1. Cockerels from meat-type (MT) and Leghorn (LG) stocks were fed on diets containing 0, 20, or 30 mg corticosterone/kg from 1 to 22 d after hatching. 2. Within MT and within LG stocks, responses to dietary corticosterone inclusion rates were similar; however, patterns differed between these major classes resulting in stock X diet interactions. 3. Traits responding to different inclusion rates of corticosterone in a dissimilar manner for MT and LG chickens included: immunoresponsiveness, body weight, efficiency of food utilisation, feathering; relative weights of liver, spleen, bursa, testes, breast and abdominal fat pad, as well as the proportion of liver lipid. 4. No differential responses occurred for relative adrenal weight, plasma xanthophyll concentrations, proportion of breast lipid and abdominal fat, or surface and cloacal temperatures. 5. Results indicated genetic differences in thresholds for response to corticosterone and in degree of response once thresholds were reached. Dietary infusion of L-[U-14C]tyrosine was used to estimate the fractional protein synthesis rates (FSR) in broiler and layer chickens. Six 2-wk-old birds of each strain were placed in individual metabolism cages and given a purified diet in agar-gel containing 2 microCi L-[U-14C]tyrosine for 6 h. The birds were sacrificed and the pectoralis major (PM) and two combined leg (LM) muscles (gastrocnemius and peroneous longus) were removed for analysis. Subgroups of chickens were sacrificed 3 d before and 3 d after infusion to observe changes in muscle composition to calculate fractional protein accretion rates (FAR). Fractional protein breakdown rates (FBR) were calculated by difference (FBR = FSR-FAR). Protein, ribonucleic acid (RNA) and deoxyribonucleic acid (DNA) concentrations were determined to observe relationships between these cellular constituents and FSR. Fractional whole body growth rate and FSR in PM was greater (P less than .05) in broiler than layer birds. The FSR in LM of the layer was not different (P greater than .05) from that of broilers, and from the FSR of the PM in each bird-type. The calculated FBR in the layer PM was at least 17% higher than that of the other muscles. Ratios of FSR to FBR indicated that 16% of the protein synthesized in the layer PM was retained, compared with 45% in the broiler PM. The RNA activity of the layer PM was less (P less than .05) than that of the other muscles investigated. Isolated porcine and bovine muscle strips were incubated in Krebs Ringer bicarbonate buffer to determine in vitro protein synthesis (PS) and protein degradation (PD) rates to validate the in vitro system for use with livestock species. The addition of 5X plasma concentrations of amino acids to the medium stimulated PS 30%. Addition of 3.5 mM leucine to a leucine-deficient buffer supplemented with amino acids decreased PD 37% and stimulated PS 24%. The addition of .1 U/ml insulin reduced PD 28% and increased PS 30%. Protein degradation was elevated in longitudinally split rat soleus and extensor digitorum longus muscles compared to their contralateral intact muscles. Muscle strips must be removed within 15 min of exsanguination because PD rates become greatly elevated thereafter. ATP concentrations declined during incubation, but the addition of ATP or creatine had no effect on either PD or PS. Neither PD nor PS was affected by the addition of transferrin, fetuin, ascorbate, dexamethasone or indomethacin to the incubation medium. However, muscle strips were sensitive to the addition of triiodothyronine (T3), PD was increased up to 75% as T3 concentration was increased, and PS rates doubled compared to controls. Serum from mature barrows or gilts had no effect on protein turnover, but the addition of 10% and 15% serum from boars increased both PD and PS. With fasted pigs a continual decline in PS occurred over 5 d, whereas PD was elevated at 3 d and then declined to rates comparable to the fed state after 5 d. These data suggest that the in vitro system has application for assessing relative changes that occur in vivo following nutritional, physiological and endocrinological manipulation. 相似文献

Reducing dietary protein concentration in isocaloric diets consistently decreased nitrogen and energy excretion, and increased dry matter (DM), non-protein DM (NPDM), energy retention and fatness. There were significant correlations, negative between dietary energy-to-protein (E:P) ratio and nitrogen excretion and positive between the E:P ratio and the retention of DM, NPDM and energy. Nitrogen excretion was correlated with energy excretion, and NPDM retention with energy retention. Corticosterone injections increased fatness despite significantly increasing nitrogen and energy excretion. A positive relationship was observed in corticosterone-treated birds between nitrogen excretion on the one hand and the retention of DM and NPDM on the other. Increased food intake because of a low dietary protein concentration was not evident when a sorghum-based diet was used. Moreover, corticosterone injections increased neither the food intake nor the fatness of chicks fed this diet. Decreasing the protein concentration in diets containing identical metabolisable energy (ME) levels slightly, but significantly increased apparent ME values in three out of four experiments. This study determined the Pectoralis (P) major mixed muscle protein turnover (PT) in two meat broiler lines, Line A and Line B, during the finishing grow-out feeding period (21–42 days) as affected by the dietary metabolizable energy (ME) levels and ambient temperatures. Experimental finishing diets consisted of 80, 90, 100, 110 and 120% ME of recommended nutrient guidelines for energy level. Fractional synthesis rates (FSR) or fractional degradation rates (FDR) were measured in P. major at day 36 and 42. Protein and fat mass gain were measured, and respective energy retention efficiencies as protein and fat (EREp and EREf) were determined. Metabolic heat production (HP) was also reported. Experimental feeding studies were conducted in cool season (24 hr mean: 69.91˚F and 63.98% RH) and in hot season (24 hr mean: 77.55˚F and 86.04% RH). Results showed that FSR or FDR values were not affected by dietary ME levels at day 36, whereas reduced FSR ( p < .05) were observed at day 42 fed diets with reduced ME levels (≤100% ME) which could have resulted from greater maintenance energy requirement of maturing broilers at that age. Broilers fed reduced ME diets (≤100% ME) maintained protein mass (equivalent to broilers fed ≥100%–120% ME) by reduced FDR and increased feed intake. Grow-out ambient temperature did not affect FSR or FDR values across ME levels. Line B retained higher protein mass, lower fat mass and greater HP compared to Line A. This was followed by higher feed intake in Line B. Further, Line B exhibited higher EREp and lower EREf across dietary ME levels. In summary, PT homeostasis and body composition changes in broiler lines studied seemed to be regulated by the birds’ intent to normalize energy intake as per physiological need by controlling feed intake. 相似文献

In Exp. 1, 36 individually penned steers (initial BW = 294 +/- 3.8 kg) were used to determine effects of dietary CP percentage and programming gain on performance and carcass characteristics. Steers were fed to achieve a predicted gain of 1.13 kg/d for the first 84 kg of gain and 1.36 kg/d for the next 124 kg of gain and were offered feed for ad libitum consumption for the final 58 kg of gain before slaughter. In these three phases of growth, steers were fed diets, sequentially, with the following CP percentages: HHH (16, 13.5, and 12.5%), LHH (9, 13.5, and 13%), or LLL (9, 9, and 9%). When predicted gain was 1.13 kg/d, ADG was greater (P < 0.01) for steers in the HHH (1.09 kg/d) vs LHH and LLL (0.83 kg/d) systems. When predicted gain was 1.36 kg/d, ADG and gain efficiency were greatest (P < 0.01) for steers in the LHH system. Overall ADG and gain efficiency were greater (P < 0.01) for steers in the HHH (1.46 kg/d, 0.194) and LHH systems (1.38 kg/d, 0.190), compared with steers in the LLL (1.21 kg/d and 0.166) system. Carcass fat thickness was lower for steers in the LHH (0.74 cm) system than for steers in the LLL system (1.09 cm). In Exp. 2, 18 individually penned steers (initial BW = 225 +/- 5.8 kg) were either offered a 13% CP diet for ad libitum intake (AL) throughout the 134-d experiment or fed a high- (16% CP; PI-HH) or low- (10% CP; PI-LH) CP diet and fed to achieve a predicted gain of 1.13 kg/d for the first 85 d of the experiment. Steers in the PI-HH and PI-LH feeding regimens were then offered a 13% CP diet for ad libitum consumption from d 86 to 134. Fractional protein accretion rate was greater (P < 0.01) for steers in the PI-HH and PI-LH feeding regimens than for steers in the AL regimen at d 92, 106, and 120. Fractional breakdown and synthesis rates were not affected (P = 0.63) by feeding regimen. Increased ADG and gain efficiency of steers during compensatory growth periods may in part be due to greater fractional accretion rates of skeletal muscle protein. 相似文献

The cysteine proteinases, mu- and m-calpain, along with their inhibitor, calpastatin, have been hypothesized to play a role in skeletal muscle protein degradation. Because nutrition has previously been shown to influence the expression of calpastatin, the working hypothesis of this study was that the quantity and source of dietary protein could influence regulation of the calpain system in muscle. The objectives to support this hypothesis were to determine the effects of dietary protein (amount and source) on the expression of calpastatin in canine skeletal muscle. This study comprised eight diets with seven dogs per diet. A biopsy was taken from the biceps femoris of all 56 dogs before and after 10 wk on their respective diets. This experimental design allowed examination of change within individual dogs. Diets 1 to 4 contained 12% total protein derived from chicken and/or corn gluten meal in ratios of 100:0, 67:33, 33:67, and 0:100%, respectively. Diets 5 to 8 contained 28% total protein with protein sources and ratios identical to Diets 1 to 4. Differences in calpastatin were examined qualitatively using SDS-PAGE and immunoblotting, and semiquantitatively with densitometric analyses. The majority of the calpastatin blots showed three distinct calpastatin bands, the uppermost appearing at approximately 110 kDa. Diet 5 (28% CP, 100% chicken) resulted in an increase in the expression of the 110-kDa calpastatin band compared with the other two lower molecular weight bands in the same samples. Muscle from dogs fed Diet 5 showed greater increase in (P < 0.05) calpastatin intensity of the topmost band than those fed Diet 8 (0:100; chicken:corn gluten meal). Diet 5 (100:0; chicken:corn gluten meal) showed greater total calpastatin intensity than Diet 8 (0:100; chicken:corn gluten meal). These data suggest that dogs fed a diet containing a higher total percentage of chicken protein may have a greater potential to regulate calpain-mediated degradation of muscle protein than dogs fed diets containing corn gluten meal. 相似文献

1. Male and female broiler chickens (144 in total) were given diets supplemented with clenbuterol (CB) at 0 (control) and at 1 mg/kg between 28 and 49 d of age to study the effect of CB on growth, carcase and skeletal muscle. 2. CB improved growth in males by increasing daily weight gain and final live weight and by lowering food conversion ratio. In females it changed the carcase composition by reducing abdominal fat pad and by increasing the proportion of protein. Consequently, carcase protein gain was increased in both sexes (11% and 16%, respectively). 3. Skeletal muscle weights were enhanced by between 6% and 22%. Muscle fibre diameters were increased in extensor hallucis longus (EHL) but not in gastrocnemius (GAS) muscle. This increase was more pronounced in females. EHL total muscle fibre number remained unchanged. The proportion of fast‐twitch glycolytic fibres was increased at the expense of fast‐twitch oxidative fibres in males only. Nuclear/cytoplasm and DNA/protein ratios tended to be decreased by CB. 4. From the elevated EHL muscle RNA/DNA, unchanged protein/RNA and translation activity it is suggested that CB stimulated protein synthesis at the pretranslational level. Reduced protein degradation is deduced from decreased neutral calcium‐dependent proteolytic activity. 5. It is concluded that broiler chickens respond to long‐term CB treatment as has been shown in various mammals. However, the sex‐specific response in growth, carcase composition and skeletal muscle cellularity is more clearly apparent in broiler chickens. 相似文献

1. Muscle protein turnover was measured in broilers fed on rye-based diets containing either beef tallow (T) or soybean oil (S) at an inclusion rate of 100 g/kg. Each of these diet types was tested either in the absence (S[-], T[-]) or presence (S[+], T[+]) of a xylanase-containing enzyme preparation. Protein turnover was measured in gastrocnemius muscle (GM) and pectoralis major muscle (PM). 2. Fractional rate of protein synthesis (FSR) was measured by the large dose technique using [15N]-labelled phenylalanine whereas fractional protein growth rate (FGR) was estimated by regressing tissue protein content over time. Fractional breakdown rates (FBR) were calculated by the difference between FSR and FGR. 3. In PM, FSR (%/d) was 22.1, 23.4, 21.5 and 24.4 in groups S[-], S[+], T[-] and T[+], respectively, and FBR (%/d) was 8.3, 9.8, 4.5 and 10.8 with the xylanase effect being significant. The FGR of 17.0%/d calculated for the broilers fed on the T[-] diet tended to be higher than for the other groups (13.6 to 13.8%/d). No significant effects were detected for these parameters in the GM. 4. The absolute amounts of protein which were synthesised daily and accreted in both muscles were significantly higher with xylanase supplementation in both fat type diets but at a significantly higher level when soybean oil was the dietary fat type. 5. The observed effects on protein turnover have to be seen in the context of an overall adverse effect of dietary soluble pentosans from rye in combination with tallow on physico-chemical chyme conditions, digestion and absorption of energy and nutrients and tissue-specific metabolic changes. 相似文献

The aim of the current study was to investigate the effects of a porcine growth hormone releasing hormone (pGHRH) gene plasmid injection in piglets on growth performance and whole body protein turnover. Sixty male Canadian Landrace × Chinese Taihu piglets were assigned to an intramuscular injection of 0 (control), 0.25, 0.5, 1 and 2 mg. All pigs were fed with the same diet (crude protein: 239.8 g/kg, digestible energy: 14.28 MJ/kg) at ad libitum intake. Protein turnover was determined on the 22nd day with a three-pool model by using a single-dosage, end-product analysis method with 15 N-glycine as a tracer. Injection of the pGHRH gene plasmid increased the piglets' growth rate, altered feed intake and decreased feed conversion ratio. It increased plasma growth hormone releasing hormone (GHRH), growth hormone (GH), insulin-like growth factor-I (IGF-I) and somatostatin but reduced serum urea and triglyceride. It reduced the urinary nitrogen excretion and led to higher nitrogen retention as well as the efficiencies of nitrogen retention and digestible N utilization. It increased the rates of protein synthesis, protein breakdown and net protein gain. Excretion of endogenous urinary nitrogen was reduced and nitrogen reutilization rate was improved. Conclusions: Injection of the pGHRH gene plasmid in skeletal muscle stimulated GHRH, GH and IGF-I excretion in piglets. Protein deposition was increased by an increase in protein synthesis and a smaller increase in protein breakdown, which was accompanied by reducing amino acid oxidation and increasing nitrogen reutilization. 相似文献

1. The present study was undertaken to determine the effects of dietary spirulina on growth performance and pigmentation in the muscle of growing broiler chickens and to examine the possibility that zeaxanthin in spirulina may affect yellow colour development in the meat. 2. Twenty-four, 21-d-old, male broiler chicks were fed an experimental diet containing spirulina at 0, 40, or 80 g/ kg for 16 d. No significant differences among treatments were observed in body weights, nor weights or yields (as a percentage of body weight) for any of the selected traits, including liver, abdominal fat, kidney and Pectoralis profundus. 3. Spectrocolourimetric analyses revealed that the redness of Pectoralis superficialis, profundus and Sartorius muscles reached a maximum in chicks fed the 40 g/kg spirulina diet, while the yellowness of all fillets, including the Semitendinosus muscle, increased in a sub-linear fashion with increased spirulina in the diet. The overall correlation between the yellowness and zeaxanthin content in the Pectoralis muscle was significant. 4. This study provides the first conclusive evidence that dietary spirulina influences both the yellowness and redness of broiler flesh and that the increments in yellowness with dietary spirulina content may possibly be reflected in the common yellow pigment related to the accumulation of zeaxanthin within the flesh. 相似文献

1. The aim of the study was to investigate the effects of dietary threonine (Thr) and crude protein (CP) in maize-soybean meal based diets on the growth performance, carcase traits and meat composition of broiler chickens and to determine the dietary Thr requirement for optimum performance (weight gain and feed conversion efficiency (FCE)) at 0 to 3 weeks and 3 to 6 weeks of age. 2. Two basal diets that differed in CP (191.3 or 179.7 and 176.7 or 165.4 g/kg at 0 to 3 and 3 to 6 weeks, respectively) were formulated to have identical contents of Thr (6.0 and 5.4 g/kg), energy (12.97 and 13.39 MJ ME/kg) and other essential amino acids except for Gly + Ser. Basal diets were supplemented with L-Thr from 0.6 to 1.8 g/kg in 0.6 g/kg increments. Broiler chicks (540) were randomly allocated to 9 dietary treatments with 6 replicates of 10 (5 female, 5 male) chicks. 3. A significant interaction between dietary CP and Thr was found for feed intake, body weight (BW) gain and FCE. Increasing Thr supplementation improved feed intake, BW gain and FCE, especially in high CP diets in both feeding periods. 4. Incremental increases in dietary Thr increased breast yield at both CP levels and drumstick yield only on high CP diets. The proportion of thigh decreased with Thr concentration. Liver weight was significantly reduced by Thr supplementation; abdominal fat was not affected. 5. Estimated Thr requirements for FCE increased as dietary CP increased according to an exponential model. 1. In an experiment on broiler cockerels, the influence of tannic acid (TA) and polyethylene glycol (PEG) on body weight gain (BWG), feed conversion ratio (FCR), weight of intestine and liver, the activities of serum enzymes LDH, AST, ALT and intestinal absorption function were investigated. 2. Broiler cockerels were given either a commercial diet alone (control group) or a commercial diet with TA (20 g/kg), PEG (10 g/kg) or TA plus PEG (20 + 10 g/kg), for 10 d. 3. On the last day of the experiment, all birds and remaining feed were weighed individually and a sample of blood was taken to measure the serum activities of lactate dehydrogenase (LDH), aspartate aminotransferase (AST) and alanine aminotransferase (ALT). The capacity of intestinal cells for the absorption of D-xylose was measured. Finally all birds were killed humanely and the intestine and liver were weighed. 4. The results showed that TA significantly reduced BWG and FCR, as well as the activity of LDH, AST and ALT. 5. TA also increased the relative weight of the intestine. Adding PEG alone had no effect on any of the measured parameters. 6. However, PEG improved significantly BWG, FCR and the activity of LDH and AST of TA-fed birds. 7. The plasma D-xylose concentration of experimental birds was similar for all dietary treatments most likely because of temporal separation between feeding the dietary TA and administering the D-xylose. 8. It was concluded that the presence of tannins in the GI lumen of the bird was necessary to affect the processes involved in the absorption of simple sugars such as D-xylose, at the level of intestinal absorptive cells. 相似文献
