Abstract

The anatomy of the gaster of Trichosurolaelaps crassipes is illustrated. Protonymphs, deutonymphs and adults were all seen with a scarlet gut and gave strongly positive results to occult blood tests. Host blood cells were identified in a gut smear from a female mite. An adult female gut could hold about 7.1 × 10?6 ml. The mite was seen frequenting small open skin lesions. Gram-negative bacteria identified in the mite's gut are most likely to be endosymbionts. Mite larvae have their gastric caeca invaded bv these bacteria before birth. There is a notential for this mite to act as a weak vector of disease from one possum to another.  相似文献

Skin biopsies from two Merino sheep heavily infested with Psorergates ovis were immersed in liquid nitrogen and cut into vertical frozen sections stained with lipophilic Sudan IV or Oil Red O and haematoxylin. A survey of the lateral distribution of mite sections showed a majority (ca 80%) were in or within 0.2 mm of the follicle mouth. A survey of vertical distribution showed no mite penetration deeper than inner stratum corneum where 57% of mite sections were seen; 30% were within outer stratum corneum or scurf; 13% were on the outer surface and less than 1% were detached. Lipid was the only material seen within stained mites at a location considered to be gut. This was supported by dosing sheep with quinacrine, taking biopsies at Day 6 and Day 14 and examining frozen sections under blue light. Fluorescent lipid was seen at a location considered to be mite gut. From these results it was recommended that acaricide treatments against P. ovis be lipophilic and administered transepidermally because less than 15% of mites were at a superficial location likely to be reached by topical application.  相似文献

The mite Psoroptes ovis causes sheep scab disease in flocks throughout much of the world. A serious impediment to the development of novel control measures for this mite is our inability to produce in vitro colonies of the mite. Here, we describe the alimentary canal of the mite with the particular aim of determining what it feeds on, as part of a longer term goal, to develop in vitro culture techniques for P. ovis. The alimentary canal of P. ovis consists of a cuticle-lined foregut and hindgut separated by a microvilli-lined midgut. The foregut is divided into a pre-oral cavity and a muscular pharynx and oesophagus. The midgut consists of three ultrastructurally discrete areas: a stomach with bi-lobed ventriculi, a colon and a post-colon. The stomach is composed of two cell types. The most common cells (Type 1) are either cuboidal or squamous depending on the degree of gut distension and possess short microvilli, a single basally located nucleus, extensive rough endoplasmic reticulum and other components suggesting active secretion. The less common cells (Type 2), possess an extensive apical network of tubules and basally food vacuoles suggesting intracellular digestion. These cells extend into the gut lumen, become free-floating and degenerate. The colon and post-colon are composed of cells similar to Type 1 stomach cells but the post-colon epithelium possesses significantly longer microvilli. Cells from these areas have not been observed to leave the surrounding epithelium and enter the gut lumen, but it is suggested that significant absorption occurs in these two areas. Faecal pellets, often containing a significant number of bacteria, leave the digestive system through the cuticle-lined anal atrium.  相似文献

A case of feline demodicosis is described in this report. A 13-year-old spayed female domestic short hair cat weighing 4.5 kg was being treated with cefovecin and alternately with prednisone or methylprednisolone. On further physical examination, the cat showed mild erythema and hair loss on the bridge of the nose, around the eyes, on the chin, on the side part of the breast and on the abdomen. A large number of Demodex mites were found in deep skin scrapings from the affected areas. The cat was then treated with ivermectin at 600 μg/kg administered SC daily. After 4 weeks of treatment, the cat was clinically normal with no mites detected in the skin scrapings from the face or breast areas. The mite responsible may represent a previously seen but as yet unnamed new species. This is third report that describes a case of feline demodicosis caused by a different, unnamed mite species that has different morphological characteristics to those of known Demodex mites and may represent a previously seen but as yet unnamed species.  相似文献

Ovine IgG was detected in homogenates of repeatedly washed Psoroptes ovis. Some of the immunoglobulin in the homogenates was fragmented although the host IgG present in mite washings was largely intact. The host immunoglobulin was immuno-localised to the surface or cytoplasm of the gut cells of feeding stages of freshly harvested P. ovis examined by cryosectioning. A similar distribution of rabbit IgG was detected in P. cuniculi. The IgG demonstrated in the mite gut represented partially digested as well as intact immunoglobulin. The presence of intact host immunoglobulin suggests that P. ovis may be susceptible to vaccination by the gut antigen approach, a method used successfully for blood-feeding ectoparasites like Boophilus microplus.  相似文献

(1) The effect of infections with Dermanyssus gallinae (poultry red mite or chicken mite) and Ascaridia galli (roundworm) on the behaviour and health of laying hens was investigated. (2) Six groups of 15 pullets (Isa Brown) were kept in indoor pens from 18 weeks of age. Two groups were artificially infected with D. gallinae, two groups with A. galli and two groups were kept as uninfected controls. The hens were observed for behavioural reactions and physiological changes (weight gain and various blood variables) to the parasitic infections. (3) Infections with D. gallinae resulted in reduced weight gain, anaemia and even death of some of the hens. Behavioural changes were also observed, as the mite-infected hens showed higher self-grooming and head scratching both during the day and night. (4) A. galli resulted in a lower weight gain but no significant changes were seen in blood variables or behavioural activities.  相似文献

Background: Identification of the bacterial organism in dogs with endocarditis is challenging. Human studies have reported the utility of the polymerase chain reaction (PCR) to amplify and identify bacterial nucleic acid from infected valvular tissue and blood. Hypothesis/Objectives: We hypothesized that PCR using primers designed to amplify the bacterial 16s gene would identify circulating bacteria in dogs with suspected bacterial endocarditis more consistently than standard blood culture techniques. Animals: Eighteen dogs with suspected bacterial endocarditis based upon clinical and echocardiographic findings. Fifteen clinically normal dogs served as negative controls. Methods: Prospective study of dogs evaluated for suspect endocarditis at 6 veterinary hospitals. A blood sample was drawn from all dogs and evaluated with both a single‐sample PCR and standard 3‐sample blood culture techniques. Results: Blood culture identified noncontaminant bacteria in 6/18 study animals (33%) and 1 control dog; PCR identified noncontaminant bacteria in 7/18 study animals (39%). There were no study animals in which the 2 tests identified different bacteria (κ= 1.0). However, bacteria were identified by both techniques in only 2/18 study animals. When results from both PCR and blood culture were considered together, a noncontaminant bacterial organism was identified in 11/18 study animals (61%). Conclusion and Clinical Importance: The results of this study suggest that although single sample PCR with 16s primers was not more sensitive than blood culture for detection of bacteremia in dogs with suspect endocarditis, performing both techniques simultaneously did increase the likelihood of identification of bacteria in blood.  相似文献

Eight out of 10 lambs died within six to 11 hours after occlusion of the cranial mesenteric blood vessels by a snare placed around both the artery and vein. Death resulted from shock and was accompanied by post mortem findings identical to those seen in red gut. It is suggested that the cause of death in the naturally occurring disease is from arrested mesenteric blood flow following intestinal torsion.  相似文献

Atopic dermatitis is a chronic inflammatory and pruritic skin disease commonly seen in dogs and humans that is characterised by the presence of allergen-specific IgE. Data from skin tests and serological analysis suggest that the house dust mite Dermatophagoides farinae is the most important allergen in dogs with atopic dermatitis. The aim of this study was to determine if D. farinae specific peripheral blood mononuclear cell (PBMC) responses could be detected in dogs with atopic dermatitis. PBMCs were isolated by the density centrifugation from dogs with atopic dermatitis that were skin test positive for D. farinae, dogs with atopic dermatitis that were skin test negative for D. farinae, and healthy dogs. Cells were cultured with increasing concentrations of the D. farinae extract, no antigen, vaccine antigens or concanavalin A (ConA). There was significantly greater responsiveness of PBMCs from the D. farinae positive dogs than from either the D. farinae negative or healthy dogs (ANOVA, P<0.05). In contrast, no significant differences were observed in the control responses between the three groups. This is the first study to demonstrate that D. farinae specific circulating memory cells are involved in the pathogenesis of canine house dust mite hypersensitivity.  相似文献

The goal of prebiotic applications from different sources is to improve the gut ecosystem where the host and microbiota can benefit from prebiotics. It has already been recognized that prebiotics have potential roles in the gut ecosystem because gut microbiota ferment complex dietary macronutrients and carry out a broad range of functions in the host body, such as the production of nutrients and vitamins, protection against pathogens, and maintenance of immune system balance. The gut ecosystem is very crucial and can be affected by numerous factors consisting of dietary constituents and commensal bacteria. This review focuses on recent scientific evidence, confirming a beneficial effect of prebiotics on animal health, particularly in terms of protection against pathogenic bacteria and increasing the number of beneficial bacteria that may improve epithelial cell barrier functions. It has also been reviewed that modification of the gut ecosystem through the utilization of prebiotics significantly affects the intestinal health of animals. However, the identification and characterization of novel potential prebiotics remain a topical issue and elucidation of the metagenomics relationship between gut microbiota alteration and prebiotic substances is necessary for future prebiotic studies.  相似文献

Naive and previously exposed young Hereford cattle were infested with Psoroptes ovis and maintained in stanchions for 7 weeks. Skin biopsy samples were examined at 1, 2, 5, and 7 weeks after infestation was induced, compared with those from noninfested control calves, and correlated with changes in the mite populations and development of dermatitis. Gross and microscopic appearance of lesions were qualitatively similar in both groups of infested calves. The lesions occurred early in the previously exposed calves, but progressed slowly, whereas lesions in the naive calves appeared later, but were rapidly progressive. The basic histopathologic pattern was chronic exudative superficial perivascular dermatitis compatible with allergy. The mite populations on the naive calves revealed exponential growth and high fecundity, and reached high population densities, whereas mite populations on previously exposed calves had low growth rates and low fecundity and reached population densities 100 to 1,000 times less than those on naive calves. Seemingly, there was a hypersensitivity to mite antigens. The hypersensitivity response may have been a mechanism by which humoral immunity was delivered to the mite microenvironment and may have contributed to acquired resistance by controlling mite populations. The mechanism of population control seems to be related partially to decreased fecundity of female P ovis.  相似文献

The poultry red mite (Dermanyssus gallinae) is a serious problem for the poultry industry worldwide. However, the relationship between the mite population and the damage that they cause is still unclear. In this study, the mite population in poultry houses was examined using an established trap method, and the risk of blood‐stained eggs caused by the mites was assessed. Traps were placed once a week outside the egg channels and/or on the floor in two poultry farms in Fukuoka Prefecture, Japan, from April 2012 to July 2014. The numbers of blood‐stained eggs and total eggs were counted at weekly intervals. The results showed that the number of mites increased from April to May, and reached a peak around the beginning of June when the average temperature and humidity were >24°C and 70–90%, respectively. In the segmented model, the correlation between the proportion of blood‐stained eggs and the number of mites or temperature was positive over a threshold. In conclusion, our established trap method is useful for monitoring mites and can be used to predict when poultry farms should be treated to prevent appearance of blood‐stained eggs.  相似文献

Blood glucose was measured with a portable glucose meter in 907 rabbits, including 238 clinically healthy ones. Blood glucose concentrations ranged from 1.2 to 30.1 mmol/l. Diabetes mellitus was not encountered. No significant effect of sex or sedation was found. Hypoglycaemia was seen in 16 rabbits including one with an insulinoma. There was a significant relationship between blood glucose, food intake, signs of stress and severity of clinical disease. Rabbits showing signs of stress had higher blood glucose than rabbits with no signs and rabbits that were totally anorexic had higher blood glucose values than those that were eating normally or those with reduced food intake. Severe hyperglycaemia (>20 mmol/l) was associated with conditions with a poor prognosis. Rabbits with confirmed intestinal obstruction had a mean blood glucose of 24.7 mmol/l (n=18). This was significantly higher than the rabbits with confirmed gut stasis, which had a mean value of 8.5 mmol/l (n=51). The conclusion of the study was that blood glucose is a measurable parameter that can be used to assess the severity of a rabbit's condition and help to differentiate between gut stasis and intestinal obstruction in rabbits that are anorexic.  相似文献

White leghorn hens were experimentally infested with northern fowl mites (Ornithonyssus sylviarum) and antibody responses to mite immunogens were monitored over 12 weeks. Mite burdens increased during the early phase of infestation and declined over the latter weeks of the study. Antigen was prepared from homogenized whole mites, which were then sonicated and extracted with non-ionic detergent. Antigen extract was fractionated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and antibody-reactive polypeptides were identified by immunoblotting. At the start of infestation, hens had natural, pre-existing antibodies that reacted with several mite-extract components. Individual hens had different natural antibody reactivities; however, all birds had immunoglobulins reactive with extract polypeptides of 117,000, 77,000 and 36,000 molecular weight. A variety of mite extract components reacted with hen antibodies generated in response to experimental infestation. The number of antibody-reactive mite polypeptides increased through week 8 of infestation and then decreased by week 12. Fifteen polypeptides of northern fowl mite extract were reactive with antibodies developed by the majority of infested birds. These commonly reactive polypeptides had molecular weights ranging from 40,000 to 160,000. Glycoconjugates of fractionated mite extract were identified by blotting with lectins that have different carbohydrate binding specificities. Also identified were lectins that bound extract components with the same molecular weights as those moieties complexed by immunoglobulins of infested birds.  相似文献

Newcastle disease virus (NDV) specific antigen in the gut contents and NDV specific antibody in blood circulation were seen in day old chicks belonging to nine different commercial hatcheries of Tamil Nadu, India. Antigen disappeared by 4th week and antibody by 6th week of age. Fourteen NDV isolates obtained from the gut contents of day old chicks of different commercial hatcheries, one NDV isolate from dead in shell eggs and one NDV isolate from breeder hen were characterized and grouped under velogenic, mesogenic and lentogenic pathotypes. Four isolates were grouped under F and another four isolates were grouped under E based on reaction with monoclonal antibodies (Mabs) but found to be velogenic based on pathogenicity tests. In one particular flock velogenie NDV was isolated from breeder hen, dead in shell embryos and day old chicks and they all belong to Mabs group E. Vertical transmission of velogenic, mesogenic and lentogenic NDVs and role of NDVs in the gut contents have been discussed.  相似文献

Immunological control of ixodid ticks requires development of a vaccine that stimulates an effective anti-tick response. However, it is important to avoid induction of intense host skin reactivity to tick feeding. Salivary-gland-derived molecules are introduced into the host during ixodid engorgement; therefore, use of these moieties might not be an optimal immunization strategy. Antigens not normally involved in acquired resistance can be used to induce anti-tick immunity. These "novel" antigens, obtained from tick gut absorptive surface, are not introduced into the host during tick feeding, but are exposed to host-immune effector elements in the blood meal, resulting in ixodid rejection, prevention of ova production and death. Anti-tick immunity is induced with microgram quantities of this ixodid gut antigen preparation. Sera obtained from immunized animals were used to identify antibody-reactive components of the resistance-inducing extract. Tick gut absorptive surface antigen glycoconjugates were identified by lectin blotting, using a series of probes with different carbohydrate specificities. The lectins, peanut and wheat germ agglutinin, and immunized host antibodies bound extract components with similar molecular weights. Solubilization and fractionization of tick gut proteins with the non-ionic detergent Triton X-114 is described.  相似文献

Young willow ptarmigan chicks were fed an artificial chick diet (ACD), and different supplements. One group got ascorbic acid supplements, another neomycin and Oxytetracyclin in the water, and a third was given ascorbic acid and sprouts of Vaccinium myrtillus. Chicks that were fed ACD only, showed low weight gain and high mortality rate. The numbers of bacteria in the gut were high and the incidence of villous epithelium erosion and bacteria colonizing the gut epithelium were high.Admixture of ascorbic acid to the diet improved the viability of the chicks and reduced the number of coliforms in the gut.Antibiotics in the water reduced the number of C. perfringens, enterococci and coliforms in the gut, and improved the gut epithelium conditions.Combined supplements of ascorbic acid and Vaccinium myrtillus resulted in strongly viable chicks with healthy gut epithelium. C. perfringens was not demonstrated in the gut, and the numbers of enterococci and coliforms were low.  相似文献
