In an attempt to increase the accuracy of roost emergence counts for a monitoring programme, the exits of two Pipistrellus pygmaeus roosts were illuminated with light of different colours and intensities. Light intensity affected bat emergence more than light colour. At one roost there was no significant difference in the bat emergence pattern between when the roost exit received no illumination and when it was illuminated with red light. The use of the latter is proposed to increase the accuracy of bat roost emergence counts. 相似文献

The soprano pipistrelle (Pipistrellus pygmaeus) is a house-dwelling bat, whose maternity colonies are often affected by exclusion from buildings in southern Europe. Bat boxes with a suitable thermal behaviour can provide alternative roosts. However, this is particularly difficult to achieve in hot climates, where boxes may overheat. We studied roost temperatures and roost thermal preferences of this bat in attics and roofs in a Mediterranean region. Roosts reached high temperatures, experienced great thermal fluctuations and had wide spatial thermal ranges. Bats preferred the warmest locations within roosts, indicating that they are a heat tolerant species. However, they avoided temperatures of 40 °C and above, suggesting that this is the upper limit of the species thermally neutral zone. The study of experimental sets of white, grey and black boxes demonstrated that the latter were the ones that most often reached the temperatures observed in roosts in buildings. They were also the ones occupied by larger numbers of bats. These results suggest that, even in hot Mediterranean climates, black boxes are the best option as alternative roosts for evicted colonies. However, on particularly hot days, bats left them, suggesting that black boxes could be improved by creating a wider thermal range. 相似文献

We studied habitat selection by Rhinolophus euryale in a rural area of southern Italy in 1998-2000 by radio-tracking. Two comparisons were carried out, one between habitat occurrence within individual home ranges and within the study area, the other between time spent in each foraging habitat and habitat occurrence within the home range. The first analysis showed that olive groves and conifer plantations were, respectively, the most and the least important habitats. The second analysis highlighted the importance of woodland for R. euryale, while urban sites, open areas and conifer plantations were avoided. We recommend that clearing of continuous, large areas of woodland for tree harvesting should be avoided. Conifers should not be used for reforestation. Urbanisation should be limited in the areas of greatest importance for the species, and linear landscape elements such as tree lines and hedgerows should be maintained. 相似文献

Protection of roosting habitat is essential to the conservation of bats in human-dominated landscapes. To help define bat roosting needs in suburban settings, we used radio telemetry to locate day roosts of a common North American species (Myotis yumanensis) within a residential area in California. Between June and August 2000, we tracked 16 bats to 20 roosts in two buildings and 18 trees. We used multiple logistic regression to assess roost selectivity at multiple spatial scales. Of 15 tree, plot, and site characteristics considered, only three helped distinguish roosts from random comparisons: tree diameter, distance to water, and forest cover. Myotis yumanensis preferred large trees (mean diameter 115 cm), and roosted only in the five species of largest mean diameter (Sequoia sempervirens, Pseudotsuga menziesii, Quercus agrifolia, Quercus lobata, and Acer macrophyllum). At the site level, these bats selected roosts that were close to water and had substantial forest cover in the surrounding 100-m radius. Unlike other North American bats, they often roosted in live trees (89% live). Relatively high roost fidelity (mean 4.8 days) and large travel distances between consecutive roosts (mean 1130 m) and between capture sites and roosts (mean 2007 m) may indicate a greater degree of roost limitation at this site versus other research sites. We recommend the preservation of large trees and forested parkland, particularly along stream corridors, to help maintain bat populations in urbanizing landscapes. 相似文献

The location of roosts of two species of vespertilionid bats, the lesser long-eared bat (Nyctophilus geoffroyi) and Gould’s wattled bat (Chalinolobus gouldii), was investigated in a remnant vegetation-farmland mosaic and adjacent floodplain forest in south-eastern Australia. A total of 45 individuals of N. geoffroyi and 27 C. gouldii were fitted with radio transmitters, which resulted in the location of 139 and 89 roosts respectively. Selection of roosting habitat showed both inter- and intra-specific differences. At the landscape level, locations of roosts used by male and female N. geoffroyi differed markedly. Most males roosted in the farmland mosaic within 3 km (mean 1.9±2.9 km) of where they were captured while foraging in remnant vegetation. In contrast, roost sites of females were predominantly in the floodplain forest, 6-12 km from their capture site in farmland (mean for all females, 6.7±2.9 km). All maternity roosts were in the extensive floodplain forest, 4-10 km from foraging areas. Distances moved by this species were greater than previously recorded and further than predicted by flight morphology. Most C. gouldii roosted in the floodplain forest, 4-10 km from their capture site (6.9±1.6 km). Within the floodplain forest, both species roosted in areas that had higher densities of hollow-bearing trees than generally available; dead hollow-bearing trees for N. geoffroyi and large, live trees for C. gouldii. The scale of movements undertaken by these species means that they can obtain resources from multiple landscape elements. Both species used different habitats for roosting and foraging despite the energetic costs of commuting relatively large distances. Conservation of bat populations in rural environments needs to be considered at the landscape scale, with particular attention to identifying landscape elements that provide key resources. 相似文献

The roosts of many IUCN-listed cave-roosting bat species are under threat from tourist development in SE Europe and other regions of the world. Much-needed conservation strategies require, among other information, an understanding of their roost movements and population dynamics, which can now be obtained relatively quickly using advanced models. We have studied the long-fingered bat, Myotis capaccinii, an obligate cave-dweller, in Dadia National Park, Greece. The species formed colonies of up to a few thousand individuals and was highly mobile, frequently switching summer roosts up to 39 km apart, even during late pregnancy. The bats migrated to distant hibernacula including a cave in Bulgaria 140 km NW of the Park. Adult recapture probabilities varied with season and sex: low female recapture rates in autumn, relative to spring and summer, indicated non-random temporary emigration following nursery colony dispersal. The opposite pattern was seen in males: increasing recapture rates in the autumn suggest that males gather in these roosts to mate with females in transit. Adult survival (0.86-0.94) was similar in females and males, similar in winter and summer, and comparable to recent estimates for other bats based on similar modelling techniques. Sex-based differences in juvenile recapture suggest female philopatry and male-biased dispersal. Our work shows that protection of M. capaccinii roosts must extend beyond the Park’s and indeed the country’s boundaries: its conservation requires large-scale, trans-national integrated conservation plans. Monitoring programs serve to track changes in the distribution and abundance of species. A major problem with most monitoring programs is that species detection is imperfect and some populations are inevitably missed. Therefore, in most monitoring programs the true distribution of a species will be underestimated. Here, we report a field test of the reliability and performance of a commonly used method to monitor the distribution of amphibians (anuran call surveys). We surveyed the distribution of four anuran species in western Switzerland, and estimated detection probabilities to account for imperfect species detection and used these estimates to adjust our estimate of site occupancy (i.e., distribution). Next, we assessed how detection probabilities were affected by weather and how site occupancy was affected by site specific covariates. For one species (Hyla arborea), call surveys proved efficient in determining the regional distribution with only few site visits because detection probabilities were relatively high. The call surveys apparently missed many populations of another common species (Bufo calamita) because detection probabilities were lower. Two other species (Bombina variegata and Alytes obstetricans) were uncommon and strong inference from the analysis is not possible. Thus, multispecies surveys may be inefficient for rare species. Estimates of detection probabilities were used to calculate how many site visits are necessary to infer the absence of a species with some predetermined statistical certainty. The implications of “false absences” are important in ecology as they are known to bias usual habitat suitability models and overestimate extinction/colonization events in metapopulations. Large-scale monitoring programs would benefit from the application of an estimation-based approach to monitoring the distribution of species. 相似文献

The abundance and population structure of pseudomonads in soils collected from long-(1006 years) and short-(54 years) term grapevine monocultures in Switzerland were examined across five soil horizons within the 1.20-1.35 m range. Soil samples were baited with grapevine, and rhizosphere pseudomonads containing the biocontrol genes phlD (2,4-diacetylphloroglucinol synthesis) and/or hcnAB (hydrogen cyanide synthesis) were analyzed by MPN-PCR. The numbers of total, phlD+ and hcnAB+ pseudomonads decreased with depth by 1.5-2 log (short-term monoculture) and 3-3.5 log (long-term monoculture). In addition, the percentages of phlD+ (except in short-term monoculture) and hcnAB+ pseudomonads were also lower in deeper horizons. RFLP-profiling of phlD+ and hcnAB+ pseudomonads revealed three phlD and twelve hcnAB alleles overall, but the number of alleles for both decreased in relation to depth. The only phlD allele found in deeper horizons was also found in topsoil, whereas one hcnAB allele (k) found in deeper horizons in long-term monoculture was absent in the topsoil. This suggests that certain Pseudomonas ecotypes are adapted to specific depths. Four hcnAB alleles enabled discrimination between monocultures. We conclude that soil depth is a factor selecting phlD and hcnAB genotypes, and that the allelic diversity of the two biocontrol genes decreases with depth. 相似文献

We investigated roost selection by Barbastella barbastellus in a mountainous area of central Italy. Twenty-five bats, mostly lactating females, were radio-tracked to 33 roost trees. Trees in unmanaged woodland were favoured as roost trees; woodland subject to limited logging was used in proportion to availability, and areas where open woodland and pasture occurred were avoided. Selection depended on tree condition (dead beech trees were preferred) and height (roost trees were taller than random ones). Cavity selection was based on cavity type, height and entrance direction: roost cavities were mainly beneath loose bark, at a greater height above ground and facing south more frequently than random cavities. Untouched areas of mature woodland should be preserved to provide roosting conditions for B. barbastellus. In logged areas, harvesting protocols should save dead and mature trees; frequent roost switching and small colony size imply that large numbers of roost trees are needed. 相似文献

Two root-colonizing Fusarium strains, Ls-F-in-4-1 and Rs-F-in-11, isolated from roots of Brassicaceae plants, induced the resistance in Lepidium sativum seedlings against Pythium ultimum. These strains caused an increase in the content of benzyl isothiocyanate, and of its precursor glucotropaeolin, in the roots of the host plants. The increased isothiocyanate content is one of the factors contributing to the resistance of L. sativum against P. ultimum. To be transformed into the fungitoxic compound benzyl isothiocyanate, glucotropaeolin has to be hydrolyzed by myrosinase, which can be produced either by plants or microorganisms. The Fusarium strain Ls-F-in-4-1 has a myrosinase activity but the strain Rs-F-in-11 has not. These results suggest that both strains are able to trigger the metabolic pathway leading to benzyl isothiocyanate production in the plant. In the case of the myrosinase-negative strain Rs-F-in-11, hydrolyzation into isothiocyanate is only due to the myrosinase activity of the plant, and in the other case, the myrosinase produced by the strain Ls-F-in-11 also would contribute to the production of isothiocyanate. Livingstone's fruit bats, Pteropus livingstonii, (Pteropodidae: Chiroptera) are endemic to only two islands, Moheli and Anjouan of the Comoros Islands in the West Indian Ocean. With an estimated population of less than 2000 individuals, P. livingstonii is listed as Critically Endangered on the International Union for the Conservation of Nature's Red List. Yet the habitat characteristics of these giant fruit bats, or flying foxes, are poorly known. I conducted field research during 5 months in 1998 focused on finding new roost sites, measuring characteristics of both roost sites and roost trees at new as well as previously identified roost sites, and recording the same characteristics for a set of absence sites. I identified six new roost sites allowing me to measure the characteristics of 15 known roost sites and seven absence sites. The study spanned both dry and rainy seasons and I found a significant increase in the number of individuals at roost sites during the rainy season. Bat roost sites were associated with presence of water, southeast aspect, steep slopes, and presence of several native tree species. Roost habitat was characterized based on biotic, abiotic and human factors. This research on habitat characteristics is contributing to the development of a conservation action plan for the Livingstone's fruit bats. However, further research on dry season roost sites and distribution as well as feeding sites is necessary in order to better understand their seasonal habitat requirements and therefore ensure the conservation of P. livingstonii. 相似文献

The possible transfer of genes from Bacillus thuringiensis subsp. kurstaki (Btk) to indigenous Bacillus spp. was investigated in soil samples from stands of cork oak in Orotelli (Sardinia, Italy) collected 5 years after spraying of the stands with a commercial insecticidal preparation (FORAY 48B) of Btk. Two colonies with a morphology different from that of Btk were isolated and identified as Bacillus mycoides by morphological and physiological characteristics and by 16S rDNA analysis. Amplification by the polymerase chain reaction (PCR) of the DNA of the two isolated B. mycoides colonies with primers used for the identification of the Btk cry genes showed the presence of a fragment of 238 bp of the cry1Ab9 gene that had a similarity of 100% with the sequence of the cry1Ab9 gene present in GenBank, indicating that the isolates of B. mycoides acquired part of the sequence of this gene from Btk. No cells of Btk or B. mycoides carrying the 238-bp fragment of the cry1Ab9 gene were isolated from samples of unsprayed control soil. However, the isolates of B. mycoides were not able to express the partial Cry1Ab protein. Hybridization with probes for IS231 and the cry1Ab9 gene suggested that the inverted repeated sequence, IS231, was probably involved in the transfer of the 238-bp fragment from Btk to B. mycoides. These results indicate that transfer of genes between introduced Btk and indigenous Bacillus spp. can occur in soil under field conditions. 相似文献

Populations of rare or elusive large mammals are difficult to monitor, because they usually are secretive, solitary, occur at low densities, and have large home ranges. The global trend of generally decreasing large carnivore populations necessitates new, feasible, reliable, and cost-effective monitoring methods. We evaluate an index method developed for monitoring populations of moose (Alces alces) based on voluntarily and systematically collected observations from hunters, corrected for effort, for use in monitoring populations of large carnivores in Sweden. For our evaluation, we used independent estimates of minimum brown bear (Ursus arctos) densities from DNA-based scat surveys and brown bear distribution from mandatory reports from successful bear hunters. We verified that the index correctly reflected bear distribution. We also found strong linear relationships between the indices and the independent density estimates for bears at the scale of local management units (about 1000-2000 km2) in all three regional study areas (adjusted R2 = 0.88-0.60). Our results suggest that systematic, effort-corrected reports of observed animals can be an alternative and accurate monitoring method for the conservation and management of large mammals occurring over large areas when large numbers of willing volunteers are available (effort >30,000 h). 相似文献

Two indole-producing Paenibacillus species, known to be associated with propagules of arbuscular mycorrhizal (AM) fungi, were examined for their mycorrhization helper bacteria activity at pre-symbiotic and symbiotic stages of the AM association. The effects were tested under in vitro and in vivo conditions using an axenically propagated strain of the AM fungus Glomus intraradices and Glycine max (soybean) as the plant host. The rates of spore germination and re-growth of intraradical mycelium were not affected by inoculation with Paenibacillus strains in spite of the variation of indole production measured in the bacterial supernatants. However, a significant promotion in pre-symbiotic mycelium development occurred after inoculation of both bacteria under in vitro conditions. The Paenibacillus rhizosphaerae strain TGX5E significantly increased the extraradical mycelium network, the rates of sporulation, and root colonization in the in vitro symbiotic association. These results were also observed in the rhizosphere of soybean plants grown under greenhouse conditions, when P. rhizosphaerae was co-inoculated with G. intraradices. However, soybean dry biomass production was not associated with the increased development and infectivity values of G. intraradices. Paenibacillus favisporus strain TG1R2 caused suppression of the parameters evaluated for G. intraradices during in vitro symbiotic stages, but not under in vivo conditions. The extraradical mycelium network produced and the colonization of soybean roots by G. intraradices were promoted compared to the control treatments. In addition, dual inoculation had a promoting effect on soybean biomass production. In summary, species of Paenibacillus associated with AM fungus structures in the soil, may have a promoting effect on short term pre-symbiotic mycelium development, and little impact on AM propagule germination. Field pea (Pisum sativum L.) is widely grown in South Australia (SA), often without inoculation with commercial rhizobia. To establish if symbiotic factors are limiting the growth of field pea we examined the size, symbiotic effectiveness and diversity of populations of field pea rhizobia (Rhizobium leguminosarum bv. viciae) that have become naturalised in South Australian soils and nodulate many pea crops. Most probable number plant infection tests on 33 soils showed that R. l. bv. viciae populations ranged from undetectable (six soils) to 32×103 rhizobia g−1 of dry soil. Twenty-four of the 33 soils contained more than 100 rhizobia g−1 soil. Three of the six soils in which no R. l. bv. viciae were detected had not grown a host legume (field pea, faba bean, vetch or lentil). For soils that had grown a host legume, there was no correlation between the size of R. l. bv. viciae populations and either the time since a host legume had been grown or any measured soil factor (pH, inorganic N and organic C). In glasshouse experiments, inoculation of the field pea cultivar Parafield with the commercial Rhizobium strain SU303 resulted in a highly effective symbiosis. The SU303 treatment produced as much shoot dry weight as the mineral N treatment and more than 2.9 times the shoot dry weight of the uninoculated treatment. Twenty-two of the 33 naturalised populations of rhizobia (applied to pea plants as soil suspensions) produced prompt and abundant nodulation. These symbioses were generally effective at N2 fixation, with shoot dry weight ranging from 98% (soil 21) down to 61% (soil 30) of the SU303 treatment, the least effective population of rhizobia still producing nearly double the growth of the uninoculated treatment. Low shoot dry weights resulting from most of the remaining soil treatments were associated with delayed or erratic nodulation caused by low numbers of rhizobia. Random amplified polymorphic DNA (RAPD) polymerase chain reaction (PCR) fingerprinting of 70 rhizobial isolates recovered from five of the 33 soils (14 isolates from each soil) showed that naturalised populations were composed of multiple (5-9) strain types. There was little evidence of strain dominance, with a single strain type occupying more than 30% of trap host nodules in only two of the five populations. Cluster analysis of RAPD PCR banding patterns showed that strain types in naturalised populations were not closely related to the current commercial inoculant strain for field pea (SU303, ≥75% dissimilarity), six previous field pea inoculant strains (≥55% dissimilarity) or a former commercial inoculant strain for faba bean (WSM1274, ≥66% dissimilarity). Two of the most closely related strain types (≤15% dissimilarity) were found at widely separate locations in SA and may have potential as commercial inoculant strains. Given the size and diversity of the naturalised pea rhizobia populations in SA soils and their relative effectiveness, it is unlikely that inoculation with a commercial strain of rhizobia will improve N2 fixation in field pea crops, unless the number of rhizobia in the soil is very low or absent (e.g. where a legume host has not been previously grown and for three soils from western Eyre Peninsula). The general effectiveness of the pea rhizobia populations also indicates that reduced N2 fixation is unlikely to be the major cause of the declining field pea yields observed in recent times. 相似文献

A population viability analysis is important for the management of endangered populations and requires the estimation of survival parameters. The long-tailed bat (Chalinolobus tuberculatus) is one of only two native terrestrial mammals currently found in New Zealand and is classed as vulnerable. Its viability in temperate beech (Nothofagus) forest, Eglinton Valley, Fiordland, New Zealand was estimated using mark-recapture data collected between 1993 and 2003 using the Program MARK. Survival was estimated based on a total of 5286 captures representing 1026 individuals. Overall annual survival varied between 0.34 and 0.83 but varied significantly among three sub-populations and with sex and age. Females generally had a higher survival rate compared to males; and adults had higher survival relative to juveniles. Survival of all bats was lower in years when the number of introduced mammalian predators was high and when the winter temperature was warmer than average. High numbers of introduced predators occurred during three of the 10 years in the study. Climate change may mean that the conditions that promote high predator numbers may occur more frequently. A preliminary population viability analysis using a projection matrix on the overall adult female population showed an average 5% decline per year (λ = 0.95). Increased predator control targeting a range of predators is required in years when their numbers are high in order to halt the decline of this population of long-tailed bats. A collection of 299 isolates of rhizobia nodulating Medicago truncatula was isolated from 10 Tunisian soils and was characterized by restriction fragment length polymorphism analysis of polymerase chain reaction (PCR/RFLP) of 16S rRNA gene. Results showed that 227 and 72 isolates were assigned, respectively, to Sinorhizobium meliloti and Sinorhizobium medicae. In 9 out of 10 soils S. meliloti was detected, whereas S. medicae was recovered from only 5 out of 10 soils. The cross-nodulation of three populations of M. truncatula grown on Bulla Regia soil, which contained naturally the two Sinorhizobium species, showed that M. truncatula population collected from Amra site was selective to S. meliloti at least in soil conditions. Forty-eight isolates of each Sinorhizobium species trapped by M. truncatula populations collected from Bulla Regia, Soliman and Rhayet sites on Bulla Regia soil were characterized by repetitive extragenic palindromic-PCR (REP-PCR) and showed a clear distinction between the two Sinorhizobium species and a higher diversity for S. meliloti. 相似文献

The capability of native bacterial strains isolated from Lolium perenne rhizosphere to behave as plant growth promoting bacteria and /or biocontrol agents was investigated. One strain (BNM 0357) over 13 isolates from the root tips of L. perenne resulted proved to be nitrogenase positive (ARA test) and an IAA producer. Conventional tests and the API 20E diagnostic kit indicated that BNM 0357 behaves to the Enterobacteriaceae family and to the Enterobacter genus. Molecular identification by 16S rRNA sequence analysis indicated that BNM 0357 had the highest similarity to Enterobacter ludwigii (EN-119). Isolate BNM 0357 had the capability to solubilize calcium triphosphate and to antagonize Fusarium solani mycelial growth and spore germination. Strain BNM 0357 also showed the ability to improve the development of the root system of L. perenne. This study disclosed features of E. ludwigii BNM 0357 that deserve further studies aimed at confirming its putative importance as a PGPR. 相似文献

A cultivation-based approach was used to determine the in vitro antagonistic potential of soil bacteria towards Rhizoctonia solani AG3 and Fusarium oxysporum f. sp. lini (Foln3). Four composite soil samples were collected from four agricultural sites with previous documentation of disease suppression, located in France (FR), the Netherlands (NL), Sweden (SE) and the United Kingdom (UK). Similarly, two sites from Germany (Berlin, G-BR; and Braunschweig, G-BS) without documentation of disease suppression were sampled. Total bacterial counts were determined by plating serial dilutions from the composite soil samples onto R2A, AGS and King's B media. A total of 1,788 isolates (approximately 100 isolates per medium and site) was screened for antifungal activity, and in vitro antagonists (327 isolates) were found amongst the dominant culturable bacteria isolated from all six soils. The overall proportion of antagonists and the number of isolates with inhibitory activity against F. oxysporum were highest in three of the suppressive soils (FR, NL and SE). Characterization of antagonistic bacteria revealed a high phenotypic and genotypic diversity. Siderophore and protease activity were the most prominent phenotypic traits amongst the antagonists. The composition and diversity of antagonists in each soil was site-specific. Nevertheless, none of the antimicrobial traits of bacteria potentially contributing to soil suppressiveness analyzed in this study could be regarded as specific to a given site. 相似文献

The effect of the soil yeast, Rhodotorula mucilaginosa LBA, on Glomus mosseae (BEG n°12) and Gigaspora rosea (BEG n°9) was studied in vitro and in greenhouse trials. Hyphal length of G. mosseae and G. rosea spores increased significantly in the presence of R. mucilaginosa. Exudates from R. mucilaginosa stimulated hyphal growth of G. mosseae and G. rosea spores. Increase in hyphal length of G. mosseae coincided with an increase in R. mucilaginosa exudates. No stimulation of G. rosea hyphal growth was detected when 0.3 and 0.5 ml per petri dish of yeast exudates was applied. Percentage root length colonization by G. mosseae in soybean (Glycine max L. Merill) and by G. rosea in red clover (Trifolium pratense L. cv. Huia) was increased only when the soil yeast was inoculated before G. mosseae or G. rosea was introduced. Beneficial effects of R. mucilaginosa on arbuscular mycorrhizal (AM) colonization were found when the soil yeast was inoculated either as a thin agar slice or as a volume of 5 and 10 ml of an aqueous solution. R. mucilaginosa exudates (20 ml per pots) applied to soil increased significantly the percentage of AM colonization of soybean and red clover. 相似文献
