The effect of reproduction was investigated on females of Hungarian strain W, French strain F, and their cross‐breed 1X whose ovulation was stimulated with carp pituitary (0.3 mg kg?1and, after 12 h, 2.7 mg kg?1) or Ovopel (one‐fifth of a pellet per kg and, after 12 h, one pellet per kg). It was found that in the case of Ovopel treatment, the percentage of spawning females of strain F and the cross‐breed 1X was higher than in the hypophysed fish compared. The applied ovulation stimulators did not significantly affect the weight of obtained eggs, whereas the significant (P ≤ 0.01) effect was recorded with respect to the quality of eggs after 12‐, 24‐, 36‐ and 48‐h of incubation. After Ovopel stimulation, the quality of eggs was better. The origin of the females had no statistically significant effect on the weight of eggs although the yield of eggs from fish of strain W was much smaller than that from females of strain F and the 1X cross‐breed. The interaction between the ovulation stimulator and the provenance of the females was significant (P ≤ 0.05) for the percentage of live embryos after 48‐h of incubation of eggs. Eggs of the best quality (and highest weight) were obtained from fish of strain F and cross‐breed 1X treated with Ovopel. In females of strain F that spawned within 6 and 10 h after the second Ovopel injection, the effect of the ovulation time on the weight of eggs was non‐significant. It was significant with respect to the percentage of egg fertilization and of live embryos after 36‐h of incubation (P ≤ 0.01 and P ≤ 0.05 respectively). Chloramphenicol (CAP) is a banned substance in fish farming; yet, residues are sometimes detected in farmed fish products. Therefore, it is important that tissue distribution and elimination rates of CAP are measured to allow monitoring of fish farms. The tissue distribution and residue of CAP in carp (Cyprinus carpio L.) after muscle injection was investigated. Chloramphenicol concentrations were determined using high‐performance liquid chromatography. Pharmacokinetics were used for the studies of CAP residue and the results showed that the CAP was easily assimilated in the liver, serum and gill, with the absorption rates of 6383, 3.02 and 1.39 h?1, respectively, following a single‐dose injection of 80 mg kg bw?1. The elimination half‐lives (t1/2) were 22.28, 15.47, 14.87, 9.28 and 5.32 h for the liver, serum, gill, muscle and kidney respectively. The t1/2 after 5 days of multi‐doses of repeated CAP injection with 40 mg kg bw?1 was 1.09, 0.887, 0.872, 0.476 and 0.617 days for the five tissues respectively. This study focuses on reducing total phosphorus loading (T-P) from carp culture through improved feed formulation. Since phosphorus (P) contained in fish meal (FM) mainly in the form of tricalcium phosphate is not available to carp, which lack a stomach, the reduction of FM from their diets is effective for lowering T-P. Thus in this experiment, six diets (crude protein < 35%, digestible energy > 3.5 kcal g−1) were designed by substituting FM (10%−25%) with alternate protein ingredients such as poultry feather meal (PFM; 5%−10%), blood meal (BM; 5%−7%) and defatted soybean meal (dSBM; 4%−9%). All diets followed the Kasumigaura ‘Feed Standard’. The total dietary P was 1.0%−1.4% and water extractable P available to carp was 0.66%−0.71%, the levels meeting the dietary requirement of carp. A feeding trial was conducted with juvenile carp (4.6 ± 0.7 g) for 12 weeks at a mean water temperature of 23.7°C. The T-P loading from fish fed the different diets was estimated based on absorption and retention of dietary P. Growth performance corresponded to increasing levels of FM inclusion, being highest in the fish fed 25% FM diet; however, the decrease in T-P was achieved at the lower FM levels. The T-P (based on P retention) ranged from 8.9 to 11.7 kg t−1 production, much lower than that from the commercial diets (9.1–26.4 kg t−1 production). These results indicated that the reduction of FM levels in carp diets to 15%−20% through the combined use of PFM, BM and dSBM effectively lowered T-P. Moreover, the formulated diets were also found to be better than commercial diets in lowering the N loading from carp culture. 相似文献

Investigations of the effects of controlled reproduction were conducted on carp females of Polish strain 6 and Hungarian strain W after ovulation stimulation with carp pituitary homogenate (CPH) (0.3 mg+2.7 mg kg?1 body weight), Ovopel (1/5+1 pellet kg?1 body weight) or Dagin (1 dose kg?1 body weight). In both strains, the highest percentage of spawning females was found after the Ovopel treatment. After treatments with CPH and Dagin this percentage did not differ in strain 6; however, after Dagin, it was higher in fish of strain W. After CPH or Ovopel stimulation the females of line 6 yielded eggs of higher weight compared with line W. After the Dagin treatment the weight of eggs obtained from females, both strains was similar but much lower than after the two remaining agents. The quality of eggs expressed both by the percentage of fertilization and that of living embryos after 24 and 36 h incubation was similar in the two strains only in the group treated with Ovopel. After CPH application eggs of better quality were obtained from strain 6 compared with strain W; however, after the treatment with Ovopel, eggs of much better quality were noted in strain W. In general, the best effects of reproduction were recorded in the case of Ovopel and the poorest in that of Dagin. Females of strain 6 yielded eggs of a higher weight but the percentages of spawning females were higher in strain W after each of the applied spawning agents. 用经紫外线灭活的团头鲂精子激活橙黄色锦鲤的卵子,在4℃的水温下冷休克处理卵子30 min使其染色体加倍,获得了孵化率为30.6%、存活率为24.2%的雌核发育锦鲤,表明该雌核发育锦鲤研制方法具有较高的效率.利用外周血细胞培养方法制备了雌核发育锦鲤染色体并利用核型分析方法对其核型进行了分析,发现雌核发育锦鲤染色体数目为100,核型公式为22 m+34 sm+22 st+22 t;表明雌核发育锦鲤为二倍体,其核型与锦鲤母本一致.利用石蜡切片方法,随机选取10尾1龄雌核发育锦鲤性腺进行检测,发现其性腺全部为卵巢;并且在繁殖季节随机检测100尾2龄雌核发育锦鲤,没有l尾能挤出精液,表明雌核发育锦鲤全部为雌性,为证明雌性锦鲤的性别决定类型为XX提供了重要证据.此外,还对雌核发育锦鲤与母本的的SOX基因遗传关系、外形、体色等进行了比较,结果表明雌核发育锦鲤与母本有相同的SOX基因扩增片段,为证明其为雌核发育锦鲤而非杂交后代提供了重要证据;同时,雌核发育锦鲤保留了母本体态优美的特点,且具有色泽更鲜艳等优点,表明该雌核发育方法产生了遗传改良效果.雌核发育锦鲤的获得为该观赏鱼的提纯复壮、遗传改良以及性别决定和性别控制研究奠定了基础. 相似文献

In an in vitro study it has been shown that HCG at a dose of 100 i.u./ml does not cause an increase in the percentage of mature oocytes (follicles) compared to a control incubation. However, HCG together with carp pituitary homogenate (c.p.h.) at ratios of 6:4, 5:5, 4:6 (dosages of 60, 50, 40 i.u./ml of HCG and 40, 50, 60 μg/ml of c.p.h.) gave a higher mean percentage of mature follicles than did c.p.h. alone at a dose of 100 μg/ml.Based on the in vitro results, in vivo experiments with HCG and c.p.h. were conducted in order to decrease the amount of c.p.h. used in the induction of artificial spawning in carp.Experiments were carried out on 200 adult female carp. The use of HCG and c.p.h. in two injections (ratios 6:4, 5:5 and 4:6) at a dose 10 times higher than in the in vitro experiment did not increase either the percentage of mature oocytes or the number of ovulated females compared to groups injected with c.p.h. alone at the same dose. Classical hypophysation induced ovulation in the majority of injected females (60–80%).The results indicate that HCG with c.p.h. is ineffective in reducing the amount of c.p.h. required for artificial spawning of carp. 相似文献

The physiological response of common carp, Cyprinus carpio (L.) to increased stocking density and an additional acute net confinement stressor was investigated. Stocking densities were increased from 28.4 to 56.8 or 113.6 kg m?3 by the use of crowding screens and fish were sampled from the crowded groups after 15, 39 and 87 hours of crowding (hc). A transient elevation of plasma cortisol was found in the higher density group after 15 hc before values returned to control levels. Increased stocking density also increased plasma levels of glucose, free fatty acids (FFA) and lactate during the experimental period. No effect was found on oxygen radical production in the blood, haemoglobin or haematocrit levels, but leucocrit values were lower after 39 hc in both groups. Net confinement resulted in a significant increase in plasma cortisol levels in all groups. However, after 15 hc, cortisol and FFA levels in both crowded plus confinement groups were higher than in the control and confined groups. Confinement resulted in increased oxygen radical production in the crowded plus confined groups at all times. Results indicate that although carp responded with a mild stress response to increased stocking density and adaptation occurred by 87 hc, the crowded fish were more sensitive to an additional acute stressor. 相似文献

The interactioneffect was demonstrated as insignificantbetween altitude*sex and betweenstrain*sex.The comparative test of analysis of variance(ANOVA) revealed significantly (P < 0.05)higher weight of fish after killing on lowaltitude (1611.6 g) than on high altitude(1090.3 g), but processed body yield andpercentage of fillet without skin was notsignificantly different (64.0 and 34.2 for lowaltitude, 64.2 and 33.4 for high altitude). Thepercentage of processed body weight andpercentage of fillet without skin in analysisboth altitudes were significantly better infemales (65.5 and 34.6%) than in males (62.5and 33.00%), respectively. The highestsignificant differences in weight of fish afterkilling were found between HSM (890.8 g) andHSM × AC (1283.3 g) in high altitude. - 1. In Australia, the carp Cyprinus carpio L. is regarded as a threat to the native fish and the aquatic environment. In recent years, Botany Wetlands, a significant coastal wetland in the Sydney region, has been invaded by the undesirable cyprinids, carp and goldfish (Carrasius auratus L.).
- 2. In 1996 a cyprinid removal programme commenced at Botany Wetlands with the objective of managing the invasive species, increasing fish diversity, reducing cyanobacterial blooms and hence enhancing the aquatic habitat. Using electrofishing and gill netting, 4073 carp and 261 goldfish, amounting to 10 117 kg of cyprinid biomass were removed between 1996 and 2004. The captured carp ranged from 60 to 835 mm. Males matured before females. Carp between 350 and 680 mm in length grew at the rate of 1.66±0.38 g day?1.
- 3. The success of the programme was monitored by assessing four indicators related to carp populations and two related to habitat. The former included the pattern of length–frequency distribution, mean weight per size class, condition factor (CF) and the catch per unit effort (CPUE), and the latter the Secchi disc transparency and cyanobacterial counts. After 8 yr of removal, the plots of carp length–frequency distribution flattened, CF decreased from 2.86 to 1.82 and CPUE decreased from 97 kg day?1 to 50 kg day?1. A 10‐fold decrease occurred in cyanobacterial counts and the Secchi disc transparency increased by 20%. More than 20 000 fingerlings of native Australian bass Macquaria novemaculeata (S.) have been introduced to the wetlands, increasing the potential for juvenile carp predation and biodiversity.

The F1 progeny from parents selected randomly for differences in performance traits from different families of mirror carp were used as a mapping panel. Linkage maps constructed using Join Map 4.0 software with 992 microsatellite polymorphic markers ultimately yielded 51 groups. Composite interval mapping (CIM) of superoxide dismutase (SOD) trait of three tissues (liver, anterior and posterior intestine) was conducted for quantitative trait loci (QTL) location using Map QTL 6.0 software. Results shows that seven QTLs significantly (P < 0.05) explained 8.2–18.5% of the phenotypic variations in liver, five QTLs explained 7.9–11.1% of the variation in anterior intestine (P < 0.05) and six QTLs were located and explained 6.1–15.8% of the phenotypic variations in posterior intestine (P < 0.05). NCBI BLASTn was used for homology analysis between the sequences from markers used in the located QTL regions and sequences from the zebrafish database, and many protein relating with the SOD expression were blasted (similarity >85%). These results showed that it is highly probable that these QTLs are the main regions of SOD expression. The data also support the value of forward genetic studies on organism development and serve as a basis for selection. 相似文献

The increasing need for aquafeed resources and the finite availability of conventional feed resources are making it necessary to search for alternative high‐protein resources that are not used as human food. The earthworm Perionyx excavatus was tested as a feed ingredient in diets for common carp. An experiment was conducted to evaluate the potential of earthworm powder as a replacement for fishmeal. In a recirculation aquarium system, triplicate groups of five common carp (Cyprinus carpio L.) were fed a control feed (fishmeal based protein), or experimental diets in which 30% (EW30), 70% (EW70), or 100% (EW100) of fishmeal protein was replaced by earthworm protein. Fish growth, feed digestibility and feed utilization were monitored. Growth rate, protein efficiency and energy retention in fish were similar (EW30, EW100) or higher (EW70) for diets containing earthworm meal compared to the control diet. Protein digestibility in EW30, EW70 and EW100 was higher than in the control diet, but in (EW100), lipid conversion was lower. We conclude that earthworm is a suitable partial replacement for fishmeal in feeds for common carp. 相似文献

This study investigated the lipid metabolism responses of common carp (Cyprinus carpio L.) to mulberry leaf meal (MLM) replacing of some fish meal in diet. Six iso‐nitrogenous and iso‐energetic diets containing different levels of MLM (0%, 6.3%, 12.6%, 18.9%, 25.2% and 31.5%) were prepared and fed to common carps in tanks for 56 days. Analysis showed that serum total protein, albumin contents and glutamic‐pyruvic transaminase, glutamic oxalacetic transaminase activities were not significantly different among treatments, blood glucose, cholesterol, high‐density lipoprotein and low‐density lipoprotein but triglyceride concentration depicted a declining trend in MLM supplementation groups compared with control. Liver and muscle lipid content decreased with increased supplementation of MLM in the diet. Hepatic lpl showed an up‐regulated trend in the MLM groups, apo‐a1 was up‐regulated in common carp fed 31.5% MLM diet, pparα was up‐regulated in 18.9% and 25.2% MLM groups, while apo‐a4, pparβ and pparγ were down‐regulated in all the MLM‐treated groups compared with control, fabp1 was down‐regulated in fish fed high‐MLM diets (≥25.2%). The results revealed that half of fish meal in common carp diet could be replaced by MLM with no hepatotoxicity; MLM promoted the reversal of cholesterol transport thereby reducing the cholesterol in blood; MLM elevated the ability of fatty acid oxidation of the common carp, but did not activated the activity of lipoprotein lipase fully, and the suppressed transportation of fatty acid affected the degradation of triglyceride in blood together; MLM reduced liver lipid contents might through suppressing the isolation and proliferation of adipocytes. 相似文献

The Japanese ornamental (koi) carp is a popular decorative fish all over the world. In koi, clones have not yet been obtained, although production of fish with identical colour patterns could be of commercial interest. Mitotic gynogenetic progenies are essential for subsequent production of clones in fish. However, resulting late‐shocked progenies may be contaminated with meiotic gynogens from spontaneous suppression of the second meiotic division in eggs. In this study, microsatellite DNA markers were used to confirm mitotic gynogenetic origin of obtained late‐shocked progenies. Recombination rate (y) and mapping distance relative to centromere (M‐C) of 10 microsatellite loci were determined based on percentage of heterozygotes in meiotic gynogenetic progenies. The range of y varied from 0.01 to 0.96 and the M‐C map ranged from 0.5 to 48 cM. The mean value of y over the 10 loci was 0.481. Six loci, which had y 0.47 and higher, were used as markers in two late‐shocked gynogenetic progenies. Complete homozygosity was revealed at all six microsatellite loci indicating mitotic gynogenetic origin of analysed progenies. 相似文献

Random amplified polymorphic DNA (RAPD) markers have been used in a variety of genetic studies in fisheries and aquaculture. Most population studies are performed without preliminary data demonstrating the Mendelian inheritance and reproducibility of RAPD markers. In this study, the inheritance and reproducibility of RAPD markers was examined in two consecutive generations of common carp, Cyprinus carpio L. Variability and segregation of RAPD markers were investigated in one F1 progeny and three F2 progenies. Seventy-four RAPD markers were generated by five primers using DNA extracted from the initial ornamental (koi) common carp female and wild-type colour common carp male. Fifty-five of these RAPD markers were transmitted to the F1 progeny and the inheritance patterns were analysed. Twenty RAPD markers were fully reproducible and demonstrated dominant simple Mendelian inheritance patterns in two consecutive generations. Twenty-four RAPD markers were not reproducible in all progenies. Thirteen markers displayed inheritance ratios in the progenies that did not fit simple Mendelian inheritance patterns. Non-reproducibility of RAPD markers and distorted ratios may be caused by the absence of amplification, poor amplification or by the appearance of artefact bands. Random amplified polymorphic DNA markers with poor reproducibility and non-Mendelian inheritance can lead to misinterpretations of data in population studies, resulting in errors in the estimation of genetic diversity within and between individual populations. Therefore, it is recommended to first identify the set of reproducible RAPD markers that demonstrate Mendelian inheritance before application of the RAPD technique in population studies. 相似文献

鲤鱼(体重约16克)腹腔注射乙酸棉酚0,7.3,54,100,200,400mg/kg体重,经四天(96小时)后,死亡率分别是0,0,20%,70%,90%,100%。96小时半数致死剂量是63.6mg/kg体重。注射剂量高于54mg/kg体重时,发生腹腔红肿、体壁溃烂,甚至死亡。实验结果说明虽然棉籽水平的棉酚不引起鲤鱼的任何可见的中毒症状,但是腹腔注射大剂量棉酚可引起与哺乳类相似的中毒症状。 相似文献

The effects of a single injection of mammalian superactive analogue [d ‐Ser(tBu)6,Pro9‐NEt]‐LHRHa (20 μg/ kg?1) combined with the dopamine antagonist, haloperidol (HAL, 0.5 mg kg?1), for induction of ovulation in the koi carp broodstocks were determined under routine hatchery conditions. The results were compared with classic carp pituitary extract (CPE, double injection) application (water temperature 22 °C). Physiological saline (0.7% NaCl)‐injected fish were used as a control group and no ovulation occurred in this group. The spawning ratio was high in the LHRHa+HAL treatment group and in the CPE treatment group (6/7 and 5/7 respectively). The latency period was 14–16 h in the LHRHa+HAL treatment group and 12–14 h in the CPE treatment group (after the second injection). There was no difference between the two ovulating groups with respect to the spawning index (the weight of eggs as a percentage of female body weight) and fertilization rate of eggs (P>0.05). As a result, ovulation can be induced successfully in koi carp broodstocks with 20 μg kg?1 [d ‐Ser(tBu)6,Pro9‐NEt]‐LHRHa+0.5 mg kg?1 HAL treatment in a single injection without decreasing the egg quality. Application of this combination can be useful for hatchery and broodstock management in koi carp culture. 相似文献
