to focus on methods to improve the delivery of specific FA and adequately powered studies should be designed to critically evaluate their effects on establishment and maintenance of pregnancy in cattle.  相似文献

One of the problems encountered in embryo transfer (ET) in cattle is the wide variation in pregnancy rates. Many factors, of either embryonic, maternal or environmental origin, influence the result of the ET (Sreenan & Diskin 1987). An important environmental factor is the method of transfer. Pregnancy rates are generally lower following non-surgical, compared with surgical transfer. In addition, the skill and experience of the operator is of great importance. Furthermore, pregnancy rates tend to decrease the longer the transfer catheter remains in the uterus (Rowe et al. 1980). One suggested reason for this is uterine trauma or irritation caused by the catheter with a subsequent endometrial inflammation. As the uterine environment is of great importance for the establishment of pregnancy, an inflammatory reaction would certainly diminish the chances for embryo survival. There is a possibility that the endometrium becomes damaged mechanically during the transfer procedure. This might be more pronounced in animals where ET is difficult to perform, due to anatomical reasons and/or, an inexperienced operator. Furthermore, when a longer time is needed for passing the cervix and placing the embryo in the tip of the uterine horn, the pregnancy rate seems to be influenced negatively.  相似文献

A transvaginal ultrasound-guided intrauterine injection (IUI) technique was developed for embryo transfer and for injection of small quantities of sperm in mares. The target area of a horn was positioned by transrectal manipulation against the wall of the vaginal fornix over the face of a transvaginal transducer. A needle with a catheter containing the embryo or semen was inserted through the needle guide of the transducer into the uterine lumen. The tips of the needle and catheter, the movement of the catheter in the uterine lumen, and the ejection of fluid was monitored on the ultrasound screen. Pregnancy rate 15 days after ovulation for the IUI embryo transfer technique (30/39, 77%) was similar to the pregnancy rate for transcervical (TC) embryo transfer (30/38, 79%). The pregnancy rate for IUI insemination of 20 × 106 progressively motile sperm into the tip of the uterine horn ipsilateral to ovulation was 5/10 (50%). Results indicated that the IUI approach is a viable alternative for embryo transfer. Results also supported the potential of IUI for insemination of low numbers of sperm, but more extensive studies with various doses of sperm are needed.  相似文献

Synchronization of development between the embryo and uterus is required for successful pregnancy establishment.Transfer of early embryos requires synchrony with the recipient uterus of 2 days or less in sheep,because asynchrony of 3 days or more results in failure of pregnancy recognition signaling for maintenance of corpus luteum (CL) and progesterone (P4) production and/or uterine support of the embryo.The objective was to determine if P4 treatment of recipient ewes would obviate the need for pregnancy recognition signaling and maintain a uterine environment conducive to embryo survival after asynchronous transfer,thereby establishing a universal recipient.Embryos (morulae/blastocysts) were recovered on day 6 from super-ovulated donor ewes.Recipient ewes received 25 mg P4 daily from day 6 post-estrus until 60 days after embryo transfer.Embryos were transferred into recipients on day 6,9,12,18,or 30 post-estrus.The pregnancy rate on day 22 post-transfer was 60% for synchronous transfers to day 6 ewes,44% and 22% for asynchronous transfers to day 9 and 12 ewes,and 0% for asynchronous transfers to day 18 and 30 ewes.On day 39 post-transfer,pregnancy rates remained 60% for day 6 ewes,33% for day 9 ewes,and 0% for day 12,18,and 30 ewes.The P4 treatment did extend the window of uterine receptivity to early embryos in ewes by one day,but did not create a universal recipient.Available results support the idea that a window of uterine receptivity to the conceptus exists in sheep that is independent of pregnancy recognition signaling.  相似文献

Mycobacterium paratuberculosis was isolated from the uterine flush fluids obtained from 3 to 4 cows with clinical paratuberculosis. Four cows with clinical paratuberculosis were subjected to uterine lavage, using an established embryo recovery technique, and the recovered fluids were cultured for M paratuberculosis. Mycobacterium paratuberculosis has previously been demonstrated to adhere to bovine ova. Embryos within the uterus of a superovulated cow infected with M paratuberculosis could be exposed to the organism.  相似文献

The objectives of the study were to characterize an ultrasound reproductive tract scoring (URTS) system to assess suitability for breeding in dairy cows, to describe the prevalence of these scores in commercial dairy herds and to examine their relationship with subsequent fertility. Ultrasound examinations (7797) were performed on 5751 Holstein–Friesian cows prior to breeding in 62 seasonally calving herds over 2 years. Data recorded from images of both ovaries and the uterus were combined into a six point scoring system and the prevalence of cows with cystic ovarian follicles and uterine abscesses and adhesions was recorded separately. The prevalence of ovulatory cows which had completed uterine involution (score 1), or had mild (2) or moderate endometritis (3) was 62.5%, 21.7% and 2.2%, respectively. The prevalence of anovulatory cows with moderate endometritis (4), ovulatory cows with pyometra (5) and anovulatory cows which had completed uterine involution (6) was 3.3%, 2.2% and 8.1%, respectively. The interval between calving and examination differed between cows with each of the scores 1, 2, 5 and 6 (61, 46, 53 and 50 days, respectively, p < 0.05) but not between cows with scores 3 and 4 (37 and 35 days, respectively). Ovulatory cows which had completed uterine involution (score 1) had a higher likelihood of pregnancy to first service than ovulatory or anovulatory cows which had not completed uterine involution (p ≤ 0.10). The prevalence of cows with cystic ovarian follicles or abscesses or adhesions of the reproductive tract was 3.9% and 1.2%, respectively. In conclusion, 29% and 11% of cows in seasonally calving and breeding dairy herds had not completed uterine involution or were anovulatory prior to the mating start date, respectively. Both conditions, detected using a URTS system, significantly reduced first service pregnancy rate in these pasture-based dairy herds.  相似文献

Uterine disease after parturition is present in up to 40% of dairy cows and can cause cow infertility,which restricts efficiency of the cow production and leads to large economic loss.System administration and intrauterine infusion are widely utilized to treat cow endometritis.In recent years,the balloon technique is used gradually.Uterus washing pipe is used to inject liquid into the uterus and then the liquid and the inflammatory substances are discharged by the veterinarian.After the inflammatory substances cleaning up,the drug is delivered into the uterus.In order to provide a new method that can treat the infertility in dairy cows with metritis,323 cows which were diagnosed with metritis were used to detect the efficiency of balloon technique,the location of the balloon and the different drugs perfusion against infertility in dairy cows with metritis.The results showed that the balloon technique was showed remarkable effect against cow metritis comparing to the conventional technique.The pregnancy rate of the balloon technique was more than 60%,by contrast,the conventional technique was about 35%.Compared with being placed in the cornua uteri,the efficacy was more obvious when the balloon was placed in the uterine body,and the pregnancy rate was more than 60%.However,the pregnancy rate of another method was almost 40%.There were prominent curative effect of the three drug combinations.The pregnancy rates of the saline+Oxytetracycline,Lutajing+Gongdekang and saline were more than 70%,60% and 55%, respectively.Therefore,compared to the conventional technique and cornua uteri technique,balloon technique(uterine body)significantly increases the pregnancy rate,which is beneficial to be widely used in production.So it is significant that the new technique used widely is important for improving the production efficiency of dairy cows.  相似文献

分娩后奶牛发生子宫炎症的概率高达40%,子宫炎引发的不孕症是制约奶牛业生产效率的主要因素之一,造成了巨大经济损失。对于子宫炎的治疗主要有传统的子宫内灌注给药和系统给药方法。近年来,气囊子宫冲洗法逐渐被应用,其利用专用子宫冲洗管,将气囊置于奶牛子宫,注入液体冲洗子宫后,人工把液体及炎性物质全部排出,冲洗干净后投药。本研究利用323头被诊断为子宫炎的奶牛,进行气囊法与常规法治疗效果、气囊所放位置和冲洗用药对治疗效果的影响研究。结果表明,与常规法相比,气囊子宫体冲洗法提高了子宫炎奶牛的配种率,气囊法配种率达60%以上,而常规法配种率仅为35%左右。气囊子宫体冲洗法治疗效果明显优于子宫角放置法,气囊子宫体冲洗法配种率达60%以上,而气囊子宫角冲洗法治疗后其配种率仅为40%左右,且子宫体冲洗法节省时间、易于操作,安全系数高。3种药物组合都取得明显效果,生理盐水+土霉素子宫注射剂法的配种率达70%以上,露它净+宫得康法的配种率达60%以上,只用生理盐水冲洗的配种率也达55%以上。因此,与子宫内灌注给药、气囊子宫角放置冲洗等方法相比,气囊子宫体冲洗法显著提高了子宫炎奶牛的配种率,适用于在规模化生产中进行应用。这一新方法的推广对提高奶牛生产效率具有重要意义。  相似文献

This study aimed to evaluate the effect of oestrous synchrony between donors and recipients and the embryo quality on the pregnancy rate in beef cow recipients. The experiment was performed over two years at an embryo transfer (ET ) centre in Southern Brazil. Ninety Aberdeen Angus cows were subjected to superovulation (SOV ) protocols, resulting in the recovery of 1,048 transferable embryos. Eleven groups were formed with intervals of 6 hr, from ?30 to +30 hr, with respect to recipient versus donor oestrous detection. Evaluation of embryo quality was according to the IETS guidelines. The overall pregnancy rate was 52%. Effects related to donor and recipient oestrous synchronization on pregnancy rate were observed (=  .01), ranging from 36% to 50%. The embryo quality rate affected the pregnancy rate, where Grade I resulted in 57% and Grade III in 43% of pregnancy (<  .001). The embryonic state (frozen or fresh) showed no (>  .05) effect on pregnancy rate: 53% for fresh embryos and 44% for frozen embryos. The odds ratio for explanatory variables causing pregnancy indicated that Grade III embryos had 31% less chance of conception compared to Grade I. Thus, oestrous synchrony between donor and recipient, considering ±30 hr apart, can affect the pregnancy rate along with embryo quality.  相似文献

The endometrial epidermal growth factor (EGF) profile is an indicator of uterine function and fertility in cattle. The present study aimed to investigate the effects of heat stress on the endometrial EGF profile and fertility in lactating Holstein cows. The endometrial EGF profiles of 365 cows in the Hokkaido and Kyushu regions were examined between June and September (heat stress period, n = 211) and between October and January (control period, n = 154). EGF profiles were investigated using uterine endometrial tissues obtained by biopsy 3 days after estrus (Day 3). The proportion of cows with an altered EGF profile was higher between June and September than between October and January (41.2 vs. 16.2%, P < 0.05). The effects of rectal temperature on Days 0 and 3 on the endometrial EGF profile were also assessed in cows (n = 79) between June and September in the Kyushu region. A single embryo was transferred to cow on Day 7 to evaluate fertility (n = 67). Regardless of the rectal temperature on Day 3, the proportion of cows with an altered EGF profile was higher (64.1 vs. 30.0%, P < 0.05) and the pregnancy rate after embryo transfer (ET) was lower (26.7 vs. 51.4%, P < 0.05) in cows with a rectal temperature ≥ 39.5°C on Day 0 than in cows with a rectal temperature < 39.5°C on Day 0. The present results indicate that alterations in the endometrial EGF profile induced by an elevated body temperature on Day 0 contributed to reductions in fertility in lactating dairy cows during the heat stress period.  相似文献

The present experiment was conducted to evaluate the effects of feeding allowance level on embryonic survival, uterine development and reproductive hormone secretion in early gestation gilts. A total of 54 F1 crosses of Landrace x Large white gilts were randomly allocated to three treatment groups of high (H, 2 x maintenance), medium (M, 1.2 x maintenance) and low (L, 0.6 x maintenance) feeding level after mating. Blood samples and uterine flushings were collected on day 12, 25 and 35 of pregnancy, and embryonic survival rate was estimated. Concentrations of insulin-like growth factor I (IGF)-1, insulin, growth harmone (GH), leptin and progesterone in serum and uterine flushings were determined by enzyme-linked immunosorbent assay. Embryonic survival was affected by dietary treatment; total number of viable embryos and embryo survival of group M were higher than other groups in the early pregnancy (p < 0.05). Greater foetal weight in M gilts was achieved when gestation advanced to day 35 (p < 0.05), though there was no difference on day 25 of pregnancy among treatments. No appreciable differences were observed in the crown-rump length on day 25 and 35 of pregnancy among groups. Greater weight of uterus and products of conception were identified in M gilts compared with group H and L (p = 0.024 and p = 0.005, respectively) on day 25 of pregnancy. The hormone level was greatly affected by feeding allowance level. In serum, concentrations of IGF-1, leptin and insulin tended to be greater in H than in M and L during the early gestation, while concentrations of GH were greater in M and progesterone were the lowest in H. At the same time, feed allowance level affected the concentration of IGF-1, insulin, GH, leptin and progesterone in uterine flushings. These data demonstrated that feed allowance level after mating has important consequence on embryo survival, embryo development and uterine development, possibly mediated by nutrition level inducing changes in concentrations of reproductive hormones and/or intermediary metabolites in early pregnancy.  相似文献

Proteins that cross-react with antiserum to the major progesterone-induced proteins found in the pregnant sheep uterus, the uterine milk proteins (UTM-proteins), were detected as radiolabelled secretory products of endometrium from pregnant cows. Cross-reactive proteins included a form at 57,000 molecular weight as well as other lower-molecular-weight variants found in lower amounts. Similar proteins were also detected in uterine fluid from a cow at day 270 of gestation. Using immunohistochemical procedures, proteins that cross-reacted with antiserum to sheep UTM-proteins could be localized to the epithelial cells of endometrial glands in the cow. Results indicate that UTM-protein-like molecules are secreted by the endometrium of the cow during pregnancy.  相似文献

The effects of supplemented protein level (PL) during the periconceptional period and their interaction with body condition were evaluated in sheep. Multiparous Rambouillet ewes (n = 12) received two PL of rumen undegradable protein (UIP) during a 30‐day pre‐mating and 15‐day post‐mating period: low [LPL, 24% crude protein (CP), 14 g UIP and 36 g/CP animal/day] and high [HPL, 44% CP, 30 g UIP and 50 g/CP animal/day]. While ovulation rate (OR) did not differ between treatments (1.6 ± 0.5, mean ± SEM), a lower fertility rate, a decreased embryo number and a reduced uterine pH (UpH) was observed in the HPL group (p < 0.05), irrespective of BC. Luteal tissue weight, volume and progesterone secretion did not differ among treatments. Sheep with lower UpH also had lower conceptus weight (Cwt; p < 0.05, r = 0.65) and conceptuses with lower mass tended to secrete less INF‐τ and IGF‐1, and the correspondent endometrial explants had a higher basal PGF release. Current study indicates that high protein diets during the periconceptional period in sheep modify uterine and embryonic relationships, increasing early pregnancy losses and inducing embryo growth retardation. Surviving embryos were affected by weight reductions, which could compromise later foetal growth and birth weight. Results evidence the key role of a balanced diet in reproductive success and indicate that the quality and nutrient composition of the maternal diet are essential for an adequate establishment of pregnancy, having paramount effects on the interplay of the embryo and the uterus.  相似文献

Reasons for performing the study: During embryo transfer (ET) the equine embryo can tolerate a wide degree of negative asynchrony but positive asynchrony of >2 days usually results in embryonic death. There is still confusion over whether this is due to the inability of the embryo to induce luteostasis or to an inappropriate uterine environment. Objectives: To assess embryo survival and development in an advanced uterine environment. Hypothesis: Embryo–uterine asynchrony, not the embryo's inability to induce luteostasis, is responsible for embryonic death in recipient mares with a >2 days chronologically advanced uterus. Methods: Experiment 1: Thirteen Day 7 embryos were transferred to the uteri of recipient mares with luteal prolongation, occasioned by manual crushing of their own conceptus, such that donor–recipient asynchrony was between +13 and +49 days. Experiment 2: Day 7 embryos were transferred to recipient mares carrying their own conceptus at Days 18 (n = 2), 15 (n = 2), 14 (n = 4), 12 (n = 4) or 11 (n = 4) of gestation. In addition, Day 8 embryos were transferred to 4 pregnant recipient mares on Day 11 of gestation. Results: No pregnancies resulted following transfer of Day 7 embryos to recipients in prolonged dioestrus with asynchronies between +13 and +49 days. However, the use of early pregnant mares as recipients resulted in 5/20 (25%) twin pregnancies, 4 of which came from the transfer of a Day 8 embryo to a Day 11 recipient. All transferred embryos showed retarded growth, with death occurring in 4/5 (80%). Conclusions and potential relevance: The results emphasise the importance of an appropriate uterine environment for embryo growth and the inability of equine embryos to survive transfer to a uterus >2 days advanced even when luteostasis is achieved. It is possible that in normal, non‐ET equine pregnancy, embryo–uterine asynchrony may account for some cases of embryonic death.  相似文献
