Pinctada margaritifera is an economically important marine bivalve species for cultured pearl production in French Polynesian aquaculture. In order to evaluate the influence of donor oyster age on pearl quality traits, experiments were conducted over 6 years using both grafts and surgreffe operations. At harvest, six pearl quality traits were recorded and compared: surface defects, lustre, grade, darkness level and visual colour. Analysing the quality traits of pearls harvested in the initial graft process and those of pearls obtained from surgreffe experiments allowed a comparison of the influence of pearl sac cells originating from the initial mantle graft, which aged together with their recipient oysters. The results demonstrated a significant decrease between these successive grafts in lustre, grade (A‐B‐C), darkness level, and green colour – traits that are of major importance in the pearl market. The duplicated graft experiment allowed the comparison of donor oyster families at 2 and 5 years old, where a mantle graft was inserted into recipient oysters aged 2.5 years. The results showed the same tendencies to a lesser extent, with (i) an improved pearl grade, predominantly through a most important rate of 0 surface defect category, and (ii) a green/grey ratio in favour of the younger donor. A comparison between the graft‐surgreffe and the duplicated graft experiments also highlighted: (i) the indirect role played by the younger recipient oysters, which must be optimized for optimal pearl quality realization, and (ii) the complex interplay between the donor and recipient oysters.  相似文献

The black‐lipped pearl oyster, Pinctada margaritifera, is the most important farmed species in French Polynesia and the basis of the most valuable export industry. Mass production of black pearls relies on a surgical operation requiring tissue from a donor pearl oyster to be grafted, together with a nucleus made of shell, into the gonad of a recipient oyster. Improving pearl size through family selection remains one of the main challenges for future aquaculture development. This study analyses the relative contribution of donor and recipient oysters to pearl size. To this end, hatchery‐produced donor oysters of two batches, large and small (based on shell height), were used to supply grafts for recipients, which were then monitored individually for their growth performance by recording shell height, width, and thickness, and total live weight (flesh + shells) every 6 months (four biometric measurement times) over 20 months of culture. Pearls issued from the two batches of donors showed no significant differences in nacre weight or thickness. In contrast, recipient oyster shell height and total weight were increasingly positively correlated with these pearl size parameters over the culture period, becoming significant at 8 months post‐grafting. Potential therefore exists to use shell height and oyster weight as phenotypic indicators for selective breeding of recipient oysters with high growth performance to increase pearl size in P. margaritifera.  相似文献

In French Polynesia, the aquaculture of Pinctada margaritifera is carried out in numerous grow‐out sites, located over three archipelagos (Gambier, Society and Tuamotu). To evaluate the impact of macro‐geographical effects of these growing sites on pearl quality traits, five hatcheries produced families were used as homogeneous donor oysters in an experimental graft. The molluscs were then reared in two commercial locations: Tahaa Island (Society) and Rangiroa atoll (Tuamotu). At harvest, eight pearl quality traits were recorded and compared: surface defects, lustre, grade, circles, shape categories, darkness level, body and secondary colour and visual colour categories. Overall inter‐site comparison revealed that: (1) all traits were affected by grow‐out location except for lustre and round shape, and (2) a higher mean rate of valuable pearls was produced in Rangiroa. Indeed, for pearl grade, Rangiroa showed twice as many A‐B and less reject samples than Tahaa. This was related to the number of surface defects (grade component): in Rangiroa, twice as many pearls had no defects and less pearls had up to 10 defects. Concerning pearl shape, more circled and baroque pearls were found in Tahaa (+10%). For colour variation, 10% more pearls have an attractive green overtone in Rangiroa than in Tahaa, where more grey body colour were harvested. Lustre does not seem to be affected by these two culture site (except at a family scale). This is the first time P. margaritifera donor family have been shown to vary in the quality of pearls they produce depending on their grow‐out location.  相似文献

A 10‐day experiment was performed to examine different mono, binary and ternary dietary combinations on survival and growth of D‐shaped and umbone black‐lip pearl oyster, Pinctada margaritifera, larvae. The three tropical microalgae species were the flagellate Isochrysis galbana clone T. Iso (CS‐177) and diatoms Chaetoceros calcitrans (CS‐178) and Chaetoceros muelleri (CS‐176) which were fed to D‐shaped and umbone larvae at a density of 7000 and 14 000 cells mL?1, respectively. A second experiment was performed to investigate the feasibility of replacing T. Iso with a lipid emulsion for both D‐shaped and umbone larvae for 10 and 12 days, respectively. The treatments included only T. Iso, unfed and lipid emulsion to substitute T. Iso at levels of 10% (LIP10), 30% (LIP30) and 100% (LIP100). In the first experiment, results showed that a monospecific diet of T. Iso led to significantly higher (< 0.05) survival and growth of D‐shaped larvae than all the other treatments. Meanwhile, D‐shaped larval survival was significantly lower when only fed C. calcitrans as well as growth for those fed C. calcitrans or in combination with C. muelleri. However, for umbone larvae, survival and growth were significantly higher when fed a binary combination of T. Iso and C. muelleri or the ternary combination of T. Iso, C. muelleri and C. calcitrans compared with all other treatments. For the second experiment, results showed that with increasing lipid emulsion replacement, survival of both D‐shaped and umbone larvae significantly decreased (< 0.05); however, the LIP100 treatment was not significantly different (> 0.05) from the unfed treatment. For D‐shaped larvae, no significant growth difference was detected (> 0.05) between the T. Iso and LIP10 fed treatments while for umbone larvae, the T. Iso, LIP10 and LIP30 were not significantly different (> 005). These results indicate that microalgae combinations appear more necessary for later staged P. margaritifera larvae. In addition, the use of a lipid emulsion appeared to provide some nutrition to the larvae, although more research should be conducted to improve the use of such replacements.  相似文献

为探讨育珠对壳基质蛋白基因表达的影响,开展了合浦珠母贝(Pinctada fucata)N19和Prismalin-14基因在育珠贝和非育珠贝中的荧光定量表达研究。结果显示,N19和Prismalin-14在育珠贝与非育珠贝的闭壳肌、肝胰脏、性腺、肠、鳃等组织中均无表达,在外套膜均有表达;N19在珍珠囊表达,而Prismalin-14则不表达。N19勺Prismalin.14在育珠贝外套膜的表达量均大于非育珠贝,表明育珠可能对某些壳基质蛋白的表达具有一定的促进作用。其中Prismalin-14在育珠贝外套膜的表达量最高(P〈0.01),是非育珠贝外套膜Prismalin-14的1.23倍,是育珠贝外套膜N19的25.04倍,是非育珠贝外套膜N19的41.04倍,推测在珍珠贝生长过程中,对Prismalil7-14的需求量可能比N19大。N19在育珠贝珍珠囊中表达量最高(P〈0.01),是育珠贝N19外套膜的11.58倍,是非育珠贝外套膜的18.97倍,推测在珍珠形成的过程中,对N19的需求量可能比贝壳自身生长对N19的需求量大。  相似文献

Cultivated pearls frequently exhibit morphological irregularities making obvious that mineral deposition was irregularly distributed onto nucleus surface. Taking advantage of experimental cultivations with short durations (from 10 days to few months), these irregular deposits predating occurrence of the nacre were investigated in Polynesian pearls by biochemical characterizations and a series of physical methods. Diversity in the resulting data suggests that various in‐depth alterations of the biomineralization mechanism may have occurred during the grafting process, leading to diversity in the biochemical pathways to nacreous deposition. This allows a precise discussion of current views about pearl formation. The “reversed shell theory” is formally disproved through point to point comparison with development of the shell growing edge. Similarity of pearl formation with “regeneration” or “shell repair” is also discussed, emphasizing the differences between these concepts.  相似文献

This paper reports on 5 experiments conducted to assess the effect of cleaning regime and predation on growth and survival of blacklip pearl oyster (Pinctada margaritifera) juveniles in north Queensland, Australia. P. margaritifera juveniles with a mean (±SE) dorso-ventral shell height (DVH) of 4.5 ± 0.1 mm were placed into plastic mesh trays and cleaned either every 4 or 8 weeks or left uncleaned for 16 weeks. Cleaning regime had a significant effect on growth and survival (P < 0.005). Lowest DVH (16.2 ± 1.0) was shown by oysters in uncleaned trays during 16 weeks compared to oysters in cleaned trays; however, there was no significant difference in DVH between oysters held in trays cleaned every 4 (19.4 ± 1.2) or 8 weeks (21.2 ± 0.8). In contrast lowest survival was shown by oysters held in trays that were cleaned every 4 weeks (30 ± 5%), but no differences were noted between oysters cleaned every 8 weeks (63 ± 4%) and oysters that were left uncleaned for 16 weeks (75 ± 8%). Predators of P. margaritifera in northern Australia included crabs, stomatopods, flatworms, gastropods and fish. The stomatopod, Gonodactylus falcatus, was the most destructive predator with individuals consuming in excess of 20 juvenile pearl oysters per week. The leather jacket, Paramonocanthus japonicus, did not kill pearl oysters, but trimmed the margin of oysters shells significantly reducing DVH when compared to control groups cultured without fish. Removing predators monthly had a significant effect on growth of pearl oysters compared to oysters in non-inspected trays; however monthly inspection of culture trays did not significantly improve oyster survival. Clearance rate (CR) and ingestion rate (IR) of different sizes (89, 125 and 188 μm shell length) of Pinctada margaritifera larvae were determined when feeding on various microalgae. The microalgae tested were the diatoms, Chaetoceros muelleri and C. simplex, and flagellates, Tahitian Isochrysis aff. galbana, Pavlova lutheri and P. salina at 5 or 10 cells μL–1. Both CR and IR of microalgae tested in this study increased with increasing larval size; but at all larval sizes, diatoms resulted in lower CR and IR. Of the microalgae tested, P. margaritifera larvae showed greatest CR and IR with the two Pavlova spp. Maximum CR for P. salina was 10.5, 21.2 and 29.7 μL h–1 for larvae with shell lengths of 89, 125 and 188 μm, respectively. The highest IR values for P. margaritifera larvae with shell lengths of 89, 125 and 188 μm were 8.7, 81.0 and 165.7 cells·larva–1 h–1, respectively. CR and IR of P. salina were approximately five times higher than those recorded for C. muelleri and C. simplex.  相似文献

The effects of glycopeptides, prepared from pearl oyster Pinctada fucata, on embryonic development, larval and juvenile growth and adult non‐specific immunity of P. fucata were investigated in this study. Glycopeptides had a pronounced stimulatory effect on embryonic development and larval and juvenile growth of P. fucata. enhancing with increased glycopeptide concentrations. All of haemocytes, phagocytosis, aggregation, serum microbiostatic activity and bacteriocidal activity all showed significant increases after 60‐day feeding, relative to unfed controls. The major conclusion is that glycopeptides had a pronounced stimulatory effect on the non‐specific immunity of pearl oysters.  相似文献

The effects of replacing dietary microalgae with artificial diet on pearl production traits and biomineralization‐related gene expression were investigated in pearl oyster (Pinctada fucata martensii). Three experiment groups (EG1, EG2 and EG3) were set. EG1 and EG3 were separately fed Platymonas subcordiformis and artificial diet (D4), respectively, and EG2 was fed with mixed P. subcordiformis and D4. A control group (CG) was cultured in natural sea. All groups were continuously fed for 150 days. The results showed that the survival rates of EG1, EG2 and EG3 were significantly higher than that of CG. No significant differences in retention rate and pearl thickness were observed among the four groups, but CG had the lowest values of both parameters. The relative expression levels of EGFR and FGF18 mRNA did not significantly differ among EG1, EG2 and EG3. Moreover, the relative expression levels of GHITM and TβR I mRNA significantly varied among the groups and EG3, and EG1 had the highest and lowest relative expression levels of GHITM and TβR I mRNA respectively. The relative expression levels of nacrein and chitin‐binding mRNA significantly differed among the groups, and EG1 had the lowest expression levels of nacrein and chitin‐binding mRNA. No significant differences in the relative expression levels of MSI60 and TIMP mRNA were observed among the groups. The results suggested that D4 can replace part of microalgae, which is helpful for further studies on developing artificial diet of pearl oyster.  相似文献

In an investigation of the potential for pearl production in New South Wales (NSW), Australia, changes in the physical and reproductive condition of the pearl oyster, Pinctada imbricata, were monitored for over 2 years. Using wild oysters gathered from close to the southern extent of the species' range in Port Stephens, NSW, a series of macroscopic and histological observations were made. Reproductive activity in P. imbricata was greatest from late spring to early autumn with oysters in poor reproductive condition during winter. Peaks in reproductive indices occurred in November 1998, March 1999, December 1999 and April 2000. Four indices of physiological condition were used: shell growth, byssal attachment, mantle thickness and mucoprotien layer. With the exception of the thickness and extent of the mucoprotein layer, these indices either showed little variation or the variation that occurred was not related to seasonal or reproductive changes. Changes in the mucoprotein layer were correlated with water temperature and suggest that this layer is metabolized during periods of high demand such as during gonadogenesis. Collectors deployed at two sites in Port Stephens demonstrated that spatfall occurs largely in the months of December and January following the spring and early summer peaks in reproductive activity (November 1998 and December 1999). Spatfall was not observed following the autumnal peaks (March 1999 and April 2000) in reproductive activity. Overall, reproductive patterns in P. imbricata are poorly suited to spat supply in Port Stephens. Farmers require spat in early spring (September) to allow maximum use of the ‘growing’ season (September–May). Reproductively capable oysters are not available from the wild until September and quantity of natural spatfall is too variable and occurs too late in the season (December–January). As a result, oysters are being conditioned in the hatchery in July, spawned in August and spat are supplied to farmers in mid September.  相似文献

The winged pearl oyster, Pteria penguin, is cultured primarily to produce half‐pearls (mabé). The mabé quality is influenced by culture techniques, but there is limited information in this field. P. penguin with mean (±SE) dorso‐ventral height of 250 ± 6.5 mm were used to investigate the influence of culture period and nucleus position on mabé quality. Oysters were relaxed using 1‐propylene phenoxetol, and five nuclei were glued at different positions to the inner surfaces of the oyster shells; three on the more concave left valve and two on the right valve. Nucleated oysters were then cultured for 10 months under commercial pearl farming conditions at Savusavu in Fiji. Nacre deposited at the base and top of the nuclei was measured monthly, from the 6th to the 10th months of culture and the different qualities of mabé produced at different positions were scrutinized. Nacre thicknesses at the base and top of the resulting mabé were significantly different at different months (P < 0.05) and the rate of nacre deposition was highest during the warmer months. The different positions of nuclei on the valve greatly affected the quality of mabé formed. After a 10‐month culture period, around 1 mm of nacre covered the nuclei although the best quality mabé were obtained after 9 months.  相似文献

1. A population of the freshwater pearl mussel Margaritifera margaritifera L. was found in the River Narcea in the province of Asturias (north‐western Spain). Primary analysis of the population shows that it is well distributed along the river and is particularly abundant in shady river banks with a gravel substrate. 2. The length of living mussels ranges between a minimum of 22 mm and a maximum of 116 mm, with most in the size class between 65 mm and 70 mm. There is a good proportion of young mussels, although no fertile females were found in the course of the investigation. 3. Although there is no specific legislation in Spain for the protection of M. margaritifera, it has recently been proposed to include M. margaritifera in the Spanish List of Threatened Species as ‘vulnerable’. 4. Although the River Narcea seems to provide suitable conditions for the pearl mussel (water quality, available salmonid hosts, etc.), the main conservation measures must be directed at avoiding river fragmentation from dams, channelization and organic enrichment from individual farms or towns situated close to the river bank. To understand the relationships between shell growth and some environmental factors, we examined the relationships between water temperature or chlorophyll abundance and the shell growth of the Japanese pearl oyster, Pinctada fucata martensii, suspended at three different depths at two sites. Growth in height, length and thickness of the shells were limited by water temperature during winter (< 20 °C), whereas growth in thickness correlated with food abundance, measured as chlorophyll, during early summer (> 20 °C). These results suggest that the shell of P. fucata martensii could grow well at locations with greater abundance of food and adequate water temperatures (20–26 °C), resulting in a longer growing season.  相似文献

We isolated microbiota from the digestive tract of Nodipecten subnodosus and Pteria sterna and determined in vitro their haemolytic activity, antagonism against Vibrio spp., bacterial hydrophobicity, production of extracellular enzymes and molecular identification. Five bacterial strains were selected: RL5 and C3 (Lactobacillus spp.) and PB1‐1, PB1‐5 and PB1‐6 (Bacillus spp.). The RL5 and C3 isolates showed antimicrobial activity against Vibrio spp. and the PB1‐1, PB1‐5 and PB1‐6 isolates showed enzymatic activity for amylase, protease, lipase and cellulose; the C3 and PB1‐5 isolates were highly hydrophobic. The selected strains of bacteria were tested in vivo as probiotics, together with a treatment of ampicillin and a control without bacteria on juvenile Kumamoto oysters Crassostrea sikamea. The strains were provided individually and as mixes of isolates. Survival, growth and biochemical composition of the juveniles were determined as in vivo indicators. Juveniles grew significantly larger and faster when treated with a specific mix of isolates (MIX‐B), compared with the control. The protein, lipid and carbohydrate concentrations were also significantly higher in oysters exposed to probiotic treatments, compared with the control and the antibiotic treatment. The selected microbiota showed probiotic proprieties for cultivating C. sikamea juveniles.  相似文献

We evaluated growth and survival rates of Pinctadaimbricata in relation to environmental changes during nine months insuspended and bottom culture in the Golfo de Cariaco, Venezuela. Juveniles,measuring 13 mm in length, were cultured using two methods, (1) inSpanish-type baskets suspended at 3–4 m in depth from a longline and (2) in baskets embedded on the seabed (7–8 m indepth). At monthly intervals, we quantified mortality and took a sample ofoysters to determine shell length (dorsal-ventral axis) and the dry mass of theshell, muscle and remaining tissues. We also quantified the dry mass of foulingon the shells (a potentially important environmental factor). Survival rateswere slightly higher in suspended culture (98–100%), although notsignificantly higher than on the bottom. The growth rate was higher in suspendedculture than on the bottom culture, and the difference between culture methodswas greater for tissue and shell biomass than for shell dimensions. At the endof the study, oysters measured 55 mm in shell length for oysters insuspension and 45 mm for those on the bottom. Although growth wasnot significantly correlated with any environmental factor, it tended toincrease with increases in chlorophyll a during periods ofupwelling, thus suggesting that phytoplankton abundance enhanced the growth ofPinctada imbricata. The pearl oyster Pinctadaimbricata should be an excellent species for aquaculture activities,given its high rates of growth and survival in suspended culture and itseconomic importance in the Caribbean region.  相似文献
