Background

Measurement of salivary cortisol has been used extensively as a non-invasive alternative to blood sampling to assess adrenal activity in ruminants. However, there is evidence suggesting a considerable delay in the transfer of cortisol from plasma into saliva. Previous studies in cattle have used long sampling intervals making it difficult to characterise the relationship between plasma and salivary cortisol (PLCort and SACort, respectively) concentrations at different time points and determine whether or not such a time lag exist in large ruminants. Therefore, the objective of this study was to characterise the relationship between plasma and salivary cortisol and determine if there is a significant time lag between reaching peak cortisol concentrations in plasma and saliva across a 4.25 h time-period, using short sampling intervals of 10–15 min, following social separation in dairy cattle.Five cows were separated from their calves at 4 days after calving, and six calves were separated from a group of four peers at 8 weeks of age. Following separation, the animals were moved to an unfamiliar surrounding where they could not see their calves or pen mates. The animals were catheterised with indwelling jugular catheters 1 day before sampling. Blood and saliva samples were obtained simultaneously before and after separation.

Results

In response to the stressors, PLCort and SACort increased reaching peak concentrations 10 and 20 min after separation, respectively. This suggested a 10 min time lag between peak cortisol concentrations in plasma and saliva, which was further confirmed with a time-series analysis. Considering the 10 min time lag, SACort was strongly correlated with PLCort (P < 0.0001).

Conclusions

Salivary cortisol correlates well with plasma cortisol and is a good indicator of the time-dependent variations in cortisol concentrations in plasma following acute stress. However, there is a time lag to reach peak cortisol concentrations in saliva compared to those in plasma, which should be considered when saliva samples are used as the only measure of hypothalamic-pituitary-adrenal axis response to stress in cattle.  相似文献

To assess the validity of laboratory parameters in blood and faeces as indicators of stress in cows, concentrations of cortisol, non-esterified fatty acids (NEFAs), 3-hydroxybutyrate, glucose, triglycerides, cholesterol, serum amyloid A (SAA) and haptoglobin in serum, as well as corticosterone in faeces, were determined in two breeds of cattle (Alberes and Bruna dels Pirineus) under different systems of housing and feeding. Serum cortisol concentrations were markedly elevated in the Alberes group, probably because they were less habituated to human handling. Corticosterone concentrations in faeces were significantly increased in the Bruna dels Pirineus cattle on Alberes pastures. Concentrations of NEFAs and cholesterol were significantly elevated in the Alberes cows, indicating an adrenergic stimulus of lipolysis or the existence of nutritional stress. SAA concentrations were significantly higher in groups living in hardy conditions, whereas there were no significant differences in haptoglobin between the three groups.  相似文献

In addition to its basic role in the metabolism of purine nucleotides, xanthine oxidoreductase (XOR) is involved in the generation of oxygen-derived free radicals and production and metabolic fate of nitric oxide (NO). Growth hormone (GH) and Vitamin E (E) have been shown previously to modify immune response to infection. Our objective was to determine in heifers the effect of endotoxin challenge (LPS; 3.0 μg/kg BW, i.v. bolus, Escherichia coli 055:B5) on xanthine oxidase (XO) activity in plasma and liver and the modification of this response by daily treatment with recombinant GH (0.1 mg/kg BW, i.m., for 12 days) or GH+E (E: mixed tocopherol, 1000 IU/heifer, i.m., for 5 days). In experiment 1, 16 heifers (348.7±6.1 kg) were assigned to control (C, daily placebo injections), GH, or GH+E treatments and were challenged with two consecutive LPS injections (LPS1 and LPS2, 48 h apart). After LPS1, plasma XO activity increased 290% (P<0.001) at 3 h, reached peak (430%) at 24 h and returned to basal level by 48 h after LPS2. XO responses (area under the time×activity curve, AUC) were greater after LPS1 than LPS2 (P<0.001). Total plasma XO responses to LPS (AUC, LPS1+LPS2) were augmented 55% (P<0.05) over C with GH treatment but diminished to C responses in GH+E. There was a linear relationship (r2=0.605, P<0.001) between total response in plasma XO activity and plasma nitrate+nitrite concentration. In experiment 2, 24 heifers (346±6 kg) were assigned to C or GH treatments and liver biopsy samples were obtained at 0, 3, 6, and 24 h after a single LPS challenge. Hepatic XO activities increased 63.3% (P<0.05) 6 h after single LPS challenge and remained elevated at 24 h (100.1%, P<0.01) but were not affected by GH treatment. Results indicate that LPS-induced increases in plasma XO activity could be amplified by previous GH treatment but attenuated by E administration. The data also suggest that E may be effective in controlling some mediators of immune response associated with increased production of NO via the effect on XO activity and its production of superoxide anion as well as uric acid.  相似文献

The objective was to assess the effect of banding or burdizzo castration performed on farms on plasma testosterone, acute-phase proteins, scrotal circumferences, growth, and well-being of bulls. 243 Continental bulls (12 months; 399.2 ± 5.72 kg) from three different farms were allocated at random, after stratification on weight within breed type, to one of three treatment groups: banding castration (BAND; n = 80), burdizzo castration (BURD; n = 83), or controls (CON; n = 80). The castration methods were conducted under local anaesthesia, and tetanus toxoid vaccine and antibiotic were also injected at castration. BAND and BURD castrates had lower (P < 0.001) plasma testosterone concentration than control bulls, with no difference between BAND and BURD castrates on 28 d post-castration. From days 0 to 14 post-castration, BAND (P = 0.0002) and BURD (P < 0.0001) castrates had lower average daily gain (ADG) than CON bulls, no difference (P = 0.46) was found between BAND and BURD castrates. From days 15 to 28, BAND castrates had lower ADG compared with BURD castrates (P = 0.03) and CON bulls (P = 0.01), while no difference (P = 0.76) was found between BURD and CON. From days 29 to 56, BAND (P = 0.01) and BURD (P = 0.002) castrates had lower ADG than CON bulls, no difference (P = 0.55) was found between BAND and BURD. From days 57 to 84, the ADG of BAND castrates was not different compared with BURD castrates (P = 0.12) and CON bulls (P = 0.38), while BURD had lower (P = 0.02) ADG compared with CON. The integrated ADG from day 0 to 112 of BAND (P = 0.0001) and BURD (P = 0.02) groups were lower compared with CON, while there was no difference (P = 0.09) between BAND and BURD castrates. On d 14 post-castration, BAND castrates had lower scrotal temperature than BURD (P < 0.0001) and CON (P < 0.0001), and BURD castrates had greater (P < 0.006) scrotal temperature than CON; BAND castrates had lower scrotal latitudinal and longitudinal circumferences than BURD castrates (P < 0.001) and CON bulls (P < 0.001), and BURD castrates had greater (P < 0.001) scrotal latitudinal and longitudinal circumferences than CON bulls. BAND (P < 0.0001) and BURD (P = 0.01) castrates had greater glucose concentration than CON bulls, and BAND castrates had greater (P = 0.04) glucose concentration than BURD. In conclusion, BAND or BURD castration significantly reduced plasma testosterone concentration; reduced average daily weight gain mainly during the first 2 weeks, which was not compensated during the subsequent 16 weeks; increased withdrawal of stored energy and increased plasma protein concentration. BURD showed an advantage over BAND in growth during days 15 to 28 following castration.  相似文献

Previously we have shown that resistance to Eimeria acervulina (EA) infection in broiler chickens was enhanced by a probiotic treatment. In the present studies, we examined cytokine and oocyst production under similar conditions using a commercial Lactobacillus-based probiotic. Day-old male broiler chicks were fed control or probiotic diets and were orally challenged with either 2×104 (Experiment 1) or 1×104 (Experiment 2) oocysts of EA at 3 weeks of age. For the first experiment, fecal oocyst shedding and IFN-γ levels in the culture supernatants of ConA-stimulated spleen lymphocytes were evaluated. Humoral and local cell-mediated immunity in the second experiment were assessed by evaluating antibody and cytokine (IFN-γ and IL-2) levels in sera and intestinal secretions on a 3-day interval post inoculation. Results showed small but significant (P<0.05) differences in cytokine levels and oocyst production but not antibody levels between the probiotic-treated and control groups. Collectively, these data suggest a positive impact of the probiotic on cellular immune responses of infected broilers as compared to control chickens resulting in enhanced resistance to EA as shown in reduced fecal oocyst shedding. The results showed an immunoregulatory effect of probiotic diets on the local cell-mediated immunity in poultry and provide a rationale for further study to investigate the beneficial effects of Lactobacillus-based probiotics in food animals.  相似文献

The effect of weaning the 4-5 heaviest piglets in the litter on day 33 of lactation and the remainder 2 days later (fractionated weaning) on plasma levels of prolactin, cortisol, oestradiol-17 beta (E2), progesterone (P4) and LH, as well as on the weaning to oestrus interval in primiparous sows was studied. Twelve crossbred sows were grouped into 6 pairs according to farrowing date and litter size. The litter of 1 sow in each pair (F) was weaned in 2 stages, and the other conventionally weaned at 35 days (C). Blood samples were collected via a permanent jugular vein catheter every 3 h from 9 a m to 9 p m daily throughout the experimental period, and intensively at 15 min intervals for 12 h on the day of first and final weaning and for 6 h on the day after each weaning. All sows were slaughtered following their first post-weaning oestrus and the reproductive organs were macroscopically examined. Lactational oestrus was not observed in any of the sows. Sows from 5 out of 6 pairs showed oestrus within 8 days of weaning and post-mortem examination showed normal ovulation. There was a tendency for the F sows to have a shorter weaning to oestrus interval, as compared with the C sows (5 of 6 pairs, 4.8 days v 5.6 days). The plasma levels of prolactin around weaning were not significantly different between the 2 groups. Within 6 h after final weaning, the prolactin concentrations decreased gradually from 7.6 and 8.7 to 1.6 and 1.7 microgram/l in the control and treatment groups, respectively. The plasma levels of cortisol, showing a diurnal rhythm (with the lowest level at 6 and/or 9 p m), did on no occasion differ between the 2 groups. On the day of final weaning, no diurnal rhythm was observed, with cortisol remaining high at 6 and 9 p m. The plasma levels of E2 and P4 were low until final weaning in both groups. After final weaning the E2 levels rose faster in the F sows than in the C sows, to 44.3 and 34.8 pmol/l, respectively, on day 2 (p less than 0.01). No significant differences in levels of plasma LH and the number of LH pulses were observed between the groups. After final weaning the average and base levels of LH and the number of LH pulse(s) increased significantly.  相似文献

Four controlled trials were conducted to evaluate the therapeutic and persistent efficacy of a new moxidectin formulation (moxidectin 1% nonaqueous injectable) against nematode parasites in cattle. This injectable moxidectin formulation, given as a single subcutaneous injection at a dose rate of 0.02 ml/kg BW to provide 0.2 mg moxidectin/kg BW, was highly efficacious (>90–100%) against larval and/or adult stages of many species of nematodes in cattle including, Dictyocaulus viviparus, Ostertagia spp., Trichostrongylus axei, Haemonchus placei, Trichostrongylus colubriformis, Cooperia spp., Nematodirus helvetianus, Strongyloides papillosus, Oesophagostomum radiatum and Trichuris spp. This formulation had persistent efficacy of >90% against D. viviparus for at least 6 weeks post-treatment, H. placei and Oe. radiatum for 5 weeks post-treatment, and Ostertagia spp. and T. axei for 2 weeks post-treatment.  相似文献

When the ratio of lean to fat deposition is improved, so is feed conversion efficiency. Net benefits may include lower production costs, better product quality, less excretion of nitrogenous wastes into the environment, decreased grazing pressure on fragile landscapes, and reduced pressure on world feed supplies. However, finding a way to achieve these goals that is reliable, affordable, and acceptable to the majority of consumers has proved to be a major challenge. Since the European Union banned hormonal growth promoters (HGPs) 15 years ago, countries such as Australia and the United States have licensed new products for livestock production, including bovine growth hormone (GH), porcine and equine GH, and the beta-agonist ractopamine. There has also been considerable research into refining these products, as well as developing new technologies. Opportunities to improve beta-agonists include lessening their effects on meat toughness, reducing adverse effects on treated animals, and prolonging their duration of action. In the last regard, the combined use of a beta-agonist with GH, which upregulates beta-adrenoceptors, can produce an outstanding improvement in carcass composition and feed efficiency. Insulin-like growth factor-1 (IGF-1) mediates many of the actions of GH, but has proved to be of more use as a growth reporter/selection marker in pigs, than as a viable treatment. However, a niche for this product could exist in the manipulation of neonatal growth, causing a life-long change in lean:fat ratio. Another significant advance in endocrinology is the discovery of hormones secreted by muscle and fat cells, that regulate feed intake, energy metabolism, and body composition. Leptin, adiponectin and myostatin were discovered through the study of genetically obese, or double-muscled animals. Through genetic manipulation, there is potential to exploit these findings in a range of livestock species, although the production of transgenic animals is still hampered by the poor level of control over gene expression, and faces an uphill battle over consumer acceptance. There are several alternatives to HGPs and transgenics, that are more likely to gain world-wide acceptance. Genetic selection can be enhanced by using markers for polymorphic genes that control fat and lean, such as thyroglobulin, or the callipyge gene. Feed additives of natural origin, such as betaine, chromium and conjugated linoleic acid, can improve the fat:lean ratio under specific circumstances. Additionally, 'production vaccines' have been developed, which alter the neuro-endocrine system by causing an auto-immune response. Thus, antibodies have been used to neutralise growth-limiting factors, prolong the half-life of anabolic hormones, or activate hormone receptors directly. Unfortunately, none of these technologies is sufficiently well advanced yet to rival the use of exogenous HGPs in terms of efficacy and reliability. Therefore, further research is needed to find ways to control fat and lean deposition with due consideration of industry needs, animal welfare and consumer requirements.  相似文献

To evaluate the effects of endogenously secreted cortisol on mineral homeostasis and bone metabolism in cows, 4 ovariectomized Holstein cows were infused for 12 h with either an adrenocorticotropic hormone (ACTH) solution (0.5 mg/2 L isotonic NaCl solution per cow) or isotonic NaCl solution in a 2 × 2 crossover design. ACTH infusion stimulated cortisol secretion and increased plasma cortisol concentrations for 18 h (P < 0.001), leading to an elevated plasma glucose concentration until 36 h (P < 0.001). Plasma calcium and magnesium concentrations in ACTH-infused cows fluctuated within normal ranges, whereas hypophosphatemia was observed unequivocally. The biochemical bone resorption markers tartrate-resistant acid phosphatase 5b and hydroxyproline decreased following ACTH infusion (P < 0.001 and P = 0.003, respectively). Similarly, the bone formation marker, bone-specific alkaline phosphatase, decreased continuously until 72 h after the ACTH infusion (P < 0.001). These results demonstrate that increased secretion of cortisol via a 12-h ACTH infusion disrupted homeostasis of inorganic phosphate and suppressed bone metabolism in ovariectomized cows without involving gonadal steroid hormones.  相似文献

The effect of biotin supplementation on various foot lesions and hoof ceramide composition of toe (wall) and sole portions of hooves was studied in crossbred dairy cattle. Biotin supplementation was done for five months in 14 cattle at a farm and the other 14 animals kept as control. A significant decline was observed in heel erosions and sole avulsions along with total disappearance of white line fissures and double soles in the biotin supplemented cattle resulting in decrease in the overall disease score. Thin layer chromatographs of the hoof lipids revealed 11 types of ceramides in sole lipids and 6 types of ceramides in toe (wall) lipids. The ceramides were typed and identified according to their Rf values. A qualitative increase in the density of thin layer chromatographs of sole lipids was observed in biotin supplemented cattle whereas a non-significant difference in density of thin layer chromatographs of toe lipids was observed after supplementation of biotin.  相似文献

The effect of growth and parturition on hair cortisol concentrations of cattle was investigated. Plasma, saliva, and hair (black and white from the shoulders and hip) samples were collected from calves at 6 and 24 weeks old and from dairy cattle at the dry (1 and 2 months prepartum) and lactation (10, 50, 150, and 250 days postpartum) periods. Plasma and saliva cortisol concentrations were lower in 24-week-old calves than those of 6-week-old calves, and hair cortisol concentrations decreased regardless of color and position. In 6-week-old calves, hair cortisol concentrations differed between sampling positions, but this difference was not observed in 24-week-old calves. Plasma and saliva cortisol concentrations increased before parturition until 10 days postpartum then decreased until 50 days postpartum. The same trend was observed in the cortisol concentrations of white hair. Contrarily, cortisol concentrations in black hair remained unchanged and was lower than that in white hair. Hair cortisol concentration can vary greatly depending on the location on the body, hair color, cattle age, or parturition. When this method is used, all of the above factors must be considered.  相似文献

Local and systemic cytokine responses were studied in 3 groups of cattle, with 4 animals each, experimentally infested with Hypoderma lineatum (De Villers) first instars (L1). The first group was undergoing a primary infestation (G-1), the second group was undergoing a secondary infestation (G-2) and the third group was infested for their third consecutive year (G-3). Cattle were infested with 25 L1 deposited on the skin. Blood and skin samples were taken at 0, 6, 12, 48, 96 and 144 h post-infestation (h.p.i.). Interleukin 10 (IL-10), IL-4 and interferon gamma (IFN-γ) production was studied by immunohistochemistry and sandwich ELISAs. IL-4+ cells showed a significant increase at 6 h.p.i. in both reinfested groups (G-2 and G-3) when compared with G-1. In all groups the number of IL-4+ cells decreased significantly at 48 h.p.i. IL-10+ cells increased in G-1 at 6 and 48 h.p.i., whereas in both reinfested groups increased at 12 h.p.i. with a peak at 48 h.p.i. IFN-γ+ cells showed a significant increment at 6 h.p.i. in all groups, followed by a rapid descent at 12 (G-1 and G-2) and 48 h.p.i. (G-3). Penetration of the skin by H. lineatum did not have any significant effect on IFN-γ serum concentrations and, except for IL-10 there were no correlation between local production and serum concentrations of cytokines. The increase of both Th1 (IFN-γ) and Th2-type cytokines (IL-4 and IL-10) indicates that bovine T-cell response during the first phases of the infestation by H. lineatum is apparently a Th0 response.  相似文献

The aims of this study were to examine whether mouflons exposed to constant long and short day photoperiods are able to exhibit an annual cycle of hair growth and moult, and prolactin (PRL) secretion. Mouflon ewes were assigned to three groups of treatment. Ewes were maintained, either under natural photoperiod (control, n=9), or received a series of subcutaneous melatonin implants from December to April (n=8), or were exposed to a constant long day photoperiod (16-h light:8-h dark; 16L:8D) during 18 months (n=7). Blood was collected weekly to determine PRL concentrations, and hair samples were clipped weekly from the base of the neck to measure the length of predominant hair. Under constant long days and with melatonin implants, mouflons expressed an annual rhythm of PRL secretion, even though these treatments modified the times of rise or falling of PRL concentrations throughout the year. Hair growth initiation was almost coincident with the summer solstice in both control and melatonin-implanted mouflons but occurred two months earlier in long day hold mouflons (P<0.001). Long day hold mouflons had a lower hair growth rate than control and melatonin-implanted mouflons (P<0.001), and at the end of the experiment, a shorter hair length (3.4±0.24 cm; P<0.01) than control (4.3±0.17 cm), and melatonin-implanted mouflons (4.2±0.12 cm). Our data support the conclusion that in mouflon, an endogenous circannual rhythm of PRL secretion exists, and that the seasonal cycle of hair growth and moult appears to depend, at least in part, on circulating levels of PRL.  相似文献

Limited data exist on age-related physiological variations in plasma concentrations of cortisol and dehydroepiandrosterone (DHEA) in dogs, despite their potential role in the pathophysiology of ageing. This study examined plasma cortisol and DHEA concentrations and cortisol/DHEA ratio variations, according to age and sex in 311 dogs, aged from two months to 16 years. Before adulthood, DHEA concentrations were higher in peri-pubertal males. During adulthood, cortisol and DHEA were higher in males than females. Among females, DHEA was lower in older dogs, but the decrease was observed at an older age in intact than ovariectomised females. Variations in the cortisol/DHEA ratio inversely reflected those of DHEA. Results indicate that testicles are an important source of DHEA in males, and that DHEA is mainly secreted by the adrenal glands in females. The ovaries’ contribution to circulating DHEA appears to be limited, although it may partially compensate an age-related decrease in adrenal secretion.  相似文献

This study aimed to assess the impact of seasonal thermal stress on oxidative stress, immune response, and stress hormones of lactating dairy cows in subtropical regions with different levels of temperature-humidity index (THI). A total of 32 healthy lactating Holstein dairy cows experienced 4 seasons (8 cows/season). The physiological parameters were categorized into low THI (LTHI, THI = 42.97 ± 0.95) in winter, moderate THI (MTHI, THI = 61.84 ± 0.42) in spring and autumn, and high THI period (HTHI, THI = 86.09 ± 0.23) in summer. The blood samples were collected twice in each season to measure oxidative stress, inflammatory and hormonal parameters. Our results showed THI had a positive correlation with the rectal temperature (R2 = 0.821, P < 0.001) and respiratory rate (R2 = 0.816, P < 0.001). Dry matter intake, milk yield and fat percentage also significantly differed among groups (P < 0.05). Compared with the MTHI group, the LTHI group exhibited a significant increase in malondialdehyde (MDA) level (P < 0.001), and the HTHI group displayed a significant increase in levels of cortisol, interleukin (IL)-10, IL-1β and tumor necrosis factor-α (P < 0.001). Opposite changes in serum endotoxin and immunoglobulin G levels were observed with the increasing THI (P < 0.001). LTHI notably increased the triiodothyronine level, although the thyroxine level was reduced by LTHI and HTHI compared with the MTHI group. In conclusion, LTHI and HTHI conditions may induce different degrees of oxidative stress, inflammation response, and stress hormone imbalances on lactating dairy cows, therefore environmental management is necessary for the health of dairy cows in extreme weather conditions.  相似文献

Pregnancy is a physiology state accompanied by high energy and oxygen demand that may lead to increased level of oxidative stress and development of metabolic and reproductive disorders in pregnant water buffaloes. In the present study, the alterations in serum cortisol and erythrocyte lipid peroxides and superoxide dismutase activities were examined in 28 pregnant water buffaloes supplemented with antioxidant nutrients, Vitamin E and selenium. Another 14 buffaloes did not receive any treatment during pregnancy to serve as negative control. Results of the present study suggested that pregnancy is associated with oxidative stress and supplementation of vitamin E and selenium may be beneficial by alleviating oxidative stress in water buffaloes.  相似文献

The association between sole ulcer found at routine claw trimming 2–6 months after calving and reproductive, health and productive traits in the same lactation, was investigated in an observational study of 2368 dairy cows in 102 Swedish herds. The data were analysed by mixed multivariable linear-, logistic- and Poisson-regression modelling at the cow level, accounting for clustering within herd. Associations were found between sole ulcer and first-service conception risk in the first study year (OR 0.59), calving interval (2% longer), treatment for anoestrus (OR 1.61), and 305-day milk yield (479 kg ECM higher). No associations could be shown between sole ulcer and first-service conception risk in the second study year, the number of services per conception, clinical mastitis, high milk somatic-cell counts or culling.  相似文献

The temporal pattern and sex effect of immune and stress hormone responses to a lipopolysaccharide (LPS) challenge were assessed using a pig model. Secretion of the pro-inflammatory cytokines tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6 increased in a time-dependent manner following LPS infusion. There was also a time-dependent increase in secretion of the stress-related hormones cortisol, epinephrine (E), and norepinephrine (NE) following LPS, with peak concentrations attained within 30 min. The magnitude of the TNF-α and IL-1β responses were both positively associated (P < 0.05) with the magnitude of cortisol response following LPS, whereas serum IL-1β and IL-6 were positively correlated with the magnitude of E and NE responses following LPS. Acute-phase protein production was also time-dependently increased following LPS. The concentration of immune cells in circulation was decreased (P < 0.05) at 5.5 h post-LPS and negatively correlated with pro-inflammatory cytokine production. By 24 h post-LPS, immune cell counts increased (P < 0.05) and were positively associated with both pro-inflammatory cytokine and stress hormone production. The amplitude of pro-inflammatory cytokine response following LPS was affected (P < 0.05) by sex classification; however, the magnitude of elevated cytokine concentrations was not. The magnitude of the NE response, but not of the E and cortisol responses, to LPS was influenced by sex (P < 0.05). Similar to the pro-inflammatory cytokines, the magnitude of exposure to the stress hormones following LPS was not influenced by sex. The production of serum amyloid A (SAA) was influenced by sex, with barrows producing more SAA than gilts at 24 h post-LPS (P < 0.05). Chronic elevation of glucocorticoid concentrations is detrimental to health. We investigated effects of chronic increase in plasma cortisol concentrations on energy balance and endocrine function in sheep. Because food intake and reproduction are regulated by photoperiod, we performed experiments in January (JAN) and August (AUG), when appetite drive is either high or low, respectively. Ovariectomized ewes were treated (intramuscularly) daily with 0.5 mg Synacthen Depot® (synthetic adrenocorticotropin: ACTH) or saline for 4 wk. Blood samples were taken to measure plasma concentrations of cortisol, luteinising hormone (LH), follicle-stimulating hormone (FSH), growth hormone (GH), leptin, insulin, and glucose. Adrenocorticotropin treatment increased concentrations of cortisol. During JAN, treatment reduced food intake transiently, but increased food intake in AUG. Leptin concentrations were reduced and glucose concentrations were greater in AUG, and insulin concentrations were similar throughout the year. Treatment with ACTH increased leptin concentrations in AUG only, whereas insulin concentrations increased in JAN only. Synacthen treatment increased glucose concentrations, with a greater effect in JAN. Changes in truncal adiposity and ACTH-induced cortisol secretion were positively correlated in JAN and negatively correlated in AUG. Treatment reduced the plasma LH pulse frequency in JAN and AUG, with an effect on pulse amplitude in JAN only. Treatment did not affect plasma GH or FSH concentrations. We conclude that chronically elevated cortisol concentrations can affect food intake, adiposity, and reproductive function. In sheep, effects of chronically elevated cortisol concentrations on energy balance and metabolism depend upon metabolic setpoint, determined by circannual rhythms.  相似文献
