Elicitors of systemic acquired resistance are well known to reduce severity of several plant pathogenic diseases caused by fungi, bacteria and viruses. Their field applications for management of plant diseases are, however, limited because of yield penalties. Our studies on affect of Benzo (1,2,3)-thiadiazole-7-carbothioic acid S-methyl ester (BTH), an elicitor of systemic acquired resistance, on chickpea blight caused by a fungal pathogen Didymella rabiei showed that multiple foliar applications of the chemical were effective in management of the disease under economic threshold levels. Multiple applications, however, affected chickpea grain yield adversely. The BTH induced yield penalties could be prevented by foliar spray schedule comprised of BTH and a contact fungicide mancozeb. One spray of BTH (50 ppm) followed by another of mancozeb (0.2%) was less effective (8.3% severity) than three sprays of BTH (4.2% severity) in blight control, however, this treatment enhanced grain yield significantly (1.241 t ha−1) over three sprays of BTH (0.922 t ha−1).  相似文献

This study aimed to determine the resistance/susceptibility to fire blight of apple germplasm resources from the province of Erzincan in eastern Turkey. In total, 32 native apple accessions from four regions within the province were tested under greenhouse conditions by inoculating the shoot tips with pathogenic bacterium. Shoot tips were wounded for inoculation. Genotypical susceptibility index (GSI%) values were computed for each genotype based on the length of the lesion that developed on each shoot. Accessions were grouped into five classes of resistance/susceptibility, as follows: Class A (resistant); Class B (moderately resistant); Class C (moderately susceptible); Class D (susceptible); and Class E (highly susceptible). GSI% values differed significantly among accessions (p < 0.01). Five accessions received a rating of Class A, and 7 accessions received a rating of Class B. This was followed by 9 accessions with a rating of Class C, 5 accessions with a rating of Class D and 6 accessions with a rating of Class E. The findings of this study are expected to contribute to breeding efforts with respect to apple resistance to fire blight.  相似文献

Factors influencing in vitro regeneration through direct shoot bud induction from hypocotyl explants of Jatropha curcas were studied in the present investigation. Regeneration in J. curcas was found to be genotype dependent and out of four toxic and one non-toxic genotype studied, non-toxic was least responsive. The best results irrespective of genotype were obtained on the medium containing 0.5 mg L−1 TDZ (Thidiazuron) and in vitro hypocotyl explants were observed to have higher regeneration efficiency as compared to ex vitro explant in both toxic and non-toxic genotypes. Adventitious shoot buds could be induced from the distal end of explants in all the genotypes. The number of shoot buds formed and not the number of explants responding to TDZ treatment were significantly affected by the position of the explant on the seedling axis. Explants from younger seedlings (≤15 days) were still juvenile and formed callus easily, whereas the regeneration response declined with increase in age of seedlings after 30 days. Transient reduction of Ca2+ concentrations to 0.22 g L−1 in the germination medium increased the number of responding explants.Induced shoot buds, upon transfer to MS medium containing 2 mg L−1 Kn (Kinetin) and 1 mg L−1 BAP (6-benzylamino purine) elongated. These elongated shoots were further proliferated on MS medium supplemented with 1.5 mg L−1 IAA (indole-3-acetic acid) and 0.5 mg L−1 BAP and 3.01-3.91 cm elongation was achieved after 6 weeks. No genotype specific variance in shoot elongation was observed among the toxic genotypes except the CSMCRI-JC2, which showed reduced response. And for proliferation among the toxic genotypes, CSMCRI-JC4 showed highest number of shoots formed. Among the rest, no significant differences were observed. The elongated shoot could be rooted by pulse treatment on half-strength MS medium supplemented with 2% sucrose, 3 mg L−1 IBA (indole-3-butyric acid), 1 mg L−1 IAA, 1 mg L−1 NAA (α-naphthalene acetic acid) and subsequent transfer on 0.25 mg L−1 activated charcoal medium. The rooted plants could be established in soil with more than 90% success. No significant differences were observed in rooting of shoots in the different toxic genotypes. However, rooting response was reduced in non-toxic genotype as compared to toxic genotypes.  相似文献

A novel bacterial blight (BB) resistance gene, Xa23, identified from Oryza rufipogon was introgressed into three popular restorer lines (Minghui63, YR293 and Y1671) for wild abortive cytoplasmic male sterility by marker-assisted backcross breeding approach in combination with artificial inoculation and stringent phenotypic selections. The three derived BB resistant restorer lines (Minghui63-Xa23, YR293-Xa23 and Y1671-Xa23) and their hybrid combinations with Zhenshan97A (Shanyou63-Xa23), NongfengA (Fengyou293-Xa23) and Zhong9A (Zhongyou1671-Xa23) demonstrated similar BB resistance spectrum as the donor parent, CBB23 (B). The newly developed BB resistant restorers and their derived hybrids were identical to their respective original versions for agronomic traits especially under disease free condition. However, under severe disease condition, the three BB resistant restorer lines exhibited significantly higher grain weight and spikelet fertility as compared to the respective original restorer lines thus further resulting in BB resistant hybrids with significantly higher grain yields than their respective popular original hybrids. The results indicated that the Xa23 gene could completely express its dominant broad spectrum resistance in different backgrounds of both restorer and male sterile lines across different growth stages, suggesting its immense breeding value in BB resistance improvement for hybrid rice. Moreover, a reasonable utilization and deployment of Xa23 gene for efficient control of BB disease in hybrid rice production was recommended.  相似文献

Talaromyces flavus a fungal antagonist, was isolated from soil samples collected from potato fields in Varamin and Karaj districts, Tehran province, Iran. Antagonistic effects of T. flavus isolates against Verticillium albo-atrum, the causal agent of potato wilt disease were investigated in the laboratory and greenhouse conditions. T. flavus colonies were recovered after three weeks from soil samples cultured on selective medium. Antagonistic effects of volatile and non-volatile extracts of T. flavus isolates on V. albo-atrum growth were investigated in the laboratory and five that caused higher growth inhibition of V. albo-atrum, were selected for greenhouse experiments. Infection index was compared in the greenhouse in a split plot trial with five isolates applied to soil, seed, or both arranged in a randomized complete block design with four replications. The minimum infection index was observed when seed were treated with T. flavus with the most effective isolate being Tf-Po-V-52. On seed, the minimum infection index was observed with Tf-Po-V-50. The most effective T. flavus isolate was also evaluated in a field experiment. Results indicated that infection index in seed treatment contained this isolate was 0.15 whereas that of control was 3.5. The overall results of this study showed that it may be possible to manage potato Verticillium wilt disease effectively by using T. flavus, a biocontrol fungus.  相似文献

The legume pod borer Helicoverpa armigera (Hubner) (Lepidoptera: Noctuidae) has developed high levels of resistance to conventional insecticides, and therefore, efforts are being made to develop transgenic chickpea expressing toxin genes from the bacterium Bacillus thuringiensis (Bt) for controlling this pest. However, there is an apprehension that acid exudates in chickpea might interfere with the biological activity of Bt. Therefore, we studied the biological activity of Bt (BiolepR) on four chickpea genotypes with different levels of resistance to H. armigera under field conditions, and by incorporating lyophilized leaf and pod tissue into the artificial diet with and without Bt. The pH of the acid exudates varied from 2.1 to 2.9, and malic and oxalic acids were the major components of the acid exudates in different chickpea genotypes. There was no survival of H. armigera larvae in chickpea plants sprayed with 0.1, 0.2 and 0.5% Bt. There was a significant reduction in larval survival, larval and pupal weights and fecundity, and prolongation of larval and pupal periods in chickpea plots sprayed with Bt (0.05%) as compared to the unsprayed plots. Biological activity of Bt was lower on artificial diets with leaf or pod powder of chickpea genotypes, which might be because of a low intake of Bt toxins due to the antifeedant effects of acid exudates in the chickpea or reduction in biological activity of Bt due to the interaction of biochemical constituents in chickpea with the Bt toxins. Larval survival, larval and pupal weights, pupation and adult emergence were significantly lower on diets with leaf or pod powder of the H. armigera-resistant genotypes than on the susceptible check. Chickpea genotypes with resistance to H. armigera acted in concert with Bt to cause adverse effects on the survival and development of this insect. The results suggested that development of transgenic chickpeas expressing toxin genes form Bt will be quite effective for controlling of the pod borer, H. armigera.  相似文献

Root nodulating Sinorhizobium fredii KCC5 and Pseudomonas fluorescens LPK2 were isolated from nodules of Cajanus cajan and disease suppressive soil of tomato rhizosphere, respectively. Both strains produced IAA, siderophore, solubilized insoluble phosphate, showed chitinase and β-1,3-glucanase activities, and strongly inhibited the growth of Fusarium udum. It also caused degradation and digestion of cell wall components, resulting in hyphal perforations, empty cell (halo) formation, shrinking and lysis of fungal mycelia along with significant degeneration of conidia. LPK2 produced volatile cyanogen (HCN). Combinations of S. fredii KCC5 and P. fluorescens LPK2 with half dose of chemical fertilizers showed a significant increase in seed germination (94%) while seed germination with co-inoculated strains (KCC5 + LPK2), KCC5 and LPK2 alone was 90, 84 and 82% respectively as compared to control 77%. After 120 days of sowing, per plant number of pods, nodules, shoot length, root length, shoot weight and root weight were greater for the combination with half dose of chemical fertilizers compared to the control. Combinations of S. fredii KCC5 and P. fluorescens LPK2 with half dose of chemical fertilizers resulted in an 82% increase in grain yield per hectare compared to the control. Both strains KCC5 and LPK2 led to proto-cooperation as evidenced by synergism, aggressive colonization of the roots, and enhanced growth, suggesting potential biocontrol efficacy against Fusarium wilt in C. cajan.  相似文献

The effects of extracts of different parts of the perennial tropical plant Balanites aegyptiaca (L) Del., including various solvent extracts of roots, methanol extracts from leaves, fruits, flowers and roots, partially purified saponins obtained from its roots and a standard saponin were studied on the life cycle (adult longevity, number of eggs, crawlers, adults, weight of adults and % wax content) of a laboratory-reared parthenogenic line of the mealy bug, Maconellicoccus hirsutus (Homoptera: Pseudococcidae). Extracts derived from various parts of B. aegyptiaca (leaves, fruits, flowers, and roots in methanol) affected the life cycle of M. hirsutus with a methanol root extract being the most effective at a concentration of 500 μg ml−1. Partially purified saponin of B. aegyptiaca and the commercial bark saponin extract (Sigma) from Quillaja saponaria at a concentration of 500 μg ml−1 were effective in reducing the longevity of M. hirsutus. Significant reductions in oviposition by M. hirsutus were found for all the extracts at a concentration of 500 μg ml−1. Extracts also affected the number of emerging crawlers, number of adults as well as the weight and wax content of emerging adults. These studies suggest that B. aegyptiaca plant extracts and saponins can be useful botanical insecticides for the protection of crops from mealy bugs.  相似文献

A local isolate of Trichoderma asperellum was tested for its antagonistic activity against Thielaviopsis paradoxa (telemorph = Ceratocyctis paradoxa). The highest antagonistic activity was achieved when the concentration of T. asperellum conidia was 1 × 107 conidia/mL. The highest biomass and number of colony forming unit/mL of the T. asperellum peaked at 144 h after incubation in yeast waste residue medium. The minimum inhibition concentration value of the formulation was observed as 1% on growth of Th. paradoxa incubated at 28 ± 2 °C for 10 d. In the soil fungicide-screening test, the effect of concentrations 100-1600 μg/mL on mycelia growth was not significant (P < 0.05). Complete mycelial growth inhibition occurred at concentration above 52,600 μg/mL. Results of the fruit application tests clearly showed that all treated fruits were free of disease at the end of the incubation period. No significant differences (P > 0.05) in pH, total soluble solids and titratable acidity were observed between fruits treated with formulation of T. asperellum and the control formulation treated pineapples.  相似文献

Polianthes tuberosa is a commercially valuable flower crop in the Mekong Delta of Vietnam that is propagated by the harvesting and planting of bulbs. Cultivation of P. tuberosa is infected by an endemic Aphelenchoides besseyi nematode that damages a high proportion of plants and persists within the bulbs. Here we report on the comparison of hot water and pesticide treatments as control methods to protect P. tuberosa from A. besseyi damage, and conclude that a hot water treatment consisting of soaking bulbs in water for 30 min at 57 °C is the most efficacious method to produce healthy flowers in a cost effective manner.  相似文献

The overreliance on and overuse of fungicides is not only a health hazard but also induces natural resistance in plant pathogens, resulting in an economic burden on agricultural producers and a potential threat to natural systems across the globe. It is therefore necessary to identify natural substitutes of fungicides. This study was designed to evaluate the inhibitory effect of the root exudates of two different garlic cultivars, cv. Gailiang (G064) and Cangshan (G025), against Phytophthora capsici, a pepper fungus. All treatments (T1 to T4, i.e., 25%, 50%, 75% and 100% root exudate concentrations) of garlic cultivar G025 showed lower inhibition effects than cultivar G064. An intervarietal inhibition effect comparison at 100% concentration (T4) exhibited a 69.24% decline in hyphal growth for G064 compared with 49.06% for G025. The mycelial growth measured in the control was found to be significantly greater compared with the garlic treatments. The results of high performance liquid chromatography (HPLC) revealed that G064 possessed a large amount of allicin compared with G025. Scanning and transmission electron microscopy showed that, compared with the controls and hyphae treated with G025, the fungal hyphae treated with G064 exudates were misshaped, fragmented and had a smaller diameter, as well as empty cytoplasmic contents in the cell wall. Thus, the root exudates of the G064 cultivar had a significant fungicidal effect on P. capsici.  相似文献

The effects of four host plants (Agave sisalana, Ag. americana var. marginata, Ananas comosus Baili and Ancomosus Smooth Cayenne) on the biology of the mealybug Dysmicoccus neobrevipes Beardsley were studied in the laboratory at 26 ± 1 °C, 75-90% RH and 14:10 (L:D) photoperiod. The development, survivorship, longevity, reproduction and life table parameters of D. neobrevipes differed among the host plants. The shortest developmental period (from the first instar nymph to adult) was recorded on Ancomosus Smooth Cayenne (22.4 days for females and 21.3 days for males), whereas the longest was recorded on An. comosus Baili (25.6 days for females and 24.7 days for males). The highest survivorship was found on An. comosus Baili (98% for both females and males) and the lowest was on Ag. americana var. marginata (39.6% for females and 50% for males). Meanwhile the sex ratio and fecundity were highest and the pre-lay period was shortest on Ag. sisalana. The longest longevity of females was 62.5 days on Ancomosus Baili, whereas the other host plants did not differ significantly with grand mean longevities of 51.0 days for females, while the longest and shortest longevities of males were 4.6 days and 2.3 days on Ag. americana var. marginata and Ag. sisalana, respectively. Values for net reproductive rate, intrinsic rate of increase and finite rate of increase were highest on Ag. sisalana, whereas the mean generation time was shortest on An. comosus Smooth Cayenne. The results indicated that Ag. sisalana is the most suitable host for D. neobrevipes among the four tested plants. When reared on Ag. sisalana, D. neobrevipes had a short developmental period (females 22.7 days and males 23.8 days), high reproduction (418 nymphs/female) and a high intrinsic rate of increase (0.106). Results of this study indicated that host plant can largely influence the population dynamics of D. neobrevipes, and our findings are useful in understanding the roles of host plants in integrated management of this pest, including exploitation of these host plants in push-pull control.  相似文献

The influence of leaf surfaces of different host plants on the biology and life table parameters of the predacious mite, Agistemus exsertus Gonzalez was studied in the laboratory. A. exsertus successfully developed and reproduced on all tested plant leaves. Smooth, glabrous pear leaves accelerated the development of A. exsertus and showed the shortest mean generation time. Slightly rough, slightly hairy leaves of mulberry and acalypha were the most suitable surface for oviposition of the predator. These leaves exhibited the highest fecundity and net reproductive rate. On the other hand, leaves of rough, pubescent apple and scabrous, bristly lantana were the least suitable surface for oviposition of A. exsertus. They showed the lowest fecundity and net reproductive rate. Acalypha leaves exhibited the highest intrinsic rate of increase for the predator, whereas lantana leaves showed the lowest value. This difference in the predator response to leaf surfaces of host plants should be considered to enhance the role of the predator in biological control programs.  相似文献

European tarnished plant bug, Lygus rugulipennis, is a serious pest on late season strawberries in the UK. Feeding by the pest on the developing fruits causes severe malformation, and over 50% of fruit may be downgraded as a result of capsid feeding in unsprayed crops. A range of compounds was tested against the pest over two years at East Malling Research. In 2008 experiments were conducted on weed plots with high L. rugulipennis populations, and in 2009 were conducted on an everbearer strawberry planting. The 2008 experiments showed that etoxazole had no effect on L. rugulipennis and that although flonicamid and indoxacarb significantly reduced populations they were not as effective as thiacloprid, either with or without a surfactant, or acetamiprid. In the 2009 experiment on strawberry thiacloprid + surfactant, applied twice, was more effective than this treatment applied once, which in turn was more effective than thiacloprid alone. In both years the most effective treatment was the pyrethroid bifenthrin. There was no suggestion that different developmental stages of the pest were more susceptible to the tested insecticides. Thus there was no evidence to suggest that timing applications against specific life stages would significantly improve pest control.  相似文献

The effect of electrolyzed oxidizing (EO) water in combination with ozone to control postharvest decay of tangerine cv. “Sai Num Pung” was investigated. The spore suspension containing 105 conidia ml−1 of Penicillium digitatum was prepared. EO water was generated by electrolysis of various concentrations of NaCl solution (5, 25, 50% and saturated NaCl). The spore suspension was inoculated into EO water and incubated at 27 °C for 1, 2, 4, 8 and 32 min. It was found that the EO water with saturated NaCl completely inhibited the spore germination of the fungus within 1 min. When the fruits inoculated with P. digitatum were washed in EO water at the same concentrations as previous experiment for 4, 8 and 16 min and stored at 5 °C for 18 days, it was found that immersion of the fruit in EO water for 8 min was the most effective to reduce disease incidences. Moreover, washing fruit in EO water and kept in a refrigerated chamber at 5 °C with continuous ozone exposure at a concentration of 200 mg l−1 for 2 h day−1 to extend storage life suppressed the disease incidence until 28 days. However, none of the treatments had any effect on the quality of fruit such as total soluble solids, titratable acidity, percent weight loss and peel color. Therefore EO water may be useful for surface sanitation and ozone has potential to control the recontamination of postharvest diseases in tangerine fruit in storage room.  相似文献

The aims of this work were to introduce Lippia junelliana into cultivation, to compare the essential oil accumulation between cultivated and wild plants, and to reintroduce micropropagated plants in the location of the original population. The leaves and inflorescences of cultivated plants accumulated, on a dry weight basis, higher amounts of essential oil than their wild counterparts. Thus, total essential oil accumulation of cultivated plant parts was also significantly higher than that of wild counterparts. The cultivated plants showed the same essential oil profile than the wild plants. This work demonstrates that cultivation can be a more efficient vehicle to both preserve and exploit L. junelliana, than collection from the wild, because higher yields of biomass and oil accumulation can be achieved, while essential oil composition is less affected by the different treatments. The reintroduction of new plants into the species’ original location has proved to be a viable alternative for their in situ preservation or enrichment planting. This model of introduction of aromatic plants into cultivation through micropropagation could be a useful technique to recover valuable chemotypes from the wild in the search for new alternatives in the agriculture and for the preservation of natural resources for future generations.  相似文献
