Papillomatous digital dermatitis (PDD) is a growing cause of lameness of dairy cattle worldwide. Farms with PDD-afflicted cows experience economic loss due to treatment costs, decreased milk production, lower reproductive efficiency and premature culling. Cows exhibit both humoral and cellular immune responses to PDD-associated spirochetes. This study was undertaken to further characterize the bovine humoral response to PDD-associated spirochetes. Forty-seven sera samples collected from cattle (Field cattle) on three different dairy operations in Iowa were analyzed. In addition, sera were obtained from six young steers (Test cattle) that received a mixed inoculum of four previously isolated Treponema phagedenis-like spirochetes (1A, 3A, 4A and 5B) on two separate occasions. Relative levels of total IgG, IgG1, IgG2 and IgM reactive to each individual spirochete were determined. Field cattle had a higher mean antibody response to 5B compared to the other isolates and T. phagedenis. Test cattle reacted most strongly with 4A following initial exposure, shifting to a greater reactivity with 5B and a reactivity profile similar to field cattle following secondary exposure. No measurable IgM was detected. IgG1 was produced predominately in all cattle. Low to moderate levels of total IgG reactivity to T. phagedenis occurred with sera from all cattle. 相似文献

Outbreaks of papillomatous digital dermatitis (PDD) have been reported from many countries, but there is little information on prevalence and geographic distribution of the disease. Our objectives were to describe prevalence of PDD on dairies belonging to the two main dairy-producer associations in Chile, and to evaluate a milking-parlor screening method for detection of PDD in dairy cows. First, a self-administered questionnaire was sent to all 214 association dairy managers. Seventy percent of the 119 respondents had observed PDD on their dairies. About 83% of the 63 responding managers had first seen PDD during the previous 10 years. Subsequently, a random sample of 43 dairies stratified by association was selected from the total 214 for PDD screening. During milking, we inspected cows' feet for PDD lesions; 91% of dairies had cows affected by PDD; median PDD prevalence for milking cows in dairies was 6.1%. Finally, in one dairy, PDD screening at the parlor was compared to PDD diagnosis by inspecting restrained cows in a chute. The screening method had a sensitivity of 0.72, and a specificity of 0.99. This study demonstrated that PDD was widespread on study dairies in Chile. The screening test presented can be used as a cost efficient method to estimate prevalence of PDD either on individual dairies or in large-scale surveys. 相似文献

Lactating Holstein-Friesian dairy cows in four separately housed groups in a herd with endemic digital dermatitis (DD) were monitored weekly for 4 weeks in December 2004 for the presence of and transition between five stages (M0-M4) of DD. Cows were also monitored for the presence of heel horn erosion (HHE) and interdigital hyperplasia. Prior to the study, two groups had been housed indoors on a high or low energy ration, one group had been grazed and one was a dry cow group. All cows received the same ration during the period of investigation. 'Active infection' was defined as transition from M0, M1, M3 or M4 to M2 and 'resolving M2 lesions' were defined as transition from M2 to another stage. M2 lesions were diagnosed on 106 occasions in the hind claws of 49 (36%) of 138 dairy cows; both hind claws were affected in nine cases (18%). M2 lesions were more often painful on palpation than other stages; 94% of M2 lesions were located plantar-medially near the interdigital cleft and 71% had a diameter of 2-4 cm. More M1 lesions than other stages were found within the interdigital space. When interdigital hyperplasia was present, claws were always concurrently affected by DD. The baseline incidence for 'active infection' was 6% per week, increasing to 11% when HHE was present, 14% when cows were previously housed indoors and fed a high energy ration and 16% when cows were 60-120 days in lactation. Topical treatment with chlortetracycline resulted in resolution from M2 of 79% per week. There were no significant effects of group, stage of lactation, parity or HHE on resolution of M2 lesions. 相似文献

Papillomatous digital dermatitis (PDD) is a polymicrobial infection causing lameness in dairy cattle. Culture-independent analysis has shown that Treponema phagedenis is present consistently and predominantly in the lesions. However, the pathogenesis of PDD, especially the tissue penetration pathway, has not been examined. In the present study, we investigated whether T. phagedenis strains isolated from PDD produce proteolytic enzyme (s) for disruption of the epithelial cell barrier and have the ability to translocate in polarized normal human epidermal keratinocytes (NHEK) in vitro. Ten strains of T. phagedenis isolated from lesions did not show proteolytic activity on modified skim milk agar, although a human strain of T. denticola used as a control showed such activity. The integrity of tight junctions was monitored by measurement of transepithelial electrical resistance (TER). The TER values after inoculation of the T. phagedenis strains examined did not change during the experimental period; however, apical to basolateral translocation of T. phagedenis was confirmed after 24 hr by microscopy and Treponema-specific PCR. We further confirmed that translocation of T. phagedenis was accelerated by co-inoculation with live T. denticola, but not with heat-killed organisms. Furthermore, tight junction ZO-1 protein was not lost intensity after inoculation with T. phagedenis and the organism was observed in NHEK cells using a florescence microscope. These results suggest that T. phagedenis strains may translocate via a transcellular route in vitro and that the invasion is accelerated by other bacteria, such as T. denticola, producing proteolytic activity. 相似文献

Infrared thermography (IRT) was used to detect digital dermatitis (DD) prior to routine claw trimming. A total of 1192 IRT observations were collected from 149 cows on eight farms. All cows were housed in tie-stalls. The maximal surface temperatures of the coronary band (CB) region and skin (S) of the fore and rear feet (mean value of the maximal surface temperatures of both digits for each foot separately, CB max and S max) were assessed. Grouping was performed at the foot level (presence of DD, n = 99; absence, n = 304), or at the cow level (all four feet healthy, n = 24) or where there was at least one DD lesion on the rear feet, n = 37). For individual cows ( n = 61), IRT temperature difference was determined by subtracting the mean sum of CB max and S max of the rear feet from that of the fore feet.Feet with DD had higher CB max and S max ( P < 0.001) than healthy feet. S max was significantly higher in feet with infectious DD lesions (M-stage: M2 + M4; n = 15) than in those with non-infectious M-lesions (M1 + M3; n = 84) ( P = 0.03), but this was not the case for CB max ( P = 0.12). At the cow level, an optimal cut-off value for detecting DD of 0.99 °C (IRT temperature difference between rear and front feet) yielded a sensitivity of 89.1% and a specificity of 66.6%. The results indicate that IRT may be a useful non-invasive diagnostic tool to screen for the presence of DD in dairy cows by measuring CB max and S max. 相似文献

OBJECTIVE: To determine the most relevant aero-allergens involved in canine atopic dermatitis in southeastern Australia and provide information about these aero-allergens to the general practitioner. PROCEDURE: Dogs presented to the Animal Skin & Allergy Clinic with history and clinical signs of atopic dermatitis were injected intradermally with 38 different allergens and negative and positive control. Intradermal skin tests in 1000 dogs were retrospectively evaluated. RESULTS: One third of all patients reacted to the house dust mite Dermatophagoides farinae. Allergens reacting in more than 15% of the patients were wheat (Triticum aestivum), sweet vernal (Anthoxanthum odoratum), English couch (Agropyron repens), yellow dock (Rumex crispus), Mexican tea (Chenopodium ambrosioides), plantain (Plantago lanceolata), melaleuca (Melaleuca quinquenervia) and peppercorn (Schimus spp). CONCLUSION: House dust mites are the most common allergens in canine atopic dermatitis in southeastern Australia and D farinae is involved most frequently. However, a number of grass, weed and tree pollens also are involved regularly. 相似文献

Digital dermatitis (DD) is a major infectious foot disease of cattle worldwide. Some DD stages are associated with lameness, and the disease has significant economic and animal welfare consequences. The pathogenesis of the disease is not yet fully understood, but Treponema spp. have been associated consistently with clinical cases. Isolation of these fastidious bacteria is difficult and cumbersome. We describe an improved method enabling the culturing of the 3 Treponema spp. ( T. pedis, T. phagedenis, and T. medium) from bovine foot specimens derived from DD lesions, using a combination of membrane filtering and subsequent growth on selective agar media. The entire procedure from sampling to verification of individual Treponema spp. takes up to 24 d. In addition, we established a MALDI-TOF MS–based identification method to be applied for confirmation of the different Treponema spp. This scheme provides an unambiguous, simple, and straightforward identification procedure for DD-associated Treponema spp. 相似文献

A study was carried out to determine whether spirochaetes are frequently associated with digital dermatitis in United Kingdom (UK) dairy cattle. Histopathological examination of lesions using a silver stain showed a large number of unidentified spirochaete-like organisms present in digital dermatitis hoof skin tissue in all examined biopsies. Immunocytochemical staining demonstrated that spirochaetes in skin lesions were identified by polyclonal antisera to Borrelia burgdorferi, Treponema denticola and Treponema vincentii (again all biopsies were positively stained), whereas monoclonal antibodies to B. burgdorferi and any Treponema pallidum did not stain any organisms in all biopsies. A PCR of 16S rRNA, previously shown to be specific for a new treponeme, was employed and produced positive results from 82.4% of digital dermatitis tissues. It is concluded that this spirochaete (or related spirochaetes), which is similar to human oral treponemes, is frequently associated with, and may be responsible for, pathological changes in digital dermatitis. 相似文献

Aims: To assess the inter-observer agreement for detecting bovine digital dermatitis (BDD) lesions in digital colour photographs of the hind feet of cows, which had been taken while the animals were standing to be milked, between two trained observers. Methods: Thirty-six photographs were selected from a total of 184 photographs held by the first author (R1), who had classified them as negative (n=11) or positive (n=25) for BDD. They were delivered to a technician (R2) who had previously visually inspected cattle for BDD lesions, and who then recorded the photographs as being either BDD-positive or BDD-negative. The percentage agreement between R1 and R2, and two other inter-observer agreement statistics, Cohen’s κ and Gwet’s first-order chance correction agreement coefficient (AC1), were calculated. The cumulative membership probabilities of Cohen’s κ and Gwet’s AC1 were then calculated for different benchmark ranges of κ. Results: The percentage agreement between R1 and R2 was 33/36 (92%), Cohen’s κ was 0.80 (95% CI=0.57–1.0) and Gwet’s AC1 was 0.86 (95% CI=0.69–1.0). Based on the cumulative membership probabilities for Gwet’s AC1, there was 75% probability that the two observers had almost perfect agreement (κ≥0.81). For both Cohen’s κ and Gwet’s AC1, there was >95% probability that the two observers had at least substantial agreement (κ≥0.61). Conclusions: The two trained observers had at least substantial agreement in identifying from a digital photograph as to whether BDD lesions were present or absent. Therefore results from the two could be used interchangeably. Clinical Relevance: Visual assessment for BDD lesions in the milking parlour can be subjective. However a high agreement between these two trained BDD inspectors means BDD prevalence reported from different regions in New Zealand by these two can be directly compared. 相似文献

Objective Evaluate the survival and prognostic indicators (i.e. breed predilection, season, blood transfusion, and the prevalence of autoagglutination) of dogs with immune-mediated haemolytic anaemia (IMHA) in Victoria, Australia. Design Retrospective study of 110 diagnosed with primary IMHA at the University of Melbourne Veterinary Clinic and Hospital. Results In total, 80 of the dogs (72.7%) were discharged from hospital and 48 of 65 (73.8%) dogs available for follow-up were known to be alive at 1 year, giving an overall 1-year survival of 48 (50.5%) of 95 dogs for which survival data were available. Regarding breed, 80 (18.2%) of the 110 dogs were Maltese-breed dogs compared with 81 (7.4%) of 1100 control dogs (P < 0.001). Springer Spaniels (P = 0.02), Hungarian Vizslas (P = 0.02) and Airedale Terriers (P < 0.001) were also over-represented compared with the control sample. There was no evidence of an association between the occurrence of IMHA in dogs and season in this study. Receiving one or more blood transfusions did not affect survival to the time of discharge from hospital. On initial blood smear examination, 57 (51.8%) of the 110 dogs had spontaneous autoagglutination and its presence was associated with decreased survival to discharge from hospital (P = 0.03). Packed cell volume, white cell count, platelet count and serum total bilirubin on admission had no effect on survival to the time of discharge from hospital or 1 year later. Conclusion Dogs with IMHA have a guarded prognosis as only half are still alive 1 year after the acute event. 相似文献

This retrospective study of papillomatous digital dermatitis (PDD) was performed using data for cows that calved between July 1993 and June 1994 on a Mexicali, Mexico, dairy farm in order to calculate incidence rates, and to compare milk yield and reproduction data between affected and nonaffected dairy cows. A total of 190 out of 577 (33%) cows were affected during lactation and six (1%) during the dry period. Fifty-two of 77 (68%) cows that had lesions in the previous lactation and were also present on the farm in the current lactation had lesions in the current lactation. The highest risk for PDD occurred during the first month of lactation (9%). The yearly estimated cumulative incidence risk was 35% and the incidence density rate was 44.6 cases per 1000 cow-months. More animals were affected in summer and fall than in winter and spring. Purchased animals were 3.4 times more likely to be affected than animals born on the farm. Survival analyses indicated healthy cows conceived 93 days after calving (median), but affected cows conceived 113 days after calving (median) ( P < 0.01). PDD-affected animals produced less milk than healthy cows, but the difference was not statistically significant in the multiple regression. 相似文献

Objective To evaluate if methicillin‐resistant Staphylococcus aureus (MRSA) is present in the horse population in Australia. Design A two‐part retrospective study of laboratory submissions of microbial culture results from horses. Methods Part A: medical records of 216 horses that had MRSA screening performed on nasal swabs collected over a 30‐day period at admission to the Scone Equine Hospital Clovelly Intensive Care Unit were retrieved. Part B: laboratory records from 2004 to 2009 of culture submissions to the Scone Veterinary Laboratory were reviewed and cultures that grew MRSA were identified. The MRSA isolates from Parts A and B were genotyped over an 18‐month period. Results MRSA screening of 216 horses identified eight (3.7%) positive samples. MRSA was isolated from cultures of 80 (0.002%) clinical bacteriology samples over a 6‐year period. Genotypic analysis was performed on 36 isolates. All MRSA characterised had the same pulse field gel electrophoresis pattern (type 1), with eight closely related subtypes identified (subtypes A–F and H) and 66% of isolates classified as subtype D, which multilocus sequence and staphylococcal cassette chromosome mec typing analysis identified as ST612‐MRSA‐IVa, a clonal complex (CC) 8 S. aureus strain. Antimicrobial resistance to more than two classes of antimicrobials was common. Conclusions MRSA was present in a population of horses in Australia. Genotypic analysis of the isolates identified the MRSA strain as CC8 S. aureus. Further research needs to be undertaken to evaluate MRSA infection and colonisation of horses and personnel in Australia. 相似文献

Objective To estimate the prevalence of carriage of methicillin‐resistant Staphylococcus aureus (MRSA) among Australian veterinarians. Methods Individuals attending veterinary conferences in Australia in 2009 were recruited to provide nasal swabs and complete a questionnaire about their professional activities. Swabs were processed by standard methods for detecting MRSA and questionnaire responses were used to group veterinarians according to their areas of major work emphasis (species and practice type). Prevalence was estimated for each of these grouping and contingency tables and regression tree analysis used to explain the variation in MRSA carriage. Results Among the 771 respondents ‘industry and government veterinarians’ (controls) had the lowest prevalence of MRSA carriage at 0.9%. Veterinarians with horses as a major area of work emphasis had a prevalence of 11.8% (13‐fold that of controls) and those whose only major emphasis was horses had a prevalence of 21.4% (23‐fold that of controls). Veterinarians with dogs and cats as a major activity had a 4.9% prevalence (5‐fold that of controls). Prevalence rates for other major activities (pigs, dairy and beef cattle, avian and wildlife) were also increased, but were estimated from smaller numbers of respondents. Regression tree analysis clearly isolated equine veterinarians and dog and cat practitioners as groups at increased risk of carriage of MRSA. Conclusion Carriage of MRSA is a notable occupational health issue for veterinarians in clinical practice in Australia, particularly those who work with horses. 相似文献

The Constitution forming the Australian Commonwealth Government on 1 January 1901 provided that animal and animal products imported into and exported from Australia would be under the authority of the national government. By mutual agreement, the Quarantine Act 1908 provided for the states to continue the delivery of services under contract until 1995 when the Commonwealth took back full responsibility for quarantine services. In the 1940s, 50s and 60s there were world pandemics of livestock diseases and Australia ceased the import of many species. By the 1970s, the livestock industries sought relaxation of import restrictions to gain access to diversified genetic stock. By the use of new technologies, many species can now be imported into Australia through tight importation protocols. With the advent of the World Trade Organization and implementation of the Sanitary Phytosanitary Agreement, Australia has developed a risk-based framework to support the development of import conditions for animals and animal products. Australia's 'Acceptable Level of Protection' has been set to provide a low likelihood of disease entry. Being an island continent, Australia can apply strong controls over imports and exports of all commodities and relatively few outbreaks of exotic animal diseases have occurred by breach of quarantine, but the outbreaks of rinderpest in 1923 and equine influenza in 2007 were notable exceptions. 相似文献

Acute radiation‐induced dermatitis (ARID) is a common sequela of radiation therapy in dogs. There is no consensus regarding ARID management in human medicine and the standard of care in veterinary medicine has not been reported. The objective was to report the practice standards for ARID management in dogs in North America. The design used was a questionnaire survey. Fifty‐eight private and university teaching veterinary hospitals were contacted, 54 participated. The topical and oral medications used to treat ARID, prevent or treat bacterial infection, control pain and the indications for and timing of treatment initiation were the outcome measures. A minority of facilities (4/54, 7.5%) use exactly the same protocol regarding all parameters. There was agreement (>75% of facilities) with respect to the general use of oral antibiotics (77.8%), the need for pain control (92.6%) and the use of tramadol for pain control (76%), although the details of their use varied widely. There is a divergence of opinions regarding all details of ARID management in dogs except the general use of oral antibiotics and pain control medications. 相似文献

In 2012, 2000 questionnaires were sent to a random sample of Welsh sheep farmers. The questionnaire investigated farmers’ knowledge and views on contagious ovine digital dermatitis (CODD) – an emerging disease of sheep responsible for causing severe lameness, welfare and production problems. The overall response rate was 28.3% with a usable response rate of 25.6%. The between farm prevalence of CODD was 35.0% and the median farmer estimated prevalence of CODD was 2.0%. The disease now appears endemic and widespread in Wales. Furthermore, there has been a rapid increase in reports of CODD arriving on farms since the year 2000. Risk factors for CODD identified in this study include the presence of bovine digital dermatitis (BDD) in cattle on the farm and larger flocks. Farmers also consider concurrent footrot/interdigital dermatitis, buying in sheep, adult sheep, time of year and housing to be associated with CODD. Further experimental research is necessary to establish whether these observations are true associations. 相似文献
