Authenticity assessment of gamma-decalactone (1) and delta-decalactone (2) from peach (Prunus persica var. persica), apricot (Prunus armeniaca), and nectarine (Prunus persica var. nectarina) was performed using gas chromatography-isotope ratio mass spectrometry (GC-IRMS) in the combustion (C) and pyrolysis (P) mode. In addition, commercially available synthetic (nature-identical) 1 and 2 as well as biotechnologically produced samples (declared to be "natural") were characterized by their delta(2)H(V)(-)(SMOW) and delta(13)C(V)(-)(PDB) values. For the Prunus fruits under study, rather narrow ranges of delta(13)C(V)(-)(PDB) and delta(2)H(V)(-)(SMOW) data of 1, varying from - 34.6 per thousand to - 38.4 per thousand and -160 per thousand to -206 per thousand, respectively, were obtained. Synthetic references of 1 showed delta(13)C(V)(-)(PDB) and delta(2)H(V)(-)(SMOW) data ranging from -27.4 per thousand to -28.3 per thousand and -151 per thousand to -184 per thousand, respectively. Samples of 1 declared to be "natural" exhibited ranges from -28.1 per thousand to -29.2 per thousand and -192 per thousand to -286 per thousand for delta(13)C(V)(-)(PDB) and delta(2)H(V)(-)(SMOW), respectively. For 2 from peach, apricot, and nectarine, delta(13)C(V)(-)(PDB) values ranging from -34.0 per thousand to -37.9 per thousand were determined; the delta(2)H(V)(-)(SMOW) values ranged from -171 per thousand to -228 per thousand. The delta(13)C(V)(-)(PDB) and delta(2)H(V)(-)(SMOW) data for synthetic 2 were -28.2 per thousand and -171 per thousand, respectively, that is, similar to those of 2 from "natural" origin, ranging from -27.7 per thousand to -30.1 per thousand and -185 per thousand to -230 per thousand for delta(13)C(V)(-)(PDB) and delta(2)H(V)(-)(SMOW), respectively. GC-C/P-IRMS allowed clear-cut analytical differentiation of the synthetic and "ex-plant" origin of 1 and 2, whereas narrow ranges of delta(13)C(V)(-)(PDB) and delta(2)H(V)(-)(SMOW) data were found for samples of synthetic and "natural" origin. 相似文献

Based on (2)H/(1)H ratio measurements of commercial synthetic and "natural" references, the recently developed on-line gas chromatography pyrolysis isotope ratio mass spectrometry (HRGC-P-IRMS) technique was used to determine the delta(2)H(SMOW) values of the flavor compounds decanal, linalool, and linalyl acetate, as well as those of E-2-hexenal and E-2-hexenol in foods and essential oils. In preceding model studies, the influence of sample preparation steps (simultaneous distillation extraction, SDE; solvent extraction, SE; liquid liquid extraction, LLE) on the delta(2)H values was found to be negligible. For decanal, the typical (2)H abundance, with higher content of (2)H for synthetic material (delta(2)H(SMOW) from -90 to -156 per thousand) and lower (2)H content for natural references (delta(2)H(SMOW) from -138 to -262 per thousand) was observed. Although the delta(2)H data recorded for linalool did not allow one to distinguish between synthetic (delta(2)H(SMOW) from -207 to -301 per thousand) and natural (delta(2)H(SMOW) from -234 to -333 per thousand) materials, the situation was somewhat more encouraging for linalyl acetate; delta(2)H(SMOW) values from -199 to -239 per thousand and from -213 to -333 per thousand were found for synthetic and natural samples, respectively. E-2-Hexenal and E-2-hexenol showed clear-cut origin-dependent differences in their (2)H/(1)H ratios; that is, delta(2)H(SMOW) values from -14 to -109 per thousand and from -263 to -415 per thousand as well as from -41 to -131 per thousand and from -238 to -348 per thousand were recorded for products from synthetic and natural origins, respectively. 相似文献

Authenticity assessment of trans-anethole is deduced from (2)H/(1)H and (13)C/(12)C isotope ratios, determined by gas chromatography-isotope ratio mass spectrometry (GC-IRMS). For that purpose, self-prepared anise and fennel oils, and synthetic and "natural" samples of trans-anethole, as well as commercially available anise and fennel oils have been investigated. Authenticity ranges of (2)H/(1)H and (13)C/(12)C isotope ratios of trans-anethole were defined. Scope and limitations of the applied online GC-IRMS techniques are discussed. 相似文献

Thiolysis of a wine tannin fraction yielded trihydroxylated flavanol units (as previously observed in grape skins) in addition to the well-known procyanidins (dihydroxylated units), usually described in the literature for grape condensed tannins. To determine how they occur in condensed tannins, the wine fraction was analyzed by liquid chromatography coupled to electrospray ionization mass spectrometry. Thus, various series of ion peaks containing a variable number of trihydroxylated units were detected as monocharged ions from dimers up to pentamers. From pentamers, oligomers were found as doubly charged ions. Heptamer species corresponded to the highest mass detected. These results showed that wine condensed tannins consist of, besides procyanidins, mixed tri- and dihydroxylated flavanol units and also of pure trihydroxylated flavanol units. These new data should be taken into account to interpret organoleptic properties of wines. 相似文献

The qualitative and quantitative composition of free amino acids in a typical Finnish peat bog at various depths down to 5.3 m below the surface was studied using capillary gas chromatography and mass spectrometry. Sixteen amino acids were identified at each depth: α-alanine, β-alanine, glycine, valine, leucine, proline, isoleucine, serine, threonine, glutamic acid, aspartic acid, phenylalanine, tyrosine, γ-aminobutyric acid, ornithine and lysine. Their amounts decreased markedly at a depth of 40–100 cm. The total amount of amino acids varied between 0.6 and 5.6 g kg?1 dry matter (i.e. 0.06–0.56%) depending on the depth. The proportion of neutral amino acids was greatest at all depths studied, except at the surface layer where it ranged between 41 and 72% by mass. The acidic amino acids decreased with depth from 56 to 23% of the total. The proportion of aromatic amino acids was very small, 3.2–5.5% by mass. In samples from aerobic conditions, where the microbial production of free amino acids was the greatest, α-ala, gly, glu and asp were most abundant. In peat from anaerobic conditions, where the microbiological activity was low, the proportion of the most chemically stable amino acid was exceptionally high. This may have been because glycine was a degradation product of other amino acids or peptides. Peat type and degree of decomposition had a strong influence on the total amount of free amino acids and their qualitative composition. 相似文献

A rapid multiresidue method for the simultaneous determination of 25 fungicides and insecticides in soil was developed. Soil samples are extracted by sonication with a water-acetonitrile mixture, and the pesticides are partitioned into dichloromethane. Final determination was made by gas chromatography (GC) with nitrogen-phosphorus detection (NPD). Confirmation analysis of pesticides was carried out by GC-MS in the selected ion monitoring (SIM) mode. The identification of compounds was based on retention time and on comparison of the primary and secondary ions. The average recovery by the GC-NPD method obtained for these compounds varied from 68.5% to 112.1% with a relative standard deviation between 1.8% and 6.2%. The GC-NPD method presents good linearity over the range assayed 50-2000 microg/L, and the detection limit for the pesticides studied varied from 0.1 to 10.4 microg/kg. The proposed method was used to determine pesticide levels in soil samples from experimental greenhouse pepper cultivation. 相似文献

During an evaluation of the gas chromatography/mass spectrometry (GC/MS) confirmatory procedure of Lynch and Bartolucci for pyrantel residues in swine tissues, we developed a GC flame ionization method for quantitating pyrantel residues in extracts of swine liver. The method was subjected to trial principally in the laboratories of Biospherics, Inc., using control liver, fortified control liver, and incurred liver tissue samples. Although the method does not meet all of the current Food and Drug Administration criteria, it compares favorably to the official determinative method. Portions of the same extract can be used for quantitation and for GC/MS confirmation, true recoveries appear to be slightly higher, and an internal standard is not required. The precision of this method equals or exceeds that of the official determinative method. 相似文献

A method using gas chromatography/chemical ionization mass spectrometry (GC/CIMS) for the determination of daminozide residues in apples has been developed. Daminozide was separated from the sample matrix by water extraction and cation exchange, converted to the methyl ester by treatment with HCl-methanol, and determined by GC/CIMS using succinonitrile as an internal standard. The detection level was 0.05 ppm. Recoveries were 92-104% from apples spiked at the 0.05-0.5 ppm levels. Of the 25 apple samples analyzed, only 2 were positive for daminozide (1.04 and 0.32 ppm). 相似文献

An on-line solid-phase extraction (SPE) following a liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) method was established for the simultaneous analysis of bisphenol A (BPA), nonylphenol (NP), and octylphenol (OP) in cereals (including rice, maize, and wheat). The target compounds were extracted by acetonitrile, purified by an automated on-line SPE cartridge, and analyzed by LC-MS/MS under the negative-ion mode. Mean recoveries fortified at three concentration levels ranged from 81.6 to 115.7%, and the coefficient of variation ranged from 4.6 to 19.9% (n = 6). The limits of quantification (LOQs) of the method were 0.5, 0.5, and 0.25 μg/kg for BPA, NP, and OP, respectively, in both rice and maize, while the LOQs in wheat were 0.5, 1.25, and 0.5 μg/kg for BPA, NP, and OP, respectively. This method was applied in the analysis of rice, maize, and wheat from a local market. As a result, NP occurred in all cereal samples at the concentration range of 9.4-1683.6 μg/kg and BPA was detected in a few samples. 相似文献

The present work describes a solid-phase microextraction (SPME) gas chromatography-tandem mass spectrometry (MS/MS) method to quantify 24 pesticides in fortified white wine and fortified red wine. In this study "fortified wine" refers to a wine in which fermentation is arrested before completion by alcohol distillate addition, allowing sugar and alcoholic contents to be higher (around 80-100 g/L total sugars and 19-22% alcohol strength (v/v)). The analytical method showed good linearity, presenting correlation coefficients (R(2)) ≥ 0.989 for all compounds. Limits of detection (LOD) and quantitation (LOQ) in the ranges of 0.05-72.35 and 0.16-219.23 μg/L, respectively, were obtained. LOQs are below the maximum residue levels (MRL) set by European Regulation for grapes. The proposed method was applied to 17 commercial fortified wines. The analyzed pesticides were not detected in the wines tested. 相似文献

Capillary gas chromatography-electron impact mass spectrometry is used to confirm the presence of levamisole in cattle and swine livers at the 0.1 ppm tolerance level. Use of a fused silica capillary column from injector to detector solved chromatographic problems encountered with the analyte. Of the mass spectrometric techniques evaluated, electron impact mass spectrometry provided the most satisfactory data for a confirmatory method. Recoveries from swine and cattle livers fortified at 0.1 ppm averaged 74.9 and 72.7%, respectively, indicating potential utility of this methodology as a quantitative method. Apparent levamisole residues in control livers were less than 0.01 ppm. 相似文献

A method is described for determining ethyl carbamate at low microgram/kg levels in several types of alcoholic beverages by capillary column gas chromatography with Hall electrolytic conductivity detection and confirmation by mass spectrometry. Samples are diluted to obtain a uniform concentration of ethanol (ca 10%) then saturated with NaCl and extracted with methylene chloride. Extracts are evaporated to a small volume and injected in ethyl acetate solution for chromatographic analysis. The method was evaluated by 5 laboratories, 4 employing the Hall detector and one using mass spectrometric detection. Overall between-laboratory mean percent recoveries were: wine, 85.3 +/- 21.0% coefficient of variation (CV) (spiking level 20-45 micrograms/kg); sherry, 83.8 +/- 16.1% CV (spiking level, 81-142 micrograms/kg); whiskey, 79.5 +/- 13.9% CV (spiking level 127-190 micrograms/kg); and brandy, 85.0 +/- 12.5% CV (spiking level 297-446 micrograms/kg). Mass spectrometric results agreed well with the Hall results for all commodities. Detection limits were about 5 micrograms/kg for the Hall detector and about 0.5 microgram/kg for mass spectrometric detection. 相似文献

A new method based on comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC×GC-ToF MS) has been developed for the first time for the analysis of complex mixtures of disaccharides previously converted to their trimethylsilyl oximes (TMSO). Among the different experimental parameters considered for optimization, both the column set combination and the dimensions of the second-dimension column were found to be the most significant with regard to the complete resolution of structurally similar disaccharides. Application of the optimized method to honey analysis allowed the separation of most of the honey disaccharides previously described in the literature. Furthermore, 12 other unknown disaccharides have been separated by this method and characterized from their mass spectral data. 相似文献

A method for quantitative analysis of acrylamide has been developed for use with headspace solid-phase microextraction (SPME). In the method, acrylamide undergoes silylation with N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA) to form the volatile N,O-bis(trimethylsilyl)acrylamide (BTMSA). Once formed, BTMSA is readily extracted from the headspace over the silylation reaction using a 100 microm poly(dimethylsiloxane) SPME fiber. A series of experiments was undertaken to optimize the amount of BSTFA, the silylation reaction temperature, the silylation reaction duration, and SPME sampling duration to maximize the analytical sensitivity for BTMSA. Acrylamide levels were quantified relative to a [13C3]-acrylamide internal standard using gas chromatography/ion-trap mass spectrometry (GC/MS) in the single ion monitoring mode. An analytical working curve was constructed and found to be linear over the 4 to 6700 ppb acrylamide range investigated with a limit of detection of 0.9 ppb. The native acrylamide levels of three commercial cereals were measured using the optimized analytical method. Quantitative standard additions of acrylamide to the cereal matrixes demonstrated complete recovery of the spiked acrylamide. 相似文献

Yeast autolysis affects membrane stability and induces a release of vacuolar enzymes into the cell cytoplasm. Consecutively, it was important to study the evolution of sterol content in Saccharomycescerevisiae for a fourteen day period of accelerated autolysis. Unesterified and esterified sterols were analyzed both in the biomass and in the autolysis medium. Ten sterols were identified by gas chromatography/mass spectrometry. A second group of six sterols was separated and partially characterized. Among the first group of 10 sterols, a dehydroergosterol was identified as ergosta-5, 7,9(11),22-tetraen-3beta-ol, not yet charaterized in S. cerevisiae. Yeast autolysis induced a decrease of esterified sterol content, especially first intermediates in the sequence of the ergosterol biosynthesis, as zymosterol. In contrast, the yeast autolysis resulted in the release of a low quantity of sterols into the medium. At the end of the fourteenth day of autolysis, 0.015% of the total sterol content of the initial biomass was found in the medium. 相似文献

A procedure has been developed and validated for measuring the concentration of pentobarbital residues in dry, extruded animal feed in the range of 3-200 ng/g (ppb) with an estimated limit of quantitation of 2 ppb. The method was developed for surveillance purposes: to measure the concentration of euthanizing agent which might be present in feeds incorporating rendered products which themselves might include some fraction of euthanized animals. A previously published qualitative procedure was modified by adding isotopically labelled pentobarbital as an internal standard. Dry feed was ground and extracted with methanol. The extract was loaded on a mixed-mode (C-18, anion exchange) solid-phase extraction cartridge designed for barbiturate residues. Pentobarbital was eluted and derivatized for gas chromatography/mass spectrometry in positive ion chemical ionization mode. Quantitation was based on the ratio of dimethyl-pentobarbital MH+ (m/z 255) vs dimethyl-pentobarbital-d(5) (m/z 260) in standards and extracts. Accuracy ranged from 112% at 3 ppb to 96% at 200 ppb, with relative standard deviations ranging from 4% at 3 ppb to 2% at 200 ppb. 相似文献

Benzene, a carcinogen that can cause cancer in humans, may form at nanogram per gram levels in some beverages containing both benzoate salts and ascorbic or erythorbic acids. Through a series of reactions, a hydroxyl radical forms that can decarboxylate benzoate to form benzene. Elevated temperatures and light stimulate these reactions, while sugar and ethylenediaminetetraacetic acid (EDTA) can inhibit them. A headspace gas chromatography/mass spectrometry method for the determination of benzene in beverages was developed and validated. The method was used to conduct a survey of 199 soft drinks and other beverages. The vast majority of beverages sampled contained either no detectable benzene or levels below the U.S. Environmental Protection Agency's drinking water limit of 5 ng/g. Beverages found to contain 5 ng/g benzene or more were reformulated by the manufacturers. The amount of benzene found in the reformulated beverages ranged from none detected to 1.1 ng/g. 相似文献

Abstract The isotopic signal provided by differential discrimination against atmospheric carbon dioxide (13CO2) by C3 and C4 plant photosynthetic pathways is being widely used to study the processes of carbon (C) fixation, soil organic matter formation, and mineralization in nature. These studies have been facilitated by the availability of automated C and nitrogen (N) combustion analyzers (ANCA) combined with continuous flow isotope ratio mass spectrometers (CFIRMS). Analysis of 13CO2 in these instruments requires consistent sample mass for best precision, a requirement that is easily satisfied for soil and tissue samples by adjusting sample weight. Consistent CO2 sample size is much more difficult to achieve using gas handling systems for samples of headspace gases when CO2 concentrations vary widely. Long storage of gaseous samples also is difficult. Extended respiration studies are most easily conducted by trapping CO2 in alkali and conversion to an insoluble carbonate. Thermal decomposition of the carbonate in an on‐line ANCA allows consistent and optimal CO2 sample mass to be obtained. The use of precipitated carbonates also facilitates storage of samples and enables full automation of sample analysis using an ANCA interfaced to a CFIRMS. Calcium (Ca), strontium (Sr), and barium (Ba) carbonates were tested. Strontium carbonate (SrCO3) with the addition of vanadium pentoxide (V2O5) as a combustion catalyst was found most suitable. 相似文献

An electron capture gas chromatographic method for the determination of sulfamethazine was modified to separate and quantitate simultaneously sulfamethazine and 2 of its metabolites, N4-acetylsulfamethazine and desaminosulfamethazine. The modified method was applied to incurred residues in a veal calf depletion study and to incurred residues in swine tissues. With capillary column gas chromatography-positive ion chemical ionization mass spectrometry, confirmation of the identities of incurred desaminosulfamethazine, N4-acetylsulfamethazine, and sulfamethazine in tissues was obtained from a single injection. 相似文献
