Chickens and Japanese quail (Coturnix japonica) have traditionally been the primary avian models in developmental biology research. Recently, the blue‐breasted quail (Coturnix chinesis), the smallest species in the order Galliformes, has been proposed as an excellent candidate model in avian developmental studies owing to its precocious and prolific properties. However, data on the embryonic development of blue‐breasted quail are scarce. Here, we developed a normal developmental series for the blue‐breasted quail based on developmental features. The blue‐breasted quail embryos take 17 days to reach the hatching period at 37.7°C. We documented specific periods of incubation in which significant development occurred, and created a 39‐stage developmental series. The developmental series for the blue‐breasted quail was almost identical to that for chickens and Japanese quail in the earlier stages of development (stages 1–16). Our staging series is especially useful at later stages of development (stages 34–39) of blue‐breasted quail embryos as a major criterion of staging in this phase of development was the weight of embryos and the length of third toes.  相似文献

Poultry diets are formulated with additional animal fat or vegetable oils to improve growth rate and feed conversion efficiency. High‐fat diet feeding in rats and fish has been shown to result in alterations in the phospholipid composition and cholesterol content of the erythrocyte membrane, in turn affecting erythrocyte osmotic fragility. In contrast, the few studies performed using high‐fat diet feeding in avian species show no changes in erythrocyte osmotic fragility. This study made use of the Japanese quail as no data exists on investigation of this species with respect to high‐fat diet feeding and erythrocyte osmotic fragility. Fifty‐seven male quail were randomly divided into six groups and fed either a standard diet (commercial poultry feed) or one of five high‐fat diets (commercial poultry feed with 22% of either coconut oil, lard, palm oil, soya bean oil or sunflower oil on a weight/weight basis) for 12 weeks. All birds on the high‐fat diets were significantly heavier (p < 0.05) after the 12‐week feeding period, than when commencing the dietary intervention. Serum triglyceride concentrations of birds in all high‐fat diet groups were significantly lower (p < 0.05) than birds in the standard diet group, whereas only birds in the palm oil group had significantly lower (p < 0.05) serum cholesterol concentrations compared to the standard diet group. Fragiligrams of erythrocytes from birds in the various dietary groups were similar. High‐fat diet feeding with different types of additional fat did not affect the osmotic fragility of the quail erythrocytes. Feeding quail high‐energy diets of varying degrees of fatty acid saturation was well tolerated and did not seem to affect the overall health status of the birds. Resistance of avian erythrocytes to modification by excess dietary fat may be a general characteristic of avian erythrocytes.  相似文献

Adipose triglyceride lipase (ATGL) is the rate-limiting enzyme of lipolysis in chicken adipose tissue. Its regulation is not fully understood. Recent studies suggest ATGL may be regulated by physical protein-protein interactions. Comparative gene identification 58 (CGI-58) has been identified as an activator of ATGL in mice. The purpose of the current study was to clone and sequence the CGI-58 gene in avian species and to investigate its regulation during development, fasting, and refeeding. Here, we report the cloning and sequencing of the complete coding sequence of CGI-58 and the deduced AA sequences for the domestic chicken, turkey, and Coturnix quail. The CGI-58 protein is a 343-AA protein in the chicken and quail, and a 344-AA protein in the turkey. Sequence comparisons with the human and mouse show that the CGI-58 gene is highly conserved among avian and mammalian species, with complete identities at the predicted lipid-binding site. Cell fractionation of chicken fat cells and stromal-vascular cells revealed that CGI-58 is expressed primarily in mature adipocytes (P < 0.01). When compared in multiple organs and tissues, avian CGI-58 is expressed predominantly in the adipose tissue (P < 0.001), similar to ATGL. To understand CGI-58 expression during adipose tissue development, its mRNA expression was measured along with ATGL and stearoyl CoA desaturase (SCD-1) mRNA, an adipogenic marker, in embryos and adults. Messenger RNA expression of CGI-58 increased (P < 0.05) immediately after hatching, concurrent with peak ATGL expression. It is interesting that CGI-58 remained somewhat increased at posthatch d 11 and 33 as SCD-1 mRNA expression increased (P < 0.05). To evaluate the response of CGI-58 to nutritional status, chickens and quail were fasted for 24 h and subsequently refed. After the fasting period, CGI-58 mRNA was induced (P < 0.05) for both chickens and quail and was returned to control levels upon refeeding. The ATGL mRNA responded similarly, increasing dramatically after fasting and quickly decreasing with refeeding. The direct relationship between CGI-58 and ATGL mRNA expression indicates a role for CGI-58 in activating ATGL-mediated lipolysis in avian species.  相似文献

Harvesting, handling and transporting quails to the slaughterhouses, other farms and laboratories might covertly reduce their welfare. The aim of this study was to evaluate the effects of two major sources of energy in poultry nutrition on reducing transportation stress in Japanese quail (Coturnix Coturnix japonica). Male quails (n = 60) were divided into two groups. The first group was fed corn‐based diet, and the second was fed wheat‐based diet supplemented with xylanase and phytase. At the end of the experiment (day 35), quails were subjected to 80 km of transportation. Immediately on arrival and after 24 h, heterophil counts, lymphocyte counts and H:L ratios were measured. On arrival, H counts were lower, L counts were higher, and H:L ratios were lower for corn‐fed group. After 24 h, wheat‐fed group showed lower increment of H counts, greater increment of L counts and also decrement of H:L ratios rather than corn‐fed group which showed increment of H:L ratios. However, these ratios were still lower in corn‐fed group. Results indicate that corn‐based diets can help Japanese quail to better resist transportation stress, although it seems that feeding wheat‐based diets supplemented with xylanase and phytase could have positive effects for coping better with stress after journeys.  相似文献

Contrary to humans, most ovarian tumors in other species do not arise from the ovarian surface epithelium but are of follicular‐, stromal‐ or germ‐cell origin. One of the few species where ovarian cancer arises from the ovarian surface epithelium (OSE) is chicken (Gallus domesticus). Little is known about the morphology of the OSE in other avian species. In our study we analyzed the OSE morphology of Japanese quail (Coturnix japonica) using ultrastructural and histochemical techniques. Carbohydrate residues have been studied by using a panel of fluorescein isothiocyanate labeled lectins. Japanese quails are commonly used animal models in biomedical research as their housing is comparatively inexpensive and they show a short generation interval. Our ultrastructural and histochemical results demonstrate that the quail ovarian surface epithelium shows characteristic features which resemble the epithelia of both chicken and human. Additionally, the ovarian surface epithelium of the Japanese quail contains cytokeratin as well as vimentin intermediate filaments in their cytoplasm. Therewith and among other parts the quail OSE shows many characteristic features also seen in those of humans, which may qualify quail's ovary as a potential animal model for human ovarian carcinomas.  相似文献

A comparative analysis of skeletal muscle structure reveals that production species (nine species, representing three mammalian families and an avian family) have mitochondrial volume fractions (MVF) 37% lower than the non‐production species at equivalent size (17 species, with representatives from 10 mammalian families) ( Fig. 1 ; F1,25 = 4.79; p = 0.039). As MVF provides evidence of oxidative capacity, this comparative analysis indicates that production animals share an exceptionally low oxidative capacity muscle phenotype. A possible bioenergetic reason for this observation, relating to a reduction in the cost of maintaining trans‐membrane ion gradients is briefly discussed. This discussion is framed within a biological economic design theory called symmorphosis and makes predictions about avenues for improvements in livestock bioenergetics.

Liver‐expressed antimicrobial peptide 2 (LEAP‐2) is a cationic peptide that plays an important role in innate immunity for host defense. The aim of this study was to characterize the LEAP‐2 gene in the Japanese quail (Coturnix japonica). Japanese quail LEAP‐2 (CjLEAP‐2) was identified from the Japanese quail draft genome database by a local BLAST analysis using chicken LEAP‐2 (GgLEAP‐2). The exon‐intron structure of CjLEAP‐2, analyzed from three quails, is composed of three exons, as is the chicken LEAP‐2 homolog (GgLEAP‐2). An analysis of the coding sequence revealed that CjLEAP‐2 is 231 bp long, like GgLEAP‐2, and 93% identical to GgLEAP‐2 at the nucleic acid level. The predicted amino acid sequence of CjLEAP‐2 contained the liver‐expressed antimicrobial peptide 2‐precursor domain and four cysteine residues characteristic of the LEAP‐2 protein. The amino acid sequence of the mature peptide of CjLEAP‐2 was 100% identical to that of GgLEAP‐2. We confirmed that CjLEAP‐2 was transcribed in at least seven tissues, including the digestive system. Additionally, the mature peptide region of CjLEAP‐2 exhibited no polymorphisms in 99 quails from six strains. Taken together, these findings indicate that CjLEAP‐2 is non‐polymorphic and therefore, it likely plays an important role in the innate immunity of quail as it does in chicken.  相似文献

Skeletal chondrofication, ossification and growth of turkey embryos were investigated and analysed to enable assessment of the developmental status and evaluation of the experimental effects on skeletal development, skeletal mutations and development of cultured embryos. Ten embryos were prepared every 24 h from 8 to 28 days of incubation. The fixed embryos were cleared and stained in toto with Alcian blue & Alizarin red for cartilage and ossified components, respectively. Observation of the skeleton was performed under a stereoscopic microscopy, with special attention to the timing of chondrofication and ossification of the bones. The first occurrence of the primary ossification centres was observed in the femur, tibiotarsus, and the dentary and supra-angular of the mandible on the 12th day, followed immediately by the other long bones. Skeletal features of the skull were determined to show the latest appearance of cartilage and ossification. Hence, all elements of the hyolingual apparatus remained cartilaginous until hatching took place except for the ceratobranchial. Even though the vertebral column chondrified earlier as compared with the ribs and sternum, they ossified later. While chondrofication was present in all the regions of the vertebral column at the same time, ossification progressed from the cervical through caudal regions. The growth rate of the femur was eminently higher than that of the humerus with increase in time, particularly after the 20th day of incubation. This seems to be obviously natural because the eggs used in the study are from the broiler turkey, which gains giant muscle mass at a very short period; precocity is probably at the expense of the bones of the leg rather than those of the wing.  相似文献

On day 9 or 12 of the hatching period different pesticides (parathion, methyl-parathion, carbendazim, 2,4-D-amine Na, phosmethylane) were applied in ecotoxicological trials. The formulations were either injected into the air space of pheasant, quail or hen eggs or hen eggs were treated by the immersion technique. The residues of pesticides were measured in samples on days 13, 14 and 16 of incubation of chicken and pheasant embryos, while the Japanese quail embryos were analysed on days 10-14 of incubation. Analytical chemistry data showed a varying degradation rate of the compounds in avian embryos of the same species. The residues directly affect the embryos, disturbing their normal development and causing pathophysiological and morphological changes.  相似文献

The onset of ossification centres of the pectoral girdle and wing skeleton of the quail (Coturnix coturnix japonica) has been studied in embryos and juvenile birds, which were sampled daily from the 4th day of incubation up to the 12th day after hatching. Specimens, which were cleared and were stained with Alcian Blue and Alizarin Red S, were examined at the stereomicroscope. The furcula ossified with intramembranous ossification at the 7th day of incubation. The first centres of perichondral ossification in the scapula and the coracoid bones were observed at the 7th day of incubation. In the humerus, radius and ulna, perichondral ossification was observed at the 6th day of incubation. The carpal bones were ossified between the 8th and 10th day of the post-hatching period. The 2nd metacarpal bone began to ossify at the 6th day of the post-hatching period, whereas the 3rd and 4th metacarpal bones were ossified at the 7th day of incubation. At the 9th day of incubation, ossification was observed in the 1st phalanx of the alular digit and in the phalanges of the major digit. The 2nd phalanx of the alular digit began to ossify at the 12th, whereas the phalanx of the minor digit at the 14th day of incubation. The data presented here provide useful baseline information on the normal sequential pattern of ossification in the pectoral girdle and wing skeleton in this species.  相似文献

This study was conducted in two separate experiments. Experiment I was conducted to investigate the effects of hemp seed (HS) on meat quality traits, and experiment II was designed to determine egg fatty acid (FA) composition in Japanese quail (Coturnix coturnix japonica). In experiment I and experiment II treatments, groups were as follows: (i) control (not included HS0), (ii) 5% HS inclusion to diets, (iii) 10% HS inclusion to diets and (iv) 20% HS inclusion to diets. In experiment I, a total of 192 7‐day‐old‐quail were fed with HS diets for 5 weeks, and end of the experiment I, a total of 64 quail (16 each) slaughtered and meat samples were analysed for meat colour, pH, cooking and thawing loss and FA composition. In experiment II, a total of 120 8‐week‐old laying quail were fed with experimental diets for 6 weeks and egg FA composition were determined. Breast meat cooking loss was significantly lower in 20% supplemented group (p < 0.01). Hemp seed inclusion to diets caused a linear and cubic increase in redness (a*) of hip meat (p < 0.01). Palmitoleic and oleic FAs were decreased with HS addition in breast meat (p < 0.05); however, linoleic and linolenic acid contents of meat were linearly increased (p < 0.01). In experiment II, the linoleic and linolenic FA contents of egg samples were linearly increased with increasing the dietary HS ratio (p < 0.01). Hemp seed addition was provided lower palmitoleic and oleic FAs in quail eggs (p < 0.05). In conclusion, HS supplementation to quail diets may increase quail meat redness and meat and egg yolk omega‐3 FA content and decrease cooking loss of quail meats.  相似文献

Quail (Coturnix coturnix japonica) and broiler (Gallus domesticus) chicks were inoculated experimentally with IBH virus (avian adenovirus-1) derived from quails to determine its pathogenicity. Quail chicks were inoculated by the intraperitoneal route at 3, 4, 5, 6 or 7 weeks of age. Lesions were encountered most frequently in the liver, kidneys and lungs. These included pale, swollen and mottled liver, swollen nephrotic kidneys, and congested and pneumonic lungs. The lesions were severe in birds inoculated at 5 weeks of age. Large basophilic intranuclear inclusion bodies were seen in hepatocytes and occasionally in the renal epithelium. The results showed that this isolate is pathogenic for quails above 3 weeks of age. Broiler chicks were inoculated at 4 weeks of age by the intraperitoneal route. The lesions produced in these chicks were similar to those of adenovirus-induced inclusion body hepatitis. The present study sought to assess the potential application of avian models with different developmental modes to studies on cognition and neuroscience. Six altricial Bengalese finches (Lonchura striata var. domestica), and eight precocial blue‐breasted quails (Coturnix chinensis) were presented with color discrimination tasks to compare their respective faculties for learning and memory retention within the context of the two developmental modes. Tasks consisted of presenting birds with discriminative cues in the form of colored feeder lids, and birds were considered to have learned a task when 80% of their attempts at selecting the correctly colored lid in two consecutive blocks of 10 trials were successful. All of the finches successfully performed the required experimental tasks, whereas only half of the quails were able to execute the same tasks. In the learning test, finches required significantly fewer trials than quails to learn the task (finches: 13.5 ± 9.14 trials, quails: 45.8 ± 4.35 trials, P < 0.05), with finches scoring significantly more correct responses than quails (finches: 98.3 ± 4.08%, quails: 85.0 ± 5.77% at the peak of the learning curve). In the memory retention tests, which were conducted 45 days after the learning test, finches retained the ability to discriminate between colors correctly (95.0 ± 4.47%), whereas quails did not retain any memory of the experimental procedure and so could not be tested. These results suggested that altricial and precocial birds both possess the faculty for learning and retaining discrimination‐type tasks, but that altricial birds perform better than precocial birds in both faculties. The present findings imply that developmental mode is an important consideration for assessing the suitability of bird species for particular experiments.  相似文献

In this study, we identified a cluster of 14 avian β‐defensins (AvBD; approximately 66 kbp) in the Japanese quail, Coturnix japonica. Except for AvBD12 (CjAvBD12) and ‐13, the CjAvBDs coding sequences exhibited greater than 78.0% similarity to the respective orthologous chicken AvBD genes (GgAvBD). The putative amino acid sequence encoded by each CjAvBD contained six cysteine residues and the GXC (X1‐2) motif considered essential for the β‐defensin family. Each CjAvBDs also formed a sub‐group with the respective orthologous genes of various bird species in a phylogenetic tree analysis. Synteny between the CjAvBD cluster and GgAvBD cluster was confirmed. The CjAvBD cluster was mapped on the long‐arm end of chromosome 3 by linkage analysis based on single nucleotide polymorphisms (SNPs) of CjAvBD1 and CjAvBD12 (approximately 46kbp), as well as GgAvBD cluster. We also confirmed that CjAvBD1, ‐4, ‐5, ‐9, and ‐10 are transcribed in 20 tissues, including immune and digestive tissues. However, our experimental data indicated that the CjAvBD cluster lacks the AvBD3 and ‐7 loci, whereas the CjAvBD101α, ‐101β, and ‐101θ loci arose from gene duplication of the AvBD6 orthologous locus in the CjAvBD cluster after differentiation between Coturnix ‐ Gallus.  相似文献

ABSTRACT

Fast-growing and highly adaptable avian species such as the Japanese quail (Coturnix coturnix japonica) have greater potential for use in alleviating food and nutrition insecurity. However, the continued use of soybeans as a protein source may reduce the contribution of quails to food and nutrition security. It is, therefore, prudent that alternative dietary proteins, with no direct food value for humans, be evaluated to allow for sustainable intensification of quails. One such protein source is canola meal, a by-product of oil extraction from canola seed. This review explores the potential of canola meal as a replacement for soybean meal in quail diets by presenting a comparative analysis of the composition and functional properties of the two proteins. A balanced assessment of in vivo responses to canola meal in quails and other avians is presented with the objective of enhancing the quail's contribution to food and nutrition security.  相似文献

Cell‐cell fusion has been a great technology to generate valuable hybrid cells and organisms such as hybridomas. In this study, skeletal muscle myoblasts were utilized to establish a novel method for autonomous xenogenic cell fusion. Myoblasts are mononuclear myogenic precursor cells and fuse mutually to form multinuclear myotubes. We generated murine myoblasts (mMBs) expressing green fluorescent protein (GFP) termed mMB‐GFP, and the chick myoblasts (chMBs) expressing Discosoma red fluorescent protein (DsRed) termed chMB‐DsRed. mMB‐GFP and chMB‐DsRed were cocultured and induced to differentiate. After 24 h, the multinuclear myotubes expressing both GFP and DsRed were observed, indicating that mMBs and chMBs interspecifically fuse. These GFP+/DsRed+ hybrid myotubes were able to survive and grew to hyper‐multinucleated mature form. We also found that undifferentiated mMB‐GFP efficiently fuse to the chMB‐DsRed‐derived myotubes. This is the first evidence for the autonomous xenogenic fusion of mammalian and avian cells. Myoblast‐based fusogenic technique will open up an alternative direction to create novel hybrid products.  相似文献

Age-related patterns of tissue lipid peroxidation were characterised in brain, liver and heart tissue of developing Japanese quail (Coturnix japonica) embryos between 9 and 15 d of age, over which embryo mass increased by a factor of 6. Lipid peroxidation was quantified in each tissue by spectrophotometric measurement of malondialdehyde using the thiobarbituric acid reactive substances assay.

  3. In all tissues, lipid peroxidation increased greatly as development proceeded. Concentrations of malondialdehyde increased in parallel with the cumulative amount of oxygen consumed by the developing embryo consistent with the hypothesis that oxidative stress results from the production of free radicals due to oxidative metabolism.

  4. This study describes in vivo oxidative stress in developing avian embryos and suggests that rates of embryonic growth, oxidative metabolism and oxidative damage likely vary in parallel.

Computer‐assisted systems for the assessment of sperm morphometry (ASMA systems) have been used successfully with several mammalian species. Unfortunately, they have so far been of little use for assessing bird semen, a consequence of the filiform shape of avian spermatozoa. This study compares two staining techniques (Hemacolor® and aniline blue staining) for the morphometric analysis of rooster and red‐legged partridge spermatozoa as part of a computer‐assisted light microscopy method. For both species, Hemacolor® staining provided a significantly higher percentage of measurable cells (93.7 ± 11.7% in roosters and 71.9 ± 15.3% in red‐legged partridges). Hemacolor® also showed greater repeatability (lower coefficients of variation) for length and area in roosters' sperm and for width in the case of red‐legged partridge's sperm. In the roosters, the Hemacolor® technique returned significantly (p < 0.05) larger sperm head width and area values than did the aniline blue technique, while the latter resulted in greater sperm head length values (p < 0.05). In the red‐legged partridge, no differences were seen in the results for sperm head width and area provided by the two techniques, but aniline blue staining was associated with longer length measurements. In conclusion, the morphometric values recorded differed depending on the staining method and species. However, the Hemacolor® technique might be deemed the more appropriate for computerized sperm assessment systems as it provides larger percentages of measureable cells and shows greater repeatability.  相似文献

Long non‐coding RNAs (lncRNAs) participated in growth and development of skeletal muscle; however, little is known about their response to testosterone deficiency in porcine skeletal muscle. We compared lean mass related carcass traits and lncRNAs expression files in Longissimus dorsi (LD) muscle between intact and castrated Huainan male pigs. The results showed that castration significantly reduced eye muscle area and lean meat percentage (P < 0.05), but increased the fat mass weight (P < 0.05). Meanwhile, 8946 lncRNAs, including 6743 intergenic lncRNAs (lincRNAs), 498 anti‐sense lncRNAs, and 1705 intronic lncRNAs, were identified in porcine LD, among which, 385 lncRNAs were considered as the differentially expressed candidates between intact groups and castrated groups (q‐value < 0.05). Functional analysis indicated that these differently expressed lncRNAs and their target genes were involved in the estrogen receptor signaling pathway and skeletal and muscular system development and function. We first detect porcine muscular lncRNA response to castration, and the results suggested that lncRNAs and their target genes participated in the regulation of testosterone deficiency‐related skeletal muscle growth.  相似文献

One of the main hypotheses proposed to explain the evolution of fruit color deals with a preference of avian frugivores for specific colors, mainly black and red, which are the most common fruit colors in many of the studied habitats. I analyzed fruit color preferences by wild birds belonging to 2 species of the highly frugivorous genus Turdus (Eurasian Blackbird Turdus merula Linnaeus, 1758 and Redwing Turdus iliacus Linnaeus, 1758) by means of captivity experiments with artificial fruits. Despite important within‐individual (i.e. temporal) and among‐individual variability, consistent patterns of species‐specific color preferences emerged. Eurasian Blackbirds tended to prefer red over blue, green and black, whereas Redwings seemed to prefer black over the rest. Green was systematically avoided by both species, suggesting that it might signal unripeness of fruits. Both preferred colors have been previously reported as the most common among fleshy‐fruited plants. The high variability, both within and between individuals, in preferences suggests that they can be subject to changes through experience and learning and, therefore, are not likely to drive the evolution of fruit color. The main differences between both species could be related to the most common fruit color they fed upon during the last months before capture.  相似文献
