Summary In this study genetic variation for resistance to the greenhouse whitefly (Trialeurodes vaporariorum) of four genotypes of tomato (L. esculentum) and two subspecies of L. hirsutum was investigated. Resistance was quantified by the whitefly life history components adult survival, oviposition rate, pre-adult survival and developmental period, measured on plants inoculated with whiteflies in clip-on cages.The largest differences between species were found when life history components were measured on adult plants of about four months old. On L. hirsutum f. glabratum whiteflies had the lowest adult survival, oviposition rate and pre-adult survival. On L. hirsutum these components were intermediate whereas on all L. esculentum genotypes they were highest. The variation between plants was low compared to the variation within plants. These results indicate that single plant tests can be used to determine accurately genetic variation between individual plants in a segregating population.  相似文献

Summary The variation among single and three-way crosses derived from seven lines of pepper (Capsicum annuum) selected for resistance to Phytophthora capsici was analyzed into statistical-genetic components due to general and specific combining abilities and additive-by-additive epistasis. Indirect evidence of higher order epistasis was also estimated. Epistasis was a principal source of variation. It was correlated with the level of aggressiveness of the pathogen. A breeding method such as recurrent selection seems to be the most appropriate to improve resistance to the fungus.  相似文献

Breeding for reniform nematode (Rotylenchulus reniformis) resistance is hindered by the lack of resistance in upland cotton (Gossypium hirsutum) cultivars. Resistance has been frequently identified in accessions from the Gossypium arboreum germplasm collection with accession PI 529740 rated as highly resistant. Accession PI 529740 was crossed with the susceptible Garboreum accession PI 529729 to develop an F2 population for genetic characterization. The population showed quantitative variation suggesting multiple genes conferred the resistant phenotype. Thirteen of the 216 F2 plants showed resistance similar to the resistant parent and these data supported a two recessive gene model. Sixty plants were classified as resistant or moderately resistant, indicating a single recessive gene conferred the moderately resistant phenotype. The classification of 24 F2:3 families for nematode resistance generally supported the classification of the corresponding F2 plants; however, most families were highly variable for infection with no families rated as resistant. This information will aid in the introgression of resistance into upland cotton as larger populations will be required to successfully recover resistance conferred by multiple recessive genes.  相似文献

Summary Raparadish, x Brassicoraphanus, the amphidiploid hybrid between Brassica rapa (syn. B.campestris) and Raphanus sativus (fodder radish) was made by Dolstra (1982). Primary hybrid plants grew vigorously, suggesting that the amphidiploid AARR might be useful as a fodder crop. Three populations of this new material were studied, with special attention to improvement of fertility and resistance to beet cyst nematode (Heterodera schachtii), whilst preserving genetic variability. For lack of progress one of the populations was abandoned after the fourth generation. The other two populations were observed through nine or ten generations. Apart from the last two generations mass selection for seed set was carried out on the basis of single plants. This led to a considerable increase in average seed production, without losing a wide variation for this trait. Thus more progress is being expected. Five cycles of mass selection for resistance to beet cyst nematodes led to a considerable increase of the level of resistance of both populations. The prospects of this new agricultural crop are discussed.  相似文献

Inheritance of resistance to Karnal bunt was investigated in the crosses of four resistant synthetic hexaploid wheats (SH; Triticum turgidum×T. tauschii) and two susceptible T. aestivum cultivars. The resistance was dominant or partly dominant over susceptibility. The SH cultivars Chen/T. tauschii (205) and Chen/T. tauschii (224) have single dominant resistance genes which could be allelic to each other. ‘Altar 84’/T. tauschii (219) appeared to have two dominant genes for resistance. ‘Duergand’T. tauschii (214) possessed two complementary dominant genes for resistance. The work is being extended to involve diverse Karnal bunt-resistant SH and bread wheat cultivars.  相似文献

The genetics of resistance to Phomopsis stem blight caused by Diaporthe toxica Will., Highet, Gams & Sivasith. in narrow-leafed lupin (Lupinus angustifolius L.) was studied in crosses between resistant cv. Merrit, very resistant breeding line 75A:258 and susceptible cv. Unicrop. A non-destructive glasshouse infection test was developed to assess resistance in the F1, F2, selected F2-derived F3 (F2:3) families, and in selfed parent plants. The F1 of Unicrop × 75A:258 (and reciprocal cross) was very resistant, and the F2 segregated in a ratio of 3:1 (resistant: susceptible), which suggested the presence of a single dominant allele for resistance in 75A:258. In Merrit × Unicrop (and reciprocal), the F1 was moderately resistant, and the F2 segregated in a ratio of 3:1 (resistant: susceptible). Thus Merrit appeared to carry an incompletely dominant resistance allele for resistance. The F1 of Merrit × 75A:258 (and reciprocal) was very resistant and the F2 segregated in a ratio of 15:1 (resistant: susceptible), which supported the existence of independently segregating resistance alleles for resistance in 75A:258 and Merrit. Alleles at loci for early flowering (Ku) and speckled seeds (for which we propose the symbol Spk) segregated normally and independently of the resistance alleles. Resistant F2 plants gave rise to uniformly resistant or segregating F2:3 families, whereas susceptible F2 plants gave rise only to susceptible F2:3 families. However, the variation in resistance in the F2 and some F2:3 families of crosses involving 75A:258, from moderately to extremely resistant, was greater than that expected by chance or environmental variation. We propose the symbols Phr1 to describe the dominant resistance allele in 75A:258, and Phr2 for the incompletely dominant resistance allele in Merrit. Phr1 appears to be epistatic to Phr2, and expression of Phr1 may be altered by independently segregating modifier allele(s). Sclerotinia sclerotiorum (Lib.) de Bary is a major limiting factor for sunflower (Helianthus annuus) production. This study was conducted to (1) evaluate a large number of current sunflower lines for their reaction to Sclerotinia, and (2) determine the optimum allocation of resources for their selection. A set of 85 inbred lines was screened under artificial leaf infection in three environments. The test revealed ample variation for all resistance traits in the current germplasm. A line selected from NDBLOS, an oilseed sunflower germplasm poo, and its derivatives were most resistant to Sclerotinia. Lines derived from interspecific crosses with the wild species Helianthus tuberosus and Helianthus argophyllus also showed satisfactory Sclerotinia resistance. The evaluation of five plants per plot in two replications and four environments appears to be a reasonable allocation of resources in order to optimize the selection response.  相似文献

Thrips are damaging pests in pepper worldwide. They can cause damage directly by feeding on leaves, fruits or flowers, and also indirectly by transferring viruses, especially tomato spotted wilt virus (TSWV). Although thrips are among the most damaging pests in pepper, until now there is no commercial variety with a useful level of resistance to thrips. This is at least partly due to the lack of knowledge on resistance levels in pepper germplasm of QTLs and/or genes for resistance, and of information about resistance mechanisms to thrips in pepper. This paper describes our research aimed at developing practical and reliable screening methods for thrips resistance in pepper and at identifying pepper accessions showing a strong resistance to thrips. Thirty-two pepper accessions from four species of pepper (Capsicum annuum, C. baccatum, C. chinense and C. frutescens) and two species of thrips (Frankliniella occidentalis and Thrips parvispinus) were used in this study. Our results indicate that the laboratory based leaf disc test and the detached leaf test can be used as reliable screening methods for thrips resistance in pepper. We observed a large variation for resistance to thrips in Capsicum that can be exploited in breeding programs.  相似文献

The amount of genetic variation among inbred lines and testcrosses, and covariation between both genetic materials, are of crucial importance for selection efficiency in hybrid breeding. To estimate these quantitative genetic parameters for resistance of winter rye (Secale cereale) to head blight caused by Fusarium culmorum, 88 three-way cross hybrids, produced by crossing each of 44 S2 Carsten inbred lines with two unrelated Petkus single-cross testers, were evaluated along with the parental lines over 2 years. Resistance traits were head-blight rating and grain weight per spike relative to the non-inoculated control. Significant genotypic variation occurred among lines and in both testcross series. S2 lines displayed considerably more variation than testcross series. Genotype × environment interaction was more marked among the inbred lines, while estimates of heritability were similar for both genetic materials. Testcrosses showed heterosis for head-blight resistance. No relationship existed between S2 lines and the two testcross series for any resistance trait. This might be caused by an association between inbreeding and Fusarium-head-blight susceptibility and different inbreeding depression among the S2 population. The phenotypic correlations between the testcross series were moderate for both traits (r = 0.58, P < 0.01). In conclusion, Fusarium-head-blight resistance has to be selected at the respective heterozygosity levels.  相似文献

Eighty-three third backcross lines which comprise a set of near isogenic lines (NIL's) of the barley cultivar ‘Clipper’ but each carrying a different chromosomal segment from Hordeum spontaneum, marked with a distinct isozyme, were tested for resistance to three races of the barley leaf rust pathogen (Puccmia hordei). Fourteen lines showed resistance to at least one race and three showed resistance to all three races. The resistance in two of these lines was controlled by separate, single partially dominant genes. In one case the resistance gene named Rph1O was on chromosome 3 and linked (r = 0.15 ±0.05) with the isozyme locus Est2. In the second case, the gene (Rph11) was on barley chromosome 6 and linked (r = 0.07±0.02) with the isozyme locus Acp3 and (r = 0.11±0.02) with Dip2.  相似文献

Asian rice gall midge (Orseolia oryzae) is a major pest across much of south and southeast Asia. This pest is genetically diverse and many gall midge biotypes are known to exist in each country. During the last three decades, host plant resistance has proved to be the most effective mechanism of controlling the Asian rice gall midge. Seven genes conditioning resistance to gall midge larvae have been identified in rice (Oryza sativa) and are being used in cultivar improvement programs. However, some of these genes are rendered ineffective by new gall midge biotypes. Increased understanding of genetics, inheritance, allelic relationships and linkage is necessary to maximise the durability of major gene resistance by the pyramiding of these genes. The two genes, Gm-2 and Gm-6(t), are known to confer resistance against a number of biotypes in India and China, respectively. An F3 population derived from a cross between Duokang #1 (donor of Gm-6(t)) and Phalguna (donor of Gm-2) was screened against Chinese gall midge biotype 4 at Guangdong, China, and Indian gall midge biotype 1 at Raipur, India. At each location, separately,a single gene governed resistance. The parallel segregation of 417 F3progenies for both biotypes at two locations revealed that recombination had occurred between the two genes, establishing that the two genes are not allelic. However, the two genes Gm-2 and Gm-6(t), were found to be linked with a distance of ∼16.3 cM. A number of lines homozygous at one locus and segregating for the other locus were identified and selected. These lines were selfed to obtain lines homozygous for the favourable alleles at both loci (two locus pyramids). This is the first report on use of conventional host-pest interaction method for pyramiding two closely located Gm-resistance loci of dissimilar effects. The implications of deployment of these pyramids within and across country borders, with reference to the prevailing gall midge populations are discussed. Summary In studies of the inheritance of resistance, pea seedlings of seven lines in which stems and leaves were both resistant to Mycosphaerella pinodes were crossed with a line in which they were both susceptible. With seven of the crosses resistance was dominant to susceptibility. When F2 progenies of five crosses were inoculated on either stems or leaves independently, phenotypes segregated in a ratio of 3 resistant: 1 susceptible indicating that a single dominant gene controlled resistance. F2 progenies of one other cross gave ratios with a better fit to 9 resistant: 7 susceptible indicating that two co-dominant genes controlled resistance. The F2 progeny of another cross segregated in complex ratios indicating multigene resistance.When resistant lines JI 97 and JI 1089 were crossed with a susceptible line and leaves and stems of each F2 plant were inoculated, resistance phenotypes segregated independently demonstrating that leaf and stem resistance were controlled by different genes. In two experiments where the F2 progeny of the cross JI 97×JI 1089 were tested for stem and leaf resistance separately, both characters segregated in a ratio of 15 resistant:1 susceptible indicating that these two resistant lines contain two non-allelic genes for stem resistance (designated Rmp1 and Rmp2) and two for leaf resistance (designated Rmp3 and Rmp4). Evidence that the gene for leaf resistance in JI 1089 is located in linkage group 4 of Pisum sativum is presented.  相似文献

Summary Seedling resistance to wheat stem rust was determined in populations of wild emmer wheat, Triticum dicoccoides, and characterized by means of ecological factors and allozyme genotypes. Reactions to wheat stem rust were studied in 102 single plant accessions of T. dicoccoides from ten populations by inoculation with Puccinia graminis tritici race 14, isolate GSR-739. Six populations displayed different degrees of response polymorphism with reactions ranging from high resistance to complete susceptibility, whereas four populations contained only susceptible plants. In some of the accession, unexplained intrasib variation in resistance and intraplant variation of infection-types were found. Resistance to stem rust was negatively correlated with two ecological factors, altitude and number of Sharav (hot-dry) days which are unfavorable to disease development. Variation in stem rust response was shown to exist in ecogeographic regions where climatic variables enhanced the development of the fungus, conceivably maintained by natural selection. Likewise, allozyme genotypes, single or in multiple loci combinations, appeared to be associated with resistance or susceptibility to rust. Such association need to be verified by genetic studies in order to become established as useful markers.  相似文献

Late blight (Phytophthora infestans) can have devastating effects on tomato production over the whole world. Most of the commercial cultivars of tomato, Solanum lycopersicum, are susceptible. Qualitative and quantitative resistance has been described in wild relatives of tomato. In general qualitative resistance can more easily be overcome by newly evolved isolates. Screening of three S. habrochaites accessions (LA1033, LA2099 and LA1777) through a whole plant assay showed that accession LA1777 had a good level of resistance to several isolates of P. infestans. To explore the potential in this wild species, an introgression line (IL) population of S. habrochaites LA1777 was used to screen individual chromosome regions of the wild species by a detached leaf assay. Two major isolates (T1,2 and T1,2,4) were used and two parameters were measured: lesion size (LS), and disease incidence (DI). Substantial variation was observed between the individual lines. QTLs were identified for LS but not for DI. The presence of five QTLs derived from LA1777 (Rlbq4a, Rlbq4b, Rlbq7, Rlbq8 and Rlbq12) results in unambiguous higher levels of resistance. All QTLs co-localized with previously described QTLs from S. habrochaites LA2099 except QTL Rlbq4b, which is therefore a novel QTL.  相似文献

The genetics of race-non-specific foliage resistance against Phytophthora infestans, of foliage maturity type, and of their association in potato (Solanum tuberosum) were studied. Six progenies were derived from a half-diallel set of crosses between diploid potato clones that represented a broad pool within the genus Solanum and were free of any of the 11 known R genes for late blight resistance. The progenies were evaluated for resistance to late blight and for foliage maturity type, and five of them showed a significant correlation between the two traits. The correlation did not account for all variation that was present for both traits, as reflected in the analysis in which the relative AUDPC values were adjusted for foliage maturity type. The present study adds to previous results: resistance against P. infestans always coincides with late foliage maturity. However, the results also indicate that some selection for late blight resistance without affecting the foliage maturity type should be possible.  相似文献

Phomopsis helianthi (sexual stage Diaporthe helianthi) is a major disease in most sunflower growing countries. The objectives of this study were (1) to investigate whether resistance to Phomopsis differs between the leaf and stem tissue and (2) to examine the genetic variation for resistance to Phomopsis in sunflower inbreds. A set of 16 inbred lines was tested using artificial leaf infection with mycelial explants. The lines were screened for their reaction on leaf and stem in four environments to a French Phomopsis isolate and in two environments to a Yugoslavian isolate. Significant genetic variance and mostly high genetic ratios (analogous to heritability) were found for all resistance traits. While fungal growth reflected resistance to Phomopsis in the leaf, petiole score was associated with leaf length. Stem lesion proved to be the most suitable trait for evaluating resistance in the petiole-stem passage. No correlation existed between resistance in the leaf and stem, suggesting that both resistance factors are inherited independently. Both isolates resulted in similar resistance rankings of the inbreds and no difference in their aggressiveness was detected with regard to leaf resistance. Screening for Phomopsis consisting of several steps is recommended, beginning with petiole score or fungal growth to select the most susceptible plants in the early testing, and measuring stem lesion in further generations of breeding.  相似文献

Genetic analysis of common wheat cultivar PBW343 confirmed temperature-sensitive leaf rust resistance and adult plant stripe rust resistance. At low temperatures, PBW343 was resistant to P. triticina (Ptr) pathotype (pt.) 121R63-1, and at high temperature it was resistant to Ptr pt. 121R127. The low temperature resistance to pt. 121R63-1 was attributed to interaction between dominant and recessive genes. The dominant gene involved in low-temperature resistance to pt. 121R63-1 also conferred resistance to pt. 45R35. The high-temperature resistance to Ptr pt. 121R127 was governed by a different single partially dominant gene. Agra Local (a commonly used susceptible check) and IWP94 (a leaf rust differential used in India) are also resistant to pt. 121R127 at high temperatures. An allelism test indicated that PBW343 and IWP94 possessed a common gene for high temperature resistance to this pathotype. The adult plant stripe rust resistance against P. striiformis (Pst) was possibly conferred by one gene in addition to Yr27.  相似文献

‘Minghui 63’ is a restorer line widely used in hybrid rice production in China for the last two decades. This line and its derived hybrids, including ‘Shanyou 63’, are susceptible to bacterial blight (BB), caused by Xanthomonas oryzae pv. oryzae (Xoo). To improve the bacterial blight resistance of hybrid rice, two resistance genes Xa21 and Xa7, have been introgressed into ‘Minghui 63’ by marker‐assisted selection and conventional backcrossing, respectively. The single resistance gene‐introgressed lines, Minghui 63 (Xa21) and Minghui 63 (Xa7) had higher levels of resistance to bacterial blight than their derived hybrids, Shanyou 63 (Xa21) or Shanyou 63 (Xa7). Both Xa21 and Xa7 showed incomplete dominance in the heterozygous background of rice hybrids by infection with GX325 and KS‐1‐21. The improved restorer lines, with the homozygous genotypes, Xa21Xa21 or Xa7Xa7, were more resistant than their hybrids with the heterozygous genotypes Xa21xa21 or Xa7xa7. To further enhance the bacterial blight resistance of ‘Minghui 63’ and its hybrids, Xa21 and Xa7 were pyramided into the same background using molecular marker‐aided selection. The restorer lines developed with the resistance genes Xa21 and Xa7, and their derived hybrids were evaluated for resistance after inoculation with 10 isolates of pathogens from China, Japan and the Philippines, and showed a higher level of resistance to BB than the restorer lines and derived hybrids having only one of the resistance genes. The pyramided double resistance lines and their derived hybrids have the same high level of resistance to BB. These results clearly indicate that pyramiding of dominant genes is a useful approach for improving BB resistance in hybrid rice.  相似文献

Pvr4 locus, commonly associated with a co-dominant CAPS marker, confers monogenic dominant resistance to potyvirus complex in Capsicum spp. Aiming to investigate whether the resistance found in resistant genotypes not bearing Pvr4 marker is due to allelism in the Pvr4 locus, or due to a new locus of the pvr series, segregation analyses of an F2 population obtained from a cross between two pepper lines “Myr-29-10” (P1) (resistant to PepYMV, showing a single band of 444 bp, Pvr4/Pvr4) and “PIM-025” (P2) (resistant to PepYMV, showing a single band of 458 bp, Pvr4+/Pvr4+) were performed. According to the results, there is strong evidence that the locus controlling PepYMV resistance in PIM-025 (P2) is not Pvr4. These results provide evidence that those resistant genotypes, bearing a susceptible band pattern in the Pvr4 locus (458 bp, Pvr4+/Pvr4+), carry a different gene from those described in the literature up to the present time.  相似文献

A new resistance (R) gene to powdery mildew has been identified and characterized in a population derived from the wild potato species, Solanum neorossii under natural infection in the greenhouse. The segregation of resistance has revealed that this R gene is controlled by a single monogenic and dominant gene designated Rpm-nrs1. Analysis of the DNA sequence on an internal transcribed spacer (ITS) region of the pathogen genome suggests that the pathogen causing the powdery mildew disease is either Golovinomyces orontii or G. cichoracearum. The resistance locus was localized to the short arm of chromosome 6 where several disease R genes already identified in potato and tomato are known to reside. The resistance locus cosegregated in 96 progeny with three AFLP markers and one PCR marker. The sequences of the two cosegregating AFLP markers are highly homologous to Mi-1 conferring resistance to nematode, potato aphid and whitefly and Rpi-blb2 conferring resistance to late blight. The results in this study will facilitate the cloning of this gene conferring resistance to powdery mildew.  相似文献
