为快速测定黄瓜叶片面积,开发了基于数字照片解析技术的叶片面积测定法,通过与坐标纸法、复印比重法对比,分析数字照片解析法的准确性与稳定性。结果表明:数字照片解析法与坐标纸法测定的结果无显著差异(配对检验P(p>|t|)=0.872>0.05),复印比重法测定的结果较坐标纸法显著偏高。数字照片解析法较坐标纸法更为便捷、准确,可作为一种新的叶面积测定法推广使用。  相似文献

一种简单高效的聚丙烯酰胺凝胶DNA回收方法   总被引:2,自引:1,他引:1  
本文介绍了一种简单的聚丙烯酰胺凝胶DNA回收方法(直取法),通过与传统煮沸法比较以证明直取法回收聚丙烯酰胺凝胶DNA的高效性。以木薯cDNA为材料,将直取法与常规煮沸法从实验方法、杂带污染率以及回收扩增率三个方面进行比较。实验结果表明:直取法较煮沸法简单、快速;煮沸法的杂带污染率为59.09%,直取法为43.08%;煮沸法的平均产物量约为40 ng/μL,直取法约为60 ng/μL。研究结果表明,直取法相对煮沸法简单,快速,扩增效率高,杂带污染几率小。  相似文献

为了获得板栗子叶总RNA提取的理想方法,为后续的分子生物学研究打下基础,以板栗幼果子叶为材料,采用改良CTAB法、硼砂-CTAB法、酚-SDS法、皂土法和2种植物总RNA提取试剂盒共6种方法提取板栗子叶总RNA。结果表明,除硼砂-CTAB法外,其他5种方法均能获得板栗子叶总RNA。但是,与酚-SDS法、皂土法和2种植物总RNA提取试剂盒相比较,改良CTAB法提取的板栗子叶总RNA完整性好,纯度高,且无DNA污染,能够满足cDNA-AFLP等分子生物学研究的要求。  相似文献

以250份宁夏粳稻为材料,根据7个农艺性状的遗传多样性,研究粳稻核心种质的构建方法.采用混合线性模型统计分析方法无偏预测材料各性状的基因型值,根据2种遗传距离(马氏距离和欧氏距离),3种取样方法(多次聚类随机取样法、多次聚类优先取样法、多次聚类偏离度取样法),8种聚类方法(最短距离法、最长距离法、中间距离法、重心法、类平均法、可变类平均法、可变法、离差平方和法),用均值、方差、极差、变异系数及秩次评价不同遗传距离、取样方法、聚类方法的优劣.结果表明,马氏距离优于欧氏距离;偏离度取样法优于优先取样法和随机取样法,聚类方法最好的是最短距离法、可变类平均法,适宜取样比例是15%.用马氏距离、偏离度取样法、15%取样比例、最短距离聚类法构建了37份宁夏粳稻核心种质.  相似文献

Based on the practical project a study on analysis methods for rock slope foundation stability of highrise-building project criterion transfer coefficient method, rotation limit equilibrium method (Sweden slices method) and finite element method was carried out.The authors found that safety factor of criterion transfer coefficient method is larger than that of rotation limit equilibrium method, but is similar to the results of finite element method. So, when rockslope foundation stability of highrise building is to be analysed,the rotation limit equilibrium method is suggested to use.  相似文献

目的:意在找出适合秸秆纤维素提取的方法。[方法]:分别采用氢氧化钠-醋酸-亚氯酸钠-丙酮法、高压蒸煮法、硝酸-氢氧化钠法、氢氧化钠-氢氧化钠法、硝酸-乙醇法、过氧酸体系法等方法提取秸秆纤维素,采用重铬酸钾-硫酸亚铁铵法测定纤维素的含量,以纤维素含量与所需时间为评价指标,选出最适合提取玉米秸秆纤维素的方法。[结论]:高压蒸煮法和硝酸-乙醇法较优越,具有操作简单、速度快捷、提取率较高的优点。高压蒸煮法处理量大,适合工业生产需要,硝酸-乙醇法提取的纤维素量少,但其含量高,适用于科研机构及相关部门。  相似文献

Taking into the feature of computer computing method, using Stress solution of elasticity theory and according to different method of solving of system of second order ordinary differential equations , this article provides two numerical value computing method as for the stress and deformation of the turbine impeller which is within the scope of elasticity . One combines the initial value computing method of system of ordinary differential equations (Runge-Kutta Method) with optimization method, another combines the boundary value computing method of system of ordinary differential equations difference method-with optimization method and three points interpolation method . The proposed method can eliminate the deficiencies of Secondary Calculation method and is particularly suited for programmable computer-based solution . The sample show that results gotten by two methods is nearly equal to the precise results,so they are practicable. They completes the quick and precise calculating of stress and deformation . A new efficient method named matrix method is presented based on the traditional calculation methods of stabilizing force. The method is derived from the thrust force method and designed to address the deficiency of traditional method. It cannot only get the best pile location with minimum stabilizing force when is applied to the design calculation of single row anti slide piles, but also be applied to the optimal calculation of multi row anti slide piles. The cases comparing results show the method is more reasonable. For major projects of landslide control, the combination of matrix method and finite element method can make the result safer and more accurate.  相似文献

样品处理是双向电泳实验最基础也是最关键的环节之一,样品制备成功与否直接关系到双向电泳图谱的质量和蛋白质组信息的完整性。应用于双向电泳中提取植物全蛋白的常用方法有:TCA-丙酮沉淀法、Tris-HC1法、酚法、Trizol沉淀法、Tris-丙酮-酚法、尿素/硫脲提取法。不同的方法适合不同特征的植物样品,比较了用于双向电泳的六种植物全蛋白提取方法的优缺点,并对每种方法适用的植物组织作了简要概述。  相似文献

为了选择一种省时省力、简单有效的皱边石杉内生真菌菌丝体总DNA提取方法,采用氯化苄法提取其总DNA,对比研究石英砂研磨与超声波振荡2种破壁方式对其DNA的提取效能。结果表明,采用石英砂研磨法提取的DNA,其点样孔附近残余杂质较多、拖尾现象严重;超声波破壁法提取的DNA质量总体较好,DNA条带在23.1 kb处集中,多糖、蛋白质等杂质污染少,拖尾现象轻微。比较而言,超声波破壁法可快捷、高效提取皱边石杉内生真菌菌丝体DNA,适于植物内生真菌DNA大规模快速提取,尤其对质地坚硬的菌落较石英砂研磨更有效。提取的DNA经琼脂糖凝胶电泳、ITS序列扩增,表明超声波破壁法提取的DNA与石英砂研磨法一样,均可获得良好的PCR扩增结果。  相似文献

以枳壳(酸橙)叶片为试验材料,分别采用改良CTAB法、CTAB-硅珠法、SDS法、高盐低pH值法和周世良法5种方法提取枳壳基因组DNA,并通过紫外分光光度计、琼脂糖凝胶电泳及SSR-PCR扩增3种方法对所提取的DNA样品进行检测。结果表明,5种方法中SDS法提取的DNA浓度最高(平均为900 ng/ul),纯度最好(OD260/OD280平均值为1.90),且SSR-PCR扩增结果与紫外分光光度计和凝胶电泳检测结果基本一致。因此,SDS法是枳壳DNA最佳提取方法,可用于分子检测试验。  相似文献

This paper presents a new hierarchical and coordinative control method for reactive power optimization in large-scale interconnected power systems. It combines the interconnection forecast method for hierarchical control in large-scale steady state industrial systems with the incremental linear programming method for reactive power optimization in real time. The results of applying the method to power system examples show that the method not only solves the problem of high dimension in order, but also keeps the advantages of the incremental linear programming method. The proposed method is also suitable for the security analysis of reactive power correction in large-scale interconnected power systems.  相似文献
