alpha-klotho was identified as a gene associated with premature aging-like phenotypes characterized by short lifespan. In mice, we found the molecular association of alpha-Klotho (alpha-Kl) and Na+,K+-adenosine triphosphatase (Na+,K+-ATPase) and provide evidence for an increase of abundance of Na+,K+-ATPase at the plasma membrane. Low concentrations of extracellular free calcium ([Ca2+]e) rapidly induce regulated parathyroid hormone (PTH) secretion in an alpha-Kl- and Na+,K+-ATPase-dependent manner. The increased Na+ gradient created by Na+,K+-ATPase activity might drive the transepithelial transport of Ca2+ in cooperation with ion channels and transporters in the choroid plexus and the kidney. Our findings reveal fundamental roles of alpha-Kl in the regulation of calcium metabolism.  相似文献

Cadherins are a family of cell adhesion receptors that are crucial for the mutual association of vertebrate cells. Through their homophilic binding interactions, cadherins play a role in cell-sorting mechanisms, conferring adhesion specificities on cells. The regulated expression of cadherins also controls cell polarity and tissue morphology. Cadherins are thus considered to be important regulators of morphogenesis. Moreover, pathological examinations suggest that the down-regulation of cadherin expression is associated with the invasiveness of tumor cells.  相似文献

Administration of glucose, fructose, and glycerol to fasted rats produced a significant depression of liver phosphoenolpyruvate carboxykinase activity within 4 to 8 hours; galactose and ribose were much less effective. All the compounds yielded appreciable quantities of liver glycogen. The depression of phosphoenolpyruvate carboxykinase activity by glucose and glycerol was diminished by the concomitant administration of 2-deoxyglucose. The latter depressed glycogen formation from administered carbohydrate in muscle but not in liver. In rats made diabetic by alloxan, depression of elevated phosphoenolpyruvate carboxykinase activity by insulin was dependent upon a dietary source of carbohydrate. These results were interpreted to indicate that depression of certain gluconeogenic enzymes after carbohydrate ingestion is initiated by the metabolism of carbohydrate in some extrahepatic site.  相似文献

Vestibulospinal reflexes as a function of microgravity   总被引:2,自引:0,他引:2  
The idea that sustained microgravity will result in altered otolith input requiring a modification of postural control was explored by using the pathway that links the otolith organs and spinal motoneurons. Two related methods were used. First, the Hoffmann reflex was used to measure at specific times the excitability of the soleus-spinal motoneuron pool during a brief unexpected linear acceleration. Second, extensive dynamic postural testing with a moving platform was done before and after flight. The Hoffmann reflex amplitude, reflecting otolith-modulated motoneuron sensitivity, was low in flight after adaptation, and its postflight potentiation may have been dependent on rate of adaptation. The strength of inflight motion sickness symptoms was related to postflight Hoffmann reflex amplitude. Dynamic posture tests showed significant deviations from the results obtained before flight. The strategy used for balance on the moving platform was modified, and the behavior of the subjects suggested a decrease in awareness of the direction and magnitude of the motion.  相似文献

The neural cell adhesion molecule (NCAM) can influence a number of diverse intercellular events, including junctional communication, the association of axons with pathways and targets, and signals that alter levels of neurotransmitter enzymes. These pleiotropic effects appear to reflect the ability of NCAM to regulate membrane-membrane contact required to initiate specific interactions between other molecules. Such regulation can occur through changes in either NCAM expression or the molecule's content of polysialic acid (PSA). When NCAM with a low PSA content is expressed, adhesion is increased and contact-dependent events are triggered. In contrast, the large excluded volume of NCAM PSA can inhibit cell-cell interactions through hindrance of overall membrane apposition.  相似文献

Neural crest cells, which generate peripheral nervous system and facial skeleton, arise at the neural plate/ectodermal border via an inductive interaction between these tissues. Wnts and bone morphogenetic proteins (BMPs) play roles in neural crest induction in amphibians and zebrafish. Here, we show that, in avians, Wnt6 is localized in ectoderm and in vivo inhibition of Wnt signaling perturbs neural crest formation. Furthermore, Wnts induce neural crest from naive neural plates in vitro in a defined medium without added factors, whereas BMPs require additives. Our data suggest that Wnt molecules are necessary and sufficient to induce neural crest cells in avian embryos.  相似文献

Clinically successful hematopoietic cell transplantation is dependent on hematopoietic stem and progenitor cells. Here we identify the matricellular protein Nephroblastoma Overexpressed (Nov, CCN3) as being essential for their functional integrity. Nov expression is restricted to the primitive (CD34) compartments of umbilical vein cord blood, and its knockdown in these cells by lentivirus-mediated RNA interference abrogates their function in vitro and in vivo. Conversely, forced expression of Nov and addition of recombinant Nov protein both enhance primitive stem and/or progenitor activity. Taken together, our results identify Nov (CCN3) as a regulator of human hematopoietic stem or progenitor cells.  相似文献

Ras p21 as a potential mediator of insulin action in Xenopus oocytes   总被引:36,自引:0,他引:36  
The oncogene protein product (p21) of the ras gene has been implicated in mediating the effects of a variety of growth factors and hormones. Microinjection of monoclonal antibody 6B7, which is directed against a synthetic peptide corresponding to a highly conserved region of p21 (amino acids 29 to 44) required for p21 function, specifically inhibited Xenopus oocyte maturation induced by incubation with insulin. The inhibition was dose-dependent and specific since (i) the same antibody had no effect on progesterone-induced maturation, (ii) immunoprecipitation and Western blotting indicated that the antibody recognized a single protein of molecular weight 21,000 in oocyte extracts, and (iii) inhibition was not observed with identical concentrations of normal immunoglobulin. Thus, p21 appears to be involved in mediating insulin-induced maturation of Xenopus oocytes. Furthermore, the mechanism may involve phosphorylation of p21, as p21 was found to be a substrate of the insulin receptor kinase.  相似文献

Ethanol-drinking was induced in laboratory rats that were maintained in total darkness. The established preference for ethanol was not reversed under conditions of constant illumination although a decrease in ethanol intake occurred. Administration of melatonin to rats maintained under "normal" photoperiods (9 hours of darkness during a 24-hour day) also induced ethanol-drinking.  相似文献

Rats in a low-ceiling shuttlebox initially show a lower level of learning than rats in a high-ceiling box. After an hour's interruption of conditioning the performance of animals in low-ceiling boxes improves and avoidance is slightly more efficient than in the unimproved performance of animals in high-ceiling boxes. The size and number of aerial spray drops impinging on spruce budworm in its conifer forest habitat were determined by means of a new tracer method that uses fluorescent particles in a liquid spray. Examination of 1113 larvae affected by an experimental insecticide that had been applied to a 5000-acre (2024-ha) test area in Montana showed that 93 percent had not been contacted by any droplets larger than 50 micro in diameter. Small numbers of droplets 50 to 100 micro in diameter were found on 7 percent of the larvae, along with lethal numbers of smaller drops. No evidence was found that significant numbers of drops larger than 100 micro reached the target insects. Because about 95 percent of the spray applied to forests by current methods consist of droplets larger than 50 micro, the biologically effective portion of the drop spectrum is only a few percent. The data foreshadow a major potential reduction in insecticide requirements for the successful control of spruce budworm.  相似文献
