The objective of this study was to investigate the growth- and breed-related changes of muscle fiber characteristics in cattle and their importance to meat quality. Four cattle breeds with different growth impetus and muscularity were reared and slaughtered under experimental conditions. German Angus as a beef type, Galloway as a hardy type, Holstein Friesian as a dairy type, and double-muscled Belgian Blue as an extreme type for muscle growth were used. Between 5 and 17 bulls of each breed were slaughtered at 0, 2, 4, 6, 12, 18, and 24 mo of age. Muscle fiber traits were determined and classified by computerized image analysis, and several measures of meat quality were also determined, including shear force value, meat color, and i.m. fat content. The postnatal growth of semitendinosus muscle in cattle was characterized by a nearly 10-fold increase of muscle fiber area from birth to 24 mo of age. In the first few months after birth, a transformation of type IIA fibers into IIB fibers was found, whereas type I fibers were nearly unaffected by age. The apparent total muscle fiber number of semitendinosus muscle did not increase during postnatal life. These results confirm that the fiber number is determined in embryonic development. Throughout the study, the double-muscled Belgian Blue (BBDM) bulls had almost twice the fiber number of the other breeds, emphasizing a more extensive hyperplasia of muscle fibers during embryonic development in BBDM compared with the other three breeds. The apparent number of type I fibers was, however, not affected by breed, which suggests that the additional fibers found in BBDM postnatally were type IIB and IIA fibers. We did not find significant differences in muscle fiber total number, muscle fiber type frequencies, or meat quality characteristics among breeds, with the exception of BBDM. Having pooled the four breeds, paler meat was related to a higher frequency of type IIB fibers, a lower area of type IIA and type I fiber, and a higher total muscle fiber number. These findings based on data of double muscling give us some hints for biological causes for the variation of meat quality. Further investigation, in particular within each breed, is necessary to identify the superior fiber traits for bovine meat production.  相似文献

The objective of this study was to investigate the changes in muscle fiber bundles of cattle of different breeds during growth. Different numbers of muscle fibers are surrounded by connective tissue to form bundles macroscopically visible as meat fibers or meat grain, a common meat quality trait. To determine the influence of breed and age on morphological characteristics of muscle fiber bundles, 4 cattle breeds with different growth impetus and muscularity were reared and slaughtered under experimental conditions. German Angus, a typical beef cattle; Galloway, a smaller beef type; Holstein Friesian, a dairy type; and double-muscled Belgian Blue, an extreme type for muscle growth, were used. Between 5 and 15 bulls of each breed were slaughtered at 2, 4, 6, 12, or 24 mo of age, and slices of semitendinosus muscle were removed. Muscle structure characteristics were determined by computerized image analysis. During growth, the muscle cross-sectional area enlarged (P < 0.001) about 5-fold in double-muscled Belgian Blue bulls and about 4-fold in the other breeds. This was a result of the enlargement (P < 0.001) of primary bundles and muscle fibers. The bundle size was similar (P > or = 0.15) in bulls of German Angus and Galloway in all age groups and was doubled (P < 0.001) in double-muscled Belgian Blue animals from 4 mo of age on. The Holstein Friesian bulls had the smallest (P < 0.001) muscle fiber bundles at 24 mo of age. The number of muscle fibers per bundle and the number of bundles per muscle remained nearly constant (P > 0.05) during growth. This supports the existing view that the structure of the muscle is already fixed in prenatal life. The double-muscled Belgian Blue bulls showed a more than 2.5-fold greater (P < 0.001) number of muscle fibers per primary bundle compared with the other breeds investigated. The larger muscle fiber bundles led to a smaller amount of connective tissue per muscle area in double-muscled cattle. The coarser grain of meat in double-muscled Belgian Blue bulls and in older animals was not related to greater shear force values.  相似文献

Serial ultrasound estimates of backfat thickness and marbling score were obtained on two groups of steers fed high-energy rations for averages of 166 and 148 d. These measurements were used to develop mathematical models that described the changes in those two attributes as functions of days on feed. An exponential model best described the increase in backfat thickness, and the rate coefficients were .0117 and .0096 for the two groups (doubling times = 59 and 72 d, respectively). A modified power function was better than either a linear or exponential model for describing the development of marbling. In this model, marbling increased very slowly, approximately one degree every 100 d, in the interval from low Select to low Choice but then increased at faster rates as initial marbling scores became higher. Ultrasound backfat measures could be used to predict days to reach a target carcass backfat level with an average error of 30 d or less when backfat thickness averaged more than 3 mm at evaluation. Projections from measurements taken when cattle were started on feed were not accurate from one group, which averaged only 1.7 mm backfat at that time. Projections of carcass marbling improved as evaluation date neared slaughter date. Projections from the initial evaluation date allowed tentative categorization of candidates for Choice or not Choice but were only 64% accurate. Projections from evaluations made later in the feeding period exceeded 75% accuracy in distinguishing Choice from Select. Only small relationships (r2 = .07 or .16) existed between carcass backfat thickness and marbling score. Results indicate that ultrasound estimates of backfat and marbling made during the feeding period can be used to predict carcass merit at slaughter.  相似文献

A subjective, but repeatable, scoring system was devised to quantitate ultrasound speckle in tomograms of the longissimus muscle over the 12th rib. Speckle scores in 11 of 14 groups of cattle (n = 619) were highly correlated (P less than .001) with carcass marbling score. Live animal speckle scores classified carcasses as Select or Choice with 77% accuracy. Similar accuracy was achieved as much as 148 d before slaughter by adjusting for the regression of speckle scores with days on feed. Marbling scores were not distributed normally with both positive skewness and kurtosis (P less than .001). The procedure exploited an unmodified low-cost ultrasound system; scores were estimated instantly from the monitor image. This ability to predict carcass grade at distant dates might be incorporated into a strategy to cluster feedlot cattle into outcome groups for more effective marketing. The results suggest that mathematical analysis of the ultrasound radio frequency signal or pixel map statistics might reveal parameters that could improve accuracy and precision of the method and, conceivably, could predict specific attributes of beef palatability.  相似文献

We investigated changes in structures and mechanical properties of the intramuscular connective tissue during the fattening of Japanese Black steers, using the cell maceration method for scanning electron microscopy. During the early fattening period, from 9 to 20 mo of age, collagen fibrils of the endomysium in longissimus muscle associated more closely with each other, and collagen fibers in the perimysium increased in thickness and their wavy pattern became more regular. These changes were closely related to the increase in mechanical strength of the intramuscular connective tissue and resulted in a toughening of the beef during the period. The shear force value of longissimus muscle decreased after 20 mo of age, concomitantly with the rapid increase in the crude fat content. Scanning electron micrographs of the longissimus muscle dissected from 32-mo-old steers clearly showed that the adipose tissues were formed between muscle fiber bundles, that the honeycomb structure of endomysia was partially broken, and that the perimysium separated into thinner collagen fibers. In semitendinosus muscle, in which the crude fat content was lower (P<.05) than that in longissimus muscle, the structure of the intramuscular connective tissue remained rigid at 32 mo of age. The shear force value of the muscle increased even in the late fattening period, from 20 to 32 mo of age. Thus, the development of adipose tissues in longissimus muscle appears to disorganize the structure of the intramuscular connective tissue and contributes to tenderization of highly marbled beef from Japanese Black cattle during the late fattening period.  相似文献

The deposition of fat in muscle, recognized by the consumer as marbling, is an important meat quality trait. The objective of the study was to provide additional insights into the quantitative extent of marbling by means of computer image analysis. Fifty-one F(2) generation German Holstein and Charolais crossbreed cattle, 18 mo of age, were used to determine relationships among marbling traits, adipocyte size, and the amount of adipose tissue in different depots. Differences were recorded among the size of i.m. adipocytes in different groups of marbling flecks, divided according to the location in the muscle cross-section and to the size of the marbling flecks. The results showed positive correlation between i.m. adipocyte size and the weight of s.c. fat, intestinal fat, omental fat, and perirenal fat (r = 0.50, 0.61, 0.70, and 0.63, respectively, P < 0.001). The i.m. adipocyte size was correlated with i.m. fat content, number of marbling flecks, proportion of marbling fleck area, and total length of marbling flecks (r = 0.71, 0.44, 0.62, and 0.55, respectively, P < 0.01). The number of marbling flecks was also correlated with i.m. fat content, proportion of marbling fleck area, and total length of marbling flecks (r = 0.58, 0.62, and 0.91, P < 0.01, respectively). The ventral marbling flecks had a 5-fold larger fleck area, 4-fold more adipocytes, and larger adipocytes (P < 0.001). Larger marbling flecks contained larger adipocytes (P < 0.001). Moreover, compared with the small marbling flecks, there was a 48-fold larger fleck area and 26-fold more adipocytes in the large marbling flecks. The results indicate that i.m. fat deposition increases concurrently with the other fat depots but is still independent. Furthermore, the i.m. fat is preferentially deposited in the ventral area of LM. Although the i.m. adipocyte size has an important effect on the traits of marbling flecks, cell number plays a greater role in i.m. fat deposition than cell size.  相似文献

Marbling is an important criterion for beef quality grading in many countries. The purpose of the current study was to utilize the natural genetic variation to identify major metabolic indicators of marbling in cattle differing in genotypes. Rectus abdominis (RA, oxidative), semitendinosus (glycolytic), and longissimus thoracis (LT, oxido-glycolytic) muscles were taken from steers of different genotypes that expressed high [Angus, n = 16; and crossbred (Angus x Japanese Black), n = 10] or low (Limousin, n = 12) levels of marbling in their meat. Muscles from Angus and crossbred steers were characterized, as expected, by a greater triacylglycerol (TAG) content (P < 0.001) and also by greater protein contents of fatty acid-binding protein specific for heart and muscles (H-FABP; P < 0.001 for RA and P < 0.05 for LT muscle) or for adipocytes (A-FABP; P < 0.001 for RA and LT muscles). Moreover, oxidative enzyme activities (beta-hydroxyacyl-CoA dehydrogenase, citrate synthase, isocitrate dehydrogenase, cytochrome-c oxidase) were greater (P < 0.01 to 0.001) in the 3 muscles studied, whereas glycolytic enzyme activities (phosphofructokinase and lactate dehydrogenase) were lower (P < 0.001) in RA muscle in Angus and crossbred steers compared with Limousin steers. Significant correlations were observed between TAG content and H- and A-FABP protein contents, and oxidative (r > or = +0.55, P < 0.001) or glycolytic enzyme activities (r > or = -0.47, P < 0.001), when the 3 genotypes and muscles studied were considered as a whole. In addition, A-FABP protein content and some oxidative enzyme activities were significantly correlated with TAG content independently of the genotype and muscle effects. In conclusion, A-FABP protein content, as well as oxidative enzyme activities, may be used as indicators of the ability of steers from extreme genotypes to deposit intramuscular fat.  相似文献

The objective of this study was to develop mathematical equations for describing the change in marbling in Japanese Black steers using longitudinal measurements. Serial ultrasound measurements were taken at 14, 16, 20, and 26 months of age and analyzed using an image analysis software. The longitudinal marbling measurements from the ultrasound images and carcasses were fitted into a nonlinear logistic curve. Data used for the analysis consisted of 749 steers that converged in nonlinear curve fitting and showed reasonable estimated parameters of the logistic curves. The average predicted mature beef marbling score (BMS) and maturation rate were 6.26 and 0.353, respectively, and the average maturity levels at 24 months of age were 83.9%. The heritability estimates for the predicted maturity traits were moderate, indicating that these traits may have potential for genetic improvement. There was a negative relationship between the expected progeny differences between carcass BMS and maturity traits, suggesting that genetic improvement by carcass BMS may lead to the selection of bulls with late maturity for marbling. The results indicate that ultrasound and model building for marbling can be useful tools to correctly select candidate bulls with high marbling in the early fattening period.  相似文献

OBJECTIVE: To compare incidence of and breed-related risk factors for gastric dilatation-volvulus (GDV) among 11 dog breeds (Akita, Bloodhound, Collie, Great Dane, Irish Setter, Irish Wolfhound, Newfoundland, Rottweiler, Saint Bernard, Standard Poodle, and Weimaraner). DESIGN: Prospective cohort study. ANIMALS: 1,914 dogs. PROCEDURE: Owners of dogs that did not have a history of GDV were recruited at dog shows, and the dog's length and height and depth and width of the thorax and abdomen were measured. Information concerning the dogs' medical history, genetic background, personality, and diet was obtained from owners, and owners were contacted by mail and telephone at approximately 1-year intervals to determine whether dogs had developed GDV or died. Incidence of GDV based on the number of dog-years at risk was calculated for each breed, and breed-related risk factors were identified. RESULTS AND CLINICAL RELEVANCE: Incidence of GDV for the 7 large (23 to 45 kg [50 to 99 lb]) and 4 giant (> 45 kg [> 99 lb]) breeds was 23 and 26 cases/1,000 dog-years at risk, respectively. Of the 105 dogs that developed GDV, 30 (28.6%) died. Incidence of GDV increased with increasing age. Cumulative incidence of GDV was 5.7% for all breeds. The only breed-specific characteristic significantly associated with a decreased incidence of GDV was an owner-perceived personality trait of happiness.  相似文献

Marbling, defined by the amount and distribution of intramuscular fat, is an economically important trait of beef cattle in Japan. The endothelial differentiation , sphingolipid G-protein-coupled receptor , 1 ( EDG1 ) gene has been considered as a positional functional candidate for the gene responsible for marbling. We have recently reported that 2 single nucleotide polymorphisms (SNPs), c.-312A > G in the 5' untranslated region (UTR) and c.*446G > A in the 3' UTR in EDG1 were associated with marbling in Japanese Black beef cattle, but this was not functional and a causal mutation for marbling. In the present study, we detected 2 novel SNPs, referred to as g.1475435G > A and g.1471620G > T , in the 5' flanking region of the EDG1 between low-marbled and high-marbled steer groups, which were previously shown to have EDG1 expression differences in musculus longissimus muscle. The g.1475435G > A SNP seemed not to segregate in Japanese Black beef cattle. The g.1471620G > T SNP was associated with the predicted breeding value for beef marbling standard number by the analyses using Japanese Black beef cattle population. Based on these findings, we hypothesized that the g.1471620G > T SNP might have an impact on EDG1 expression and also marbling.  相似文献

Fatty acid composition, marbling score and melting point data collected between 1994 and 1996 were analysed. The data were from the adipose tissue of 764 Angus, Belgian Blue, Hereford, Jersey, Limousin, South Devon and Wagyu crossbred cattle slaughtered after lot‐feeding at 500 days of age. The aim was to investigate sire‐breed differences and to estimate heritability and genetic and phenotypic correlations. Significant breed differences were found: Jersey crosses had the highest marbling score and Belgian Blue crosses had the lowest. Limousin crosses had the highest melting point and Jersey crosses the lowest. South Devon crosses had the highest proportion of stearate and Jersey crosses the lowest. Desaturation indices in C16 and C18 fatty acids were highest in Jersey crosses and lowest in Limousin and South Devon crosses. In contrast, there were no breed differences in the proportions of palmitate, oleate, total saturated, total mono‐unsaturated fatty acids and elongation index. Heritability estimates of individual fatty acids and their summations, melting point and marbling were low to moderately low (0.05–0.27). Strong genetic correlations of melting point and desaturation index in C16 fatty acids (‐0.93), melting point and stearate (0.62), marbling and stearate (‐0.71) and marbling and desaturation index in C18 fatty acids (0.62) were observed. Phenotypic correlation were generally low. The results imply that fatty acids in the adipose tissue of lot‐fed cattle have a moderately low heritability, hence genetic progress might be slow.  相似文献

Marbling, defined by the amount and distribution of intramuscular fat, is an economically important trait of beef cattle in Japan. The c2‐11#2 expressed sequence tag (EST) has been previously shown to possess expression difference in musculus longissimus muscle between low‐marbled and high‐marbled steer groups, and to be located within genomic region of a quantitative trait locus for marbling. Thus, the ribosomal protein L27a (RPL27A) gene containing the c2‐11#2 EST sequence was considered as a positional candidate for the gene responsible for marbling. In the present study, a single nucleotide polymorphism (SNP) in the promoter region of the RPL27A, referred to as g.3109537C>T, was detected between the 2 steer groups. The SNP was associated with the predicted breeding value for beef marbling standard number by the analyses using Japanese Black beef cattle population. The effect of genotypes of the SNP on the predicted breeding value for subcutaneous fat thickness was not statistically significant. These findings suggest that the RPL27A SNP may be useful for effective marker‐assisted selection to increase the levels of marbling in Japanese Black beef cattle.  相似文献

The objective of the study was to determine the prevalence of cataracts in dogs presented to veterinary medical teaching hospitals in North America between 1964 and 2003. A retrospective study of all dogs presented with cataracts to veterinary medical teaching hospitals in North America between 1964 and 2003 was conducted to determine cataract prevalence. The different decades, breeds, gender, and age at time of presentation with cataract were compared. The prevalence of dogs presented with cataract varied by decade and ranged from 0.95% (1964-73), 1.88% (1974-83), 2.42% (1994-2003), to 3.5% (1984-93). The total number of dogs presented with cataracts over the 40-year period was 39,229. From 1964 to 2003 the prevalence of cataract formation in this patient population increased by about 255%. Fifty-nine breeds of dogs were affected with cataracts above the baseline prevalence of 1.61% seen in mixed-breed/hybrid dogs. The breeds with the highest cataract prevalence included: Smooth Fox Terrier (11.70%), Havanese (11.57%), Bichon Frise (11.45%), Boston Terrier (11.11%), Miniature Poodle (10.79%), Silky Terrier (10.29%) and Toy Poodle (10.21%). The breeds with the largest number of cataractous dogs during the entire four decades were the Boston Terrier (11.11%), Miniature Poodle (10.79%), American Cocker Spaniel (8.77%), Standard Poodle (7.00%), and Miniature Schnauzer (4.98%). Gender ratios of cataractous dogs seemed to affect limited breeds. Age of presentation with cataract diagnosis varied among several breeds. In the mixed-breed/hybrid baseline population, cataract formation appeared to be age related with a higher frequency of cataract formation in dogs after 4-7 years. Cataract formation is one of the most prevalent eye diseases in the dog population, and in about 60 breeds of dogs the prevalence of cataract exceeds that of the baseline mixed-breed/hybrid group. The prevalence of cataract is also influenced by age in most purebred dogs and affects 16.80% of the 7-15+-year-old mixed-breed/hybrid dog population. Total and age-related cataract prevalence in dogs seems very similar to that in man.  相似文献

Selection criteria for yearling bulls commonly include indicators of fertility and carcass merit, such as scrotal circumference (SC) and intramuscular fat percentage (IMF). Genetic correlation estimates between ultrasound traits such as IMF and carcass marbling score (MS) with fertility traits SC and heifer pregnancy (HP) have not been reported. Therefore, the objective of this study was to estimate the genetic parameters among the indicator traits IMF and SC, and the economically relevant traits MS and HP. Records for IMF (n=73,051), MS (n=15,260), SC (n=43,487), and HP (n=37,802) were obtained from the Red Angus Association of America, and a 4-generation ancestral pedigree (n=10,460) was constructed from the 8,915 sires represented in the data. (Co)variance components were estimated using a multivariate sire model and average information REML to obtain estimates of heritability and genetic correlations. Fixed effects included contemporary group and the linear effect of age at measurement for all traits, and an additional effect of age of dam for both HP and SC. The random effect of sire was included to estimate additive genetic effects, which were assumed to be continuous for IMF, MS, and SC, but a probit threshold link function was fitted for HP. Generally moderate heritability estimates of 0.29 ± 0.01, 0.35 ± 0.06, 0.32 ± 0.02, and 0.17 ± 0.01 were obtained for IMF, MS, SC, and HP on the underlying scale, respectively. The confidence interval for the estimated genetic correlation between MS and HP (0.10 ± 0.15) included zero, suggesting a negligible genetic association. The genetic correlation between MS and IMF was high (0.80 ± 0.05), but the estimate for HP and SC (0.05 ± 0.09) was near zero, as were the estimated genetic correlations of SC with MS (0.01 ± 0.08) and IMF (0.05 ± 0.06), and for HP with IMF (0.13 ± 0.09). These results suggest that concomitant selection for increased fertility and carcass merit would not be antagonistic.  相似文献

Hemochromatosis is an inherited defect in Salers cattle, characterized by increased iron absorption and wasting in young animals. Loss of teeth and soft bones has been reported. This study details bone changes in a prospective study of an affected Salers heifer and an age-matched, half-sibling control. The heifer fell 2 weeks before euthanasia, developed non-weight bearing lameness, and then became recumbent. At necropsy, the heifer was smaller than the control and had fractures of the humerus, femur, and ribs. Distinctive circumferential laminations in the outer third of the cortex were seen radiographically as layers of variably decreased density. In midshaft cross sections, the cortex was wider in some long bones, and fluorescent labeling revealed irregular and intermittent mineralization in the outer cortical layers. Mineral apposition rates in osteons of the inner cortex were decreased as compared with the control, and longitudinal bone formation at the growth plate was markedly decreased. Histologically, there were buried layers of osteopenic, poorly mineralized plexiform bone in outer circumferential lamellae. There was stainable iron in woven bone cores of the matrix. Iron was found in occasional osteoclasts as well. The mandible was severely affected, with loss of cortical bone as well as trabecular bone around the teeth and resorption of dental roots. Bone analysis revealed iron levels in the affected calf that were 30-50 times greater than the control and decreased percent ash in the outer cortex. These results indicate that periosteal dysplasia and osteopenia are responsible for pathologic fractures and tooth loss.  相似文献

Objective To determine the prevalence of the breed‐related glaucomas in pure‐bred dogs presented to the veterinary medical teaching hospitals in North America that participate in the Veterinary Medical Data Base (VMDB). Materials and methods In this retrospective study, age of first diagnosis, breed, and gender data for all breeds of dogs were collected from the VMDB with the clinical diagnosis of primary glaucoma (glaucoma‐NOS) at 5–10 year intervals from 1964 to 2002. The prevalence for each breed (affected dogs compared to all dogs of each breed), any changes over the 38 years, and any gender differences for these glaucomas were determined. Results The prevalence of the primary breed‐related glaucomas has gradually increased from 0.29% (1964–1973); 0.46% (1974–1983); 0.76% (1984–1993); to 0.89% (1994–2002). Breeds that consistently featured among the highest 10 for glaucoma prevalence from four different periods (1964 to 2002) included American Cocker Spaniel, Basset Hound, Wire Fox Terrier, and Boston Terrier. During the last observation period (1994–2002), 22 different breeds had 1% or higher prevalence of the glaucomas. The highest prevalence of glaucomas in 1994–2002 by breed included: American Cocker Spaniel (5.52%); Basset Hound (5.44%); Chow Chow (4.70%); Shar‐Pei (4.40%); Boston Terrier (2.88%); Wire Fox Terrier (2.28%); Norwegian ElkHound (1.98%); Siberian Husky (1.88%); Cairn Terrier (1.82%); and Miniature Poodle (1.68%). A predominance of females with glaucoma occurred in the American Cocker Spaniel, Basset Hound, Cairn Terrier, Chow Chow, English Cocker Spaniel, Samoyed, and perhaps the Siberian Husky, and a predominance of males in the Australian Cattle dog and St Bernard. Age affected the time for first presentation of the glaucomas in the pure‐bred dog. In the majority of breeds the glaucomas were presented for initial diagnosis in dogs between 4 and 10 years of age. Conclusion Breed‐related glaucomas in pure‐bred dogs are frequently presented to the veterinary medical teaching hospitals in North America. The prevalence of the breed‐related glaucomas in the dog appears similar to humans, and in some breeds exceeds that in humans. In many breeds the high prevalence of the glaucomas suggests a genetic basis.  相似文献

肉牛双肌基因(double muscle gene)是可使产肉性能大大提高,但同时也使母牛难产率有所提高。该基因位于第2号染色体上,是由生长抑素(myostatin)基因在第3外显子处核甘酸突变造成的,为不完全显性遗传。牛的双肌性状是一个综合性状。作者论述了牛双肌性状的遗传特征、基因定位、多形性及双肌牛的生理、繁殖、胴体特性,并对双肌牛在牛肉生产中的利用方式及在我国肉牛生产中的应用前景进行阐述。  相似文献
