An entomopathogenic strain of Bacillus sp. was isolated from diseased red slug caterpillars of the leaf-feediug pest of tea, Eterusia magnifica, from the Darjeeling foothill region. Analysis of the bacterimn based on polyphasic approach such as growth phase, biochemical tests, whole body" protein, crystal protein profiles along with bioassay (i.e. LC50 and LT50 values) established it as a different strain but close to Bacillus thuringiensis kurstaki (Btk), the commercial microbial pesticides of lepidopterans. Among biochemical parameters differences were noted between the new strain and Btk in ONPG, lysine decarboxylase, omithin decarboxylase, urease, nitrate reduction, V-P and glucose utilization tests. PAGE analysis of the whole body protein for the new strain recorded a 34 kDa band which was absent in Btk (used as reference). Crystal protein profile of the newly isolated bacterial strain showed 53 and 49 kDa bands whereas in Btk only 52 kDa band was evident. Although the LC50 values of the new strain and Btk were close, their LT50 values were much different, the new strain showing a lower value than Btk. In light of the above differences and in absence of any report of entomopathogenic bacterial strain of E. magnifica, the isolated strain of Bacillus appeared to be new to science and hence was designated as RS01. The new strain opens up the possibility of its futttre use as microbial pesticide after standardizing its formulation and checking its safety aspects.  相似文献

The changes in humic substances (HS) is fundamental in detecting soil carbon sequestration mechanisms in natural and cultivated environments. Based on a long-term trial, the amounts of water dissolved substances (WSS), humic acid (HA), fulvic acid (FA) and humin (HU) were determined to explore the impact of long-term fertilization on HS. Increases in the amounts of WSS, HA, FA and HU were significant different among the treatments with manure. A significant correlation was found between the increased soil organic carbon (SOC) and HS (R^2=0.98, P〈0.01). The change in the E4/E6 ratio was significantly correlated with the increased SOC (R2=0.88, P〈0.01), HA (R^2=0.91, P〈0.01), FA (R^2=0.91, P〈0.01) and HU (R^2=0.88, P〈0.01). The cluster was mainly divided into two parts as manure fertilization and inorganic fertilization, based on the increases in HA, FA and HU. These results suggest that long term fertilization with manure favours carbon sequestration in HS and is mainly stabilized as HU, while the HA becomes more aliphatic. We conclude that increases in SOC can be linked to changes in the molecular characteristics of HS fractions under long term fertilization.  相似文献

对甘蔗斑茅杂种BC1后代崖城01-21和崖城01-36进行内转录间隔(ITS)鉴定和染色体计数与核型分析,探讨甘蔗斑茅杂交后代的染色体遗传行为.结果表明,崖城01-21的体细胞染色体核型公式为:2n=104=100m+4 sm(4SAT),最长与最短染色体的长度比为4.25,平均臂比1.35,染色体属2C型,分子鉴定结合细胞学研究表明其为斑茅的真实后代,其染色体遗传为n+n;崖城01-36经ITS鉴定为真实杂种,其体细胞染色体核型公式为:2n=132=130m+2 sm,最长与最短染色体的长度比为3.94,平均臂比1.27,染色体属2B型,结合其性状的进展,推断亲本向其传递染色体可能以2n+n的方式进行.  相似文献

Bemisia tabaci (Gennadius) biotypes B and Q are two invasive biotypes in the species complex. The comparison of the population genetic structure of the two biotypes is of significance to show their invasive mechanism and to their control. The intersimple sequence repeats (ISSR) marker was used to analyze the 16 B-biotype populations and 4 Q-biotype populations worldwide with a Trialeurodes vaporariorum population in Shanxi Province, China, and a B. tabaci non-B/Qbiotype population in Zhejiang Province, China, was used as control populations. The analysis of genetic diversity showed that the diversity indexes of biotype Q including Nei's gene diversity index, Shannon informative index, and the percentage of polymorphic loci were higher than those of biotype B. The high genetic diversity of biotype Q might provide the genetic basis for the excellent ecological adaptation. Cluster analysis suggested that the ISSR could not be used in the phylogenetic analysis though it could easily distinguish the biotypes of B. tabaci. The difference of the population genetic structure between the biotype B and the biotype Q exists based on the ISSR marker. Meanwhile, the results suggested that the molecular marker has its limitation in the phylogenetic analysis among the biotypes of B. tabaci.  相似文献

Whether the biochar amendment could affect soil organic matter (SOM) turnover and hence soil carbon (C) stock remains poorly understood. Effects of the addition of ^13C-labelled rice straw or its pyrolysed biochar at 250 or 350℃ to a sugarcane soil (Ferrosol) on soil labile C (dissolved organic C, DOC; microbial biomass C, MBC; and mineralizable C, MC) and soil organic C (SOC) were investigated after 112 d of laboratory incubation at 25℃. Four treatments were examined as (1) the control soil without amendment (Soil); (2) soil plus ^13C-labelled rice straw (Soil+Straw); (3) soil plus 250℃ biochar (Soil+B250) and (4) soil plus 350℃biochar (Soil+B350). Compared to un-pyrolysed straw, biochars generally had an increased aryl C, carboxyl C, C and nitrogen concentrations, a decreased O-alkyl C and C:N ratio, but similar alkyl C and δ^13C (1 742- 1 877 %). Among treatments, significant higher DOC, MBC and MC derived from the new C (straw or biochar) ranked as Soil+Straw〉Soil+B250〉Soil+B350, whilst significant higher SOC from the new C as Soil+B250〉Soil+Straw≈Soil+B350. Compared to Soil, DOC and MBC derived from the native soil were decreased under straw or biochar addition, whilst MC from the native soil was increased under straw addition but decreased under biochar addition. Meanwhile, native SOC was similar among the treatments, irrespective of the straw or biochar addition. Compared to Soil, significant higher total DOC and total MBC were under Soil+Straw, but not under Soil+B250 and Soil+B350, whilst significant higher total MC and total SOC were under straw or biochar addition, except for MC under Soil+B350. Our results demonstrated that the application of biochar to soil may be an appropriate management practice for increasing soil C storage.  相似文献

Our expedition have identified location in the central part of Kosovo where were collected a samples (accessions) at coordinates; Sub-Locality SL-1 (N42°34′36″, E21°07′45″), altitude 574 m, SL-2 (N42°34′29″, E 21°07′45″altitude 591 m and SL-3 (N42°34′30″, E21°07′46″) on altitude 565 m. The aim of present investigation was carried out to study genetic variability of quantitative and qualitative parameters. The populations from SL-2 were characterized of higher significance on seed oil (23.74%) and protein content (16.74%). The estimation average value of seed protein and oil content was 15.52% and 23.48% respectively. The variability of plant height ranged from 182.33 cm to as much as 200.89 cm, while distinction's between them were + 18.56 cm which was a significant higher at P〈0.05 probability level. The SL-3 showed the higher value of HWK (34.6 g), head per plant (39.33) and head diameter (9.5 cm) but, was characterized significantly lowest on seed protein content (13.99%). The all traits included in our investigations showed a higher genetic variability with majority of extreme values. The sunflower populations differed significantly in several parameters measured, they have a different genetic and morphological backgrounds. Results from wild sunflower form indicated notable differences in dispersion of important genes for different traits and showed the potential for using in future for breeding program.  相似文献

The study aims at finding alternative interventions for the prevention ofE. coli infection in piglets so as to boost farmers' benefits and improve environment in the area. Data were collected from one experimental group consisting of 210 small pig holder farms and one control group of 70 farms. All of 280 litters and sows were analyzed and the techniques of early vaccination at 60-80 d of pregnant (time 1) were applied and repeated after 10 d with E. coli vaccine to produce antibodies through milk. The percentage of piglets with diarrhea caused by E. coli significantly decreased from 75.7% to 18.9%. Injection of E. coli vaccine for the pregnant sows created antibodies for young piglets through colostrums. The treatment of E. coli cost 45,420 Vietnam Dong (VND) per litter in the experimental group and that was 156,000 VND/litter in the control group. Use of betel-bokashi (bio-product as plant antibiotics and Lactobacillus) to treat piglet diarrhea was proved safe and effective. Litter size and average weight at birth and weaning were improved significantly (P 〈 0.05). The average weaned weight of piglets and sows were 11.2 kg and 118.6 kg in the experimental group, compared to 9.4 kg and 89.9 kg in the control group, respectively. Piglets in the experimental group had good health, no diarrhea and a shorter weaning time (2 d) and mating interval 2.22 days earlier than those in the control group. The average profit of 210 households was significantly higher in the experiment group: feed-cost was much lower than that in the control group. Small-holder farms implementing the experiments of applying the techniques received more financial profit, which was 1,502,000 VND per year per sow, while those who did not apply the techniques gained only 223,000 VND per year per sow.  相似文献

Erianthus arundinaceus is an important, closely related genus of Saccharum officinarum L. It is therefore important to understand how the chromosomes are transmitted when it hybridizes with sugarcane. The hybrids and backcross progenies of S. officinarum and E. arundinaceus and their parents were used for Karyotype analysis and to study the law of chromosome transmission. The results showed that the somatic chromosome number of both of the E. arundinaceus Hainan92-105 and Hainan92-77 were 2n = 60 = 60sm, belonging to type 1 A, and the BC1 YC01-21 was 2n = 104 = 100m + 4sm, belonging to type 2C. The other six tested clones belonged to type 2B. The both F1s YC96-66 and YC96-40 that originated from Badila (2n = 80 = 70m + 10sm) with E. Arundinaceus were 2n = 70 = 68m + 2sm, which suggests an n + n transmission. The cross between YC96-66 (female parent) and CP84-1198 (male parent, 2n = 120 = 114m + 6sm) also followed the same genetic law and the somatic chromosome number of their progeny, YC01-3 (2n = 105 = 95m + 10sm). The cross derived from YC96- 40 (female) and CP84-1198 (male), YC01-21 had 2n = 104 = 100m + 4sm chromosomes, following the same genetic law of n + n. However, YC01-36 had 2n = 132 = 130m + 2sm chromosomes, which suggests a 2n + n chromosome transmission. It can be inferred that the inheritance of chromosomes was very complex in the BC1. The difference in chromosome number between clones was as high as 28. This could be explained by the 2n + n transmission of chromosomes. In addition, as there was not be a regular number of haploids, this phenomenon is termed as disequilibrium hybridization.  相似文献

【目的】研究3种除草剂不同配比防除狗牙根草坪内泽漆的效果,筛选出最优防除方案,为有效防除泽漆提供科学依据。【方法】用77.7%草甘膦铵盐可溶粒剂、56% 2甲4氯钠可溶性粉剂、40%苄·二氯(苄嘧磺隆+二氯喹啉酸)可溶性粉剂正交组合的16种不同配比对泽漆进行防除,在施药后3,6,12,20,30 d分别观察记载泽漆外观状态,统计死亡植株数,计算药效指数、株防效,并在施药后3,6,12,20,30,45 d观测记载狗牙根生长情况。【结果】3种除草剂不同配比对泽漆的防效差异显著(P<0.05),3种除草剂混用对泽漆的防除效果比2种混用效果好,2种混用的效果比单一好。当3种除草剂混用时,以40%苄·二氯可溶性粉剂85 mg/m2+56% 2甲4氯钠可溶性粉剂300 mg/m2+77.7%草甘膦铵盐可溶粒剂420 mg/m2组合防效最好,药后3~6 d,药效指数达到96.0%~97.6%,株防效可达到88.9%,药后20 d药效指数和株防效均达到100%,且在30 d内没有再生能力。单一除草剂的使用对狗牙根生长无影响或影响甚微,2种及3种除草剂混用在药后3~20 d对狗牙根生长造成可见影响,但在30 d后均可减轻,45 d后均可消失。【结论】可用40%苄·二氯可溶性粉剂85 mg/m2+56%2甲4氯钠可溶性粉剂300 mg/m2+77.7%草甘膦铵盐可溶粒剂420 mg/m2组合对狗牙根草坪内的泽漆进行防除。  相似文献

The objective of this study was to evaluate the effect of a novel feed additive on chicken intestinal colonization and carcass contamination by Campylobacterjejuni. The feed additive was composed of microencapsulated organic acids and essential oils (OA/EO). The feed additive tested was provided by Jefo Nutrition Inc., St-Hyacinthe, Quebec, Canada. Day-old birds were separated into two rooms and subdivided into two groups. Chicken were fed with OA/EO or not fed with OA/EO until they reached 35 d of age. At 14 d of age, chickens received an oral suspension of two well characterized C. jejuni strains, depending on the room they were housed in. The levels of C. jejuni were periodically monitored in the caecum and on the carcasses. C. jejuni colonization was further characterized by the use of high-resolution melt analysis of the C. jejuniflaA gene (HRM-flaA). The effect of the feed additive was strain-dependent. In room two, the feed additive had no effect on the caecal counts. In room one, at 35 d of age, caecal C. jejuni counts were higher with OA/EO, as opposed to carcasses counts which were lower in the treated group. The HRM-flaA analysis showed that an amplification profile was predominant in birds fed with OA/EO at 35 d of age in room one, suggesting the selection of a C. jejuni strain. In conclusion, the OA/EO seemed to be effective to reduce C. jejuni levels but this effect appeared strain dependent.  相似文献

Expression of insecticidal protein for transgenic Bacillus thuringiensis (Bt) cotton is unstable and related to nitrogen metabolism. The objective of this study was to investigate the relationship between leaf carbon nitrogen ratio (C/N) and insecticidal efficacy of two Bt cotton cultivars. C/N ratio and Bt protein content were both measured at peak square period and peak boll period respectively under 5-7 d high temperature and different nitrogen fertilizer rates on the Yangzhou University Farm and the Ludong Cotton Farm, China. All plants were grown in field. The results showed that the C/N ratio enhanced slightly and the Bt protein content remained stable at peak square period, but significant increases for the C/N ratio and decreases markedly for the leaf Bt protein concentration were detected at the peak boll period. The similar patterns at the two growth periods were found for the leaf C/N ratio and Bt protein content by different N fertilizer treatments. When nitrogen rate was from 0 to 600 kg ha-l, the C/N ratio was reduced by 0.017 and 0.006 for Sikang 1 and Sikang 3 at peak square period, compared to the 1.350 to 1.143 reduction for Sikang 1 and Sikang 3 at peak boll period, respectively. Correspondingly, the leaf Bt protein contents were bolstered by 2.6-11.8 and 26.9-36.9% at the two different growth periods, respectively. The results suggested that enhanced C/N ratio by high temperature and nitrogen application may result in the reduction of inseetiocidal efficacy in Bt cotton, especially in peak boll period.  相似文献

Eight strains of nervous necrosis virus (NNV) isolated in Vietnam were used to detect the pathogenicity and immune response in sea bass (SB). All strains induced cytopathic effect in SB cell line, complete destruction of monolayer of cells appeared after seven days post infection (dpi). Virus titer was different for each strain, TCIDso ranged from 102.7 to 1069, and LDs0 from 1015 to 1075. Five NNV strains named QN 02, QN 05, QN 07, ND 11 and KH 05 had higher virulence than the other three, the first causing 100% mortality in experimental fish 3-5 dpi. NNV KH 05 had the highest antigenic similarity, and it was inactivated completely by 0.2% formalin, 0.002 mol/L binary ethylenimine (BEI) and 0.1% beta-propiolactone. The neutralization antibody titer obtained in fish of groups immunized by BEI 0.002 M and beta-propiolactone 0.1% inactivated virus was four to eight times higher than that of the group treated with the formalin inactivated virus. The antibody titer in fish immunized with beta-propiolactone inactivated virus was more persistent. The efficacy of vaccines developed from beta-propiolactone inactivated virus and aluminium hydroxide (AH) or aluminum phosphate (AP) was observed by intramuscularly immunizing Epinephelusfuscoguttatus size 1.5 cm. Neutralizing antibodies appeared in vaccinated fish on 10th day post-immunization (dpi) at a dilution of 1:16; 1:32 and highest levels were reached on 30-45 dpi, at dilutions of 1:256 and 1:512, after treatment with AH and AP vaccine, respectively. The relative percent of survival (RPS) of vaccine at 30 dpi was highest with challenge doses 0.2-1 × 10^6.8 TCIDs0, the RPS varied from 80%-83.3% in both groups of AH and AP immunization. This result provides the basis for developing a vaccine against NNA disease.  相似文献

Biogas production generates digested slurry as a byproduct. It can be used as a fertilizer especially after its conversion into digested liquid. A pot based study was conducted in order to evaluate the effect of the application of digested liquid on CH4 and N2O flux, and plant biomass in paddy. Analysis revealed that digested liquid treated soils released more CH4 compared to ammonium sulphate and the control. Ammonium sulphate treated soil emitted the highest N20 whereas digested liquid application decreased its emission significantly. Further, the cumulative emission over 101 d of the experiment was found to be higher for CHa (16.9 to 29.9 g m^-2) compared to N20 (-49.3 to 18.9 mg m^-2) for all treatments. Digested liquid application had positive impact on plant variables such as panicle number and weight of panicles. This study suggests that digested liquid application significantly decrease N20 emission and increase CH4 emission possibly due to affecting the availability of organic C in the soil to microbial activity for methanogenesis. Another possibility for enhancing CH4 emission by following biogas digested liquid could be attributed to the increase in plant biomass.  相似文献

[目的]构建一种适宜的陈皮精油提取方法。[方法]利用超临界CO2萃取技术提取陈皮中的挥发性精油,以提取率为指标,首先研究了原料粒度、温度、压力及时间各单因素对提取率的影响,然后用响应面分析法优化提取工艺。[结果]研究表明,超临界CO2萃取技术能有效地提取陈皮中的精油,提取率模型为:Y=-53.39667+2.57611A+0.062000B+0.046000C-0.031098A2。优化后的提取工艺为温度41.43℃、压力25 MPa和粒度60目,精油提取率模型预测值为4.28%,试验验证值为4.18%,预测值与实际值无显著差异(P=0.01)。[结论]超临界CO2萃取技术是一种从陈皮中提取精油的有效方法。  相似文献

It is predicted that the current atmospheric CO2 concentration will be doubled and global mean temperature will increase by 1.5-6°C by the end of this century. Although a number of studies have addressed the separate effects of CO2 and temperature on plant-insect interactions, few have concerned with their combined impacts. In the current study, a factorial experiment was carried out to examine the effect of a doubling CO2 concentration and a 3℃ temperature increase on a complete generation of the brown planthopper (Nilaparvata lugens) on rice (Oryza sativa). Both elevated CO2 and temperature increased rice stem height and biomass of stem parts. Leaf chlorophyll content increased under elevated CO2, but only in ambient temperature treatment. Water content of stem parts was reduced under elevated temperature, but only when coupled with elevated CO2. Elevated CO2 alone increased biomass of root and elevated temperature alone enhanced leaf area and reduced ratio of root to stem parts. Brown planthopper (BPH) nymphal development was accelerated, and weight of and honeydew excretion by the F1 adults was reduced under elevated temperature only. Longevity of brachypterous females was affected by a signiifcant interaction between CO2 and temperature. At elevated temperature, CO2 had no effect on female longevity, but at ambient temperature, the females lived shorter under elevated CO2. Female fecundity was higher at elevated than at ambient temperature and higher at elevated CO2 than at ambient CO2. These results indicate that the combined effects of elevated temperature and CO2 may enhance the brown planthopper population size.  相似文献

Essential oils (EOs) from Rosemary, Rosmarinus officinalis (Lamiaceae), Orange, Citrus sinensis (Rutaceae), Lavandula, Lavandula angustifolia (Lamiaceae), False Yellowhead, Dittrichia viscosa (Asteraceae) and their major components were evaluated against root knot nematode, Meloidogyne incognita and Meloidogynejavanica. Second stage juveniles' (J2) paralysis and egg hatch inhibition were studied, while pulverized plant parts were tested for nematodes biological cycle arrest. All EOs paralyzed J2 and M. incognita were found more sensitive than M javanica with the EC50/4 days calculated at 250, 3,650 and 4,260 12g/mL for 1). viscosa, L. angustifolia and R. officinalis, respectively. Similarly, all EOs inhibited nematodes egg hatch and D. viscosa exhibited the highest inhibition on egg hatch (100% inhibition at 5 μg/mL). A significant influence of constituent terpenes (limonene, 1,8-cineole, linalool, camphor, L-borneol, caryophyllene oxide, β-eudesmol) dose and exposure time was indicated on egg hatch inhibition (56% to 100% at 500 μg/mL and 1,000 μg/mL), while only β-eudesmol achieved paralysis of J2 and specifically against M. incognita (EC50/1d = 50μg/mL). Interestingly, the most active botanical species arresting Meloidogyne spp. biological cycle in soil was C. sinensis (EC50 = 2 mg/g) and the most sensitive nematode species was M. javanica. The larvicidal and egg hatch inhibition activity holds promise towards the optimization of artificial terpene mixtures as novel and effective natural nematicides. Complex interactions of primary compounds and subsequent decomposition derivates compose efficacy profile of soil amendments.  相似文献

A pot experiment was conducted to evaluate the efficiency of farm yard manure (FYM) and lime (calcium carbonate) on reducing the uptake of Cd by rice in two Cd contaminated soils of Chittagong. The effects of farm yard manure, lime and farm yard manure mixed with lime (FYM + Lime) on rice (Oryza sativa L.) growth, Cd uptake by rice and 0.1 N HC1 extractable Cd in soil were investigated. Results showed that addition of lime and farm yard manure mixed with lime significantly decreased 0.1 N HCl extractable Cd in the city sewage soil. Application of farm yard manure significantly decreased Cd concentration in rice in the city sewage soil. Lime and farm yard manure mixed with lime significantly reduced Cd concentration in rice m both the contaminated soils. Lime was more efficient than farm yard manure in reducing the uptake of Cd by rice.  相似文献

Phosphorus (P) is a major growth-limiting nutrient, and unlike the case of nitrogen (N), there is no large atmospheric source that can be made biologically available. Moreover, P governs crucial role in plant as it stimulates root development and growth, gives plant rapid and vigorous start leading to better tillering and essential for many metabolic processes for seed formation. Soil microbes play very important role in bio-weathering and biodegradation. The microorganisms produce low molecular mass organic acids, which attack the phosphate structure and transform phosphorus from non-utilizable to the utilizable for the plants form. The test of the relative efficiency of isolated strains is carried out by selecting the microorganisms that are capable of producing a halo/clear zone on a plate owing to the production of organic acids into the surrounding medium. It is a well-known fact that as the particle size of rock phosphate decreases, the microbe mediated solubilization of rock phosphate increases in soil. In the present investigation, microbial solubilization of nano rock phosphate (〈 100 nm) particles was studied. Experimental results revealed that Pseudomonas striata solubilized 11.45% of the total P after 24 h of incubation from nano rock phosphate particles while 28.95% and 21.19% of the total P was solubilized by Aspergillus niger (black pigmented) and Aspergillus niger (green pigmented), respectively. It was also observed that Aspergillus niger has the higher ability to dissolve Udaipur rock phosphate than Pseudomonas striata.  相似文献
