Forty-two ewes had an intravenous catheter sewn in place in a prepared area over the jugular vein and beginning at 60 days of gestation received an infusion 3 times daily. The infusion consisted of sterile saline or sterile saline containing arginine. Twenty-six ewes in both control and treatment groups aborted between 81 days of gestation and term. Fetuses from 16 ewes that aborted were examined. Most were autolyzed or had early mummification. Macroscopic placentitis and noncollapsing lungs were noted. Large numbers of coagulase-positive Staphylococcus were isolated from fetal abomasal content, lungs, brains, or placentas. Histologically, suppurative placentitis with necrosis and pulmonary aspiration of meconium and amniotic debris often with suppurative bronchopneumonia were observed in abortuses. Four ewes euthanized and examined after abortion had suppurative endometritis. Three ewes had severe, chronic, jugular thrombophlebitis from which coagulase-positive Staphylococcus was isolated. The fourth ewe had mild phlebitis, and Staphylococcus aureus was isolated from both the catheter and the blood. Catheter-associated staphylococcal abortion was diagnosed.  相似文献

Conventionally-delivered colostrum-deprived lambs were inoculated with either parainfluenza virus type 3 (PI3) alone or PI3 followed, 6 days later, by Pasteurella haemolytica (P.h.). Six out of 20 lambs died or were killed in extremis within 3 days of the inoculation of P.h.; the remainder were selected at random and killed from 1 to 28 days after the inoculation of P.h.Extensive pneumonic lesions developed in a large proportion of lambs inoculated with both agents but in none of those inoculated with the virus alone. Histologically, the pneumonic lesions fell into two categories: necrotic lesions, demarcated by a zone of either oat-cell or neutrophil infiltration, and a milder, purulent bronchopneumonia. Bacterial numbers tended to be higher in necrotic lesions than they were in purulent lesions. Virus titres in nasal secretions, on the day of inoculation of P.h. (day 6), also were higher in animals that developed necrotic lesions than they were in those that developed milder lesions. Nevertheless, titres were similar in both groups on day 4.Necrotic lesions persisted for at least 21 days as residual encapsulated abscesses which still contained viable P.h. whereas the milder, purulent bronchopneumonia was not detected later than 3 days after the inoculation of P.h..  相似文献

Abortions due to Campylobacter fetus fetus (C. fetus fetus) were diagnosed in three Canterbury ewe flocks 6 weeks prior to lambing. In two of the flocks, two inoculations of a C. fetus fetus bacterin, 10 days apart, reduced the incidence of abortions in the treated ewes to about one third and one half respectively of the level in the control ewes in the same flock. The treatment had no effect in a third flock where an outbreak had been in progress for 2 weeks before investigations started. The results confirm earlier Scottish work where pregnant sheep were inoculated with a C. fetus fetus vaccine following artificial challenge with C. fetus fetus. This inoculation significantly reduced the number of C. fetus fetus abortions. The results also indicate that treatment must start very early in an outbreak.  相似文献

The passage of non-Toxoplasma antibodies from dam to fetus through damaged placenta was studied in sheep inoculated with Toxoplasma gondii. Six ewes were inoculated with chicken globulins and Leptospira bacterins 2 months before oral inoculation with Toxoplasma gondii oocysts. Ewes were euthanatized between 42 and 62 days after T gondii inoculation. Antibody titers against chicken globulins, Leptospira spp, Haemonchus contortus, Sarcocystis spp, and T gondii were measured in the maternal and fetal sera. All ewes became infected with T gondii and had grossly visible necrotic foci in the placentas, and T gondii antibodies were found in the fetuses and the ewes. Appreciable amounts of antibodies to Haemonchus contortus, Sarcocystis sp, Leptospira spp, and chicken globulins did not cross the placental barrier. Seemingly, serologic examination of the fetus was reliable for the diagnosis of ovine congential toxoplasmosis.  相似文献

An immunohistochemistry (IHC) procedure for the detection of Campylobacter fetus antigens using an avidin-biotin complex technique was performed on formalin fixed bovine and ovine fetal tissues from 26 natural cases of Campylobacter spp. abortion (four ovine and 22 bovine). The species of Campylobacter isolated included C. fetus ssp. venerealis from 13 bovine fetuses, C. fetus ssp. fetus from two ovine and one bovine fetus, Campylobacter jejuni from seven bovine fetuses, Campylobacter lari from two ovine fetuses and an unspeciated Campylobacter species in one bovine fetus. Histologic lesions identified in the aborted fetuses included placentitis, serositis, pneumonia, gastroenteritis, hepatitis and encephalitis. Campylobacter fetus antigens were identified by IHC in 13 of 13 bovine fetuses from which C. fetus ssp. venerealis was isolated and in two of two ovine fetuses from which C. fetus ssp. fetus was isolated. The IHC stains were negative in tissues from seven bovine fetuses from which C. jejuni was isolated, one bovine fetus infected with C. fetus ssp. fetus, one bovine fetus infected with the unspeciated Campylobacter and two ovine fetuses infected with C. lari. In positive cases, the IHC stain most frequently identified bacteria in the lung and gastrointestinal tract. The C. fetus IHC procedure performed on formalin fixed tissues is a practical tool for the diagnosis of natural cases of ovine and bovine abortion caused by C. fetus.  相似文献

An outbreak of acute encephalopathy occurred in pregnant ewes and their newborn lambs associated with consumption of Talisia esculenta fruits and bark. Clinical signs in 5 adult pregnant ewes included drooling, bloat, tachypnea, depression, ataxia, body shaking, difficulty in rising, and recumbency. Three neonatal lambs born to some of those ewes had similar clinical signs. No significant gross abnormalities were observed on autopsy. Histologically, neuronal necrosis, axonal and dendritic swelling, and loss of Purkinje neurons were observed in the cerebellum. The observation of similar neurologic clinical signs and lesions in pregnant ewes and their neonatal lambs suggests that the toxic principle of T. esculenta crosses the placenta and reaches the fetus.  相似文献

The present study was aimed at investigating the effect of experimental infection by Trypanosoma vivax in different stages of pregnancy, determining the pathogenesis of reproductive failure, and confirming transplacental transmission. We used 12 pregnant ewes distributed into four experimental groups: G1, was formed by three ewes infected with T. vivax in the first third of pregnancy (30 days); G2 comprised three infected ewes in the final third of pregnancy (100 days); G3 and G4 were composed of three non-infected ewes with the same gestational period, respectively. Each ewe of G1 and G2 was inoculated with 1.25 × 105 tripomastigotes. Clinical examination, determination of parasitemia, serum biochemistry (albumin, total protein, glucose, cholesterol, and urea), packed cell volume (PCV), serum progesterone, and pathological examination were performed. Placenta, amniotic fluid, blood and tissues from the fetuses and stillbirths were submitted to PCR. Two ewes of G1 (Ewe 1 and 3) presented severe infection and died in the 34th and 35th days post-infection (dpi), respectively; but both fetuses were recovered during necropsy. In G2, Ewe 5 aborted two fetuses on the 130th day (30 dpi) of pregnancy; and Ewe 6 aborted one fetus in the 140th day (40 dpi) of gestation. Ewes 2 and 4 delivered two weak lambs that died five days after birth. Factors possibly involved with the reproductive failure included high parasitemia, fever, low PCV, body score, serum glucose, total protein, cholesterol, and progesterone. Hepatitis, pericarditis, and encephalitis were observed in the aborted fetuses. The presence of T. vivax DNA in the placenta, amniotic fluid, blood, and tissues from the fetuses confirms the transplacental transmission of the parasite. Histological lesion in the fetuses and placenta also suggest the involvement of the parasite in the etiopathogenesis of reproductive failure in ewes.  相似文献

Seventeen mature, pregnant, anaplasmosis-susceptible and 3 anaplasmosis-carrier ewes were used in Anaplasma ovis in utero transmission studies. Susceptible ewes were arbitrarily allotted to 3 groups, 4 each in groups A and C and 9 in group B, and were inoculated with whole blood from a carrier ewe. Each group was Anaplasma-exposed once during one of the thirds of pregnancy. In all ewes, resulting parasitemias were low and anemias were mild. Three carrier ewes comprised group D. Blood samples were obtained once during gestation from each fetus at various stages of development for evaluation and inoculation into splenectomized lambs. Blood obtained from neonates before nursing was also evaluated for Anaplasma presence. Parasitemia was not detected in any fetus or neonate; however, 3 of 16 splenectomized lambs inoculated with fetal or neonate blood developed acute anaplasmosis. Dams of fetuses/neonates with infective blood had been Anaplasma-exposed during their 2nd or last 3rd of pregnancy. Infective Anaplasma agents crossed the placental barrier as early as 130 days of gestation.  相似文献

Of five pregnant cows inoculated intravenously with 5 X 10(8) viable 'Haemophilus somnus', one aborted within 5 days and excreted 'H. somnus' from the vagina for a further 7 weeks. A second cow proceeded to full term parturition but both it and its apparently healthy calf persistently excreted 'H. somnus'. The other animals underwent normal full term calvings and 'H. somnus' was not isolated from them or their calves. Lesions attributable to 'H. somnus' were detected only in the aborted fetus which showed an acute generalized inflammatory cell response consistent with a systemic Gram-negative bacterial infection. 'H. somnus' was isolated from all fetal tissues, including the placenta. The fetus and placenta also showed evidence of damage prior to inoculation. The placental damage may have predisposed the fetus and placenta to infection with 'H. somnus'. The placental epithelial cells contained intracytoplasmic organisms with the morphological and antigenic properties of 'H. somnus'.  相似文献

Pasteurella multocida was isolated from the lungs of calves that died on a farm in the south of England. This organism was inoculated experimentally into 13 calves by the intratracheal route: in all but two of the calves mild clinical disease resulted and at necropsy, three or four days later, pneumonic consolidation involving up to 22 per cent of the lung was observed. P multocida was isolated from all but two of the lungs. Of two calves inoculated intravenously with P multocida, one showed mild clinical disease and slight pneumonic consolidation at necropsy and the other remained normal. Control calves inoculated intratracheally and intravenously with sterile broth showed no signs of illness and no pneumonic consolidation. Histologically the lung lesions comprised a fibrinous bronchopneumonia with variable sized areas of coagulative necrosis, extensive deposition of fibrin and massive dilatation and oedema of the interlobular and pleural lymphatics. It is concluded that P multocida should receive more recognition as a primary pathogen.  相似文献

A strain of Yersinia enterocolitica of O serogroup 6,30 isolated from the liver of an aborted ovine fetus was inoculated intravenously into a group of pregnant ewes at about 90 days gestation and produced placentitis with abortion or delivery of infected lambs about 50 days later. Y. enterocolitica of the same serogroup was recovered from the necrotic placental cotyledons and most other fetal tissues and could be isolated from vaginal discharges of the ewes for a least 2 weeks after abortion. Histological changes were consistent with an acute bacterial necrotizing placentitis and systemic infection of the fetus. Subsequent pregnancies in the ewes proceeded to term without evidence of infection.  相似文献

Experiences, including results of original experimental work on Campylobacter fetus, C. jejuni and C. coli induced diseases of cattle, sheep, dogs, rabbits poultry and men in Hungary are reviewed. Out of 31 cases of abortion in cows 29 (93.5%) were causes by C. fetus subsp. venerealis and only one case each (3.2%) by C. fetus subsp. fetus and C. jejuni, respectively. Out of the 29 strains of C. fetus subsp. venerealis, 26 belonged to serogroup 01 (A) and only 3 to serogroup 02 (B). Campylobacter abortions in sheep flocks were caused in 18 cases (78.3%) by C. fetus subsp. fetus and in 5 cases (21.7%) by C. jejuni. The latter strains belonged to Penner's serogroup 1 (6 strains), 5 (4 strains) and 8 (5 strains), respectively. In scouring dogs 12.7% of the cases were caused by C. jejuni. The same pathogen caused diarrhoea also in young rabbits. Isolated strains belonged to serogroup 2. In cases of Campylobacter hepatitis of laying hens, egg production has been reduced by 8 to 15% for 2 to 3 weeks. Row poultry meat represents often the source of infection for men. The 32 strains of C. jejuni isolated from faecal samples of men affected with diarrhoea belonged to 12 serogroups.  相似文献

Twenty-eight pregnant ewes were inoculated IV with approximately 6 X 10(8) nonclassified, anaerobic, flagellated bacteria (NAFB) that had been isolated from an aborted lamb. Abortion occurred in 3 of the ewes and 1 ewe gave birth to a weak lamb. The remaining 24 ewes and 3 other ewes inoculated orally with NAFB did not develop clinical signs of illness. Suppuration and vasculitis were seen in the placentas of the 3 aborted lambs, 1 of which had necropurulent hepatitis indistinguishable from that usually attributed to Campylobacter fetus infection. The NAFB was isolated from fetal placenta, abomasal content, or internal organs of 2 aborted lambs and the weak lamb. A morphologically similar organism was seen in the abomasal content of the other aborted lamb, but the organism did not grow on bacteriologic culture medium. Therefore, in susceptible pregnant ewes, NAFB can cause fetal placentitis and hepatitis and subsequent birth of weak lambs or abortion.  相似文献

Nine pregnant heifers were inoculated intravenously with infectious bovine rhinotracheitis virus (IBRV) in the sixth month of pregnancy. Tissues were collected from the fetus of a heifer killed 13 days postinoculation (PI), from fetuses of 6 heifers that aborted 16-27 days PI, and from mummified fetuses of 2 heifers that aborted 53 and 85 days PI, respectively. Control tissues were obtained from the fetus of a non-inoculated heifer that was killed in the seventh month of gestation. Tissues were fixed in 10% formalin, embedded in paraffin, and examined for viral antigen by immunohistochemistry, using biotinylated second antibody and alkaline phosphatase-labeled avidin-biotin complex. Antigen was detected in at least 1 tissue from the fetus of each inoculated heifer. Positive tissues included lung, liver, spleen, kidney, adrenal, and placenta. In several fetuses, antigen was identified in tissues from which virus was not isolated in cell culture. This appeared to occur when tissues had only a few small foci of infection or when tissues were severely autolyzed. The observation of viral antigen in tissues from mummified fetuses indicates that this technique may be useful in diagnostic laboratories to detect IBRV infection in tissues that are not suitable for virus isolation or for examination by the cryostat tissue section-fluorescent antibody technique.  相似文献

ELISA was adapted for the study of antigenic relations among important Campylobacters and for the presence of anti-campylo-bacter antibodies in 394 sheep and 265 cattle. Rabbit anti-C. jejuni, C. coli, G. fetus subsp. fetus and C. laridis heat-stable antigen sera were evaluated against 29 Campylobacter strains and 6 other bacteria. Anti-C. jejuni and G. coli reacted strongly with homologous antigens and weakly with C. fetus subsp. fetus, C. laridis and C. fecalis antigens. C. fetus subsp. fetus serum reacted mainly with its homologous antigen. C. laridis serum showed closer reactivity to C. jejuni than to C. fetus subsp. fetus, C. coli and C. fecalis. Insignificant cross-reactions were observed with Y. enterocolitica, S. dublin and E. aerogenes heat-stable antigens, Ewes vaccinated with C. fetus subsp. fetus bacterin showed higher ELISA titers against C. fetus subsp. fetus antigens than non-vaccinated ewes or rams. Twenty-five percent of the vaccinated animals showed titers as low as 95 % of the non-vaccinated animals. In cattle the lowest antibody titers against C. fetus subsp. fetus, C. jejuni, C. coli and C. laridis antigens were exhibited by the precolostrum sera followed by the postcolostrum and adult sera. These studies demonstrated the applicability of the ELISA test in seroepidemiological investigations concerning the distribution and significance of Campylobacter antibodies in food animal sera.  相似文献

Chlamydia psittaci is a major cause of ovine abortion in the fourth to fifth months of gestation. During the lambing seasons of 1986, 1987, and 1988, fetuses from 52 cases of ovine abortion, stillbirth, or perinatal death were submitted to the laboratory for necropsy examination. Placenta or fetal tissues from 34 cases were cultured on mouse L cells for C. psittaci. Chlamydia psittaci was identified by immunofluorescence on cultures in 20 of these cases. The major gross lesion consistently associated with chlamydial abortion was placentitis with multifocal cotyledonary necrosis and accumulation of red-brown exudate in the intercotyledonary placenta. Chlamydiae appeared as spherical organisms, less than 1 micron in diameter, in the cytoplasm of trophoblasts in impression smears of cotyledons. Histologically, placentitis was sometimes accompanied by pneumonia or encephalitis in the fetus. Chlamydia psittaci was considered the cause for fetal death when chlamydial isolation was associated with placentitis or inflammation of other fetal tissues and when other abortifacient agents were not detected.  相似文献

To determine the presence of tissue cysts in ovine placentas, 6 ewes were inoculated orally with 10,000 Toxoplasma gondii oocysts at 60 days of gestation. The ewes were euthanatized and necropsied 21, 52, 56, 57, 57, and 62 days after T gondii inoculation, and placental cotyledons from each ewe were collected and homogenized. To distinguish between the presence of tachyzoites that are killed by acid pepsin solution and bradyzoites (from cysts) unaffected by this solution, a portion of each homogenate was inoculated into mice and another portion was inoculated into mice after digestion in acid pepsin solution. Toxoplasma gondii was isolated in 26 of 34 (76.4%) of mice inoculated with nondigested placentas of all 6 ewes and in 16 of 34 (47%) mice inoculated with digested placenta of 5 of 6 ewes. Seemingly, cysts do occur in placental tissue, but the digestion method was inferior, compared with the nondigestion method for recovery of T gondii from placenta.  相似文献

Homogenised tissues or tissue culture supernatant fluid containing a noncytopathic pestivirus obtained from a lamb with a neurologic form of border disease, were inoculated into ewes at different stages of pregnancy. Foetal death occurred in 9 ewes of those inoculated between 19 and 47 days of pregnancy while 3 ewes did not lamb. Eight of the foetuses were aborted between 77 and 132 days of pregnancy; of these 6 were autolysed or mummified and one had arthrogryposis. The one full-term dead lamb had a hairy birth coat and lissencephalic micrencephaly. Foetal death occurred in only 7 of 14 ewes inoculated between 57 and 72 days of pregnancy. Four of these ewes aborted between 77 and 108 days of pregnancy and 3 gave birth to full-term, dead, hairy lambs. The remaining 7 ewes gave birth to live hairy lambs with severe inco-ordination. All lambs carried to term and aborted foetuses or lambs that could be examined had a range of intracranial malformations including focal leucomalacia, micrencephaly, hydranencephaly, porencephaly, lissencephaly and cerebellar hypoplasia. Some lambs also had skeletal abnormalities including arthrogryposis, scoliosis and brachygnathia inferior. The pestivirus isolate used in these trials produced more severe effects on the ovine foetus than previously observed in similar inoculation trials using pestivirus isolates from border disease lambs without nervous signs.  相似文献

Equine herpesvirus 9 (EHV-9), a new neurotropic equine herpesvirus, was inoculated intranasally at 107 plaque-forming units in five dogs to assess its pathogenicity. Dogs showed weight loss, pyrexia, anorexia, and neurologic signs on the fourth day. The EHV-9 virus was recovered from the examined brains. Histologically, dogs had a fulminant nonsuppurative encephalitis characterized by severe neuronal degeneration and loss, with intranuclear inclusions, slight glial reactions, perivascular cuffing, and multifocal hemorrhage. The olfactory bulb and the frontal and temporal lobes were predominantly affected. Immunohistochemistry revealed reactivity for EHV-9 antigen in neurons. All dogs had mild bronchopneumonia and various degrees of lymphoid necrosis. These findings indicate that dogs are fully susceptible to EHV-9 and that EHV-9 can cause fulminant encephalitis with high mortality in dogs, as in gazelles and goats.  相似文献

We describe here the gross and microscopic lesions in 18 experimentally induced and 120 natural Campylobacter abortions. In natural Campylobacter abortions, gross lesions were reported infrequently; placentitis was recorded in 6% and hepatic lesions in 4% of our field cases. Placentitis was the microscopic lesion identified most consistently in natural abortions (93%) and was often observed in association with abundant bacterial colonies in chorionic villi (54%) and less often with placental vasculitis (13%). In natural abortions, suppurative fetal pneumonia (48%), necrosuppurative hepatitis (16%), and purulent meningitis (7%) were also observed. The better-preserved specimens from experimentally induced abortions were utilized to define placental changes more precisely. Placentitis was identified in all 18 experimentally induced abortions and was observed most consistently in the chorionic villus stroma (100%), often accompanied by suppurative surface exudate (89%). An inflammatory infiltrate was less commonly identified in the cotyledonary hilus (39%) and intercotyledonary placenta (22%). Bacteria were visualized in H&E-stained sections in 89% of placentas from experimentally infected ewes, primarily as well-demarcated bacterial colonies within subtrophoblastic, sinusoidal capillaries (89%), in the cotyledonary villus stroma (89%), and within the cytoplasm of trophoblasts (22%). Transmission electron microscopy and immunohistochemistry confirmed that the vast majority of the well-demarcated bacterial colonies characteristic of Campylobacter abortion were within subtrophoblastic sinusoidal capillaries. The most characteristic microscopic lesions identified in cases of Campylobacter abortion in sheep were placentitis with placental bacterial colonies, placental vasculitis, and fetal pneumonia.  相似文献
