Maiden heifers and lactating cows of known ovarian status and of several breeds were treated with a synthetic prostaglandin, cloprostenol, or a synthetic progestagen, norgestomet, at the start of an artificial insemination (AI) program. Animals in the cloprostenol treatment received 2 injections 10 days apart. Over the next 26 days those animals that showed oestrous behaviour were inseminated. Synchronisation rates and calving rates to insemination over the first 7 days were calculated. Those in the norgestomet treatment received an implant of norgestomet plus an injection of norgestomet and oestradiol valerate. The implant was removed 10 days later and the animals were given an injection of pregnant mare serum gonadotrophin (PMSG). They were inseminated at 48 h (maiden heifers) or 56 h (lactating cows) after implant removal. Calving rates to fixed-time insemination were recorded. After completion of the AI program the animals in both treatments were joined with bulls. Overall calving rates (AI plus bulls) were calculated. By day 7 of the program, 82% of the maiden heifers and 76% of the lactating cows in the cloprostenol treatment had been detected in oestrus. By day 21 the respective figures were 99% and 81% Norgestomet treatment had an immediate and a prolonged effect on ovarian activity in those females classified as having inactive ovaries at the start of the AI program. Calving rates of those females to fixed-time AI and overall were similar to those of the females with active ovaries in both treatments. Their calving rates to fixed-time insemination, and overall calving rates for the lactating females, were significantly higher than the corresponding values of their contemporaries treated with cloprostenol and inseminated on observed oestrus over 7 days. For those females classified as having active ovaries at the start of the AI program, calving rates to first insemination and overall were similar for both treatments. Overall calving rates of lactating cows of each breed were, with one exception, higher in the norgestomet treatment than in the cloprostenol treatment. Although norgestomet treatment was more expensive than cloprostenol treatment, the advantage in calf crop resulted in an overall monetary advantage to the norgestomet treatment. 相似文献

Cloprostenol, 100 micrograms, given intramuscularly to the nanny, with 50 micrograms 10 hours later, precipitated parturition in goats after 36 +/- 1 hours (mean +/- SEM), when administered at 137 +/- 0.5 days gestation. All kids were born alive and survived to weaning. Milk yield over 40 weeks post partum was not significantly different from that after spontaneous parturition. Three hundred micrograms cloprostenol (200 micrograms with 100 micrograms 10 hours later) also initiated parturition at 137 +/- 0.5 days gestation but caused a significant (P less than 0.01) suppression of lactation. Cloprostenol-induced parturition in more than one pregnancy had no adverse effects except for an increased incidence of placental retention, which was treated successfully with intrauterine pessaries containing oestrogen. During the first eight days after spontaneous parturition efficiency of milk secretion was inversely related to udder mass, suggesting a gradual maturation of the secretory alveolar epithelium over this time. When parturition was induced by cloprostenol there was a four to eight day delay before the establishment of this relationship which appeared essential for a successful lactation. Cloprostenol proved to be a useful tool for the control of parturition in goats, having applications to both general animal husbandry and for the study of mammary development and secretory competence. 相似文献

Reproductive management programmes based on strategic use of prostaglandin F2 alpha (PGF2 alpha) to induce and synchronize oestrus in post-partum dairy cows are widespread. Repeated shortening of the oestrous cycle during early lactation in high-yielding dairy cows, however, could impair corpus luteum function and thus decrease fertility. The objective of this study was to analyse the effect of repeated treatments with the prostaglandin F2 alpha analogue D (+) cloprostenol sodium on progesterone concentrations indicative of a functional corpus luteum in post-partum dairy cows. Furthermore, the influence of milk production, parity and endometritis on progesterone concentrations under these circumstances were studied. Eighty-four cows of a commercial dairy operation were treated three to four times with D (+) cloprostenol sodium (Preloban; Hoechst Roussel Vet, Wiesbaden, Germany) at 14-day intervals, starting 22-28 days post-partum. Blood samples were collected prior to treatment 1 (sample 1) and 14 days after treatments 1, 2 and 3 (samples 2-4) and serum progesterone (P4) levels were determined. The percentage of cows with P4 levels < 1 ng/ml decreased from 51% in sample 1 to 23% in samples 3 and 4. More primiparous cows had low P4 levels 14 days after the second treatment than older cows (P < 0.05). Cows with low progesterone levels in sample 3 or 4 had lower protein contents in milk on the second milk test day post-partum and in their cumulative milk yield of the first 100 days of lactation. Clinical endometritis at post-partum examination did not influence progesterone levels after treatment with PGF2 alpha. Repeated application of PGF2 alpha (more than twice) in the post-partum period does not influence serum progesterone levels 14 days after treatment. Failure to develop luteal tissue after treatment contributed to the conception failures after first service. 相似文献

Cloprostenol, a prostaglandin analogue, was administered intramuscularly to a total of 35 cycling buffalo cows and heifers in two doses, each of 0.5 mg, given 11 days apart. Out of five cows and 12 heifers subjected to observations after the second injection of cloprostenol (day 0), all except one heifer responded. Signs of oestrus were most marked on days 3 or 4. Eighteen treated heifers were kept with buffalo bulls for four days after the second injection while a control group of nine heifers was kept with bulls for 21 days. The first-service conception rate, diagnosed by rectal palpation at 60 days, was 33 1/3 per cent in both groups. Twelve treated heifers were artificially inseminated at 72 and 96 hours after the second injection of cloprostenol, using fresh semen diluted in egg yolk--citrate extender. Data from 3276 cows with 6598 lactations in 85 herds participating in a herd health monitoring programme were selected for an observational study. Cases of endometritis were diagnosed based on a clinical examination after 21 days postpartum. The cases were grouped by the severity of endometritis (mild, severe) and by the treatment performed (intrauterine infusion, prostaglandin injection, no treatment). The conception rates and the calving-to-conception intervals of these groups were compared statistically. Conception rate was lower in cows with severe endometritis compared to cows with mild endometritis while it did not differ between treatment groups. In cows with mild endometritis a significantly longer calving-to-conception interval was observed if intrauterine infusions were performed compared to the other two treatment options. The occurrence of other fertility problems during the same service period had a significant effect on conception rate and calving-to-conception intervals. In cows with severe endometritis intrauterine or prostaglandin treatments appeared to be beneficial, whereas intrauterine infusions in cows with slight endometritis had a negative effect on reproductive efficiency. 相似文献

In an experiment involving 161 farrowings, cloprostenol was injected on day 112 or 113 of gestation at the recommended dosage (175 μg) or a lower dosage (125 μg). Cloprostenol treatment did not result in abnormally high body temperatures of sows at parturition. Farrowing began within 29 hours of injection in 94% and 88% of the sows treated with 175 μg and 125 μg cloprostenol respectively, as compared to 15% of saline-injected controls. The duration of farrowing and number stillborn were not affected by treatment. Sows farrowing within 19 hours of treatment tended to have a large number of piglets and a higher body temperature postpartum. 相似文献

A series of experiments was conducted to investigate the potential of an analogue of prostaglandin-F2 α, namely ICI 80996, to control ovulation in the cyclic ewe. A single intramuscular injection of 100 μg of this material during the mid-luteal phase of the oestrous cycle resulted in rapid luteolysis with a consequent synchronised return to oestrus. Moreover, the endocrine changes during the induced cycle were not significantly different from those during a natural cycle. Two injections given 9 days apart controlled ovulation in all ewes no matter what stage of the cycle treatment commenced. There was a high degree of synchrony in the return to oestrus following the second injection (38.6 ± 0.8 h) and the time interval from injection to ovulation (72.9 ± 1.5 h). Fertility of treated ewes to natural service was not significantly different from controls when assessed in terms of both the conception rate (68 vs 710, respectively) and prolificacy (147 vs 141%, respectively). 相似文献

AIM: To evaluate the efficacy of a programme using oestradiol benzoate, progesterone and the prostaglandin-F2 (PG) analogue, cloprostenol, to synchronise oestrus and ovulation in dairy cows, compared with a programme using a gonadotropinreleasing hormone (GnRH) agonist, buserelin, and cloprostenol. METHODS: Twenty non-lactating dairy cows, at random stages of the oestrus cycle, were randomly assigned to 1 of 2 treatments. In Treatment 1 ( OPPG; n=10), cows were injected with 2 mg oestradiol benzoate intramuscularly (IM) plus 200 mg progesterone subcutaneously (SC) on Day 0, followed by 500 microg cloprostenol IM on Day 9 and 1 mg oestradiol benzoate on Day 10. In Treatment 2 (GPG; n=10), cows were injected with 10 microg buserelin IM on Day 0, 500 microg cloprostenol IM on Day 7 and 10 microg buserelin on Day 9. The ovaries of all cows were examined by ultrasonography, using an 8 MHz probe, from 5 days before the initial treatment until ovulation. Cows were observed for oestrus 3 times daily for 7 days after cloprostenol treatment. Blood samples were collected daily for determination of progesterone, and 6-hourly for 36 h after the second oestradiol or buserelin injection for the determination of follicle stimulating hormone (FSH) and luteinising hormone (LH) concentrations. RESULTS: The percentage of cows observed in oestrus was higher in the OPPG group than in the GPG group (100% vs 55.6%, p=0.018). Treatment with either short-acting progesterone plus oestradiol benzoate or buserelin was followed by atresia or ovulation of the dominant follicle. Emergence of a new follicular wave occurred earlier (p>0.001) in the GPG group (2.2+/-0.2 days) than in the OPPG group (3.6+/-0.2 days). There was no significant difference between treatment groups in the variation of time of follicular wave emergence or size of the largest follicles at either the time of initial treatment (10.8+/-1.4 mm vs 11.1+/-0.8 mm), cloprostenol treatment (13.8+/-0.7 mm vs 14.0+/-1.3 mm) or of ovulation (15.4+/-0.7 mm vs 17.6+/-1.1 mm; p=0.10). The LH surge occurred sooner after the second injection of buserelin (4.0+/-1.0 h) than after the second injection of oestradiol benzoate (22.8+/-1.2 h; p>0.001). The interval between the second injection of oestradiol benzoate or buserelin and ovulation did not differ significantly between treatment groups (1.7+/-0.3 days vs 1.6+/-0.2 days; p=0.69). CONCLUSIONS: The use of short-term progesterone treatment, combined with oestradiol benzoate for follicular wave synchronisation, and cloprostenol to cause lysis of residual luteal tissue, is a promising alternative to established methods of oestrus synchronisation in cows. 相似文献

The oxytocic effect of a prostaglandin F2 alpha analogue, fenprostalene, was assessed in 4 ovariectomized ewes fitted with electrodes in both uterine horns and in the cervix. In the absence of estradiol priming, significant motility changes were not elicited by fenprostalene. Conversely, when ewes were primed with 17-beta-estradiol, fenprostalene markedly increased the electrical activity in the uterus and cervix. After a single subcutaneous fenprostalene administration (5 micrograms/kg), activity values remained about twice that of the control values during 8.52 +/- 3.31 hours. When the same dosage was administered IM, similar post-injection activity values were obtained, but only during 5.88 +/- 0.72 hours. Oxytocic effects of fenprostalene were far longer than those elicited by a single IM administration of 50 micrograms of prostaglandin F2 alpha (tham salt)/kg (0.91 +/- 0.32 hours) or by a single IM administration of 1 microgram of cloprostenol/kg (1.88 +/- 0.81 hours). Using the dose-effect relationship curve obtained from the same ewes by continuous IV infusions of oxytocin (OXT), the postinjection activity values reached after a single subcutaneous administration of fenprostalene were equivalent to those of an IV infusion of OXT at an average dose of 4.09 ImU of OXT/kg/hr for 6 to 13 hours, according to the values of the particular ewe concerned. These long-lasting oxytocic properties, in addition to its luteolytic capabilities, would make fenprostalene a suitable drug for promoting effective evacuation of the uterus when required in daily veterinary practice. 相似文献

Prostaglandins (PGs) are essential to trigger the cascade of events that degrade the extracellular matrix of follicles leading to follicular rupture and ovulation. In mares, systemic administration of flunixin meglumine (FM), a PG synthetase inhibitor, blocks ovulation by inducing luteinized unruptured follicles (LUF). In the rat, the administration of PGF(2α) (PGF) and PGE restored ovulation in indomethacin treated animals. The mares were treated with FM 0, 12, 24 and 36 h after human chorionic gonadotrophin (hCG) administration to induce experimentally LUF (n = 15) or were left untreated (controls, n = 5). In addition, 250 μg of cloprostenol were administered intravenously to the mares 33, 35 and 36 h (CLO 33, n = 5) or 48, 49 and 50 h (CLO 48, n = 5) after hCG. One group was treated with FM but not with cloprostenol (FM-control, n = 5). The ovulation rate, follicular diameter and progesterone concentration were compared amongst groups. The ovulation rate at 48 h was higher (p < 0.05) in the controls (100%) than in the FM-control (0%), CLO 33 (0%) or CLO 48 (20%) mares. All but one FM treated mares developed LUF by 48 h after hCG administration. Two LUF collapsed between 48 and 60 h and 72 and 84 h in one mare from FM-control and from the CLO 33 group each, respectively. Progesterone concentration was significantly higher (p < 0.05) in the control mares than in any of the FM treated mares 5, 9 and 13 days after hCG. In conclusion, FM administered during the periovulatory period blocked ovulation in the mares. In contrast, the administration of cloprostenol, a PGF analogue, in the previously FM treated mares failed to restore ovulation. 相似文献

Oestrus synchronization was studied in samples from six cows of the Black-Pied Lowland breed. Three cows four to five days from oestrus were used as the control; three animals with marked periodic corpora lutea were given an i. m. injection of 0.5 mg cloprostenol. The eighth day from the administration of the preparation, the ovaries of the cows were excised and, after histological processing in a simultaneous series in a 4mm interval, the preparations were subjected to qualitative and quantitative microscopic evaluation. The structure of non-atretic and atretic follicles was described in different stages of the atretic process. The lymphoid cells of atretic follicles were observed to penetrate into the granulosa membrane. A multiplication of non-atretic tertiary follicles was observed after the administration of cloprostenol. This multiplication was more pronounced on the right ovary where the preceding ovulation had taken place (P less than 0.01). The treated animals, compared with the controls, showed a significant multiplication of tertiary follicles at early atresia and at total collapse atresia (P less than 0.001), whereas the number of follicles with contractive atresia showed a significant decrease (P less than 0.001). The results suggest that cloprostenol can influence follicle population mostly through the stimulation of the growth and ripening of tertiary follicles; its modulation effect seems manifest itself in cooperating relation with gonadotrophic hormones, mainly with the follicular secondary hormone (FSH), in the theory of the complex effect of proteohormones . 相似文献

Two groups of lactating dairy cows in each of 4 dairy herds were injected with either 0.5 mg (219 cows) or 0.3 mg (118 cows) of Ooprostenol (I.C.I.) The first group of cows were injected from Day 6 to Day 17 of the oestrous cycle (Day 0 — oestrus) and the second group from Day 6 to Day 12. The four herds were selected specifically because oestrus-detection procedures were thorough and reliable. Of the cows receiving the 0.5 mg dose rate, 54.3% were observed in oestrus within 4 days of treatment. The remainder were in groups which were: (i) in oestrus from 5 to 10 days post-treatment (20.5% with prolonged response intervals), and 11 to 20 days post-treatment (15.5% with unaltered cycle lengths); or (ii) experienced a “silent oestrus” in the immediate post-treatment period and either did not show oestrus for a further 3 weeks (6.4% in oestrus from 21 to 29 days post-treatment) or conceived to a single set-time insemination 72 h after treatment (0.9%). Use of the lower dose rate resulted in fewer cows being observed in oestrus within 72 h (43.8% v 31.4%) and a corresponding rise in the percentage experiencing a “silent oestrus” and returning to service (6.4% v 13.6%) or conceiving to set-time insemination (0.9% v 3.4%). Most of the unaltered cycle lengths occurred with cows being treated on Day 6 of the oestrous cycle. The pregnancy rate for the 171 cows observed in oestrus within 4 days of treatment and inseminated at 72 h, or 72h and 96 h, was 57.3%. Records from this trial, as well as previous trials in which cows were injected with 0.5 mg of Cloprostenol, showed that 446 of 524 cows (85%) injected at from Day 6 to Day 17 of the oestrous cycle were observed in oestrus within 5 days (144 h) of treatment. Amongthese 446 cows, the interval to post-treatment oestrus was influenced by the stage of the cycle at injection. Whereas 89% of the oestrous responses in cows treated on Days 6 to 9 were observed within 72 h of injection, the comparable figures for cows treated on Days 10 to 13 and Days 14 to 17 were 48% and 70% respectively. Variation in the post-treatment interval to oestrus is greater in lactating dairy cows than in heifers and is not resolved by using double-injection regimes. The reasons for this variation have not been identified. The most effective use of prostaglandins for oestrus synchronisation in lactating dairy cows requires the treatment of groups of animals at similar stages of theiroestrous cycles and their post-treatment insemination at observed oestrus. These requirements may limit the usefulness of this technology in herds with poor breeding management. 相似文献
