The effect of recombinant somatotropin (rbST), Synovex (Syn), and their combination (rbST+Syn) on intact male calves was examined in an experiment that lasted an average of 238 d. Holstein-Friesian bull calves were allotted to one of four subtreatments (n = 14/treatment) in a factorial arrangement. There were two levels of rbST (0; rbST) and two levels of the estrogenic growth promoter Synovex (0; Syn). The rbST was administered once every 2 wk as injections of 500 mg of Posilac. Synovex (C and S) was implanted at 90-d intervals. The animals were fed for ad libitum consumption a diet with a metabolizable energy concentration of 11.7 MJ/kg DM and 15% crude protein. The hot carcasses were weighed after the removal of kidney, pelvic, and cod fats, which were weighed separately. The 12th rib cut was saved for analysis. Average daily gain and feed conversion efficiency were increased by rbST treatment by 9% (P < .005) and 10% (P < .016), respectively. There was no significant effect of Syn treatment, nor was there a rbST x Syn interaction. The proportion of the fat of the large depots in the carcass was reduced by 34% (P < .0001) and in the longissimus muscle by 32% (P < .16) owing to the rbST treatment. The plasma concentrations of GH, insulin, and thyroxin were increased by rbST treatment (P < .001, P < .01, and P < .03, respectively). The concentration of IGF-I was not affected. Synovex had no effect on plasma hormone concentration. Plasma essential and nonessential amino acid concentrations were reduced by 14 and 9%, respectively, when rbST was injected. Concentrations of cholesterol and fatty acids in muscle and subcutaneous fat were not affected (P > .072) by the rbST treatment. Synovex increased the monounsaturated fatty acids (MUFA), and the combinaton of Syn with rbST reduced polyunsaturated fatty acid (PUFA) concentration in the longissimus muscle (at the 12th rib). The reduced muscle fat content of the rbST-treated animals was associated with a trend toward an increase in polyunsaturated fatty acids.  相似文献

One hundred twenty crossbred beef steers averaging 377 kg were used in a 2 x 4 factorial experiment to determine the dose-response effects of recombinant bovine somatotropin (rbST) implants on growth performance and carcass characteristics. The implant dosages were 0 (sham), 40, 80, or 160 mg/wk administered s.c. in the tailhead region on a weekly basis throughout the experiment. The steers were fed a high-concentrate diet, which contained either a degradable (DP; soybean meal) or an escape (EP; corn gluten and blood meal) protein source. No dietary protein effect or dietary protein x rbST level interactions were detected. Recombinant bST decreased both DMI (P less than .10) and feed/gain (P less than .05) in a linear dose-dependent manner. Dosage of rbST did not significantly affect (P greater than .10) ADG or final weight of the steers. Recombinant bST decreased backfat depth (P less than .10), marbling score (P less than .05), and quality grade (P less than .10) and increased yield grade (P less than .10) in a linear dose-dependent manner. Soft tissue composition of the 9-10-11th rib section was altered (P less than .01) by rbST administration in a linear dose-dependent manner. The percentage of protein in the rib section was increased by 9.4% and fat was decreased by 11.8% at the 160 mg/wk rbST level compared with the sham-implanted steers. During two studies, effects of recombinant bovine somatotropin (rbST) on plasma and milk IGF's in cows adapted to summer (S; 12 cows) or winter (W; 12 cows) conditions were evaluated. Each study consisted of on-farm periods (30 days) followed by climatology chamber periods (CC; 30 days). Cows were given daily injections of rbST, Sometribove, USAN (25mg/day; 6 cows each study) or saline (control; 6 cows each study). During on-farm periods, blood and milk (am and pm) samples were collected once weekly. During CC periods, blood samples were collected every 2 days and milk samples (am and pm) were collected daily. Plasma IGF-I and IGF-II were increased in cows treated with rbST. A pronounced seasonal pattern in basal and rbST-stimulated plasma IGF-I but not IGF-II was detected. Higher basal and rbST-stimulated plasma IGF-I concentrations in S occurred despite large decreases in feed intake and energy balance. Milk IGF-I and IGF-II was not affected by rbST treatment or season. Although milk IGF-I and IGF-II concentrations were unaffected by rbST treatment, total IGF-output increased due to increased milk yield. The observed seasonal patterns in plasma IGF-I may be indicative of seasonal differences in the coupling of the somatotropin-IGF axis. In particular, we failed to detect an uncoupling of the somatotropin-IGF-I axis in S despite an induced negative energy balance during thermal stress.  相似文献

Effects of recombinant bovine somatotropin (rbST) on ovarian and uterine function and the production of components of the insulin-like growth factor (IGF) system were examined during the period of maternal recognition of pregnancy in cattle. Lactating dairy cows were treated with 25 mg/d rbST (n = 8) or saline (n = 8) for 16 d after estrus. Ovaries, uteri, and conceptuses were collected on Day 17 after estrus. The length (millimeters) of the conceptus was recorded. The concentration of IGF-I and the content of IGF-binding proteins (BP) in uterine flushings were determined. Corpora lutea (CL) were weighed, and the number of follicles (⩾2 mm in diameter) were counted. Follicular fluid from the largest and second-largest follicles was assayed for the concentration of IGF-I, IGFBP, progesterone, and estradiol. The length of the conceptus and the total amount of IGF-I in uterine fluid were similar for rbST and control. Recombinant bST increased 1) the weight of the CL, 2) the number of largest follicles (10 to 15 mm in diameter), 3) the concentration of IGF-I in the follicular fluid, 4) the follicular fluid content of IGFBP of the largest estrogenic follicle, and 5) the quantity of IGFBP in uterine flushings. The concentration of progesterone in the follicular fluid tended to be increased in rbST-treated cows, whereas the concentration of estradiol was similar to that of control cows. The concentration of progesterone in plasma was similar for rbST compared with control. In conclusion, the administration of rbST in lactating dairy cows for 16 d after estrus did not alter the growth of the conceptus collected on Day 17. The greatest responses to rbST were found within the ovary, where rbST increased the weight of the CL and altered the amount of IGF-I and IGFBP in the follicular fluid.  相似文献

Thirty-six primiparous heifers were used to determine the effect of dietary energy on postpartum interval, milk production, serum concentrations of insulin, insulin-like growth factor I (IGF-I), growth hormone, and cerebrospinal fluid (CSF) concentrations of neuropeptide-Y (NPY). Low-quality hay was fed during the last trimester of pregnancy to achieve suboptimal calving weight (370 +/- 5 kg) and condition score (4.0 +/- .1). After calving, cows were allotted to one of four dietary treatments that differed in metabolizable energy. Experimental diets were fed at 2.5% of shrunk body weight and formulated to provide 1.8 (low), 2.1 (maintenance), 2.4 (maintenance high), or 2.7 (high) Mcal of ME/kg DM. Daily milk production was estimated at approximately 30, 60, and 90 d postpartum. Condition score change and weight change were defined as change from calving to d 90 postpartum. As energy intake increased, condition score change (P < .001), IGF-I (P < .001) and insulin (P < .01) increased and postpartum interval decreased (P = .04). No relationship existed between postpartum interval and CSF concentration of NPY (P > .1). Condition score change was positively associated with IGF-I and insulin (r = .71, P < .001; r = .38, P = .02, respectively) and negatively associated with GH (r = -.67, P < .001). Weight change and serum concentrations of GH did not differ (P > .10) among treatments. Increasing dietary energy intake was associated with a curvilinear increase in milk yield (P = .04) and percentage milk fat (P = .03) and a linear increase (P = .04) in energy available for milk production. Greater milk yield at d 30 was associated with a longer postpartum interval (r = .34, P < .05). In conclusion, a greater proportion of net energy is partitioned to maternal tissue realimentation when cows receive high compared to low-, maintenance-, or moderate-energy diets.  相似文献

This study was performed with growing chickens (4 to 22 d of age) to evaluate the effects of feeding a rye-based diet supplemented with commercial enzyme preparation containing xylanase and beta-glucanase (Quatrazyme HP, Nutri-Tomen, France) on small intestine wall morphology, bile acid composition, nutrient digestibility, and bird performance compared with unsupplemented rye- or corn-based diets. The rye-based diet decreased (P < or = 0.05) weight gain, feed intake, and feed efficiency and increased water intake compared with the corn-based diet. Moreover, rye consumption reduced crude fat and protein digestibility as well as apparent metabolizable energy (P < or = 0.05). The small intestine wall showed that villus length, width, and surface were decreased in broiler chickens fed the rye-based diet compared with those fed the corn-based diet. However, crypt morphometry parameters were not affected by diet type. The concentration of conjugated bile acids in the small intestine contents of broiler chickens fed the rye-based diet was decreased (P < or = 0.05) compared with those fed the corn-based diet. These findings suggest that feeding a rye-based diet reduces villus capacity for nutrient absorption and bile acid capacity for fat solubilization and emulsification, resulting in decreased bird performance. The addition of xylanase and beta-glucanase to the rye-based diet improved (P < or = 0.05) weight gain, feed intake, and feed efficiency, and decreased water intake. The digestibility of nutrients and apparent metabolizable energy were also increased (P < or = 0.05). Addition of xylanase and beta-glucanase increased (P < or = 0.05) villus size and the villus height-to-crypt depth ratio, as well as the concentration of conjugated bile acids (P < or = 0.05) in the small intestine contents. Exogenous enzymes improved nutrient digestibility and broiler chicken performance, probably by improving the absorption capacity ofthe small intestine through increased villus surface and intestinal concentration of conjugated bile acids.  相似文献

The objective of this study was to determine the effects of level of feeding on growth, feed efficiency (gain:feed; G:F), body composition (BC), and serum concentrations of somatotropin (ST), IGF-I, and IGF-binding proteins (BP) in growing beef cattle supplemented with bovine (b) ST. In each of two consecutive years, 40 growing beef cattle were blocked by weight (average BW: yr 1 = 316 kg, yr 2 = 305 kg) and used in a 2 x 2 factorial arrangement with main effects of bST (0 or 33 microg x kg BW(-1) x d(-1)) and level of feed intake (ad libitum [AL] or 0.75 AL). Relative to uninjected cattle, treatment with bST increased ADG 9.6% (1.14 vs 1.25 kg/d; P < 0.05) and increased G:F 8.1% (12.3 vs 13.3 gain [g]:feed [kg]; P < 0.05), whereas ADG in AL animals was 39% greater than that in 0.75 AL animals (1.39 vs 1.00 kg/d; P < 0.05). There was a tendency (P = 0.10) for a bST x level of feeding interaction, such that the increase in ADG with bST was greater in AL cattle than in 0.75 AL cattle (10.6 vs 7.8%; P = 0.10). Serum concentrations of ST were greater in 0.75 AL cattle than in AL cattle (13.0 vs 8.6 ng/mL; P < 0.05) and in bST-treated cattle than in uninjected cattle (16.3 vs 5.2 ng/mL; P < 0.05). Due to a bST x level of feeding interaction (P < 0.01), the magnitude of the increase in serum ST to exogenous bST was greater (P < 0.01) in 0.75 AL cattle than in AL cattle. Relative to uninjected cattle, treatment with bST increased (P < 0.05) serum concentrations of IGF-I and IGFBP-3 and reduced (P < 0.05) concentrations of IGFBP-2. Similarly, AL cattle had greater (P < 0.05) serum concentrations of IGF-I and IGFBP-3 and reduced (P < 0.05) IGFBP-2 compared with 0.75 AL cattle. In summary, treatment with bST increased growth rate and G:F and stimulated serum IGF-I and IGFBP-3 while reducing IGFBP-2. Feeding at 0.75 ad libitum intake reduced the magnitude of response for each of these variables. Thus, limit-feeding may reduce the effect of exogenous bST on growth rate by blunting bST-induced increases in IGF-I and IGFBP-3 and bST-induced decreases in IGFBP-2.  相似文献

One-hundred-three multiparous sows were randomly assigned to one of two lactation diets containing either no supplemental animal fat (C) or 10% added fat (F) during two seasons, summer (S) and winter (W), in a 2 X 2 factorial arrangement of treatments. Sows were placed on their respective dietary treatments 1 wk prior to farrowing and were fed these diets ad libitum throughout the 28-d lactation period. Weekly feed intake and total feed intake were not affected by diet or season, while weekly metabolizable energy (ME) intake tended to be higher during week 1 and 3, and was higher (P less than .04) during wk 2 for sows fed diet F. Sow weight loss from farrowing to 21 d of lactation and to weaning (28 d) were unaffected by diet or season. Average pig birth weight was .15 kg higher (P less than .01) for pigs born during S compared with those born in W. Sows receiving diet F had heavier litters at 21 d (P less than .01) and heavier average pig 21-d weights (P less than .01). This was primarily due to the 13.1% increase (P less than .04) in estimated milk yield and the higher fat concentration (P less than .001) of milk consumed by the pigs nursing sows fed diet F. Interval between weaning and rebreeding was shortened by 5.9 d (P less than .01) for sows during W than during S, and tended to be lower for sows fed diet F (7.3 d) compared with that of sows fed diet C (9.7 d).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Research was conducted to examine growth rates, circulating concentrations of IGF-I, and mRNA abundance levels of IGF-I and IGF-II in channel catfish (Ictalurus punctatus) given recombinant bovine ST (rbST; Posilac, Monsanto Co., St. Louis MO). In the first study, juvenile catfish (5.5 +/- 0.5 g) were randomly assigned to one of three treatments: 1) sham-injected control (one needle puncture per week); 2) rbST (30 microg x g BW(-1) x wk(-1); Posilac); and 3) nonhandled control (control). At the end of the 6-wk study, the fish were weighed, measured for length, and G:F was determined. Compared with sham and control treatments, rbST-treated fish had 48% greater final BW, 14% greater total length, and 52% greater G:F (P < 0.001). In the second study, juvenile catfish (41.1 +/- 1.5 g) were assigned randomly to one of two treatments: 1) sham or 2) rbST. Eight fish per treatment were sampled on d 0, 1, 2, 7, 14, and 21 for blood, muscle, and liver. Relative expression of IGF-I and IGF-II mRNA was determined by real-time PCR and plasma concentrations of IGF-I were measured using a validated fluoroimmunoassay. Circulating concentrations of IGF-I were increased (37.9 +/- 5.5 vs. 22.0 +/- 6.6 ng/mL; P < 0.05) in rbST-injected fish compared with sham-injected controls by d 14. Liver IGF-I and IGF-II mRNA was increased 4.3-and 14.4-fold, respectively, by d 1 in rbST-injected fish compared with controls (P < 0.05); however, abundance of liver IGF-I and IGF-II mRNA did not differ from controls on d 0, 2, 7, 14, and 21. Abundance of muscle IGF-I and IGF-II mRNA did not differ in rbST-injected fish compared with controls throughout the study. Results of the first study demonstrated that rbST improves growth performance of channel catfish. Results of the second study showed that the growth-promoting effects of rbST were not mediated by the expression of IGF-I or IGF-II mRNA in the muscle. Instead, the results suggest that rbST promotes growth by stimulating plasma IGF-I release, possibly through its direct effect on the liver or on local tissues to synthesize IGF-I. The changes in mRNA abundance and plasma concentrations of IGF-I support the role of IGF-I in growth regulation of channel catfish.  相似文献

Effects of daily injectable or sustained-release bovine somatotropin (bST) administrations on plasma and milk bST and insulin-like growth factor-I (IGF-I) concentrations were monitored in 74 lactating cows through early, mid- and late lactation. Treatments beginning at wk 4 of lactation were excipient (CO, 24 cows) at 2 wk intervals, daily injections of 10.3 mg bST (DI, 25 cows) and 350 mg sustained-release bST at 2 wk intervals (SR, 25 cows). The duration of treatments was 40 wk. Data were first analyzed for the overall mean concentrations covering the 40 wk treatment period. Overall mean plasma bST, milk bST and plasma IGF-I concentrations were significantly increased by both bST treatments (p<0.05). On the other hand, milk IGF-I concentrations were significantly increased (p<0.05) only in the DI group. Next, data were analyzed according to stage of lactation. The bST treatments resulted in significant increases (p<0.05) in plasma and milk bST concentrations for all early, mid- and late lactation periods. Even though plasma IGF-I concentrations were higher (p<0.05) in all lactation periods for bST treatment groups, higher milk IGF-I concentrations (p<0.05) occurred only in mid- and late lactation periods for the DI group. The patterns of bST and IGF-I concentrations in milk follows those of the plasma after bST treatments.  相似文献

In humans the IGF binding proteins (BP) are closely related to metabolic status. In this paper we have examined the influence of controlled feed intake and GH treatment on IGF binding proteins in growing lambs. Analyses were performed on plasma samples from animals maintained on two levels of feed intake (1.75% body weight as lucerne pellets or 3% body weight which is approximately equivalent to an ad libitum intake) either with or without recombinant bovine growth hormone (BST; 0.25 mg/kg body weight/day) administration. Samples used for the analyses reported in this paper were collected at 9.00 hr following 41 d of treatment. Total plasma IGF-I was increased on the higher plane of nutrition (P less than .01) and by BST (P less than .001) but only on high feed intake. IGF is associated with BP of 150 kDa and 40-50 kDa in sheep plasma. 150 kDa bound IGF-I was increased on the higher plane of nutrition (P less than .05) and by BST treatment (P less than .001) but only on the higher feed intake. By contrast no change in 40-50 kDa bound IGF-I was observed with treatment. Unbound IGF-I was also found in sheep plasma (2-5% of total) but demonstrated only minor changes in relation to treatment. Saturation analysis gave estimates of total binding capacity and saturation of the IGF-BP. In ovine plasma the binding capacity of the 150 kDa species is in excess of bound IGF (P less than .001). Saturation did not change with treatment despite the observed differences in 150 kDa bound IGF-I. Thus BP(s) contained in the 150 kDa fraction were responsive to treatment. By contrast large differences in saturation of the 40-50 kDa species were observed (P less than .001) despite little treatment dependent change in bound IGF-I. IGF-BP(s) in the 40-50 kDa fraction were elevated in the low nutrition group and suppressed on the higher feed intake resulting in near saturation. These data strongly suggest that the IGF BP are modulated according to metabolic status in the sheep.  相似文献

Recombinant bovine somatotropin (bST) was administered at 0, 10.3, or 20.6 mg per cow per day to 32 Holsteins for 38 wk. Fifteen currently treated cows had been treated in the previous lactation. Eighteen hematological variables and blood concentration of insulin-like growth factor I (IGF-I) were measured at five sample periods. The objectives of the study were to test the effects of bST treatments on hematological profiles and to relate blood IGF-I concentration to these variables. Results indicated little influence (P greater than .10) of previous bST treatments on hematological profiles measured in the current lactation. Current bST treatments, however, altered hematocrits (P = .10), mean corpuscular volume (P = .03), mean corpuscular hemoglobin (P = .009), and fraction of blood lymphocytes (P = .09). A week x bST treatment interaction also contributed to variation in hematocrit (P = .09), mean corpuscular hemoglobin (P = .05), fraction of neutrophils (P = .02), and fraction of lymphocytes (P = .04). Blood IGF-I concentration influenced fractions and counts of neutrophils (P = .06, .09), lymphocytes (P = .04, .02), and monocytes (P = .08, .05). Further in vivo and in vitro studies are warranted because this preliminary evidence suggests that bST and(or) IGF-I contribute to regulation of hematopoiesis in mature dairy cows.  相似文献

Yearling Angora wethers (n = 24; 24+/-1.0 kg BW) were used in an experiment with a 2 x 2 factorial arrangement of treatments to investigate effects of bovine somatotropin (bST) treatment and dietary level of ruminally undegraded protein on DMI, ADG, and mohair production. Untreated casein (UC) or casein treated with formaldehyde (TC) was included at 7% DM of a diet containing 11% CP and 46% concentrate. A slow-release bST form was administered weekly to deliver 0 (Control) or 100 microg/ (kg BW.d) of bST. Plasma concentrations of bST and IGF-I were increased (P < .05) during the 7-d period following bST injection. Ruminal fluid ammonia N concentration was lower (P < .01) for TC than for UC before feeding (6.6 vs 7.5 mg/dL) and 4 h later (8.2 vs 12.2 mg/dL), and total VFA concentration was lower (P < .01) for TC than for UC. Treatment with bST decreased (P = .08) DMI with UC (1.15 vs .91 kg/d) and increased (P = .08) DMI with TC (.95 vs 1.06 kg/d). Formaldehyde treatment of casein increased ADG (65, 74, 55, and 91 g/d; P = .03) and clean fleece production (P < .01; 14.1, 17.3, 15.0, and 18.4 g/d for UC-Control, TC-Control, UC-bST, and TC-bST, respectively), with no effect of bST during the 8-wk period of treatment or for the 8 wk thereafter (P > .10). In conclusion, with yearling Angora wethers, bST does not seem useful to enhance mohair production and may not alter effects of dietary level of ruminally undegradable protein on mohair production.  相似文献

Nutritional balance studies were conducted to determine the effect of Eimeria acervulina infection on the retention of nutrients and the efficiency of utilization of protein and energy in the diet during the course of intestinal coccidiosis in chickens. Both light and electronmicroscopic studies were conducted to elucidate the pathogenesis of fat malabsorption in the infected chickens. The infection with E. acervulina during the acute phase of the disease (day 4 to day 8 postinfection) caused a reduction (P smaller than 0.01) in the retention of percent protein, percent ether extract and percent gross energy of the diet with a concomitant increase (P smaller than 0.01) in the concentration of ether extract and gross energy of the excreta. The metabolizable energy value of the diet was also depressed during this phase. During the recovery phase of the disease (day 9 to day 13 postinfection) infected chickens voided excreta containing a slightly greater concentration of ether extract when compared to the controls but differences in the retention of percent protein and percent gross energy were no longer observed. An increased retention of percent phosphorus also occured during the recovery phase. Studies involving the carcass composition revealed that the efficiency of utilization of protein and metabolizable energy of the diet for tissue deposition was reduced (P smaller than 0.05) during day 0 to day 8 postinfection but only the efficiency of metabolizable energy utilization was found to be depressed (P smaller than 0.05) when data for day 0 to day 14 postinfection were analyzed. Both light and electronmicroscopic studies of the duodenal villi of infected chickens during day 5 to day 6 postinfection revealed accumulation of large globules of fat in the villus epithelial cells parasitized by the gammonts of E. acervulina indicating a possible blackade of "fat exit" from these cells.  相似文献

Chopped hay supplemented with a volatile fatty acid salt mixture at 20% of metabolizable energy (ME) was fed to 30 Suffolk X Corriedale lambs (mean wt 29.6 kg) to determine the effects of dietary propionate on weight gain, body composition and plasma metabolites. Propionate accounted for 0, 25, 50, 75 or 100% of the salt mixture energy, and acetate accounted for the remainder. Each diet was fed at two levels of daily intake (158.7 and 130.6 kcal ME/W.75). Lambs on the high level of intake lost more (P less than .05) energy in feces, lost less (P less than .05) energy in CH4 and had carcasses with more (P less than .10) water and fat than lambs on the low intake level. Percentage of propionate in the salt mixture had no effect on ME or body composition of lambs. Increasing the percentage of propionate in the salt mixture increased plasma propionate in blood samples taken at 1 and 2 h after feeding (linear, P less than .05). Empty body weight gain per megacalorie of ME above maintenance increased as the percentage of propionate in the salt mixture increased, but not until propionate reached 75% of the salt mixture (linear, P less than .05; cubic, P less than .05). These results show that propionate in the blood has a positive, but nonlinear, effect on weight gain, even when energy intake is held constant.  相似文献

Previous research indicates that the neonatal pig does not alter feed intake in response to changes in the energy density of manufactured liquid diets. Also, the limited response of IGF-I to exogenous porcine ST (pST) previously observed in young pigs may be influenced by the source of dietary energy. Our objectives were to 1) determine the effect of a high-fat (HF; 25% fat and 4,639 kcal/kg ME; DM basis) or low-fat (LF; 2% fat and 3,481 kcal/kg ME; DM basis) manufactured liquid diet on pig performance; and 2) determine whether the limited response to exogenous pST in young pigs depends on the source of dietary energy. Two replicates of 60 pigs (n = 120; barrows and gilts distributed evenly), with an initial BW of 4,207 +/- 51 g, were weaned from the sow at 10 d of age and used in a randomized complete block design. Pigs were assigned by BW to one of six pens. Diets were formulated to provide a constant lysine:ME ratio and were fed on a pen basis for a duration of 9 d. On d 5, barrows and gilts within a pen were assigned randomly to receive either 0 or 120 microg of pST.kg BW(-1).d(-1) for 4 d. Pigs gained 336 +/- 9 g/d, which resulted in an ending BW of 7,228 +/- 120 g, regardless of dietary treatment (P > 0.15). Pigs fed the LF diet consumed 17% more DM per pen daily than pigs fed the HF diet (2,777 +/- 67 vs. 2,376 +/- 67 g/d, P < 0.01), but calculated ME intake did not differ between dietary treatments (P > 0.20). The G:F was 24% greater in HF- than in LF-fed pigs (P < 0.01). Plasma urea N concentrations were higher in the HF-fed pigs (11.0 +/- 0.6 mg/dL) than in pigs fed the LF diet (6.2 +/- 0.6 mg/dL; P < 0.05). Treatment with pST increased circulating IGF-I (P < 0.01) and decreased PUN (P < 0.01) concentration 32 and 25%, respectively, regardless of dietary treatment (P > 0.30). Circulating leptin averaged 1.8 +/- 0.1 ng/mL and was not affected by dietary treatment (P > 0.35) or pST (P > 0.40). These results suggest that the ST/IGF axis is responsive in the young pig and the increase in circulating IGF-I and growth is independent of the source of dietary energy. Also, young pigs respond to a lower energy density liquid diet with increased feed intake, without altering growth performance, apparently utilizing a mechanism other than circulating leptin.  相似文献

Seven multiparous Holstein cows (>150 d postpartum) were used to evaluate the time course of the chronic adaptation in lipolytic response to adrenergic challenge with bovine somatotropin (bST) treatment. Cows received daily bST (sometribove; 40 mg/d) or excipient injections for 7 d (single reversal design) with a 7-d interim between periods. Epinephrine challenges (1.4 μg/kg body weight intravenously) were administered on Days −2, −1, 1, 2, 3, 5, and 7 of treatment at 10:00 a.m. (15 hr after bST or excipient injection). Frequent blood samples were collected, and concentrations of plasma glycerol (GLY) and nonesterified fatty acids (NEFA) were determined. Treatment with bST increased milk yield 23% (P < 0.05) and milk fat content 33% (P < 0.001) compared with controls. Somatotropin-treated cows entered negative energy balance by Day 3 and had higher basal plasma concentrations of GLY and NEFA than did controls by Day 2 and Day 3, respectively. Response to epinephrine, expressed as area under the response curve corrected for basal, was enhanced by bST treatment, regardless of energy balance. GLY response was greater than control by Day 1 of bST treatment (P < 0.01), and had plateaued by Day 2 (P < 0.001). The NEFA response area was higher than control and had plateaued by Day 1 of bST treatment (P < 0.001). Day 1 represented 15 hr after the first bST injection. Results illustrate that bST treatment results in enhanced in vivo lipolytic response to catecholamine challenge, and the metabolic adaptation is in place by 15 hr after the first bST injection.  相似文献

The effect of exogenous bovine somatotropin (bST) treatment on the temporal pattern of insulin-like growth factor-I (IGF-I) in serum of four multiparous Holstein cows was examined. Cows (190 +/- 24 days postpartum) were treated with daily subcutaneous injections of recombinant bST (40 mg) or excipient for 12-day periods in a crossover experimental design. During excipient treatment, concentrations of IGF-I in serum were relatively constant throughout the day and averaged 70 ng/ml. Following the first bST injection, serum IGF-I began increasing after a lag of 5 to 7 hr and progressively increased over the first 2 days of treatment. Serum IGF-I levels were approximately 2-fold greater than control values at the end of day 1 of bST treatment, with a 3-fold elevation observed at the end of day 2. Concentrations of IGF-I in serum plateaued by day 3 of bST treatment. Serum concentrations of IGF-I did not follow the oscillating pattern of bST in serum resulting from daily bST injections. Milk yield (3.5% fat-corrected) plateaued after 6 days of bST treatment and was increased 61% (+15.3 kg). Both IGF-I and milk yield remained essentially constant across days for the remainder of treatment. Following cessation of treatment, serum IGF-I and milk yield gradually declined, returning to control values after approximately 4 days. The temporal pattern of circulating concentrations of IGF-I is consistent with a role for IGF-I in mediating a portion of the effects of exogenous bST in lactating cows.  相似文献

Eight steers (327 kg average) were fed diets containing either nontreated or anhydrous NH3-treated wheat straw in a two-period crossover design. Intake and digestibilities of dry matter, gross energy, neutral detergent fiber, acid detergent fiber and crude protein were measured by total collection. Energy losses in urine and methane were determined. Heat production was determined by indirect respiration calorimetry on steers fed both diets and after they were fasted. Ammoniation of the straw raised its N content from .49 to 1.59% (P less than .001). Dry matter (DM) intake was increased (P less than .01) by the process from 1.0 to 1.3% of body weight. Digestibility coefficients of dry matter and energy, as well as those for fiber, were increased (P less than .001) by four percentage units or more. Crude protein digestibility, however, was depressed (P less than .001) from 67.8% to 53.5% by ammoniation. Urinary energy as a percentage of gross energy intake (GEI) was reduced (P less than .05) by NH3 treatment from 4.10 to 3.74%. Methane energy was not different (P greater than .10). Metabolizable energy was improved (P less than .001) by ammoniation, increasing from 45.2 to 50.0% of GEI. Daily heat production was higher (P less than .01) for steers consuming the ammoniated straw diet, increasing from 113.7 to 125.3 kcal/wt.75), and was due to higher metabolizable energy intake (MEI) since partial efficiency of MEI used for maintenance did not differ (P greater than .10) between diets. The higher net energy value of the ammoniated wheat straw diet (1.45 vs 1.26 kcal/g DM) was due mainly to decreased fecal loss and a slight decline in urinary loss.  相似文献

Administration of exogenous bovine ST (bST) increases growth rate, feed efficiency, and carcass quality in beef cattle. The magnitude of response to bST in beef cattle is variable and related to the age of the animal. Our objective was to determine the response of the somatotropic axis, in particular IGF-I, IGFBP-2, and IGFBP-3, to bST treatment from birth to 1 yr of age. Blood samples were collected before and after a single injection of bST (500 mg) every 50 d from birth to 1 yr of age in male and female Hereford calves. Body weights and serum concentrations of ST, IGF-I, IGFBP-2, and IGFBP-3 were determined. At birth, serum concentrations of ST, IGF-I, and IGFBP-3 increased (P < 0.05) following bST treatment. From 50 to 350 d of age, average concentrations of ST and IGF-I were greater (P < 0.05) in males, whereas IGFBP-2 concentrations were greater (P < 0.05) in females. No gender differences in IGFBP-3 concentrations were observed. Following bST treatment, IGF-I increased (P < 0.05) from 50 to 350 d of age, IGFBP-2 decreased (P < 0.05) from 50 to 200 d of age, and IGFBP-3 increased (P < 0.05) at 250 d of age. At 250 d of age, baseline concentrations of IGFBP-2 decreased (P < 0.05). Due to the positive response of IGFBP-3 and decreased baseline IGFBP-2 at 250 d of age, we conclude that this is an age at which the somatotropic axis is most responsive to exogenous bST, and it therefore may be an appropriate age to begin bST treatment in beef calves to realize the positive influence of bST on BW gain, feed efficiency, and carcass composition.  相似文献
